The survival of Kluyveromyces thermotolerans and Torulaspora delbrueckii in mixed cultures with Saccharomyces cerevisiae was examined at low oxygen availability in a defined grape juice medium. In these fermentations, K. thermotolerans and T. delbrueckii died off earlier than S. cerevisiae, and K. thermotolerans and T. delbrueckii exhibited parabolic death kinetics. Furthermore, the early deaths seemed to be non-apoptotic in nature. In order to understand the mechanism causing the early deaths, various single- and mixed-culture fermentations were carried out. The early deaths could not be explained by nutrient depletion or the presence of toxic compounds. Rather, they seemed to be mediated by a cell-to-cell contact mechanism at high cell densities of S. cerevisiae, and to a lesser ability of K. thermotolerans and T. delbrueckii to compete for space, as compared to S. cerevisiae. These results contribute to an increased understanding of why K. thermotolerans and T. delbrueckii die off before S. cerevisiae in wine fermentations.  相似文献

The yeast Saccharomyces cerevisiae synthesises a variety of volatile aroma compounds during wine fermentation. In this study, the influence of fermentation temperature on (1) the production of yeast-derived aroma compounds and (2) the expression of genes involved in aroma compounds’ metabolism (ADH1, PDC1, BAT1, BAT2, LEU2, ILV2, ATF1, ATF2, EHT1 and IAH1) was assessed, during the fermentation of a defined must at 15 and 28°C. Higher concentrations of compounds related to fresh and fruity aromas were found at 15°C, while higher concentrations of flowery related aroma compounds were found at 28°C. The formation rates of volatile aroma compounds varied according to growth stage. In addition, linear correlations between the increases in concentration of higher alcohol and their corresponding acetates were obtained. Genes presented different expression profiles at both temperatures, except ILV2, and those involved in common pathways were co-expressed (ADH1, PDC1 and BAT2; and ATF1, EHT1 and IAH1). These results demonstrate that the fermentation temperature plays an important role in the wine final aroma profile, and is therefore an important control parameter to fine-tune wine quality during winemaking.  相似文献

Wine proteins play an important role in the quality of wine, because they affect taste, clarity and stability of product. The majority of wine proteins are in the range of 20–30 kDa. Different mass spectrometry (MS) techniques have been successfully applied to study the grape and wine proteins. By liquid chromatography (LC) electrospray ionization (ESI) MS and nano-LC/MS, nine dipeptides and 80 peptides were unambiguously identified in Champagne and Sauvignon Blanc wines, respectively. Using matrix-assisted laser desorption/ionization (MALDI) time-of-flight (TOF) and surface-enhanced laser desorption/ionization TOF, the protein and peptide fingerprints in Chardonnay, Sauvignon Blanc and Muscat of Alexandria wines were determined. MALDI-TOF identified the mesocarp proteome of six Vitis grape varieties. Proteins in different grape tissue extracts were also studied. The major grape pathogenic-related proteins are chitinases and thaumatin-like proteins, which both persist through the vinification process and cause hazes and sediments in bottled wines. ESI-MS, LC/ESI-MS and MALDI-TOF analysis of these proteins in grape and wine were also used to characterize different grape varieties.  相似文献

The roles of hybridization and of introgression as potential threats to small native populations are current topics of interest both for evolutionary and conservation biologists. Two factors are mainly involved in the threat to natural populations through hybridization: demographic swamping and genetic assimilation. At prealpine locations, twoSaxifraga species with very different growth habits,S. aizoides andS. mutata, form a hybrid,S. ×hausmannii. Investigation of meiosis and crossing experiments revealed regular pairing of the 26 chromosomes and development of viable seeds in the hybrid. Analysis of hybrid indices, GPI allozymes, germination experiments and seed sizes showed an intermediate position ofS. ×hausmannii between its parental species.Saxifraga aizoides andS. mutata were separated by only weak hybridization barriers. Introgression occurred mostly in direction ofS. aizoides, although to a small amount. There is no actual threat to small, prealpine populations ofS. aizoides andS. mutata through interspecific gene flow because hybridization is localized, resulting from different flowering phenologies of the parental species and from the narrow habitat requirements ofS. ×hausmannii, which are similar to those ofS. aizoides. The major threat to local, prealpine populations ofS. aizoides andS. mutata is the loss of landscape dynamics. Both species need open erosion slopes for successful recruitment.  相似文献

