Growth and development of plants are known to be affected by exposure to red and blue light. Mechanisms by which light quality influences gene expression in maize (Zea mays L.) embryos have not been explored. Maize kernels can be cultured in vitro allowing experimental manipulation of environmental factors during seed development. We used the in vitro kernel culture system to investigate the response of developing maize seeds, which normally develop without exposure to light, to controlled light quality. Kernels grown under red light accumulated more dry weight than those grown in darkness, whereas kernels grown under blue light accumulated less. Reciprocal color shift experiments showed that light quality during the first week in culture had more influence on kernel weight than during the subsequent three weeks in culture. Soluble sugars were higher in both light treatments than in darkness. Blue-grown kernels had higher amino acid and lower lipid levels than red-or dark-grown kernels. Embryo morphology was markedly affected by red light, under which the upper shoot axis was longer than under blue light or in darkness. Embryo morphology was influenced by light quality during the later stages of development rather than the first week. We suggest, based on these results, that gene expression in the embryo and endosperm of developing maize seeds is sensitive to light quality, and the mechanism and time dependence of this effect warrant further study. In vitro maize kernel culture affords a convenient system for such light quality experiments.  相似文献

通过盆栽试验,研究了内生真菌拟茎点霉B3(Phomopsis liquidambari)及苍术(Atractylodes lancea)粉联合施用对连作花生根际土壤微生物区系、酶活性及有效态微量元素(Mo、B、DTPA-Fe、Zn、Cu、Mn)含量的影响。结果表明:内生真菌B3和苍术粉复合处理比内生真菌B3处理的荚果和秸秆产量分别增加10.28%和14.11%,内生真菌B3处理与正常施肥相比显著提高了根瘤数量、荚果和秸秆产重,各处理组与正常施肥对照相比分枝数和根长无显著差异。B3处理与对照相比显著提高了种子期、结荚期和成熟期根际土壤可培养细菌和放线菌数量,B3和苍术粉复合处理与对照相比显著提高种子期、花期和成熟期可培养真菌和放线菌数量;细菌DGGE指纹图谱聚类分析表明,B3和苍术粉复合处理相对于正常施肥处理,显著改变种子期、苗期、花期和成熟期花生根际土壤细菌群落结构,同时苗期、花期和结荚期的细菌条带数和香农指数也有所提高,真菌DGGE指纹图谱聚类分析表明,B3和苍术粉复合处理对真菌群落影响较大,除种子期以外的生育期真菌条带数和香农指数都有明显提高,花期真菌群落结构变化最大,相似度仅为49.6%。花生关键生育期(花期和结荚期)根际土壤脲酶和蔗糖酶活性B3处理和复合处理都显著高于正常施肥对照,促进了连作花生生态系统的物质循环和能量流动。B3和苍术粉复合处理促进了花生生长发育必需微量元素Mo、B、Fe、Zn、Mn的活化,花生叶片和籽粒中微量元素Mo、B、Fe的积累显著增加。研究结果表明,内生真菌和苍术粉联合施用能有效改善连作花生根际微生物区系,提高土壤酶活性,促进微量元素的活化和吸收,对缓解花生连作障碍具有重要意义。  相似文献

The bioactivity of powder from the leaves of Aristolochia ringens, Dalbergia saxatilis, Dioclea reflexa, Ocimum gratissimum, Chrysophyllum albidum, Moringa oleifera, Citrus limon and Newbouldia laevis was tested against the larger grain borer (LGB) in maize kernels and the effect of the powder on the nutritional composition of the kernels was determined. A total of 72 glass bottles containing 100?g maize kernels was divided into four groups. In the first group of 32 glass bottles, 5?g powder of the experimental plants was admixed uniformly with the kernels (5%?w/w). Ten 1–5-day-old LGB adults were introduced into each of the glass bottles. A second group of 32 glass bottles contains 100?g maize kernels admixed with the plant powder as described above, but LGB were not added. In the third group of four glass bottles, 100?g maize kernels received insects only, while in the fourth group of four glass bottles 100?g maize kernels were neither mixed with plant products nor infested with LGB. The treatments were arranged on worktables using completely randomised design and left for three months. The plant products-treated samples (insect free) were passed through a 30?mesh sieve to sieve out the powder and the clean kernels were analysed for proximate composition. The untreated maize kernel had a significantly (p?<?0.05) higher % of grain weight loss (17.33), grain damage % (70.96), number (122.0) of adult LGB, weight (14.46?g) of grain dust and lower % of grain germination. The adsorption and absorption of some of the plant powder by the kernels had significant effect on the kernel composition of protein, fibre and carbohydrate, but not on moisture, dry matter and fat and ash content. Control of LGB with the powder of the test plants could be used as green insecticide for the management of LGB.  相似文献

