A product of the processing of precursor rRNA ("excess" RNA)has been indirectly found to be unmethylated in mammalian systems,but direct measurement was precluded because of its instability.The "excess" RNA of duckweeds is relatively stable allowingdirect estimation of its methylation by polyacrylamide gel electrophoresis.The "excess" RNA with an apparent molecular weight 0.5?106 wasunmethylated. A pulse labeled RNA with an apparent molecularweight of 1.2?106 (presumably from chloroplasts) was also unmethylated.Under similar conditions the presumed cytoplasmic rRNA precursors,and the mature cytoplasmic and chloroplast rRNAs were methylated. 1Present address: Department of Biological Sciences, S.U.N.Y.Binghamton, N.Y. 13901, U.S.A. We developed a polyethylene glycol (PEG)-mediated direct DNAtransfer method from intact Saccharomyces cerevisiae spheroplastsinto Arabidopsis thaliana protoplasts. To monitor the DNA transferfrom yeast to plant cells, ß-glucuronidase (GUS) reportergene in which a plant intron was inserted was used as a reporter.This intron-GUS reporter gene on a 2µm-based plasmid vectorwas not expressed in yeast transformants, while it expressedGUS activity when the plasmid DNA was introduced into plantcells. When a mixture of 1 x 108 of S. cerevisiae spheroplastsharboring the plasmid and 2 x 106 of A. thaliana protoplastswas treated with PEG and high pH-high Ca2+ solution (0.4 M mannitol,50 mM CaCl2, 50 mM glycine-NaOH pH 10.5), GUS activity was detectedin the extract of the plant cells after a three-day culture.The GUS activity was higher than that of a reconstitution experimentin which the mixture of 1 x 108 of S. cerevisiae spheroplastswhich did not carry the reporter gene, 2 x 106 of A. thalianaprotoplasts and the same amount of the reporter plasmid DNAas that contained in 1 x 108 of S. cerevisiae spheroplasts,was treated with PEG and high pH-high Ca2+ solution. Moreover,the GUS gene expression was resistant to micrococcal nucleasetreatment before and during PEG treatment. From these results,we concluded that plasmid DNA can be directly transferred fromintact yeast spheroplasts to plant protoplasts by a nuclease-resistantprocess, possibly by the cell fusion. 2Deceased on September 15, 1992.  相似文献

Gibberellin-like substances of stems and leaves from Steviarebaudiana were analyzed and gibberellin A20 was identifiedby gas chromatography-mass spectrometry. The presence of GA20in S. rebaudiana is significant for the interpretation of thegibberellin activity of steviol. It indicates that steviol,the C-13 hydroxykaurenoic acid, may function as a precursorfor C-13 hydroxy-gibberellins and not as a gibberellin analog. 1 This work was supported by National Science Foundation GrantGB 17304 to M. The total number of planktonic bacteria in the upper mixed layerof the Bering Sea during the late spring-early summer periodranged between 1 and {small tilde}4 x 106 ml–1 (biomass10–40mg C m–3). In the northern Pacific, along 47–526N,the corresponding characteristics of the bacterioplankton densityin the upper mixed water layer were: total number 1–2x 106 cells ml–1 and biomass 15–46mg C m–3Below the thermocline at 50–100 m, the density of bacterioplanktonrapidly decreased. At 300 m depth, it stabilized at 0.1–0.2x 106 cells ml–1. The integrated biomass of bacterioplanktonin the open Bering Sea ranged between 1.2 and 3.6 g C m–2(wet biomass 6–18 g m–2) Its production per dayvaried from 2 to 23 mg C m–3 days–1 in the upper0–100 m. The numerical abundance of planktonic ciliatesin this layer was estimated to be from 3 to l0 x 103 cells l–1,and in the northern Pacific from 0.4 to 4.5 x 103 l–2.Their populations were dominated by naked forms of Strombidium,Strombilidium and Tontonia. In some shelf areas, up to 40% ofthe total ciliate population was represented by the symbioticciliate Mesodinium rubrum. The data on the integrated biomassof basic groups of planktonic microheterotrophs are also presented,and their importance in the trophic relationships in pelagiccommunities of subarctic seas is discussed.  相似文献

