Bracomorpha ninghais sp. n. A new method was developed for loading the pinewood nematode, Bursaphelenchus xylophilus (Steiner et Buhrer) Nickle, on the beetle Monochamus alternatus Hope. Postdiapause beetle larvae were sterilized with 70 and 99.9% aqueous ethanol and placed singly in flasks where B. xylophilus reproduced on the fungus Ophiostoma minus (Hedgcock) H. et P. Sydow that had been grown on autoclaved barley grain and Pinus densifiora Sieb. et Zucc. wood chips. The fungus produced a large nematode population that developed to a high proportion of third-stage dispersal juveniles that molted to the fourth-stage dispersal juveniles. The survival rate was 80%, and the mean nematode load was 10,096. It took a mean of 5 wk to obtain the nematode-infested beetles after the initiation of nematode rearing.  相似文献

Inhibitory effects of Bursaphelenchus mucronatus on the number of B. xylophilus carried by an adult Monochamus alternatus were investigated using artificial pupal chambers. When pupal chambers were infested with either B. xylophilus or B. mucronatus, the load of B. xylophilus onto the beetle was greater (P < 0.001) than that of B. mucronatus. However, within the pupal chamber there was no difference in the abundance of the third-stage dispersal juveniles, which would molt to the fourth-stage dispersal juveniles to board beetles. The nematode load on beetles that emerged from pupal chambers infested with both Bursaphelenchus species was smaller (P = 0.015) than that of beetles with B. xylophilus alone but greater (P < 0.001) than that of beetles with B. mucronatus alone, suggesting an inhibitory effect of B. mucronatus. As a result of this study, the rate of inhibition of B. mucronatus on molting of third-stage dispersal juveniles of B. xylophilus to fourth-stage dispersal juveniles was 0.65, which resulted in great inhibition on boarding beetles at a rate of 0.7.  相似文献

Dastarcus helophoroides adults, a coleopteran parasitoid of Monochamus alternatus, were reared in the laboratory. The objectives of this paper are to investigate the host foraging behavior of D. helophoroides, and the evidence for learning in such behaviors. Choice and no-choice experiments were carried out. The choice experiments show that the number of D. helophoroides adults choosing the Pinus massoniana log infested with M. alternatus larvae was significantly higher than that choosing the blank (a flavorless simulative log made of brown kraftpaper), and the number of D. helophoroides adults choosing the uninfested P. massoniana log was also significantly higher than that choosing the blank. The no-choice experiment shows no significant difference between the number of D. helophoroides adults that chose the infested log and that chose the uninfested log, despite previous experience with uninfested log, infested log, or neither of them. No evidence of learning in host foraging behavior in D. helophoroides adults was found. The results indicate that the volatile chemicals emitted by uninfested P. massoniana logs and infested logs with M. alternatus larvae are equally attractive to D. helophoroides.  相似文献

The presence of alien invasive species has serious negative impact on endemic biodiversity, especially on native species that occupy the same niche in the ecosystem. To study the influence of the alien invasive species Bursaphelenchus xylophilus on its native sister species B. mucronatus, the two nematode species were mix-cultured in a fungal mat and mix-inoculated into a susceptible host. By comparing the propagation parameters of both species under competitive and noncompetitive conditions it was shown that the propagation level of B. xylophilus was clearly higher than that of B. mucronatus under laboratory culture. Furthermore, the propagation capacity of B. xylophilus under competitive conditions was much higher than that under noncompetitive conditions, both in laboratory culture and with host inoculation. Bursaphelenchus xylophilus also excluded B. mucronatus when the two species were cultured as a mixture for a longer time. The relative abundance ratios of the two species in natural pinewoods were also determined by random sampling of dying pine trees from regions with different invasion histories. It was noted that with an increase in invasion years the distribution frequency of B. xylophilus increased while that of B. mucronatus decreased. Experimental tests verified our hypothesis that because of its high fecundity and strong competitive ability, the invasive species B. xylophilus out-competed the native species B. mucronatus and displaced it in natural ecosystems. The successful invasion of B. xylophilus is attributed to competitive displacement, which may be one of the ecological invasive mechanisms.  相似文献

Allophycocyanin (APC) is a minor component of phycobiliproteins in cyanobacteria and red algae. This paper describes a simple and inexpensive extracting method for isolating APC from Spirulina (Arthrospira) platensis with high efficiency. The crude phycobiliprotein extract was pretreated by ammonium sulfate fractionation. Then, by adding hydroxylapatite into crude phycobiliprotein extract dissolved in 20 mM phosphate buffer (pH 7.0), APC was selectively adsorbed by hydroxylapatite but C-phycocyanin (C-PC) was not. The hydroxylapatite was collected and APC was extracted from the crude phycobiliprotein extract. Then, the enriched APC was washed off from the hydroxylapatite using 100 mM phosphate buffer (pH 7.0). In this simple extracting method it was easy to remove C-PC and isolate APC in large amounts. The absorbance ratio A 650/A 280 of extracted APC reached 2.0. The recovery yield was 70%, representing 4.61 mg · g−1 wet weight. The extracted APC could be further purified by a simple anion-exchange chromatography with a pH gradient from 5.6 to 4.0. The absorbance ratio A 650/A 280 of the purified APC reached 5.0, and the overall recovery yield was 43%, representing 2.83 mg · g−1 wet weight. Its purity was confirmed by native polyacrylamide gel electrophoresis (PAGE) and sodium dodecyl sulfate-PAGE.  相似文献

Columba livia is an important reservoir and carrier of Cryptococcus neoformans, Cryptococcus uniguttulatus, Cryptococcus laurentii and Cryptococcus albidus. Upper digestive tract of this species is also known as a habitat for Cryptococcus neoformans. Given the increasing clinical interest of this microorganism, 331 swabs from crop and 174 dropping samples from pigeon lofts in Grand Canary Island have been studied. The obtained results show an extensive presence samples 81 positive (24.47%) of Cryptococcus spp. in analysed crops: 32 (9.66%) for C. neoformans, 24 (7.2%) for C. uniguttulatus, 23 (6.9%) for C. albidus and 2 (0.6%) for C. laurentii. In the same way, Cryptococcus spp was also isolated in 82 (47.13%), dropping samples: C. neoformans in 59 (33.9%), C. uniguttulatus, in 9 (5.17%), C. laurentii in 8 (4.59%) and C. albidus in 6 (3.44%) of the investigated samples, respectively. The cryptococcosis produced by species of cryptococci other than C. neoformans has become more important during the last decade, supporting the study on the role of pigeon in the epidemiology of this disease.  相似文献

Following a re-examination of the material treated under Barleria brevispina (Fiori) Hedrén in the recent Flora of Somalia account of the Acanthaceae, it is concluded that two distinct species are involved and Barleria compacta Malombe & I. Interaction of Bdellovibrio bacteriovorus 100NCJB with bacteria Campylobacter jejuni (strains 1, 2, 3, 4, and 5) and Helicobacter pylori, strain TX30a, was confirmed. The results indicate that lytic activity of bdellovibrios both in liquid media and cells attached to a surface was observed. The potential use of the antimicrobial activity of predatory bacteria for environmental bioprotection and public health is discussed.  相似文献
