Adenosine deaminase and 5'-nucleotidase activities as well as chemiluminescence emission were measured in peritoneal macrophages of Syrian hamsters in the growth process of tumours with different grade of malignancy. The adenosine deaminase activity was established to decrease, while the 5'nucleotidase activity--to increase in macrophages after the subcutaneous injection of tumour cells with high level of malignancy as compared with these values in normal cells. This is accompanied by a decrease of the macrophage chemiluminescence during the whole experimental period. At the same time adenosine deaminase and 5'-nucleotidase activities as well as chemiluminescence emission in peritoneal macrophages of hamsters treated with low-malignancy cells do not differ from these values in the control group.  相似文献

Cytochemistry was used to measure the activity of succinate dehydrogenase (SOD), lactate dehydrogenase (LDH) and glucose-6-phosphate dehydrogenase (G-6-PDH) in rat peritoneal macrophages under the action of the endogenous immunostimulant tuftcin (tre-lys-pro-arg) during phagocytosis of latex particles and at rest. Tuftcin did not affect the activity of the study enzymes in non-phagocytic cells. Elevation of the peptide concentration to 0.25 micrograms/ml and higher in phagocytic macrophages activated G-6-PDH and lowered the activity of LDH. Tuftcin did not alter the activity of SOD in phagocytic macrophages.  相似文献

In experiments on rabbits immunized with soluble protein antigen immune reactions were found to be accompanied by the production of lipofuscin in macrophages. This process was the morphological manifestation of the digestion of antigen by macrophages which thus acquired the ability to migrate in the organ and to form lymphoid follicules in the medullary zone of lymph nodes. The newly formed follicules seem to be the basis of pronounced specific immune response. In staphylococcal bacteriemia the phagocytic activity of macrophages was delayed, thus causing disturbances in lipofuscin production; as a result, the subsequent phases of immune response also lagged somewhat behind in time.  相似文献

The present study investigated the effect of staphylococcal enterotoxin type A (SEA) and endotoxin Serratia marcescens (LPS) on the phagocytosis and killing of Staphylococcus aureus by mouse peritoneal macrophages. Two hours after enterotoxin intraperitoneal injection phagocytic and bactericidal activity were depressed. 24 hours later there was increased functional activity of macrophages by SEA and LPS, apart. But when two toxins were administered together (LPS four hours later enterotoxin) marked inhibition of bacterial killing was observed. When peritoneal macrophages were treated in vitro for 24 hours with the same toxins they were also markedly suppressed in bactericidal activity.  相似文献

A complex quantitative study of changes of proliferative processes in lymphatic vessels on the one hand and changes of the level of corticosteroids in the peripheral blood in staphylococcal intoxication and infection in the same rats on the other hand has been carried out. In the work H3-thymidine has been used. The comparison of the obtained morphological data with the dynamics of the change in the corticosteroid level shows that the decay of lymphocytes, the devastating of light centers and the cortical substance of lymph nodes, an abrupt lowering of the amount of DNA-synthesizing and mitotically dividing cells take place in the period associated with a release of a great amount of corticosteroids into the blood. The drop of their level to control values goes in parallel with increasing the amount of DNA-synthesizing and mitotically dividing cells. The obtained data suggest that the leading role in those changes of the lympoid tissue which had been described by a number of authors in infectional processes is played by corticosteroids.  相似文献

A study was made by histochemical methods of the activity of the enzymatic systems of macrophages from normal rabbits and those immunized with staphylococcus alpha-toxoid per se and infected with the strains of staphylococcus--producers of alpha-toxin or leukocydin. Immunization of rabbits was accompanied by a reduction in macrophages of the activity of the group of lysosomal enzymes and by an increase in the activity of the redox enzymes. In infection of "immune" macrophages with the living culture of the alpha-toxigenic strains the mentioned changes were more pronounced; no such changes were found after the infection with the leukocydin-active strain. The data obtained suggested that the lysosomal enzymes played a definite role in the process of phagocytosis.  相似文献

Bacterial cell wall component-induced tolerance represents an important protective mechanism during microbial infection.Tolerance induced by the TLR2 agonist bacterial lipoprotein (BLP) has been shown to attenuate the inflammatory response,and simultaneously to augment antimicrobial function,thereby conferring its protection against microbial sepsis.However,the underlying mechanism by which BLP tolerance augments bactericidal activity has not been fully elucidated.Here,we reported that the induction of BLP tolerance in murine macrophages upregulated the expression of Rab20,a membrane trafficking regulator,at both the mRNA and protein levels upon bacterial infection.The knockdown of Rab20 with Rab20 specific siRNA(siRab20) did not affect the phagocytosis of Escherichia coli (E.coli),but substantially impaired the intracellular killing of the ingested E.coli in BLP-tolerized macrophages.Furthermore,Rab20 was associated with GFP-E.coli containing phagosomes,and BLP tolerization resulted in the enhanced maturation of GFP-E.coli-containing phagosomes associated with Rab20 and strong lysosomal acidification.The knockdown of Rab20 substantially diminished lysosome acidification and disturbed the fusion of GFP-E.coli containing phagosomes with lysosomes in BLP-tolerized macrophages.These results demonstrate that Rab20 plays a critical role in BLP tolerization-induced augmentation of bactericidal activity via promoting phagosome maturation and the fusion of bacteria containing phagosomes with lysosomes.  相似文献

