Anaerobic co-digestion of grease trap and sewage sludge from a wastewater treatment plant is evaluated. Enzyme-lipase application, both addition and dosage, are evaluated by fitting the methane production of biochemical potential tests with the first order model. The enzyme addition effect, at 2, 5 and 10% of grease trap (%GT VSFED?1) and the enzymes doses, between 0.25 and 1.67% (v/v), without and with grease trap presence were studied. Grease trap addition showed a negative effect on the waste biodegradability, which was completely overcome by the addition of lipase. Enzyme addition improved notably the methane production for all grease trap fractions studied. In regards to the dosage, the best result was achieved between 0.33 and 0.83% (v/v) of enzyme. The co-digestion of sewage sludge and grease trap may be a feasible process by using lipases due to the saving in operational costs and the increase in the biogas production  相似文献

Electric field induced conformational changes of bacteriorhodopsin were studied in six types of dried film (randomly and electrically oriented membranes of purple as well as cation-depleted blue bacteriorhodopsin) by measuring the frequency dependence of the optical absorbance change and the dielectric dispersion and absorption. For the purple bacteriorhodopsin the optical absorbance change induced by alternating rectangular electric fields of ±300 kV/cm altered the sign twice in the frequency range from 0.001 Hz to 100 kHz (around 0.03 Hz and 100 kHz), indicating that the electric field induced conformational change in these samples consists of, at least, three steps. Similarly, it was found for the blue bacteriorhodopsin that at least two steps are involved. In accord with optical measurements, the dielectric behaviour due to alternating sinusoidal electric fields of±6kV/cm in the frequency range from 10 Hz to 10 MHz showed two broad dispersion/absorption regions, one below 1 kHz and the other around 10–100 kHz. This suggests that the conformational change of bacteriorhodopsin is also reflected by its dielectrical properties and that it is partially induced at 6 kV/cm. Including previous results obtained by analysis of the action of DC fields on purple membrane films, a model for a field-induced cyclic reaction for purple as well as blue bacteriorhodopsin is proposed. In addition it was found that there are electrical interactions among purple membrane fragments in dried films.  相似文献

Electric fields induced in a conductive body by the magnetic field of a current-carrying wire were analyzed theoretically and experimentally to assess the dosimetric importance of highly nonuniform, field-exposure conditions. Experimentation revealed that a 60-Hz magnetic field was inversely proportional to the radius of a wire bundle carrying 100 A within a 0.5-m2 test area. A miniaturized electric field probe was used to measure the electric fields induced in 5-cm-deep, saline-filled models. In the theoretical analysis, numerical estimates of induced fields were made by a spreadsheet method. The theoretical calculations and the measured values of induced electric fields were generally in good agreement. The induced fields were in a plane perpendicular to a vertically incident magnetic field; the maximally induced fields were in areas nearest the wire bundle. The strength of the induced field increased with model size: from 96 microV/cm in a 10 x 10 cm model to 176 microV/cm in a 40 x 40 cm model. The strength of the field induced in a 20 x 20 cm model decreased with increasing model-to-wire spacing: from 132 microV/cm for a 1-cm spacing (2-mT maximum, incident field) to 50 microV/cm for a 6-cm spacing (0.33-mT maximum). The results indicate that increases in local values of nonuniformly incident fields produce relatively small increases in induced electric fields. This finding may be important in dosimetric consideration of circumstances, such as use of electric blankets, in which fields of low average strength are accompanied by intense local fields.  相似文献

A 5‐year field survey examined western corn rootworm (WCR) (Diabrotica virgifera virgifera LeConte) beetle density in Serbia from 2002 to 2006. First‐, second‐, third‐, fourth‐ and fifth‐year maize fields were sampled; they represented 64.61%, 21.66%, 9.45%, 3.53% and 0.75% of all sampled fields respectively. Results showed that the mean WCR beetle population density from 794 maize fields differed depending on cropping history. Minimum mean WCR/trap/day was 0.0 in the first‐year maize fields in 2002 and 2006. Maximum mean WCR/trap/day was registered in the fourth‐year and the fifth‐year maize fields (27.8 and 21.2 respectively). Mean population density of WCR adults increased with the number of years of continuous maize from 1.17, 4.61, 6.41, 10.30 up to 13.53 WCR/trap/day for first‐fifth‐year maize fields respectively. Mean WCR/trap/day ± SE exceeded the economic population threshold of >6 WCR/trap/day in third‐year continuous maize fields. Out of 794 maize fields, 697 (87.78%) registered a mean population density below the <6 beetles/trap/day threshold. In only 97 fields was WCR population density >6 beetles/trap/day, a finding that predicts a risk of economic damage to a subsequent maize planting. These data are representative of the Serbian situation from 2002 to 2006; they indicate that WCR are well dispersed across commercial maize fields in Serbia. These results provide new insight into the current low WCR population densities in maize fields managed by crop rotation, a finding that can help in creating long‐term management strategy.  相似文献

