This experiment was conducted to evaluate the effect of administering increasing doses of royal jelly (RJ) on reproductive parameters in ewes. Additionally, this study compared using RJ vs. equine chorionic gonadotropin (eCG) in estrous cycle control. In May (transitional period between anestrous and the breeding season) 37 multiparous, winter-lambing Awassi ewes 3-6 years of age (average body weight of 53+/-1.2 kg) were fitted with intravaginal flourogestone acetate-impregnated sponges (FGA, 40 mg) for 12 days. Ewes were randomly assigned into five treatment groups to receive no RJ (CON, n=7), 250 mg RJ/d (RJ250, n=8), 500 mg RJ/d (RJ500, n=8), 750 mg RJ/d (RJ750, n=7), or 600IU eCG (eCG, n=7). Royal jelly was administrated orally on daily basis when sponges were in place while eCG was administered on the day of sponge withdrawal (d 0). Behavioral estrus was checked using fertile Awassi rams at 6h intervals for 5 days beginning on d 1. Interval from d 0 to onset of estrus was shorter (P<0.05) in eCG than in CON and RJ250 groups. No differences in the onset of estrus were detected among the RJ-treated groups. The intervals from d 0 to first progesterone rise were shorter (P<0.05) in the eCG-treated compared with RJ-treated and control ewes (100+/-15.3, 138.4+/-14, 135.7+/-15, 155.6+/-15, 154.4+/-15.1h in eCG, CON, RJ250, RJ500, and RJ750, groups, respectively). The overall pregnancy rate from mating at induced estrus was 75.7% (28/37). Of these ewes, 23/37 (64.8%) lambed within 155 days following d 0. Lambing rate was higher (P<0.05) in the RJ500 group compared with controls. Lambing rate from mating at induced estrus was 2/7 (28.5%), 4/8 (50%), 8/8 (100%), 4/7 (57%), and 5/7 (71%) in CON, RJ250, RJ500, RJ750, and eCG groups, respectively. Results of the present study demonstrate that eCG but not RJ was effective in improving estrus expression in ewes during the transition between the non-breeding and breeding seasons. Royal jelly may be effective in improving pregnancy and lambing rates but further studies are required to confirm such findings.  相似文献

An experiment was conducted to determine whether natural royal jelly (RJ) paste administered orally or intramuscularly (i.m.) in conjunction with exogenous progesterone is associated with improved reproductive responses in ewes. Thirty 3-6-year-old Awassi ewes were randomly allocated into three (RJ-capsule, RJC; RJ-injection, RJI and control, CON) groups of 10 ewes each. All ewes were treated with intravaginal progesterone sponges for 12 days. Ewes in the RJC and RJI were administered orally or i.m. with a total of 3g of RJ given in 12 equal doses of 250 mg per ewe per day starting at the time of sponge insertion. At the time of sponge withdrawal (day 0, 0 h), ewes were exposed to three rams and checked for breeding marks at 6-h intervals for 3 days. Blood samples were collected from all ewes for analysis of progesterone concentrations. Pretreatment progesterone levels were <0.5 ng x ml(-1) in 16/30 and >1.3 ng x ml(-1) in the remaining ewes indicating luteal function and cyclicity. Similar reproductive responses and progesterone levels occurred in ewes of the RJC and RJI; therefore, data of the two groups were pooled. Following sponge insertion, progesterone levels increased rapidly and reached maximum values of 5.8+/-0.2 ng x ml(-1) within 2 days among ewes of the three groups, and then declined gradually to day 0 values of 1.6+/-0.1 and 1.9+/-0.1 ng x ml(-1) for the RJ-treated and CON ewes, respectively. The rate of progesterone decline was greater (P<0.001) in RJ-treated than in CON. Mean progesterone levels during the 12-day period were lower (P<0.001) in RJ-treated than in CON (2.8+/-0.2 ng x ml(-1) versus 3.3+/-0.2 ng x ml(-1)). Treatment with RJ resulted in greater (P<0.05) incidence of oestrus and shorter (P<0.05) intervals to onset of oestrus than CON. Based upon progesterone levels, ovulation occurred following day 0 in all ewes. Progesterone increased on day 3 in RJ-treated and on day 4 in CON ewes. Progesterone remained elevated through day 18 in 8/20 RJ-treated and 1/10 CON ewes (P=0.09). All pregnant ewes exhibited oestrus 14 h earlier (P<0.02), ovulated approximately 1 day earlier and had higher (P<0.001) luteal phase progesterone levels than non-pregnant ewes. Non-pregnant had higher (P<0.04) body weights than pregnant ewes. In conclusion, results demonstrate that both RJ treatments in conjunction with exogenous progesterone were equally capable of improving oestrus response and pregnancy rate.  相似文献