The protein content of apices and cotyledons in Morally inducedor vegetative plants of Pharbitis nilwas examined using isoelectricfocusing. A sediment trap study was conducted at Daya Bay, South ChinaSea, to investigate the relationships between encystment andpopulation dynamics of Scrippsiella trochoidea from December1999 to January 2001. A dense bloom of S. trochoidea occurredduring the study period from August to September 2000, withthe maximum cell number of 3.18 x 104 cells mL–1.Two morphotypes of cysts, one with a thick calcareous wall (calcifiedcyst) and another without the obvious calcareous cover (non-calcifiedcyst), were observed during this investigation. The morphologicaland excystment characteristics of these two cyst types werestudied as well. Mass encystments of S. trochoidea, with themaximum of 3.05 x 105 cysts m–2 d–1for calcified cyst, and 1.54 x 107 cysts m–2 d–1for non-calcified cyst, coincided with the maximum abundanceof the vegetative cells. Encystment caused the transfer of atotal of 2.24–4.49 x 108 cells m–2 vegetativecells from the water column to the sea bottom during the bloomand resulted in a considerable loss of the bloom population.High assemblages of cysts of S. trochoidea were detected inthe surface sediments as well. This rich ‘seed bed’in the surface sediments caused by the high efficiency of encystmentafter blooms acting as a benthic reservoir for future vegetativepopulation, together with the short dormant period (15–26days) and high germination rate (50–90%), may explainthe repeated occurrence of S. trochoidea blooms in Daya Bay.  相似文献

A monospecific bloom of the coccolithophorid Emiliania huxleyi(Lohmann) Hay & Mohler (Prymnesiophyceae) was detected offRio de La Plata at 36°S and 54°W in November 1989. Thecell densities observed were up to 6x105 cells I–1. Thisis the first record of a bloom of E.huxleyi in the area.  相似文献

Direct somatic embryogenesis can be obtained from epidermaland cortical cells in roots from in vitro Cichorium plantlets.The first embryogenic cells are seen after six days of culturein darkness, at 35 °C, in a liquid medium supplemented withNAA (1 x 10–7 M), 6-dimethylallyl-amino-purine (2·5x 10–6 M), sucrose (0.03 M) and glutamine (1·7x 10–3 M). Embryogenic cells undergo first a linear andthen a globular segmentation, with increasing cytoplasmic density.These cells and young embryoids show aniline blue fluorescence.SEM allows the same microglobular pattern to be seen on thesurface of young embryoids and on young microspores of Cichoriumused as controls. Five inbred strains (129/J, BALB/cByJ, C3HeB/FeJ, C57BI/6J andDBA/2J) were examined with two-bottle (48 h) preference ratiotesting across concentrations of sodium saccharin (3 x 10–4M, 10–3 M, 3 x 10–3 M and 10–2 M), d-phenylalanine(10–3 M, 10–2 M and 10–1 M), and l-glutamine(10–2 M, 3 x 10–2 M, 10–1 M and 3 x 10–1M). Three consistent groupings of strains were observed acrosssubstances and concentrations:
  1. C57BI/6J (preference at low andhigh concentrations);
  2. BALB/cByJ and C3HeB/FeJ (preferenceat high concentrations);
  3. 129/J and DBA/2J (preference at highconcentration for sodiumsaccharin and indifference to d-phenylalanineand l-glutamine).
