Beta-carotene functions independently of vitamin A in the reproductive performance of dairy cows. The concentrations of beta-carotene in plasma decrease during the dry period, and reach a nadir in about the first week postpartum. This coincides with a negative energy balance, which affects the onset of the first ovulation in early postpartum cows. Thus, we hypothesised that plasma beta-carotene concentrations during the peripartum period may affect ovulation in the first follicular wave postpartum in dairy cows. The aim of the present study was to investigate changes in the profiles of plasma beta-carotene concentrations during the peripartum period in ovulatory and anovulatory cows during the first follicular wave postpartum. We used 22 multiparous Holstein cows, which were fed a total mixed ration consisting of grass, corn silage and concentrate, and collected blood samples for beta-carotene and progesterone analysis from week 3 prepartum to week 3 postpartum when the period of day 0-6 after parturition was regarded as the parturient week (week 0). The first ovulation was confirmed using the profile of plasma progesterone concentrations and colour Doppler ultrasound. Thirteen cows ovulated during the first postpartum follicular wave. Parity, the dry-off period, calving interval, mastitis episodes, and actual 305 days' milk yield during the previous lactation, and milk composition in the last month during the previous lactation in this study did not differ between ovulatory and anovulatory cows. Differences in the plasma beta-carotene profile were observed between ovulatory and anovulatory cows. Plasma beta-carotene concentrations at week 3 prepartum were greater in ovulatory cows (2.97+/-0.24 mg/L) than in anovulatory cows (1.53+/-0.14 mg/L; P<0.001), after that its concentrations in ovulatory cows decreased and reached the lowest level at week 1 postpartum, although its concentrations in anovulatory cows remained unchanged. No differences in plasma beta-carotene concentrations between the two groups were observed postpartum. The present study indicates for the first time that the lower beta-carotene concentrations in plasma during the prepartum period is associated with anovulation during the first follicular wave postpartum. 相似文献

Two studies were conducted to evaluate the concentration of serum progesterone in pyometritic cows and relate it to palpation of ovarian structures per rectum . In Trial 1, serum samples from 34 pyometritic cows were assayed for progesterone. Mean serum progesterone concentrations were 6.8 +/- 0.7 ng/ml. In Trial 2, each of 54 pyometritic cows was paired with a control cow on the basis of days post partum (18-50 days). Mean concentration of progesterone was 9.7 +/- 1.0 ng/ml for the pyometritic cows and 5.7 +/- 0.8 ng/ml in control cows (P<0.005). Progesterone concentration was greater (P<0.005) in both groups of cows with palpable corpora lutea (CLs). Ninety-six percent of the pyometritic cows had palpable CLs compared to 57% of the control group. Comparing serum progesterone only in cows with a palpable CL, the mean concentration was still greater (P<0.005) in the pyometritic group (10.6 +/- 1.0 ng/ml) than the control group (6.6 +/- 1.0 ng/ml). Compatability of rectal palpation findings and concentrations of serum progesterone were 92.6% for the pyometritic group and 72.2% for the control group. Progesterone concentration increased (P<0.05) by increased days post partum in Trial 2 (n=54) but not in Trial 1 (n=23). In both Trials 1 and 2, uterine size due to pyometra increased (P<0.05) with increased days post partum. No other associations were found. 相似文献

