Campanumoea is a small genus in the family Campanulaceae, with species divided into sections Campanumoea and Cyclocodon. Sixteen accessions from Campanumoea and related genera native to China were used to study their karyotype. The results showed that chromosome characteristics were different between the two sections. For Campanumoea, the karyotypic formula was 2n = 2X = 2m + 12sm + 2st = 16,3A and for Cyclocodon it was 2n = 2X = 6m + 12sm = 18,3B. These data, combined with chromosomal length characteristics, support the restoration of section Cyclocodon as a genus. However, the incorporation of section Campanumoea into Codonopsis requires more evidence. Comparison of chromosomal length and haploid set length revealed that chromosomal segment rearrangements occurred within sections of Campanumoea and between genera, with the difference within sections being greater than that between genera. Therefore, chromosomal segment rearrangements are present in Campanulaceae, implying that chromosomal segment rearrangement plays an important role in the evolution of diversity in Campanulaceae. By comparing the chromosomal characteristic in section Campanumoea and the genus Adenophora, we concluded that the secondary chromosome type such as n = 17, 18 would be derived by autopolyploidization of n = 9, and by chromosome fusion.  相似文献

The chromosome analysis of Santolina rosmarinifolia subsp. rosmarinifolia, S. oblongifolia, S. semidentata subsp. semidentata, S. semidentata subsp. melidensis, S. canescens and the hybrid complex (S. rosmarinifolia subsp. rosmarinifolia, S. oblongifolia and their putative hybrids) shows that all the taxa are diploids (2n = 2x = 18; 18 + 1 or more B chromosomes, with 2n = 19, 20 only in the hybrid complex). The results show a conserved general structure of the karyotype (14m + 2sm + 2st), but in S. semidentata subsp. melidensis it is variable, with 14m + 2sm + 2st in ten individuals, 14m + (1m ? 1sm) + (1 m ? 1st) in nine individuals and 12m + (1m ? 1sm) + (1m ? 1st) + 2st + 1B in five individuals. Tetraploid individuals occurred in the diploid populations of S. rosmarinifolia subsp. rosmarinifolia and S. canescens, and their autopolyploid origin is discussed. Multivalent configurations at diakinesis, simple and double chromosome bridges and delayed disjunction of homologous and non-homologous chromosomes at anaphase I have negative effects on pollen stainability. The mean fructification percentage is moderate. The results suggest that the complex is a mosaic of introgressive hybrids.  相似文献

Heptapteridae is one of the fish families of the order Siluriformes with a wide distribution throughout the basins of the Neotropical region. The genera Imparfinis and Pimelodella comprise few species and/or populations with some chromosome information. Specimens of Imparfinis schubarti, Imparfinis mirini, and Pimelodella meeki from different sites located in the Paranapanema River Basin/PR/Brazil were cytogenetically analyzed. The two species of the genus Imparfinis exhibited 2n = 58 and FN = 116: I. schubarti, with a karyotypic formula of 30m + 28sm and I. mirini with a karyotype formula of 36m + 22sm. P. meeki presented a karyotype of 2n = 46 characterized by 26m + 14sm + 6st and FN = 92, confirming a variability in the 2n of the Heptapteridae family. Both Imparfinis species exhibited interstitial NORs in pair 1, coincident with a secondary constriction; P. meeki presented NORs located in the terminal position on the short arm of pair 17. All AgNORs were coincident with 18S rDNA probe and CMA3 positive. P. meeki showed a small amount of heterochromatin rich in AT and GC bases. The heterochromatin in I. schubarti was CMA3 positive. In I. mirini the heterochromatin was DAPI-positive. Furthermore, the long arm of one of the chromosomes of pair 19 revealed the presence of heterochromatic heteromorphism only in male individuals. After meiotic analyses, this heteromorphism could be easily identified in the pachytene and metaphase I stages, and was heteropyknotic and DAPI positive. This feature may be an indication of initial differentiation of sex chromosomes in I. mirini, increasing the great karyotypic variability within this family of fish.  相似文献

