Development of in vitro culture protocol for early stage ovarian follicles of zebrafish is important since cryopreserved early stage ovarian follicles would need to be matured in vitro following cryopreservation before they can be fertilised. Development of molecular markers for zebrafish (Danio rerio) ovarian follicle growth assessment following in vitro culture of early stage zebrafish ovarian follicles in ovarian tissue fragments is reported here for the first time although some work has been reported for in vitro culture of isolated early stage zebrafish ovarian follicles. The main aim of the present study was to develop molecular markers in an optimised in vitro culture protocol for stage I and stage II zebrafish ovarian follicles in ovarian tissue fragments. The effect of concentration of the hormones human chorionic gonadotropin and follicle stimulating hormones, and additives such as Foetal Bovine Serum and Bovine Serum Albumin were studied. The results showed that early stage zebrafish ovarian fragments containing stage I and stage II follicles which are cultured in vitro for 24 h in 20% FBS and 100mIU/ml FSH in 90% L-15 medium at 28 °C can grow to the size of stage II and stage III ovarian follicles respectively. More importantly the follicle growth from stage I to stage II and from stage II to stage III were confirmed using molecular markers such as cyp19a1a (also known as P450aromA) and vtg1 genes respectively. However, no follicle growth was observed following cryopreservation and in vitro culture.  相似文献

There have been no reported methods for in vitro growth of early stage ovarian follicles for fish and their cryopreservation is still under investigation. If cryopreservation of early stage ovarian follicles can be achieved, in vitro procedures for ovarian follicle culture, development, ovulation and fertilisation after cryopreservation would be needed. The aim of the present study was to develop an in vitro culture method for early stage zebrafish ovarian follicles for use after their cryopreservation. Procedures for in vitro culture of stage I (primary growth) and stage II (cortical alveolus) ovarian follicles were developed. The effects of concentration of L-15 medium, pH and the concentration of human chorionic gonadotropin (hCG) and activin A were studied. The results demonstrated that early stage zebrafish ovarian follicles can be cultured in vitro for 24 h, stage I and II ovarian follicles can grow to the sizes of early stage II and stage III ovarian follicles after hCG treatment. The method developed here is effective for assessing early stage zebrafish ovarian follicles growth competence in vitro. The results from the present study indicated that in vitro culture is the most reliable method for assessing ovarian follicle viability when compared with vital dye staining methods.  相似文献

The uptake of l-methionine-methyl-3H and l-leucine-3H from completely defined medium into acid-soluble fractions of preimplantation mouse embryos has been studied. Late four-cell embryos and early blastocysts raised in vitro can concentrate both amino acids by processes which exhibit saturable, Michaelis-Menten type kinetics, characteristic of carrier-mediated active transport systems. This uptake is temperature-sensitive and inhibited by certain amino acids which compete for the same uptake sites. Methionine uptake seems to be mediated by a single transport system (Km = 6.25 × 10?5M) at the four-cell stage. Complex kinetics suggest that two distinct transport systems exist at the early blastocyst stage (Km = 6.25 × 10?5M; 8.9 × 10?4M). Vmax values (mg/embryo/15 min) for methionine and leucine transport increase significantly from the late four-cell stage to the blastocyst stage, suggesting that additional carriers are produced or activated during development.Most importantly, leucine and methionine transport is Na+-independent at the four-cell stage, methionine transport is partially dependent at the morula stage, and both amino acids are completely Na+-dependent at the blastocyst stage. The cumulative results suggest that preimplantation embryos accumulate leucine and methionine by specific, chemically mediated, active transport systems. The qualitative and quantitative developmental changes in cell membrane function may represent preparatory steps for subsequent growth of embryonic and/or trophoblastic cells.  相似文献

Haploid Saccharomyces cerevisiae cells of mating type a, but not α, produce and secrete a diffusible substance, designated a factor. The a factor transiently arrests cells of mating type α, but not a, at a very early stage of the cell cycle, prior to budding and to the initiation of DNA synthesis. While the cells are arrested at this stage, few, if any, of the functions required for the ensuing cell cycle are carried out. This stage of the cell cycle coincides with the stage at which α factor, produced by cells of mating type a, specifically arrests cells of mating type a [2]. It seems probable that the reciprocally acting a and α factors together provide the mechanism by which haploid cells are synchronized to the appropriate stage of the cell cycle as a prelude to conjugation.  相似文献