This study was conducted to explore systemic infection by the Aspergillus flavus group into corn ears via the stalk. An A. parasiticus mutant which produces norsolorinic (NOR) acid (a visible orange intermediate of the aflatoxin biosynthetic pathway) was used in field studies to monitor systemic infection of corn stalk and ear tissues. Corn hybrids resistant and susceptible to aflatoxin contamination were grown in the field and inoculated prior to tasseling by inserting A. parasiticus infested toothpicks into stalks between the 5th and 6th node below the lowest ear shoot. Beginning 2 weeks after inoculation, systemic infection by the NOR mutant was assessed weekly by collecting ear shank tissue and stalk tissue from the nodes between the infection sites and the developing ears. Ears were collected at the end of the growing season to determine the level of kernel infection by the NOR mutant. In two separate studies, the A. parasiticus NOR mutant was isolated from stalk tissues at all of node positions and ear shank tissue from several susceptible corn hybrid plants at the first harvest date 2 weeks after inoculation. The NOR mutant was also isolated from stalk and ear tissue of a resistant hybrid. The NOR mutant was only isolated from kernels of susceptible hybrids in 2003 and 2004. Infection rates of kernels in infected ears were very low (<1%). In 2005, the fungus was found in only one kernel from an ear of the resistant hybrid. The NOR mutant was not isolated from stalks, ears, or kernels from control (uninoculated) plants grown in the plots with inoculated plants. Although infection levels of corn kernels were low, systemic movement of the A. parasiticus up the stalk appears to be another possible route to infection of developing corn ears.  相似文献

The antagonistic activity of two yeast strains (Pichia anomala (E.C. Hansen) Kurtzman, strain K and Candida oleophila Montrocher, strain O) against the parasitic complex responsible for banana crown rot was evaluated. The strains were applied at three different concentrations (106, 107, 108 cfu/ml) and their efficacy tested in vivo on three separate fungi (Colletotrichum musae (Berk. & Curt.) Arx, Fusarium moniliforme Sheldon, and Cephalosporium sp.) and on a parasitic complex formed by association of these three fungi. At the concentrations used C. musae appeared to be the most pathogenic. The complex showed intermediate aggressiveness between C. musae and both other fungi.Statistically significant antagonistic effects were observed on C. musae, F. moniliforme, and the fungal complex. The highest protection level (54.4%) was observed with strain O added at 108 cfu/ml on crowns previously inoculated with the fungal complex. The level was lower when the fungi were inoculated separately.Furthermore, the antagonistic effect was strongly reinforced when strain O at 108 cfu/ml was applied 24 h before fungal complex inoculation (59.9%), as compared to its application 15 min (24.3%) or 3 h (27.3%) after fungal complex inoculation. Bananas showed increased susceptibility to the fungal complex from March to June, and this influenced the level of protection by yeast, which decreased over the same period. A strict negative correlation (R2 = 0.83) was highlighted between susceptibility of banana to crown rot and protection provided by yeast.  相似文献

The objective of this work is to evaluate the ability of some additive substances in protecting the biocontrol agent Candida oleophila (strain O) against the adverse effects of environmental factors, such as water activity (aw, 0.93 and 0.98) and relative humidity (75% and 98%). The protection obtained with various protectant substances, skimmed milk (SM), peptone, maltose, sucrose, sorbitol, lactose and polyethylene glycol was assayed under in vitro and in vivo conditions. The yeast cells with the highest level of protecting agents (1%) had higher viability than those with low protectant levels (0.1% and 0.5%). SM, sucrose and sorbitol improved significantly the C. oleophila survival on apple fruit surface by 80.8%, 42.26% and 37.27% and gave a significant protection (from 96% to 100%) against Penicillium expansum under dried conditions. The highest strain O density and efficacy was obtained with SM. Under experimental conditions reflecting practical conditions, SM applied in combination with the strain O resulted in improved biocontrol efficacy by 74.65%. Therefore, SM could be used as material substrate with the best sugar protectants during the formulation process of this antagonistic yeast for eventual pre-harvest application.  相似文献

