The effect of molybdenum concentration upon the growth of Anabaenacylindrica is studied. Within the range of molybdenum concentrationthat it was found possible to achieve by the methods described,the element does not affect growth on ammonium chloride, butit is required for healthy growth on nitrate or gaseous nitrogen,the optimal concentrations being about 0·075 and 0·20p.p.m., respectively. Molybdenum appears to be unnecessary fornitrate uptake.  相似文献

To investigate the nutritional value of the diatom Cyclotella cryptica Reimann, Lewin, and Guillard as an alternative feed for the use in the aquaculture industry, the heterotrophic growth characteristics, total fatty acids, and the resultant fatty acid profile of the microalga were studied when cultivated with sodium nitrate, ammonium chloride, or urea. All three nitrogen sources supported growth under heterotrophic conditions, and their uptake affected the pH of the cultivation medium, even when buffered. The use of sodium nitrate or urea resulted in a significant increase in the pH of the cultivation media, whereas the use of ammonium chloride caused a minor decrease in the pH of the cultivation media. The maximum specific growth rate was highest when urea and ammonium chloride were supplied at a low concentration; however, the total fatty acid content was not significantly affected (P = 0.101) by the nitrogen source when supplied at 10.7 mM nitrogen. The total fatty acid content and fatty acid profile of C. cryptica was more affected by the growth phase (predominately influenced by the initial nitrogen concentration) than by the source of nitrogen.  相似文献

Plant acid-base homeostasis is achieved when the mmoles of hydroxylions produced in the plant equal the mmoles of protons. Reductionof nitrate to ammonia is the major source of hydroxyl ions whereasammonium uptake-assimilation and the metabolism of neutral sugarsto organic acids are the primary sources of protons. Soybean[Glycine max (L.) Merr plants were grown hydroponically on mediumsupplemented with 3·0 mol m–3 nitrogen providedas various combinations of KNO3 and NH4NO3 Plant growth consumedessentially all available nitrogen in each case; however, onlyin flasks supplemented with approximately 1·8 minolesof KNO3 plus 0·6 mmole of NH4NO3 was the pH of the mediumunchanged. Thus, for every mmole of nitrogen assimilated, approximately0·6 mmole of dissociable protons must have been producedby the conversion of neutral sugars to carboxylic acids. Also,it was shown that a plant obtaining all of its nitrogen fromnitrate must neutralize or excrete approximately 0·5mmole of hydroxyl ion d–1. Conversely, the plant derivingall of its nitrogen from dinitrogen must excrete or neutralizeat least 0·8 mmole of hydrogen ion d–1 whereasthe plant deriving all of its nitrogen from aminonium must excreteor neutralize approximately 2·1 mmoles of hydrogen iond–1. Nevertheless, plants grown on medium supplementedwith 2·4 mol m–1 nitrate plus 0·6 mol m–3ammonium did not achieve a higher growth rate than plants grownon 3·0 mol m–3 nitrate. The effect of inorganic nitrogen source on population growth of Euglena mutabilis, an acidophillic benthic protozoa colonizing on the sediment of acid mine drainage, was investigated. Sodium nitrate, ammonium chloride, and ammonium sulfate were tested as nitrogen sources. The population density of E. mutabilis at equilibrium density cultivated in ammonium chloride‐ and ammonium sulfate‐containing media was 9–11 times higher than that in sodium nitrate‐containing medium at the optimal salt medium concentration. The population growth of E. mutabilis in ammonium sulfate‐containing medium was rapid and reached half of the equilibrium density after ca. 228 h, which was ca. 77 h earlier than that in ammonium chloride‐containing medium. Culture medium with ammonium sulfate as the nitrogen source achieved the highest maximum population density and the fastest growth rate among the three nitrogen salts used as nitrogen sources.  相似文献

