Summary Sequence comparisons were made for up to 667 bp of DNA cloned from 14 kinds of HawaiianDrosophila and five other dipteran species. These sequences include parts of the genes for NADH dehydrogenase (subunits 1, 2, and 5) and rRNA (from the large ribosomal subunit). Because the times of divergence among these species are known approximately, the sequence comparisons give insight into the evolutionary dynamics of this molecule. Transitions account for nearly all of the differences between sequences that have diverged by less than 2%; for these sequences the mean rate of divergence appears to be about 2%/Myr. In comparisons involving greater divergence times and greater sequence divergence, relatively more of the sequence differences are due to transversions. Specifically, the fraction of these differences that are counted as transversions rises from an initial value of less than 0.1 to a plateau value of nearly 0.6. The time required to reach half of the plateau value, about 10 Myr, is similar to that for mammalian mtDNA. The mtDNAs of flies and mammals are also alike in the shape of the curve relating the percentage of positions at which there are differences in protein-coding regions to the time of divergence. For both groups of animals, the curve has a steep initial slope ascribable to fast accumulation of synonymous substitutions and a shallow final slope resulting from the slow accumulation of substitutions causing amino acid replacements. However, the percentage of all sites that can experience a high rate of substitution appears to be only about 8% for fly mtDNA compared to about 20% for mammalian mtDNA. The low percentage of hypervariable sites may be a consequence of a functional constraint associated with the low content of guanine and cytosine in fly mtDNA.  相似文献

The relevance of three-dimensional structures of proteins, determined by X-ray crystallography, is an important issue that is becoming even more critical in light of the Structural Genomics Initiative. As a case study, a detailed comparison of functional properties of the T quaternary states of genetically or chemically modified human hemoglobins (Hbs) in solution and in the crystal was performed. Oxygen affinities of Hbs in crystals correlate with the rate constants of their initial combination with carbon monoxide (CO) in solution, indicating that changes in ligand affinity caused by the modifications are similarly observed in both physical states.  相似文献

Summary By using complete sequence data of mitochondrial DNAs, three Markov models (Day-hoff, Proportional, and Poisson models) for amino acid substitutions during evolution were applied in maximum likelihood analyses of mitochondrially encoded proteins to estimate a phylogenetic tree depicting human, cow, whale, and murids (mouse and rat), with chicken, frog, and carp as outgroups. A cow/whale clade was confirmed with a more than 99.8% confidence level by any of the three models, but the branching order among human, murids, and the cow/whale clade remained uncertain. It turned out that the Dayhoff model is by far the most appropriate model among the alternatives in approximating the amino acid substitutions of mitochondrially encoded proteins, which is consistent with a previous analysis of a more limited data set. It was shown that the substitution rate of mitochondrially encoded proteins has increased in the order of fishes, amphibians, birds, and mammals and that the rate in mammals is at least six times, probably an order of magnitude, higher than that in fishes. The higher evolutionary rate in birds and mammals than in amphibians and fishes was attributed to relaxation of selective constraints operating on proteins in warm-blooded vertebrates and to high mutation rate of bird and mammalian mitochondrial DNAs.Offprint requests to: M. The present study was performed to investigate the influence of the intake of selective oestrogen receptor modulators on the urinary endogenous steroids profile. For this purpose the circadian variability of luteinizing hormone, follicle-stimulating hormone, testosterone, 5α-androstan-3α,17β-diol, 5β-androstan-3α,17β-diol, epitestosterone, 4-androstenedione, androsterone and etiocholanolone were measured on eight subjects (four males and four females) by gas chromatography–mass spectrometry and chemiluminescent immunometric assay techniques before and after oral administration of multiple doses of either tamoxifen (80 mg for 2 days) or toremifene (120 mg for 2 days) or clomiphene (100 mg for 2 days). The individual baseline variability of the steroids studied was set up by collecting the urine samples every 3 h, for 3 days prior to the treatment; whereas the evaluation of the effects of the oral administration of multiple doses of selective oestrogen receptor modulators on the steroid urinary profile was assessed by collecting urine samples every three hours for at least five days from the first administration.The results of our measurements showed that, only in male subjects, the relative urinary concentrations of testosterone, epitestosterone and 4-androstenedione were significantly altered generally after the second day of drug administration. While no significant effects were recorded in both sexes on the luteinizing hormone, follicle-stimulating hormone, androsterone, etiocholanolone, 5α-androstan-3α,17β-diol and 5β-androstan-3α,17β-diol urinary levels and on testosterone/epitestosterone, 5α-androstan-3α,17β-diol/5β-androstan-3α,17β-diol and androsterone/etiocholanolone ratios.  相似文献

