This research was conducted to assess the water quality and the contamination of heavy metals in water, sediment, fish, and frogs, as well as bioaccumulation factors (BAFs) in fish and frogs around the gold mine area. The water samples were analyzed for water quality (temperature, pH, and dissolved oxygen). The samples were analyzed for heavy metals by inductively coupled plasma optical emission spectrometry. The water quality was within the standard. The concentrations of heavy metals, including As, Cr, Cd, Pb, Ni, Zn, Fe, Mn, and Cu, in water and sediment samples were measured. Three species of fish were collected: Rasbora tornieri, Brachydanio albolineata, and Systomus rubripinnis. The mean heavy metal concentrations of fish were as follows: Fe>Zn>Mn>Cr>Ni>Cu>As>Pb>Cd. The As, Cr, and Pb concentrations in all the fish species exceeded the standard levels. Five species of frogs were collected: Kaloula pulchra, Microhyla heymonsi, Fejervarya limnocharis, Hoplobatrachus rugulosus, and Microhyla pulchra. The mean heavy metal concentrations of frog were as follows: Fe>Cu>Mn>Zn>Cr>Ni>Pb>Cd>As. The Cr, Cd, and Cu concentrations exceeded the standard levels. The BAFs in fish were in order of Cr>Zn>Ni>Cu>Pb>Fe>Cd>As. The BAFs in frogs were Cr>Zn>Ni>Cu>Fe>Pb>Cd>As. The accumulation of heavy metals was higher in the sediment than in the water. Many aquatic organisms take up heavy metals directly from the environment around the gold mine.  相似文献

以桃儿七种子诱导的无菌苗为材料,研究了外源激素对芽诱导、增殖和生根的影响,建立了桃儿七离体培养再生体系。结果表明:芽诱导阶段,采用3.0 mg·L-1 6-BA+0.5 mg·L-1 GA3激素组合,出芽率最高可达85-71%,缩短出芽时间30~40 d,在添加3.0 mg·L-1 6-BA+0.2 mg·L-1 IAA的MS培养基上利于增殖,增殖速度快,增殖系数1.63。以WPM+1.5 mg·L-1 IAA+0.5 mg·L-1 NAA的培养基培养30 d 后,生根率可达60.1%。  相似文献

The synthesis, structure and spectroscopic properties of novel palladium(II) chloro complexes with a series of (aminoalkyloxymethyl)dimethylphosphine oxides (AOPO) are reported. The complexes with general formula PdCl2(N,N′-AOPO2) were obtained by the reaction of PdCl2(CH3CN)2 with the ligands in dry ethanol. The crystal structure of the trans-bis[2-(dimethylphosphinoylmethoxy-1,1-dimethylethylamine)]palladium(II) dichloride has been determined from single-crystal X-ray diffraction data. The compound crystallizes in monoclinic crystal system with P21/n space group. The square-planar coordination sphere of palladium consists of two N atoms from two aminoalkyldimethylphosphine ligands and two Cl atoms in trans-arrangement. The AOPO ligand has monodentate coordination through the NH2 group. The Pd-N and Pd-Cl distances are 2.0610(14) and 2.3225(4) Å, respectively. The preparation of complexes with a composition PdCl2(AOPO)2 in chloroform solution are also reported.  相似文献

基于16属65种琵甲(含新记述7属)的防御腺特征, 探讨了琵甲族(鞘翅目: 拟步甲科)的属级系统发育关系。通过对这些种的防御腺着生位置、 形状、 长度、 宽度、 囊体间距、 囊壁厚度、 囊壁花纹、 囊壁褶皱等特征进行解剖测量和分析, 归纳出族、 属级特征。利用SPSS19.0和Hennig86(1.5版)两个软件对选定特征进行聚类分析和系统进化分析, 得出琵甲族16属的进化关系为:Prosodes>Blaptogonia>Tagonoides>Thaumatoblaps>Caenoblaps>Agnaptoria>Asidoblaps>Coelocnemodes>Dila>Gnaptor>Blaps>Pseudognaptorina> Nalepa>Belousovia>Gnaptorina>Itagonia。基于防御腺形态学数据的琵甲族系统发育分析结果将琵甲族16个属清楚地分开, 表明其在分类上具有重要价值。  相似文献