The effect of ketone body status on occurrence of first ovulation during early lactation was assessed in 84 multiparous dairy cows under field conditions. Animals were equally distributed across 8 farms and were controlled by the same herd fertility monitoring program. Cows were visited twice antepartum and 6 times postpartum at weekly intervals between 5:30 and 8:30 AM. On these occasions, body condition scores and milk yields were measured, blood and milk samples were taken, cows were gynecologically examined, and parameters of reproduction were determined. The onset of first ovulation was specified by milk progesterone determination and rectal palpation. Cows starting postpartum ovarian cyclicity within or after 30 d were classified as early and late responders (ER and LR, respectively). Resumption of the estrous cycle within 30 d postpartum is considered optimal under practical conditions, and classification based on this threshold value resulted in groups of equal size and equal distribution of ER + LR cows within farms. Ketone bodies measured were beta-hydroxybutyrate in serum and acetoacetate and acetone in serum and milk. Blood serum and milk ketone body concentrations during the first 6 wk of lactation were higher in LR than in ER, whereas plasma glucose and nonesterified fatty acid and milk fat, protein and urea concentrations did not differ between groups. Maximal concentrations of ketone bodies from parturition to first ovulation were better predictors of the onset of the estrous cycle than mean or minimal concentrations over the same period. Milk acetone and serum beta-hydroxybutyrate concentrations provided the most reliable information with regard to resumption of ovarian activity of all ketone bodies. 相似文献

Luteolysis in the cow depends upon an interaction between prostaglandin F(2alpha) (PGF(2alpha)) and oxytocin. The objectives of our study were 1) to determine oxytocin concentrations in postpartum dairy cows and 2) to identify the temporal relationship between oxytocin and PGF(2alpha) release patterns during luteolysis in normal and abbreviated estrous cycles in the postpartum period. Serum oxytocin and PGF(2alpha) metabolite (PGFM) concentrations from nine cows which had short estrous cycles (< 17 d) were compared with those of six cows which had normal estrous cycles. Serum basal oxytocin concentrations in short estrous cycle cows (23.7 to 31.1 pg/ml) were higher (P<0.05) than those of normal estrous cycle cows (14.6 to 19.8 pg/ml). Oxytocin concentrations increased to peak values in both short and normal cycle cows, during luteolysis. Basal PGFM concentrations (112.2 to 137.4 pg/ml) were higher in cows with short cycle (P<0.05) than in cows with normal cycles (62.9 to 87.5 pg/ml). The increase in PGFM concentrations during luteolysis was significant in both normal cycle and short cycle cows (P<0.05). Increases in serum PGFM concentrations were always associated with increases in serum oxytocin concentrations in normal cycle and short cycle cows and the levels decreased simultaneously before the subsequent estrus. Results support the idea of a positive relationship between PGF(2alpha) and oxytocin concentration during the estrous cycle as well as a possible synergistic action of these hormones in the induction of luteolysis in dairy cattle. 相似文献

The objective of this study was to investigate the effect of time of first postpartum ovulation after calving on uterine involution in dairy cows with and without uterine puerperal disease. Transvaginal follicular puncture (FP) of follicles >6 mm suppressed ovulation and development of a CL until Day 42 after calving. Fifty-three lactating Holstein Friesian cows (3.4 ± 1.2 years old, parity 2.5 ± 1.0 [median ± mean absolute deviation]) were divided into groups on the basis of the presence (UD+) or absence (UD−) of uterine disease and whether FP was carried out (FP+) or not (FP−). Uterine disease was defined as the occurrence of retained fetal membranes and/or metritis. This resulted in the following groups: UD−FP− (n = 15), UD−FP+ (n = 13), UD+FP− (n = 13), and UD+FP+ (n = 12). A general examination, vaginoscopy, transrectal palpation, and transrectal B-mode sonography of the reproductive organs were conducted on Days 8, 11, 18, and 25 and then every 10 days until Day 65 after calving. After hormonal synchronization of ovulation (cloprostenol between Days 55 and 60 postpartum and GnRH 2 days later), cows were inseminated in the next spontaneous estrus. On average, the cows ovulated on Day 21.0 ± 6.0 (UD−FP−), 50.0 ± 4.0 (UD−FP+), 16.0 ± 3.0 (UD+FP−), and 48.0 ± 2.0 (UD+FP+) postpartum. Calving-to-conception interval and first-service conception rates were not affected by FP (P > 0.05). Healthy cows with FP had smaller (P < 0.05) uterine horn and cervical diameters assessed sonographically than cows without FP. FP reduced the prevalence of purulent vaginal discharge and uterine size assessed transrectally in UD+ cows (P < 0.05). The results showed that suppression of an early ovulation by transvaginal FP improved uterine involution in cows with and without uterine disease. 相似文献

