The present study describes the preparation of poly(HEMA-co-GMA) reactive membranes that were grafted with polyethylenimine (PEI) following UV photo-polymerization. The immobilization of tyrosinase was carried out via multi-point ionic interactions based on ---NH2 groups of PEI and Cu(II) ions. Tyrosinase is a copper-dependent enzyme, which should show a binding affinity for the chelated Cu(II) ions on the membrane surfaces. The tyrosinase immobilization was positively correlated with the input enzyme amount in the immobilization medium. The maximum tyrosinase immobilization capacities of the poly(HEMA-co-GMA)–PEI and poly(HEMA-co-GMA)–PEI–Cu(II) membranes were 19.3 and 24.6 mg/m2, respectively. The enzyme activity when assessed at various pH and temperatures gave broader range for immobilized preparations when compared to free enzyme. The poly(HEMA-co-GMA)–PEI–Cu(II) tyrosinase membranes retained 82% of their initial activity at the end of 120 h of continuous reaction. Moreover, upon storage for 3 months the activity of the immobilized membranes retained 46% of their initial levels. After deactivation of the enzyme, the poly(HEMA-co-GMA)–PEI membrane was easily regenerated, re-chelated with the Cu(II) ions and reloaded with the enzyme for repeated use. The mild immobilization conditions, easy and rapid membrane preparation, one-step enzyme adsorption at substantially higher levels and membrane reusability are the beneficial properties of such systems and offers promising potential in several biochemical processes.  相似文献

Research indicates that exposure to hypoxia is associated with oxidative stress. In this investigation, healthy subjects were exposed to hypoxia by inhalation of 10% oxygen for 2 h (corresponding to 5500 m above sea level). The levels of strand breaks and oxidatively damaged purine bases, measured by the comet assay, and the expression of genes involved in DNA repair of oxidatively damaged DNA were investigated in mononuclear blood cells (MNBC) at baseline, after 2 h of hypoxia, 2 h of reoxygenation, and 1 day and 8 days after the exposure. The level of strand breaks and oxidized purine bases in MNBC increased following both the 2 h of hypoxia and the 2 h reoxygenation period, whereas this effect was not observed in unexposed subjects. The expressions of oxoguanine DNA glycosylase 1 (OGG1), nucleoside diphosphate linked moiety X-type motif 1 (NUDT1), nei endonuclease VIII-like 1 (NEIL1), and mutY homolog (MUTYH) were unaltered throughout the experiment in both groups of subjects, indicating that DNA repair genes are not up-regulated by the hypoxia and reoxygenation treatment. Taken together, this report shows that inhalation of 10% oxygen for 2 h is associated with increased number of oxidized DNA lesions in MNBC, but acute hypoxia may not inflict upon the regulation of genes involved in repair of oxidized DNA.  相似文献

The photokilling activity of a porphyrin-C60 (P-C60) dyad was evaluated on a Hep-2 human larynx-carcinoma cell line. This study represents the first evaluation of a dyad, with high capacity to form a photoinduced charge-separated state, to act as agent to inactivate cells by photodynamic therapy (PDT). Cell treatment was carried out with 1 μM P-C60 incorporated into liposomal vesicles. No dark cytotoxicity was observed using 1 μM P-C60 concentration and during long incubation time (24 h). The uptake of sensitizer into Hep-2 was studied at different times of incubation. Under these conditions, a value of 1.5 nmol/106 cells was found after 4 h of incubation showing practically no change even after 24 h. The cell survival after irradiation of the cells with visible light was dependent upon light exposure level. A high photocytotoxic effect was observed for P-C60, which inactivated 80% of the cells after 54 J/cm2 of irradiation. Moreover, the dyad kept a high photoactivity even under argon atmosphere. Thus, depending on the microenviroment where the sensitizer is localized, this compound could produce a biological photodamage through either a 1O2-mediated photoreaction process or a free radical mechanism under low oxygen concentration.