The total protein concentrations in the hemolymph of noninfected Biomphalaria glabrata and those parasitized by Schistosoma mansoni have been compared. That of the former, averaging 1.42 g/100 ml, is greater than that of the latter which averages 0.96 g/100 ml. The hemoglobin concentrations in the two categories of snails have also been compared and that in noninfected specimens, averaging 0.956 g/100 ml, is higher than that in infected snails which average 0.650 g/100 ml. It has also been ascertained that both the total protein and hemoglobin concentrations in snails of different sizes vary; specimens measuring between 10 and 16 mm in diameter tend to include more hemoglobin than larger ones. On the other hand, larger snails tend to include greater total protein concentrations.  相似文献

The complete mitochondrial genome sequence of the Chinese hook snout carp, Opsariichthys bidens, was newly determined using the long and accurate polymerase chain reaction method. The 16,611-nucleotide mitogenome contains 13 protein-coding genes, two rRNA genes (12S, 16S), 22 tRNA genes, and a noncoding control region. We use these data and homologous sequence data from multiple other ostariophysan fishes in a phylogenetic evaluation to test hypothesis pertaining to codon usage pattern of O. bidens mitochondrial protein genes as well as to re-examine the ostariophysan phylogeny. The mitochondrial genome of O. bidens reveals an alternative pattern of vertebrate mitochondrial evolution. For the mitochondrial protein genes of O. bidens, the most frequently used codon generally ends with either A or C, with C preferred over A for most fourfold degenerate codon families; the relative synonymous codon usage of G-ending codons is greatly elevated in all categories. The codon usage pattern of O. bidens mitochondrial protein genes is remarkably different from the general pattern found previously in the relatively closely related zebrafish and most other vertebrate mitochondria. Nucleotide bias at third codon positions is the main cause of codon bias in the mitochondrial protein genes of O. bidens, as it is biased particularly in favor of C over A. Bayesian analysis of 12 concatenated mitochondrial protein sequences for O. bidens and 46 other teleostean taxa supports the monophyly of Cypriniformes and Otophysi and results in a robust estimate of the otophysan phylogeny.  相似文献

Hybridization may threaten the conservation status of rare species through genetic assimilation and may confound the ability to distinguish among taxa. We studied these issues in an endangered shrub, Purshia subintegra (Rosaceae), known from four populations growing on limestone outcrops in central Arizona (USA). Using amplified fragment length polymorphisms (AFLP) and the Bayesian clustering algorithm implemented in STRUCTURE, we identified three distinct genetic lineages among Arizona Purshia subintegra and P. stansburiana. An initial split divided San Carlos Basin P. subintegra (considered P. pinkavae by Schaack) from northern P. stansburiana populations (FST = 0.394). A subsequent split separated northern P. stansburiana from two P. subintegra populations at Horseshoe Lake and Burro Creek (FST = 0.207), which comprised a nearly perfect admixture of the two lineages identified in the initial analysis. In the Verde River Valley P. subintegra is sympatric with P. stansburiana and exhibited an average 27% P. stansburiana genes for 5 of 6 stands analyzed, indicating ongoing hybridization and backcrossing with P. subintegra. Individuals carrying >90% P. subintegra markers are identifiable 68% of the time based on morphology, with leaf lobing, leaf size, and leaf length acting as the most reliable indicators of taxonomic status. However, the genetic and morphological distance correlation among individuals was low (r = 0.17, P = 0.0002), indicating that morphology cannot always accurately predict genetic admixture or taxonomy. Overall, our study confirmed the genetic distinctiveness of the San Carlos Basin population, an ancient natural hybrid origin of P. subintegra, and the presence of a hybrid swarm in the Verde Valley, whose conservation value may lie in its heightened genetic diversity.  相似文献