An experiment was conducted to determine whether factors affecting pregnancy rate out-of-season are associated more with transcervical artificial insemination (T-AI) procedures or with the reproductive state of the ewe. Twenty Finncross ewes were treated with progesterone sponges, and at sponge removal (0 h) 10 ewes were treated with eCG. Blood samples were collected for LH and progesterone analyses, and follicular development was monitored using ultrasonography. Ewes were inseminated from 48 to 52 h with 200 million motile frozen-thawed spermatozoa. The incidence of estrus, LH surges and ovulation was greater (P < 0.01) and intervals to these responses were shorter (P < 0.01) in the eCG-treated ewes. The number of follicles > 5 mm was higher (P < 0.05) in eCG-treated than control ewes. Progesterone concentrations increased and remained elevated through Day 19 in 7 eCG-treated and in 1 control ewe, and these ewes were pregnant based upon ultrasonographic examination. The results demonstrate that the T-AI technique using frozen-thawed semen produces a relatively high (70%) pregnancy rate out-of-season. The pregnancy rate was found to reflect primarily the reproductive condition of the ewe.  相似文献

A standard dose of 500 IU of eCG is commonly given to progestogen pre-treated anestrous ewes for induction of estrus. Twelve seasonally anestrous and 12 cyclic Western White Face ewes were treated for 12 days with intravaginal sponges impregnated with medroxyprogesterone acetate (MAP). In trials in both the breeding and nonbreeding seasons, six randomly selected ewes were given 500 IU of eCG at sponge removal to determine the effects of low dose of eCG on ovarian antral follicular dynamics and ovulation. Ultrasound scanning and blood sampling were done daily. Treatment with eCG did not have marked effects on antral follicular growth. All ewes ovulated, except for five of six control anestrous ewes. Luteal structures and progesterone secretion were confirmed in all but the control anestrous ewes. In the breeding season, peak progesterone concentrations were greater (P<0.05) in eCG-treated compared to control ewes. Daily serum estradiol concentrations were greater in the periovulatory period in eCG-treated compared to control ewes (treatment-by-day interaction; P<0.05), particularly in anestrus. Progestogen-treated ewes ovulated follicles from several follicular waves, in contrast to ovulations of follicles from the final wave of the cycle in untreated, cyclic ewes. Anestrous ewes exhibited more frequent follicular waves and FSH peaks compared to cyclic ewes after a progestogen/eCG treatment. In conclusion, 500 IU of eCG given after 12 days of progestogen treatment had limited effects on the dynamics of ovarian follicular waves. However, eCG treatment increased serum concentrations of estradiol during the periovulatory period, particularly in anestrous ewes; this probably resulted in the synchronous estrus and ovulation in anestrous ewes.  相似文献

The objectives were to evaluate, in anestrous ewes, the effectiveness of a CIDR-G device (0.3 g progesterone) administered for 5 d to induce estrus; and FSH (Folltropin; 55 mg NIH-FSH-P1 equivalent) in saline:propylene glycol (1:4) 24 h before insert removal (Day 0), to increase ovulation rate and prolificacy. Ewes of mixed breeding were assigned at random to 3 treatments: control (C; n = 125), 5 d progesterone (P5; n = 257) and 5 d progesterone plus FSH (P5F; n = 271). Intact rams were joined at insert removal and ewes were observed every 24 h for 3 d. On Day 14, the ovulation rates of all ewes detected in estrus in the treated groups were determined using transrectal ultrasonography. Rams were removed on Day 26 to 31. Ewes were examined for pregnancy then, and again 20 to 25 d later to detect ewes that conceived to the second service period. Percentage of ewes marked by rams was higher in progesterone-treated (77%) than in C (20%; P < 0.01), but did not differ between P5 and P5F. The ovulation rate (1.95+/-0.04) did not differ due to FSH. Conception (68%) and pregnancy (52%) rates were higher in progesterone-treated (P < 0.01) than in C (0%) ewes. Estrous response varied quadratically with time after ram introduction, and the conception rate varied quadratically with the time of observation of onset of estrus. Over two service periods more progesterone-treated than C ewes lambed (65 vs 45%; P < 0.01). Lambs born per ewe exposed (0.7+/-0.1, 1.0+/-0.1, and 1.1+/-0.1 for C, P5 and P5F, respectively) was increased by progesterone (P < 0.05). Litter size to the first service period (1.59+/-0.04) and overall (1.54+/-0.03) did not differ among treatment groups. FSH-treated ewes tended to have more lambs (1.67+/-0.1) than did ewes receiving progesterone alone (1.5+/-0.1; P = 0.06) and than did ewes lambing to the second service period (1.5+/-0.1; P = 0.06). In summary, a 5-d progesterone pre-treatment of anestrous ewes induced estrous cycles and increased the pregnancy rates. A single injection of FSH only tended to increase litter size.  相似文献