In the present study we investigated the effect of hCG administration on Day 7 (Day 0 = day of standing estrus) to ovulate the dominant follicle of the first wave and the associated increase in progesterone concentration on subsequent superovulatory response in dairy cows. Twenty cyclic lactating cows were allocated at random to 2 groups: control (n = 10) and hCG-treated (n = 10). The ovaries of each cow were scanned using an ultrasound scanner on Day 7, to confirm the presence of the dominant follicle and thereafter every other day until embryo recovery. All cows received a total dose of 400 mg Folltropin-V in decreasing amounts for 5 days (Days 9 to 13) and 35 mg PGF(2alpha) on Day 12. In addition, the treated cows received 1000 IU hCG on Day 7. All cows were inseminated twice during estrus, and the embryos were collected 7 days later by a nonsurgical procedure. Blood smaples were taken at different times of the treatment period for progesterone determination. All cows possessed a dominant follicle at Day 7, and all but one of the hCG-treated cows ovulated the dominant follicle and formed an accessory corpus luteum. Plasma progesterone concentrations were significantly higher (P<0.01) in hCG-treated cows than control cows on the first day of Folltropin treatment and on the day of PGF(2alpha) injection. The mean number of follicles at estrus, the number of ovulations, the total number of embryos and the number of transferable embryos were not different (P>0.05) between control and hCG-treated cows. 相似文献

The efficiency of a synthetic prostaglandin analog (ICI 80996) for the control of the estrus cycle in dairy cows has been tested with and without simultaneous use of progestagen implants (SC 21009, Searle). Synchronization of estrus is closer after combined progestagen-prostaglandin treatments than after prostaglandins alone (78.7 % in estrus in 48h vs 60.2 % ; P<0.001).The percentage of cows not observed in estrus during the first 96h after the end of treatment was also lower after combined treatments (20.7 % vs 31.8 % ; P<0.05). The best estrus synchronization was obtained by injecting prostaglandin analog two days before implant removal (86 % in estrus in 48h and 14.0 % not observed in estrus within 96h).Fertility was studied in a field trial after one or two inseminations at a predetermined time for each treatment. A higher pregnancy rate was obtained with two inseminations after combined progestagen-prostaglandin treatment (49.1 %). This pregnancy rate differed significantly (P<0.05) from those obtained with one A.I. after the same treatment or two A.I. after the prostaglandin treatment but did not differ from the pregnancy rate obtained with one A.I. 80h after the second prostaglandin injection. The most important problem in dairy cows is the fall in pregnancy rate between 21 days and 6 months (19.7 to 35.1 % depending on treatment ; P<0.05). 相似文献

Eleven 3-yr-old crossbred Angus cows that had raised one calf each were mastectomized at 6 to 7 mo of their second gestation. All calves were removed from cows within 12 h after birth. Cows were kept in a drylot with a fertile bull for 60 d after parturition. Cows were observed for estrus every 6 h, and blood serum was collected daily and assayed for progesterone. The average first ovulation and first estrus occurred at 13.9 and 20.1 d after parturition, respectively. Nine of 11 cows conceived, and the average time of conception was 34.3 d after parturition. The next year, eight of the same mastectomized cows were allocated to two equal groups. In one group, calves were removed from cows within 12 h after birth, whereas in the other group, calves remained with cows for 46 to 53 d. Calves that remained with their dams were hand fed from a bottle or bucket every 12 h. The two groups of cows were kept in separate drylots about 50 m apart, and a fertile bull was kept with each group. Blood samples were obtained from all cows, and they were observed for estrus as before. Cows withcut calves ovulated before 22 d (average 16.0), exhibited estrus by 33 d (average 24.0), and conceived by 40 d (average 30.5) after calving. None of the cows with calves ovulated or exhibited estrus earlier than 49 d after calving. However, all cows ovulated by 4 d (average 3), exhibited estrus by 10 d (average 5.5), and conceived by 11 d (average 9.3) after calf removal. We conclude that cow-calf interaction can suppress ovulation and estrus even when suckling and lactation do not take place. 相似文献

