Improvement in prosthetic training using intermanual transfer (the transfer of motor skills from the trained, “unaffected” hand to the untrained, “affected” hand) has been shown in previous studies. The aim of this study is to determine the influence of the inter-training interval on the magnitude of the intermanual transfer effects. This was done using a mechanistic, randomized, single-blinded pretest-posttest design. Sixty-four able-bodied, right-handed participants were randomly assigned to the Short and Long Interval Training Groups and the Short and Long Interval Control Groups. The Short and Long Interval Training Groups used a prosthesis simulator in their training program. The Short and Long Interval Control Groups executed a sham training program, that is, a dummy training program in which the same muscles were trained as with the prosthesis simulator. The Short Interval Training Group and the Short Interval Control Groups trained on consecutive days, while the Long Interval Training Group and Long Interval Control Group trained twice a week. To determine the improvement in skills, a test was administered before, immediately after, and at two points in time after the training. Training was performed with the “unaffected” arm; tests were performed with the “affected” arm. The outcome measurements were: the movement time (the time from the beginning of the movement until completion of the task); the duration of maximum hand opening, (the opening of the prosthetic hand while grasping an object); and the grip-force control (the error from the required grip-force during a tracking task). Intermanual transfer was found in movement times, but not in hand opening or grip-force control. The length of the inter-training interval did not affect the magnitude of intermanual transfer effects. No difference in the intermanual transfer effect in upper-limb prosthesis training was found for training on a daily basis as compared to training twice a week.

The radial arm water maze (RAWM) contains six swim paths (arms) extending out of an open central area, with an escape platform located at the end of one arm (the goal arm). The goal arm location remains constant for a given mouse. On day 1, mice are trained for 15 trials (spaced over 3 h), with trials alternating between visible and hidden platform. On day 2, mice are trained for 15 trials with the hidden platform. Entry into an incorrect arm is scored as an error. The RAWM has the spatial complexity and performance measurement simplicity of the dry radial arm maze combined with the rapid learning and strong motivation observed in the Morris water maze without requiring foot shock or food deprivation as motivating factors. With two sessions each day, 16 mice can be tested over 2 days.  相似文献

Background

Emotion can either facilitate or impair memory, depending on what, when and how memory is tested and whether the paradigm at hand is administered as a working memory (WM) or a long-term memory (LTM) task. Whereas emotionally arousing single stimuli are more likely to be remembered, memory for the relationship between two or more component parts (i.e., relational memory) appears to be worse in the presence of emotional stimuli, at least in some relational memory tasks. The current study investigated the effects of both valence (neutral vs. positive vs. negative) and arousal (low vs. high) in an inter-item WM binding and LTM task.

Methodology/Principal Findings

A five-pair delayed-match-to-sample (WM) task was administered. In each trial, study pairs consisted of one neutral picture and a second picture of which the emotional qualities (valence and arousal levels) were manipulated. These pairs had to be remembered across a delay interval of 10 seconds. This was followed by a probe phase in which five pairs were tested. After completion of this task, an unexpected single item LTM task as well as an LTM task for the pairs was assessed. As expected, emotional arousal impaired WM processing. This was reflected in lower accuracy for pairs consisting of high-arousal pictures compared to pairs with low-arousal pictures. A similar effect was found for the associative LTM task. However, the arousal effect was modulated by affective valence for the WM but not the LTM task; pairs with low-arousal negative pictures were not processed as well in the WM task. No significant differences were found for the single-item LTM task.

Conclusions/Significance

The present study provides additional evidence that processes during initial perception/encoding and post-encoding processes, the time interval between study and test and the interaction between valence and arousal might modulate the effects of “emotion” on associative memory.  相似文献

A single exposure to the elevated plus-maze test of anxiety reduces or abolishes the anxiolytic efficacy of benzodiazepines. The present study was designed to examine whether this phenomenon of "one-trial tolerance" resulted from a motivational deficit on trial 2. We hypothesized that whereas there is a motivational conflict on trial 1 in relation to the open arms (exploration drive X natural fear of open spaces), there is no "reason" for an animal to explore it on trial 2. A motivational conflict was introduced on trial 2 by rendering the enclosed arms of the apparatus aversive on trial 1. Thus, every time rats entered the enclosed arms, an aversive situation (light and hot air blow) was produced until they left the arm. On trial 2, rats did not receive this aversive stimulation. Chlordiazepoxide significantly enhanced the percent open arm time as well as the percent open arm entries on trial 2 in rats that had been submitted to the aversive stimulation in the enclosed arms on trial 1, but was not effective in rats which had been exposed to the apparatus in the absence of the aversive stimulation on trial 1. In addition, there was no difference in the percent open arm time and entries on trial 2 between saline-treated rats submitted to the aversive or non-aversive condition on trial 1. The aversive condition on trial 1 did not modify the number of total arm entries on trial 2, either. The results suggest that the anxiolytic effect of chlordiazepoxide in the elevated plus-maze depends on the presence of a motivational conflict situation.  相似文献