The objective of this study was to evaluate the human NCI-N87 cell line as a model for gastric permeability drug studies under pH conditions of the stomach. The optimal conditions that led NCI-N87 cells to form a typical differentiated gastric epithelial barrier were a seeding density of 2.5 × 105 cells/cm2 on porous inserts and growth in serum-complemented RPMI-1640 medium until 18–27 days post-confluency. The resulting cell monolayers showed moderately high transepithelial electrical resistance (TEER) values of about 500 Ω cm2, cells of polygonal morphology expressing E-cadherin and ZO-1 proteins at their contact surfaces, and production of mucus clusters. The monolayers withstood apical pH of 7.4 down to 3.0 with the basal pH fixed at 7.4. The apparent permeability coefficients (Papp) of model compounds were evaluated in the apical-to-basolateral and basolateral-to-apical directions under different pH gradients. The monolayers were impermeable to the integrity marker Lucifer Yellow (low Papp of 0.3–1.1 × 10−6 cm/s). The furosemide Papp (0.4–1.5 × 10−5 cm/s) were slightly dependent on pH but remained moderate. The caffeine Papp (4.2–5.0 × 10−5 cm/s) were higher and insensitive to pH changes. The NCI-N87 cell line provides a useful in vitro tool to assess gastric drug permeability and absorption under physiologic conditions prevailing in the human stomach.  相似文献

Summary The meiotic behaviour of chromosomes was studied in four inbred lines and their F 1 hybrids of P. typhoides. The inbred lines showed a decrease in mean chiasma frequency when compared with the population plants, but differed from one another in their mean chiasma frequencies. Together with the decrease in mean chiasma frequency the inbreds showed variation in mean chiasma frequencies. The inbred lines showed a number of meiotic abnormalities such as extra chromosomes, extra fragments, desynapsis, persistent nucleoli and differential condensation of chromosomes. The F 1 hybrids of these inbreds exhibited heterosis for chiasma frequency. All the F 1's had mean chiasma frequencies higher than the means of the respective participating parents. The F 1's, however, differed in the degree of heterosis exhibited. In the F 1 hybrids, the variation in mean chiasma frequency, both between plants and between PMC's within plants, was significantly lower than that of the inbred lines. The effect of environment was studied in the inbred lines and their F 1 hybrids. The mean chiasma frequencies of the inbred lines were significantly lower, and the variation in mean chiasma frequencies was greater, in the stress season. The mean chiasma frequencies of F 1's did not show any significant differences between the two seasons. The F 1's exhibited less variation in mean chiasma frequency than the inbred lines, showing that F 1's were developmentally more stable. The F 1's did not show any meiotic abnormalities in either season.  相似文献

An inverse and unusual relationship was found between preferred temperature and acclimation temperature in the bullseye puffer, Sphoeroides annulatus. The final preferendum temperature (PT) was 26.8 °C. The critical thermal maxima (CTMax) were 37.7, 38.8, 40.0, 40.8 and 41.3 °C where the temperatures of acclimation were 19, 22, 25, 28 and 31 °C±1 °C, respectively, and the endpoint of CTMax was loss of the righting response. The acclimation response ratio presented an interval of 0.22-0.38; these values are in agreement with results for other subtropical and tropical fishes. The temperature significantly affected the oxygen consumption of bullseye puffer juveniles. The oxygen consumption rate (OCR) increased significantly with an increment in the temperature from 19 to 31 °C. The range of the temperature coefficient Q10 in bullseye puffer individuals was lowest between 25 and 28 °C, at 1.37. The optimal temperature for growth was 26 °C. The results of this study will be useful for optimizing the culture of bullseye puffer juveniles in controlled conditions.  相似文献