Acidification of phagosomes has been proposed to have a key role in the microbicidal function of phagocytes. Here, we show that in alveolar macrophages the cystic fibrosis transmembrane conductance regulator Cl- channel (CFTR) participates in phagosomal pH control and has bacterial killing capacity. Alveolar macrophages from Cftr-/- mice retained the ability to phagocytose and generate an oxidative burst, but exhibited defective killing of internalized bacteria. Lysosomes from CFTR-null macrophages failed to acidify, although they retained normal fusogenic capacity with nascent phagosomes. We hypothesize that CFTR contributes to lysosomal acidification and that in its absence phagolysosomes acidify poorly, thus providing an environment conducive to bacterial replication.  相似文献

The effects of adenosine were examined on the isolated perfused heart of the frog Rana ridibunda. Adenosine produced negative chronotropic and inotropic effects on frog ventricle in a concentration-dependent manner. The effects of adenosine on cardiac metabolism were also investigated by measuring the tissue content of adenine nucleotides, lactate, pyruvate, adenosine and inorganic phosphate, during adenosine perfusion. Adenosine had no effect on the tissue content of metabolites. No net synthesis of adenine nucleotides was observed during perfusion with increasing concentrations of adenosine. Lactate output from the heart decreased significantly with adenosine perfusion. Correlation of adenosine effects on cardiac muscle with the effects of hypoxia are discussed.  相似文献

【目的】解析氮源浓度对米根霉木糖代谢途径及产物的影响,提高木糖利用率。【方法】以木糖为碳源,考察不同氮源浓度下米根霉的生物量、有机酸积累量、木糖代谢关键酶(木糖还原酶、葡萄糖-6-磷酸脱氢酶)活力以及胞内还原力(NADH/NAD+、NADPH/NADP+)的差异。【结果】富氮条件下(2.4 g/L尿素),木糖代谢速率达2.03 g/(L·h),木糖还原酶、葡萄糖-6-磷酸脱氢酶的活力以及胞内还原力较高,生物量达18.01g/L,几乎不积累有机酸;限氮条件下(0.15 g/L尿素),木糖还原酶、葡萄糖-6-磷酸脱氢酶的活力以及胞内还原力水平降低,生物量仅4.02 g/L,富马酸积累量为6.55 g/L,残余木糖量较高;氮源浓度为0.6 g/L时,木糖还原酶和葡萄糖-6-磷酸脱氢酶的活力以及NADPH/NADP+处于前二者之间,此时生物量9.11 g/L,有机酸积累量较大,其中富马酸为12.28 g/L。【结论】充足的氮源可使米根霉通过木糖代谢关键酶与胞内还原力的协同效应强化木糖代谢活力,通过优化氮源浓度后,米根霉可积累更多有机酸。  相似文献

The study of the phosphatase activity of chick fibroblasts in the process of staphylococcal infection by the electron-histochemical method has revealed the presence of correlation between the degree of cytoplasmic destruction in fibroblasts and the level of acid phosphatase activity. Chronic inflammation of the lung, as a consequence of persistent bacterial infections by several opportunistic pathogens represents the main cause of mortality and morbidity in cystic fibrosis (CF) patients. Mechanisms leading to increased susceptibility to bacterial infections in CF are not completely known, although the involvement of cystic fibrosis transmembrane conductance regulator (CFTR) in microbicidal functions of macrophages is emerging. Tissue macrophages differentiate in situ from infiltrating monocytes, additionally, mature macrophages from different tissues, although having a number of common activities, exhibit variation in some molecular and cellular functions. In order to highlight possible intrinsic macrophage defects due to CFTR dysfunction, we have focused our attention on in vitro differentiated macrophages from human peripheral blood monocytes. Here we report on the contribution of CFTR in the bactericidal activity against Pseudomonas aeruginosa of monocyte derived human macrophages. At first, by real time PCR, immunofluorescence and patch clamp recordings we demonstrated that CFTR is expressed and is mainly localized to surface plasma membranes of human monocyte derived macrophages (MDM) where it acts as a cAMP-dependent chloride channel. Next, we evaluated the bactericidal activity of P. aeruginosa infected macrophages from healthy donors and CF patients by antibiotic protection assays. Our results demonstrate that control and CF macrophages do not differ in the phagocytic activity when infected with P. aeruginosa. Rather, although a reduction of intracellular live bacteria was detected in both non-CF and CF cells, the percentage of surviving bacteria was significantly higher in CF cells. These findings further support the role of CFTR in the fundamental functions of innate immune cells including eradication of bacterial infections by macrophages.  相似文献

Like neutrophils, phagocytizing macrophages undergo a "respiratory burst" in which significant quantities of oxygen are drawn into the cell. The consumed oxygen is not used in oxidative phosphorylation but, rather, in the formation of superoxide anion (O2) and H2O2. These oxygen metabolites and the products of their interaction, in particular hydroxyl radical (OH), have been implicated in the killing of ingested bacteria by neutrophils. Their role in macrophage microbicidal activity has not been fully defined. However, activated macrophages, which mediate increased resistance to infection in vivo, have a markedly increased capacity to generate O2 and H2O2 in vitro when stimulated by phagocytosis or surface perturbation. The enhanced capacity of activated macrophages to generate highly reactive oxygen metabolites during phagocytosis could contribute to the improved microbicidal and tumoricidal activity of these cells.  相似文献

Terrilytin and immobilized terrilytin enhance the activity and intensity of phagocytosis and increase the concentration of lysozyme in nonimmunized animals. Both preparations increase the production of antibodies to staphylococcal alpha-hemolysin, the titers of beta-lysins, the activity and intensity of the phagocytosis of bacterial cells by peripheral blood leukocytes in animals immunized with staphylococcal toxoid and challenged with live staphylococcal culture. In healthy animals terrilytin and immobilized terrilytin induce an increase in total proteolytic activity and in the activity of alpha-1-antitrypsin and alpha-2-macroglobulin, decreased as the result of staphylococcal infection.  相似文献