Fos-like antigens (Fosl) including Fosl1 and Fosl2 exclusively heterodimerize with Jun members to form AP-1 complex, thereby participating in various cellular progresses including cell cycle regulation. However, expression patterns of these two genes during embryonic development remains largely unknown. In the present study, both temporal and spatial expression patterns of fosl1 and fosl2 were examined during embryonic development of Xenopus tropicalis. Real-time quantitative PCR results showed that the expression of the two genes was increased from stage 2 to stage 42. However, expression level of fosl1 is much higher than that of fosl2 at stage 42. Whole-mount in situ hybridization showed that fosl1 was expressed in eyes, branchial arch, notochord, otic vesicle, and liver. However, fosl2 was expressed in lung primordium from stage 34 to stage 38, in addition to the moderate expression in eyes and branchial arch at stage 42. Thus, the developmental expression patterns of these two fosl genes is different in Xenopus embryos. These results provide a basis for further functional study of these two genes.  相似文献

The distributions of Haemonchus contortus, Ostertagia circumcincta and Trichostrongylus axei during their development in lambs were studied by freezing and sectioning the abomasum and then recovering the worms from the contents and mucosa of each section. In previously worm-free lambs, the distribution of fourth stage H. contortus was very regular, the frequency of larvae being highest in the anterior fundic area and then decreasing in each section towards the pylorus. The distributions of the early larval stages of O. circumcincta and T. axei were variable but tended to be biomodal, with one peak of larvae in the fundic area and a second in the pyloric area. It is suggested that the difference in distribution between H. contortus and the other two species may be due to a difference in the rate of response of exsheathed third stage larvae to stimuli in the abomasum. In the later stages of development, H. contortus and O. circumcincta tended to migrate towards the pyloric area. In previously infected lambs, the distribution of inhibited early-fourth stage H. contortus was similar to that of normally developing fourth stage larvae in previously worm-free lambs and appeared to be unaffected by the host response which caused an altered distribution and rejection of late-fourth stage and adult worms. The rate of rejection of adult O. circumcincta differed between the fundic and pyloric areas of the abomasum, which suggests that the host response in these two areas may be different.  相似文献

This paper contains observations and experiments which collectively demonstrate a requirement for cell-cell interactions among limb bud mesenchyme cells during chondrogenic differentiation. Limb bud cells isolated from brachypodismH (bpH) and wild-type mouse embyros between Thieler stage 16–17 and midstage 21 were compared with respect to their abilities to undergo chondrogenic differentiation in high-density micromass cultures. Nodules formed by dissociated Day 12 (stage 20) bpH limb bud cells have been reported previously to be abnormally reduced in size and number, and delayed in formation. We corroborate these results, but find that bpH cultures prepared from earlier-stage limb buds (between stages 16–17 and early stage 21) are progressively more like wild-type cultures. Stage 16–17 bpH cultures at 72 hr actually contain normal numbers of and size nodules, while stage 18 bpH cultures are intermediate between stages 16–17 and stage 21 in nodule formation. On the other hand, we also find that the initial rate of aggregate formation is normal even in bpH cultures prepared from stage 20 cultures in which nodule formation is not normal. Preparation of cultures composed primarily of early stage 21 bpH limb bud cells mixed with small quantities (e.g., 5%) of stage 16–17 wild-type limb bud cells showed significant increases in cartilage nodule formation over control cultures composed only of early stage 21 bpH cells. Greater proportions of wild-type cells obtained from embryos older than stages 16–17 were required for the same degree of normalization, supporting the hypothesis that a specific cell type, whose proportion decreases normally in the limb bud over time, is required to increase in vitro chondrogenesis by bpH cells. Additionally, cultures containing stage 23 chick limb cells and early stage 21 bpH cells at a ratio of 1:20 contained wild-type levels of nodules per square millimeter of culture. Thus, bpH cells appear to respond to chondrogenic inductive signals from normal limb mesenchyme cells. In order to test for the ability of bpH limb bud mesenchyme to induce chondrogenesis, stage 16–17 bpH and wild-type limb bud cells, which form identical numbers of aggregates and nodules in culture, were each mixed with early stage 21 bpH cells at ratios of 1:20, 1:10, and 1:3. Although low proportions of wild-type stage 17 cells significantly increased the number of aggregates and nodules in these mixed cultures, low proportions of bpH stage 16–17 cells did not. It is, therefore, concluded that the primary defect of the bpH mutation is likely to reside in the reduced ability of a specific mesenchyme cell subpopulation to provide an inductive stimulus for chondrogenesis.  相似文献