Summary The genome structure of the temperateRhizobium meliloti phage and the attachment site of this phage on the host chromosome were examined by genetic means. The heat-sensitive mutants used in 2 and 3 point crosses gave a linear chromosome map. There was no evidence for map circularity. The immunity region has a distal position on the phage chromosome. The functional grouping of the used 23 phage mutants was made byin vivo andin vitro complementation tests and 20 cistrons were obtained. The cistrons, near to the immunity region, were identified as early genes, the remaining ones as morphogenetic cistrons. The latter inin vitro complementation tests gave two complementing groups, presumably as head and tail donors. The attachment site of the prophage on the host chromosome was localized by pulse mutagen treatments in synchronously replicating cultures. Summary The rhizines of the lichens, Parmelia caperata, Parmelia trichotera and Lobaria pulmonaria were studied with the Stereoscan scanning electronmicroscope and in ultrathin sections with the transmission electronmicroscope. The rhizines are composed of fungal hyphae.The fungal hyphae in the rhizines of the thallus of Parmelia caperata and in the cilia at the thallus border of Parmelia trichotera are connected by a glue-like substance. The ends of the Parmelia caperata rhizines are flattened and enlarged. With these footlike rhizines the thalli are in good connection with the substratum. The cilia at the thallus border of Parmelia trichotera have a tip by which the thallus is fixed on bark or rocks. The cell walls of the fungi hyphae in the Parmelia caperata rhizines and in the Parmelia trichotera cilia are 150–400 nm thick.The rhizines of Lobaria pulmonaria consist of fungi hyphae which are interlaced without a gluey substance. The thallus of Lobaria pulmonaria is connected to the substratum through the tips of single hyphae. The hyphae walls of Lobaria pulmonaria are 800–1800 nm thick.  相似文献

To measure genetic relatedness between populations, for breeding purposes, we analyzed 170 birds from six crossbred populations of three pure lines of White Leghorn chickens, using 14 microsatellite markers. All the microsatellites were polymorphic, with 2–6 alleles. The mean number of alleles per locus was 3.21. The effective number of alleles varied from 1.14 to 3.94. The observed heterozygosity varied from 0.133 to 1.00, with a mean of 0.748. The F IS values were mostly negative, with an average of ?0.345. The mean F ST value was 0.056. The Nm values ranged from 1.91 to 42.17. The highest genetic identity was observed between IWI × IWK and IWK × IWI. The relation between any two groups of crosses was more than 85%. The results suggest that the crossbred populations were very closely related.  相似文献

The ultrastructure of the marine predatory flagellate Metromonas simplex Larsen et Patterson was studied. The cell is surrounded by a low-contrast fibrous layer composed of thin hairs covered by a thin bilayer membrane and an outer layer of thin short fibers. The plasmalemma lies under these layers. The predator captures whole cells of the prey, usually bodonids or chrysomonads. The cytostome as a cell pocket is undetectable. The long flagellum bears very thin mastigonemes (hairs) with lengths of 0.8–1.0 μm; the short flagellum is naked and reduced in length. The transitional zone lacks spirals or other additional elements. The transversal plate is elevated on the cell surface. The flagellar root system is very simple and has one microtubular band which originates near the kinetosomes. The latter are parallel to each other and interconnected by fibrous bridges. The vesicular nucleus, Golgi apparatus, and endoplasmic reticulum are of typical structures. The oval mitochondria of 0.6–2.5 μm contain lamellar cristae. The cylindrical extrusomes (trichocysts) found in the cytoplasm have lengths of 1.0–1.4 μm and diameters of 0.12–0.08 μm. The trichocysts have a wheel-shaped structure with 13 spokes visible in cross-sections. The contractile vacuole is absent. The similarity that M. simplex shares with Metopion fluens Larsen et Patterson, cryothecomonads, and other predatory flagellates is discussed.  相似文献

