The mass of tubulin protein in developing embryos of the sea urchin Lytechinus pictus was measured using a radiodilution immunoassay based on densitometric analysis of immunoprecipitated tubulins resolved electrophoretically. The tubulins constitute an average of 360 +/- 35 pg per egg, or 0.66% of the total protein, and there is no significant change in their concentration during embryogenesis. The masses of soluble and polymerized tubulin were measured for extracts prepared under conditions that stabilize microtubules. In eggs, a maximum of 14% of the tubulin is insoluble, and this increases throughout embryogenesis to 67% at pluteus stage (72 hr). The concentration of tubulin in eggs is at least 500 micrograms/ml, well above the critical concentration for tubulin assembly in vitro, yet microtubules have not been observed in eggs. The mass of newly synthesized tubulin, estimated from the mass of tubulin mRNA per embryo, accounts for a small fraction of the total tubulin by the end of gastrulation but for over half of the tubulin by the 72-hr pluteus stage. These observations are consistent with a model in which the declining level of unpolymerized tubulin controls the stability of tubulin mRNa, providing an autogenous regulation of the ontogenetic pattern of tubulin synthesis during sea urchin embryogenesis (Gong and Brandhorst, Development 102: 31-43).  相似文献

Egg masses from wild populations of the screwworm, Cochliomyia hominivorax (Coquerel), were obtained by collecting females from rotted liver, holding them for 3 d until egg maturation and then placing them on heated ground beef for oviposition. Nearly 50% of the females oviposited. Fertility of egg masses was 66 and 95% at the two collection sites. Decreased fertility was associated with collections of virgin females. Average weight +/- SD of the egg masses was 13.7 +/- 1.40 mg; the number of eggs per mass averaged 343. This method of obtaining egg masses from wild screwworm females is a cost-effective, sensitive alternative to the use of sentinel animals in the field. The use of rotted liver costs less, takes less time, and takes less equipment than the traditional sentinel animal technique.  相似文献

Synopsis The influence of current velocity on the survival and development of lingcod embryos was investigated in the field and laboratory. Examination of egg masses at five lingcod spawning sites indicated that embryo mortalities were high (up to 95%) at low-current sites because of inadequate ventilation and resulting hypoxia. Development of embryos near the center of poorly ventilated egg masses was retarded relative to development of embryos near the periphery. Hatching of embryos from poorly ventilated eggs was protracted; embryos from the interior of egg masses hatched later and were significantly smaller than embryos from eggs near the periphery. Oxygen levels measured in egg masses at low-current velocity sites during tidal flow average 16% air saturation, corresponding to a Median Tolerance Limit (LT50) of about 73 h. Oxygen levels measured in egg masses at high-current velocity sites during slack water average 69% air saturation, a level that did not adversely affect the embryos. Current velocities of 10–15 cm s–1 were needed to maintain interstitial oxygen levels in egg masses near that of the ambient water. Water movement may be an important stimulus for spawning site selection by lingcod. In areas where tidal currents were weak, spawn deposition occurred in shallow water where waves and vertical tide motion created water movement. In areas where tidal currents were strong, spawns were consistently deposited in deeper water.  相似文献

Coastal spawning grounds of the squid Doryteuthis gahi were identified for the first time at three sites in Central Chile during 2014 and 2015. A total of 15 egg masses were collected from between 10 and 15?m depth and brought to the laboratory for evaluating capsular, embryonic and when possible, paralarval characters. Egg capsules from all spawning areas were similar in size (~25?mm in length) and number of embryos per capsule (~15). Egg-laying patterns, in addition to field observations, suggest that relatively small D. gahi may be using shallow waters in semi-protected environments to reproduce and spawn. Additionally, the differential use of healthy versus defoliated kelp Lessonia trabeculata suggests that egg-laying females selectively choose substrata that limit mechanical damage to the fragile egg capsules and that also allow for adequate water flow, which could be reduced in a mass of kelp fronds. These findings highlight the opportunistic behaviour of D. gahi which use overgrazed L. trabeculata, a condition that is widespread in areas with high herbivory pressure.  相似文献

