Pigeons sitting on a turntable are exposed to horizontal -ramp-and-hold stimuli (= -RP-stimuli; angular velocity) (Fig. 1). At the same time, the mean rateE(t) of impulses/0.1 s or 0.2 s ofM. abductor indicis, M. abductor pollicis orM. flexor pollicis is registered. If the rotation axis lies in or in front of the pigeon's head the muscles studied respond phasic-tonicly to an ipsilaterad -RP-stimulus; and purely tonicly or also phasic-tonicly to a contralaterad -RP-stimulus (Figs. 2C and 4). The heightE P of the tonic response component is independent from the direction of rotation and is only determined by the height of the -plateau (Fig. 2A). The heightE D of the phasic response component is dependent upon the rotation direction and is determined, at a given rotation direction, by (Fig. 2A) and by the ramp slope ( angular acceleration) (Fig. 3A). This and the dependence of the tonic component and, with some pigeons, also of the phasic component uponr (distance between pigeon and rotation axis) lead to the conclusion that the tonic component is caused by the centripetal accelerationb p , and the phasic component is caused by the angular acceleration and, with some pigeons, by the tangential accelerationb t. The influence of the acceleration components uponE D andE P is dependent upon the pigeon's position angle relativ to the radiusvectorr (Figs. 4 and 5). The tonic component is the response to a quasi-pitch or a quasi-roll of the pigeon during steady rotation. This results from a comparison of the -RP-responses of the pigeon in a radial or tangential position (Fig. 6A) to the pitch and roll responses (Fig. 6B). A model has been developed on the basis of the measured stimulus-response relations (Fig. 7). It contains three input channels (x -,u t- andu p-channel) with low-pass filters for the angular, tangential and centripetal acceleration. The channel gain depends upon . The sum(t) of all channel outputs is sent through a pure-time-delay element and finally through a half-wave rectifier with a threshold and an upper limitation. The transfer characteristics of the model agree rather well with those of the pigeon. The model can be interpreted in terms of neurophysiology.

A long-term field experiment was initiated to simulate chronic atmospheric N deposition, a widespread phenomenon in industrial regions of the world. Eight years of experimental nitrate ( ) additions (3 g -N m–2 per year) to four different northern hardwood forests located along a 500 km geographic gradient dramatically increased leaching losses of -N, dissolved organic carbon (DOC), and dissolved organic nitrogen (DON). During the last two water years, the average increase in solution -N and DON leaching from the -amended plots was 2.2 g N m–2, equivalent to 72% of the annual experimental N addition. Results indicate that atmospheric N deposition may rapidly saturate some northern hardwood ecosystems across an entire biome in the upper Great Lakes Region of the USA. Changes in soil C and N cycling induced by chronic N deposition have the potential in this landscape to significantly alter the flux of DOC and DON from upland to aquatic ecosystems. Michigan Gradient study site characteristics are similar to those of European forests most susceptible to N saturation.  相似文献

Summary Linkage data, using the polymorphic markers 52A (DXS51), F9, 4D-8(DXS98), and St14(DXS52), are presented from 14 fragile X pedigrees and from 7 normal pedigrees derived from the collection of the Centre d'Étude du Polymorphisme Humaine. A multipoint linkage analysis indicates that the most probable order of these four loci in normal families is DXS51-F9-DXS98-DXS52. Recombination frequencies ( ) corresponding to maximum LOD scores ( ) were obtained by two-point linkage analysis for a nuber of linkage groups, including: DXS51-F9 ( =5.94, =0.03), F9-DXS98 ( =0.51, =0.26), F9-DXS52 ( =0.84, =0.27), and DXS98-DXS52 ( =0.32, =0.20). A multipoint linkage analysis of these loci, including the fragile X locus, was also performed for the fragile X population and the data support the relative order (DSX51, F9, DXS98)-FRAXA-DXS52. Recombination frequencies and maximum LOD scores, which again were derived from two-point linkage analyses, were obtained for the linkage groups DXS51-F9 ( =9.96, =0) and F9-DXS52 ( =0.07, =0.45), as well as for the groups DXS51-FRAXA ( =2.42, =0.15), F9-FRAXA ( =1.30, =0.18), DXS98-FRAXA ( =0.05 =0.36), and DXS52-FRAXA ( =2.42 =0.15). The linkage data was further tested for the presence of genetic heterogeneity both within and between the fragile X and normal families for the intervals DXS51-F9, F9-DXS52, F9-FRAXA, and DXS52-FRAXA using a modification of the A test. Except for the interval F9-FRAXA (P<0.10) there was no evidence of genetic heterogeneity for each of the various linkage groups examined. The heterogeneity detected for the interval F9-FRAXA, however, was most likely due to one family (Fx-28) that displayed very tight linkage between these two loci.  相似文献

