动脉平滑肌细胞(sm ooth m uscle cell,SMC)是动脉粥样硬化(atherosclerosis,AS)斑块中的主要细胞,它的增殖在AS形成过程中极其重要.利用体外培养的人主动脉SMC,观察了天然高密度脂蛋白(native high density lipoprotein,N-HDL)及氧化修饰HDL(oxidized HDL,OX-HDL)对培养人主动脉SMC cyclin D1(细胞周期蛋白D1)基因转录表达的影响.结果表明:(1)N-HDL对SMCcyclin D1基因表达无影响(P> 0.05);(2)OX-HDL使SMCcyclin D1基因表达显著增强(P<0.01),其表达量随时间(2、12、24 h)延长而增加.上述结果表明,OX-HDL的致AS作用可能与其刺激SMCcyclin D1基因表达增加有关.  相似文献

本研究观察到10-7~10-5kg/L脂多糖(lipopolysacharide,LPS)可显著促进血管平滑肌细胞(VSMC)的增殖及DNA的合成(P<005)。5×10-4~10-3kg/LLPS却抑制VSMC的增殖及DNA的合成,降低其活力(P<001),并呈时间依赖效应。一氧化氮合酶抑制剂NNitroLArginine(LNNA)可拮抗LPS的抑制作用。大剂量LPS作用组VSMC上清液中一氧化氮(NO)代谢产物NO-3和NO-2的含量与对照组相比显著增加(P<001),48h组比24h组增加91%,72h组比48h组增加45%;同时,诱导性一氧化氮合酶(inductivenitricoxidesynthase,iNOS)免疫组化染色呈阳性。结果表明,低浓度LPS促进VSMC增殖和DNA合成,而高浓度LPS却明显抑制VSMC增殖和DNA合成,降低其活力。这种抑制作用可能与LPS诱导VSMC产生的NO有关。  相似文献

为探讨饥饿及饥饿与骤冷对动物肝脏的影响,本实验用健康昆明种小鼠25只,随机分成正常组5只,饥饿组10只,饥饿后再予冷刺激组10只(下称骤冷组)。采用组织化学及酶组织化学方法观察糖原(PAS反应)、SDH(琥珀酸脱氢酶),LDH(乳酸脱氢酶),ChE(胆碱酯酶)、Mg2+-ATPase(镁激活三磷酸腺苷酶),ACP(酸性磷酸酶)。结果提示:饥饿时肝脏PAS反应,SDH,Mg2+-ATPase、ChE活性显著下降,而ACP活性明显增强;饥饿后骤冷时PAS(反应)、SDH、ChE更显著下降,而ACP及Mg2+-ATPase活性反而增强。  相似文献

半胱胺对大鼠泌乳量及血液,组织几种激素含量的影响   总被引:12,自引:1,他引:11  
选用40只Wistar泌乳大鼠进行配对实验。哺乳期第8天起,实验母鼠灌服半胱胺(cysteamine,CS)80mg/kgB.W,每隔5天一次,直至哺乳第30天。实验中记录泌乳量、仔鼠体重及采食量,并用放免法测定血液及组织生长抑素(SS)、生长激素(GH)、催乳素(PRL)的含量。结果表明,CS使母鼠哺乳期第10、15、20天的泌乳量明显提高(P<0.05),第25天泌乳量有所增加(P>0.05),第30天差异不明显;母鼠哺乳期采食量略有增加(P>0.05),料/奶比明显下降(P<0.05);哺乳期第15天的血液、下丘脑和大脑皮层SS含量明显下降,血液GH含量显著升高(P<0.05),但第25天无明显变化;母鼠哺乳期血液PRL含量明显下降(P<0.05)。提示,CS能促进大鼠泌乳,而抑制体内SS,提高内源性GH水平,可能为CS的主要作用机制。  相似文献

