The distribution of mitochondria, their content and concentration (expressed as the ratio of the mean volume of mitochondria and the surface of the sensory axon) were determined in group-III and-IV nerve fibres innervating the knee joint capsule in the cat. Mitochondria mainly accumulated in axonal swellings (beads) and end bulbs of the terminal branches. Between single nerve fibres, marked differences in the content and the concentration of mitochondria were obtained in proximal portions (inside of the perineurium) and in distal portions (unmyelinated sensory endings). In group-III nerve fibres, the mitochondrial concentration ranged from 0.005 to 0.030 m3/m2 (proximal portion) and from 0.016 to 0.080 m3/m2 (distal portion). In unmyelinated group-IV nerve fibres, the values also showed a broad variation ranging from 0.001 to 0.011 m3/m2 (proximal portion) and from 0.003 to 0.019 m3/m2 (distal portion). The wide range of mitochondrial concentrations may reflect different energy consumption during receptive processes: nerve fibres with a low mechanical threshold and a high probability of excitatory events may be rich in mitochondria, whereas fibres with a high mechanical threshold and a low probability of excitatory events may be poor in mitochondria.  相似文献

Whilst reliable estimates of chiasma frequencies can usually not be obtained, the probability (b) of a chromosome arm to be bound by at least one chiasma can often be determined. In the absence of interference this probability equals (1–e –2), where 2 is the average chiasma frequency of the chromosome arm and the average crossover frequency or map length. In the presence of interference is shown to retain its genetic meaning as an additive metric that may describe the chromosome arm or other distinctive chromosome segment in terms of genetic recombination. It is a form of potential map length, comparable to, but numerically different from the regular map length. It is termed provisionally crossing-over potential.A chromosome with armsm andn with crossing-over potentials and will form ring bivalents with a frequency (1–e –2).(1–e –2); open bivalents with a frequency (1–e –2).e –2+(1–e –2).e –2; univalent pairs with a frequencye –2.e –2. Estimates of these frequencies yield equations from which and may be solved. In rye (Secale cereale) their ratio (q) is approximately two and differs from the mitotic arm length ratio of 1.4, indicating localization of chiasmata in the long arms.Graphs are given to show how, with constantq, the relation between the probabilitiesb m andb n of the two arms being bound changes with changing averageb.Data are presented on chiasma frequencies in M I, and compared with the frequencies expected in the absence of interference to give an impression of the degree of interference. Summary A method for the determination of exchangeable lithium using 0.5M NH4Cl is described. The range of exchangeable Li in the fifty Papua New Guinea (PNG) soils analyzed was 0.002 to 0.409 gg–1 in contrast to five Australian soils which ranged from 0.032 to 0.830 gg–1. The PNG soils were divided into hill and alluvial soils with average exchangeable Li contents of 0.062 and 0.263 gg–1 respectively. No significant correlation between total and exchangeable Li was found in either group of soils althoughr=0.67 for the comined data and was significant at the 5% level. From the analysis of three profiles exchangeable Li was found to be at least twice as high (0.27 gg–1) in surface soils as in subsurface samples (0.10 gg–1). The average value of the deeper subsoil samples was 0.18 ppm.R mode cluster analysis of the data for village garden soils collected on a sampling grid showed that exchangeable Li was more strongly assoicated with Ca and Mg than with pH, 0.05M EDTA soluble Zn, 0.5M NaHCO3 soluble P or exchangeable Na and K. Computer constructed isographs using the analyses of grid samples from a garden illustrated the association between Li, Ca and Mg and the inverse association with Na.The correlation coefficient between Ca and Li in the ash of three food plants (Gnetum gnemon, Hibiscus abelmoschus andStenochlaena plustris) while not significant on an individual basis, was significant when the data was combined suggesting that the association between these elements in the soil may reflect an association in the ash returned to the soil when the garden was cleared. The correlation coefficient between soil exchangeable Li and Li in plant ash was positive, but not significant.Adsorption experiments over a five-day period demonstrated that Li was strongly adsorbed from solution. On average 63–75% of the adsorbed Li was fixed in a form which was not exchangeable with 0.5M NH4Cl or soluble in 0.05M EDTA.  相似文献