Bombacoideae is one out of nine subfamilies of Malvaceae and encompasses 160 tree species. The subfamily is karyotypically characterized by small and numerous chromosomes and is traditionally known by a remarkable inter- and intraspecific chromosome number variation. We conducted a comparative cytogenetic analysis to investigate karyotype diversity and chromosome evolution within Bombacoideae. To achieve this, we performed new chromosome counts, CMA/DAPI double staining, genome size estimations, and localization of 5S and 45S rDNA by fluorescence in situ hybridization for 21 species distributed across the Bombacoideae phylogeny. We performed ancestral states reconstruction analyses to elucidate chromosome evolution and provide insights into the systematics and evolution of Bombacoideae in comparison with other Malvaceae species. Newly generated data on chromosome number on Bombacoideae revealed diploids (Ochroma (2n = 84), Cavanillesia, Pochota, Pseudobombax (2n = 88), and Pachira (2n = 92)) and polyploids (Adansonia digitata (2n = 160) and Eriotheca species (2n = ca. 194 and 2n = 276)). For most species, in situ hybridization revealed karyotype, with two pairs of 45S rDNA sites co-located with CMA+ bands, and 5S rDNA sites in only one chromosome pair. Taken together, our results provide support to the hypothesis of karyotypic stability in Bombacoideae. Only the Pachira s.l. clade displayed some variability in ploidy level, number of CMA+ bands and 45S rDNA sites, and genome size compared to other Bombacoideae clades. The Striated bark clade was characterized by comparatively small genomes and low cytomolecular variability. Karyotypic data were related to biogeographic and species richness patterns of Bombacoideae.  相似文献

The subgenus Xiphium is one of the six infrageneric divisions of the genus Iris. Chromosome numbers of six of the seven Xiphium species are known. Here the aim was to infer genetic and phylogenetic relationships based on chromosome numbers, chromosome markers and plastid sequences. Chromosomal locations of 5S and 45S rDNA loci were determined in 19 populations of the 7 species by fluorescence in situ hybridization (FISH). Additionally, the trnLtrnF plastid spacer was sequenced and a phylogenetic analysis performed. Based on chromosome markers, subgenus Xiphium species were classified into four groups that differed in the number and locations of both types of nrDNA: (1) I. tingitana (2n = 28), I. filifolia (2n = 30, 34) and I. xiphium (2n = 34), (2) I. juncea (2n = 32) and I. boissieri (2n = 36), (3) I. serotina (2n = 34) and (4) I. latifolia (2n = 42). Although the trnLtrnF phylogeny was not fully resolved, the sequence analysis showed a well-supported subgroup of I. filifolia, I. tingitana and I. xiphium, as well as I. juncea. FISH physical maps of the Iris subgenus Xiphium taxa are species dependent. I. filifolia, I. tingitana and I. xiphium are very closely related species and share cytogenetic characteristics. Disploidy appears to have been central in the evolution of this subgenus, given a series of chromosome numbers (2n = 28, 30, 32, 34, 36, 42) and our phylogenetic results. Clear differences were found among European and African populations of I. filifolia. A different taxonomic treatment of I. filifolia is supported for populations on both sides of the Strait of Gibraltar.  相似文献

Fish of the genus Selene, known as lookdowns or moonfish, are one of the most morphologically derived groups of the family Carangidae, whose phylogenetic relationships are still largely unknown. In this study, we discuss karyoevolutionary aspects of three representatives of this genus from the Western Atlantic: Selene brownii (2n = 48; FN = 48), Selene setapinnis (2n = 46; FN = 48), and Selene vomer (2n = 48; FN = 50). Their body patterns were also investigated and compared to one another and in relation to two other species of different genera. Two mechanisms of karyotypic evolution seem to have acted in the diversification of this genus, namely pericentric inversions and centric fusions. Mapping of rDNA sequences showed that chromosome pairs bearing 5S rDNA sites are similar, whereas those bearing 18 rDNA sites are morphologically distinct while apparently also exhibiting interspecies synteny. Although the nucleolar organizer-bearing chromosomes are extremely efficient cytotaxonomic markers among Selene species, others cytogenetic patterns of these species are relatively conserved. Hybridization with telomeric probes (TTAGGG)n did not exhibit interstitial telomeric sites (ITS), especially in S. setapinnis, where, along with a reduction in diploid number, a large metacentric pair derived from centric fusion is present. Data obtained by geometric morphometrics enable a clear morphological distinction among the three species, as well as in relation to two other species of the genus Caranx and Oligoplites. Data obtained suggest that morphologic evolution in Selene species was primarily dissociated from visible changes that occurred at the chromosomal level.  相似文献