A strategy widely adopted in the modern dairy industry is the introduction of postpartum health monitoring programs by trained farm personnel. Within these fresh cow protocols, various parameters (e.g., rectal temperature, attitude, milk production, uterine discharge, ketones) are evaluated during the first 5 to 14 days in milk (DIMs) to diagnose relevant diseases. It is well documented that 14% to 66% of healthy cows exhibit at least one temperature of 39.5 °C or greater within the first 10 DIM. Although widely adopted, data on diagnostic performance of body temperature (BT) measurement to diagnose infectious diseases (e.g., metritis, mastitis) are lacking. Therefore, the objective of this study was to identify possible factors associated with BT in postpartum dairy cows. A study was conducted on a commercial dairy farm including 251 cows. In a total of 217 cows, a vaginal temperature logger was inserted from DIM 2 to 10, whereas 34 cows did not receive a temperature logger as control. Temperature loggers measured vaginal temperature every 10 minutes. Rectal temperature was measured twice daily in all cows. On DIM 2, 5, and 10, cows underwent a clinical examination. Body temperature was influenced by various parameters. Primiparous cows had 0.2 °C higher BT than multiparous cows. Multiparous cows that calved during June and July had higher BT than those that calved in May. In primiparous cows, this effect was only evident from DIM 7 to 10. Furthermore, abnormal calving conditions (i.e., assisted calving, dead calf, retained placenta, twins) affected BT in cows. This effect was more pronounced in multiparous cows. Abnormal vaginal discharge did increase BT in primiparous and multiparous cows. Primiparous cows suffering from hyperketonemia (beta-hydroxybutyrat ≥ 1.4 mmol/L) had higher BT than those not affected. In multiparous cows, there was no association between hyperketonemia and BT. The results of this study clearly demonstrate that BT is influenced by various parameters in dairy cows. Therefore, these parameters have to be considered when interpreting measurements of BT in dairy cows. This information helps to explain the high incidence of type I and II errors when measuring BT and clearly illustrates that measures of BT should not be used as a single criterion to decide whether or not to provide antibiotic treatment to dairy cows. However, research-based test characteristics of other parameters (e.g., vaginal discharge) alone or in combination with BT are still lacking. 相似文献

This paper briefly reviews recent data and concepts on the development and mitigation of infection and inflammation in the reproductive tract of dairy cows during the first 2 mo after calving. The incidence of metritis is typically between 10 and 20%, of clinical endometritis or purulent vaginal discharge (PVD) approximately 15%, and of subclinical or cytological endometritis a further 15%. Worse postpartum negative energy balance is associated with more severe or prolonged uterine inflammation. Changes in feed intake, expression of genes for pro-inflammatory cytokines, notably interleukin (IL) 1, IL6 and IL8, circulating concentrations of beta-hydroxybutyrate (BHBA) or nonesterified fatty acids (NEFA), and innate immune function precede both metritis and endometritis by several weeks. Infections with Escherichia coli and Arcanobacterium pyogenes are associated with both metritis and PVD. There are new data to suggest that specific virulence factors in E. coli associated with adherence may be important in metritis and PVD. Cytological endometritis and PVD are overlapping but largely distinct conditions, and there are emerging data that cervicitis exists both concurrent with and separate from endometritis. Much remains to be learned about what initiates and sustains harmful inflammation of the reproductive tract. Such information is necessary to develop effective treatments for the various forms of disease and, more importantly, to develop means to prevent endometritis and cervicitis. In particular, vaccination against specific uterine pathogens and interventions to modulate innate immune response appear to be important avenues for investigation. Presently, commonly recommended best management practices for cows in the transition period are likely to be helpful to mitigate the risk of reproductive disease. 相似文献