Merr.] is inhibited by continuous growth in the presenceof 4· mol m–3 KNO3 The presence of 4·0 molm–3 ‘starter nitrate’ for 3-6 d during noduledevelopment, however, subsequently stimulates nodule dry weightaccumulation and nitrogenase activity. These stimulations occureven though 4· mol m–3 nitrate temporarily delaysnodule development, i.e. the late steps of nodule developmentare reversibly inhibited by a short-term exposure to 4·0mol m–3 nitrate. On the other hand, treatment with 4·0mol m–3 nitrate in excess of 14 d significantly reducesnodule dry weight Thus, extended growth in the presence of 4·0mol m–3 KNO3 seems to block both early and late stepsof nodule development. Nodulation of hydroponically grown soybeansis also inhibited by continuous growth in the presence of 2·0mol m–3 (NH4)2SO4 This inhibition is not caused by acidityof the growth medium. On the other hand, nodule development6 d after inoculation with Rhizoblum japonicum is not delayedby a 7-d exposure to 2·0 mol m–3 (NH4)2SO4 butis partially inhibited by a prolonged exposure to (NH4)2SO4Because repression of nodulation by 4·0 mol m–3KNO3 is more severe than that by 2·0 mol m–3 (NH4)2SO4and because ammonium taken up by the soybean plant is not activelyoxidized to nitrate, it is suggested that there are at leasttwo mechanisms by which nitrate utilization represses noduleformation in soybean. In vitro carotenoid pigment production in callus cultures of Calendula officinalis L. was investigated using two basal media, semi-solid versus liquid media and varied concentrations of sucrose, ammonium, and nitrate nitrogen. Of the two explants that were evaluated, floret explants were best for callus induction using Murashige and Skoog (MS) medium supplemented with 2.0 mg l−1 2,4-dichlorophenoxyacetic acid under complete darkness. Carotenoid pigment induction was significantly augmented when the sucrose concentration was increased. Low sucrose concentrations in the culture medium deferred the onset of pigment induction and reduced the overall levels of carotenoid pigments produced. The highest amount of carotenoid pigments was observed when the callus was grown on the MS medium without ammonium nitrogen. The quantity of carotenoids was slightly elevated in cultures grown on semi-solid medium than those grown in liquid medium. In vitro carotenoid production was optimized by modifying the concentration of ammonium nitrogen to nitrate nitrogen in the culture medium and enhancing the sucrose concentration.  相似文献

Phototrophic growth of the moderate halotolerant Rhodobacter capsulatus strain E1F1 in media containing up to 0.3 M NaCl was dependent on the nitrogen source used. In these media, increased growth rates and growth levels were observed in the presence of reduced nitrogen sources such as ammonium and amino acids. When the medium contained an oxidized nitrogen source (dinitrogen or nitrate), increases in salinity severely inhibited phototrophic growth. However, the addition of glycine betaine promoted halotolerance and allowed the cells to grow in 0.2 M NaCl. Inhibition of diazotrophic growth by salinity was due to a decrease in nitrogenase activity which was no longer synthesized and reversibly inactivated, both effects being alleviated by the addition of glycine betaine. In R. capsulatus E1F1, inhibition of cell growth in nitrate by salt was due to a rapid inhibition of nitrate uptake, which led to a long-term decrease in nitrate reductase activity, probably caused by repression of the enzyme. Addition of glycine betaine immediately restored nitrate uptake, but the recovery of nitrate reductase activity required several hours. Neither ammonium uptake nor ammonium assimilation through the glutamine synthetase-glutamate synthase pathway was affected by NaCl.  相似文献

Summary The effect of ammonium, nitrate, and organic nitrogen on growth and sporulation of 18 Aspergilli was examined in a chemically defined medium in surface culture under controlled conditions. All three forms of nitrogen were metabolized by all the Aspergilli tested. Ammonium nitrogen was not good both for growth and fruiting. This was due to the sharp fall in the pH level which resulted due to the rapid utilization of anions of the ammonium nitrogen than cations. The effect of adding succinic acid in the medium containing ammonium nitrogen has been discussed.Good growth of Aspergilli in media containing nitrate nitrogen with the accompanying rise in the pH of the medium showed that these species are capable of reducing nitrate nitrogen to the level of ammonia. The role of succinic acid in the utilization of nitrate nitrogen was investigated. All fungi accomplished good growth on a medium containing asparagine.  相似文献