The effects of melatonin implants on out-of-season breeding in New Zealand Romney composite ewes, was determined by comparison of reproductive performance in ewes treated with progesterone+equine chorionic gonadotrophin (eCG) (control; n=107), melatonin+progesterone+eCG (n=97) or melatonin+progesterone (n=96). Conception rates in melatonin+progesterone+eCG-treated ewes (67%) were higher than in the control ewes (P<0.01; 47%). Pregnancy rates were higher in melatonin+progesterone+eCG-treated ewes (55%; P<0.001) compared with the control ewes (40%). Fewer melatonin+progesterone-treated ewes displayed oestrus (14%; P<0.001) and subsequently became pregnant (6%). Oestrus rates in melatonin+progesterone-treated ewes (14%) were lower than both the melatonin+progesterone+eCG-treated (82%) and control ewes (86%; P<0.001), which were similar to each other. The number of foetuses per pregnant ewe was similar in all three treatment groups. Serum melatonin concentrations at Day -9 were higher in the ewes treated with melatonin and there was a large variation between individual ewes, but concentrations were similar for pregnant and nonpregnant ewes. The combination of higher conception rate and the trend for more lambs per pregnant ewes resulted in more lambs being born per ewe treated in melatonin+progesterone+eCG-treated ewes compared to the other two treatment groups. These results suggest that melatonin implants, in conjunction with administration of progesterone and eCG, may be suitable as a means of increasing the number of lambs born per ewe treated in an out-of-season breeding program in New Zealand sheep flocks while melatonin and progesterone is not.  相似文献

The main objective of this study was to investigate the effectiveness of certain progestagen-gonadotrophin treatments on synchronization of estrus in sheep. In Experiment I, 30 Chios ewes were treated at the beginning of the breeding season with medroxyprogesterone acetate (MAP) intravaginal sponges for 12 days and a single i.m. treatment of either FSH (Group 1,10 IU, n = 8; Group 2, 5 IU, n = 8; Group 3, 2.5 IU, n = 8) or eCG (Group 4, 400 IU, n = 6) at the time of sponge removal. Ten days after sponge removal laparotomy was performed to record ovarian response. Clinical estrus was observed in more (though not at a significant level) FSH treated than eCG treated sheep (62.5% versus 33.3%). Administration of 400 IU eCG resulted in the highest mean number of CL perewe ovulating (2.8 +/- 0.2), with administration of 10 IU FSH producing the next best results (2.1 +/- 0.3). Statistically significant differences in the mean number of CL per ewe ovulating were found only between ewes in Group 3 (1.7 +/- 0.4) and Group 4 (2.8 +/- 0.2) (P < 0.05). In Experiment II, 53 Chios and 30 Berrichon ewes were treated during the mid-breeding season with MAP intravaginal sponges for 12 days and a single i.m. treatment of either 10 IU FSH (27 Chios and 16 Berrichon ewes) or 400 IU eCG (26 Chios and 14 Berrichon ewes), at the time of sponge removal. Ewes that were in estrus on Days 2-4 and 19-23 after sponge removal were mated to fertile rams. No significant differences were recorded between treatment or breed groups in the proportions of ewes observed in estrus after treatment. In the Berrichon breed, FSH administration resulted in higher lambing rates (93.8% versus 57.1%, P < 0.05) and higher mean number of lambs born per ewe exposed to rams (1.4 +/- 0.2 versus 0.8 +/- 0.2, P < 0.05) than that of eCG. After treatment with eCG, the mean number of lambs born per ewe exposed to rams was higher in the Chios than the Berrichon breed (1.4 +/- 0.2 versus 0.8 +/- 0.2, P < 0.05). After treatment with FSH, the lambing rate was higher in the Berrichon than the Chios breed (93.8% versus 63.0%, P < 0.05). In conclusion, a single FSH treatment (5 or 10 IU) at the end of progestagen treatment appears to be more effective than eCG for the induction of synchronized estrus in sheep at the beginning of the breeding season, with no cases of abnormal ovarian response observed. During the mid-breeding season FSH (10 IU) appears to be equally as effective as eCG (400 IU) in respect of lambing rate and mean number of lambs born per ewe.  相似文献