Although human gamma activity (30-80 Hz) associated with visual processing is often reported, it is not clear to what extend gamma activity can be reliably detected non-invasively from frontal areas during complex cognitive tasks such as long term memory (LTM) formation. We conducted a memory experiment composed of 35 blocks each having three parts: LTM encoding, working memory (WM) maintenance and LTM retrieval. In the LTM encoding and WM maintenance parts, participants had to respectively encode or maintain the order of three sequentially presented words. During LTM retrieval subjects had to reproduce these sequences. Using magnetoencephalography (MEG) we identified significant differences in the gamma and beta activity. Robust gamma activity (55-65 Hz) in left BA6 (supplementary motor area (SMA)/pre-SMA) was stronger during LTM rehearsal than during WM maintenance. The gamma activity was sustained throughout the 3.4 s rehearsal period during which a fixation cross was presented. Importantly, the difference in gamma band activity correlated with memory performance over subjects. Further we observed a weak gamma power difference in left BA6 during the first half of the LTM rehearsal interval larger for successfully than unsuccessfully reproduced word triplets. In the beta band, we found a power decrease in left anterior regions during LTM rehearsal compared to WM maintenance. Also this suppression of beta power correlated with memory performance over subjects. Our findings show that an extended network of brain areas, characterized by oscillatory activity in different frequency bands, supports the encoding of word sequences in LTM. Gamma band activity in BA6 possibly reflects memory processes associated with language and timing, and suppression of beta activity at left frontal sensors is likely to reflect the release of inhibition directly associated with the engagement of language functions.  相似文献

Place memory is relevant for exploration and forage behaviour. When food supply is dispersed, a win-shift has advantage over a win-stay strategy. In the Olton Octagonal Maze, the rat follows a win-shift strategy using working memory. However, in the Olton 4x4 version, the rat follows a win-stay strategy, using both working and long-term memories. It has been suggested that the neocortex is required for the resolution of tasks demanding long-term, but not for that demanding working memory alone. The role of anteromedial/posterior parietal cortex (AM/PPC) was investigated here, using a reversible lesion induced by intracerebral lidocaine infusion. Long-Evans rats were implanted with guide cannulae into the AM/PPC and trained in an Olton 4x4 maze, counting working and long-term memory errors after a delay. Then, the animals were infused with lidocaine or saline during the delay phase and tested for three days. Another series of animals, treated as before, was tested in an Olton Octagonal Maze and subjected to the same injection schedule. In the Olton 4x4 Maze, lidocaine produced a significant increase in working and long-term memory errors, compared to saline and post-lidocaine conditions. In contrast, in the Olton Octagonal Maze, lidocaine did not induce any effect on working memory errors. Thus, AM/PPC is required when both working with previous information and long-term memories are needed, but not when only working memory is required, as it happens under ethological conditions. Whenever food supply is dispersed, a win-shift strategy is preferable.  相似文献

焦虑是当今社会最普遍的精神障碍之一.药物疗法和心理疗法是焦虑症的主要治疗手段,后者因无副作用且不易反复而倍受重视.在焦虑症的心理行为疗法中,奖赏记忆缓解焦虑的模型及机制是一个重要的科学问题.对这一问题的研究将有助于理解其内在机制并开发更好的治疗焦虑症的方法.基于经典的小鼠焦虑行为评测工具——高架十字迷宫,我们首先考查了焦虑评测结合行为训练的可行性,发现小鼠在高架十字迷宫上反复评测后行为达到稳态,则可通过在开放臂末端设置食物奖赏来研究奖赏记忆对焦虑行为的影响;然后通过实验证明短期奖赏训练和无奖赏的强制开放臂暴露不能改善焦虑行为;进而确定并验证了奖赏训练改善焦虑行为的正确方式;最后在此行为模型中应用有致遗忘效应的蛋白激酶C的抑制剂白屈菜赤碱(chelerythrine),发现奖赏记忆形成被抑制伴随着焦虑行为改善的削弱.初步揭示了此行为模型涉及的细胞分子机制.  相似文献

