Summary Pseudomonas 135, a facultative methylotroph, was cultivated on methanol as a sole carbon and energy source for the accumulation of poly--hydroxybutyric acid (PHB). The cells grew fairly well on minimal synthetic medium containing 0.5% (v/v) of methanol at pH 7.0 and 30° C. The maximum specific growth rate was determined to be 0.26–0.28 h–1 with a growth yield of 0.38 in the optimized growth medium. For stimulation of PHB accumulation in the cells, deficiency of nutrients such as NH inf4 sup+ , Mg2+ and PO inf4 sup3– was crucial even though cell growth was significantly suppressed. The PHB content of a 40-h culture was determined to be 37% of the total cell mass in NH inf4 sup+ -limited medium, 42.5% on Mg2+-deficient medium, and 34.5% on PO inf4 sup3– -deficient medium. The maximum content of PHB in the cells could reach 55% in NH inf4 sup+ -limited fed-batch culture. The average relative molecular eight determined by gel permeation chromatography was 3.7 × 105 in NH inf4 sup+ -limited culture, 2.5 × 105 in Mg2+-deficientmedium, and 3.1 × 105 in PO inf4 sup3– -deficient medium. Polydispersity determined in each culture was relatively high (about 10–11). The solid PHB had a melting temperature of 173° C. Correspondence to: J. M. Lebeault  相似文献

The short-term effects of a simulated cattle dung pat on N2 fixation and total uptake of N in a perennial ryegrass/white clover mixture was studied in a container experiment using sheep faeces mixed with water to a DM content of 13%. We used a new 15N cross-labelling technique to determine the influence of dung-pat N on N2 fixation in a grass/clover mixture and the uptake of dung N in grass and clover. The proportion of N in clover derived from N2 fixation (%Ndfa) varied between 88–99% during the 16 weeks following application of the dung. There was no effect of dung on the %Ndfa in clover grown in mixture, whereas the %Ndfa in clover grown in pure stand decreased (nominal 2–3%) after dung application. Dung did not influence the amount of N2 fixed, and the uptake of dung N in grass and clover proceeded at an almost constant rate. After 16 weeks, 10% of the applied dung N was taken up by grass and clover, 57% had been incorporated in the soil by faunal activity and 27% remained in residual dung on the soil surface. The dung N unaccounted for (7%) was probably lost by ammonia volatilisation and denitrification. The uptake of dung N in grass/clover mixtures in the field was similarly followed by using simulated 15N-labelled dung pats. The total dry matter production and N yields increased in the 0–30 cm distance from the edge of the dung patch, but the proportion of clover decreased. Thirteen months after application of the dung 4% of the applied dung N was recovered in the harvested herbage, 78% was recovered from the soil and the residual dung, and 18% was not accounted for. It is concluded that N2 fixation in the dung patch border area in grass/clover mixtures is not influenced directly by the release of N from dung pats in the short term. However the amount of N2 fixed may be reduced, if the growth of clover is reduced in the patch border area.  相似文献

Aims: The purpose of this study was to determine the effect of moisture on thermal inactivation of Salmonella spp. in poultry litter under optimal composting conditions. Methods and Results: Thermal inactivation of Salmonella was studied in fresh poultry compost by simulating early phase of composting process. A mixture of three Salmonella serotypes grown in Tryptic soy broth with rifampin (TSB‐R) was inoculated in fresh compost with 40 or 50% moisture at a final concentration of c. 7 log CFU g?1. The inoculated compost was kept in an environmental chamber which was programmed to rise from room temperature to target composting temperatures in 2 days. In poultry compost with optimal moisture content (50%), Salmonella spp. survived for 96, 72 and 24 h at 50, 55 and 60°C, respectively, as compared with 264, 144 and 72 h at 50, 55 and 60°C, respectively, in compost with suboptimal moisture (40%). Pathogen decline was faster during the come‐up time owing to higher ammonia volatilization. Conclusions: Our results demonstrated that Salmonella spp. survived longer in fresh poultry compost with suboptimal moisture of 40% than in compost with optimal moisture of 50% during thermophilic composting. High nitrogen content of the poultry compost is an additional factor contributing to Salmonella inactivation through ammonia volatilization during thermal exposure. Significance and Impact of the Study: This research validated the effectiveness of the current composting guidelines on Salmonella inactivation in fresh poultry compost. Both initial moisture level and ammonia volatilization are important factors affecting microbiological safety and quality of compost product.  相似文献