This study was conducted to develop a bacterial glucan as an animal feed additive. A novel glucan-producing bacterium.Paenibacillus polymyxa JB115, was isolated from Korean soil. The glucan, JB115-BG, produced byP. polymyxa JB115, was confirmed by TLC to be composed of glucose only. By examining FT-IR,1H NMR, and13C NMR spectra, it was proven that JB115-BG has a β-(1→3)- and β-(1→6)-linked glucan structure. The particle size of JB115-BG was distributed in the range of 4–800 μm, with a mean value of 149.1 μm, and its molecular distribution ranged from 6.9∼3,103.7 kDa. It was also observed that 80% of the purified JB115-BG had a molecular distribution above 100 kDa. The obtained results suggest that the glucan JB115-BG can be used as an animal feed additive for the purpose of enhancing immunity.  相似文献

由禾谷镰刀菌(Fusarium graminearum, Fg)引起的赤霉病是限制小麦生产的主要病害之一。生物防治是一种高效且可持续的防治方法。【目的】从小麦种子内筛选具有抑制禾谷镰刀菌的菌株并对其生防潜力进行评估,为小麦赤霉病生防制剂的开发与利用提供菌种资源及理论支撑。【方法】采用平板对峙、孢子萌发法和无菌上清液抑菌试验筛选小麦种子内对禾谷镰刀菌具有拮抗活性的内生菌株;利用扫描电镜(scanning electron microscope, SEM)和共聚焦扫描电镜(confocal laser scanning microscope, CLSM)观察并分析无菌上清液对Fg的分生孢子形态、膜完整性以及胞内活性氧的影响;通过盆栽试验验证内生菌对小麦赤霉病的生防效果;应用二代Illumina HiSeq测序平台进行全基因组测序。【结果】从小麦种子中分离出一株高效抑制Fg生长的内生菌株JB7,其衰亡期无菌上清液对Fg孢子萌发抑制率高达85.23%。菌株JB7的无菌上清液使Fg孢子表面凹陷,破坏其细胞膜,造成核酸和蛋白质的渗漏,诱导Fg菌丝活性氧的累积,引起Fg菌丝可溶性蛋白和丙二醛含量的显著升高。该菌株具有分泌蛋白酶、纤维素酶、葡聚糖酶和产铁载体的能力。盆栽试验表明菌株JB7能显著降低小麦赤霉病的病情指数(P<0.05)。经全基因组学鉴定为甲基营养型芽孢杆菌(Bacillus methylotrophicus) JB7,该菌株基因组中含有12个抑菌功能的次级代谢产物合成基因簇。【结论】菌株JB7能抑制禾谷镰刀菌的生长,对小麦赤霉病有较强的防效,可作为生物防治小麦赤霉病的候选菌株。  相似文献

The changes in moisture content, storage mycoflora and aflatoxin B1 (AFB1) in bran from untreated or raw rice (Rr) and parboiled rice (Pbr) stored in small lots in polyethylene bags were studied at 15-day intervals up to 60 days, using five lots of each type of bran. Deterioration was more rapid with reference to all the three parameters, in Rr bran compared to Pbr bran, the former becoming completely overgrown and caked with fungi by the end of 60 days.Aspergillus flavus was the dominant fungus in Pbr bran, whereasA. candidus andTrichoderma viride were abundant in Rr bran. The frequency of incidence as well as concentration of AFB1 increased with storage time in both types of bran, but the rate of increase as well as overall concentration were much higher in Rr bran. Solarization of soil, (potting mix = coarse sand:Eucalyptus marginata fines = 1∶1) infested with 3 fungi pathogenic to gerbera (Phytophthora cryptogea, Fusarium oxysporum andRhizoctonia solani), for 3 to 4 weeks within transparent polyethylene bags controlled root rot of gerbera. Solarization for 2 weeks however, was less effective. All plants grown in the infested potting mix which had been kept in shade for 2, 3 or 4 weeks were severely attacked. Solarization of soil within plastic bags for 4 weeks also increased availability of nutrients such as NH4 +-N, PO4 and K+ in comparison to bagged soil kept in the shade for the same period.  相似文献