We investigated the use of extramaze and intramaze cues on the eight-arm radial maze. The rats received daily training consisting of forced-choice visits to four baited arms, a retention interval, and the availability of all eight arms with baits available at arms that did not appear in the forced-choice phase. The radial maze was placed in a featureless octagonal enclosure to minimize the availability of extramaze cues. Intramaze cues were provided at the distal end of each arm by placing a small object in front of the food trough; unique objects were randomly sampled from a large pool of objects. The use of extramaze and intramaze cues was assessed by rotating the objects, after the retention interval, on occasional non-rewarded probes, thereby dissociating the location of extramaze and intramaze cues. The rats used extramaze rather than intramaze cues. Implications for spatial representations are discussed.  相似文献

The EMG activity of upper limb muscles during human gait has rarely been studied previously. It was examined in 20 normal volunteers in four conditions: walking on a treadmill (1) with unrestrained natural arm swing (Normal), (2) while volitionally holding the arms still (Held), (3) with the arms immobilized (Bound), and (4) with the arms swinging in phase with the ipsilateral legs, i.e. opposite-to-normal phasing (Anti-Normal). Normal arm swing involved weak rhythmical lengthening and shortening contractions of arm and shoulder muscles. Phasic muscle activity was needed to keep the unrestricted arms still during walking (Held), indicating a passive component of arm swing. An active component, possibly programmed centrally, existed as well, because some EMG signals persisted when the arms were immobilized during walking (Bound). Anti-Normal gait involved stronger EMG activity than Normal walking and was uneconomical. The present results indicate that normal arm swing has both passive and active components.  相似文献

Lesions to the hippocampal system in rats result in a profound impairment of place or locale spatial learning although other learning strategies remain unaltered. The main objective of the present study was to investigate whether the spatial knowledge preserved in the hippocampal animals can be expressed flexibly under conditions different from those of the acquisition period. Rats with neurotoxic lesions to the dorsal hippocampus and sham-operated subjects were trained to reach the goal arm in a four-arm plus-shaped maze using a constant starting arm. During the training a transparent plexiglas barrier divided the maze in two equal halves in such a way that the animals could only travel from the starting arm to the goal arm, not having access to the remaining 50% of the maze. After seven days of training, a transfer test was used in which the starting arms were the two arms from which the animals had not started during the training phase. Results indicated that the lesioned rats made significantly more errors than the control subjects. But the most interesting results revealed that the kind of error made by the lesioned animals was congruent with the turn that they had to make during the acquisition phase in order to access the goal arm (reinforced). These results suggest that when the hippocampus is damaged a preserved highly inflexible egocentric strategy is employed to solve the spatial problem.  相似文献

Memory is a complex and dynamic process that is composed of different phases. Its evolution under natural selection probably depends on a balance between fitness benefits and costs. In Drosophila, two separate forms of consolidated memory phases can be generated experimentally: anaesthesia-resistant memory (ARM) and long-term memory (LTM). In recent years, several studies have focused on the differences between these long-lasting memory types and have found that, at the functional level, ARM and LTM are antagonistic. How this functional relationship will affect their evolutionary dynamics remains unknown. We selected for flies with either improved ARM or improved LTM over several generations, and found that flies selected specifically for improvement of one consolidated memory phase show reduced performance in the other memory phase. We also found that improved LTM was linked to decreased longevity in male flies but not in females. Conversely, males with improved ARM had increased longevity. We found no correlation between either improved ARM or LTM and other phenotypic traits. This is, to our knowledge, the first evidence of a symmetrical evolutionary trade-off between two memory phases for the same learning task. Such trade-offs may have an important impact on the evolution of cognitive capacities. On a neural level, these results support the hypothesis that mechanisms underlying these forms of consolidated memory are, to some degree, antagonistic.  相似文献