以疏花卫矛(Euonymus laxiflorus)、丝棉木(E.maackii)和卫矛(E.alatus)3种卫矛属植物为试验材料,测定叶片转色期叶色参数和相关生理生化指标,探讨各项指标的变化规律和内在联系,为优良色叶植物的筛选提供依据。结果表明:(1)转色期,疏花卫矛的明亮度参数L~*和色素参数b~*(黄/蓝)呈上升趋势,色素参数a~*(红/绿)变化不大;丝棉木的L~*和b~*呈先上升后下降的单峰曲线,a~*呈上升趋势;卫矛的L~*和b~*变化不大,a~*呈上升趋势。(2)3种植物的叶绿素含量在转色期呈明显下降趋势;疏花卫矛的花色素苷相对含量、花色素苷/叶绿素的值较为平稳,类胡萝卜素/叶绿素的值呈上升趋势;丝棉木和卫矛的花色素苷相对含量、花色素苷/叶绿素的值均呈上升趋势,类胡萝卜素/叶绿素的值保持平稳。(3)3种植物苯丙氨酸解氨酶(PAL)活性呈下降趋势,查尔酮异构酶(CHI)活性呈先上升后下降的单峰曲线;卫矛的过氧化物酶(POD)和多酚氧化酶(PPO)活性保持平稳,疏花卫矛和丝棉木的POD活性呈上升趋势,疏花卫矛的PPO活性保持平稳,丝棉木的PPO活性呈下降趋势。(4)疏花卫矛和卫矛的可溶性糖和淀粉的质量分数呈先上升后下降的单峰曲线,丝棉木的可溶性糖和淀粉的质量分数呈上升趋势。(5)相关性分析显示,疏花卫矛呈现黄色主要是因为叶绿素的分解,丝棉木和卫矛呈现红色是因为花色素苷的合成,可溶性糖、淀粉、CHI对花色素苷的合成有一定作用,POD对叶片呈现红色有促进作用,PAL和PPO活性对花色素苷的合成无显著影响。  相似文献

The xmnIRM genes from Xanthomonas manihotis 7AS1 have been cloned and expressed in Escherichia coli. The nucleotide (nt) sequences of both genes were determined. The XmnI methyltransferase (MTase)-encoding gene is 1861 by in length and codes for 620 amino acids (aa) (68660 Da). The restriction endonuclease (ENase)-encoding gene is 959 by long and therefore codes for a 319-aa protein (35275 Da). The two genes are aligned tail to tail and they overlap at their respective stop codons. About 4 × 104 units/g wet cell paste of R·XmnI was obtained following IPTG induction in a suitable E. coli host. The xmnIR gene is expressed from the T7 promoter. M·XmnI probably modifies the first A in the sequence, GAA(N)4TTC. The xmnIR and M genes contain regions of conserved similarity and probably evolved from a common ancestor. M·XmnI is loosely related to M·EcoRI. The XmnI R-M system and the type-I R-M systems probably derived from a common ancestor.  相似文献

The galvanotactic response of human granulocytes was investigated theoretically and experimentally. The basic results are: (i) The granulocytes move towards the anode. (ii) The directed movement has been quantified by two different polar order parameters-the McCutcheon index and the average of cos . (iii) The polar order parameters are a function of the applied electric field (= dose-response curve). (iv) The inverse of the galvanotactic constant of migrating cells (analogous to the Michaelis-Menten constant) has a value of-0.2±0.03 V/mm. (v) The galvanotactic response of granulocytes is a non-cooperative process with a cooperativity coefficient of 1±0.2. (vi) The galvanotactic constant is a function of pH. (vii) The protein essential for the galvanotactic response is very likely a G-protein.  相似文献