Karyotypic characters, mitotic metaphase chromosomes, monoploid idiograms and karyograms of Minuartia anatolica (Boiss.) Woronow var. phrygia (Bornm.) McNeill, Minuartia anatolica (Boiss.) Woronow var. scleranthoides (Boiss. & Noe) McNeill, Minuartia corymbulosa (Boiss. & Balansa) McNeill var. gypsophilloides McNeill and Minuartia aksoyi M.Koç & Hamzao?lu were investigated for the first time. Analysis of somatic metaphases showed that the chromosome numbers and the formulas of these taxa were 2n = 24 = 14m + 6sm + 4st for Minuartia anatolica var. phrygia, 2n = 14 = 6m + 8sm for Minuartia anatolica var. scleranthoides, 2n = 14 = 6m + 4sm + 4st for Minuartia corymbulosa var. gypsophilloides and 2n = 30 = 14m + 10sm + 6st for Minuartia aksoyi. No satellites were observed in the karyotypes of these taxa. Karyotype asymmetry was estimated by many different methods, namely the Stebbins classification, the karyotype asymmetry index (As K %), the total form percent (TF %), the Rec and Syi indices, the intrachromosomal asymmetry index (A1) and interchromosomal asymmetry index (A2), the dispersion index (DI), the degree of asymmetry of karyotype (A index) and the asymmetry index (AI).  相似文献

Chromosome data are important for elucidating intergeneric relationships and delimiting infrafamilial tribes in plants. This paper reports karyological data for 12 Seriphidium (Bess.) Poljak. species from China, of these, five count as new reports and the others have been reported elsewhere. We also report the tetraploid level in S. schrenkianum for the first time. The karyotype formulae and types for the five new reports are: S. schrenkianum (Ledeb.) Poljak. 2n = 4x = 36 = 22m + 12sm + 2st, S. sublessingianum (Kell.) Poljak. 2n = 2x = 18 = 14m + 4sm and S. transiliense (Poljak.) Poljak. 2n = 2x = 18 = 4M + 6m + 2m(SAT) + 4sm + 2sm(SAT), belong to 2A type; S. cinum (Berg. ex Poljak.) Poljak. 2n = 2x = 18 = 8m + 10sm and S. sawanense Y. R. Ling et C. J. Humphries 2n = 2x = 18 = 10m + 2m(SAT) + 6sm, belong to 2B type. Furthermore, we estimated karyotype asymmetry in the 12 species by using an intrachromosomal asymmetry index and an interchromosomal asymmetry index. Solanum species were cytogenetically analysed to localize species-specific markers. Solanum atropurpureum Schrank, S. dulcamara L., S. gilo Raddi., S. melongena L., S. nitidibaccatum Bitter and S. paniculatum L. showed 2n = 24, whereas S. luteum Mill., S. nigrum L. and S. laciniatum Ait. showed 2n = 48, 2n = 72 and 2n = 92, respectively. All species demonstrated a symmetrical karyotype. The average chromosome size varied between diploid (1.95 μm) and polyploid (1.31 μm) species. Application of the CMA3/DAPI technique showed two CMA3 +/DAPI? terminal blocks in S. dulcamara, S. atropurpureum and S. luteum, indicating one homologous pair. In S. nitidibaccatum two large CMA3 +/DAPI? segments were observed apart from several terminal blocks in almost all chromosomes. The same CMA3 + telomeric standard was also found in prometaphases of S. luteum. Similar results were obtained with FISH with 45S rDNA probes: fluorochrome CMA3 + telomeric blocks associated with satellites, with two blocks in S. atropurpureum, S. dulcamara, S. nitidibaccatum and S. luteum and four blocks in S. nigrum and S. lacinatum. Localization of species-specific markers was successful, allowing recognition of particular cytological features in most of the species analysed.  相似文献

A new species of Myrsinaceae, Lysimachia jinzhaiensis S.B. Zhou & K. Liu, is described and illustrated from Dabieshan mountain, Anhui, China. It is similar to L. christiniae in the prostrate stem, opposite leaves, yellow flowers born singly in leaf axils, but differs by being glabrous throughout or glandular on young parts, and having quadrangular stem, corolla with densely transparent glandular, orange-red corolla base with lobes being significantly overlapping and contorting to left in bud. Moreover, L. jinzhaiensis have a different karyotype, formulated as 2n = 2m + 2sm + 10st (2SAT) + 10t.  相似文献

