The ability of a cell-free secretion from germinating conidia(germination fluid) and heat-killed conidia of Botrytis cinereaPers. ex Pers. to induce resistance in exposed carrot root tissueswas unaffected during freezing to –25 °C and rapidthawing, and by heating to 50 °C for 10 min. Activity waslost by boiling for 10 min or at room temperature after 24 h. There was less cell death (Evans blue and neutral red staining)in the surface tissue of slices pretreated with heat-killedconidia and subsequently exposed to infection than in similartissue of untreated, inoculated slices. Both inducing preparationsalone caused maceration and cell death in one or two cell layersof the slice surface. Healthy tissue killed by freezing to –25°Cfor 2 h and rapid thawing, released a factor which, when similarlytested on slices, resulted in germ-tube inhibition and reducedvisual symptoms following inoculation with live spores. Thepossibility that cell death triggered the release of a hostelicitor which caused enhanced accumulation of phytoalexinsand suberin deposition is discussed. The incorporation of radioactivity from precursors of methylester and galacturonosyl residues into pectin was investigatedusing tissue slices cut from ripening pear fruits. Incorporationfrom 14CH3 methionine into methyl ester of water soluble pectinincreased 10 fold in 4 d at 18 °C and declined in laterstages of ripening. Activity from [3H]inositol could not bedetected in gaJacturonic acid released enzymically from solublepolysaccharides. When l3H]glucose was used as a precursor, activitycould be detected in galacturonic acid released from both thesoluble and insoluble polysaccharide fractions. Methionine wasa more efficient precursor of methyl ester groups than S-adenosylmethionine or S-methyl methionine; incorporation from all threeprecursors was inhibited under nitrogen. Radioactively labelledmethyl ester did not decline during a 225 min ‘chase’following a 15 min ‘pulse’ of [14CH3]methionine;the total pectin content of slices increased by 20% during this4 h incubation.  相似文献

The levels of purine and pyrimidine nucleotides in suspensioncultures of Catharanthus roseus were determined 24 h after stationary-phasecells were transferred to fresh complete (‘+Pi’)or phosphate-deficient (‘–Pi’) Murashige-Skoogmedium. The levels of ATP, GTP, UTP and CTP were from approx.3 to 5-fold greater in the cells grown in ‘+Pi’medium than in the cells grown in ‘–Pi’ medium.The levels of almost all other nucleotides were slightly higherin the cells in ‘+Pi’ medium. The rates of de novoand salvage biosynthesis of purine and pyrimidine nucleotideswere estimated from the rates of incorporation of radioactivityfrom [14C]formate, [2–14C]glycine, NaH14CO3, [6–14C]orotate,[8–14C]adenine, [8–14C]adenosine, [2–14C]uraciland [2–14C]uridine. The results indicated that the activityof both the de novo and the salvage pathway was higher in thecells in ‘+Pi’ medium than in the cells in ‘–Pi’medium. The rate of degradation estimated from the rate of releaseof 14CO2 from labelled purines and pyrimidines indicated thatdegradation of uridine was significantly reduced in the cellsin ‘+Pi’ medium, but no significant difference wasfound in the degradation of adenine, adenosine and uracil. Thepossible role of Pi in the control of the biosynthesis of nucleotidesand in the degradation of uridine is discussed. Development of seeds following pollination with irradiated pollenwas studied inActinidia deliciosa(kiwifruit) ‘Hayward’.Pollinations were carried out using two different sources ofpollen (‘Tomuri’ and ‘Matua’) irradiatedwith gamma rays at doses of 700 and 900 Gy. Non-irradiated crosseswere used as controls. Pollen irradiation had little effectonin vitropollen germination. Irradiated pollen affected seedset and seed content, and induced the formation of parthenogeneticembryos. In comparison to the control, the embryo growth ratewas slower and the endosperm contained very low amounts of storageproducts. Seed set was significantly reduced following bothdoses of irradiation. Two types of seeds were observed: (1)seeds with endosperm only; and (2) seeds with both embryo andendosperm. The