The relationships between population densities ofCalepitrimerus vitis (Nalepa),Colomerus vitis (Pagenstecher) andTetranychus urticae (Koch), and grape weight and sugar content were studied during 3-year-long trials. In the case ofCalepitrimerus vitis, the influence of population density on leaf area, length of spring shoots and number of grapes in the following year was studied. Significant relationships between population densities of bothCalepitrimerus vitis andT. urticae in late summer and grape weight were determined. The sugar content of the grapes was markedly less influenced by mite infestation. The negative effect ofColomerus vitis on grape weight was not confirmed. Significant relationships for crop-protection decisions are presented.  相似文献

One hundred and fifteen Saccharomyces cerevisiae strains from Aglianico del Vulture, a red wine produced in Southern Italy, were characterized for the production of some secondary compounds involved in the aroma and taste of alcoholic beverages. The strains exhibited a uniform behaviour in the production levels of n-propanol, active amyl alcohol and ethyl acetate, whereas isobutanol, isoamyl alcohol and acetaldehyde were formed with a wide variability. Only five strains produced wines close to the reference Aglianico del Vulture wine for the traits considered. Of these, two strains were selected, underwent to tetrad analysis and the single spore cultures were tested in grape must fermentation. The progeny of one strain showed a significant metabolic variability, confirming the necessity to test starter cultures for the segregation of traits of technological interest. Our findings suggest the selection of specific strains for specific fermentations as a function of the vine variety characteristics in order to take the major advantage from the combination grape must/S. cerevisiae strain.  相似文献

Aims: The objective of this study was to investigate the presence of genes coding for enzymes of oenological relevance in wine Lactobacillus strains isolated from South African grape and wine samples during the 2001 and 2002 harvest seasons. Methods and Results: A total of 120 wine lactobacilli isolates belonging to Lactobacillus plantarum, Lactobacillus hilgardii, Lactobacillus brevis, Lactobacillus pentosus, Lactobacillus paracasei, Lactobacillus sakei and Lactobacillus paraplantarum were genetically screened for enzyme‐encoding genes using PCR with primers specific for β‐glucosidase, protease, esterase, citrate lyase and phenolic acid decarboxylase. The results of PCR screening showed that the Lactobacillus strains possessed different combinations of enzymes and that some strains did not possess any of the enzymes tested. Confirmation analysis with gene sequencing also showed high similarity of genes with those available in GenBank database. Conclusion: In this study, we have demonstrated the existence of genes coding for wine‐related enzymes in wine lactobacilli that could potentially hydrolyse wine precursors to positively influence wine aroma. Significance and Impact of the Study: An expansion of knowledge on the genetic diversity of wine‐associated lactic acid bacteria will enable the selection of novel malolactic fermentation starter cultures with desired oenological traits for the improvement of the organoleptic quality of the wine, and hence wine aroma.  相似文献

