During the cheese-making process, water activity (aw) is one of the essential environmental parameters acting on bacterial growth and metabolic pathways. The influence of aw on Streptococcus diacetylactis growth and lactic acid production was studied. The specific growth rate was linearly related to water availability in the milk medium. The cell behaviour was quite different above and below aw=0.95, which can be considered a limiting value. Below this value, the lactic acid production reached 1.4–6.1 mg·g–1, whereas the specific productivity was 2.0–2.6 mg·10–10 cells·h–1. Changes in the consumption of lactose and amino acids during the different growth phases was completely modified by decreasing the water availability in the medium. Correspondence to: N. Summary The growth parameters ofPenicillium cyclopium have been evaluated in a continuous culture system for the production of fungal protein from whey. Dilution rates varied from 0.05 to 0.20 h–1 under constant conditions of temperature (28°C) and pH (3.5). The saturation coefficients in the Monod equation were 0.74 g l–1 for lactose and 0.14 mg l–1 for oxygen, respectively. For a wide range of dilution rates, the yield was 0.68 g g–1 biomass per lactose and the maintenance coefficient 0.005 g g–1 h–1 lactose per biomass, respectively. The maximum biomass productivity achieved was 2 g l–1 h–1 biomass at dilution rates of 0.16–0.17 h–1 with a lactose concentration of 20 g l–1 in the feed. With a cell concentration of 125 g dry biomass 1–1 and a dilution rate of 0.1 h–1,Propionibacterium acidipropionici produces 30 g propionic acid 1–1 from sugar with a productivity of 3 g 1–1 h–1. The yield of propionic acid is approx. 0.36–0.45 g propionic acid g–1 sucrose and is independent of the dilution rate and cell concentration. Acetic acid is an unwanted by-product in the production of propionic acid. The concentration of acetic acid only increases slightly when the cell concentration is increased. A two-stage fermentation process was developed for the conversion of sugar or molasses of various types to propionic acid and vitamin B12. By fermentation of blackstrap molasses (from sugar beet and sugar cane) in the first fermentation stage 17.7 g propionic acid 1–1 with a yield of 0.5 g propionic acid g–1 carbohydrate was produced with a dilution rate of 0.25 h–1. Summary The nitrogen fixation rate in a 4-year-old stand of the woody legumeLeucaena leucocephala (Lam.) de Wit. was estimated in the field at a rather dry site in Tanzania by use of an acetylene reduction technique. The diurnal mean value during April–May was 35 nmol C2H4 mg–1 (dry weight) nodules h–1, with a variation between 22±8 and 48±12 nmol C2H4 mg–1 (dry weight) nodules h–1 in early morning and at midday, respectively. The nodule biomass was determined by auger sampling to be 51±16 kg (dry weight) ha–1. Most of the nodules were found at the 10–30 cm soil depth level. A rough calculation of the amount of nitrogen fixed annually arrived at 110±30 kg ha–1. The results give strong support for the use ofL. leucocephala for soil enrichment in less humid areas of tropical Africa.  相似文献

Summary Optimal growth conditions for Zymomonas mobilis have been established using continuous cultivation methods. Optimal substrate utilization efficiency occurs with 2.5 g l–1 yeast extract, 2.0 g l–1 ammonium sulfate and 6.0 g l–1 magnesium sulfate in the media. Catabolic activity is at its maximum with glucose uptake rates of 16–18 g l–1 h–1 and ethanol production rates of 8–9 g l–1 h–1, Qg values of 22–26 and Qp values between 11 and 13, which results in 40 g l–1 h–1 ethanol yields using a 100 g l–1 substrate feed. Any increase in these parameters goes on cost of substrate utilization efficiency. The capsular polysaccharide (PS) is the most important pneumococcal virulence factor and is currently used as antigen in all pneumococcal vaccines. Despite its physiological and epidemiological importance, meager studies have been devoted to improve PS production and understand its relationship with pneumococcal central metabolism. In this study, kinetics of growth and production of PS by Streptococcus pneumoniae serotype 14 (PS14) in batch and continuous cultivation were investigated. Strong cell lysis was observed in batch cultivation, while accumulation of organic acids and autolysis was avoided in continuous cultivation. In the continuous cultivation was possible to achieve higher concentration of biomass and PS14. Calculation of kinetic parameters demonstrated that PS14 is a cell-associated product. The coefficients for growth-associated stoichiometric true yield and maintenance were determined as 0.25 gglucose gbiomass−1 and 1.24 gglucose (gbiomass h)−1, respectively. The maximum productivity of PS14 released in the supernatant (PS14R) and cell-bound PS14 (PS14C) were obtained at a dilution rate of 0.8 h−1, respectively, 85 and 122 mg  (gbiomass h)−1. Compared to batch fermentation, both PS14R and PS14C productivities were increased by about 300% in the continuous process. These findings demonstrate that continuous cultivation is a promising strategy for PS production to be used in pneumococcal vaccines.  相似文献