The nitric oxide synthase inhibitor N(G)-nitro-L-arginine methyl ester (l-NAME) increased vascular resistance (VR) 10% above baseline of 3.08+/-0.08 (n=11) mmHg/mL/min at 10 mg/kg and 20% above 3.05+/-0.08 (n=9) at 50 mg/kg in anesthetized toads (Bufo marinus). Blood pressure was unaffected by either dose of L-NAME. Blood flow decreased at the higher dose of L-NAME. L-arginine (300 mg/kg) reversed the effects of L-NAME on VR and blood flow in toads treated with 10 mg/kg but not with 50 mg/kg. Injection of 50 mg/kg L-NAME into empty-bladder toads produced a 10% decrease in water uptake, J(v), resulting in a J(v) of 1,267+/-11 cm(3)/cm(2)/s x 10(-7) (n=9) compared to 1,385+/-12 (n=8) for controls. Injection of 10 microg/kg angiotensin II (ANG II) increased J(v) 15% across the pelvic patch (J(v), cm(3)/cm(2)/s x 10(-7)), resulting in a J(v) of 1,723+/-12 cm(3)/cm(2)/s x 10(-7) (n=8) compared to 1,471+/-12 (n=8) for controls. It is hypothesized that during cutaneous drinking blood flow into the capillary bed of the pelvic patch is regulated by nitric oxide and ANG II.  相似文献

Agriotes obscurus (L.) wireworms assembled in increasing numbers at rows of treated (Agrox DL Plus seed treatment) and untreated wheat, Triticum aestivum L., planted at increasing densities (0, 0.15, 0.30, and 0.60 seeds/cm). In treated wheat plots at all planting densities, no wireworm damage to seedlings was apparent, and total wireworms taken in core samples in wheat rows increased according to the asymptotic equation y = B0(1 -e(-Blx)), where B0 is the asymptote, B1 is the slope of the initial rise, and x is the seeding density. The number of dead wireworms in treated plots increased linearly and intercepted the asymptotic models (theoretical point at which 100% mortality of assembled population occurs) at 0.95 seeds/cm on 11 June and 1.14 seeds/cm on 18 June 1996. Untreated wheat at all densities planted had severe wireworm damage and significantly reduced stand. Populations that had assembled at the surviving untreated wheat were fewer than in the treated wheat plots, and although increasing with seeding density, did not follow the asymptotic model. The data suggest that A. obscurus populations can be assembled and killed in fallowed fields in large numbers at treated trap crops of wheat over a 19-d period when planted in rows spaced 1 m apart at a linear seeding density of 1.5 seeds/cm.  相似文献