Summary Although membranes are often viewed as barriers to diffusing particles, in many cases their presence does not slow down diffusion. Investigations of the transit time (mean diffusion time) for cases where the source and the target of diffusing particles are separated by various arrangements of membranes reveal the following facts: (i) The transit time is composed of the sum of the times to diffuse each of the membrane and aqueous regions separately and terms representing the time spent at the vicinity of the interfaces between these regions. (ii) In cases of one dimensional diffusion between aqueous and membranal phases, the transit time is governed by the parameter whereD m andD w are the diffusion coefficients in the membrane and water, respectively, and is the membrane/water partition coefficient of the particles. While the former ratio depends mostly on the viscosities of the two phases, the latter parameter is very strongly dependent on the identity of the particles. The diffusion from water to the membrane is faster than from the membrane to water whenever 1$$ " align="middle" border="0"> . The opposite is true when this parameter is smaller than 1. (iii) In case of one dimensional transmembranal diffusion, the transit time shows a minimum when wherel w1 andl w2 are the net diffusion distances in the aqueous phases on both sides of the membrane. In this case, if the diffusion proceeds through pores in the membrane, represents the fraction of membrane area that is occupied by the pores.The transit times for three dimensional diffusion into and from a spherical cell are also presented in a simple form. In addition, some of the relations between transit times and other measurable time parameters, such as the course of the decay of gradients and the time lag to establish steady states, are discussed briefly.The conclusions emerging from this analysis, together with the simple expressions for the transit times can make these investigation useful for the understanding of diffusion in systems containing natural or artificial membranes.  相似文献

Summary Six Standardbred horses were used to evaluate the time course of pulmonary gas exchange, ventilation, heart rate (HR) and acid base balance during different intensities of constant-load treadmill exercise. Horses were exercised at approximately 50%, 75% and 100% maximum oxygen uptake ( max) for 5 min and measurements taken every 30 s throughout exercise. At all work rates, the minute ventilation, respiratory frequency and tidal volume reached steady state values by 60 s of exercise. At 100% max, the oxygen consumption ( ) increased to mean values of approximately 130 ml/kg·min, which represents a 40-fold increase above resting . At the low and moderate work rates, showed no significant change from 30 s to 300 s of exercise. At the high work rate, the mean at 30 s was 80% of the value at 300 s. The HR showed no significant change over time at the moderate work rate but differing responses at the low and high work rates. At the low work rate, the mean HR decreased from 188 beats/min at 30 s to 172 beats/min at 300 s exercise, whereas at the high work rate the mean HR increased from 204 beats/min at 30 s to 221 beats/min at 300 s exercise. No changes in acid base status occurred during exercise at the low work rate. At the moderate work rate, a mild metabolic acidosis occurred which was nonprogressive with time, whereas the high work rate resulted in a progressive metabolic acidosis with a base deficit of 16 mmol/l by 300 s exercise. This paper investigates the relationship between the hydroclimatic parameters (rainfall and flood index) and the catch, stock abundance and recruitment of the catfish, Chrysichthys nigrodigitatus (Bagridae) and the bonga, Ethmalosa fimbriata (Clupeidae) of southeastern Nigeria's inshore waters. For C. nigrodigitatus, most peaks in the mean biomass and recruitment curves occurred during the 'wet' years, i.e., years for which the percentage deviations of rainfall and flood index from their means remained above their averages. Catch and abundance respectively showed good positive linear correlation with the flood index. E. fimbriata behaved differently; some peaks in the and curves occurred in the 'wet' and some in the 'dry' years; and there was no correlation between the annual catch of bonga and either the rainfall or the flood index. The hypothesis, that linear relationships exist between the interannual variations in the hydroregime and the yearly fluctuations in the catch and population structure of some coastal and estuarine fishes, holds true for the catfish, C. nigrodigitatus, but not for bonga, E. fimbriata.  相似文献