The CO2 production of individual larvae of Apis mellifera carnica, which were incubated within their cells at a natural air humidity of 60–80%, was determined by an open-flow gas analyzer in relation to larval age and ambient temperature. In larvae incubated at 34 °C the amount of CO2 produced appeared to fall only moderately from 3.89±1.57 µl mg–1 h–1 in 0.5-day-old larvae to 2.98±0.57 µl mg–1 h–1 in 3.5-day-old larvae. The decline was steeper up to an age of 5.5 days (0.95±1.15 µl mg–1 h–1). Our measurements show that the respiration and energy turnover of larvae younger than about 80 h is considerably lower (up to 35%) than expected from extrapolations of data determined in older larvae. The temperature dependency of CO2 production was determined in 3.5-day-old larvae, which were incubated at temperatures varying from 18 to 38 °C in steps of 4 °C. The larvae generated 0.48±0.03 µl mg–1 h–1 CO2 at 18 °C, and 3.97±0.50 µl mg–1 h–1 CO2 at 38 °C. The temperature-dependent respiration rate was fitted to a logistic curve. We found that the inflection point of this curve (32.5 °C) is below the normal brood nest temperature (33–36 °C). The average Q10 was 3.13, which is higher than in freshly emerged resting honeybees but similar to adult bees. This strong temperature dependency enables the bees to speed up brood development by achieving high temperatures. When an initial cell loading of about 30–40 µg chlorophyll (Chl)·g–1 gel and alginate suspension of 3% (w/v) were used for immobilization of Chlamydomonas reinhardtii, the resulting cell beads showed optimum nitrite uptake rate, at 30° C and pH 7.5, of 14 µmol NO inf2 sup– ·mg–1 Chl·h–1, the photosynthetic and respiratory activities being about 120 µmol O2 produced·mg–1 Chl·h–1, and 40 µmol O2 consumed ·mg–1 Chl·h–1, respectively. The nitrite uptake activity required CO2 in the culture and persisted after 8 days of cells immobilization, or in the presence of 0.2 mm ammonium in the medium. Our data indicate that alginate-entrapped C, reinhardtii cells may provide a stable and functional system for removing nitrogenous contaminants from waste-waters.Correspondence to: C. A fermentation medium based on millet (Pennisetum typhoides) flour hydrolysate and a four-phase feeding strategy for fed-batch production of baker's yeast,Saccharomyces cerevisiae, are presented. Millet flour was prepared by dry-milling and sieving of whole grain. A 25% (w/v) flour mash was liquefied with a thermostable 1,4--d-glucanohydrolase (EC 3.2.1.1) in the presence of 100 ppm Ca2+, at 80°C, pH 6.1–6.3, for 1 h. The liquefied mash was saccharified with 1,4--d-glucan glucohydrolase (EC 3.2.1.3) at 55°C, pH 5.5, for 2 h. An average of 75% of the flour was hydrolysed and about 82% of the hydrolysate was glucose. The feeding profile, which was based on a model with desired specific growth rate range of 0.18–0.23 h–1, biomass yield coefficient of 0.5 g g–1 and feed substrate concentration of 200 g L–1, was implemented manually using the millet flour hydrolysate in test experiments and glucose feed in control experiments. The fermentation off-gas was analyzed on-line by mass spectrometry for the calculation of carbon dioxide production rate, oxygen up-take rate and the respiratory quotient. Off-line determination of biomass, ethanol and glucose were done, respectively, by dry weight, gas chromatography and spectrophotometry. Cell mass concentrations of 49.9–51.9 g L–1 were achieved in all experiments within 27 h of which the last 15 h were in the fedbatch mode. The average biomass yields for the millet flour and glucose media were 0.48 and 0.49 g g–1, respectively. No significant differences were observed between the dough-leavening activities of the products of the test and the control media and a commercial preparation of instant active dry yeast. Candida utilis was grown on a pineapple cannery effluent in a chemostat at dilution rates ranging between 0.05 and 0.65 h–1 to establish optimal conditions for biomass production and chemical oxygen demand (COD) reduction. Sucrose, fructose and glucose were the main sugars in the effluent. Maximum value for cell yield coefficient and productivity were (0.686, gx/gs) and (2.96, gx/l/h) at a dilution rate of 0.425 and 0.475 h–1, respectively, while maximum COD reduction (98%) was attained at a dilution rate of 0.1 h–1. The maintenance coefficient attained a value of (0.093, gs/gx/h). An increase in dilution rate produced a higher protein content of the biomass.  相似文献