Transgenic corn expressing Bacillus thuringiensis Berliner (Bt corn) (Maximizer and Yieldgard hybrids, Novartis Seeds), non-Bt isolines and high-performance (check) hybrids were evaluated for European corn borer, Ostrinia nubilalis (Hübner), damage and grain yield in commercial strip plots across Ontario in 1996 and 1997. Bt corn hybrids reduced stalk tunneling damage by 88-100%. In 1996, minimal damage was found in locations where only one generation of European corn borer occurred per year. Bt corn proved its greatest potential for reducing the number and length of cavities below the primary ear in locations where two generations of European corn borer were present. A yield response to using Bt hybrids only occurred when levels of tunneling damage exceeded 6 cm in length. European corn borer infestations resulted in a 6 and 2.4% reduction in yield for 1996 and 1997, respectively, when Bt hybrids were compared with their non-Bt isolines. A linear relationship was found between tunnel length per plant in centimeters (x) and yield protection (%) obtained from using Bt corn (y) (y = 1.02 + 0.005x, r2 = 0.7217). At a premium of $34.58 Canadian (CDN) perhectare for Bt corn seed, an infestation of at least 6 cm of corn borer tunneling per plant was required to break even at a market price for corn of $2.50 per bushel CDN. During the period of study, low infestations (0-2 cm) of European corn borer occurred at 25% of the locations assessed, moderate infestations (4-6 cm) occurred at 42% of the locations, and high infestations (>6 cm) occurred at 33% of the locations. At a corn price of $3.00 per bushel CDN and seed premiums of $34.58 per hectare CDN, 5 cm of tunneling was required for a return on investment in Bt seed, comprising only 55% of the growers in the study. With infestations of more than 6 cm of tunneling occurring only 33% of the time, a return on seed investment would be realized in only one of three growing seasons. At a seed premium of $24.70 per hectare CDN per year, at least $74 per hectare CDN in the year of infestation would be required to make up for the two years of no return. In this study, a $74 per hectare CDN return at a corn price of $9.26 per hectare CDN with >16 cm of tunneling damage would have occurred only 7.3% of the time.  相似文献

Conventional electroporation (EP) by 0.1 to 1 kV/cm pulses longer than 100 micros, and supra-electroporation by 10 to 300 kV/cm pulses shorter than 1 micros cause different cellular effects. Conventional EP delivers DNA, proteins, small drugs, and fluorescent indicators across the plasma membrane (PM) and causes moderate levels of phosphatidylserine (PS) translocation at the PM. We hypothesize that supra-EP is central to intracellular effects such as apoptosis induction and higher levels of PS translocation. Our cell system model has 20,000 interconnected local models for small areas of the PM and organelle membranes, small regions of aqueous media, appropriate resting potentials, and the asymptotic EP model. Conventional EP primarily affects the PM, but with a hint of endoplasmic reticulum involvement. Supra-EP can involve all of a cell's membrane at the largest fields. Conventional EP fields tend to go around cells, but supra-EP fields go through cells, extensively penetrating organelles.  相似文献

Traps have been used extensively to provide early warning of hidden pest infestations. To date, however, there is only one type of trap on the market in the U.K. for storage mites, namely the BT mite trap, or monitor. Laboratory studies have shown that under the test conditions (20 °C, 65% RH) the BT trap is effective at detecting mites for at least 10 days for all three species tested: Lepidoglyphus destructor, Tyrophagus longior and Acarus siro. Further tests showed that all three species reached a trap at a distance of approximately 80 cm in a 24 h period. In experiments using 100 mites of each species, and regardless of either temperature (15 or 20 °C) or relative humidity (65 or 80% RH), the most abundant species in the traps was T. longior, followed by A. siro then L. destructor. Trap catches were highest at 20 °C and 65% RH. Temperature had a greater effect on mite numbers than humidity. Tests using different densities of each mite species showed that the number of L. destructor found in/on the trap was significantly reduced when either of the other two species was dominant. 1995年在山东和安徽棉区4个地点比较研究了性信息素笼罩诱捕器和水盆诱捕器对棉铃虫雄蛾的诱捕效果。试验结果表明,4地点笼罩诱蛾量分别是水盆诱蛾量的2.8、3.5、2.5和2.0倍,平均2.7倍。两种诱捕器诱蛾量倍数的变异系数分别为22.9%、31.5%、50.7%和51.2%。可见笼罩诱捕器比水盆诱捕器对棉铃虫雄虫具有更高的诱捕效率。考虑到笼罩诱捕器高效、稳定及简便,建议在我国尽快用其取代水盆进行棉铃虫成虫的标准化监测。  相似文献

A double-vented serial tetraphasic capillary column approach is applied to proteomic MuDPIT-type analysis using extended length capillary reverse-phase columns. The heart of the tetraphasic device consists of a triphasic MuDPIT trap located upstream of a venting tee. The trap is followed by a 60 cm high-resolution capillary column. A conventional high-flow HPLC is used to develop gradients at standard flow rates and pressures. The double-vented triphasic MuDPIT trapping device relieves the capillary separation column from the salt burden during the on-line cation-exchange portion of the analysis. Two configurations are presented, a double-vented continuous column model and a discontinuous model in which the triphasic MuDPIT trap is installed on a six-port valve; both configurations were tested with 60 and 10 cm capillary columns. All four systems were challenged with a trypsin digest of undepleted human serum, and a matrix of proteomic results for the different models and column lengths are compared.  相似文献