Summary The in vivo phototransformation kinetics of mustard hook and cotyledon phytochrome exhibit a deviation from a single first order curve, quite similar to that for pumpkin hooks as reported in a previous paper (Boisard, Marmé and Schäfer, 1971). The P frPrkinetics can be characterized by the ratios fr, I · P fr I / fr, II · P fr, II and where P fr I and P fr II are two populations of phytochrome molecules which convert to P rwith a first order half-life of and . These ratios depend on the length of time of etiolation. The ratio fr, I · P fr I / fr, II · P fr, II is independent of the amount of total P frpresent at the beginning of the P frPrphototransformation after a non-saturating dose of red light. Summary The resting membrane potential data existing in the literature for the giant axon of the squid, frog muscle and barnacle muscle have been analyzed from the standpoint of the theory of membrane potential due to Kobatake and co-workers. The average values derived for the effective charge density (where is a constant, , and represents the fraction of counterions that are free, and is the stoichiometric charge density in the membrane) present on the different biomembranes existing in their normal ionic environment are 0.3, 0.325 and 0.17 M for the squid axon, frog and barnacle muscles, respectively. On the assumption that the values of are 0.4 and 0.2 for nerve and muscle membranes, respectively, values of 0.75, 1.62 and 0.85 M have been derived for the stoichiometric charge density present in the respective biological membranes. These correspond to 1 negative charge per 222, 103 and 195 Å of the membrane area of the squid axon, frog and barnacle muscles, respectively.  相似文献

The authors have developed a continuous recycle reactor which efficiently performs emulsion type enzymatic reactions. The reactor column is filled with immobilised lipase and the reactions are effected by pumping the pre-prepared oil-water emulsion through the bottom of the reactor. A part of the product was recycled back and this type of recycling greatly improves the productivity of fatty acid compared to continuous once-through reactor without recycling. The recycle reactor could be continuously run for 35 days without decrease in conversions. Respiratory chain phosphorylation has been investigated in the methylotrophic bacterium Methylophilus methylotrophus following the addition of oxidisable substrates to aerobic, whole cell suspensions. Initial-rate experiments showed that ATP synthesis occurred at the overall expense of AMP and inorganic phosphate via the sequential action of the ATP phosphohydrolase and adenylate kinase; some of the nascent ATP was rapidly used to synthesis nonadenine nucleoside triphosphates. After being corrected for ATP turnover, Pi/O quotients of 0.46 to 0.54, 0.77 and 1.37 nmol/ng-atom O were obtained for the oxidation of methanol dehydrogenase-linked substrates (methanol, ethanol and acetaldehyde), duroquinol and formate (NAD+-linked) respectively. These values were proportional to the H+/O and/or K+/O quotients exhibited by these substrates, and yielded an average H+/ATP (H+/Pi) quotient of 4.2 ng-ion H+/nmol. Steady-state experiments showed that the extent of cellular energisation varied with the respiration rate but was always in the order methanol > duroquinol > acetaldehyde, thus indicating that under these longer-term conditions methanol was completely oxidised to yield PQQH2 and 2NAD(P)H. An analytical model is developed to describe the performance of a packed-bed immobilized enzyme reactor in which parallel processes take place. In particular, two-substrate reaction, inhibition of the enzyme by one of the reaction products, and binding of one substrate and/or one product to an added ligand are taken into account. In addition, substrates and product diffusion into the porous catalyst are also considered. Using this model, numerical simulations were performed. The results point to the fact that, when all the above processes occur concomitantly, a variety of performance characteristics can be obtained, depending on the particular values of the related parameters. The and the Gp have been measured in whole cells ofMethylophilus methylotrophus during the oxidation of various respiratory chain substrates. The magnitude of the depended on the external pH and the composition of the assay medium, and varied from-109 to-165 mV. The relative contributions of the and the pH to the were found to vary with the external pH such that the internal pH remained constant; depending on the composition of the assay medium, this value was between 6.6 and 7.0. A Gp of approximately-46 kJ/mol was generated during the oxidation of methanol, and either the or pH alone was fully competent to drive ATP synthesis. Respiration and ATP synthesis were found to be poised far from equilibrium under the conditions of these experiments, and the value of the Gp was thus controlled kinetically. Summary Adelie penguins (Pygoscelis adeliae) experience a wide range of ambient temperatures (T a) in their natural habitat. We examined body temperature (T b), oxygen consumption ( ), carbon dioxide production ( ), evaporative water loss ( ), and ventilation atT a from –20 to 30 °C. Body temperature did not change significantly between –20 and 20°C (meanT b=39.3°C).T b increased slightly to 40.1 °C atT a=30°C. Both and were constant and minimal atT a between –10 and 20°C, with only minor increases at –20 and 30°C. The minimal of adult penguins (mean mass 4.007 kg) was 0.0112 ml/[g·min], equivalent to a metabolic heat production (MHP) of 14.9 Watt. The respiratory exchange ratio was approximately 0.7 at allT a. Values of were low at lowT a, but increased to 0.21 g/min at 30°C, equivalent to 0.3% of body