Glucose repressed xylose utilization inCandida tropicalis pre-grown on xylose until glucose reached approximately 0–5 g l–1. In fermentations consisting of xylose (93 g l–1) and glucose (47 g l–1), xylitol was produced with a yield of 0.65 g g–1 and a specific rate of 0.09 g g–1 h–1, and high concentrations of ethanol were also produced (25 g l–1). If the initial glucose was decreased to 8 g l–1, the xylitol yield (0.79 g g–1) and specific rate (0.24 g g–1 h–1) increased with little ethanol formation (<5 g l–1). To minimize glucose repression, batch fermentations were performed using an aerobic, glucose growth phase followed by xylitol production. Xylitol was produced under O2 limited and anaerobic conditions, but the specific production rate was higher under O2 limited conditions (0.1–0.4 vs. 0.03 g g–1 h–1). On-line analysis of the respiratory quotient defined the time of xylose reductase induction.  相似文献

An efficient fedbatch process for the production of Lactobacillus kefir DSM 20587 cells was developed. An improvement in space time yield of 270% (3.7 gDCW l–1 day–1) and in final enzyme activity of 440% (9.1 U/ml) was achieved on a 150 l scale by controlling the oxygen transfer rate to 7–9 mmol l–1 h–1. The cells exhibited good and highly stereoselective reducing activities against tert-butyl 6-chloro-3,5-dioxohexanoate. tert-Butyl (3R,5S)-6-chloro-dihydroxyhexanoate, a chiral building block for HMG-CoA reductase inhibitor synthesis, was produced with 47.5% yield and >99% ee at C33 and C55 in a simple batch biotransformation process.  相似文献

3-Hydroxypropionic acid (3-HP) is a platform molecule whose biological production was carried out by the bacterium Limosilactobacillus reuteri according to a two-step process: first, a growth phase in batch mode on glucose, then a glycerol bioconversion into 3-HP in fed-batch mode. With the objective of improving 3-HP bioproduction, this study aimed at defining the operating conditions during the bioconversion phase that increases the bioproduction performance. A central composite rotatable design allowed testing various pH levels and specific glycerol feeding rates. By establishing response surfaces, optimal conditions have been identified that were different depending on the considered output variable (final 3-HP quantity, 3-HP production yield and production rate). Of them, 3-HP final quantity and 3-HP production yield were maximized at pH 6.0 and at specific glycerol feeding rates of 60 and 55 mggly gCDW−1 h−1, respectively. The specific 3-HP production rate was the highest at the upper limit of the specific substrate feeding rate (80 mggly gCDW−1 h−1) but was not affected by the pH. An additional experiment was carried out at pH 6.0 and a specific glycerol feeding rate of 80 mggly gCDW−1 h−1 to validate the previous observations. In conclusion, the results showed a significant improvement of 3-HP concentration by 13%, of specific production rate by 34% and of 3-HP volumetric productivity by 39%, as compared to the initial values.  相似文献

Summary The kinetics ofBordetella pertussis growth was studied in a glutamate-limited continuous culture. Growth kinetics corresponded to Monod's model. The saturation constant and maximum specific growth rate were estimated as well as the energetic parameters, theoretical yield of cells and maintenance coefficient. Release of pertussis toxin (PT) and lipopolysaccharide (LPS) were growth-associated. In addition, they showed a linear relationship between them. Kinetic data of ferrous iron oxidation by Thionacillus ferrooxidans were determined. The aim was to remove H2S (<0.5 ppm) from waste gas by a process proposed earlier. Kinetic data necessary for industrial scale-up were investigated in a chemostat airlift reactor (dilution rate 0.02–0.12 h–1; pH 1.3). Due to the low pH, ferric iron precipitation and wall growth could be avoided. The maximum ferrous iron oxidation rate of submersed bacteria was 0.77 g 1–1 h–1, the maximum specific growth rate about 0.12 h–1 and the yield coefficient was found to be 0.007 g g–1 Fe2+. The specific O2 demand of an exponentially growing, ironoxidizing batch culture was 1.33 mg O2 mg–1 biomass h–1. Summary A study of the ammonium transport system of Zygosaccharomyces bailii was carried out using methylammonium as a non-metabolizable analogue. Benzoic acid in the growth medium decreased the capacity of the transport system from 1.46 ± 0.11 mmol.g–1.h–1 to 0.41±0.04 mmol.g–1.h–1, while the affinity for ammonium was not significatively affected. Although ammonium uptake was inhibited by benzoic acid, the ammonium transport system was still active at preservative concentrations which fully inhibited growth suggesting that inhibition of growth was not governed by the uptake of this nutrient.  相似文献