proportion of seeds containing endosperm onlywas almost ten-fold higher than those containing both embryoand endosperm. Embryo production by gamma-irradiated pollenwas genotype- and dose-dependent. The induction of parthenogenesiswas higher following gamma ray doses of 900 Gy than 700 Gy,which suggests the ‘Hertwig Effect’; the best efficiencywas obtained with ‘Tomuri’ pollen. Ploidy levelof parthenogenetic embryos was evaluated by nuclear size (area)with the use of image analysis. There was a large differencein embryo nuclei size between control and parthenogenetic embryos(mean size 90.8 and 49.1 µm2, respectively). The metabolic fate of [U-14C]sucrose in suspension culturesof Catharanthus roseus cells was monitored for 96 h after thecells were transferred to fresh complete (‘+Pi’)or to phosphate-deficient Murashige and Skoog (‘–Pi’)medium. Sucrose was hydrolysed extracellularly to glucose andfructose. The rate of uptake of sugars by the cells was 1.5–3times higher in ‘+Pi’ culture than in ‘–Pi’culture. Little difference in the rate of incorporation of radioactivityinto the ethanol-soluble fraction was found between the ‘+Pi’and ‘– Pi’ cultures during the initial 24h of culture, but after 48 h the rate in ‘ +Pi’cultures was higher than that in ‘–Pi’ cultures.Incorporation of radioactivity into ethanol-insoluble macromoleculeswas always significantly higher in the cells in ‘+Pi’cultures than in those in ‘–Pi’ cultures.The results suggest that Pi strongly affects the utilizationof sugars by cultured plant cells through the stimulation oftransport of sugars as well as through the activation of metabolism. A study has been made of photosynthetic 14CO2 fixation by isolated‘mature’ internodes of Nitella translucens. Experimentalconditions were similar to those used in studies of the ionicrelations of these cells. Maximum rates of photosynthesis were33–40µµmoles CO2, fixed per cm2 of surfacearea per second (equivalent to 12–15 /xmoles fixed permg chlorophyll per hour). l4CO2 fixation was inhibited to thedark level by 3(3,4,dichlorophenyl)-1, 1-dimethylurea (at 0-6µM or 10µM) and by the uncoupler carbonyl cyanide-m-chlorophenylhydrazone(SµM). The presence of imidazole or ammonium sulphate(both of which uncouple ATP production in vitro) did not resultin an inhibition of 14CO2 fixation. These results are discussedin relation to published work on solute uptake by Nitella translucens.During photosynthesis there was rapid movement of 14C-labelledorganic compounds out of the chloroplasts. 14C-labelled sucrose,ammo-acids, and sugar phosphates were found in samples of vacuolarsap.  相似文献

A microsomal flavonoid 3'-hydroxylase (F3'H) catalyzing themetabolism of naringenin to eriodictyol in Citrus sinensis (L.)Osbeck cv. ‘Hamlin’ cell suspension cultures wasshown to be a cytochrome P450 enzyme. This reaction requiredO2 and NADPH and was inhibited by CO, with partial reversalof CO-inhibition by light at 450 nm. Cytochrome P450 contentranged from 10–20 pmol (mg microsomal protein)–1.The F3'H reaction was shown to be linear in regard to proteinconcentration between 2.5 and 25 µg of microsomal protein.The optimum pH for the reaction was 7.4–7.6 and the temperatureoptimum was between 30 and 37°C. The apparent Km and Vmaxfor naringenin were 24 µM±3.2 and 81.4±7.9pmol eriodictyol min–1 (mg protein)–1, respectively.The microsomal F3'H was also capable of forming dihydroquercetinfrom dihydrokaempferol (40 pmol min–1 (mg protein)–1)and of quercetin from kaempferol (3.25 pmol min–1 (mgprotein–1). Cytochrome c and ketoconazole were the bestinhibitors of WH activity followed by piperonyl butoxide anda-naphthoflavone. Light was shown to be an inducer of the F3'Halmost doubling the specific activity and increasing the microsomalcytochrome P450 content by 30% over that of dark grown cells.F3'H activity was also confirmed in microsomal preparationsof young (new flush) leaves from ‘Hamlin’ treesand flavedo of ‘Hamlin’ oranges, ‘Marsh’grapefruit, and ‘Lisbon’ lemon. No activity wasobserved in older, hardened leaves and albedo of all the fruittested. The effects of light, auxin and cytokinin on vessel elementformation in phloem