Accelerated lambing system is heavily reliant on reproductive technologies meant to enable off the season breeding in sheep. Therefore, the present study was programmed to assess the effect of breeding season (BS) on fertility of sheep following estrus synchronization (ES) and fixed time artificial insemination (FTAI). A total of 248 and 365 ewes were synchronized and inseminated during the BS (Febuary–March and July–September) and non-breeding season (NBS) respectively, during 2012–14. Synchronization of estrus was done by AVIKESIL-S, intra-vaginal progesterone sponges kept in situ in vagina for 12 days. 200 IU eCG was administered intramuscularly on 12th day after sponge withdrawal. FTAI was performed twice in ewes exhibiting signs of estrus at 48 and 56h after sponge removal, using liquid chilled semen of Patanwadi/Malpura rams containing 100 million sperm. Breeding season had no significant (p<0.05) effect on estrus synchronization. The estrous responses during the BS and NBS were 84.68% and 83.29% respectively. The lambing percentage during BS was 66.67%, which is significantly (p<0.05) higher than the lambing percentage of NBS (57.57%). Although, the lambing percentage in NBS maneuvered ewes were lower than the BS ewes but still the technology can be used to offset the effect of anestrus and to augment production in sheep.  相似文献

The response to superovulatory (SOV) and estrus synchronization (ES) treatments and the fertility of donor (n=68) and recipient (n=118) Saloia ewes was evaluated in the fall and spring breeding seasons. The proportion of acyclic ewes at treatment time was significantly higher in the spring than in the fall (42.6% versus 4.0%, P<0.00001). Donors treated with eCG had a significantly higher mean number of follicles over 5mm in diameter in the ovaries at embryo recovery and a significantly lower mean efficiency of recovery than FSH-treated ewes. These negative effects were more pronounced in the fall than in the spring, which resulted in a significantly lower mean number of total and fertilized ova recovered from eCG-treated ewes, compared to FSH donors in the fall, but not in the spring. Season had no significant effect on the ovulation rate and plasma P4 concentrations of recipients treated with a progestagen plus eCG combination. Although the recipient lambing and embryo survival rates were higher in the fall than in the spring the differences were not significant. No significant differences were observed in the ovulation rate or P4 concentrations of recipients that lambed compared to those that did not lamb. These preliminary results show that, in Portugal, response of Saloia ewes to SOV or ES treatments and donor fertility following the SOV treatment were similar in the spring and the fall, which suggests that in the spring acyclic ewes are in moderate anestrus. The effect of season on fertility following embryo transfer should be confirmed in further studies involving a larger number of animals. The semilaparoscopic transfer method reported here allowed lambing and embryo survival rates higher (although not significantly) than a standard surgical approach.  相似文献