Microbiological and biogeochemical investigations of the processes of methane production (MP) and methane oxidation (MO) in the coastal waters and littoral of the Kandalaksha Bay of the White Sea were carried out. The studies were conducted in the coastal zones and in the water areas of the Kandalaksha Preserve, Moscow State University White Sea Biological Station, and the Zoological Institute (RAS) biological station in August 1999, 2000, and 2001 and in March 2001. The rate of CO2 assimilation in the shallow and littoral sediments was 35–27800 g C/(dm3 day) in summer and 32.8–88.9 g C/(dm3 day) in winter. The maximal rates of MP were observed in the littoral sediments in the zone of macrophyte decomposition, in local depressions, and in the estuary of a freshwater creek (up to 113 l/(dm3 day)). The maximal level of MO was observed in the shallow estuarine sediments (up to 2450 l/(dm3 day)). During the winter season, at the temperature of –0.5 to 0.5°C, the MP rate in the littoral sediments was 0.02–0.3 l/(dm3 day), while the MO rate was 0.06–0.7 l/(dm3 day). The isotopic data obtained indicate that the Corg of the mats and of the upper sediment layers is enriched with the heavy 13C isotope by 1–4 as compared to the Corg of the suspension. A striking difference was found between the levels of methane emission by the typical littoral microlandscapes. In fine sediments, the average emission was 675 l CH4/(m2 day); in stormy discharge stretch sediments, it was 1670 l CH4/(m2 day); and under stones and in silted pits, 1370 l CH4/(m2day). The calculation, performed with consideration of the microlandscape areas with a high production, allowed the CH4 production of 1 km2 of the littoral to be estimated as 192–300 l CH4/(km2 day).  相似文献

Summary Free-living or immobilized Chlamydomonas reinhardtii cells photoproduce ammonium from nitrite in a medium containing 1 mM of l-methionine-d,l-sulphoximine (MSX). Ammonium is accumulated in the medium to 8 mM final concentration, which inhibits nitrite uptake by the MSX-treated cells and consequently the excretion of ammonium is blocked. However, if ammonium was removed from the medium and nitrite and MSX periodically restored, the photoproduction process could be maintained over 96 h, with a final ammonium concentration of about 18 mM for free-living cells and 28 mM for immobilized ones. The MSX-treated cells showed a photoproduction productivity of 1300 mol NH 4 + · mg chlorophyll (Chl)-1, with an average production rate of 14 mol NH 4 + · mg Chl-1 per hour, for calcium alginate-entrapped cells, while the corresponding data for free-living ones was 650 mol NH 4 + · mg Chl-1 and 6.7 mol NH 4 + · mg Chl-1 per hour, respectively. Immobilized cells showed a significant increase in the nitrite uptake rate, probably due to a change in membrane permeability as a consequence of cell-matrix interactions.  相似文献

The effects of plant growth regulators, light intensity, and end-of-day (EOD) light quality treatments on node and microtuber induction (% of cultures with microtubers) and development (fresh weight of microtubers) in yam (Dioscorea alata L. cv. Oriental) cultures were investigated. Nodal segments were excised from plantlets cultured on tuberization medium containing growth regulators and exposed to various light treatments. Absciscic acid (1 M) stimulated and cytokinins (2.5 M) inhibited microtuber development from yam nodal segments cultured on Mantell's and Hugo's full-strength tuberization medium under 8-h photoperiods. EOD far-red (FR) light inhibited microtuber induction and development and enhanced node formation. EOD FR light effects were nullified by immediately following the FR treatment with red light. This suggested the involvement of phytochrome in these processes. The lowest light intensity evaluated (12 mol m–2 s–1) inhibited microtuber, root and shoot production as compared to light intensities of 42, 72 and 102 mol m–2 s–1. Toxicity of 14 selected acaricides was tested for a laboratory strain of femaleD. gallinae. The most toxic were carbaryl (LC50=5.0 g/m2), deltamethrin (LC50=7.8 g/m2), bendiocarb (LC50=11.1g/m2), and permethrin (LC50=12.6 g/m2). High level resistance to DDT inD. gallinae was recorded, with factors of resistance at LC50/ LC90 reaching 57/73.  相似文献