Cadmium has been found to accumulate in peanut (Arachis hypogaea) kernels to levels exceeding the current maximum permitted concentration in Australia of 0.1 mg kg-1. Little is known of the mechanisms of Cd uptake into kernels by cultivars of peanut, so the aims of the experiments reported here were to determine if Cd is absorbed directly through the pod wall or via the main root system, and if differences exist between cultivars in this respect. Split-pot soil and sand/nutrient solution experiments were performed with two cultivars of peanut (cv. NC7 and Streeton) known to accumulate Cd to different levels in the kernel. The growth medium was separated into pod and root zones with Cd concentrations in each zone varied. In confirmation of previous field trial results, cv. NC7 had higher concentrations of Cd in kernels, given the same Cd levels in the external medium (solution or soil). Despite total Cd uptake by cv. NC7 being similar to cv. Streeton, cv. NC7 appeared to retain more Cd in the roots and translocate less Cd to shoots. Results from both soil and sand/solution culture indicated that the dominant path of Cd uptake by peanut was via the main root system, with direct pod uptake contributing less than 5% of the total Cd in the kernel. There was little difference between cultivars in this characteristic. This indicates that unlike Ca nutrition of peanuts, agronomic techniques to manage Cd uptake will require modification of soil to the full depth of root exploration, rather than just the surface strata where pods develop. Cadmium concentrations in testa were up to an order of magnitude higher than in the kernel, indicating that blanching of kernels would be effective in reducing Cd in the marketed product. High concentrations of airborne fungal spores frequently occur from spring through fall in temperate areas of the world. Although it is generally assumed that fungi on leaf surfaces are contributors to the air spora, little data are available comparing the types of fungi found on leaf surfaces with those in the atmosphere. Air sampling was carried out with a Burkard Spore Trap located on the roof of a building on the University of Tulsa campus using standard methods. Leaf samples were aseptically collected from Ulmus americana and Quercus palustris trees on campus, placed in sterile plastic bags, and brought to the lab. For each leaf, 4 cm2 areas of both upper and lower leaf surfaces were swabbed and plated on malt extract agar with streptomycin. Cultures were incubated at room temperature for 5–7 days and then examined microscopically. Results were expressed as colony forming units (CFU)/cm2. Twenty-one fungal taxa were identified from the air samples. The most abundant taxa were Cladosporium, ascospores, basidiospores, and Alternaria; together these four spore types comprised over 90% of the yearly total. Yeasts were the most abundant fungi isolated from both leaf types. Among the mycelial fungi were Phoma species, followed by Cladosporium and Alternaria. Overall twenty genera of filamentous fungi were identified. Yeasts and Phoma are normally splash dispersed and were not identified in the Burkard air samples. However, 10 taxa isolated from leaf surfaces were registered in air samples. Crude estimates of the leaf surface area of each tree suggest that the total fungal load was approximately 5.04×108 CFU for Ulmus and 2.71×108 CFU for Quercus. Of these levels, 19% were from fungi also detected in air samples. The data suggest that some leaf-surface fungi are major contributors to the air spora.  相似文献

杓兰属(Cypripedium)植物因具有较高的观赏和药用价值而长期被过度采集,已成为濒危植物。利用菌根技术进行杓兰属植物的保护和人工栽培,需要获得其可培养的菌根真菌。该研究采用分离培养法和共生回接方法,研究了云南杓兰菌根真菌菌群组成及其共生关系。结果表明:(1)从10株云南杓兰300块毛根组织中分离获得126株内生真菌,归属为3个菌属,分别是胶膜菌属(Tulasnella)73株、伏革菌属(Corticium)36株、角担菌属(Ceratobasidium)17株。其中,胶膜菌属为优势菌群,占总菌株数量的57.94%。(2)6株供试菌株中,4株菌株可显著缩短种子的萌发过程,6株菌株对幼苗的生长有显著的促进作用。(3)从中筛选获得一株CY-18高效促生真菌,对云南杓兰种子共生萌发和幼苗共生生长有极显著的促进作用。该研究结果为更好地利用菌根技术进行杓兰属植物资源的保护与可持续利用奠定了基础。  相似文献