Two experiments were conducted to examine the effect of progestagen supplement 24h prior to intravaginal pessary withdrawal on reproductive performance of seasonal anestrous ewes. Ewes in each experiment were allocated to treatment and control and all were induced to estrus using either intravaginal MAP (Exp. 1; n=24) or CIDR-G (Exp. 2; n=28) pessaries for 12 days. Half of the ewes in each experiment were supplemented 24h before withdrawal of pessaries with either 10mg oral MAP tablets (Exp. 1) or 25mg i.m. progesterone (P(4)) administration (Exp. 2; P(4)-supplement-treated group). Fertile rams were allowed with the ewes at sponge removal (Day 0, 0h) and estrus was monitored at 6-h intervals for 3 days. Blood samples were collected for measurements of P(4) (Exp. 1 and Exp. 2) and LH (Exp. 2). In both experiments, the percent of ewes in estrus was greater (P<0.05) and intervals to estrus were longer (P<0.05) in progestagen-supplement-treated than control ewes. In Exp. 2, the occurrence and magnitude of LH surges were greater (P<0.01) and intervals to onset of LH surge were longer (P<0.01) in P(4)-supplement-treated than control ewes. In Exp. 2, P(4) supplement elevated P(4) levels from 1.8+/-0.1ng/mL on Day -1 to 4.2+/-0.3 on Day 0 (P<0.001). Following pessaries removal, P(4) concentrations fell to basal values on Day 1 in both groups and remained low until Day 5. Then, P(4) concentrations increased and remained elevated through Day 19 in all (100%) progestagen-supplement-treated in Exp. 1 (12/12) and Exp. 2 (14/14) and in only 5/12 (41.7%) and 6/14 (42.9%) control ewes, respectively. These ewes were confirmed pregnant by ultrasonography and lambed on Day 149.2+/-0.2 following Day 0. In conclusion, progestagen supplement 24h prior to removal of pessary can be used successfully to improve reproductive performance of ewes bred out-of-season.  相似文献

The ability of recombinant ovine interferon-tau (roIFNtau) to extend the interestrous interval (IEI) in sheep was studied. Ewes were fitted with bilateral uterine catheters 7 or 8 days post estrus and were assigned to receive either 10 or 20 million antiviral (AV) units/day i.u. ( approximately 100 or 200 ug) of roIFNtau or ovine conceptus secretory proteins containing equivalent AV units of native oIFNtau (noIFNtau; 4 ewes/treatment). Four control ewes received ovine serum proteins (SP). Total protein injected was 6 mg per day, half at 0700 hours and half at 1730 hours. The treatments were administered from Day 11.5 (estrus=Day 0) to Day 16. Blood samples were collected by jugular vienipuncture daily from Day 11 until ewes returned to estrus. Concentrations of progesterone (P) in plasma were determined by RIA. Treatment with either noIFNtau or roIFNtau extended IEI beyond that of SP-treated ewes (19.1 vs 31.2+/-3.4 days P<0.03). Of the ewes receiving 100 mug/day of oIFNtau, 2 of 4 receiving noIFNtau (23.6+/-5.2 days) and 3 of 4 receiving roIFNtau (34.2+/-5.2 days) had an extended IEI. All ewes receiving 200 mug/day of noIFNtau or roIFNtau had an extended IEI (28.8 and 38.5+/-5.2 days. respectively). Ewes receiving roIFNtau had a longer IEI than those receiving noIFNtau (36.7 vs 26.2+/-3.4 days; P=0.07). Ewes with an extended IEI had functional corpora lutea, as assessed by P production. The results demonstrate that 10 or 20 million AV units ( approximately 100 or 200 ug) of roIFNtau extends the IEI and that the length of the IEI is longer for ewes receiving roIFNtau than noIFNtau following injection of equivalent AV units.  相似文献

The effects of monensin and progesterone priming on reproductive performance (estrous response, lambing rate and prolificacy) of grazing Boutsiko mountain breed adult and 18-mo.-old ewes at the end of seasonal anestrus were investigated. In Experiment 1 the feed supplement with or without monensin was offered for 21 d after introduction of vasectomized rams (Day 0). Progesterone was administered to the ewes in the respective groups as a single injection at Day -3. Ewes of both age groups were assigned randomly to 1 of 4 treatments: C, C+P, C+M and C+M+P. In Experiment 2 the supplement C or M was offered from Day -26 to Day 21. The treatments consisted of C, C+P and C+M+P. Blood samples were taken 50 h after ram introduction for determination of plasma concentrations of P and insulin-like growth factor-I (IGF-I). There was a greater increase in estrous response at Days 17 to 19 and at Days 0 to 19 when supplementation was offered before rather than after ram introduction in both age groups. In the adult group ewes synchronization of estrus at Days 17 to 19 was significantly increased by administration of monensin (P<0.05) and progesterone (P<0.01) compared with the control group in the first but not the second experiment. The incidence of estrus at Days 17 to 19 or at Days 0 to 19 was highest in the adult groups treated with monensin and progesterone in both experiments. In 18-mo.-old ewes progesterone was effective in synchronizing estrus only in Experiment 2. Mean plasma IGF-I concentrations were increased by monensin treatment (P<0.05) in adult ewes that were at the periovulatory stage at blood sampling time. Correlation coefficients between IGF-I and progesterone concentrations in monensin plus progesterone group adults were -0.715 (P<0.02) and -0.516 (P<0.01), respectively across all treatments. The results suggest that monensin and progesterone priming improved reproductive performance, and the monensin-induced increase in plasma IGF-I levels at the periovulatory stage may be causally related to the ability of ovulatory follicles to develop into functional corpora lutea (CL).  相似文献