slices of carrot root were examined. When slices of carrot cultivars, ‘Nakamura-senko-futo’and ‘Yamada-hyakunichisenk6- naga’, preculturedin the dark on modified Murashige and Skoog's medium for twodays were cultured on a medium containing 5x10–6 M 2,4-Din the dark, no vessel element formation occurred. When preculturedslices were cultured in the light with 5x10–6M 2,4-D,vessel element formation was remarkable. But when 5x10–7Mkinetin, benzyladenine or zeatin was added, vessel elementswere readily formed even in the dark. When slices were cultured in the light, a cytokinin-like substance(s)that causes vessel element formation was produced in the slices,then was released to the medium. The substance(s) was fairlystable to heat. In slices of carrot cultivars, kuroda-gosun, ‘Kintoki’and ‘Kokubu-senk6-6naga’, a different result forvessel element formation was obtained. When slices of thesecultivars were cultured on a medium containing 5x10–6M2,4-D in the dark, vessel element formation was remarkable.It seemed, therefore, that these cultivars contain enough ofa cytokinin-like substance(s) to form vessel elements. In two experiments, the functioning and metabolism of nodulesof white clover, following a defoliation which removed abouthalf the shoot tissue, were compared with those of undefoliatedplants. In one experiment, the specific respiration rates of nodulesfrom undefoliated plants varied between 1160 and 1830 µmolCO2 g–1h–1, of which nodule ‘growth and maintenance’accounted for 22 ± 2 per cent, or 27 ± 3.6 percent, according to method of calculation. Defoliation reducedspecific nodule respiration and nodule ‘growth and maintenance’respiration by 60–70 per cent, and rate of N2 fixationby a similar proportion. The original rate of nodule metabolismwas re-established after about 5 d of regrowth; during regrowthnodule respiration was quantitatively related to rate of N2,fixation: 9.1 µmol CO2 µmol–1N2. With the possible exception of nodules examined 24 h after defoliation,the efficiency of energy utilization in nitrogenase functioningin both experiments was the same in defoliated and undefoliatedplants: 2.0±0.1 µmol CO2 µmol–1 C2H4;similarly, there was no change in the efficiency of nitrogenasefunctioning as rate of N2 fixation increased with plant growthfrom 1 to 22 µmol N2 per plant h–1. Exposure of nodulated white clover root systems to a 10 percent acetylene gas mixture resulted in a sharp peak in rateof ethylene production after 1.5–2.5 min; subsequently,rate of ethylene production declined rapidly before stabilisingafter 0.5–1 h at a rate about 50 per cent of that initiallyobserved. Regression of ‘peak’ rate of ethyleneproduction on rate of N2 fixation indicated a value of 2.9 µmolC2H4 µmol–1 N2, for rates of N2 fixation between1 and 22 µmol N2 per plant h–1. We report that growth of Dunaliella salina at either 13°C/150µmol m–2s–1 or 30°C/2,500 µmol m–2s–1 results in the accumulation of comparable levels ofcarotenoids and the zeaxanthin-binding protein, Cbr. The layer of daytime concentration of Calanus ponticus (VC andVI C) performing daily vertical migrations and the layer of‘winter stock’ aggregation are confined to the depthof maximal gradient of the main pycnocline under an unusuallysharp oxycline. The concentration layer thickness ranges from2 to 20–30 m and the Calanus concentration in it is >250ind. m–3, sometimes being 3500 ind. m–3 and evenmore. The population in the concentration layer is divided intotwo ecological groups: I, feeding and migrating specimens ofcopepodite stages V and VI, their body lipid contents being25–60 µg min.–1; and II, non-feeding and non-migratingspecimens of copepodite stage V, their body lipid contents being100–150 µg ind.–1. The relationship with oxygenconcentration was studied in both ecogroups. The experimentsshow that specimens of ecogroup II can exit at an oxygen concentrationof 0.06 ml 1–1, but at such concentration falling intoanabiosis. They die at 0.04 ml O2 1–1. Estimates of respirationof the group II specimens (‘winter stock’) showthat lipids they store are sufficient for 7 months' survival.Depth of Calanus concentration is determined by water densityrather than concentration of oxygen.  相似文献