观察了武汉抗CD3单克隆抗体(简称WuT3)对组氨酸缺陷型鼠伤寒沙门氏菌TA97、TA98、TA100及TA102菌株的回复突变作用。结果显示在5~5000μg/皿的剂量范围内,WuT3所致的诱发回复突变菌落数与自发回复突变菌落数之比MR(Rt/Rc),无论加大鼠肝匀浆,辅酶Ⅱ及葡萄糖6-磷酸(S-9混合液)或不加S-9混合液,均不超过2。同时观察了WuT3对小鼠骨髓细胞微核率及对人外周血淋巴细胞染色体畸变率的影响,结果显示小白鼠ivWuT3,每日一次,连续2日,在25~100mg·kg-1范围内,WuT3各剂量组的微核细胞率与溶剂对照组相比均无显著性差异(P>0.05)。而环磷酰胺(CP)阳性组与溶剂对照组相比有极显著性差异(P<0.01)。WuT3在25~250μg/瓶的剂量范围内,各剂量组的染色体畸变细胞率与溶剂对照组相比无显著性差异(P>0.05),而CP组与溶剂对照组相比有极显著性差异(P<0.01)。三项试验结果均未发现WuT3有致突变性作用  相似文献

用10μg/gLa ̄(3+)、1.0%Vc、10μg/gLa ̄(3+)+1·0%Vc、蒸馏水四种处理,喷施正处在衰老过程中的黄瓜叶片,一周后提取PSⅡ颗粒,通过SDS-PAGE、薄层扫描和Clark测氧等测试技术,发现处理与对照的PSⅡ颗粒在与放氧活性密切相关的33、23、17kD三肽的相对总含量上有变化,10μg/gLa ̄(3+)处理比对照减少28.9%,而1.0%Vc和10μg/gLa ̄(3+)+1.0%Vc处理的与对照接近。前者的放氧活性降低37.3%,后者的放氧活性增加62.4%,是对照的1.6倍,共同处理的结果与对照相近。可见,在光合放氧中,1.0%Vc与10μg/gLa ̄(3+)拮抗。而且,33、23、17kD三肽总量的相对减少与放氧活性的高低呈正相关。  相似文献

测定日本大耳白兔母兔配种后10小时血清中的10项生理指标,并与母兔所产每窝仔兔的性比(雄性个体所占比率)进行对应分析(窝仔数<6的数据未参与此项分析)。结果表明:母兔血清中FSH、T(睾酮)、 Na+和M2+的浓度在高、低两个性比组间有显著差异, T3(三碘甲状腺原氨酸)的差异接近显著水平(P<0.1);并且,FSH和T3与子代性比里显著负相关(r分别为-0.50和-0.46),Mg2+与子代性比里显著正相关(r=0.39);同时,Mg2+/Ca2+、Mg2+×Na+、Mg2+×K+和T3×FSH与子代性比的相关分别为0.40、0.43、0.39和-0.53(P<0.05)。  相似文献

本文报告了进行性系统性硬化病(PSS)患者外周血单个核细胞中C-myc、Ki-ras和Ha-ras三种癌蛋白的表达和NK细胞亚群的检测结果。与对照相比,PSS患者淋巴细胞的C-myc癌蛋白阳性率显著升高(P<0.01),单核细胞的阳性率也倾向于明显升高(P=0.052);PSS组单核细胞的Ki-ras癌蛋白表达显著高于对照组(P<0.001)。另外,PSS患者NK细胞中Leu-11c~+亚群细胞数显著低于对照值,而Leu-7+亚群细胞数则显著高于对照值(P<0.05)。作者结合文献讨论了PSS患者体内癌基因表达的可能机制以及癌基因表达和NK亚群改变的临床意义。  相似文献