In order to determine the relative importance of autotrophic and heterotrophic activities in both bacterial and phytoplanktonic communities in an oligomesotrophic lake, the size fractionation by differential filtration and the use of a bacterial inhibitor (gentamycin) were combined. The study was carried out at Lake Pavin during the spring planktonic bloom. Photosynthetic and photo- and chemoheterotrophic activities were measured from the assimilation of NaH14CO3 and glucose-3H, using a double labeling technique. The bacterial community was at low cell concentration (0.6 to 7 × 105 cells ml) and represented very low biomass values (0.9 to 11.5 gC liter–1). The abundance of the phytoplankton varied between 0.5 and 1.8 × 106 cells liter–1, and biomass values ranged between 19 and 118 gC liter–1. The diatom Melosira italica sp. subarctica (O. Mueller) was the largely dominant species in the meta- and hypolimnion. Inorganic fixation by photolithotrophy (mean value: 1.66 mg C m–3 hour–1) largely predominates over assimilation by photoheterotrophy (mean value: 0.93 g C m–3 hour–1) or chemoheterotrophy (mean value: 2.42 g C m–3 hour–1). However, because of the considerable underestimation of heterotrophic assimilation due to the experimental methods used, and because of the spatial and temporal separation of photolithotrophic and photo- and chemoheterotrophic activities, it is likely that the fixation of organic carbon by microalgae plays an important role in the survival of species and/or in competitive interactions, as the results with Monoraphidium contortum (Pasch. et Korschik.), the prevailing species in the epilimnion, would suggest. Summary The wet and dry weights of both axenic and monoxenic cultures of the ciliate Tetrahymena were determined directly. These estimates are dependent upon the method of volume determination. Assuming a prolate spheroidal shape for the ciliate, we calculate a mean wet weight of 0.4157±0.0713 pg m-3 and a mean dry weight of 0.2793±0.0652 pg m-3. Using electronic cell sizing, our estimates are 0.7869±0.1659 pg m-3 and 0.5239±0.1101 pg m-3, respectively. Independent of the method of volume determination, we estimate a mean biomass conversion ratio (dry weight/wet weight) of 0.59±0.08.  相似文献

Cr(VI) tolerance was studied in four strains of Rhodosporidium toruloides and compared with that of a fifth strain, DBVPG 6662, isolated from metallurgical wastes and known to be Cr(VI) resistant. Tolerance was studied in relation to different species of sulfur (sulfates, thiosulfates, methionine, cysteine) at different concentrations. Djenkolic acid, a poor source of sulfur and an activator of sulfate transport, was also considered. In synthetic medium all strains except the Cr(VI)-resistant one started to be inhibited by 10 g ml (0.2 mm) Cr(VI) as K2Cr2O7. DBVPG 6662 was inhibited by 100 g ml (2.0 mm) Cr(VI). In Yeast Nitrogen Base without amino acids (minimal medium), supplemented with varying concentrations of chromate, all Cr(VI)-sensitive strains accumulated concentrations of total chromium (from 0.8 to 1.0 g mg cell dry wt) after 18 h of incubation at 28 °C. In minimal medium supplemented with 10 g ml Cr(VI), the addition of sulfate did not significantly improve the yeast growth. Cysteine at m levels increased tolerance up to 10 g ml, whereas methionine only reduced the Cr(VI) toxicity in the strain DBVPG 6739. Additions of djenkolic acid resulted in increased Cr(VI) sensitivity in all strains. The best inorganic sulfur species for conferring high tolerance was thiosulfate at concentrations up to 1 mm. In all cases increased Cr(VI) tolerance was due to a significantly reduced uptake in the oxyanion by the cells and not to the chemical reduction of Cr(VI) to Cr(III) by sulfur compounds.  相似文献

We determined for Vicia faba L the influence of nitrogen uptake and accumulation on the values of photon saturated net photosynthetic rate (P Nmax), quantum yield efficiency (), intercellular CO2 concentration (C i), and carboxylation efficiency (C e). As leaf nitrogen content (NL) increased, the converged onto a maximum asymptotic value of 0.0664±0.0049 mol(CO2) mol(quantum)–1. Also, as NL increased the C i value fell to an asymptotic minimum of 115.80±1.59 mol mol–1, and C e converged onto a maximum asymptotic value of 1.645±0.054 mol(CO2) m–2 s–1 Pa–1 and declined to zero at a NL-intercept equal to 0.596±0.096 g(N) m–2. fell to zero for an NL-intercept of 0.660±0.052 g(N) m–2. As NL increased, the value of P Nmax converged onto a maximum asymptotic value of 33.400±2.563 mol(CO2) m–2 s–1. P N fell to zero for an NL-intercept of 0.710±0.035 g(N) m–2. Under variable daily meteorological conditions the values for NL, specific leaf area (L), root mass fraction (Rf), P Nmax, and remained constant for a given N supply. A monotonic decline in the steady-state value of Rf occurred with increasing N supply. L increased with increasing N supply or with increasing NL.  相似文献