The aim of the present study was two-fold. First, to characterize the secretory profiles of oestradiol-17beta and progesterone in relation to the structural changes observed by ultrasonography during follicular dynamics in non-ovulating llamas. Second, to evaluate the effect of exogenous progesterone on follicular activity, in terms of follicle development and hormone production. In experiment one, six adult non-pregnant, non-lactating llamas were examined daily by rectal palpation and transrectal ultrasonography during 70 days. On day 54, intravaginal devices containing 0.33 g of progesterone (CIDR) were inserted and left in the vagina during 16 days. The mean duration of a follicular wave was 22.6+/-2.5 days. The follicular growth phase (follicles growing from 3mm to maximum size) averaged 9.2+/-2.8 days, the mature phase (follicles around maximum size) 5.2+/-1.4 days and regression phase (follicles with decreasing size) 8.2+/-2.2 days. Oestradiol-17beta plasma concentrations exhibited a similar wave pattern (P<0.05). In addition, oestradiol-17beta peak plasma concentrations (46.9+/-3.3 pmoll(-1)) were attained approximately 12 days after the beginning of the growing phase in connection with maximum follicle size (11.8+/-1.6mm). After CIDR insertion, a rapid increase in plasma progesterone concentrations was observed, with peak concentrations attained on day 1 after insertion. Thereafter, concentrations decreased gradually. Mean follicle size steadily decreased from the day of CIDR insertion to day 11 post-insertion (10.3+/-1.6 and 3.3+/-0.8mm, respectively). In order to investigate the effect of follicle size at CIDR insertion on the outcome of progesterone treatment, experiment two was designed. Sixteen adult non-pregnant and non-lactating llamas were divided into four groups according to follicle development at the time of CIDR insertion (group I: follicles < or =6 mm; group II: follicles between 6 and 9 mm; group III: follicles between 10 and 14 mm and group IV, regressing follicles). In groups II, III and IV, a significant decrease in follicle size was observed after the insertion of the CIDR device. In group I, no further development of dominant follicles was observed until the device was withdrawn. In all cases, the smallest diameter was registered between days 5 and 7 after the beginning of treatment. In conclusion, a detailed characterization of follicular waves using ultrasound and hormone determinations simultaneously in non-ovulating llamas and after the insertion of progesterone releasing devices, is presented. 相似文献

We evaluated the agreement between ultrasonographic characteristics of the corpus luteum (CL) and plasma progesterone (P4) concentration in dairy cows. In Phase I of the study, the ovaries of 8 cows were ultrasonographically examined, and P4 was analyzed daily from estrus (Day 0) to Day 4, then at Day 7 and Day 10, and again daily from Day 17 to the onset of next estrus. In Phase 2, the ovaries of 157 randomly selected Friesian cows were examined once by ultrasonography, and blood samples collected concurrently were analyzed for plasma P4. On the basis of the P4 values, the function of CLs was classified as follows: 1) non-secretory CL when plasma P4 was lower than 1 ng/mL (n=41); 2) evolving CL when plasma P4 was between 1 and 4 ng/mL (n=55); and 3) mid-cycle CL when plasma P4 was more than 4 ng/mL (n=61). On the basis of ultrasonographic examination, 3 additional groups were established (absence of CL, evolving CL, midcycle CL). Ultrasonographic characteristics and size of Day 3 to 4 CLs and their respective plasma P4 concentrations were not distinguishable from those of CLs observed 3 to 4 d before the subsequent estrus. The degree of agreement between the two classification was 72%. The data indicate that the functional classification of CLs is difficult to determine based on ultrasonography alone. 相似文献