The mechanism of cell death was analyzed by Hoechst-33258, toluidine blue staining, TUNEL and DNA fragmentation. Cell cultures treated for 24 h with P-C60 and irradiated with a dose of 54 J/cm2 showed a great amount of apoptotic cells (58%). Moreover, changes in cell morphology were analyzed using fluorescence microscopy with Hoechst-33258 under low oxygen concentration. Under this anaerobic condition, necrotic cellular death predominated on apoptotic pathway. There were more apoptotic cells under air irradiation condition than under argon irradiation condition. To determine the apoptotic pathway, caspase-3 activation was studied by caspase-3 activity detection kits. The last results showed that P-C60 induced apoptosis by caspase-3-dependent pathway. These results indicated that molecular dyad, which can form a photoinduced charge-separated state, is a promising model for phototherapeutic agents and they have potential application in cell inactivation by PDT.  相似文献

Gas vesicles are hollow, proteinaceous structures found in some strains of cyanobacteria. They have been used to increase the oxygen supply and improve the cultivation of shear-sensitive mammalian cells. However, the production and, especially, collection of cells and gas vesicles were laborious and ineffective. In this study we examined the use of the cationic polymer, polyethyleneimine (PEI), for improving the cell harvesting by flocculation and flotation. PEI was examined to determine the appropriate molecular weight, pH range, and dosage. The dose of 20–30 mg/l of PEI with molecular weight of 25,000 in the pH range of 6.0–8.5 was found to provide effective and efficient cell flocculation and flotation. As the PEI dose increased, the rate of flotation increased but the clearance (collection) efficacy declined slightly. The culture samples used in this study were taken from light-limiting continuous culture systems at different dilution rates (0.05–0.24 h−1). Without PEI addition, the cells at dilution rates lower than 0.1 h did not float while those at higher dilution rates would float slowly. With PEI addition, the flocculated cells at the dilution rate of 0.05 h−1 sank and those of higher dilution rates would float and the flotation rate increased with increasing specific growth rate. Nonetheless, PEI flocculation and flotation (or sedimentation) could be used to harvest cells at a wide range of growth states.  相似文献

Abstract:  The monoterpenoid carvone ( 1 ) has been shown to have strong antifeedant effects on Hylobius spp. However, because of the high volatility of carvone, long-time protection of conifer seedlings in the field using this compound has not been possible. We demonstrate, in several bioassay steps, that less-volatile, heavier analogues retain their pre-ingestive feeding inhibition activity in the large pine weevil, Hylobius abietis (L.) for a longer time. The first step in the evaluation of the biological activity of 12 carvone analogues was a micro-assay, a choice test lasting 4 h. Compounds active at 100 nmol/cm2 were further dose–response tested to give the effective dose needed to inhibit feeding by 50% (ED50). Of the 14 compounds tested, including both carvone enantiomers, seven heavier analogues were active at low doses (had low ED50 values). As expected from their lower vapour pressure compared with carvone, the heavier analogues proved more resistant to evaporation before testing. Thus, whereas the effect of 8-hydroxy- p -menth-en-6-one 4 declined after 2 days, some of the compounds with high molar masses, such as the alkylhydroxymenthenones 6 and 8 , retained a stable activity for 4 days. The retained activity at 4 days was strongly correlated to molecular mass and boiling point. When tested on natural material (host Pinus sylvestris L. twig sections for 48 h), the heavier analogues showed a rather low activity. Probably, the activity of the more volatile compound carvone ( 1 ) is due to a repellent effect (olfactory mode) rather than the deterrent effects (gustatory mode) of the heavier compounds. In agreement with the relatively low activity on twigs in the laboratory, the hydroxymenthenone 4 was not active in the field when tested for 2 months as a 1 : 9 mixture with a polar wax.  相似文献

Background

The patient population receiving long-term oxygen therapy has increased with the rising morbidity of COPD. Although high-dose oxygen induces pulmonary edema and interstitial fibrosis, potential lung injury caused by long-term exposure to low-dose oxygen has not been fully analyzed. This study was designed to clarify the effects of long-term low-dose oxygen inhalation on pulmonary epithelial function, edema formation, collagen metabolism, and alveolar fibrosis.