mass/h. Dry conductance increased 3.5-fold between –20 and 30°C. Evaporative heat loss (EHL) comprised about 5% of MHP at lowT a, rising to 47% of MHP atT a=30°C. The means of ventilation parameters (tidal volume [VT], respiration frequency [f], minute volume [I], and oxygen extraction [ ]) were fairly stable between –20 and 10°C (VT did not change significantly over the entireT a range). However, there was considerable inter- and intra-individual variation in ventilation patterns. The growth yield of Chlorella vulgaris, Y kJ defined as g cells harvested per kJ of light energy absorbed by the cells, was assessed in a turbidostat culture by varying CO2 and O2 partial pressures ( and ). The value of Y kJ ranged from 3.1×10-3 to 5.0×10-3 g cells/kJ under light-limited conditions [ = 1.02.4%, = 065%; total pressure of gas (composed of CO2, O2 and N2)=1 atm]. In the light-limited environment, the algal specific growth rate deteriorated appreciably with the increase of . The deterioration accounts for the above range of Y kJ observed. The growth inhibition due to oxygen that was defined by subtracting from 1.0 the ratio of at given values of to that at = 0% extended from 0.07–0.30 (7–30%). However, glycolate could not be detected in the turbidostat culture. Isotopic experiments on the specific rate of 14CO2 uptake also revealed that the inhibition due to oxygen was from 22–38% when was varied from 0 to nearly 100%. Analysis of linkage disequilibrium (=mean squared correlation of allele frequencies at different gene loci) provides a means of estimating effective population size (N e) from a single sample, but this method has seen much less use than the temporal method (which requires at least two samples). It is shown that for realistic numbers of loci and alleles, the linkage disequilibrium method can provide precision comparable to that of the temporal method. However, computer simulations show that estimates of N e based on for unlinked, diallelic gene loci are sharply biased downwards ( in some cases) if sample size (S) is less than true N e. The bias is shown to arise from inaccuracies in published formula for when S and/or N e are small. Empirically derived modifications to for two mating systems (random mating and lifetime monogamy) effectively eliminate the bias (residual bias in % in most cases). The modified method also performs well in estimating N e in non-ideal populations with skewed sex ratio or non-random variance in reproductive success. Recent population declines are not likely to seriously affect , but if N has recently increased from a bottleneck can be biased downwards for a few generations. These results should facilitate application of the disequilibrium method for estimating contemporary N e in natural populations. Direct nonenzymatic oxidation of semiquinone by oxygen is one of the main sources of superoxide radicals in mitochondria. Using all the known data on hepatocyte mitochondria, we have revealed the correlation between the rate of superoxide generation by the bc 1complex and the transmembrane potential (). Assuming that the main electrogenic stage of the Qcycle is the electron transfer between the cytochrome bhemes, then the rate of superoxide generation sharply increases when grows from 150 to 180 mV. However, this interrelation is ambiguous. Indeed, the increase of the generation rate with the growth of the potential can occur faster when succinate dehydrogenase is inhibited by malonate than when external ADP is exhausted. When the potential is changed by adding phosphate or potassium (K+), the rate of production remains constant, although the comparison of the rates at the same reveals the effect of phosphate or potassium. It turned out that the rate of generation is a function of rather than any of its components. Phosphate and K+have practically no influence on , since the change in is compensated by pH. The rate of superoxide generation by the bc 1complex is a multiple function of the electron-transfer activity of enzymes, the processes determining the membrane potential (e.g., loading), and the oxygen concentration. Summary The histochemical distribution of various hydroxysteroid dehydrogenases in human, term, foetal membranes has been investigated using the tetrazolium dye, Nitro-B.T.The trophoblastic layer was the most active, showing 3-, 3-, 11-, 16- and 17-hydroxysteroid dehydrogenase activities, a pattern of activity similar to that of the placental villous trophoblast.The amniotic epithelium showed weak 3-, 3-, 16- and 17-hydroxysteroid dehydrogenase activity; weak 3- and 3-hydroxysteroid dehydrogenase activity was noted in the connective tissue layers.All activity demonstrated was N.A.D.-linked.  相似文献

Summary The influence of local temperature changes within the posterior portion of the body on dorsal aorta blood flow ( ), femoral arterial pressure (P a ), peripheral resistance (R), skin blood flow ( ) and skeletal muscle blood flow ( ) was examined in unanesthetized lizards (Iguana iguana andTubinambis nigropunctatus). In response to local heating of the hind legs and tail and increased,P a was generally unchanged,R decreased and decreased or was unchanged (Fig. 2). It is suggested that the acquisition of heat may be favored by diverting the increase in away from the muscle to the warmer skin. In response to cooling and decreased,P a was generally unchanged, R increased and increased or was unchanged. Hence, during cooling the retention of heat may be favored by diverting blood away from the skin to the deeper muscle. The muscle-skin shunt is under sympathetic control since following blockade with phenoxybenzamine HCL (Dibenzyline) muscle blood flow changes in response to temperature were qualitatively similar to those of skin (Fig. 4).