Summary In phenotype the mycoplasmas are very different from ordinary bacteria. However, genotypically (i.e., phylogenetically) they are not. On the basis of ribosomal RNA homologies the mycoplasmas belong with the clostridia, and indeed havespecific clostridial relatives. Mycoplasmas are, however, unlike almost all other bacteria in the evolutionary characteristics of their ribosomal RNAs. These RNAs contain relatively few of the highly conserved oligonucleotide sequences characteristic of normal eubacterial ribosomal RNAs. This is interpreted to be a reflection of an elevated mutation rate in mycoplasma lines of descent. A general consequence of this would be that the variation associated with a mycoplasma population is augmented both in number and kind, which in turn would lead to an unusual evolutionary course, one unique in all respects. Mycoplasmas, then, are actually tachytelic bacteria. The unusual evolutionary characteristics of their ribosomal RNAs are the imprints of their rapid evolution.  相似文献

Species level phylogenetic hypotheses can be used to explore patterns of divergence and speciation. In the tropics, speciation is commonly attributed to either vicariance, perhaps within climate-induced forest refugia, or ecological speciation caused by niche adaptation. Mimetic butterflies have been used to identify forest refugia as well as in studies of ecological speciation, so they are ideal for discriminating between these two models. The genus Ithomia contains 24 species of warningly colored mimetic butterflies found in South and Central America, and here we use a phylogenetic hypothesis based on seven genes for 23 species to investigate speciation in this group. The history of wing color pattern evolution in the genus was reconstructed using both parsimony and likelihood. The ancestral pattern for the group was almost certainly a transparent butterfly, and there is strong evidence for convergent evolution due to mimicry. A punctuationist model of pattern evolution was a significantly better fit to the data than a gradualist model, demonstrating that pattern changes above the species level were associated with cladogenesis and supporting a model of ecological speciation driven by mimicry adaptation. However, there was only one case of sister species unambiguously differing in pattern, suggesting that some recent speciation events have occurred without pattern shifts. The pattern of geographic overlap between clades over time shows that closely related species are mostly sympatric or, in one case, parapatric. This is consistent with modes of speciation with ongoing gene flow, although rapid range changes following allopatric speciation could give a similar pattern. Patterns of lineage accumulation through time differed significantly from that expected at random, and show that most of the extant species were present by the beginning of the Pleistocene at the latest. Hence Pleistocene refugia are unlikely to have played a major role in Ithomia diversification.  相似文献

The present study examined the temporal pattern of responding in a conditioned bar-press suppression task in rats. Rats were exposed to either a 30-s or a 120-s conditioned stimulus (CS) followed by a footshock. Training took place either while the rats were lever-pressing for water (online), or with the lever removed from the box (offline). They were then exposed to the CS while they were lever-pressing for water, either in the training context or in a different context. Bar-press suppression during the CS was constant across the duration of the CS during training, but was restricted to the initial portion of the CS at the time of testing, especially when subjects were tested in a different context. Those results replicate the reactive (as opposed to anticipatory) pattern observed in a lick suppression procedure by Jozefowiez et al. (2011) and indicate that a change in context at the time of testing might be critical for its expression.  相似文献

The geographic distribution of phenotypic variation among closely related populations is a valuable source of information about the evolutionary processes that generate and maintain biodiversity. Leapfrog distributions, in which phenotypically similar populations are disjunctly distributed and separated by one or more phenotypically distinct populations, represent geographic replicates for the existence of a phenotype, and are therefore especially informative. These geographic patterns have mostly been studied from phylogenetic perspectives to understand how common ancestry and divergent evolution drive their formation. Other processes, such as gene flow between populations, have not received as much attention. Here, we investigate the roles of divergence and gene flow between populations in the origin and maintenance of a leapfrog distribution in Phyllobates poison frogs. We found evidence for high levels of gene flow between neighbouring populations but not over long distances, indicating that gene flow between populations exhibiting the central phenotype may have a homogenizing effect that maintains their similarity, and that introgression between ‘leapfroging’ taxa has not played a prominent role as a driver of phenotypic diversity in Phyllobates. Although phylogenetic analyses suggest that the leapfrog distribution was formed through independent evolution of the peripheral (i.e. leapfrogging) populations, the elevated levels of gene flow between geographically close populations poise alternative scenarios, such as the history of phenotypic change becoming decoupled from genome‐averaged patterns of divergence, which we cannot rule out. These results highlight the importance of incorporating gene flow between populations into the study of geographic variation in phenotypes, both as a driver of phenotypic diversity and as a confounding factor of phylogeographic inferences.  相似文献