The hydrozoan family Aglaopheniidae (Cnidaria) is widespread worldwide and contains some of the most easily recognizable hydroids because of their large colony size and characteristic microscopic structure. The systematics of the group has, however, been controversial and dedicated molecular analyses are lacking. We therefore analysed existing and new 16S rRNA sequences of Aglaopheniidae, in a total of 98 16S sequences corresponding to 25 putative species (25 nominal and three undescribed) from seven genera. The monophyly of the subfamilies Gymnangiinae and Aglaopheniinae, and tribes Aglaopheniini and Cladocarpini were not verified with 16S sequence data. The genera Gymnangium and Aglaophenia can only be considered valid if both Gymnangium gracicaule and Aglaophenia latecarinata are removed from their respective genera. The phenotypically similar Cladocarpus and Streptocaulus are probably monophyletic and clearly distinct genetically. The genus Lytocarpia may be polyphyletic. The nominal species Aglaophenia pluma, Aglaophenia tubiformis, and Aglaophenia octodonta are probably conspecific, as are also the species Aglaophenia acacia and Aglaophenia elongata. The 16S data revealed the existence of two potentially unnamed species of Aglaophenia respectively from the Azores and Madeira. The phylogeographical structure of the taxa with the greatest representation of haplotypes from the north‐east Atlantic and Mediterranean, revealed the influence of Mediterranean waters in Madeira and the Azores, and gene flow between deep waters of the Mediterranean and Atlantic. The last glaciations in Europe may have caused genetic bottlenecks but also high intraspecific haplotype diversity. An endoxylanase gene, xynA, was cloned from Bacillus pumilus ARA and expressed in Escherichia coli. The open reading frame of the xynA gene was 687 bp encoding a signal peptide and a mature xylanase with a molecular mass of 23 kDa. The enzyme was categorized as a glycosyl hydrolase family 11 member based on the sequence analysis of the putative catalytic domain. The recombinant XynA (Bpu XynA) was purified to homogeneity by Ni–NTA and ion exchange chromatography on DEAE–Sepharose FF. The enzyme exhibited highest activity at pH 6.6 and 50°C. The purified Bpu XynA was stable for at least 2 h at 45°C, and retained over 50% residual activity after being incubated at 60°C for 1 h. The activity of the xylanase was not significantly affected by metal ions and EDTA. The K m and K cat /K m of Bpu XynA for oat-spelt xylan were 5.53 mg/ml and 10.14 ml/mg s at 50°C and pH 6.6. The main product of hydrolysis by Bpu XynA was xylooligosaccharide. The results revealed that the consumption of grass xylan by B. pumilus ARA depended on the synergistic reactions of Bpu XynA and Bpu arabinosidase, and that a typical GH11 xylanase e.g. Tla XynA had capability to remove the side chain of xylan. The properties Bpu XynA make it promising for application in the production of Bifidobacterium growth-promoting factors and in feed industry.  相似文献

Summary The intergeneric amphiploid Hordeum chilense × Aegilops squarrosa has been synthesized. The amphiploid plants have the expected chromosome number of 28. The average meiotic chromosome pairing was 12.48 bivalents + 3.04 univalents. The morphology of the amphiploid resembles that of the Aegilops parent. Nucleoli from both H. chilense and A. squarrosa are expressed in the amphiploid. Neither chromosome instability nor homoeologous pairing was found. The amphiploid is fertile and vigorous.  相似文献

A novel enzyme, which was named Nα-benzyloxycarbonyl amino acid urethane hydrolase, was purified from a cell-free extract of Streptococcus faecalis R ATCC 8043, using Nα-benzyloxycarbonyl glycine as substrate. The enzyme was purified 1300-fold with an activity yield of 8%. The purified enzyme was homogeneous by disc electrophoresis. The molecular weight of the native enzyme is about 220,000 by gel filtration, and a molecular weight of 32,000 was determined for the reduced and denatured enzyme by gel electrophoresis in sodium dodecyl sulfate. The isoelectric point was 4.48. The enzyme was inhibited by p-chloromercuribenzoate. The presence of divalent cations (i.e., Co2+ or Zn2+) is essential for its activity.  相似文献

The final thermal preference FTP) range in parthenogenetic females of cladoceran Daphnia magna was assessed by “acute” and “chronic” methods. The first method included 4-month acclimation to different temperatures in the range of 14.2 ± 0.7 to 27.1 ± 0.3°C; the “chronic” method was characterized by long-term acclimation to +20°C. Two ranges of FTP were found for D. magna, 13.3–15.4°C and 20.2–26.2°C. The thermal preference of daphnids and the temperature of acclimation were correspondingly linearly. The range of FTP was independent of the season. The searching activity of D. magna rose in April, when the FTP range increased, and the FTP was less pronounced.  相似文献