Depending on solution conditions, beta-lactoglobulin can exist in one of its six pH-dependent structural states. We have characterized the acid and basic-induced conformational transitions between these structural states over the pH range of pH 1 to pH 13. To this end, we have employed high-precision ultrasonic and densimetric measurements coupled with fluorescence and CD spectroscopic data. Our combined spectroscopic and volumetric results have revealed five pH-induced transitions of beta-lactoglobulin between pH 1 and pH 13. The first transition starts at pH 2 and is not completed even at pH 1, our lowest experimental pH. This transition is followed by the dimer-to-monomer transition of beta-lactoglobulin between pH 2.5 and pH 4. The dimer-to-monomer transition is accompanied by decreases in volume, v degrees (-0.008(+/-0.003) cm3 x g(-1)), and adiabatic compressibility, k degrees (S) (-(0.7(+/-0.4))x10(-6) cm3 x g(-1) x bar(-1)). We interpret the observed changes in volume and compressibility associated with the dimer-to-monomer transition of beta-lactoglobulin, in conjunction with X-ray crystallographic data, as suggesting a 7 % increase in protein hydration, with the hydration changes being localized in the area of contact between the two monomeric subunits. The so-called N-to-Q transition of beta-lactoglobulin occurs between pH 4.5 and pH 6 and is accompanied by increases in volume, v degrees (0.004(+/-0.003) cm3 x g(-1)), and compressibility, k degrees (S) ((0.7(+/-0.4))x10(-6) cm3 x g(-1) x bar(-1)). The Tanford transition of beta-lactoglobulin is centered at pH 7.5 and is accompanied by a decrease in volume, v degrees (-0.006(+/-0.003) cm3 x g(-1)), and an increase in compressibility, k degrees (S) ((1.5(+/-0.5))x10(-6) cm3 x g(-1) x bar(-1)). Based on these volumetric results, we propose that the Tanford transition is accompanied by a 5 to 10 % increase in the protein hydration and a loosening of the interior packing of beta-lactoglobulin as reflected in a 12 % increase in its intrinsic compressibility. Finally, above pH 9, the protein undergoes irreversible base-induced unfolding which is accompanied by decreases in v degrees (-0.014(+/-0.003) cm3 x g(-1)) and k degrees (S) (-(7.0(+/-0.5))x10(-6) cm3 x g(-1) x bar(-1)). Combining these results with our CD spectroscopic data, we propose that, in the base-induced unfolded state of beta-lactoglobulin, only 80 % of the surface area of the fully unfolded conformation is exposed to the solvent. Thus, in so far as solvent exposure is concerned, the base-induced unfolded states of beta-lactoglobulin retains some order, with 20 % of its amino acid residues remaining solvent inaccessible.  相似文献

Diffusion of ionic and nonionic species in multilayered tissues plays an important role in the metabolic processes that take place in these tissues. To create a mathematical model of these diffusion processes, we have chosen as an example hydrogen-bicarbonate ion pair diffusion within the mammalian cornea. This choice was based on the availability of experimental data on this system. The diffusion coefficient of the hydrogen-bicarbonate ion pair in corneal stroma and epithelium is calculated from the observed change in pH in the stroma when conditions at the corneal anterior epithelial surface are changed while the posterior surface is continually bathed with a Ringer's solution in equilibrium with a CO2-gas air mixture. Matching experimental results to a mathematical model of the cornea as a two-layer diffusion system yields, at 37 degrees C, a diffusion coefficient of the hydrogen-bicarbonate ion pair of 2.5 x 10(-6) cm2/s in the stroma and 0.4 x 10(-6) cm2/s in the epithelium. Application of the Nernst-Einstein equation to these data gives the following diffusion coefficients in the two layers: 1) stroma, D(H+) = 11.8 x 10(-6) cm2/s; D(HCO3-) = 1.5 x 10(-6) cm2/s; and 2) epithelium, D(H+) = 1.9 x 10(-6) cm2/s; D(HCO3-) = 0.22 x 10(-6) cm2/s.  相似文献