If a single locus (presumably dpa or Sac) determines these phenotypes,there are likely to be three alleles. If two independent loci(presumably dpa and Sac) determine these phenotypes, an allelicassignment of Sacb/dpa+s for the C57BI/6J strain, Sacb/dpa–sfor the BALB/cByJ and C3HeB/FeJ strains, and either Sacd/dpa+sor Sacd/dpa–s for the 129/J and DBA/2J strains is suggested.Chem. The human Na+-sulfate cotransporter (hNaSi-1) belongs to the SLC13 gene family, which also includes the high-affinity Na+-sulfate cotransporter (hSUT-1) and the Na+-dicarboxylate cotransporters (NaDC). In this study, the location and functional role of the N-glycosylation site of hNaSi-1 were studied using antifusion protein antibodies. Polyclonal antibodies against a glutathione S-transferase fusion protein containing a 65-amino acid peptide of hNaSi-1 (GST-Si65) were raised in rabbits, purified, and then used in Western blotting and immunofluorescence experiments. The antibodies recognized native NaSi-1 proteins in pig and rat brush-border membrane vesicles as well as the recombinant proteins expressed in Xenopus oocytes. Wild-type hNaSi-1 and two N-glycosylation site mutant proteins, N591Y and N591A, were functionally expressed and studied in Xenopus oocytes. The apparent mass of N591Y was not affected by treatment with peptide-N-glycosylase F, in contrast to the mass of wild-type hNaSi-1, which was reduced by up to 15 kDa, indicating that Asn591 is the N-glycosylation site. Although the cell surface abundance of the two glycosylation site mutants, N591Y and N591A, was greater than that of wild-type hNaSi-1, both mutants had greatly reduced Vmax, with no change in Km. Loliun perenne L. (cv.S. 23) was grown on vermiculite in winterin a heated greenhouse for 8 weeks under factorial combinationsof two potassium regimes (nominally 6 parts/106 and 156 parts/106in Hewitt's solution) and three densities of artificially supplementedvisible radiation flux (36.1, 7.3, and 2.2 W m–2). Growthand potassium uptake were studied through the calculation ofvarious growth functions from fitted curves. There was little effect of potassium treatment but the experimentalmaterial responded markedly to light. Leaf-area ratio in thethree treatments showed extreme plasticity in increasing from2–3 x 10–2 through 6 x 10–2 to 8–9 x10–2 m2 g–1 as light intensity decreased. Correspondingdecreases in unit leaf rate, however, caused over-all reductionsin relative growth rate. Specific absorption rates for potassium (AK, dry-weight basis)were strongly reduced at the lower light intensities but alsodisplayed complex ontogenetic drifts. Values of the allometricconstant, k (the ratio of root and shoot relative growth rates),decreased from c. 0.7 at 36.1 W m–2 through c. 0.3 at7.3 W m–2 to a value not significantly different fromzero (P < 0.05) at 2.2 W m–2. In material grown under the two higher light intensities a constantinverse relationship was found between the mass ratio of rootand shoot and the corresponding activity ratio. The resultsconform to this model: Mass ratio = –0.001+45.0 (1/activityratio) where activity ratio is expressed as specific absorptionrate for potassium (in µg g root–1 h–1)/unitshoot rate (rate of increase of whole-plant dry weight per unitshoot dry weight, in mg g shoot–1 h–1). Mannitol-1-phosphate dehydrogenase (EC 1.1.1.17 [EC] ) and mannitol-1-phosphatase(EC number yet unassigned) were detected in the brown algae,Spatoglossum pacificum and Dictyota dichotoma. The enzymes wereextracted from algal fronds and their properties were investigatedusing partially purified preparations. Mannitol-1-phosphatase shows maximum activity at pH 7. The enzymehad a narrow substrate specificity. The Km value for mannitol-1-phosphateis 8.3x10–4 M (30°C, pH 7.0). The enzyme is activatedby Mg++ and Mn++and is strongly inhibited by PCMB, Hg++and NaF. Mannitol-1-phosphate dehydrogenase showed maximum activitiesat pH values 6.5 and 10.2 in reductive and oxidative reactions,respectively. The dehydrogenase also showed narrow substratespecificity; mannitol-1-phosphate and NAD or fructose-6-phosphateand NADH2 are utilized, respectively, in oxidative and reductivereactions by the enzyme. Km values for these substrates andthe coenzymes are 2.5x10–4 M and 7.1x10–5 M forthe first pair and 2.8x10–4 M and 1.3x10–5 M forthe