雄性SD大鼠33只,体重180—250g,随机分成四组。正常对照组6只;哌唑嗪组8只,于实验第1、2、3日上午用哌唑嗪灌胃(1mg/kg体重)共3次,第2日下午腹腔注射半乳糖胺(600mg/kg体重)1次,第4日上午处死动物,取肝脏及血清作有关检查;普萘洛尔组及N.S对照组分别用普萘洛尔(2mg/kg体重)及N.S(10ml/kg体重)代替哌唑嗪灌胃,处理程序同哌唑嗪组。结果表明:一、哌唑嗪能显著减轻肝脏病变及血清ALT的升高。二、哌唑嗪对肝损害后肝组织SDH、Mg2+-AT-Pase、ChE、ACP、CCo等酶活性的恢复有显著效果。三、哌唑嗪组LPO明显低于N.S对照组(P<0.01)而SOD活性明显高于N.S对照组(P<0.01)。普萘洛尔组各项指标同对照组比较均无显著差异(P>0.05)。提示:哌唑嗪对大鼠实验性肝损害有一定的保护作用。  相似文献

The present study assessed the use of Bacillus subtilis in fish as a probiotic. The bacterium was administered orally at three different doses 0.5 x 10(7) (T(2)), 1 x 10(7) (T(3)), 1.5 x 10(7) (T(4)) cfu/g feed to Labeo rohita for two weeks. The positive control group (T(1)) and negative control group (T(5,)) were fed feed without B. subtilis for the same period. On the 15th day blood and serum were sampled to determine respiratory burst activity (NBT assay), differential leukocyte counts (DLC) and serum bactericidal activity. Fishes were challenged intraperitoneally with Aeromonas hydrophila O:18 after two weeks in the treatment groups (T(2), T(3) and T(4)) and also in the positive control group(T(1)), while the negative control group (T(5)) was challenged with phosphate buffered saline (PBS, pH 7.2) only. The respiratory burst activity and DLC were assessed on the 3rd day post-challenge. B. subtilis treated fish showed significantly higher (P<0.05) respiratory burst activity and bactericidal activity during the pre-challenge compared with the control groups. The highest respiratory burst activity (0.37+/-0.03) and serum bactericidal activity were recorded in the group (T(4)) fed feed containing B. subtilis at 1.5 x 10(7)cfu/g feed. Granulocyte numbers were significantly higher (P<0.05) in treatment groups in comparison to the control in both the pre- and post-challenge periods. The result suggests that B. subtilis can enhance certain innate immune responses in rohu.  相似文献

The objective was to assess the effects of the antioxidants resveratrol and quercetin on frozen-thawed ram sperm. Semen samples (which exceeded minimum standards) from four mature crossbreed Santa Inês rams were pooled and aliquots of each pool were diluted in Tris-egg yolk-glycerol, with the addition of 0, 5, 10, 15, and 20 μg/mL of resveratrol and quercetin in Experiment 1 and Experiment 2, respectively. In Experiment 1, the proportion of sperm with a high mitochondrial membrane potential was greater (P < 0.02) in the control group than in resveratrol 20 μg/mL group. In Experiment 2, the proportion of sperm with high mitochondrial membrane potential was greater in the control group (P < 0.0001) than in the other experimental groups, and greater in the quercetin 5 μg/mL group (P < 0.05) than in the other quercetin-treated groups. Thus, addition of 5 to 20 μg/mL of either resveratrol or quercetin to the Tris-egg yolk-glycerol extender reduced sperm mitochondrial membrane potential.  相似文献