The diagnostic value of 2 plasma acute-phase proteins, haptoglobin and alpha1-acid glycoprotein, and plasma N-acetyl-beta-D-glucosaminidase enzyme activity were studied in 29 newly calved dairy cows. Nineteen had developed acute metritis with putrid vaginal discharge within 2 wk after calving; 10 were clinically healthy controls. Plasma haptoglobin concentration remained low in most cows with acute postpartum metritis. Only the 3 most severely affected cows exhibited a strong haptoglobin response. These were later culled due to poor condition and reduced fertility. This suggests that in acute uterine infection a highly increased haptoglobin concentration indicates poor prognosis for repeat conception. Plasma alpha1-acid glycoprotein concentration increased in acute postpartum metritis, the response pattern being less prominent than that for haptoglobin. The alpha1-acid glycoprotein concentrations did not correlate with severity of disease, and, consequently, the capacity of alpha1-acid glycoprotein in differentiating genital infections was relatively poor. The highest alpha1-acid glycoprotein concentrations were detected in cows with retained placenta and/or dystocia. Plasma N-acetyl-beta-D-glucosaminidase activity levels did not differ between the cows with acute postpartum metritis and healthy control cows. 相似文献

An experiment was designed to evaluate a) the effect of a progesterone-estradiol combined treatment on ovarian follicular dynamics in postpartum beef cows, and b) ovulation and the subsequent luteal activity after short-term calf removal and GnRH agonist treatment. Multiparous Angus cows (25 to 40 d after calving) were assigned to the following treatments: untreated (Control, n = 9); short term calf removal (CR, n = 8); progesterone (CIDR, n = 9) and progesterone plus estradiol-17 beta (CIDR + E-17 beta, n = 9). Progesterone treatment (CIDR) lasted 8 d and the day of device insertion was considered as Day 0. Cows in the CIDR + E-17 beta group also received an i.m. injection of 5 mg of E-17 beta on Day 1. On Day 8, calves were removed for 48 h (CR, CIDR and CIDR + E-17 beta groups) and 6 h before the end of calf removal these cows also received an i.m. injection of 8 micrograms of Busereline (GnRH). Anestrus was confirmed in all cows by the absence of luteal tissue and progesterone concentrations below 1 ng ml-1 at the beginning of the experiment. Although mean (+/- SEM) interval from the beginning of the experiment (Day 0) to wave emergence did not differ (P > 0.05) among treatment groups (Control, 1.9 +/- 1.0, range -2 to 7 d; CR, 3.9 +/- 0.7, range 0 to 6 d; CIDR, 2.8 +/- 0.5, range 0 to 4 d and CIDR + E-17 beta, 4.1 +/- 0.2, range 3 to 5), the variability was less (P < 0.05) in the CIDR + E-17 beta group. The proportion of cows ovulating 24 to 48 h after GnRH administration tended (P = 0.08) to be higher in cows from CIDR + E-17 beta group (8/9) than in those of CR (5/8) or CIDR (6/9) groups, respectively and was associated with a higher proportion (P < 0.05) of CIDR + E-17 beta treated cows (9/9) that had a dominant follicle in the growing/early static phase at the time of GnRH treatment compared to the other GnRH treated groups (5/8, and 4/9 for CR and CIDR groups, respectively). Two CR cows ovulated 0-24 h after GnRH and only one Control cow ovulated the day before the time of GnRH administration. Cows pretreated with progesterone had longer (P < 0.05) luteal lifespan (CIDR, 14.5 +/- 0.7, CIDR + E-17 beta, 13.9 +/- 0.6 d) than those not treated with CIDR (Control, 5, CR, 4.0 +/- 0.4). We conclude that progesterone plus estradiol treatment results in tightly synchronized wave emergence and high GnRH-induced ovulation rate with normal luteal activity in postpartum beef cattle. 相似文献