Methods

Guinea pigs (n = 159) were exposed to either 21% or 40% oxygen for a maximum of 16 weeks, and to 90% oxygen for a maximum of 120 hours. Clearance of inhaled technetium-labeled diethylene triamine pentaacetate (Tc-DTPA) and bronchoalveolar lavage fluid-to-serum ratio (BAL/Serum) of albumin (ALB) were used as markers of epithelial permeability. Lung wet-to-dry weight ratio (W/D) was measured to evaluate pulmonary edema, and types I and III collagenolytic activities and hydroxyproline content in the lung were analyzed as indices of collagen metabolism. Pulmonary fibrotic state was evaluated by histological quantification of fibrous tissue area stained with aniline blue.

Results

The clearance of Tc-DTPA was higher with 2 week exposure to 40% oxygen, while BAL/Serum Alb and W/D did not differ between the 40% and 21% groups. In the 40% oxygen group, type I collagenolytic activities at 2 and 4 weeks and type III collagenolytic activity at 2 weeks were increased. Hydroxyproline and fibrous tissue area were also increased at 2 weeks. No discernible injury was histologically observed in the 40% group, while progressive alveolar damage was observed in the 90% group.

Conclusion

These results indicate that epithelial function is damaged, collagen metabolism is affected, and both breakdown of collagen fibrils and fibrogenesis are transiently induced even with low-dose 40% oxygen exposure. However, these changes are successfully compensated even with continuous exposure to low-dose oxygen. We conclude that long-term low-dose oxygen exposure does not significantly induce permanent lung injury in guinea pigs.  相似文献

The capacity of tropical whitespotted bamboo sharks Chiloscyllium plagiosum to metabolically compensate, at both the whole‐animal and biochemical levels, to prolonged exposure to temperatures higher (30° C) and lower (20 and 15° C) than their native temperature (24·5° C) was examined. As expected, whitespotted bamboo shark oxygen consumption increased upon exposure to 30° C and decreased at 20 and 15° C. Initial changes in oxygen consumption were maintained even after months at the experimental temperature, indicating that whitespotted bamboo sharks did not compensate metabolically to the experimental temperatures. Maximal activities and thermal sensitivity of citrate synthase and lactate dehydrogenase from whitespotted bamboo shark white locomotor muscle were similar between control animals maintained at 24·5° C and those maintained at 15° C, indicating that cold‐exposed animals did not compensate at the biochemical level. Similarly, lactate dehydrogenase activity did not change following prolonged exposure to 30° C. White muscle from whitespotted bamboo sharks maintained at 30° C had significantly lower citrate synthase activity than did control animals. This result was surprising given the lack of metabolic compensation at the whole‐animal level. Overall, whole‐animal oxygen consumption measurements supported the hypothesis that animals from thermally stable environments lacked the capacity to metabolically compensate to altered temperatures. Enzymatic results, however, suggested that the metabolic potential of muscle could change following temperature acclimation even in the absence of metabolic compensation at the whole‐animal level.  相似文献

Objective:  To characterize the cytomorphological changes after percutaneous ethanol injection (PEI) in benign thyroid nodules, we compared the cytological features of fine needle aspiration cytology (FNAC) samples before and after PEI.
Methods:  Fifty-five cases diagnosed with adenomatous goitre (group 1, 35 cases) or cystic changes only (group 2, 20 cases) were included. In all cases, FNACs were performed by the pathologists before and after PEI. PEI was performed in all cases by a diagnostic radiologist according to the ultrasound guidelines. The following cytological features were evaluated by two pathologists: cellularity of follicular epithelial cells, background, cellular pleomorphism, nuclear/cytoplasmic (N/C) ratio, chromatin pattern, presence of nucleoli, macrophages, multinucleated giant cells, and mitosis.
Results:  In group 1, 19 cases (54%) showed changes in cellularity after PEI, with reduced cellularity in 13 cases (37%) and increased cellularity in six cases (17%). No necrotic background was observed before PEI; however, 14 cases (40%) developed necrotic background after PEI ( P  < 0.0001). Multinucleated giant cells were observed in 18 cases (51%) after PEI, which is in contrast to four cases (11%) before PEI ( P  = 0.001). In group 2, necrotic background was seen in 11 cases (55%, P  < 0.0001) and multinucleated giant cells were observed in four cases (20%) after PEI.
Conclusions:  Necrotic background and presence of multinucleated giant cells are indicative of tissue damage caused by PEI in the FNAC specimens of benign thyroid nodules. In contrast to other modalities including chemotherapy or radiation treatment for malignant tumour, no unusual cytological change is observed after PEI.  相似文献