Life-history models for populations in a single patch, in which density dependence occurs through competition between seedlings for safe-sites, suggest that timing of flowering in monocarpic perennials is such that expected lifetime reproductive success (R 0) is maximised. We discuss metapopulation models in which local populations go extinct either (1) because all rosettes die locally, or (2) because seedling recruitment is limited to restricted periods in time. In both cases there is selection for shorter optimal generation times than suggested by the single-patch model. The mechanism is that in young populations competition between seedlings for safe sites is relaxed for some years. This mostly benefits types with short generation times. Carlina vulgaris flowers earlier than the single-patch model suggests. The metapopulation effect is sufficiently strong to account for the differences but other factors cannot be outruled. Data on other monocarpic perennials are discussed. Flowering in Cirsium vulgare is also earlier than suggested by the single-patch model, but for other species the picture is far from clear. Co-evolution between phenotypic variation and other traits is of paramount importance for our understanding of the origin and maintenance of polymorphism in natural populations. We tested whether the evolution of plumage polymorphism in birds of prey and owls was supported by the apostatic selection hypothesis using ecological and life-history variables in birds of prey and owls and performing both cross taxa and independent contrast analyses. For both bird groups, we did not find any support for the apostatic selection hypothesis being the maintaining factor for the polymorphism: plumage polymorphism was not more common in taxa hunting avian or mammalian prey, nor in migratory species. In contrast, we found that polymorphism was related to variables such as sexual plumage dimorphism, population size and range size, as well as breeding altitude and breeding latitude. These results imply that the most likely evolutionary correlate of polymorphism in both bird groups is population size, different plumage morphs might simply arise in larger populations most likely because of a higher probability of mutations and then be maintained by sexual selection.  相似文献

Inferences on the evolution of human speech based on anatomical data must take into account its physiology, acoustics and perception. Human speech is generated by the supralaryngeal vocal tract (SVT) acting as an acoustic filter on noise sources generated by turbulent airflow and quasi-periodic phonation generated by the activity of the larynx. The formant frequencies, which are major determinants of phonetic quality, are the frequencies at which relative energy maxima will pass through the SVT filter. Neither the articulatory gestures of the tongue nor their acoustic consequences can be fractionated into oral and pharyngeal cavity components. Moreover, the acoustic cues that specify individual consonants and vowels are “encoded”, i.e., melded together. Formant frequency encoding makes human speech a vehicle for rapid vocal communication. Non-human primates lack the anatomy that enables modern humans to produce sounds that enhance this process, as well as the neural mechanisms necessary for the voluntary control of speech articulation. The position and shape of thermal performance curves (TPCs, the functions relating temperature to physiological performance) for ecologically relevant functions will directly affect the fitness of ectotherms and therefore should be under strong selection. However, thermodynamic considerations predict that relationships between the different components of the TPC will confound its evolutionary optimization. For instance, the “jack-of-all-temperatures” hypothesis predicts a trade-off between the breadth of the TPC and the maximal performance capacity; the “warmer is better” hypothesis suggests that low thermal optima will come with low absolute performances. Semi-aquatic organisms face the additional challenge of having to adjust their TPCs to two environments that are likely to differ in mean temperature and thermal variability. In this paper, we examine how parameters of the TPCs for maximal running and swimming speed have co-evolved in the semi-aquatic newt genus Triturus. We consider evolutionary relationships between the width and the height of the TPCs, the optimal temperatures and maximal performance. Phylogenetic comparative analyses reveal that in Triturus, swimming and running differ substantially in the (co-)variation of TPC parameters. Whereas evolutionary changes in the TPC for swimming primarily concern the shape of the curve (generalist versus specialist), most interspecific variation in running speed TPCs involves shifts in overall performance across temperatures.  相似文献