To investigate the effect of sheep dung on soil carbon (C) sequestration, a 152 days incubation experiment was conducted with soils from two different Inner Mongolian grasslands, i.e. a Leymus chinensis dominated grassland representing the climax community (2.1% organic matter content) and a heavily degraded Artemisia frigida dominated community (1.3% organic matter content). Dung was collected from sheep either fed on L. chinensis (C3 plant with δ13C = −26.8‰; dung δ13C = −26.2‰) or Cleistogenes squarrosa (C4 plant with δ13C = −14.6‰; dung δ13C = −15.7‰). Fresh C3 and C4 sheep dung was mixed with the two grassland soils and incubated under controlled conditions for analysis of 13C-CO2 emissions. Soil samples were taken at days 17, 43, 86, 127 and 152 after sheep dung addition to detect the δ13C signal in soil and dung components. Analysis revealed that 16.9% and 16.6% of the sheep dung C had decomposed, of which 3.5% and 2.8% was sequestrated in the soils of L. chinensis and A. frigida grasslands, respectively, while the remaining decomposed sheep dung was emitted as CO2. The cumulative amounts of C respired from dung treated soils during 152 days were 7–8 times higher than in the un-amended controls. In both grassland soils, ca. 60% of the evolved CO2 originated from the decomposing sheep dung and 40% from the native soil C. Priming effects of soil C decomposition were observed in both soils, i.e. 1.4 g and 1.6 g additional soil C kg−1 dry soil had been emitted as CO2 for the L. chinensis and A. frigida soils, respectively. Hence, the net C losses from L. chinensis and A. frigida soils were 0.6 g and 0.9 g C kg−1 soil, which was 2.6% and 7.0% of the total C in L. chinensis and A. frigida grasslands soils, respectively. Our results suggest that grazing of degraded Inner Mongolian pastures may cause a net soil C loss due to the positive priming effect, thereby accelerating soil deterioration.  相似文献

Aim: The pathogen growth in dairy compost was studied in a greenhouse setting under different seasons. Methods and Results: The five‐strain mixtures of each Escherichia coli O157:H7, Salmonella spp. and Listeria monocytogenes were inoculated separately into dry compost to yield c. 1 log CFU g?1. After acclimation at room temperature, the inoculated compost was initially adjusted to moisture levels of 10–50% and then kept in a greenhouse under different seasons. The populations of all three pathogens increased by 2·1–3·9 log CFU g?1 within 3 days in autoclaved compost with initial moisture content of at least 40%. Listeria monocytogenes multiplied up to 2·4 log CFU g?1 in compost with initial moisture content of 30% and was detected up to 28 days for all seasons, whereas populations of both E. coli O157:H7 and Salmonella increased by c. 1 log in compost with initial moisture content of 30% during winter months only. No pathogen growth in nonautoclaved compost was detected. Conclusion: Bacterial species, temperature, light intensity and moisture content affected the growth potential and survival of pathogens in compost when the population of background microflora was low. Significance and Impact of the Study: Keeping compost as dry as possible and maintaining certain levels of background microflora may be critical to prevent the growth of pathogens.  相似文献

The effect of moisture on the decomposition rate of cattle manure   总被引:9,自引:0,他引:9
In an incubation study the effect of moisture content on CO2-C production from manure at 25°C was determined. Different non-linear regression models were applied to describe cumulative CO2-C evolution and a simple first-order model gave the best curve fit. Derived data from the curve fitting procedure were plotted against moisture content. A linear relationship between moisture content and microbial activity up to 50% of the water-holding capacity was followed by a curvilinear response between 50% of WHC and saturation. Equations describing the effect of moisture on dry matter decomposition are given.  相似文献

Experiments conducted on samples collected from a large oligotrophic lake revealed the following: (1) excretion rates of PO inf4 sup3– by single Daphnia thorata were below detection (5 pmol animal–1 min–1) in 20 ml of oligotrophic lake water over a period of 10 min, (2) experimental addition of D. thorata to 20 ml aliquots of lake water decreased community-wide microbial uptake of PO inf4 sup3– on two occasions (as measured by 32PO inf4 sup3– incorporation), and (3) the presence of D. thorata increased uptake by organisms smaller than 1µm, and decreased uptake by large phytoplankton. The specific mechanism for these responses remains unclear, but the results imply that when phytoplankton larger than 1µm encounter cm scale patches of water recently occupied by Daphnia they may experience decreased PO inf4 sup3– availability rather than elevated concentrations of PO inf4 sup3– caused by excretion. We show that 32P uptake experiments using natural plankton assemblages can be influenced by the presence or absence of large zooplankton, and that neither grazing, turbulence, nor PO inf4 sup3– excretion can account for this influence.  相似文献