The genus Eigenmannia (Teleostei: Gymnotiformes), a widely distributed fish genus from the Neotropical region, presents very complex morphological patterns and many taxonomic problems. It is suggested that this genus harbors a species complex that is hard to differentiate using only morphological characteristics. As a result, many species of Eigenmannia may be currently gathered under a common name. With the objective of providing new tools for species characterization in this group, an analysis of the polymorphism of DNA inter-simple sequence repeats (ISSR), obtained by single primer amplification reaction (SPAR), combined with karyotype identification, was carried out in specimens sampled from populations of the Upper Paraná, São Francisco and Amazon river basins (Brazil). Specific ISSR patterns generated by primers (AAGC)4 and (GGAC)4 were found to characterize the ten cytotypes analyzed, even though the cytotypes 2n = 38 and 2n = 38 XX:XY, from the Upper Paraná basin, share some ISSR amplification patterns. The geographical distribution of all Eigenmannia specimens sampled was inferred, showing the cytotype 2n = 31/2n = 32 as the most frequent and largely distributed in the Upper Paraná basin. The cytotype 2n = 34 was reported for the first time in the genus Eigenmania, restricted to the São Francisco basin. Polymorphic ISSR patterns were also detected for each cytotype. Considering our results and the data reported previously in the literature, it is suggested that many of the forms of Eigenmannia herein analyzed might be regarded as different species. This work reinforces the importance of employing diverse approaches, such as molecular and cytogenetic characterization, to address taxonomic and evolutionary issues.  相似文献

Santolina impressa (2n = 2x = 18) is an endemic species of Portugal, with restricted geographical distribution. The present study aimed to explore its chromosomal variation in respect to chromosome morphology, meiotic behaviour, and effects on pollen stainability and fecundity. Its karyotype formula was found to be either 12m + 2msat + 2smsat + 2st (75% of the individuals) or 12m + 2msat + 3sm + 1smsat (25% of the individuals). Univalents were observed in 29.21% of the meiocytes. Chromosome fragments due to breakage in the chromosome arm were observed in 10 meiocytes only (11.23% of the meiocytes). Chains and rings of trivalents were observed in 14.60% of the meiocytes (one trivalent per meiocyte was observed). Chains and rings of quadrivalents were observed in 21.34% and 11.23% of the meiocytes, respectively, with a range of 0–1 per cell. Nine plants (40.90% of the total) with 2n = 2x = 18 + 2B showed a quadrivalent configuration in diakinesis. Twenty abnormal anaphases with delayed disjunction of the four non-homologous and the two homologous chromosomes were observed. Simple chromosome bridges without fragments and interchromosomal adhesions were observed in 35.95% of the anaphases analyzed. B chromosomes showed bivalent association in diakinesis and their segregation at anaphase I was normal. Pollen was found to be fertile (mean ± s.d. = 89.57 ± 47.14%); the effect of univalent frequency and frequency of abnormal anaphase I on pollen stainability was strong and statistically significant.  相似文献

Fish belonging to the genus Hypostomus are known for exhibiting a striking diversity in its karyotype structure, however the knowledge concerning the distribution patterns of heterochromatin and location of repetitive DNA sequences in the karyotypes is still limited. Aiming a better understanding of the chromosomal organization in this group, we analyzed three sympatric species of Hypostomus collected in the Hortelã stream, a component of the Paranapanema River basin, Botucatu/SP/Brazil. The analyses involved the cytogenetic characterization and chromosomal mapping of repetitive sequences and intra/interspecific comparisons using sequences of the cytochrome C oxidase subunit I. The results revealed that H. ancistroides presents a karyotype with 2n = 68 chromosomes, H. strigaticeps 2n = 72 chromosomes, and H. nigromaculatus 2n = 76 chromosomes. In addition to differences found in the diploid number, it was also observed variations in karyotypic formulae, amount of constitutive heterochromatin, and location of nucleolus organizer regions. The cytogenetic mapping of 5S and 18S rDNA, as well as of the H3 histone gene, disclosed a differential dispersion process among the three species. In some cases the Rex1 transposable element showed to be co-located with 5S rDNA sites. The molecular analyses support the cytogenetic data and represent an additional tool for the characterization of the analyzed species. The results evidenced that chromosomal variations are not restricted to differences in diploid number or karyotypic macrostructure in the genus Hypostomus, indicating that events such as transposition of heterochromatin and rDNA segments may participate in the differentiation process occurred in these species.  相似文献

Eighteen species belonging to family Convolvulaceae occurring in northeastern Brazil were analyzed to identify the karyotypic variability among rupicolous and non-rupicolous species. The count of 2n = 56 for Evolvulus sp., is the first report of polyploidy for the genus. The similarities among the karyotypes of species of Ipomoea, Merremia, and Operculina suggest that x = 15 is the basic secondary number for these genera, while x = 13 has been observed in Evolvulus. The genus Jacquemontia, with 2n = 18 is karyologically distinct from the other genera of the family. Intraspecific numerical chromosomal variations were not detected, nor were any karyological variations related to the rocky habitat characteristic of inselbergs. The occurrence of other plant groups with polyploid forms linked to this type of habitat suggests that the family Convolvulaceae is karyologically stable. We discuss variations in chromosome numbers, base numbers, and their implications in the evolution of the principal groups within the Convolvulaceae.  相似文献