The aim of this study was to evaluate the possible carcinogenic potential of residual DNA derived from immortalized and possibly tumorigenic cell lines due to activated oncogenic sequences (oncogenes). These cell lines have been used for the production of biologicals, i.e. monoclonal antibodies, lymphokines and vaccines. The authors used hybridoma DNA as a first model. For this reason experiments in two species were performed, namely in 3–4 week-old female Balb/c mice and newborn Riv:TOX rats. Doses of 250 μg DNA, derived from Balb/c hybridoma cells, were injected subcutaneously (s.c.) in 200 mice. These mice also received a s.c. injection of the solvent only (TE buffer) at another site of the back skin (negative control for local tumour development). An additional group of 50 mice was treated intraperitoneally (i.p.) with the solvent only to serve as a negative control group for possible systemic tumorigenic effects. Doses of 5 μg plasmid pPy1 DNA, containing the entire Polyoma virus genome, served as positive control and were injected s.c. and i.p. in 20 and 50 mice, respectively. Doses of 50 μg hybridoma DNA of 5 μg pPy1 DNA were injected s.c. in rats too, using nine animals per group. During the experiment, animals were observed weekly, especially for the occurrence of subcutaneous tumours at the injection sites. The mouse study was terminated after more than 2 years, the rat study after 1 year. Gross necropsy was performed on all animals and histopathological examination of grossly suspected neoplastic lesions was performed.In the mouse experiment, tumour development at the s.c. injection site of the DNA was observed in one out of 20 animals in the pPy1-treated positive control group (neurofibrosarcoma) and one out of 200 animals in the hybridoma DNA-treated group (haemangioma-like lesion). Tumour development at or near the s.c. injection site of the solvent only was observed in two out of 200 animals.In the rat study none out of nine hybridoma DNA-treated rats developed tumours at the injection site, while three out of nine rats of the positive control group, injected with the pPy1 DNA, showed local tumour development (benign and malignant soft tissue tumours).It is considered that, at the high dose and numbers of animals tested, parenteral administration of hybridoma DNA does not induce local tumour development. Furthermore, no indications were found for systemic carcinogenic potential of the hybridoma DNA used.Based on a worst case approach of our data, the oncogenic risk of 100 pg residual DNA was estimated to be 2×10−9, a value intermediate of the estimations of the WHO (1987) and the Dutch Health Council (1988) 5×10−11and 2×10−7, respectively. Therefore, it is unlikely that the risk of 100 pg of DNA derived from other immortalized cell lines will exceed the level of generally accepted cancer risk of 10−6.  相似文献

目的探索骨髓间充质干细胞(MSCs)对放射性肠炎(RE)肠黏膜修复的途径。 方法体外分离、培养大鼠骨髓MSCs。将RE模型的大鼠采用随机数字表法分为治疗组[经尾静脉注射干细胞悬液1 mL(细胞浓度为1×106个/mL)]和对照组(注射等量生理盐水,每日1次,连续3 d),每组10只。每日观察两组大鼠的活动量、进食和进水量、体质量变化等。1周后处死大鼠获取小肠标本,HE染色观察肠黏膜的修复情况,电镜观察上皮细胞的超微结构,免疫组化染色观察小肠隐窝Bmi-1阳性干细胞增殖情况,采用Image-Pro Plus 6.0软件对Bmi-1阳性细胞数量进行分析。组间差异的比较采用t检验。 结果成功分离得到大鼠骨髓MSCs,流式细胞仪鉴定:CD29、CD90、CD34、CD45阳性细胞比例分别为98.6﹪、99.6﹪、0.56﹪、0.89﹪。与对照组相比,治疗组大鼠移植1周后,体质量增加(190.30 g ± 13.23 g比235.00 g±14.30 g);大鼠小肠黏膜上皮得到修复,绒毛高度增高(627.50 μm ± 40.55 μm比984.33 μm ± 61.80 μm);上皮细胞超微结构较完整、清晰,隐窝Bmi-1阳性干细胞增殖数量增多[(60.67±9.63)个/mm2比(87.33 ±5.47)个/mm2],差异具有统计学意义(P < 0.05)。 结论骨髓MSCs能促进RE模型的大鼠肠黏膜的修复,这一作用可能是通过促进小肠隐窝干细胞的增殖发挥。  相似文献