Effects of soil matrix potential on longevity of egg masses of Meloidogyne incognita were determined during simulated winter conditions. Egg masses were recovered from isolated root fragments incubated in field soil at matrix potentials of -0.1, -0.3, -1.0, and -4.0 bars throughout winter survival periods of 10 weeks for tomato roots and 12 weeks for cotton roots. Egg masses were more superficial on cotton roots than on tomato roots and were more easily dislodged from cotton roots during recovery of root fragments by elutriation. The rate of decline in numbers of eggs and J2 per egg mass was greater in wet as compared to dry soils (P = 0.01), with the relationship between numbers of eggs and J2 per egg mass and time being best described by quadratic models. Percentage hatch of recovered eggs declines linearly with time at soil matrix potentials of -0.1 and -0.3 bars, but at -1.0 and -4.0 bars the percentage hatch of recovered eggs increased before declining. Effects of soil matrix potential on numbers of eggs per egg mass and percentage hatch of recovered eggs were consistent with previous reports that low soil moisture inhibits egg hatch before affecting egg development. Estimations of egg population densities during winter survival periods will be affected by ability to recover infected root fragments from the soil without dislodging associated egg masses. There is a need for procedures for extraction of egg masses not attached to roots from the soil.  相似文献

A mturation-rate relationship for Meloidogyne incognita on Lycopersicon esculentum ''Rutgers'' was derived and used to estimate harvest dates for maximmn egg hatch from laboratory cultures at ambient temperatures. Daily maturation increments were totaled (nematode maturation total, NMT) and correlated with hatch from isolated white, yellow, and amber egg masses. Hatch per mass fluctuated periodically from ca. 1.0 NMT, when egg masses were first visible, to 2.5 NMT by which time plants showed stress. Maximum yields from white and yellow masses occurred, with a shorter than expected periodicity, at 1.5-1.8 and 2.1-2.2 NMT. White masses decreased from 90% of the total masses at 1.0 NMT to 5% at 2.5 NMT, as the proportion of yellow and amber masses increased concomitantly. Harvested masses per gram of root varied from 97 to 276; hatch per gram of root, 11,000 to 86,000.  相似文献

Damaging effects of UVB in conjunction with other stressors associated with global change are well‐established, with many studies focused on vulnerable early life stages and immediate effects (e.g., mortality, developmental abnormalities). However, for organisms with complex life cycles, experiences at one life stage can have carry‐over effects on later life stages, such that sublethal effects may mediate later vulnerability to further stress. Here, we exposed embryos in benthic egg masses of the New Zealand intertidal gastropod Siphonaria australis to treatments of either periodic stress (e.g., elevated UVB, salinity, and water temperature mimicking tidepool conditions in which egg masses are commonly found during summer) or control conditions (low UVB, ambient salinity, and water temperatures). Although there was high mortality from stressed egg masses, 24% of larvae hatched successfully. We then exposed the hatching larvae from both egg mass treatments to different combinations of water temperature (15 or 20 °C) and light (high UVB or shade) 12 h per day for 10 days. The most stressful larval conditions of 20 °C/high UVB resulted in low survival and stunted growth. Carry‐over effects on survival were apparent for shaded larvae exposed to elevated temperature, where those from stressed egg masses had 1.8× higher mortality than those from control egg masses. Shaded larvae were also larger and had longer velar cilia if they were from control egg masses, independent of larval temperature. These results demonstrate that previous experience of environmental stress can influence vulnerability of later life stages to further stress, and that focus on a single life stage will underestimate cumulative effects of agents of global change.  相似文献