Peanuts grown under dryland conditions where drought stress occurred accumulated more aflatoxin before digging than peanuts grown under irrigation. Kernels became more susceptible to Aspergillus flavus and A. parasiticus invasion when the soil moisture in the pod zone approached levels at which moisture moved from the pod into the soil and the kernel moisture dropped below 31%. Isolation frequencies of these aspergilli from fresh-dug kernels were lowest in 1968 (maximum of 3%). In 1967 and 1969, maximum percentages of 100 and 74, respectively, were noted. Kernel infestation was correlated with degree of aflatoxin contamination. Dryland fresh-dug kernels contained a maximum of 35,800 parts per billion aflatoxin while a maximum of 50 parts per billion was detected in kernels from irrigated plots. In 1969 A. flavus infestation was as high as 59% in peanuts from irrigated plots; however, no aflatoxin was detected. Absence of aflatoxin in these samples is attributed to the higher kernel moisture content which reduced the aflatoxin-producing potential of A. flavus. Statistical analysis of the data revealed no significant differences in degree of fungal infestation, production levels, and grade factors between any fungicide treatments.  相似文献

The objective of this study was to determine the effect of short-term (4 days) and long-term (8 days) heat stress (35°C) on sink activity of maize (Zea mays L.) kernels. Beginning at 3 days after pollination (DAP) kernels were grown in vitro at 25°C and 24 h later were transferred to 35°C for either 4 or 8 days. Each treatment had a control that was maintained continously at 25°C. Two experiments were designed to examine the uptake and distribution of 14C among hexoses, sucrose and starch in the pedicel placento-chalazal (pedicel/p-c). endosperm, and pericarp tissues of kernels exposed to heat stress for 4 or 8 days. Kernels cultured in vitro were placed in 14C-sucrose medium either during the period of heat stress (experiment 1; 5 to 13 DAP) or immediately following heat-stress treatments (experiment 2; 10 to 22 DAP). In both experiments no significant effect of heat stress was observed on the total radioactivity accumulated in the kernels until about 17 DAP, after which heat-stressed kernels accumulated less 14C than the control. During the linear fill period, the endosperm of kernels exposed to heat stress accumulated more radioactivity associated with hexoses and sucrose and less radioactivity incorporated into starch, as compared to the control. Kernels heat stressed for 4 days showed a partial recovery in starch synthesis by 21 DAP, but to levels of only 65% of that of the control. Kernels heat stressed for 8 days did not recover. When 14C-sucrose was supplied during the heat stress period (5–13 DAP). kernels from all treatments accumulated more hexoses that sucrose in the pedicel/p-c. However, during the period following heat stress (10–22 DAP), pedicel/p-c accumulated sucrose, but only in kernels exposed to long-term heat stress. Soluble invertase activity was inhibited by both short-term and long-term heat stress, whereas the activity of insoluble invertase was affected only by long-term heat stress. These results support the hypothesis that the disruption of kernel growth and more particularly endosperm starch biosynthesis, in response to heat stress, is mainly associated with changes in carbon utilization and partitioning between the different nonstructural carbohydrates within the endosperm rather than with a limitation in carbon supply to the kernel. Therefore, the effect on sink activity does not seem to be attributable to a thermal disruption of kernel uptake of sugars, but rather it is a consequence of heat perturbation of other physiological processes such as endosperm sugar metabolism and starch biosynthesis.  相似文献

The respiration kinetics of Aspergillus oryzae on different varieties of whole wheat kernels were studied. Six wheat varieties were pretreated in two different ways. Five of the six substrates fermented similarly and independently of the pretreatment method. However, pretreatment affected fermentation of one variety of soft wheat (Apollo). T 2 1H-NMR imaging of the water inside the kernels showed a change in water binding inside the kernels when a different pretreatment strategy was used. Differences in free sugar or amino acid content or in kernel stiffness were not significant.  相似文献