latter pair. This enzyme was strongly inhibited by PCMBand Hg++, but was only slightly affected by adenosine phosphates. The uptake of L-leucine into Vinca protoplasts was studied undervarious conditions. The uptake was highly pH-dependent, withthe optimal pH between 3.0 and 4.0. The uptake was also energydependent, since azide, 2,4-dinitrophenol (DNP), carbonyl cyanidem-chlorophenyl hydrazone (CCCP), and iodoacetate inhibited theuptake. Oligomycin, N,N'-dicycIohexyI carbodiimide (DCCD) andvanadate, but not ouabain, inhibited the uptake, suggestingthat ATPase for H+ electrogenic extrusion was necessary to theuptake of L-leucine. The uptake showed stereospecificity, butwas partially inhibited by other L-amino acids. A kinetic studyof the uptake showed that the uptake was multiphasic with threesaturable phases and one unsaturable phase which occurred atconcentrations of L-leucine over 1 mM. The Km values of thethree affinity sites were 1.4 x 10–3 M, 1.3 x 10–4M, 4.3 x 10–5 M; the maximum velocity values were 3.3x 10–8, 4.5 x 10–9, 1.8 x 10–9 mol/10 min/4x 106 cells. Water permeabilities of astomatous, isolated cuticular membranesof 15 vascular epiphyte species (families: Araceae, Orchidaceaeand Piperaceae) from the moist lowland forest of Barro ColoradoNature Monument, Panama, were investigated. Permeances determinedat 30°C ranged from 0.46 x 10-6 m s-1(Aspasia principissa(Rchb.f.) R.E. Schult.) to 6.07 x 10-6 m s-1(Polystachya foliosa(Hook.)Rchb. f.). Comparison of these data with permeances of plantsfrom other habitats corroborates the notion that cuticular propertiesreflect the climatic demands of the growing sites. The studiedgroup of vascular epiphytes, living in a very drought-pronehabitat, showed the lowest cuticular permeances to water recordedto date. Sporidia of Ustilago maydis and conidia of Ceratocystis ulmipossess an antimycin A and azide-tolerant electron transportpathway which apparently diverts electrons to O2 from some pointon the substrate side of the antimycin A block. The alternatepathway (induced by 0.5 µg/ml antimycin A or 5x10–4M sodium azide) supports a respiratory rate 1.5–2 timesthat of the normal system, but has a terminal oxidase with alower than normal affinity for O2. A similarly high respiratoryrate in U. maydis is supported by the normal pathway when uncoupledby 4 µg/ml of 4,5-dichloro-2-trifluoromethylbenzimidazole,but a high affinity for O2 in this case indicates that the normalterminal oxidase is utilized. Respiration by the normal pathway in both fungi is only slightlyor moderately inhibited by 1.5x10–3 M benzohydroxamicacid (BHAM) and 5x10–4 M 8-hydroxyquinoline. The alternatepathway in U. maydis, however, is inhibited as much as 84 and92% respectively by these two compounds, while alternate respirationin C. ulmi can be inhibited as much as 86 and 76% respectively.BHAM, 8-hydroxyquinoline, 2-pyridinethiol-1-oxide, a,a'-dipyridyl,carboxin, and diphenylamine inhibit alternate respiration ata site on the alternate pathway which is not part of the normalelectron transport system. Antimycin A and azide-insensitiverespiration found in U. maydis and C. ulmi closely resemblesinhibitor insensitivity noted in several fungi and some higherplants. A dual radioactive-labelled bacteria technique using Vibrio(DRLV), developed for laboratory studies on bacterivory, hasbeen refined for use at the concentrations of prey and predatorstypcially found at sea. Experiments with estuarine water collectedin spring and in autumn showed that bacterivorous nanoflagellates(HNF) (concentration 1.38±0.35x103 HNF ml–1) ingested2.7±0.96 DRLV flagellate1–1 h–1 at concentrationsof 0.8–2.2x106 DRLV ml–1 in the presence of 2.04±0.68x106natural bacteria ml–1. The method was also applied tosamples collected in October in the Celtic Sea, when on average1 ml of water from the surface layer contained 1.41±0.16x106natural bacteria, 14.6x103 cyanobacteria, 530±170 HNF,7.3±3.0x103 phototrophic nanoflagellates (1.5–4µm), 49.0±26.5 phototrophic dinoflagellates, 36.3±12.6heterotrophic dinoflagellates and 21.3±9.5 Leucocryptosmarina. Under these conditions the grazing rate in most samplesdid not exceed the coefficient of variation of the method (2%),although we estimate the grazing rate was -1.6 DRLV HNF–1h–1 and on one occasion a rate of 2.45 was recorded. Shells from 