Prior to the baceman stage or brine fermentation in Indonesian kecap (soy sauce) production, the soybeans undergo a number of treatments, such as soaking, cooking and a fungal solid state fermentation (bungkil stage). The influence of these prebrining steps on the baceman stage was investigated. Acidification during soaking inhibited bacterial growth during the solid state fermentation, causing lower initial numbers of bacteria in and a slower acidification of the baceman. Cooking time influenced the softness of the beans and thus fungal growth during the bungkil stage. Increasing the cooking time from 1 to 3 h resulted in a better solubility of macromolecules and gave higher final concentrations of fermentation products and formol nitrogen in baceman. When cooking time was increased from 3 to 5 h only slight differences could be observed. Length of the bungkil stage had some minor influences; the amount of fermentation products and residual monosaccharides decreased by 16% when the length was increased from 2 to 7 d. The highest amounts of formol nitrogen were found with 5 d of bungkil fermentation. The addition of rice flour showed that the absence of yeast fermentation in baceman is related to the lack of sugars after the previous lactic acid fermentation. In addition to yeast fermentation, the growth of fungi during the bungkil stage was also positively influenced by the addition of rice flour. Sun drying and crumbling bungkil caused lower enzyme activities and resulted in lower amounts of formol nitrogen and glycerol. Growth of Pediococcus halophilus during the baceman stage was faster when bungkil was sun dried, although the final concentrations of fermentation products were lower due to a lower content of amino acids which resulted in less buffering. Increasing the amount of bungkil added to salty water caused a similar increase in formol nitrogen and fermentation products.  相似文献

We say that a cell of budding yeast, Saccharomyces cerevisiae, is of stage n when it has n bud scars and hence it has produced n daughters. We then divide the total population X into stage classes according to X = ΣnXn where Xn is the number of cells which have produced n daughters. The present work deals with the fractions XnX in the transient phase before the demographic equilibrium, where the fractions attain stationary values, is reached. It is found that the oscillatory (in time) stage structure is possible provided that (i) the generation time of a virgin (n = 0) cell differs from those of matured (n ≥ 1) cells by more than the sum of respective standard deviations, and (ii) both the initial stage structure and the initial distribution over the cell cycle phase differ appreciably from those prevailing in the demographic equilibrium.  相似文献

Penaeoidean shrimp pleonal muscle is a valuable economic resource worldwide, but little is known of its development during larval stages. The development of pleonal muscle in Penaeus (Litopenaeus) vannamei was studied by rhodamine-phalloidin staining and laser-scanning confocal microscopy. Dorsal pleonal muscle was first evident at the protozoea I stage while ventral pleonal muscle was present by the protozoea II stage. Identifiable ventral pleonal muscles were evident by the protozoea III stage and all ventral muscle types were present in the mysis I. The tail flex response began at the mysis stage and growth of existing pleonal muscles continued. The pleopods formed during the mysis stages, with coxal and basis muscles developed by mysis III. The pleopods became functional beginning with the first post-larval stage. We conclude that the pleonal muscle pattern of P. vannamei larvae is similar to that of adult Penaeus setiferus, and that homologous muscles are present. The major formation of dorsal pleonal muscles occurs during the protozoea II stage, while significant development of ventral pleonal muscles occurs during the protozoea III stage.  相似文献

Abies sachalinensis and Picea glehnii are co-dominant tree species and major components of the forests of Hokkaido, Japan. Recent work suggests that a reversal in potential competitive superiority at different developmental stages could be important to explain their coexistence. Such shifts in competitive advantage can be mechanistically understood by studying the corresponding physiological differences between distinct life stages. Accordingly, our objective was to investigate in the field the photosynthesis of shade-growing juveniles of these species from two different size-classes, seedlings and saplings. Our results show that seedlings of both species had higher concentrations of photoprotective xanthophylls than saplings, especially in spring, and suggest that seedlings have a lower threshold of stress tolerance than saplings. Photosynthetic capacity per needle area and lateral shoot extension rate decreased from the seedling to the sapling stage in A. sachalinensis, while in P. glehnii, both increased from the seedling to the sapling stage. Abies sachalinensis had higher photosynthetic rates at the seedling stage but lower rates at the sapling stage than P. glehnii. Nevertheless, A. sachalinensis had a higher lateral shoot extension rate than P. glehnii at both stages. Our physiological results support previous ecological observations that A. sachalinensis is a superior competitor to P. glehnii in the understory, and show that its competitive advantage is higher at the seedling stage than at the sapling stage.  相似文献