Poecilogony is a relatively uncommon life-history strategy that results in the production of two different larval forms from the same egg mass (e.g., free-swimming lecithotrophic larvae and post-metamorphic, crawling juveniles). In this study, a population of the opisthobranch gastropod Haminoea zelandiae from Pauatahanui Inlet, New Zealand, was found to exhibit poecilogony. Further, differences in development, hatching times and proportion of hatchlings that were veligers or juveniles were examined for egg masses in two temperature regimes in the laboratory: cool (15-17 °C), and warm (21-23 °C). Hatching proportions were also examined for egg masses collected from the field (where temperatures ranged from 21-23 °C) for varying lengths of time (1 d, 5 d, and 10 d post-spawning). Hatchlings from egg masses in warmer temperatures developed faster and hatched earlier than those in cool temperatures. In the laboratory, egg masses in warm conditions hatched a greater proportion of post-metamorphic juveniles (45.4%) compared to egg masses in cool conditions (24.6%) Further, egg masses that had been in the field 10 d before hatching (i.e., more days at warmer temperatures) exhibited a greater proportion of post-metamorphic juveniles (67.9%) than those that were collected after only 1 d in the field (25.1%). Together these results suggest that temperature may have an important role in mediating dispersal strategies in this poecilogonous species.  相似文献

Water molds that cause the disease saprolegniasis have been implicated in widespread mortality of amphibian embryos. However, because of the limitations of traditional identification methods, water mold species involved in die-offs or utilized in ecological studies often remain unidentified or identified only as Saprolegnia ferax. Furthermore, water mold taxonomy requires revision, so very distinct organisms may all be called S. ferax. Recent DNA-based studies indicate that the diversity of water molds infecting amphibian embryos is significantly higher than what was previously known, but these studies rely on culture methods, which may be biased towards taxa that grow best under laboratory conditions. In this study, total embryo-associated DNA was extracted from 3 amphibian species in a pond in central Washington, USA. The internal transcribed spacer (ITS) region of DNA was amplified with primers capable of amplifying a broad array of eukaryotic microorgansisms, and was used to construct clone libraries. Individual clones were sequenced and relationships among newly recovered sequences and previously studied taxa were analyzed using phylogenetics. These methods recovered several new taxa in association with amphibian embryos. Samples grouped into 11 distinct phylotypes with ITS sequence differences ranging from 4 to 28%. The water mold communities recovered differed among Rana cascadae, Bufo boreas, and Pseudacris regilla egg masses. Furthermore, the diversity of water molds increased as egg masses aged, and members comprising this diversity changed over time.  相似文献

The reproductive biology of Fidiobia dominica Evans (Hymenoptera: Platygastridae) was studied in the laboratory (25.6 +/- 1 degrees C) using host eggs of Diaprepes abbreviatus L. (Coleoptera: Curculionidae). F. dominica readily parasitized D. abbreviatus eggs on both host plant and wax paper substrates. The number of egg masses parasitized and the number of offspring produced were higher when females were offered more than one host egg mass but did not differ when either two or three egg masses were offered. Female parasitoids that were provided with host eggs and a honey food source lived significantly longer than those that were not provided a food source; however, they did not parasitize more D. abbreviatus eggs. Oviposition occurred in host eggs from 0 to 7 d old, and host mortality was relatively consistent for eggs 0-5 d old and lower for eggs 6-7 d old. Successful parasitoid emergence seldom occurred after host eggs were 4 d old, and by 7 d, no adults successfully emerged. Developmental time from egg to adult was 19.3 +/- 0.2 d for males, significantly more rapid than the females (20.4 +/- 0.1 d). The mean longevity of adult females was 8.0 +/- 0.4 d, with a mean oviposition period of 2.7 +/- 0.3 d; males survived 8.1 +/- 0.4 d. The demographic parameters including intrinsic rate of increase (r(m)), generation time (T), and net reproduction (R(o)) were 0.142/d, 22 d, and 22.4 female eggs/d, respectively.  相似文献

Dynamics of egg production of the tapeworm Diphyllobothrium dendriticum has been estimated experimentally in nestlings of the herring gull Larus argentatus per day and per reproductive period. The number of eggs in strobiles has been estimated for the maturation period. Mean egg production of the tapeworm per day (10.43 +/- 2.72 mln) is two orders of magnitude as high as the maximum number of eggs in a mature strobile (0.206 +/- 0.007). It is proposed to estimate coefficient of reproduction intensity as the ratio of egg output per unit of time and maximum egg number in a mature strobile.  相似文献