Nucleotide Binding Site/Leucine-Rich Repeat (NBS-LRR) and Serine/Threonine Kinase (STK) genes are two of the known classes of resistance (R-) genes in plants, and occur in large multigene families. Systematic identification of genes for NBS-LRRs and STKs provides a means of access to genomic regions that may be involved in disease resistance. Here we present a picture of these two families of R-gene analogs (RGAs) in grape with the aim of developing a set of resistance-related sequence-tagged-site (STS) markers. One hundred and three NBS-LRR sequences were isolated. They included members of the CC (coiled-coil) and TIR (Toll-interleukin receptor) sub-classes. A comparative analysis with other angiosperm NBSs is provided. Fifty-three genes for receptor-like kinases (RLKs) with serine/threonine specificity were identified. RLK sequences formed a putative monophyletic group within the kinase superfamily. They were similar to both cytoplasmic RLKs, such as Pto, and RLKs with LRR, S-locus, lectin-like and thaumatin-like extracellular binding-domains. The latter resembled the products of the R-related genes Xa21, FLS2, Rlk10, SFR2, and PR5K. Forty-five reference RGAs were converted into STSs by using appropriately designed specific primers. RGA-STSs were present in diverse grape genotypes, and >85% of the primers were capable of amplifying the STSs across the taxa Vitis and Muscadinia. DNA sequence polymorphism among these RGAs was assessed by SSCP (single-strand conformation polymorphism) analysis in over 20 Vitis spp. Finally, 45 universal primers for grape RGAs are proposed that should permit tagging of R-related regions in any grape genome.Communicated by R. Grape pistil has an important influence on fruit size and quality. However, there were few studies on grape ovary, and the development process of the ovary is still unclear. Therefore, in this paper, four different grape varieties with different lengths of small inflorescences, namely ‘Musct Hambourg’ grape (Vitis vinifera), ‘Concord’ grape (Vitis labrusca), ‘ShanPuTao’ grape (Vitis amurensis) and ‘GongNiang2Hao’ grape (Vitis amurensis × Vitis vinifera) were used as test materials. Four varieties ovary were significant differences by means of stereomicroscope, paraffin section. The expression of ovary determining gene VvAGAMOUS (VvAG) and its development related genes VvCRABS CLAW (VvCRC) andVvAGAMOUS-LIKE 11 (VvAGL11) with similar functions during the development of different grape varieties were preliminarily explored using fluorescence quantitative test. The relationship between VvAG and VvCRC, VvAG and VvAGL11 were analyzed using Y1H assay. Our results showed that there were obvious abdominal sutures on the surface of expect for ‘Musct Hambourg’ grape, and existing poly carpels. The ovary development of ‘ShanPuTao’ and ‘GongNiang2Hao’ grape was completed when the inflorescence length was less than 1 cm, while the ‘Concord’ and ‘Musct Hambourg’ grape were fully developed when the length of inflorescence was 3–4 and 4–5 cm, respectively. VvAG and VvCRC began to express in large quantities after the formation of stamen primordia, while VvAGL11 during the forming of ovule primordia. Therefore, VvAG and VvCRC mainly regulated the development of stamens and carpels and also promote the development of ovules, while VvAGL11 major regulated the development of ovules. The promoters of VvCRC and VvAGL11 were bound by VvAG. This study provides an important theoretical basis for further research on the molecular mechanism of grape ovary development.  相似文献

Synopsis The longitudinal distribution ofSalmo trutta andGalaxias olidus, a small salmoniform fish, was mapped over four summers (1985–1988) in the upper reaches of the Lerderderg River, an intermittent stream in central Victoria. Over the four successive summers of the study, the distribution ofS. trutta expanded upstream. Coincident with the expansion ofS. trutta was a contraction in the distribution ofG. olidus upstream. In the summer of 1988, an extended dry spell resulted in high levels ofS. trutta mortality at the upstream limits of their distribution. This suggested that the upstream distribution ofS. trutta could ultimately be constrained by an inability to tolerate prolonged periods of low stream flow and high temperatures that tended to occur in the headwaters of the stream. A study of summer mortality was conducted over three successive summers (1989–1991) at three reaches along the river. The two downstream reaches contained onlyS. trutta, the upstream reach contained bothS. trutta andG. olidus. Salmo trutta mortality was consistently higher at the most upstream reach compared with lower reaches. High levels ofS. trutta mortality at the two downstream reaches were only observed in 1991, a particularly dry year. In contrast,G. olidus survived in scattered small pools throughout the upper reaches of the stream. The results of the study suggest a shifting pattern ofS. trutta andG. olidus distribution determined by the duration of low stream flow periods and summer high temperatures.  相似文献