The nutritional requirements for ovule formation in Nigella saliva L. were investigated by growing excised pistils on defined media. Pistils grown on a medium containing the minerals of Murashige and Skoog produced significantly more ovules than on a medium containing the minerals of Bilderback. When the nitrogen, sulfate, and phosphate of the Bilderback medium were adjusted to levels comparable to those of the Murashige and Skoog medium, a similar number of ovules was formed. The effect of different forms and concentrations of nitrogen on ovule formation and pistil growth was investigated. High concentrations of nitrate (40 mil) favored pistil growth and ovule formation, but comparable levels of ammonium were toxic. When ammonium at concentrations above 10 mM was added to nitrate media, ovule formation was inhibited. A medium containing low concentrations of ammonium (10 mM) and nitrate (5 mM) supported more ovule formation and pistil growth in young pistils than a low-nitrate (5 mM) medium without ammonium. However, ovule formation on a medium containing 10 mM ammonium and 5 mM nitrate was significantly less than on a medium containing only 15 mM nitrate. Low concentrations of organic nitrogen in the form of α-alanine (1 mM) and γ-aminobutyric acid (5 mM) supported ovule formation and pistil growth similar to a high nitrate medium.  相似文献

Monascus rubiginosus ATCC 16367 was cultivated aerobically in media containing ammonium and nitrate as nitrogen source. The pH of the medium was adjusted at different times, the pH of the nitrate medium being lowered to the pH of the ammonium medium and the pH of the ammonium medium raised to that of the nitrate. More phosphate was taken up on the nitrate medium, but potassium uptake did not start until 24h. On the ammonium medium, both were taken up in parallel from the beginning, but the amount of phosphate taken up never reached the same level as on nitrate medium. When the pH was adjusted, the uptakes changed, especially on the ammonium medium where a great increase in phosphate uptake was observed. More conidia were formed on the nitrate medium and more pigment on the ammonium medium. When the pH of either media was adjusted, the development of conidia and pigment production changed to that of the other control medium where the pH evolved normally in the direction of the change, regardless of the source of nitrogen. The reasons for the development of conidia on nitrate medium or where the pH is high, and the production of pigment on ammonium medium or at low pH is discussed.  相似文献

The effects of concentration of amino acids, nitrate, and ammonium on the growth and taxol production in cultures of cell line TY-21 of Taxus yunnanensis were investigated. Addition of 20 different amino acids each at 15–20 mg l–1 to B5 medium significantly improved callus growth but inhibited taxol formation in the cultures. The optimum nitrate concentration was 20–30 mM for both growth and taxol production. Ammonium greatly suppressed growth but strongly promoted taxol formation in the cells when it was the sole inorganic nitrogen in the medium. Culturing the suspension cells in nitrate-containing medium for 15 days and then in a medium in which ammonium was the sole inorganic nitrogen for 7 days increased taxol yield by 104%, reaching up to 28.1 mg l–1.  相似文献

Nitrate and ammonium were shown to alter the growth ofRhodotorula glutinis var.salinaria in saline and non-saline media. In saline medium in which ammonium was the sole nitrogen source, ammonium inhibited growth in the presence of molybdate ions. Detailed comparisons of the growth in saline and non-saline media when nitrate was supplemented in the presence of molybdate ions showed that differences in utilizability of purine bases of nucleic acid were responsible for the differences in growth,i.e. adenine increased the growth in such saline medium, but decreased it in non-saline medium. There was not such a specific requirement for adenine in saline medium in the presence of molybdate ions when nitrate was substituted for ammonium as the sole nitrogen source. It was suggested that adenine might provide the necessary skeleton of nucleic acid for serving nitrate reduction in saline medium.  相似文献