Adult male wild rats and male laboratory rats, all Rattus norvegicus, were kept alone for 11 d is a residential maze. The maze had a central nest box and four arms radiating from it. Movement in the arms was automatically recorded. Three foods were supplied, one at the end of each of three arms; the fourth arm was empty. One food was usually much preferred to the othe two. Except on Days 1—3, access to the arms was for only 3h daily.The rate of visiting the arms declined during the first 3 d. The wild rats were more ‘active’ than the domestic in that they made more visits; but they spent less time in the arms. Visits were of two kinds: (a) short, usually < 1min; (b) long (> 4 min.: meals). The wild rats had shorter meals than the donestic.A 3-h period of access usually began with a bout of short visits to the arms. Each long visit to a food arm (a meal) was also typically followed by such a bout, sometimes after a period in the nest box (drinking). the first short visit after a meal by a wild rat was most often to the empty arm; but domestic rats distributed these visits evenly among the three arms in which they had not been feeding. In a given bout the second short visit by a wild rat, but not a domestic, tended to repeat the first.We interpret a bout of short visits as a patrol or re-exploration of the living space which may include sampling of all accessible foods. When a bout occured at the beginning of access to the maze arms, exploring was in ‘competition’ with eating; a bout after a meal may have represented ‘disinhibition’ of exploring.  相似文献

Concerning the problems of learning and memory there is a distinction of a short term memory (STM) and a long term memory (LTM). It is supposed that the STM is an electrical phenomenon, whereas the LTM depends on material changes. The assumed materials are RNA, proteins, lipids, amines etc. and the primary carrier of the information is DNA. But there is a discrepancy: learning-specifity is based on environmental changes, but not the structure of DNA. For the investigation of this, we trained goldfish in a shock-free task to take food from coloured cups under the influence of inhibitors of the proteinbiosynthesis. Two experiments were performed to look for evidence of episodic-like memory in rats. On each of a series of trials on an eight-arm radial maze, rats in two groups entered four open arms in Phase 1, with reward pellets on three arms and a favored reward (chocolate in Experiment 1 and cheese in Experiment 2) on the remaining arm. Phase 2 retention tests were given 30 min or 4 h after Phase 1, with all eight arms open. The four arms not entered in Phase 1 all contained reward pellets, and the three arms that contained pellets in Phase 1 were empty. In the replenish short group, the favored reward was replenished at the same location where it was found in Phase 1 at the 30 min retention interval but was absent (Experiment 1) or degraded (Experiment 2) at the 4 h retention interval. In the replenish long group, the favored reward was replenished at the 4 h retention interval but not at the 30 min retention interval. Over a number of daily trials that randomly mixed short and long delays, rats in both experiments learned to return earlier to the arm containing the favored reward at the retention interval when it was replenished than at the retention interval when it was absent or degraded. These results replicate earlier findings [Babb, S.J., Crystal, J.D., 2005, Discrimination of what, when, and where: implications for episodic-like memory in rats. Learn. Loss of arms has energetic consequences for stellate echinoderms (crinoids, ophiuroids, and asteroids). The energetic cost of losing an arm includes loss of investment, decrease in ability to obtain nutrients and allocation of nutrients to regeneration of the lost arms at a cost to other body compartments. The cost to other body compartments is low when food availability is very low or very high. The cost becomes apparent when food availability is sufficient to support production but not high enough that the cost of regeneration has no effect on production of other body compartments. Loss of investment is greater in asteroids than in crinoids and ophiuroids because of greater development of the body wall and presence of gonads and pyloric caeca in the arms. The cost of regeneration of organic matter in an arm can be estimated from the amount of organic matter present in intact arms and the cost of anabolism. Protein production is the primary cost of regeneration of an arm because of the high concentration of protein in the regenerated arm and the high anabolic cost of protein production. A major energetic cost of loss of arms that affects regeneration is decrease in food consumption. It is necessary to separate cost of decrease in consumption from cost of regeneration. Comparison of intact and regenerating individuals requires they consume the same amount of food. The cost of regeneration will also be affected by the quality of food because of the nutrient requirements for growth. Because the quantity and quality of the food ingested is not known, it is not possible to quantify the cost of regeneration in the field. Asteroids appear to be a good model for the study of regeneration in the laboratory because it is possible to control the quantity and quality of food they ingest. They are also a good model for the study of the evolutionary significance of regeneration by comparing individuals that have lost arms and are regenerating them to those that have lost arms and are not. The difference in the frequency of loss of arms of species is related to the difference in availability of food and the ability to feed that affect the capacity for re-investment in the lost arm. In a previous experiment, we showed that domestic pigs, Sus scrofa, unlike many other species, performed accurately in a spatial memory task, where visits to a previously baited food trough were rewarded (win-stay). We investigated whether pigs have a predisposition for this strategy, by comparing their performance in a radial arm maze under either win-stay (N=10) or win-shift (N=10) reward contingencies. Contrary to our earlier results, only one of the animals in the win-stay condition was able to reach the imposed criterion level of accuracy. The performances of the other win-stay pigs did not deviate from random. All pigs in the win-shift condition reached criterion by day 25 of the experiment, and performed better than expected by chance. Analysis of the types of errors made matched our a priori predictions that shift movements would occur more frequently, especially within visits to the maze. We suggest that the difference in learning rates may reflect the fact that win-stay pigs needed to use two different rules, stay between trials and shift within trials, while win-shift pigs only needed to use the shift rule. In our previous study, win-stay pigs did not experience a conflict of rules and this may have facilitated stay learning. We found evidence of a recency effect in win-shift animals and a primacy effect in the win-stay group. However, we discuss the unsuitability of these specific terms in this type of experiment, and propose an alternative interpretation of the results. Effect of BR-16A on various parameters of anxiety and transfer latency (TL) was studied in mice using elevated plus-maze. BR-16A (50-500 mg/kg) reduced the percentage of time spent in open arms and the percent preference of open arms for the first arm entry following acute as well as chronic drug administration. The total number of arm entries and the percentage of open arm entries remained unaffected. In combination with FG 7142 (10 mg/kg), BR-16A (100-500 mg/kg) further reduced the exploration of open arms. BR-16A reversed scopolamine (0.3 mg/kg)-induced delay in TL on 1st day. The reversal effect of BR-16A was enhanced by aniracetam (50 mg/kg). The data suggest anxiogenic and nootropic actions of BR-16A.  相似文献