14 populations of sphaeriid clams including Sphaeriumstriatinum, S. simile, Pisidium walkeri, Musculim partumeiumand M. iransversum were analyzed for organic carbon (µgCmg–1 shell), nitrogen (µg,N mg–1 shell) andCaCOs (%CaCO3 of total clam dry weight). Habitat waters wereassessed for total hardness (expressed as ppm CaCo3), ppm Ca,ppm Mg, conductivity (µmho) and suspended organic Carbon(µgCl–1). For all populations, shell organic C andN are positively correlated and there is an inverse relationshipbetween the amounts of shell CaCO3 and shell organic carbon.Trophic considerations give the best correlation with shelltype at the generic level of consideration since species ofMusculium are found at the opposite end of the trophic scale(eutrophic) from all other populations studied. For S. striatinum,the most extensively studied species, the amount of shell CaCO3is inversely related to water hardness. Grazing and ingestion rates of laboratory-born Thalia democraticaaggregates and Dolioletta gegenbauri gonozooids, phorozooidsand oozooids were determined while fed Isochrysis galbana (4–5µm diameter) alone or in combination with Peridinium trochoideum(16–18 µm diameter) at concentrations of 0.15–0.70mm3 x 1–1. Grazing rates (ml x zooid–1 x 24 h –1)ranged from 10 to 355, and at zooid weights greater than 5 µgcarbon were in order oozooid > gonozooid > aggregate.Grazing rates increased exponentially with increasing zooidweight. Weight-specific grazing rates (ml x µgC–1x 24 h–1) were independent of the four-fold initial foodconcentration. Mean weight-specific grazing rates increasedlinearly with increasing zooid weight for the aggregates andoozooids, but gonozooid mean rates were independent of zooidweight. Aggregate and gonozooid ingestion rates (106 µm3x zooid–1 x 24 h–1) ranged from 4 to 134 while oozooidrates ranged from 3 to 67. All ingestion rates were independentof the initial food concentration but increased linearly withincreasing zooid weight at similar rates. All mean weight-specificingestion rates (ml x µgC–1 x 24 h–1) wereindependent of zooid weight. The mean aggregate daily ration(µgC ingested x µg body C–1) was 59% and themean doliolid ration was 132%. Field studies indicate that normalconcentrations of D. gegenbauri in the Georgia Bight clear theirresident water volume (1 m3) in about 4 months, but that highlyconcentrated, swarm populations which occur along thermohalinefronts clear their resident water volume in less than 1 day. 1Current address: MacLaren Plansearch Ltd., P.O.Box 13250, sta.A.,St.John's, Nfld. Incorporation of polyamine-spermidine into the nutrient mediumat 10–6 and 10–5 M concentrations stimulates pollen-tubegrowth in vitro in Catharanthus roseus L. G. Don. MGBG, an inhibitorof spermidine biosynthesis, at 0.5 x 10–3 and 1 x 10–3M concentrations reduced the percentage of germination as wellas tube growth and at a concentration of 1.5 x 10–3 Mgermination was totally inhibited. Pollen grains incubated inthe medium containing 1.5 x 10–3 M MGBG, when transferredto a fresh medium with 10–5 M spermidine, resulted in80% germination recovery, along with considerable tube growth.Experiments with actinomycin-D indicate that stimulation ofpollen-tube growth by spermidine may involve de novo synthesisof protein. The theory and practice of applying the thermodynamics of irreversibleprocesses to mass-flow theories is presented. Onsager coefficientswere measured on cut and uncut phloem and cut xylem strandsof Heracleum muntegazzimum. In 0.3 N sucrose + 1 mN KC1 theyare as follows. In phloem, LEE = 5 ? 10–4 mho cm–1,LpE = 9 ? 10–6 cm3 s–1 cm–2 volt–1 cm,and LPP = 0.16 cm3 s–1 cm–2 (J cm–3)–1cm. In uncut phloem strands LEE is about 1 ? 10–3 mhocm–1. In xylem in 2 x 10–3 N KCI, Lpp = 50 to 225,LPE = 2 ? 10–4, and LEE = 4 ? 10–3. The measurementsare tentative since the blockage of the sieve plates is an interferingfactor, but if they are valid they lead to the conclusion thatneither a pressure-flow nor an electro-kinetic mechanism envisaginga ‘long distance’ current pathway can be the majormotive ‘force’ for transport in mature phloem. Measurementsof biopotentials along conducting but laterally detached phloembundles of Heracleum suggest, nevertheless, that there may bea small electro-osmotic component of at least 0.1 mV cm–1endogenous in the phloem.  相似文献
