龙须藤凝集素的分离纯化和性质

利用酸化处理的Sepharose 6B亲和柱从龙须藤（Bauhinia championii)种子中分离纯化出龙须藤凝集素（BCL）,其比活性比抽提液提高了57倍,活力回收率达63．3％。经Sphadex G-100测得BCL的分子量为64000,SDS－PAGE的结果表明BCL由两个相同的亚基组成,亚基分子量为32000,等电聚集凝胶电泳测得其等电点为4．70。BCL是一种糖蛋白,其中性糖含量为3．0％。N－乙酰－D－氨基半乳糖能强烈地抑制BCL对兔红细胞的凝集作用。

抗枯萎、抗黄萎病的棉种的浸取液,经硫酸铵分级,纤维素柱、亲和层析后,再经分子筛过滤,获得在ＰＡＧＥ、ＳＤＳ－ＰＡＧＥ或ＨＰＬＣ柱上均呈现单一蛋白带的棉花凝集素。该凝集素只凝集兔红细胞,对人Ａ、Ｂ或Ｏ型血细胞均不凝集,其凝集活性可被半乳糖或猪甲状腺球蛋白等所抑制。棉花凝集素在６５℃加热５ ｍ ｉｎ,即丧失全部凝集活性;它的凝集活性强烈地依赖于Ｃａ２＋ ;Ｍｎ２＋ 对活性也有促进作用,Ｍｇ２＋ 则无作用。经凝胶过滤或ＳＤＳ－ＰＡＧＥ测定,凝集素的分子量为６３０００,Ｎ－末端氨基酸为Ｖａｌ。凝集素含有１．５％ 的中性糖,是一种促有丝分裂原,细胞转化率为５０．３％ 。     

紫藤凝集素的分离纯化及理化性质研究

用常规方法处理的ＤＥＡＥ离子交换纤维素柱,通过线性离子强度梯度洗脱,从紫藤种子的蛋白粗提液中得到一定纯度的紫藤凝集素。纯化的凝集素凝集兔红血球的比活提高４０倍,总活力回收率为１９．２％。紫藤凝集素的分子量经ＰＡＧＥ鉴定为２０５ｋｄ,是由两种亚基构成的四聚体,这两种亚基各有２个,分子量ＳＤＳ－ＰＡＧＥ鉴定分别为７７６００ｄ和２５１００ｄ。紫藤凝集素是一种糖蛋白,等电点约为４．６０。它可凝集人的各种血     

A blood group A specific lectin from the seeds of Crotalaria striata

A lectin, monospecific for human blood group A red blood cells was extracted from seeds of Crotalaria striata and purified by molecular sieving on Sephadex G-100 and ion-exchange on DEAE-cellulose. A molecular mass of 30 kDa was determined by SDS-polyacrylamide gel electrophoresis under non-reducing and reducing conditions. Molecular sieving on a Superose 12 column indicated a molecular mass of 110 kDa, suggesting the tetrameric nature of the native protein. Amino-acid composition showed the presence of aminated carbohydrate residues on the lectin. N-terminal amino-acid sequencing showed a striking similarity with the N-terminal sequence of the lectin from Crotalaria juncea, which is blood-group non-specific. The potency order of agglutination inhibition with galactose containing monosaccharides was N-acetyl-D-galactosamine greater than D-galactose greater than D-galactosamine as found for blood-group-A-specific lectins from other species.

A new mitogenic D-galactosephilic lectin isolated from seeds of the coral-tree Erythrina corallodendron. Comparison with Glycine max (soybean) and Pseudomonas aeruginosa lectins

The lectin of Erythrina corallodendron (Caesalpiniaceae) seeds was purified by heating, ammonium sulfate fractionation, and affinity chromatography on acid-treated Sepharose. The purified lectin is similar to the soybean lectin in being a glycoprotein of molecular weight around 110 000 - 120 000 and having D-galactosephilic activity. This lectin, like the soybean and Pseudomonas aeruginosa lectins, binds to D-galactosamine, N-acetyl-D-galactosamine, alpha- and beta-galactosides as well as to D-galactose. Like these lectins it absorbs onto either untreated or enzyme (papain or neuraminidase) treated human red blood cells, but exhibits a considerable mitogenic activity towards human lymphocytes (predominantly T cells) only after their treatment with neuraminidase. This mitogenic stimulation of lymphocytes is inhibited by D-galactose and its derivatives. Despite the great similarity between them, the E. corallodendron, soybean, and Pseudomonas lectins differ in regard to the intensity of their agglutinating activity towards erythrocytes obtained from different animals and human donors of diverse ABO blood groups. This phenomenon may be attributed to the difference in the affinities of the three lectins to the various D-galactose derivatives and to their molecular properties.     