A stereoselective liquid chromatography-tandem mass spectrometry assay was developed and validated for quantification of S- and R-metoprolol at concentrations of 0.5-50 microg/L in human plasma. Metoprolol was extracted from plasma by liquid-liquid extraction with ethyl acetate (82% recovery). Chromatographic separation of the enantiomers was achieved on a chiral Chirobiotic T column using an isocratic mobile phase consisting of methanol/acetic acid/ammonia (100/0.15/0.15, v/v/v). An ion trap mass spectrometer with an electrospray interface was used for detection in the positive mode, monitoring the m/z transition 268-->191 for metoprolol. Standard curves for S- and R-metoprolol fitted quadratic functions (r(2)>or=0.9995) over the range 0.5-50 microg/L in plasma, with 0.5 microg/L representing the limit of quantification. In this range, relative standard deviations were <6% for intra-day precision and <10% for inter-day precision. The accuracy was within the range of 92-105%.  相似文献

Hemolytic delta-toxin from Staphylococcus aureus was soluble in either water, methanol or chloroform/methanol (2 : 1, v/v). The toxin spread readily from distilled water into films with pressures (pi) of 10 dynes/cm on water and 30 dynes/cm on 6 M urea; from chloroform/methanol it produced 40 dynes/cm pressure on distilled water. The toxin adsorbed barely from water (pi = 1 dyne/ cm) but it did rapidly from 6 M urea (pi = 35 dynes/cm). The protein films had unusually high surface potentials, which increased with the film pressure and decreased with increasing both pH and urea concentration in the aqueous phase. The fluorescence of 1-aniline 8-naphthalene sulfonate with delta-toxin was much greater than that with RNAase and dipalmitoyl phosphatidylcholine itself, indicating probably a marked lipid-binding character of the toxin. By circular dichroism the alpha-helix content of delta-toxin was 42% in water, 45% in methanol, 24% in 6 M urea. Infrared spectroscopy showed predominant alpha-helix in both 2H2O and deuterated chloroform/methanol as well as in films spread from either solvent on 2H2O. In spreading from 6 M [2H]urea, in which the major infrared absorption was that of [2H]urea with peaks at 1600 and 1480 cm(-1), the delta-toxin film showed prevalently non-alpha-helix structures with major peak intensities at 1633 cm(-1) > 1680 cm(-1), indicating the appearance of new beta-aggregated and beta-antiparallel pleated sheet structures in the film. The data prove that (1) high pressure protein films can consist of alpha-helix as well as non-alpha-helix structures and, differently from another cytolytic protein, melittin, delta-toxin does not resume the alpha-helix conformation in going into the film phase from the extended chain in 6 M urea; (2) conformational changes are important in the transport of proteins from aqueous to lipid or membrane phase; (3) delta-toxin is by far more versatile in structural dynamics and more surface active than alpha-toxin.  相似文献

Peculiarities of electrical responses under local illumination of filamentous cyanobacteria (blue-green algae). Phormidium uncinatum were studied by means of extracellular electrodes. Recording of electrical responses at different distances from the exposure place and comparison of these data with the results obtained on physical model of Ph. uncinatum end expected response parameters computed made it possible to estimate the parameters of the cable which can serve as the object model: RC=440 c/cm2 and lambda min greater than or equal to 0.07 cm. According to these values and taking membrane capacity as C=10(-6) phi/cm2, intracellular resistance Rm=10(7) Ohm and conductivity of the surface membrane G less than or equal to 10(-6) I/Ohm cm2 were estimated.  相似文献

A model of venous thrombosis, induced by injection of Feiba(R) (Factor Eighth Inhibitor Bypassing Activity) plus stasis, was studied in the jugular veins of anaesthetized rabbits. Right and left jugular vein segments were isolated by surgical technique for a 3 cm length, which included the bifurcation of the vessel, and left "in situ". Feiba(R) was injected through a marginal ear vein at the dose of 5 U/Kg/0.2 ml; 20 and 25 sec later the contralateral and the homolateral jugular vein segments were ligated respectively. Complete stasis lasted 10 or 20 min, then the vessels were removed, placed in a saline filled Petri dish and visually graded for red thrombus formation on a scale of 0 to 10. A correlation was found between severity of thrombosis and duration of stasis. This model appears to be suitable for testing heparin or heparin-like substances, in fact a linear correlation was found between log dose of the drug (injected i.v. 5 min before Feiba(R] and its antithrombotic effect for each duration of stasis tested. In particular the DE50 value calculated for 10 min stasis was 20.5 mcg/Kg i.v. The reproducibility of the model was good even with a small number of animals (n = 6) for each treatment group.  相似文献