Seasonal changes in the structure of the zooplankton communitiesof two large (18 500 m3) experimental enclosures (‘LundTubes’) are reviewed. The diets of the two most prominentcrustaceans, Daphnia hyalina and Diaptomus gracilis, were investigatedby direct analyses of gut contents and in a series of shortterm experimental manipulations of communities isolated in 6m3 bags, placed in one of the enclosures. The relative abundanceof Daphnia hyalina generally coincided with warmer water temperaturesbetween late spring and late summer, but its numbers fluctuatedwithin that period, apparently in response to the availabilityof smaller algae (<104 µm3 unit volume: chiefly cryptomonads,chlorellids and Ankyra). Daphnia numbers declined steadily throughthe summer in the enclosure overwhelmingly dominated by Microcystis,although it did feed on smaller colonies (< 105 µm3in volume). Diaptomus gracilis and most rotifers were also prominentin the first half of the year, but peak abundances of Keratellaquadrata followed the presence of large populations of small(<103 µm3) algae. Nitrate provision has been found to regulate the capacity forChara corallina cells to take up nitrate. When nitrate was suppliedto N sufficient cells maximum nitrate uptake was reached after8 h. Prolonged treatment of the cells in the absence of N alsoresulted in the apparent ability of these cells to take up nitrate.Chlorate was found to substitute partially for nitrate in the‘induction’ step. The effects on nitrate reductionwere separated from those on nitrate uptake by experiments usingtungstate. Tungstate pretreatment had no effect on NO3uptake ‘induced’ by N starvation, but inhibitedNO3 uptake associated with NO3 pretreatment. Chloridepretreatment similarly had no effect on NO3 uptake ‘induced’by N deprivation, but inhibited NO3 uptake followingNO3 pretreatment. The data suggest that there are atleast two mechanisms responsible for the ‘induction’of nitrate uptake by Chara cells, one associated with NO3reduction and ‘induced’ by CIO3 or NO3and one associated with N deprivation. Measurements of cavitation occurring in xylem conduits of differentstem parts in whole Chorisia insignis H.B. et. K. plants subjectedto water stress are reported. Pre-stressed plants were shownto undergo cavitation over 10 times greater than watered ones.The most vulnerable parts of plants were one-year-old twigswhere cavitation reached a peak of over 50 acoustic emissions(AE) min–1 while in two-year-old twigs AE min–1were about one half this value. Stem zones were found wherecavitation was typically very low even during water stress:these were one-year-old nodes and junctions where branches meet.Measurements of the inside diameters of xylem conduits and distributionof conduit ends in stem parts where AE were detected, showedthat nodes have a significantly larger percentage of narrowxylem conduits than internodes. Similar ‘constricted zones’were found injunctions with respect to two-year-old twigs. Hereabout 50 per cent of the xylem conduits were as narrow as 20to 50 µm in diameter. The distribution of xylem conduitends show about 3 per cent of them ending in the nodes and 1per cent in the internodes of one-year-old twigs. About 11.6per cent of xylem conduits end in the junctions and about ahalf in two-year-old internodes. Our data would give furtherexperimental evidence to the functional concept of ‘plantsegmentation’ into zones (internodes) more efficient inwater conduction, i.e. with wider xylem conduits but more vulnerableto cavitation and others (nodes and junctions) with oppositecharacteristics. Twenty-three genotypes of Indian mustard were studied for leafconductance (K1) at the floral bud initiation stage under non-stressconditions. The comparison of leaf conductance and of severalother measurements in the standard cultivar ‘Prakash’demonstrated that a screening procedure could be developed forleaf conductance prior to the flowering stage of mustard grownin the field. Errors due to short term environmental fluctuationsand to ontogenetic drift in conductance can be reduced if themeasurements are made on both the ad- and abaxial surfaces ofthe uppermost expanded leaf between 1200 h and 1400 h on cleardays. Irradiance levels can be neglected once a value of photosyntheticallyactive radiation (PAR) of about 1000 µE m–2 s–1at the adaxial surface or of 1200 µE m–2 s–1at the crop canopy has been reached. The water status of theplants is of no importance as long as the plants are not underundue stress. The different genotypes showed a genetic variabilityof 243% in leaf conductance.  相似文献