Three field experiments were undertaken concurrently at one site to evaluate a range of surface-applied nBTPT-amended urea products (0.01, 0.05, 0.1, 0.25 and 0.5% nBTPT w/w) on NH3 volatilization, grass yield and 15N recovery in the plant-soil system. Each experiment was repeated on five separate occasions over the 1992 growing season to cover a range of weather conditions. Total NH3 loss from unamended-urea ranged from 5.5% in early May to 20.8% in June. The inhibitor was highly effective in reducing ammonia volatilization and delaying the time at which maximum rate of NH3 loss occurred. Over all time periods the % inhibition was 50.4, 82.8, 89.0, 96.5 and 97.0% at the 0.01, 0.05, 0.1, 0.25 and 0.5% nBTPT levels respectively. There was no significant difference in the overall % inhibition in ammonia loss at different times suggesting that the effectiveness of the inhibitor was not dependent on climatic conditions.Over all times incorporation of nBTPT at the 0.05% level increased dry-matter yield by 9% compared to urea alone and increased the shoot recovery of N from 66.7% to 80.9%. Nitrogen saved from volatilization was taken up by the plant, however, the subsequent translation into dry-matter yield appeared to be adversely affected at the high inhibitor rates. There was no significant effect of inhibitor on 15N recovery in soil at any depth down to 15 cms. nBTPT significantly increased (p < 0.001) the % N derived from fertilizer (% N dff) in the shoot compared to unamended-urea and increased (p < 0.01) the shoot recovery of 15N from 32% up to 39%. Total 15N recovery in the soil-plant system was increased by up to 17% by amending urea with nBTPT. This urease inhibitor has been shown to improve the efficiency of urea however, its potential for the European market will be dependent on economic factors.Faculty of Agriculture and Food Science, The Queen's University of Belfast  相似文献

Knowledge of N availability from organic amendments is a key to improve N use efficiency and reduce environmental pressure from agriculture. Nitrogen mineralization from 15N-labeled cattle dung compost and rapeseed cake was investigated under flooded and upland (60% of water holding capacity) conditions in an incubation experiment for 63 d at 25 °C. The relative abundance of N in the cattle dung compost by the simple step-wise acid hydrolysis method was in the following order: labile N (37% of total N, refluxing with 1 M HCl for 3 h, H1-N) > non-hydrolyzable N (32%) > recalcitrant N (18%, 3 M HCl for 3 h, H2-N). There was no significant difference in the 15N abundance between total N and N in each fraction of the cattle dung compost. For the rapeseed cake, the H1-N accounted for 81% of total N and the 15N abundance of total N and H1-N was higher than the 15N abundance of H2-N and non-hydrolyzable N. In the cattle dung treatment, inorganic 15N was the highest at 21 d of incubation and then decreased thereafter under flooded conditions, whereas it remained constant from 21 to 63 d under upland conditions. In the rapeseed cake treatment, inorganic 15N was the highest at 42 d under flooded conditions and inorganic 15N increased until 42 d and remained stable thereafter under upland conditions. The N mineralization rate from the cattle dung compost was slow both under flooded and upland conditions. More than half of N in the rapeseed cake was mineralized during the incubation period both under flooded and upland conditions. There was no significant difference in 15N recovery in the soil between flooded and upland conditions at 63 d in the cattle dung treatment, while the 15N recovery in the soil at 63 d was higher under upland than under flooded conditions in the rapeseed cake treatment. Although N mineralization from the rapeseed cake was greater under flooded conditions than upland conditions, there was no significant difference in N mineralization from the cattle dung compost between both conditions. Therefore, N mineralization from organic amendments is not always more rapid under flooded than upland conditions depending on the amendment type.  相似文献

Aims

Effects of different soil amendments were investigated on methane (CH4) emission, soil quality parameters and rice productivity in irrigated paddy field of Bangladesh.