A series of studies was designed to identify methods of improving out-of-season breeding success in ewes. In Experiment 1, 190 mature ewes were assigned to receive in April, either: (A) a control ration of 0.3 kg corn twice daily for 8 d before ram introduction (control ewes n=49), (B) the control ration containing 0.125 mg of melengestrol acetate (MGA) in 0.3 kg corn (MGA8a ewes n=46), (C) the control ration or 7.5 d followed by 1 feeding of 0.5 mg MGA in 0.3 kg corn (MGA1 ewes n=48), or (D) the control ration plus a 20 mg i.m. injection of progesterone on D 8 (P ewes n=47). Ewes were exposed to rams for 21 d. A greater percentage of MGA8a and P ewes lambed than did control ewes (P < 0.04). The lambing rate was greatest among MGA8a (P < 0.02 vs. control), intermediate among P ewes (P < 0.19 vs. control) and least among MGA1 and control ewes (P > 0.79). In Experiment 2, 70 mature ewes were assigned to receive in June, either: (A) a control ration of 0.3 kg of corn twice daily for 8 d before to ram introduction (control ewes n=25), (B) the control ration containing 0.125 mg of MGA per 0.3 kg corn (MGA8b ewes n=21), or (C) the control ration and simultaneous treatment of ewes with a progesterone-containing controlled internal-drug releasing device (CIDR ewes n=24). Ewes were exposed to rams for 21 d. Both CIDR and MGA8b ewes exhibited estrus earlier than did control ewes (P < 0.01). The CIDR ewes exhibited estrus earlier than did the MGA8b ewes (P < 0.05). A greater percentage of ewes treated with CIDR or MGA8b lambed than did control ewes (P < 0.01), with more CIDR ewes lambing than MGA8b ewes (P < 0.01). The lambing rate was greater in CIDR ewes than in control ewes (P < 0.04). These data provide evidence that several options exist to improve pregnancy success in ewes bred out of season and that success varies with method of treatment.  相似文献

Two experiments were conducted to test the ability of clenbuterol, a beta-sympathomimetic agonist, to delay lambing. Ewes in all experiments were induced to lamb with 2.5 mg of flumethasone given intramuscularly on Day 141, 142 or 143 of gestation. Beginning on the day of flumethasone administration and continuing daily until lambing, ewes in Experment 1 were given injections intramuscularly (i.m.) with either 1) vehicle solution at 2200 h, 2) 240 ug of clenbuterol at 2200 h or 3) 240 ug of clenbuterol at 1600 and at 2200 h. There was a greater proportion of ewes lambing between 0800 h and 1600 h (P<0.01) and more stillbirths (P=0.05) in the groups which received clenbuterol than in the vehicle control group. Experiment 2 was conducted to determine if a lower dosage of clenbuterol, given as a single injection 37 h after flumethasone, would delay lambing without increasing the proportion of stillbirths. Ewes received either the vehicle solution, or 36 or 146 ug of clenbuterol. Lambing was delayed in the group which received 146 ug but not 36 ug of clenbuterol. There were no adverse effects of treatment on lamb survival in Experiment 2.  相似文献