Isolates of Cochliobolus sativus , the cause of common root rot of barley, were collected from different regions of Syria, and exposed in the laboratory to ultraviolet-C (UV-C) light (254 nm) during periods of 40 or 60 h with a dose rate of 2.52 × 10–3 W/cm2. Significant increases (P   < 0.001) in mycelial growth and sporulation were detected in comparison with the non- irradiated control, and these increased with increasing exposure to UV-C. This exposure increased the mycelial growth by 89 and 100% in the isolates CR9 and CR19, respectively, and the number of spores by 10- and 13-fold, respectively. Some of the isolates retained and some increased in virulence. These data demonstrate that variation occurred in the virulence level and the resistance level of isolates and among accessions, respectively.  相似文献

The porcine pancrease lipase was immobilized by entrapment in the beads of K-carrageenan and cured by treatment with polyethyleneimine (PEI) in the phosphate buffer. The retention of hydrolytic activity of lipase and compressive strength of the beads were examined. The activity of free and immobilized lipase was assessed by using olive oil as the substrate. The immobilized enzyme exhibited a little shift towards acidic pH for its optimal activity and retained 50% of its activity after 5 cycles. When the enzyme concentration was kept constant and substrate concentration was varied the Km and Vmax were observed to be 0.18 × 10−2 and 0.10, and 0.10 × 10−2 and 0.09 respectively, for free and for entrapped enzymes. When the substrate concentration was kept constant and enzyme concentration was varied, the values of Km and Vmax were observed to be 0.19 × 10−7 and 0.41, and 0.18 × 10−7 and 0.41 for free and entrapped enzymes. Though this indicates that there is no conformational change during immobilization, it also shows that the reaction velocity depends on the concentration. Immobilized enzyme showed improved thermal and storage stability. Hydrolysis of olive oil in organic–aqueous two-phase system using fixed bed reactor was carried out and conditions were optimized. The enzyme in reactor retained 30% of its initial activity after 480 min (12 cycles).  相似文献

Abstract Two different methods of applying Pya -AKH to long-winged (macropterous) females of the firebug, Pyrrhocoris apterus (Linnaeus) (Heteroptera) were compared: both injection and topical application increased the levels of lipids in the haemolymph and stimulated locomotor activity. Lipid mobilization was maximal when 10 pmol was applied by injection or 40–100 pmol by topical application, with the first significant responses occurring 1.5 h after injection and 2 h after topical application. The highest elevations of lipid concentration in the haemolymph were comparable between the treatments, i.e. 14.36 ± 3.59 mg/mL for injection and 14.43 ± 4.07 mg/mL for topical application. However, these maximal elevations were achieved at different times: 3 h after the injection and 7 h after the topical application.
Injection of 10 and 40 pmol of Pya -AKH stimulated locomotor activity with maximal activity 3 h later but, surprisingly, injection of 80 pmol showed no effect initially and than a slight inhibitory effect after 6–8 h.
Increased locomotor activity was found after topical application of Pya -AKH, but the response was lower than after injection and appeared later, 5–9 h after the hormone application. In addition, the greatest increase in walking activity required topical application of 300 pmol and was still less dramatic than the response to injection. The stimulatory effect of Pya -AKH on locomotion was positively correlated with its effect on lipid mobilization only for injection of the hormone. It is argued that a stress caused by injection could play a role in the appearance of the complex response to adipokinetic hormone.  相似文献