The Philodendron bipinnatifidum complex of Philodendron subgenus Meconostigma may comprise four species, which because of only slight and not very distinct morphological differences are not all unanimously recognized as good species. To find out whether these species are reproductively isolated, we studied the flowering rhythm, thermogenesis and pollination biology of three species of this complex, namely of P. bipinnatifidum, P. aff. bipinnatifidum (provisionally named “P. form selloum”) and P. mello-barretoanum in Brazil. Of the first two mentioned taxa, floral scent was collected and scent compounds were identified by GC–MS. The results showed that the coastal forest species P. bipinnatifidum has a two-, or three-night flowering rhythm, with the pistillate stage in the first night and the staminate stage lasting the second and sometimes also the third night. Strong thermogenesis with extended heating periods of several hours during the first part of the usual two subsequent nights and the maximum temperatures of up to 40 °C absolute heating of the spadices occurred in the pistillate and staminate stages. Concomitant with the heating periods, relatively low amounts of principally (Z)-2-pentenyl acetate and (Z)-jasmone were emitted by both the pistillate and staminate stage inflorescences. The dynastid scarab beetle Cyclocephala variolosa was the only pollinator attracted. The upland forest P. form selloum always had a two-night flowering rhythm with the pistillate stage in the first and the staminate stage in the subsequent night. This world-record holder of thermogenesis can heat up to the remarkable 45 °C during a relatively short period in the evening of the pistillate stage. During the thermogenic period, enormous amounts of principally 4-methoxystyrene and 3,4-dimethoxystyrene were produced and which could attract a large number of female and male individuals of the dynastid scarab beetle Erioscelis emarginata. In the staminate stage of P. form selloum, temperature elevation is significantly lower and the scent compounds are different from the pistillate stage. The cerrado biome species P. mello-barretoanum has a flowering rhythm similar to P. form selloum, reaching a maximum heating of about 40 °C during the pistillate stage. The sole pollinator attracted was Cyclocephala atricapilla. The differences observed and analyzed among the taxa, including the flowering rhythm, thermogenic activities, scent compounds emitted, pollinating dynastid scarab beetles attracted, as well as slight morphological differences and apparent geographical exclusiveness noted in these three taxa are strong indicators that P. bipinnatifidum, P. form selloum and P. mello-barretoanum are different enough to be considered good species. The morphological affinities of these species might be a hint that speciation has been a recent event and/or also that reproductive isolation based on different, non-overlapping distribution areas, different scent compounds and different pollinators was effective enough to need further morphological differentiation.  相似文献

花绒寄甲(Dastarcus helophoroides)(鞘翅目:寄甲科Bothrideridae)是寄生栗山天牛(Massicus raddei)中老龄幼虫和蛹的重要天敌,但其寄主栗山天牛世代周期长(3年1代)、发育比较整齐,不利于寄生性天敌种群数量的稳定.为了解利用花绒寄甲防治栗山天牛后,其种群能否在栎树林间长期保持较高的种群数量,达到持续控制栗山天牛的防治效果,调查研究了花绒寄甲在栎树林间的转主寄主和种群保持机制.结果表明,在东北辽东栎树干和树枝上除了栗山天牛外,还有其他8种天牛危害:双簇天牛(Moechotypa diphysis)、四点象天牛(Mesosa myops)、中华薄翅锯天牛(Megopis sinica)、锯天牛(Prionus insularis)、双带粒翅天牛(Lamiomimus gottschei)、八字绿虎天牛(Chlorophorus tohokensis)、日本绿虎天牛(C.japonicus)和拟蜡天牛(Stenygrinumquadrinotatum).其中以栗山天牛、双簇天牛、四点象天牛和拟蜡天牛数量较多,而花绒寄甲在辽东栎树干上的垂直分布与栗山天牛、双簇天牛和四点象天牛的垂直分布重叠较多.室内研究表明,花绒寄甲对四点象天牛老熟幼虫的寄生率达到26.67%,对蛹的寄生率达到了43.33%;对双簇天牛老熟幼虫的寄生率达到20.00%,对蛹的寄生率为6.67%.对双簇天牛和四点象天牛在林间的生活史调查和研究发现,花绒寄甲可寄生的这两种天牛的中老龄幼虫和蛹,在花绒寄甲不适宜寄生的栗山天牛幼龄幼虫期大量存在,表明双簇天牛和四点象天牛是花绒寄甲在栎树林中的主要转主寄主.由于这些转主寄主的存在,花绒寄甲在不利于其寄生的栗山天牛卵期、幼龄幼虫期可转移寄生这些寄主,从而在栗山天牛危害的栎树林间保持了较高的种群数量,达到对栗山天牛长期而有效的持续控制效果.  相似文献