In the current study, the effects of marginal Zn deficiency on myelin protein profiles in neonatal rats and rhesus monkeys were investigated. Following mating, rats were fed a Zn-adequate diet,ad libitum (50 μg Zn/g; 50 Zn AL), or a marginal Zn diet (10 μg Zn/g) from day 0 (10 Zn d0) or day 14 (10 Zn d14) of gestation to day 20 postnatal. An additional group of dams was restricted-fed the control diet to the food intake of the 10 Zn d0 group (50 Zn RF). Day 20 pup plasma and liver Zn concentrations in the 10 Zn groups were lower than in the 50 Zn groups. In a parallel experiment, rhesus monkeys were fed a Zn-adequatead libitum diet (100 μg Zn/g) or a marginal Zn diet (4 μg Zn/g diet; MZD) throughout gestation and lactation. Day 30 monkey infant plasma and liver Zn levels were similar in the MZD and control groups. Rat brain and monkey brain cortex weights were similar among the dietary groups. The amount of myelin recovered (mg protein/g brain) from day 20 rat pups from the 10 Zn groups was lower than that recovered from the 50 Zn rat pups. Myelin recovery from the MZD and control monkey infants was similar. When myelin protein profiles were characterized, it was found that the percentages of high-molecular-weight (HMW) proteins and Wolfgram protein were higher, whereas the percentages of small and large basic proteins were lower in myelin from the 10 Zn d0 and 50 Zn RF pups compared to the distribution in the 50 Zn AL rat pups. Results for the 10 Zn d0 and 10 Zn d14 pups were similar for all of the parameters studied. The percentage of HMW proteins was higher and that of basic protein lower in myelin from MZD monkey infants compared to the percentage of these proteins in myelin from controls. Although the interpretation of the rat data is complicated because of the anorexia associated with the Zn deficiency, the observed changes in monkey myelin protein profiles provide strong evidence that maternal Zn deficiency affects myelination in the offspring.  相似文献

It has been reported that the activity of protein improved when it was adsorbed inside the pores of mesoporous silica (MPS). The current study investigated the activity of immobilized avidin to the biotin on MPS with various pore sizes (diameter=2.4-45.0 nm). The binding amount of immobilized avidin to biotin is 123 to 160 ng biotin/10 μg avidin on 2.7- to 5.4-nm pore MPS, but that on 12- to 45-nm pore MPS was markedly decreased (33-42 ng biotin/10 μg). Moreover, the binding amount was approximately 2- and 3-fold higher on the glycidoxypropyl (Gly)-functionalized 5.4- and 45-nm pore MPS in comparison with methyl (Me)-functionalized 5.4- and 45-nm pore MPS, respectively. Furthermore, avidin immobilized in native and Gly-grafted 45-nm pore MPS retained more than 70% and 50% binding activity to biotin, respectively, after incubating at 90°C for 3 h. In contrast, the activity was greatly reduced in the native and Gly-grafted 5.4-nm pore MPS under the same conditions (<36.9%). The immobilization also protected against effects of 0.01 M HCl and 50% MeOH; all of immobilized avidin proteins showed high activity (>50%) with biotin compared with that observed with free avidin (MeOH [<18.2%] and HCl [<32.7%]).  相似文献

目的:探讨改良内皮抑素(RGDRGD-ES)对人脐静脉内皮细胞(HUVEC)的抑制作用,摸索RGDRGD-ES对HUVEC细胞抑制作用的相对最佳作用浓度和时间。方法:通过快速定点诱变PCR方法获得含有RGDRGD膜序的改良人内皮抑素基因,并构建其原核表达载体。表达、纯化改良内皮抑素(RGDRGD-ES),运用MTT法和流式细胞仪检测RGDRGD-ES对人脐静脉内皮细胞的抑制作用。结果:1.诱变了ES基因,获得了改良的RGDRGD-ES基因,并成功构建其原核表达载体。2.获得了RGDRGD-ES蛋白。3.改良的RGDRGD-ES能够有效抑制人脐静脉内皮细胞的生长(P<0.01);抑制率随着药物浓度(10μg/ml、20μg/ml、30μg/ml)的增加和作用时间(24 h、48 h、72 h)的延长而逐渐增加,具有浓度和时间依赖性(P<0.01);而30μg/ml与40μg/ml、50μg/ml组间、72 h与96 h组间无明显差异(P>0.05)。4.细胞凋亡率(作用24 h)具有药物浓度(10μg/ml、20μg/ml、30μg/ml)依赖性(P<0.01),30μg/ml与40μg/ml、50μg/ml组间凋亡率无明显差异(P>0.05)。结论:成功构建了改良RGDRGD-ES基因的原核表达载体,RGDRGD-ES蛋白在30μg/ml浓度作用72小时条件下能够有效抑制人脐静脉内皮细胞的生长,改良内皮抑素(RGDRGD-ES)对HUVEC的抑制作用较ES明显提高。  相似文献