A novel cross-linked enzyme aggregates (CLEA) concept called combi-CLEA has been described. It is based upon the fact that CLEA can be made from heterogeneous populations of proteins/enzymes. Porcine pancreatic acetone powder crude extract was used for preparing CLEA in such a way that lipase, -amylase, phospholipase A2 activities were retained upto 100%. The lipase present in the CLEA showed greater thermal stability at 50 °C as compared to free enzyme. For lipase and phospholipase A2, Vmax/Km showed no significant change upon combi-CLEA formation but decreased significantly for -amylase activity from 190 to 114 min−1. The lipase activity and -amylase activity in CLEA were completely retained upto three cycles of use. The scanning electron microscopic (SEM) studies showed that morphology of CLEA changed upon inactivation by reuses.  相似文献

Abstract: Based on surveys of species of museum pest insects commonly found in Finland, Norway, Denmark and Sweden, six species were selected for a study of the effect of exposure to anoxic treatment on the larval stage. An oxygen level of 0.3% (the rest, nitrogen) was applied and lethal exposure times were determined. Anthrenus museorum L. was the most susceptible species (LT99 = 32.2 h), while Attagenus woodroffei Halst. & Green and Attagenus smirnovi Zhantiev (LT99 = 88.1 h) were the most tolerant species. The LT99 value of At. woodroffei was not calculated, as only 50% of the larvae were killed by the treatment. The results indicate that large intraspecific variation is present in these two Attagenus species. Larvae of Anthrenus verbasci (L.) (LT99 = 43.9 h), Reesa vespulae (Milliron) (LT99 = 53.6 h), and Trogoderma angustum (Solier) (LT99 = 57.2 h) showed intermediate tolerance.  相似文献

Cationic polymers are known to have potent activity against bacteria, but their effects on viral activity have been little studied. We investigated the effect of one such polymer, polyethyleneimine (PEI), on HIV-1 infection. Although virus-cell binding was significantly inhibited by PEI, HIV-1 infection in human T-cell lines such as MT-4 and MOLT-4 was accelerated conversely when the drug treatment was carried out, after the virus had attached to the cells or PEI was simultaneously added to the virus and cell culture system. This paradoxical effect of PEI on HIV-1 infection was examined using HIV-1 chronically infected cells (MOLT-4/HIV-1). Dissociation of the glycoprotein gp120 (as revealed by exposure of transmembrane protein gp41) from MOLT-4/HIV-1 cells and the resultant fusion of these cells was shown to be induced by the addition of PEI. Accordingly, it was suggested that the binding inhibition of HIV-1 to CD4-positive cells by PEI was due to the shedding of gp120 from HIV-1 particles, and this PEI rather promoted membrane fusion between the virus and cells leading to the enhancement of HIV-1 infection. Similarly, dissociation of gp120 from MOLT-4/HIV-1 was also induced by sCD4. The effect of these reagents on changes in membrane fluidity was evaluated by polarization (p) measurements, and it was observed that the acceleration of membrane fluidity occurred only in the PEI system. Therefore, it is likely that PEI accelerates HIV-1 infection by facilitating virus entry into the host cells through an increase in membrane fluidity.  相似文献

A uricase-producing bacterium was isolated from soil with a medium containing uric acid as the only carbon source. Based on its morphological and physiological characteristics, as well as 16S rDNA sequence and phylogenetic tree analysis, this new isolate belong to the genus Microbacterium. After heat treatment at 70 °C for 30 min, the uricase retained about 100% of the initial activity. The enzyme activity remained largely unchanged when it was stored in borate buffer at pH 8.5 at 37 °C for 40 days. The effects of different factors on the enzyme production were studied. Maize milk was the best C and N resources, and the uric acid showed to be an inducer for uricase production. When the strain was cultured at 30 °C at pH 7.5 for 30–36 h, the uricase activity peaked at 1.0 U/ml.  相似文献