Methods

The experiment was laid out in a randomized complete block design with five treatments and three replications. The experimental treatments were urea (220 kg ha?1) + rice straw compost (2 t ha?1) as a control, urea (170 kg ha?1) + rice straw compost (2 t ha?1) + silicate fertilizer, urea (170 kg ha?1) + sesbania biomass (2 t ha?1 ) + silicate fertilizer, urea (170 kg ha?1) + azolla biomass (2 t ha?1) + cyanobacterial mixture 15 kg ha?1 silicate fertilizer, urea (170 kg ha?1) + cattle manure compost (2 t ha?1) + silicate fertilizer.

Results

The average of two growing seasons CH4 flux 132 kg ha?1 was recorded from the conventional urea (220 kg ha?1) with rice straw compost incorporated field plot followed by 126.7 (4 % reduction), 130.7 (1.5 % reduction), 116 (12 % reduction) and 126 (5 % reduction) kg CH4 flux ha?1 respectively, with rice straw compost, sesbania biomass, azolla anabaena and cattle manure compost in combination urea and silicate fertilizer applied plots. Rice grain yield was increased by 15 % and 10 % over the control (4.95 Mg ha?1) with silicate plus composted cattle manure and silicate plus azolla anabaena, respectively. Soil quality parameters such as soil organic carbon, total nitrogen, microbial biomass carbon, soil redox status and cations exchange capacity were improved with the added organic materials and azolla biofertilizer amendments with silicate slag and optimum urea application (170 kg ha?1) in paddy field.

Conclusion

Integrated application of silicate fertilizer, well composted organic manures and azolla biofertilizer could be an effective strategy to minimize the use of conventional urea fertilizer, reducing CH4 emissions, improving soil quality parameters and increasing rice productivity in subtropical countries like Bangladesh.  相似文献

Corn-cob was used as a substrate in the production of oxytetracycline by Streptomyces rimosus TM-55 in a solidstate fermentation. Oxytetracycline was detected on day 4, and reached its maximum on day 8. Optimal conditions for oxytetracycline production were an initial pH of 5.2 to 6.3, an initial moisture content of 64% to 67%, supplementation with 20% (w/w) rice bran or 1.5% to 2.5% (w/w) (NH4)2SO4 as sole N source, 1.0% (w/w) CaCO3, 2% (w/w) MgSO4.7H2O, and 0.5% (w/w) KH2PO4, with incubation for 8 days at 25 to 30°C. Each g substrate produced 7 to 8 mg oxytetracycline.  相似文献

Fungi able to degrade agriculture wastes were isolated from different soil samples, rice straw, and compost; these isolates were screened for their ability to produce β-glucosidase. The most active fungal isolate was identified as Talaromyces pinophilus strain EMOO 13-3. The Plackett–Burman design is used for identifying the significant variables that influence β-glucosidase production under solid-state fermentation. Fifteen variables were examined for their significances on the production of β-glucosidase in 20 experimental runs. Among the variables screened, moisture content, Tween 80, and (NH4)2SO4 had significant effects on β-glucosidase production with confidence levels above 90% (p < 0.1). The optimal levels of these variables were further optimized using Box–Behnken statical design. As a result, the maximal β-glucosidase activity is 3648.519 U g?1, which is achieved at the following fermentation conditions: substrate amount 0.5 (g/250 mL flask), NaNO3 0.5 (%), KH2PO4 0.3 (%), KCl 0.02 (%), MgSO4 · 7H2O 0.01 (%), CaCl2 0.01 (%), yeast extract 0.07 (%), FeSO4 · 7H2O 0.0002 (%), Tween 80 0.02 (%), (NH4)2SO4 0.3 (%), pH 6.5, temperature 25°C, moisture content 1 (mL/g dry substrate), inoculum size 0.5 (mL/g dry substrate), and incubation period 5 days.  相似文献

Water-use efficiency (WUE) of Leptochloa fusca (L.) Kunth (Kallar grass) and Sporobolus arabicus Boiss. was determined under different soil moisture regimes. Plants grown in lysimeters were subjected to three soil moisture regimes, viz. well-watered (100%), medium-watered (75%), and low-watered (50%) of total available water (TAW). The soil moisture was restored on alternate days by adding the required volume of water on the basis of neutron moisture meter readings taken from neutron access tubes installed in each lysimeter. The grasses were harvested after suitable intervals (4 months) to obtain maximum biomass. Leaf samples collected at each harvest were analyzed for carbon-isotope discrimination (13C) with an isotope ratio (13C/12C) mass spectrometer. Results indicated significant differences in WUE of both grasses subjected to different water regimes. Sporobolus arabicus showed higher WUE than Kallar grass. However, Kallar grass showed better value of yield response factor (k y = 0.649) compared with Sporobolus (k y = 1.06) over the entire season. The data confirm that these grasses can be grown successfully in water-limited environments by selecting an optimum soil moisture level for maximum biomass production. The mean carbon-isotope discrimination (13C) of Kallar grass (–14.4) and Sporobolus (–12.8) confirm that both are C4 plants. The carbon-isotope discrimination () was significantly and negatively correlated with WUE of the two species studied. The results of the present study confirm that 13C or of leaves can be used as good predictor of WUE in some C4 plants.  相似文献