Summary The ommatidia in the dorsal eye of male Bibio marci (March flies) are comprised of eight retinula cells (R1–8). In the distal region, the open rhabdomeres of retinula cells 1–6 are arranged in a symmetrically circular pattern with their microvilli directed radially. Immediately beneath the crystalline cone, cell 7 forms a rhabdomere that is about 1 m long and lies in the center of the circle formed by the rhabdomeres of cells 1–6. For the remaining length of an ommatidium it is replaced by the rhabdomere of retinula cell 8. The cell body of this retinula cell almost encloses its own rhabdomere by forming a deep invagination. Consequently, no ommatidial cavity is present. In the left eye rhabdomeres R 3, 5 and 6 first twist clockwise along their longitudinal axes, while rhabdomeres R1, 2, 4 and 8 twist counterclockwise. Opposite twisting is observed in the right eye. The twist rate varies along the length of the rhabdomeres. In a middle region of 60 m, within which the direction of twist does not change, the maximal twist rates are approximately 2°–5°/m in R1–6 and even higher in R 8. In a proximal region, the direction of twist is reversed, but the initial orientation of the microvilli not reestablished. Both the cross-sectional shape of the rhabdomeres and their geometric arrangement in the retinula change along with the twisting. Synopsis Foraging behaviour of lake charr, Salvelinus namaycush, exposed to 0.5 and 5.0 g l–1 cadmium concentrations for 106–112 d, was examined in a flow-through stream channel using fathead minnows, Pimephales promelas, as prey. Five modes of foraging behaviour were observed and quantified (ending at fixate-orient; ending at approach and/or chase; ending at failed attack; ending at capture and loss; successful consumption). Each mode represented an increasingly progressive end-point from initial sighting of the prey to final consumption. Compared to controls., 0.5 g l–1 Cd-exposed predators captured significantly fewer prey, while 5 g l–1 Cd-exposed predators made fewer attacks. Both Cd-exposed groups consumed fewer prey than controls.  相似文献

Cotyledonary explants of 4-day-oldCucumis melo cv. Hale's Best Jumbo in vitro seedlings showed maximum initiation of shoot buds when cultured onto a revised Murashige & Skoog medium supplemented with 5 M indole-3-acetic acid and 5 M benzylaminopurine and cultured at 25–29°C under low light intensity (5–30 mol m-2 s-1). Subculture of the shoot buds onto the same medium without auxin and supplemented with 3 M benzylaminopurine caused the development of shoots from 30% of the buds. The presence of abscisic acid significantly increased the number of explants producing shoot buds. Bud initiation was affected by genotype, seedling age, light intensity, and temperature. Addition of gibberellic acid, thidiazuron or silver nitrate to regeneration medium did not improve either bud initiation or shoot regeneration.  相似文献

Phytotoxicity and inhibitory effects of the fusarial toxins fumonisin B1 (FB1) [m.p. 103–105 °C], fusaric acid [m.p. 106–107 °C], butenolide (4-acetamido-4-hydroxy-2-butenoic acid lactone) [116–117 °C], 9, 10-dihydroxyfusaric acid [m.p. 150–155 ° C], and moniliformin on chlorophyll synthesis in the aquatic macrophyte Lemna minor (duckweed) were examined. FB1 proved to be most active, reducing the growth of L. minor fronds and their ability to synthesize chlorophyll by 53% and 59%, respectively, at 0.7 g/ml. The growth rate of L. minor was reduced 59% by 6.7 g/ml fusaric acid, 62% by 66.7 g/ml butenolide, and 22% by 66.7 g/ml 9,10-dihydroxyfusaric acid. Moniliformin was the least phytotoxic to L. minor, with only a 16% suppression of growth rate and a 54% reduction in chlorophyll at 66.7 g/ml.The mention of firm names or trade products does not imply that they are endorsed or recommended by the US Department of Agriculture over other firms or similar products not mentioned.  相似文献