We provide indirect evidence that six axonemal proteins here referred to as "dynein regulatory complex" (drc) are located in close proximity with the inner dynein arms I2 and I3. Subsets of drc subunits are missing from five second-site suppressors, pf2, pf3, suppf3, suppf4, and suppf5, that restore flagellar motility but not radial spoke structure of radial spoke mutants. The absence of drc components is correlated with a deficiency of all four heavy chains of inner arms I2 and I3 from axonemes of suppressors pf2, pf3, suppf3, and suppf5. Similarly, inner arm subunits actin, p28, and caltractin/centrin, or subsets of them, are deficient in pf2, pf3, and suppf5. Recombinant strains carrying one of the mutations pf2, pf3, or suppf5 and the inner arm mutation ida4 are more defective for I2 inner arm heavy chains than the parent strains. This evidence indicates that at least one subunit of the drc affects the assembly of and interacts with the inner arms I2.  相似文献

The tendency of nectarivorous birds to perform better on tasks requiring them to avoid previously rewarding locations (to win-shift) than to return to them (win-stay) has been explained as an adaptation to the depleting nature of nectar. This interpretation relies on the previously untested assumption that the win-shift tendency is not associated with food types possessing a different distribution. To test this assumption, we examined the specificity of this bias to different food types in an omnivorous honeyeater, the noisy miner (Manorina melanocephala). As predicted, we found that the win-shift bias was sensitive to foraging context, manifesting only in association with foraging for nectar, not with foraging for invertebrates.  相似文献

Central administration of neuropeptide Y (NPY) stimulates hyperphagia and hyperinsulinemia. Recent evidence has suggested that the Y1 and Y5 receptor subtypes may both mediate NPY-stimulated feeding. The present study attempts to further characterize the role of central NPY receptor subtypes involved in hyperinsulinemia. NPY and peptide analogs of NPY that selectively activated the NPY Y1 or Y5 receptor subtype induced feeding and hyperinsulinemia in satiated Long Evans rats, whereas NPY analogs that selectively activated the NPY Y2 or Y4 receptor subtype did not. To determine whether NPY-induced hyperinsulinemia is secondary to its hyperphagic effect, we compared the plasma insulin levels in the presence and absence of food after a 1-min central infusion of NPY and its analogs at 15, 60, and 120 min postinfusion. Our data suggest that selective activation of central NPY Y1 receptor subtype induced hyperinsulinemia independent of food ingestion, whereas the NPY Y5 receptor-induced hyperinsulinemia was dependent on food ingestion. Central administration of the selective Y1 receptor agonist D-Arg25 NPY eventually decreased plasma glucose levels 2 h postinfusion in Long Evans rats.  相似文献