The effect of shock-loading of zinc, copper and cadmium ions on the removal of total organic carbon (TOC) and phosphate in an anaerobic-aerobic activated sludge process was investigated. TOC removal was not sensitive to shock-loading of Zn2+ and Cd2+ ions, and complete removal was achieved even at 20 mg Zn2+/l and 20 mg Cd2+/l. However, with over 1 mg Cu2+/1 TOC removal efficiency decreased. PO inf4 sup3- removal, in contrast, was extremely sensitive to these metal ions, with the threshold being 1 mg Zn2+/l and 1 mg Cd2+/l. Higher concentrations adversely affected PO inf4 sup3- removal. Copper again proved detrimental; no PO inf4 sup3- removal was achieved even at 1 mg Cu/l. These results highlight the sensitivity of the removal efficiencies of TOC and PO inf4 sup3- to shock loadings of these heavy metals.Y.P. Summary Effects of fire and forest harvesting on inorganic-N in the soil, on net N-mineralization, and on the leaching of NO inf3 sup- -N and metallic cations were measured in forests of Eucalyptus regnans following a severe wildfire in 1983. E. regnans regenerates profusely by seed after fire, and this study compared unburnt forest with forests burnt at varying intensities (surface fire and crown fire), and with logged and burnt forest (slash fire). Total inorganic-N in soil (0–5 cm) increased with increasing fire intensity to a maximum of 158 g g-1 in the slash fire plot (compared with 51 g g-1 in the unburnt forest) over the first 205 days after fire. Total inorganic-N returned to a concentration equal to that in the unburnt forest after 485 days at the slash fire plot, and after only 205 days at the surface fire plot. Studies of net mineralization in situ and of NO inf3 sup- -N in soil solution support the hypothesis that inorganic-N was immobilized in all of the burnt forests; microbial immobilization after fire is identified as a key process in N-conservation, limiting the substrate available for nitrification and thereby limiting the loss of N from the system by leaching. The concentrations of NO inf3 sup- -N and metallic cations in soil solution increased with increasing fire intensity. For the first 318 days after the fire, [NO inf3 sup- -N] in soil solution at 10 cm averaged 0.6 g ml-1 in the unburnt forest, 9.7 mg l-1 in the surface fire plot, 26 mg l-1 in the crown fire plot, and 70 mg l-1 in the slash fire plot. The concentration of metallic cations in soil solution was significantly correlated with [NO inf3 sup- -N], the observed order of mobility being Ca2+>Mg2+>K+>Na+. Processes which limit the production and persistence of NO inf3 sup- -N in soil solution following disturbance will significantly reduce nutrient losses or redistribution.  相似文献

Diurnal variation in hydrological variables and dissolved inorganic nutrients such as PO inf4 sup3– -P, N O inf2 sup– -N, NO inf3 sup– -N and NH inf4 sup+ -N were studied in three interconnected biotopes including freshwater, marine and mangrove brackish water of the Kakinada coastal zone, Andhra Pradesh. Samples were collected at intervals of 3 hours, for a period of 24 hours. In the marine environment salinity varied from 26 to 32 whereas in the mangrove waters it fluctuated from 12 to 20 and in both biotopes salinity showed bimodal type of oscillation. Dissolved oxygen content was high in the mangrove waters during day time but decreased rapidly during the night hours. In the marine environment POf4 p3–-P concentration varied from 0.345 to 1.195 g at l–1, NO inf3 sup– -N from 1.03 to 6.62 g at l–1 and NO inf2 sup– -N from 0.086 to 0.506 g at l–1. The highest and the lowest concentrations of PO inf4 sup3– -P, NO inf3 sup– -N, NO inf2 sup– -N recorded in the mangrove waters were 0.790 and 0.325 g at l–1, 7.10 and 1.60 g at l–1 and 0.278 and 0.060 g at l–1, respectively. The concentration of PO inf4 sup3– -P, NO inf3 sup– -N and NO inf2 sup– -N were high in the freshwater canal, the maximum and minimum values being 1.110 and 0.730 g at l–1, 26.40 and 9.98 g at l–1 and 0.520 and 0.252 g at l–1 respectively. The concentration of ammonia was relatively high in the mangrove water. Gross and net primary production in the mangrove water was 4 times higher than in the marine biotope. There was no export of dissolved nutrients from the mangrove environment to the adjacent marine waters.  相似文献