Convoluta roscoffensis collected from the natural habitat in reproductive condition laid egg capsules for up to 16 weeks under laboratory conditions. However, both the number of capsules laid and number of embryos per capsule decreased with time in culture. When animals were maintained in an alternating light and dark regime, the capsules were laid during the dark period. Animals ceased to deposit egg capsules within five days of incubation in the photosynthetic inhibitor, DCMU. This suggests that metabolism of the algal symbionts of C. roscoffensis contributes to egg production.  相似文献

Factors affecting the orientation, reproduction, and sex ratio of the egg parasitoid Ooencyrtus kuvanae Howard were examined. Adult females were attracted to airborne volatiles from the egg mass and accessory gland of the primary host, the gypsy moth Lymantria dispar L. Visual cues also affected host selection. Background colors against which egg masses were placed affected oviposition preference. In the absence of egg masses, color variation did not affect wasp behavior. Light is required for parasitism by O. kuvanae. The age and density of both the host and parasitoid affected wasp reproduction and sex ratios. Older egg masses issued relatively fewer wasps and higher proportions of males than did young egg masses. Likewise, wasp reproduction and the proportion of females declined with wasp age. Larger egg masses produced more wasps and lower proportions of males than did smaller egg masses. The number of offspring per female, and the proportion of female offspring, were inversely related to wasp density. Implications to biological control of the gypsy moth and parasitoid ecology are discussed.  相似文献

Abstract. Gelatinous egg masses are common in a number of animal phyla. However, they are virtually unknown in marine bivalves, with structures that could be thought of as gelatinous egg masses being reported for only five species. We describe the gelatinous egg mass and intracapsular development in the tropical lucinid Phacoides pectinatus. The embryos developed within individual capsules embedded in a large flimsy, spherical mass. Swimming veligers hatch at 198 μm shell length. They did not feed, settled within several days of hatching, and metamorphosis was completed within 2 weeks of hatching. Gelatinous egg masses might be detected in members of more lucinid species if studies of development included field or in vivo observations of reproduction in addition to producing embryos by stripping the gonads.  相似文献

The reproductive phenology of two Iberian populations of midwife toads Alytes obstetricans and A cistemasii were studied in 1988 and 1989 in two different habitats where chmatological conditions restricted reproductive activity to a few months per year All the observed mating activity of A cisternasii occurred in less than a week while A obstetricans ma tings were observed for over a month A cisternasii matmg activity was explosive and correlated with rainfall No clear trends were observed m the timing of egg mass releases with male size, egg mass size, and proportion of egg mass hatched In both populations males did not complete more than one cycle of egg mass tending per season The duration of terrestnal egg development was estimated to be of 32 d for A obstetricans Alytes cistemasii showed greater flexibility on the duration of egg development, egg masses being released 23 d after the matings m 1988 and 30 d after in 1989 This flexibility is an example of the adaptive significance of terrestrial paternal care when water availabihty is unpredictable  相似文献

Egg masses of Nezara viridula (L.) are commonly parasitized by Trissolcus basalis (Woll.), and we investigated the role of size of egg masses on parasitization by T. basalis. Sentinel egg masses were exposed to parasitism in the field for 6–7 days, when they were collected for evaluation of parasitoid emergence. We recorded the number of eggs per egg mass, the number of emerged hosts, and the number of empty and parasitized eggs. We calculated the proportion of attacked host egg masses (DE), the proportion of parasitized eggs per attacked egg mass (PE), and total parasitism (PI). The total number of egg masses exposed to parasitism was 330. The minimum, mean, and maximum egg mass sizes were 25, 75.2, and 111, respectively. DE and PE varied widely between different fields, and they were independent of egg mass size. In 14.2% of all parasitized egg masses, we found simultaneous emergence of T. basalis and N. viridula independently of host egg mass size. PE exhibited low variability compared with PI and DE, which were linearly related. PI and DE values from other field studies are consistent with the linear relationship, suggesting that PI is mostly related to the proportion of the DE. This also suggests that total parasitism is independent of egg mass size, of possible differences in plant species, and T. basalis density and strains.  相似文献