The prediction that very high seed yields of dry beans (Phaseolusvulgaris L.) would be produced by the delayed transplantingof large plants has been tested in a factorial experiment withfour dates of transplanting and eight plant populations. Therewere significant differences in yield between transplantingdates and between population densities, and there was a significantdate-density interaction. At low plant densities (up to about30 plants m–2) the three transplanted treatments yieldedless than the hand-sown controls, and late transplanting yieldedless than early. At the highest density the situation was reversed;all three transplanted treatments out-yielded the controls andlate transplanting tended to out-yield plants transplanted early.The biggest yield was 340 g seed m–2 from a transplantedcrop grown at 35 plants m–2. The data on yield fitted a modified rectangular hyperbola ofthe form where y is yield per unit area, p is the number of plants perunit area, t is the number of days between sowing and transplanting,and Bo, n, m, and p are arbitrary parameters. This equationaccounted for 91 per cent of the variation in yield with t andp. It is suggested that late transplanting had adverse effects,due to transplanting ‘shock’ and which were mostmarked at low plant densities; and beneficial effects, ascribableto an effect on plant ‘plasticity’, which were mostmarked at high plant densities. The relationship between the induction of tracheary elementdifferentiation and exogenous L-methionine was examined in agar-growncultures of soya bean callus initiated from Glycine max L. ‘Wayne’and ‘Clark 63’. Although Wayne is a normal cultivarsoya bean, seedlings of Clark 63 exhibit abnormal growth at25 °C due to exessive ethylene biosynthesis at this temperature.Wayne callus showed increased xylogenesis in the presence ofexogenous L-methionine (3.7 µg 1–1) in comparisonto IAA–KN controls at both 20 and 25 °C. Clark 63callus produced greater numbers of tracheary elements in responseto exogenous L-methionine only at 25 °C. The induction ofxylem differentiation was independent of the maintenance temperatureof the stock cultures of both cultivars. Xylogenesis initiatedbyan IAA–KN medium was inhibited by the addition of AgNO3(20 mg 1–1) to the extent of 76.5 per cent in cv. Wayneand 6 per cent in cv. Clark 63. The inhibitory effect was partiallyreversed by the addition of L-methionine (3.7 µg 1–1)to the IAA–KN–AgNO2 medium. The rates of CO2 incorporation into the epidermis of C. communiswere linear and were similar during the completion of opening(2 h) and closing (1 h) movements of stomata. The kinetics of14C turnover between metabolites and the rates of ‘leakage’of metabolites were determined for opening and closing movements.When stomata were opening there was a slow turnover of 14C frommalate chiefly into sugars. Upon stomatal closure 14C was initiallymainly in sugars, malate, and sugar phosphates. Thereafter,there was a slight loss of label from sugar phosphates witha corresponding increase in malate. Starch became labelled duringopening and closing movements. Rates of incorporation of CO2found in the ‘leakage’ fraction were greatest whenstomata were opening. Of the labelled compounds Most‘from the tissue, malate was the most highly labelled whetherstomata were opening or closing. Although interpretation of the turnover patterns is difficultwithout knowledge of pool sizes for the metabolites it is suggestedthat a pool of sugars exists within the guard cells, which havefairly direct and reversible access to carbon from starch andmalate. The implications of loss of malate from guard cellsduring stomatal opening and closing are discussed.  相似文献