Animal performances were monitored in 30 Friesean dry cows (18 multiparous, MP-cows and 12 primiparous, PP-cows) starting six weeks before calving to eight weeks after calving. The cows were kept indoors and fed individually with a prepartum diet containing either low, moderate or high energy (0.75, 1.00 or 1.25 of the calculated ME requirement) and supplemented with low (0.3 kg day−1) or high (1.5 kg day−1) rapeseed meal (RSM). The diets were consisted of 1.5 kg hay, 20–25 kg wilted grass silage and grain (barley and oat, 1 : 1 DM basis) with RSM. The average ME intake during six weeks of prepartum was 75, 97 and 123 MJ day−1 on three different energy levels. After calving the cows were fed grass silage ad lib, 6.5 kg (primiparous) or 8.5 kg (multiparous) grain and 1.5 kg RSM. Liver biopsy and blood samples were taken to determine differences in metabolites due to diet and parity. Reduction of prepartum energy allowance to 0.75 of the moderate level did not affect the calf's birth weight and colostrum composition. Cows fed prepartum low energy–high rapeseed meal diet (LEHR) showed a faster increase in feed intake and milk production after parturition. Continuous low feed intake and milk production were observed in cows fed a prepartum high energy–low rapeseed meal diet (HELR). Milk yield was constantly lower for cows fed prepartum high energy diet, the difference being significant (p<0.01 and p<0.05) at six and seven weeks of lactation. Overall milk yield also showed a linear decrease with energy feeding level and an increase (p<0.10) with RSM feeding levels. Prepartum high energy feeding increased (p<0.01) milk protein content. Neither energy nor RSM feeding level affected the liver fat infiltration of the experimental cows. Cow parity however showed a significant influence on liver fat content, β-hydroxybutyrate (BHBA) and plasma glucose. A week before calving, the plasma concentration of essential amino acids (EAA) was lower for cows fed prepartum low energy diet. During the first four weeks of lactation, the concentrations of plasma non-esterified fatty acids (NEFA) and insulin were similar for all the treatment group. 相似文献

Prolonged postpartum acyclicity in suckled beef cows reduces the calf crop, and causes economic loss to beef cattle producers. Once anterior pituitary LH stores have been replenished between Days 15 and 30 post partum in suckled beef cows, methods to initiate cyclicity include non-hormonal methods such as weaning of calves (either complete, temporary or partial), or exposure to bulls, and hormonal methods such as administration of GnRH (either single injection, intermittent injections, or continuous infusion), gonadotropins (eCG, FSH, hCG), and steroids (estrogens, anti-estrogens, and progestogens). Weaning is costly, reduces growth rate of weaned calves, and short cycles are common after weaning-induced ovulation. Exposure of cows to bulls is not practical and its effect is not predictable. Repeated injections of GnRH, or a single injection of hCG are not always effective; ovulation is always followed by a short cycle, and usually a return to acyclicity. Estrogens and anti-estrogens do not consistently shorten postpartum anestrus. Exogenous progestogens include intravaginal devices, such as controlled-internal drug release (CIDR) or progesterone-releasing intravaginal device (PRID), norgestomet implants, and the feed-additive melengestrol acetate (MGA). Administration of exogenous progestogens is more practical than, and offers more advantages over, other treatments to shorten postpartum acyclicity in suckled beef cows. Mimicking the short cycle after Week 3 post partum, by maintaining circulating progesterone at subluteal concentrations or circulating progestin at intermediate concentrations, extends the life-span and allows terminal maturation of the postpartum dominant follicle as in cyclic cows, by initiating endogenous GnRH and LH pulses. This is followed by an LH surge, ovulation and normal cycles. The benefit from using exogenous progestogens after Week 3 post partum in suckled beef cows is that ovulation is induced, cyclicity is initiated, the resulting CL has a normal life-span and function, and there is no need to change management, such as weaning of calves. This study was carried out to determine the relationship between postpartum intrauterine infections, endocrine patterns and the function of corpora lutea formed following the first postpartum ovulations in dairy cows. Blood samples were collected daily starting from the day of parturition until 30 d after parturition or until the second postpartum estrus, whichever occurred first. Sera were assayed for progesterone (P(4)), prostaglandin F(2alpha) metabolite (PGFM), and luteinizing hormone (LH) concentrations. Palpations per rectum and real-time ultrasound scanning of the reproductive tracts were carried out in all cows once every 4 d for 1 mo, starting from Day 4 after parturition. In addition, endometrial swabs were collected aseptically from each cow once every 4 d during the first month postpartum. The swabs were cultured for aerobic and anaerobic bacteria. Twelve cows (60%) exhibited short estrous cycles (SC; 6 to 14 d long) following first postpartum ovulations. The mean preovulatory LH surges and LH patterns during the first postpartum cycles were similar in both groups, leading us to believe that lack of luteotrophic stimulation was not a factor in the occurrence of SC. Bacterial isolations were frequent in SC cows. The occurrence of moderate to heavy bacterial growth patterns and the repeated isolations of the similar organisms during postpartum suggests the persistence of uterine infections in SC cows. Increases in PGFM concentrations prior to luteolysis in SC cows were associated with moderate to heavy infection. Thus, postpartum uterine infections do not appear to affect ovulations, but prostaglandin (PGF(2alpha)) released in response to uterine infection may contribute to early demise of the corpus luteum formed after the first postpartum ovulation. 相似文献