Carboxymethylcellulase (CM-cellulase) and ß-glucosidaseactivities were induced in cultures of Botryodiplodia theobromae,Pat. Both growth of the fungus and CM-cellulase production werebetter with sodium nitrate as nitrogen source than with eitherammonium nitrate or ammonium chloride. Growth, per unit nitrogensupplied, was greater with glutamic acid and aspartic acid asnitrogen sources than with sodium nitrate; this was probablybecause the fungus utilized the carbon of these amino-acids.Most ß-glucosidase activity was formed with ammoniumchloride or ammonium nitrate as nitrogen source. Glucose inhibited the activity of ß-glucosidase markedly.Low glucose concentration (c. 0.003 M) stimulated CM-cellulaseactivity but concentrations above 0.05 M inhibited. The formationof both enzymic activities was repressed in the presence ofglucose. ß-glucosidase activity was more thermolabilethan that of CM-cellulase.  相似文献

Embryogenic callus from Citrus sinensis (L.) Osbeck cv. Hamlin was cultured for 28 days on 20 media arranged in a 5×2×2 factorial varying in the ratio of nitrate to ammonium nitrogen, total inorganic nitrogen, and benzyladenine. Fresh weight increase of callus and final medium pH were significantly affected by total inorganic nitrogen and the ratio of nitrate to ammonium. The nitrate to ammonium ratio accounted for 55% of the variation in the fresh weight increase of the callus and 93% of the variation in the final medium pH. Bean plants (Phaseolus vulgaris L. cv. Fardenlosa Shiny) werelabelled with carbon-11 via their first trifoliate leaves when3-weeks-old and the transient inhibitions of translocation causedby the application of ammonium chloride solutions (10 mol m–3)to a peeled region of stem were studied. At pH 6·5 theammonium was without effect. At pH 11·0 even a briefapplication inhibited translocation for many minutes, whilelonger applications inhibited translocation for considerablylonger. Solutions of 10 mol m–3 sodium chloride were withouteffect at either pH. At pH 6·5 ammonium chloride solution contains predominantlyammonium ions (NH4+) and at pH 11·0 predominantly dissolvedammonia gas (NH3). Hence we conclude that phloem transport withinbean stems is inhibited by dissolved ammonia gas but not ammoniumions. We determined the quantum requirements for growth (1/?μ) and fatty acid (FA) biosynthesis (1/?FA) in the marine diatom, Phaeodactylum tricornutum, grown in nutrient replete conditions with nitrate or ammonium as nitrogen sources, and under nitrogen limitation, achieved by transferring cells into nitrogen free medium or by inhibiting nitrate assimilation with tungstate. A treatment in which tungstate was supplemented to cells grown with ammonium was also included. In nutrient replete conditions, cells grew exponentially and possessed virtually identical 1/?μ of 40–44 mol photons · mol C?1. In parallel, 1/?FA varied between 380 and 409 mol photons · mol C?1 in the presence of nitrate, but declined to 348 mol photons · mol C?1 with ammonium and to 250 mol photons · mol C?1 with ammonium plus tungstate, indicating an increase in the efficiency of FA biosynthesis relative to cells grown on nitrate of 8% and 35%, respectively. While the molecular mechanism for this effect remains poorly understood, the results unambiguously reveal that cells grown on ammonium are able to direct more reductant to lipids. This analysis suggests that when cells are grown with a reduced nitrogen source, fatty acid biosynthesis can effectively become a sink for excess absorbed light, compensating for the absence of energetically demanding nitrate assimilation reactions. Our data further suggest that optimal lipid production efficiency is achieved when cells are in exponential growth, when nitrate assimilation is inhibited, and ammonium is the sole nitrogen source.  相似文献