Northern, Diabrotica barberi Smith & Lawrence, and western, D. virgifera virgifera LeConte, corn rootworms are major economic pests of corn, Zea mays L., in the United States. This research was conducted to determine the geographic distribution, abundance, and species composition of Diabrotica species in North Dakota, and to compare effectiveness of unbaited green Scentry™ Multigard and unbaited yellow Pherocon® AM/NB sticky traps for monitoring. Fifty-one corn fields were monitored using traps from July through October of the 2013, 2014, and 2015 growing seasons for rootworm beetle activity. The overall species composition was 61% D. barberi and 39% D. v. virgifera. Both species were frequently captured, and the highest densities (i.e. >10 beetles per trap per week) were found in southeastern North Dakota. Low densities (i.e. <0.1 beetles per trap per week) of D. barberi were found in areas further north, but no D. v. virgifera were captured in those fields. The two different coloured sticky traps were not significantly different across 38 sites for D. barberi and across 21 sites for D. v. virgifera. However, green Scentry™ Multigard traps captured more D. barberi beetles than yellow Pherocon® AM/NB traps at 68% of the 38 fields. In contrast, the yellow Pherocon® AM/NB traps captured more D. v. virgifera beetles at 57% of the 21 fields. Findings also indicated that, although D. barberi was the predominant species in surveyed fields, populations rarely reached the economic threshold. Our study observed that economic populations of corn rootworms were infrequent among the field sites trapped in North Dakota. As a result, producers should scout fields regularly for corn rootworm populations levels to make sound pest management decisions. This knowledge can enable producers to effectively protect their crop when control is economically justified, and the information can also provide input cost savings when populations do not warrant control efforts.  相似文献

Based on the re-interpretation of published data, the von Bertalanffy growth function parameters of the coelacanth, Latimeria chalumnae, are estimated as L=218cm total length, s.e. 23; K=0.059 (year–1), s.e. 0.012; t0=–3.3 (year), s.e. 0.5, corresponding to a life span of 48 years. The length–weight relationship of the form W=a*TLb, with wet weight (W) in g and total length (TL) in cm, is estimated as a=0.0278, b=2.89, r2=0.893, n=87, range=42.5–183cm TL. Using extreme value theory, the maximum length for female coelacanths is estimated as 199cm TL (95% confidence interval=175–223cm TL) and for males as 168cm TL (95% confidence interval 155–180cm TL). Based on data from seven females with embryos or mature eggs, the length-at-first-maturity for females is estimated to be about 150cm TL, corresponding to an age of about 16 years. Based on the value of t0=–3.3 years and on the presence of three scale rings found in a newborn coelacanth, the period of embryogenesis lasts for about three years, the longest known in vertebrates. The natural mortality rate is estimated at M=0.12. Population food consumption is found to be 1.4 times the existing biomass per year, and gross food conversion efficiency indicates that only 10% of the consumed food is utilized for somatic growth.  相似文献

Adult whitefly Bemisia argentifolii Bellows & Perring trap (CC trap) catches were compared with suction type trap catches. CC trap catches were significantly correlated to the suction trap catches. Higher numbers of B. argentifolii adults were caught in CC traps oriented toward an untreated, B. argentifolii-infested, cotton field as compared with traps oriented toward Bermuda grass fields, farm roads, or fallow areas. CC trap catches at five heights above ground (from 0 to 120 cm) were significantly related to each other in choice and no-choice studies. CC trap catches were low in the Imperial and Palo Verde Valleys from late October to early June each of 1996, 1997, and 1998. Trap catches increased with increasing seasonal air temperatures and host availability. Trap catches were adversely affected by wind and rain. Abrupt trap catch increases of 40- to 50-fold for 1-2 d in late June to early July followed by abrupt decreases in adult catches suggest migrating activity of adults from other nearby crop sources.  相似文献