Ipheion, as recognized by Guaglianone (Darwiniana 17:159–242, 1972), is a small genus of the family Amaryllidaceae, subfamily Allioideae, tribe Gilliesieae endemic to South America. Since 1800, the species of Ipheion were treated under Beauverdia, Brodiaea, Milla, Nothoscordum, Tristagma or Triteleia. Nowadays, some species of Ipheion are treated as Tristagma and some other under Nothoscordum. In this paper, we analyzed the morphological variation among species of Ipheion and related genera using a numerical taxonomic approach based on overall similarity, to investigate the intergeneric and infrageneric variations to the circumscribed groups. Based on 141 operational taxonomic units and 51 morphological characters, multivariate analyses were performed. Results showed that Ipheion separated into two groups according to Guaglianone’s sectional treatment: Ipheion sect. Ipheion is morphologically more similar to the studied species of Tristagma, while I. sect. Hirtellum can be separated from Nothoscordum/Zoellnerallium and from Tristagma and allies. These a priori groupings were tested using discriminant and univariate analyses. Diagnostic characters were selected to differentiate groups. Previous taxonomic treatments are discussed and results are also evaluated using additional karyological and geographical data.  相似文献

Calendula L. (Asteraceae) is a taxonomically and cytologically complex genus due to its high morphological and karyological variation. To gather consistent cytological information aiming to consolidate the existing knowledge, sustain the taxonomic revision of the genus and explore the evolutionary relationships among species, the genome size and chromosome number of the Iberian Peninsula representatives of this genus were assessed. The study included 11 taxa that occur in the Iberian Peninsula, one in Madeira and two from Morocco. Chromosome counts were made using the squash technique in root tips and flower buds, while nuclear DNA contents were assessed using propidium iodide flow cytometry. The following chromosome numbers are reported: 2n = 44 for C. arvensis, 2n = 30 for C. tripterocarpa, and 2n = 32 for the remaining Iberian taxa. The genome size of Calendula species was assessed for the first time and ranged from 1.75 pg/2C in C. maroccana to 5.41 pg/2C in C. arvensis. Within the complex formed by C. incana and C. suffruticosa, a gradient of genome size values was obtained. Intraspecific variation in genome size was detected in some taxa. The obtained genome size values and their variation are discussed in the light of the theories proposed for the speciation of the genus, with events of hybridization, genome duplication and dysploidy being hypothesized to play a major role in the evolution of this genus.  相似文献

Mitotic chromosome numbers and karyotypes of Grabowskia (G. boerhaviaefolia, G. duplicata, G. obtusa) and the monotypic genus Phrodus (P. microphyllus) were determined. Both genera were diploid with 2n = 24. The chromosome number of G. obtusa is reported here for the first time. Karyotypes of Grabowskia species were highly symmetrical with 11m + 1sm pairs. Phrodus microphyllus showed a different formula: 8m + 3sm + 1st pairs. Karyotypic features suggest that morphological differentiation in Grabowskia was not followed by chromosomal divergence, as previously found in Lycium as well. Although symmetrical karyotypes have been correlated with an ancestral condition of the taxa bearing them, in the tribe Lycieae the basal genus Phrodus showed the opposite trend.  相似文献

Studies of rDNA location in holocentric chromosomes of the Cyperaceae are scarce, but a few reports have indicated the occurrence of multiple 45S rDNA sites at terminal positions, and in the decondensed state of these regions in prometaphase/metaphase. To extend our knowledge of the number 45S and 5S rDNA sites and distribution in holocentric chromosomes of the Cyperaceae, 23 Brazilian species of Eleocharis were studied. FISH showed 45S rDNA signals always located in terminal regions, which varied from two (E. bonariensis with 2n = 20) to ten (E. flavescens with 2n = 10 and E. laeviglumis with 2n = 60). 5S rDNA showed less variation, with 16 species exhibiting two sites and 7 species four sites, preferentially at terminal positions, except for four species (E. subarticulata, E. flavescens, E. sellowiana and E. geniculata) that showed interstitial sites. The results are discussed in order to understand the predominance of terminal rDNA sites, the mechanisms involved in the interstitial positioning of 5S rDNA sites in some species, and the events of amplification and dispersion of 45S rDNA terminal sites.  相似文献