Microalgae cultures are receiving attention because of increasing biotechnological and biomedical production of active biomolecules. We evaluated various fertilizer-based culture media to scale up production of the marine microalga Phaeodactylum tricornutum for production of exocellular polysaccharides (EPS), soluble proteins, and cellular superoxide dismutase (SOD). The standard source of sodium nitrate was the same as that used in the synthetic f/2 culture medium and ammonium nitrate, urea, ammonium sulfate, and calcium nitrate as alternative sources of nitrogen. The maximum production of EPS was achieved in microalgae cells grown in the culture media containing 63 and 23% nitrogen from ammonium sulfate, and also in microalgae cells grown in the culture media containing 3% nitrogen from ammonium nitrate. The maximum production of cellular SOD was achieved in microalgae cells grown in the culture media containing 35 and 26% nitrogen from ammonium sulfate, and in the culture media containing 17% nitrogen from urea. The results suggest that it is possible to use a source of nitrogen, other than sodium nitrate, to scale up growth of P. tricornutum for production of EPS and SOD at reduced costs.  相似文献

The role of nitrate, ammonium, and culture medium pH on shoot organogenesis in Nicotiana tabacum zz100 leaf discs was examined. The nitrogen composition of a basal liquid shoot induction medium (SIM) containing 39.4 mM and 20.6 mM was altered whilst maintaining the overall ionic balance with Na(+) and Cl(-) ions. Omission of total nitrogen and nitrate, but not ammonium, from SIM prevented the initiation and formation of shoots. When nitrate was used as the sole source of nitrogen, a high frequency of explants initiated and produced leafy shoots. However, the numbers of shoots produced were significantly fewer than the control SIM. Buffering nitrate-only media with the organic acid 2[N-morpholino]ethanesulphonic acid (MES) could not compensate for the omission of ammonium. Ammonium used as the sole source of nitrogen appeared to have a negative effect on explant growth and morphogenesis, with a significant lowering of media pH. Buffering ammonium-only media with MES stabilized pH and allowed a low frequency of explants to initiate shoot meristems. However, no further differentiation into leafy shoots was observed. The amount of available nitrogen appears to be less important than the ratio between nitrate and ammonium. Shoot formation was achieved with a wide range of ratios, but media containing 40 mM nitrate and 20 mM ammonium (70:30) produced the greatest number of shoots per explant. Results from this study indicate a synergistic effect between ammonium and nitrate on shoot organogenesis independent of culture medium pH.  相似文献

Soybean cell suspension cultures grew on defined media with ammonium as the sole nitrogen source if Krebs cycle acids were added. Satisfactory growth was obtained with ammonium salts of citrate, malate, fumarate, or succinate, when compared with the regular medium containing nitrate and ammonium. Little or no growth occurred when ammonium salts of shikimate, tartrate, acetate, carbonate, or sulfate were used. The cells also grew well with l-glutamine as nitrogen source. The specific activities of glutamine synthetase and isocitrate dehydrogenase (nicotinamide adenine dinucleotide phosphate) were lower than in cells grown on a nitrate medium, but ammonium enhanced the activity of glutamate dehydrogenase. Cells of soybean, wheat, and flax have been cultured for an extended period on the ammonium citrate medium.  相似文献

选用5种不同的白葡萄酒活性干酵母,以硫酸铵、氯化铵、硝酸铵、尿素、酵母粉等5种物质为氮源,观察其生长量并称量菌体重,以此分析其对氮源利用情况及不同氮源对酵母生长的影响。研究表明:供试菌系在不同氮源中均能生长。不同氮源对酵母的生长速度和生长量有不同影响;不同酵母菌种对不同氮源的利用也有差异。在以酵母粉为氮源的培养基中生长最好。在实验提供的氮源中,酵母粉为供试菌最优氮源,其次是硫酸铵,氯化铵与硫酸铵基本相当,而硝酸铵最差。8#菌种对各种氮源的利用能力相对较强。17#菌种对各种氮源的利用能力最弱。  相似文献