Summary Cuttings of a carnation variety of the Chabaud type were rooted in a fine textured soil which had been broken down to yield 3 different soil structures,C, M andF (coarse, medium-sized and fine aggregates).Root initials were found in the basal callus of cuttings growing in all soil treatments already two weeks after planting.Throughout the rooting process, the best results were obtained with coarse aggregated soil and the worst with the medium aggregated one. The fine aggregated soil showed intermediate results, probably due to fissures formed at the point of insertion of cuttings, which improved aeration.The amount of roots at the end of the experiment was greatest with cuttings from theC treatment, while those ofM andF treatments did not differ significantly in this respect.  相似文献

Some aspects of the biologies ofTyphlodromus annectens De Leon andT. porresi McMurtry were studied at 25°C. The tetranychid mitesOligonychus punicae (Hirst),Panonychus citri (McGregor) andTetranychus pacificus McGregor were all favorable foods for development ofTy. annectens, whereas a low survival rate of immatures and no oviposition occurred on pollen.Tetranychus pacificus appeared to be a less favorable prey than the other two tetranychids forTy. porresi. Typhlodromus annectens, a very small species, andTy. occidentalis, a medium-sized species used for comparison, consumed significantly moreTe. pacificus larvae than did the medium-sizedTy. porreso. Comparing weights of fed and starved individuals suggested thatTy. occidentalis consumed the greatest volume of food material per time unit. Catinaria grossa is lichenized withDictyochloropsis splendida var.gelatinosa, var. nova. When cultured isolated from the fungus the cells of this variety are covered individually by thick gelatinous envelopes. The phycobionts ofMegalospora gompholoma subsp.gompholoma andM. atrorubicans subsp.australis belong to a new variety ofD. symbiontica, i.e. var.pauciautosporica, which preferentially reproduces by zoo- and aplanospores. The phycobiont ofPseudocyphellaria aurata does not belong toMyrmecia reticulata as formerly thought, but toDictyochloropsis symbiontica. Specimens from one locality in Tenerife and from New Zealand are lichenized with a strain ofD. symbiontica var.symbiontica, those from another locality in Tenerife with a strain ofD. symbiontica var.pauciautosporica. These strains differ in certain characters from other lichenized strains of these varieties.

The cytogeographical structures of Ixeris nakazonei, a putative hybrid between I. debilis (6x) and I. repens (2x), were investigated in the Ryukyu Archipelago and Taiwan. In the Ryukyus, I. debilis occurs on Miyakojima Island of the southern Ryukyus and northward, while I. repens occurs on all islands except for Iriomotejima and Yonagunijima Islands. I. nakazonei, comprises six polyploid cytotypes, 3x, 4x, 5x, 6x, 7x and 8x, based on x=8. Four cytotypes from 3x to 6x occur in the central Ryukyus, while four cytotypes from 5x to 8x occur in the southern Ryukyus. The higher polyploids of I. nakazonei tend to be distributed in the more southerly area. Tetraploids of I. nakazonei always co-occur with I. debilis and I. repens, supporting the hybrid origin of this cytotype. Considering the chromosome number, octoploids, which predominate in the southern Ryukyus and Taiwan, may have derived directly from hybridization between I. debilis and I. repens. Odd-numbered polyploids of I. nakazonei, 3x, 5x and 7x, are relatively rare. Their chromosome numbers indicate that triploids and heptaploids are hybrids between the tetraploid of I. nakazonei and I. repens, and between the octoploid of I. nakazonei and I. debilis, respectively. Pentaploids of I. nakazonei in the central and southern Ryukyus are, respectively, hybrids between the tetraploid of I. nakazonei and I. debilis and between the octoploid of I. nakazonei and I. repens, indicating that pentaploids of I. nakazonei have at least two independent origins.  相似文献