The objective of this experiment was to determine the effect of a 5-day progesterone priming prior to a GnRH-PGF2alpha treatment on reproductive performance of anestrous goats. Thirty-six Mountain Black goats were randomly assigned in a 2 x 2 factorial arrangement and were administered intravaginally on day -12, either with 300 mg progesterone inserts (CGPE and CGP) or with 0 mg progesterone (GPE and GP) for 5 days. On day -6, the goats were injected with 100 microg GnRH, followed 6 days later by 15 mg PGF2alpha (day 0), the time at which the goats in the CGPE and GPE groups were administered 300 IU eCG injections and those in CGP and GP groups were administered the control solution. The goats were exposed to four fertile bucks at 0 h and were checked for breeding marks at 6-h intervals for 72 h. Blood samples were collected from all goats for progesterone analysis. Progesterone concentrations increased only in CGPE and CGP during the period of device insertion but remained low in GPE and GP groups (P < 0.001). Progesterone levels at the time of GnRH injection on day -6 were basal (0.2 +/- 0.04 ng.mL-1) among the groups and began to increase starting on day -2. Day 0 progesterone concentrations differed (P < 0.05) among groups and were significantly influenced by CIDR-G (P < 0.001). A similar proportion of goats expressed estrus and intervals to detected estrus were shorter (P < 0.05) in the CGPE and GPE groups than in GP with no difference between the CGPE, CGP and GPE or between CGP and GP groups. The number of goats ovulating based upon elevated progesterone levels on day 0 was significantly greater (P = 0.002) in CGPE (9/9) and CGP (9/9) than GPE (6/9) and GP (5/9) groups and was significantly influenced by CIDR-G (P = 0.03). All pregnant goats had elevated progesterone concentration on day 0 and none of the goats with basal progesterone levels became pregnant. Pregnancy and kidding rates, twinning percentage and the number of kids born per goat exposed were greater (P < 0.05) among goats treated with progesterone and eCG. In conclusion, progesterone priming and eCG are essential for producing higher rates of pregnancy and kidding in GnRH-PGF2alpha-treated anestrous goats.  相似文献

This trial was conducted to evaluate the effectiveness of multiple eCG injections in the induction of estrus and pregnancy in Colored Mohair goats during the anestrus season. It was also aimed to determine total dose of eCG required for induction of estrus. Ten multiparous and lactating goats were used. The goats were randomly divided into two groups and treatments were started on May 22. Group eCG (n=5) was treated with eCG intramuscularly for 6 days. Daily dosages of eCG from May 22 to May 27 were 300 IU, 200 IU, 200 IU, 100 IU, 100 IU and 50 IU, respectively. Goats in control group received no treatment. Blood samples were taken from animals in each of the two groups just before and after the beginning of the treatments and serum progesterone concentrations were assayed by RIA. Starting on the fourth day after the first treatments, goats were exposed to fertile bucks twice daily for 30 min to detect standing heat. The estrus goats were allowed to be mated by the bucks. Pregnancies were determined 40 days after mating by real-time ultrasonography. One goat on day 5 and three goats on day 7 exhibited behavioral estrus in eCG group (80%) after the first eCG injection. Three of them (75%) became pregnant. None of the goats in the control group exhibited behavioral estrus. Mean serum progesterone concentrations had prominent elevations indicating ovulation in eCG group, but not in control group, after 20 days from the first treatments. Progesterone concentrations of eCG group were significantly different than those of control group on days 20 and 28 (P<0.05). The results suggest that divided multiple injections of a total 950 IU eCG are effective without progestagen pretreatment in the induction of estrus and obtaining successful pregnancy and live kids in Colored Mohair goats during the anestrus season.  相似文献

A comparison was made of the relative effectiveness of subcutaneous ear implants containing 2 mg Norgestomet or vagnial pessaries containing 60 mg medroxyprogesterone acetate (MAP) to induce estrus and conception in dry anestrous ewes. Groups of ewes were treated with one of the two progestogens for 14 d, and 500 IU pregnant mare serum gonadotropin (PMSG) was administered intramuscularly at the time of progestogen withdrawal. No significant differences in estrus induction, pregnancy rate or number of lambs born per ewe lambing were observed. Ewes treated with Norgestomet had 96% estrus, 60% pregnancy rate and 1.4 lambs per ewe lambing. Comparably, ewes treated with MAP had 94% estrus, 65% pregnancy rate and 1.7 lambs per ewe lambing. Norgestomet implants compared favorably with MAP pessaries for estrus induction and breeding of commercial, dry anestrous ewes.  相似文献