Summary The metabolic and cardiac responses to temperature were studied in two species (four subspecies) of western chipmunks (genusEutamias), inhabiting boreal and alpine environments. A specially designed (Fig. 1) implantable biopential radiotransmitter was used to measure heart rate in unrestrained animals. The estimated basal metabolic rates (EBMR) were 1.78 (E. minimus borealis), 1.64 (E. m. oreocetes), 1.50 (E. m. operarius), and 1.69 ml O2 g–1 h–1 (E. amoenus luteiventris), or 839, 752, 698, and 628 ml O2 kg–0.75 h–1, respectively, for the four subspecies (Table 1). The two alpine species (E.m.or. andE.m.op.) had significantly lower EBMR than both of their boreal counterparts. The EBMR from all animals are 120–135% of the predicted values based on body weights of the animals. The thermal neutral zone for the four subspecies ranged from 23.5 to 32°C and the minimum thermal conductances were 0.113, 0.111, 0.112 and 0.112 ml O2 g–1 h–1 °C–1, respectively, or 54.4, 54.0, 50.4 and 52.1 ml O2 kg–0.75 h–1 °C–1, respectively (Fig. 2). No interspecific diffence in conductance was observed. These values are 72 to 85% of their weight specific values. The body temperature ranged between 35.0 and 39.5°C and was usually maintained between 36 and 38°C in all subspecies between ambient temperatures of 3 and 32°C. The estimated basal heart rates were 273, 296, 273 and 264 beats/min, respectively, for the four subspecies, 49–55% of their predicted weight specific values. The resultant oxygen pulses (metabolic rate/heart rate) were 5.49, 4.50, 4.48 and 5.56×10–3 ml O2/beat, respectively, which are 2 to 2.4 times their weight specific values (Table 2).The observed reduction of basal heart rate without the corresponding decreases of basal metabolic rate and body temperature indicate sufficient compensatory increases in stroke volume and/or A-V oxygen difference at rest. Summary Submerged batch cultivation under controlled environmental conditions of pH 3.8, temperature 30°C, and KLa200 h–1 (above 180 mMO2 l –1 h–1 oxygen supply rate) produced a maximum (12.0 g·l –1) SCP (Candida utilis) yield on the deseeded nopal fruit juice medium containing C/N ratio of 7.0 (initial sugar concentration 25 g·l –1) with a yield coefficient of 0.52 g cells/g sugar. In continuous cultivation, 19.9 g·l –1 cell mass could be obtained at a dilution rate (D) of 0.36 h–1 under identical environmental conditions, showing a productivity of 7.2 g·l –1·h–1. This corresponded to a gain of 9.0 in productivity in continuous culture over batch culture. Starting with steady state values of state variables, cell mass (CX–19.9 g·l –1), limiting nutrient concentration (Cln–2.5 g·l –1) and sugar concentration (CS–1.5 g·l –1) at control variable conditions of pH 3.8, 30°C, and KLa 200 h–1 keeping D=0.36 h–1 as reference, transient response studies by step changes of these control variables also showed that this pH, temperature and KLa conditions are most suitable for SCP cultivation on nopal fruit juice. Kinetic equations obtained from experimental data were analysed and kinetic parameters determined graphically. Studies were carried out for the production of aroma compounds by Kluyveromyces marxianus grown on cassava bagasse in solid state fermentation using packed bed reactors, testing two different aeration rates. Respirometric analysis was used to follow the growth of the culture. Headspace analysis of the culture by gas chromatography showed the production of 11 compounds, out of which nine were identified. Ethyl acetate, ethanol and acetaldehyde were the major compounds produced. Lower aeration rate (0.06l h–1 g–1 of initial dry matter) increased total volatile (TV) production and the rate of production was also increased at this aeration rate. Using an aeration rate of 0.06l h–1 g–1 maximum TV concentrations were reached at 24 h and at 40 h with 0.12l h–1 g–1.  相似文献