Early life-history stages of the nudibranch Janolus fuscus (superfamily Arminoidea) were reared in the laboratory and collected from the field to document embryology, larval morphology, and pre-and post-metamorphic growth. This species produces Type B egg masses containing an average of 23,063 viable embryos. The spiral holoblastic cleavage pattern resembled that of other nudibranchs, with early cell divisions occurring roughly at 4-h intervals at 11-13 °C. Embryos progress through a gastrula stage with cellular extensions covering the blastopore and a trochophore-like stage before hatching as veliger larvae after 10-18 days. Veligers ranged in size from 125-153.8 μm at hatching, with size being positively correlated with the duration of encapsulated development. After a 36-41-day larval period, some veligers ceased growing at 266 μm and showed signs of approaching competence. Four larvae settled and two metamorphosed into 280-μm juveniles on the bryozoan prey of the adults, Bugula pacifica, 46 and 54 days post-hatching. Average growth of pre-ovipositional (<19 mm) J. fuscus was more rapid (8.79% per day) than growth of ovipositional individuals (3.52% per day). Growth continued to a maximum of 57 mm in the laboratory, with an estimated total lifespan of about 5 months. These data, which agree with concurrent field observations, suggest a subannual life cycle.  相似文献

The reproduction and survival of Podisus rostralis was evaluated under different feeding intervals using Tenebrio molitor larvae as food and a continuous supply of bean slices. There was a linear relationship between the pre-oviposition period and the periods without feeding. Females fed at intervals of 1-4 days laid their first egg mass 8-10 days after emergence, while those fed every 8 and 16 days did so after 13.6 and 20 days respectively. The oviposition period, number of egg masses, total number of eggs, number of eggs/egg mass, total number of nymphs and rate of egg consumption/day showed negative linear correlations with the increase in feeding interval. The longevity of P. rostralis was not affected by feeding interval (P> 0.05). The present results suggest a type of trade-off, where longevity is maintained at the expense of reproduction. This behaviour or attribute of P. rostralis can contribute to its establishment and survival in an environment of fluctuating food supply.  相似文献

Abstract. Galerucella nymphaeae L. , a chrysomelid that feeds on Lythrum spp., water lily and water chestnut, is closely related to two European species that were recently introduced into North America for biological control of L.salicaria , purple loosestrife. To develop a paradigm for continuously rearing these and other univoltine chrysomelids, we conducted field and laboratory studies on G. nymphaeae's development , reproduction and diapause.
A high incidence of reproduction without diapause occurred when fourth instars, pupae, and adults (held as pairs) experienced very long daylengths (LD18:6 h), i.e. longer than those G.nymphaeae encounters in nature. Under similar photoperiodic conditions, adults maintained in groups showed a significantly higher rate of reproductive diapause than those held as pairs. Females laid three to seven egg masses/week, and the size of egg masses varied between nine and nineteen eggs. During their reproductive lifetimes, individual females showed a highly significant propensity to lay a consistent number of eggs/egg mass.
After diapausing under short daylengths and low temperature (LD 10:14h, 5C), adults transferred to long days (LD 18:6h at 21C) had high rates of diapause termination and postdiapause oviposition. In contrast, those transferred to short daylengths (LD 10:14h at 21C) had low rates of reproduction.
Laboratory-derived heat-degree models accurately predicted egg and pupal, but not larval, development in the field. In nature, most females in the summer generation entered reproductive diapause without ovipositing; a small proportion of females that emerged relatively early (by mid June) oviposited before entering diapause. The overwintering population consists of adults from the first-generation and a small number from the second generation.  相似文献