The purpose of the study was to investigate the effects of different doses of equine chorionic gonadotropin (eCG) treatment on follicular development, ovulation and pregnancy rate during the breeding season in fat-tailed Chall ewes. Seventy-two cycling (62.5 ± 2.5 kg), multiparous Iranian Chall ewes were used in the trial. The ewes were randomly allocated to 6 groups (n = 12/group). Estrus was synchronized with the aid of controlled intravaginal drug release (CIDR) devices, inserted for 14 days. At the time of CIDR removal (day 14), the ewes received i.m. either 0 (control group, G0), 450 (G450), 550 (G550), 650 (G650), 750 (G750) or 850 (G850) IU eCG. Vasectomized rams were used to detect estrus in the ewes from 24 h after CIDR removal. Ovarian follicular activity was monitored with the aid of transrectal ultrasonography on the day of CIDR insertion (day 0) and daily from the day of eCG treatment (day 14), until estrus (day 16). During these days, blood samples were collected for the determination of plasma progesterone and estradiol concentrations. Laparoscopic intrauterine inseminations were conducted 54–60 h after CIDR removal. The number of CL's and pregnancy diagnosis was recorded using ultrasonography 7 and 54 days following AI, respectively. Half of ewes in control group and most of the ewes treated with eCG showed signs of estrus within 36 h of CIDR removal. The ewes in groups G750 and G850 recorded the highest number of large follicles at estrus and CL's 7 days later. The pregnancy rate in groups G550 (75.0%) and G650 (75.0%) was higher (P < 0.05) than that in the other groups. The ovarian response and estradiol concentration, as well as pregnancy rate showed that 550 or 650 IU eCG treatment is the most effective doses in improving the pregnancy rate in Iranian Chall ewes.  相似文献

Two experiments were designed to evaluate the possibility of simplifying superovulatory treatments in Corriedale ewes with use of ovine FSH (oFSH). Ewes received intravaginal progestogen sponges for 14 days. In Experiment 1, several simplified schedules were tested. Ewes were treated with 176 NIH-FSH-S1 units' oFSH given as a single injection in saline, along with 500 IU eCG 48 h before sponge removal (Group A1), in four equal doses (B1), or given as a single injection in a polyvinylpyrrolidone vehicle (C1) 24 h before sponge removal. In Experiment 2, the simplified protocol that exhibited the most desirable results in Experiment 1 (A2) was compared with the same protocol, but using less oFSH (132 units) (B2) and with the most conventional protocol (176 units of oFSH in eight decreasing doses; C2). Estrus was detected and ewes were naturally mated. The ovarian response and embryo production were assessed on Day 6 after estrus. LH was measured at 6h intervals from pessary withdrawal. The onset of estrus and the pre-ovulatory LH surge were advanced (P<0.05) in ewes treated with FSH and eCG. In Experiment 1, protocol A1 produced a greater percentage of superovulated ewes compared to C1 (100.0 compared with 58.3%; P<0.05), increased ovulation rate (13.8 corpora lutea compared with 6.2 and 4.7 for B1 and C1, respectively; P<0.05), and tended to increase the number of transferable embryos compared to B1 (P=0.08). In Experiment 2, percentages of superovulated ewes and ovulation rates were similar among groups; however, Group A2 tended to have more large follicles (P=0.07) than C2. The number of transferable embryos was similar among the three treatments. In conclusion, the reduced-dose oFSH given once along with eCG is the most appropriate superovulatory treatment because it combines simplicity and a lesser dose of gonadotropin, which also implies a reduction in cost, without reducing embryo production.  相似文献

The efficacy of using a low cost system for delivering progesterone as part of an estrous synchronization protocol in sheep was evaluated. In experiment 1, Barbados Black Belly ewes (n=34) and ewe lambs (n=35; 37.5±0.9 kg) were assigned to be untreated, control animals (C), or to receive PGF(2α) on d0 (PG), or receive two injections of progesterone (200mg, i.m. each) on D -5 and on D -2.5, prior to PGF(2α), on D 0 (2PPG). Treatment with 2PPG increased the proportion of animals lambing to the first service (P<0.05), an effect that was greater in ewe lambs than ewes (treatment × parity; P<0.05). The interval from ram introduction to lambing and the mean lambing day was less (P=0.04) in the 2PPG-treated animals compared to control animals. In Experiment 2, lactating ewes from experiment 1 (n=61) 60-85 days postpartum were assigned within parity and number of lambs reared to remain nursing (S; n=29) or weaned (W; n=32) 3 weeks prior to treatment with the 2PPG protocol. There was no effect of treatment on the proportion of animals lambing to the first service or overall, interval from ram introduction to lambing and lambing interval. An 8-month lambing interval was observed in ewes in which estrus was synchronized regardless of physiological state. In conclusion, the two-progesterone injection synchronization protocol may be used as a practical low cost and efficient method of synchronizing estrus to reduce the lambing interval and maximize productivity in tropical breeds of sheep.  相似文献