The aim of this work was to study the time course of the oxidative metabolism in mice lung after exposure to ambient particles (ROFA). Swiss mice were intranasally instilled with a ROFA suspension (0.20 mg/kg). Animals were sacrificed 1 or 3 h after the exposure. Eighty percentage of increased oxygen consumption was observed in tissue cubes after 1 h of exposure. This observation was accompanied by an increased NADPH oxidase activity (40%) and mitochondrial oxygen consumption in state 3 (19%). NO production by lung homogenates was found to be increased by 43% after 3 h of exposure. Phospholipid oxidation in lung homogenates showed a 29% increase after 1 h of exposure, while a 30% increase in the carbonyl content was found only after 3 h of exposure. Our data show the relative importance of different sources of reactive oxygen species (NADPH oxidase activity and mitochondrial respiration) to the increased tissue oxygen consumption, oxidative damage and antioxidant status observed in an acute model of ROFA particles exposure.  相似文献

Of the numerous putative fungal nitrilases available from protein databases only a few enzymes were purified and characterized. The purified nitrilases from Fusarium solani, Fusarium oxysporum f. sp. melonis and Aspergillus niger share a preference for (hetero)aromatic nitriles, temperature optima between 40 and 50 °C and pH optima in the slightly alkaline region. On the other hand, they differ in their chemoselectivity, i.e. their tendency to produce amides as by-products. The production of fungal nitrilases is increased by up to three orders of magnitude on the addition of 2-cyanopyridine to the culture media. The whole-cell and subcellular biocatalysts were immobilized by various methods (LentiKats®; adsorption on hydrophobic or ion exchange resins; cross-linked enzyme aggregates). Operational stability was examined using continuous stirred membrane bioreactors. Fungal nitrilases appear promising for biocatalytic applications and biodegradation of nitrile environmental contaminants.  相似文献

The respiration of coho salmon, Oncorhynchus kisutch , weighing between 20 and 45 g was measured at gradually declining oxygen levels and at temperatures ranging between 14 and 17 °C. The maximum and minimum oxygen concentrations tested were 235 and 41 μmol/L, respectively. Respiration rates were measured for 24 h at 235 μmol/L before the oxygen concentration was lowered stepwise to 157 and 81 μmol/L. In one single trial, the oxygen level was lowered to 66, 53, and 41 μmol/L. Respiration was highly variable in time. Peak activities always occurred during the night. The standard metabolic rate at normoxic conditions was estimated to be around 4 μmol oxygen/g/h. The highest rates reached values close to 15 μmol oxygen/g/h. At reduced oxygen levels the standard oxygen demand slightly increased to 4.5 μmol oxygen/g/h, indicating a higher demand for vital metabolic functions. Due to the decrease of swimming activity, the maximum oxygen uptake rates dropped to < 8 μmol oxygen/g/h below 81 μmol/L oxygen concentration. Under long-term conditions, physiological and behavioural adaptations play an important role for survival and need to be considered for the design and operation of fish farm facilities.  相似文献

An oxygen limitation strategy based on dynamic enzyme activity was applied to improve glycerol accumulation and decrease the residual sugar level in a fermentation of Candida krusei in a bioreactor. By applying oxygen limitation at 88 h when the activities of two glycerol synthetic enzymes cytosolic glycerol-3-phosphate dehydrogenase (ctGPD) and glycerol-3-phosphatase (GPP) were low and the activity of mitochondrial glycerol-3-phosphate dehydrogenase (mtGPD) which catalyzes the glycerol dissimilation was high, the glycerol dissimilation was efficiently reduced. The final glycerol concentration reached 51.8 g l−1 at 96 h and 54.9 g l−1 at 116 h, which was 18 and 60% higher than the control (without oxygen limitation), respectively. The residual sugar was consumed completely while it was 11.2 g l−1 at the end of fermentation in the control. Under oxygen limitation, ethanol production was detected at a final concentration of 3.6 g l−1. This work suggests a metabolic flux shift by oxygen limitation in the bioreactor.  相似文献