Composts are considered low analysis fertilizers because their nitrogen and phosphorus content are around 1% and the organic nitrogen mineralization rate is near 10%. If compost is added to agricultural land at the N requirement of grain crops (40 – 100 kg N ha–1), application rates approach 40–100 mg ha–1. Much lower rates may be advisable to avoid rapid accumulation of growth limiting constituents such as heavy metals found in some composts. Combining low amendment rates of composts with sufficient fertilizer to meet crop requirements is an appealing alternative which (a) utilizes composts at lower rates than those needed to supply all the crop N requirement, (b) reduces the amount of inorganic fertilizer applied to soils, and (c) reduces the accumulation of non-nutrient compost constituents in soils. A study was conducted to compare the effects of blends of biosolids compost (C) with 15N urea(U) or 15NH4 15NO3 (N) fertilizers to fertilizer alone on tall fescue (Festuca arundinacea L.) growth and N uptake. Blends which provided 0, 20, 40 or 60 mg N kg–1 application rate as compost N and 120, 100, 80 or 60 mg N kg–1 as fertilizer N, respectively, were added to Sassafras soil (Typic Hapludults). Fescue was grown on the blends in a growth chamber for 98 days. Fescue yields recorded by clippings taken at 23, 46 and 98 days and roots harvested after the 98-day clipping increased with increasing fertilizer level for both NH4NO3 and urea and with or without compost. Nitrogen uptake by fescue responded similarly to yield with increases recorded with increasing fertilizer levels with or without compost. Paired comparisons based on cumulative 98-day clippings data showed that yields from blends were equal to yields from fertilizer treatments containing the same percentage of fertilizer as the blends. These data indicated that compost did not provide sufficient plant-available N to increase yields or N uptake. None of the blends equaled 120 mg N kg–1 fertilizer rate except for 100 mg NH4NO3-or urea-N kg–1 –20 mg compost-N kg–1blends. The data suggest that biosolids compost blended with fertilizer at a rate of 2–6 mg ha –1 did not supply sufficient additional available N to increase yields or N uptake over those of fertilizer alone.  相似文献

The effects of slow (apatite, biotite) and fast-release nutrients (P, K, Mg) on C and N mineralization in acid forest soil were studied. These nutrients were applied alone or together with urea or urea and limestone. The production of CO2 in the soil samples taken one and three growing seasons after the application was lower in the soils treated with the fast-release nutrients than in the untreated soils. Similar reduction of microbial activity was not seen after the apatite and apatite+biotite treatments. In the first growing season, urea and urea+limestone enhanced CO2 production, but after three growing seasons, the opposite was true. Apatite and apatite+biotite added together with urea did not compensate for the decreasing effect of urea on the CO2 production. The addition of fast-release salts increased somewhat the concentration of NH inf4 sup+ in the soil and more NH4 + accumulated during laboratory incubation in the soil samples taken one growing season after the application. The urea addition immediately increased the concentrations of NH4 + and of NO3 in the soil, but, three growing seasons after application, urea had only a slight increasing effect on mineral N content of the soil. Slow-release nutrients seem to have a more favourable effect than fast-release salts on nutrient turnover in acid forest soil.  相似文献

Methanosarcina frisia accumulates phosphate up to 14% of its dry weight. The phosphate is stored as long-chain polyphosphates as shown by 31P-NMR investigations. Further results show that the accumulation of phosphates is substrate-dependent. In the presence of H2 and CO2 as the only carbon and energy source 180 mg of PO inf4 sup3- /g protein were accumulated, whereas 260 mg PO inf4 sup3- /g protein were accumulated in the presence of methanol. This is far more than necessary for the maintenance of essential metabolic pathways. In addition, the 31P-NMR studies show the occurrence of cyclic 2,3-diphosphoglycerate in Methanosarcina frisia. The role of the phosphate metabolites in cell metabolism are discussed.Abbreviations M. Methanosarcina - CCP cyclic 2,3-diphosphoglycerate  相似文献