白及( Bletilla striata)是兰科地生型多年生植物,也是我国传统中药材之一。利用菌根技术进行白及的保护和人工栽培,需要获得白及可培养的内生真菌。该研究以广西野生的白及根和叶为材料,采用分离培养法分离内生真菌,并结合真菌形态特征,及其核糖体的转录间隔区( ITS)序列分析,确定内生真菌的分类地位。结果表明:从2株白及植物90块组织中分离获得37株内生真菌,鉴定为15个分类单元,由9个属组成,分属于2门4纲7目8科,包括锤舌菌纲( Leotiomycetes)、座囊菌纲( Dothideomycetes)和粪壳菌纲( Sordariomy-cetes),伞菌纲( Agaricomycetes)。从根中分离获得内生真菌12种,蜡壳菌属为优势属;从叶中分离获得内生真菌3种,刺盘孢属为优势属;刺盘孢菌属( Colletotrichum)和蜡壳菌属( Sebacina)真菌的相对多度值均达到20%;4株担子菌均分布于根中,叶组织中未有分布。根组织中内生真菌的多样性指数(H=1.863)高于叶组织(1.098)。该研究结果及其所分离培养的担子菌类真菌,为更好地利用菌根技术进行白及等兰科植物资源的保护与可持续利用奠定了基础。  相似文献

Streptococcus bovis HC5 produces a broad spectrum lantibiotic (bovicin HC5), but S. bovis JB1 does not have antimicrobial activity. Preliminary experiments revealed an anomaly. When S. bovis JB1 cells were washed in stationary phase S. bovis HC5 cell-free culture supernatant, the S. bovis JB1 cells were subsequently able to inhibit hyper-ammonia producing ruminal bacteria (Clostridium sticklandii, Clostridium aminophilum and Peptostreptococcus anaerobius). Other non-bacteriocin producing S. bovis strains also had the ability to bind and transfer semi-purified bovicin HC5. Bovicin HC5 that was bound to S. bovis JB1 was much more resistant to Pronase E than cell-free bovicin HC5, but it could be inactivated if the incubation period was 24 h. Acidic NaCl treatment (100 mM, pH 2.0) liberates half of the bovicin HC5 from S. bovis HC5, but it did not prevent bovicin HC5 from binding to S. bovis JB1. Acidic NaCl liberated some bovicin HC5 from S. bovis JB1, but the decrease in activity was only 2-fold. Bovicin HC5 is a positively charged peptide, and the ability of S. bovis JB1 to bind bovicin HC5 could be inhibited by either calcium or magnesium (100 mM). Acidic NaCl-treated S. bovis JB1 cells were unable to accumulate potassium, but they were still able to bind bovicin HC5 and prevent potassium accumulation by untreated S. bovis JB1 cells. Based on these results, bovicin HC5 bound to S. bovis JB1 cells still acts as a pore-forming lantibiotic.  相似文献

Oviposition decisions and their fitness consequences for the seed parasite Sitophilus oryzae (L.) (Coleoptera: Curculionidae) were investigated. Female S. oryzae lay eggs inside seeds such as wheat [Triticum aestivum (L.)]. Because larvae develop to adult within a single seed, the resources available are determined by the behavior of the female parent and characteristics of the seed in which the egg was deposited. Females were demonstrated to lay more eggs in kernels 20 mg. Females initiated the chewing of oviposition holes in shriveled kernels but were less likely to oviposit in them. Progeny size increased with increasing seed size, but the probability of an adult emerging was not affected. Females accepted large kernels more quickly than small kernels and this contributed to increased oviposition in large kernels. The increase in the number of eggs per kernel appears to result from an increase in number of visits resulting in oviposition rather than an increase in the number of eggs laid during a visit.  相似文献

The ratio of milk-clotting activity to proteolytic activity (MC/PA) was used as an index to determine the quality of milk-clotting enzyme. Solid-state fermentation on wheat bran for 5 days at room temperature gave optimal production for enzyme by Rhizomucor miehei and R. pusillus. A ratio of wheat bran to moisture of 1:0.6 (w/v) gave best results. Adding skim milk powder to the media of R. miehei did not improve the MC/PA ratio but 4% (w/w) of the powder did improve the ratio with R. pusillus.