Relationships between the activity of auxin polar transport and flower formation were studied using several flower mutants ofArabidopsis thaliana. The activity of auxin polar transport in the upper portion of inflorescence axis of wildtype plants ofArabidopsis thaliana was significantly lower than that of the basal part. The activities of auxin polar transport in the upper portion of inflorescence axes ofap1 andclv1 mutants were significantly higher than that of wild-type plant. However, those of other flower mutants tested,ap3-1, ag, pi, Fl-40, Fl-54, Fl-89 andpin-formed, were extremely low as compared with that of wild one. We got some evidence that the reduction of the activity of auxin polar transport is concerned with the growth and development of plants. We could mimic it by the removal of all flowers and pods including mature or immature seeds. Moreover, artificial pollination inap3-1 andpi mutants, in which no seeds are found naturally, resulted in the partial recovery of the activity of auxin polar transport in inflorescence axis. Considering these results in this study together with the fact that inhibitors of auxin polar transport generated almost same disruptions ofpin-formed orpinoid mutants which normally had no flowers in inflorescence axis (Okadaet al. 1991, Uedaet al. 1992, Bennettet al. 1995), the systern of auxin polar transport and its activity in inflorescence axis seems to be essential for the development of flower bud in early stage ofArabidopsis thaliana, and the activity of auxin polar transport is also regulated by the formation of flowers and seeds in inflorescence axis.  相似文献

Leaf removal (LR) treatments improve the photosynthetic capacity of the remaining leaves and induce flavonoid synthesis as a stress response in the common grapevine (Vitis vinifera L.). However, excessive exposure of grape berries to UV-B radiation as a result of cultural practices in the Mediterranean climate may have negative effects on berry composition. This 2-year study determined the effects of defoliation on the autochthonous red grape variety ‘Babica’ in a Mediterranean climate (wine-growing region Dalmatia, Croatia). Six leaves were removed before flowering (FLR) and at the end of véraison (the onset of grape ripening; VerLR) and were compared to the untreated control. Yield parameters, sugar content, grape must pH, total polyphenols (TP), total anthocyanin (TA) content, and individual anthocyanin compounds were measured in grape skin extracts and wines. However, the greater mean daily temperature during the vegetation period and lesser rainfall before harvest in 2018 increased yield per vine, average cluster weights, density, and total acidity, compared to 2017. Both defoliation treatments significantly reduced TP in grape extracts, but these differences were not observed in wine. Compared to the control (NLR), VerLR treatment significantly reduced TA in grape skin extracts and wine. Significantly lesser TP concentrations, in grape skin extracts and wine, as well as TA were noticed during the 2017 season. VerLR treatment reduced the concentration of nine individual anthocyanins compared to the control in grape skin extracts, while this effect was not observed in wine. Season year was a statistically significant source of variability of the individual anthocyanin contents in wine. Under specific environmental conditions LR can decrease polyphenols, especially anthocyanins, and negatively impact grape and wine quality.  相似文献

There is a worldwide trend to understand the impact of non-Saccharomyces yeast species on the process of winemaking. Although the predominant species at the end of the fermentation is Saccharomyces cerevisiae, several non-Saccharomyces species present during the first days of the process can produce and/or release aromas that improve the bouquet and complexity of the final wine. Since no genomic sequences are available for the predominant non-Saccharomyces species selected from grapes or musts (Hanseniaspora uvarum, Hanseniaspora vineae, Hanseniaspora opuntiae, Metschnikowia pulcherrima, Candida zemplinina), a reproducible PCR method was devised to discriminate strains at the subspecies level. The method combines different oligonucleotides based on tandem repeats with a second oligonucleotide based on a conserved tRNA region, specific for ascomycetes. Tandem repeats are randomly dispersed in all eukaryotic genomes and tRNA genes are conserved and present in several copies in different chromosomes. As an example, the method was applied to discriminate native M. pulcherrima strains but it could be extended to differentiate strains from other non-Saccharomyces species. The biodiversity of species and strains found in the grape ecosystem is a potential source of new enzymes, fungicides and/or novel sustainable methods for biological control of phytopathogens.  相似文献