The objective of this study was to investigate whether monitoring progesterone concentrations in milk and blood plasma can be used to predict time of ovulation in dairy cattle. Whole milk was sampled twice daily and blood samples were collected once a day before the morning milking. Ovulation was assessed by trans-rectal ultrasonography at 4h intervals beginning from the end of estrus. For a parameter to be useful as predictor for time of ovulation, it should be precise (i.e. variation between animals should not exceed 12h). In milk, progesterone concentration dropped <15 ng/ml at 97.7+/-17.8h (range: 54-126 h) before ovulation, to <5 ng/ml at 79.7+/-11.2h (range: 54-98) before ovulation to decline further to <2n g/ml at 70.7+/-16.8h (range: 38-90 h) before ovulation (n=20). In plasma, progesterone concentration dropped to <4ng/ml 90.5+/-19.6h (range: 66-138 h) before ovulation and to <2 ng/ml at 75.0+/-12.2 h (range: 50-98) before ovulation. These intervals were not influenced by parity, milk production or days in milk. In conclusion, monitoring of progesterone alone is not sufficient to predict ovulation because of the large variation in timing of decrease of progesterone concentrations relative to ovulation between animals. The objectives of this study were to determine 1) the incidence of abnormal postpartum ovarian function in a large dairy herd in North Central Florida and 2) the effectiveness of gonadotrophin releasing hormone (GnRH) in treating this condition. The study was conducted from April 1988 to June 1989. The internal genitalia of the cows were initially examined per rectum (Day 0) between 19 and 29 (23 +/- 0.25) d after calving and again 14 d later (Day 14) for evidence of uterine involution and ovarian activity. The presence of a palpable corpus luteum (CL) and retrospective determination of plasma progesterone (P4) concentrations > 1 ng/ml were the criteria used to assess ovarian activity. Cows possessing a palpable CL and P4 concentrations > 1 ng/ml on Day 0 were determined to be cycling normally. A total of 1356 cows was used in this study. On Day 0, two groups were formed: Group 1 consisted of normal, cyclic cows, Group 2 of noncyclic cows. On Day 0, alternate cows in Group 2 were treated with GnRH (100ug i.m). On Day 14, the previously nontreated cows in Group 2 were further divided into two groups, forming Group 3, nontreated cows and Group 4, cows treated with GnRH at this time. Group 5 was comprised of cows from Group 2 that did not respond to treatment with GnRH on Day 0; these cows were treated on Day 14 with GnRH (100ug i.m). Group 6 was comprised of nontreated cows from Group 2 that responded spontaneously (presence of a CL) by Day 14. Reproductive parameters evaluated were the percentage of cows pregnant within 180 d after calving and at the end of the study, the number of days open and the number of services per conception. Data were statistically analyzed using Chisquare and survival analysis. The results of this study indicate that the incidence of abnormal postpartum ovarian function in this herd was 30.2% and that the nontreated cows experienced more days open and required more services per conception than the treated cows, those that were cycling normally on the initial examination, and those that responded spontaneously by Day 14. 相似文献