目的:探究男性精浆中白细胞介素-6(IL-6)和可溶性细胞黏附分子-1(sICAM-1)与免疫性不育的关系。方法:选择2014年6月至2015年12月我院收治不育症患者189例及100例健康体检者为研究对象,根据患者精子混合抗球蛋白反应实验(MAR)结果将不育症患者分为免疫性不育组(88例)和非免疫性不育组(101例);免疫不育组患者按照精液白细胞过氧化物酶染色情况分为免疫性阳性白细胞组(WBC≥1×10~6/mL)36例和免疫性阴性白细胞阴性组(1×10~6/m L)52例,分析并比较各组间精子质量、IL-6和sICAM-1水平。结果:免疫性不育组与非免疫性不育组患者精液精子向前运动比率、存活率均低于对照组(P0.05);免疫性不育组与非免疫性不育组精液各项参数均不存在差异(P0.05);免疫性不育组患者IL-6、sICAM-1水平均高于非免疫性不育组及对照组,差异有统计学意义(P0.05);非免疫性不育组IL-6、sICAM-1水平均高于对照组,差异有统计学意义(P0.05)。相关性分析显示,精子被Ig G黏附数比例与研究对象精液IL-6、sICAM-1水平呈正相关关系(r=0.438,0.561;P0.05);免疫性阳性白细胞组患者精液IL-6与sICAM-1水平均高于免疫性阴性白细胞组(P0.05)。结论:免疫性不育症患者精子质量下降,男性精液中IL-6和sICAM-1水平表达越高,MAR阳性率越高,男性免疫性不育发生的可能性越大。  相似文献

ObjectiveThe optimal steroid regimen in the treatment of subacute thyroiditis (SAT) is controversial. This study aims to compare low- and high-dose steroid regimens in the treatment of SAT.MethodsA single-center, retrospective observational cohort study with up to 1 year of follow-up was conducted. A total of 44 patients in the 16-mg methylprednisolone (MPS) group and 47 patients in the 48-mg MPS group were enrolled. Clinical and laboratory findings from the time of diagnosis to 1-year of the follow-up were assessed. Treatment response, recurrence, and hypothyroidism (HPT) rates were evaluated.ResultsClinical symptoms, sedimentation rates, C-reactive protein, and thyroid hormone levels of the patients were similar in the 2 groups. Recovery was achieved in all patients at the end of the treatments; however, treatment duration needed to be extended for 6 (13.6%) and 1 (2.1%) of the patients in the 16-mg and 48-mg MPS groups, respectively. The 48-mg MPS group had a higher SAT recurrence rate than the 16-mg MPS group (P = .04). Logistic regression analysis suggested that a lower thyroid-stimulating hormone level at the end of the treatment was a predictor of recurrence (β = –0.544, P = .014, 95% CI: 0.376-0.895). While the transient HPT rate was 10 (21.3%) and 10 (22.7%) in the 48-mg and 16-mg MPS groups, respectively, a permanent HPT developed in 5 (10.6%) of patients in the 48-mg MPS and 3 (6.8%) in the 16-mg MPS group. The permanent and transient HPT rates were determined to be similar in the low- and high-dose groups (P > .05).ConclusionLow-dose steroid therapy may be sufficient to achieve a complete recovery and better outcomes in SAT.  相似文献