有证据表明昆虫气味结合蛋白(odorant binding proteins, OBPs)与其嗅觉识别密切相关,起着运输外界脂溶性气味分子通过嗅觉感器淋巴液到达嗅觉受体的关键作用.为了更好地了解OBPs在苜蓿盲蝽Adelphocoris lineolatus (Goeze)嗅觉识别中的作用,本研究首次克隆了苜蓿盲蝽气味结合蛋白基因Alin-OBP1 (GenBank序列号GQ477022). 测序和序列分析结果表明,该基因开放阅读框长438 bp, 编码145个氨基酸,预测分子量为15.69 kDa,等电点为5.01,N-末端疏水区包含由18个氨基酸组成的信号肽.蛋白特征分析表明,该基因翻译后的蛋白质具有昆虫气味结合蛋白的典型特征,即氨基酸序列中有6个保守的半胱氨酸残基.利用RT-PCR和Real-time PCR技术对Alin OBP1在苜蓿盲蝽成虫不同组织和各个发育阶段的表达水平进行了测定,结果显示Alin-OBP1几乎全部在触角中表达.不同发育阶段Alin-OBP1表达量不同,在5龄若虫和成虫阶段表达水平最高.结果提示Alin-OBP1可能在苜蓿盲蝽感受包括性信息素在内的外界化合物的过程中发挥着重要作用.  相似文献

Summary Autosomal chorion geness18, s15, ands19 are shown to diverge at extremely rapid rates in closely related taxa of HawaiianDrosophila. Their nucleotide divergence rates are at least as fast as those of intergenic regions that are known to evolve more extensively between distantly related species. Their amino acid divergence rates are the fastest known to date. There are two nucleotide replacement substitutions for every synonymous one. The molecular basis for observed length and substitution mutations is analyzed. Length mutations are strongly associated with direct repeats in general, and with tandem repeats in particular, whereas the rate for an average transition is twice that for an average transversion.The DNA sequence of the cluster was used to construct a phylogenetic tree for five taxa of the Hawaiian picture-winged species group ofDrosophila. Assignment of observed base substitutions occurring in various branches of the tree reveals an excess of would-be homoplasies in a centrally localized 1.8-kb segment containing thes15 gene. This observation may be a reflection of ancestral excess polymorphisms in the segment. The chorion cluster appears to evolve at a constant rate regardless of whether the central 1.8-kb segment is included or not in the analysis. Assuming that the time of divergence ofDrosophila grimshawi and theplanitibia subgroup coincides with the emergence of the island of Kauai, the overall rate of base substitution in the cluster is estimated to be 0.8% million years, whereas synonymous sites are substituted at a rate of 1.2%/million years.  相似文献

化学感受蛋白(chemosensory proteins, CSPs)是昆虫化学感受系统中重要的组成部分之一。本研究克隆了中华蜜蜂Apis cerana cerana化学感受蛋白基因Acer-CSP1, 其核苷酸全长351 bp (GenBank登录号为FJ157352), 编码116个氨基酸残基, 预测蛋白分子量为13.85 kD, 等电点为4.89, 且含有4个保守的半胱氨酸残基, 均符合昆虫CSPs的一般特征, 且与意蜂CSP1基因具有99.1%的相似性, 与其他昆虫也有45.3%~68.0%的相似性。利用2-ΔΔCt法及绝对定量法的real-time PCR技术对Acer-CSP1在中蜂不同器官表达特征进行了研究, 得出的一致结论为Acer-CSP1显著水平地高丰度表达于中华蜜蜂触角, 其次大量表达于头部。由于触角为中华蜜蜂最主要的嗅觉器官, 而头部则具有发达的感觉神经系统和味觉系统, 这也提示Acer-CSP1极有可能参与中华蜜蜂的嗅觉以及其他化学感受功能。  相似文献

The env gene of SIV and HIV-1 encodes a single glycoprotein gp 160, which is processed to give a noncovalent complex of the soluble glycoprotein gp120 and the transmembrane glycoprotein gp41. The extracellular region (ectodomain), minus the N-terminal fusion peptide, of gp41 from HIV-1 (residues 27-154) and SIV (residues 27-149) have been expressed in Escherichia coli. These insoluble proteins were solubilized and subjected to a simple purification and folding scheme, which results in high yields of soluble protein. Purified proteins have a trimeric subunit composition and high alpha-helical content, consistent with the predicted coil-coil structure. SIV gp41 containing a double cysteine mutation was crystallized. The crystals are suitable for X-ray structure determination and, preliminary analysis, together with additional biochemical evidence, indicates that the gp41 trimer is arranged as a parallel bundle with threefold symmetry.  相似文献