中国鲎凝集素的分离纯化及性质研究

 经N-乙酰氨基葡萄糖交联琼脂糖亲和层析及以交联琼脂糖介质的高效液相分子筛层析,从中国鲎细胞溶解物中分离纯化了一种凝集素,其活性比原料鲎试剂提高128倍。鲎凝集素SDS电泳时表现出分子量为69000,和72000的二个亚基。N-乙酰氨基葡萄糖、D-半乳糖,D-甘露糖及岩藻糖等对鲎凝集素凝集鸡红细胞的活性有显著抑制作用,加热60℃,10分钟可使凝集素活性基本丧失。CaCl_2为凝集素活性所必需。鲎凝集素与肺炎球菌C多糖有沉淀反应。     

Human erythrocyte specific lectin from the seeds of Indian coral tree,<Emphasis Type="Italic">Erythrina variegata</Emphasis> Linn,var.<Emphasis Type="Italic">orientali</Emphasis> Linn,Merrill

Human erythrocyte specific lectin was isolated from the seeds ofErythrina variegata Linn. var.orientalis Linn. Merrill. The lectin preferentially agglutinated erythrocytes in the sequence of O>B>A = AB. The lectin was purified 19-fold by affinity chromatography on acid treated sepharose 4B with an yield of 81%. The purified lectin was found homogeneous on polyacrylamide gel electrophoresis. The erythroagglutination reaction was inhibited by N-acetyl-D-galactosamine, D-galactose and lactose at very low concentration. The haemagglutination by the purified lectin was not inhibited by different hexose and pentose sugars even at high concentration. The purified lectin was a glycoprotein and agglutinated leucocytes at 3 μg protein concentration. The lectin induced transformation of peripheral blood lymphocytes in cultures.     

Kluyveromyces bulgaricus yeast lectins. Isolation of N-acetylglucosamine and galactose-specific lectins: their relation with flocculation

Kluyveromyces bulgaricus is a yeast which, upon culture in a calcium-enriched glucose-peptone medium, flocculates. Its flocculation can be reversed by the addition of galactose. In this paper, it is shown that two lectins can be isolated either from the concentrated culture broth or from the supernatant of deflocculated cells suspended in galactose solution. The N-acetylglucosamine-specific lectin, at pH 7.4, agglutinates untreated sheep red blood cells, but agglutinates neither untreated rabbit red blood cells nor glutaraldehyde-fixed sheep or rabbit red blood cells. Conversely, at pH 4.5, this lectin agglutinates glutaraldehyde-fixed sheep red blood cells. The galactose-specific lectin, at pH 7.4, agglutinates both untreated and glutaraldehyde-fixed rabbit red blood cells but does not agglutinate untreated or glutaraldehyde-fixed sheep red blood cells. At pH 4.5, this lectin agglutinates both glutaraldehyde-fixed sheep and rabbit red blood cells and induces flocculation of deflocculated K. bulgaricus cells. In all cases, the agglutination and the flocculation induced by one of these two lectins were inhibited by free or conjugated N-acetyl-D-glucosamine or by free or conjugated D-galactose, respectively. No glycosylhydrolase activity could be detected in the purified lectins.     

Purification and properties of two lectins from the latex of the euphorbiaceous plants Hura crepitans L. (sand-box tree) and Euphorbia characias L. (Mediterranean spurge).

1. From the latex of two members of the plant family Euphorbiaceae, Hura crepitans L. (sand-box tree) and Euphorbia characias L. (Mediterranean spurge), two lectins were purified by affinity chromatography on acid-treated Sepharose 6B followed by elution with D-galactose. 2. The lectin from E. characias is a single molecular species with Mr 80 000, made up of two identical subunits with Mr 40 000, and is a glycoprotein containing 11% carbohydrate. 3. The lectin from H. creptians appears as a mixture of three isolectins with Mr 140 000, consisting of four different subunits with Mr values 37 500, 35 500, 31 000, and 29 000. 4. Both lectins have haemagglutinating activity, with no specificity for human blood groups. The haemagglutinating activity is inhibited by D-galactose and by galactose-containing oligosaccharides. 5. The lectin from H. crepitans is mitogenic to human T-, but not to B-, lymphocytes. The latex of E. characias is mitogenic to T- and, to a lesser extent, to B-, lymphocytes, but the purified E. characias lectin has no mitogenic activity. 6. The lectin from H. crepitans, but not that from E. characias, inhibits protein synthesis by a rabbit reticulocyte lysate.     

舍蝇(Musca domestica vicina)凝集素的分离、纯化和部分性质研究

舍蝇蛹体液经抽提后上Sepharose 4B亲和层析柱纯化制得舍蝇凝集素。制剂经聚丙烯酰胺凝胶电泳和在SDS-PAGE上均呈单一蛋白带,表观分子量为33400。它能凝集人B型红细胞,亦能凝集小白鼠及兔血红细胞。其专一结合的糖为半乳糖与D-及L-岩藻糖。     

梨形环棱螺凝集素的初步研究

通过Sepharose 4B-甲状腺球蛋白亲和层析,从梨形环棱螺Bellamya purificata体内分离到的一种凝集素,不连续PAGE显示其为单一的蛋白质谱带.它能凝集兔、猪、鸭等动物的红细胞,但不能凝集人的A、B、O及AB型血的红细胞和固定后的兔红细胞.其凝集活力可被1.0mol/L的乳糖、半乳糖和60g/L的甲状腺球蛋白抑制,但不能被碱性硼酸缓冲液抑制.对温度变化敏感,有较宽的最适pH范围.