The effect of lowered concentrations of carbon, nitrogen, and phosphorus sources in the medium on the specific growth rate of the R, S, and M dissociants of the hydrocarbon-oxidizing strain Pseudomonas aeruginosaK-2, culture pH, and the population composition was studied. Within the first 16 hours of cultivation in all of the four media tested, the R, S, and M dissociants have virtually identical . The maximal values of were reached by the 20th h of growth in the basal medium (R and S dissociants) and in the carbon-deficient medium containing 0.4% glucose (M dissociant). The R and M dissociants showed the most rapid decrease in in the nitrogen-deficient medium containing 0.55% NaNO3. By the end of cultivation in the basal medium, the pH of the R, S, and M cultures decreased to 6.3, 5.3, and 3.3, respectively. In the case of the carbon-deficient medium, the drop in the culture pH was lower. After a 2.5-day incubation of the S dissociant in the phosphorus- deficient medium containing 0.028% NaH2PO4· 2H2O and of the M dissociant in the basal medium supplemented with chalk powder, these dissociants were completely displaced from the media.  相似文献

A toxic factor released from disrupted cells of Vibrio parahaemolyticus was partially purified by gel filtration after precipitation with (NH4)2SO4 at 40% saturation. The factor, which was a thermostable protein of 63 kDa, lysed human erythrocytes at a concentration of 0.15 g ml-1. Its LD50 by intravenous injection into mice was 6.4 g. Fluid accumulated in suckling mice force-fed with the toxic material (1 to 25 g). Haemolytic activity, which occurred maximall at 37°C and pH 7.0 was enhanced by Ca2+, Cu2+ and Zn2+, each at 1 mm. Anti-toxic-factor serum agglutinated V. parahaemolyticus cells. The factor may play a role in the pathogenesis of V. parahaemolyticus infections and in the host's defence mechanisms against infection by the microorganism.  相似文献

Thylakoids isolated from cells of the red alga Porphyridium cruentum exhibit an increased PS I activity on a chlorophyll basis with increasing growth irradiance, even though the stoichiometry of Photosystems I and II in such cells shows little change (Cunningham et al. (1989) Plant Physiol 91: 1179–1187). PS I activity was 26% greater in thylakoids of cells acclimated at 280 mol photons · m–2 · s–1 (VHL) than in cells acclimated at 10 mol photons · m–2 · s–1 (LL), indicating a change in the light absorbance capacity of PS I. Upon isolating PS I holocomplexes from VHL cells it was found that they contained 132±9 Chl/P700 while those obtained from LL cells had 165±4 Chl/P700. Examination of the polypeptide composition of PS I holocomplexes on SDS-PAGE showed a notable decrease of three polypeptides (19.5, 21.0 and 22 kDa) in VHL-complexes relative to LL-complexes. These polypeptides belong to a novel LHC I complex, recently discovered in red algae (Wolfe et al. (1994a) Nature 367: 566–568), that lacks Chl b and includes at least six different polypeptides. We suggest that the decrease in PS I Chl antenna size observed with increasing irradiance is attributable to changes occurring in the LHC I-antenna complex. Evidence for a Chl-binding antenna complex associated with PS II core complexes is lacking at this point. LHC II-type polypeptides were not observed in functionally active PS II preparations (Wolfe et al. Nodal stem segments of flowering dogwood (Cornus florida L.) were cultured on media containing seven different sources of macronutrients including full- and half-strength Murashige & Skoog (MS) macrosalts, N6, Anderson's (AND), Quoirin & Lepoivre's (LP), Nitsch & Nitsch (NIT), and Woody Plant Medium (WPM). All media contained MS micronutrients, Staba vitamins, 20 g l-1 sucrose, and 6.5 g l-1 Difco Bacto-agar, and were supplemented with 2.2 M 6-benzyladenine (BA) and 0.49 M indole-3-butyric acid (IBA). Following three subcultures, the best shoot proliferation was supported on media containing WPM macronutrients. To optimize the proliferation rate, shoots were cultured on WPM macronutrients supplemented with eight combinations of BA and IBA, and 3.3 M BA without IBA was determined to be the best.Abbreviations BA 6-benzyladenine - IBA indole-3-butyric acid  相似文献

Four new species of Isospora are described from Australian geckoes. Isospora gehyrae n. sp. from Gehyra cf. variegata in South Australia have 18.5-22.5×17.5–20.0 m oöcysts with 10.0-12.5×7.5-9.0 m sporocysts; endogenous stages develop in the host cell cytoplasm. Of the two species found in Heteronotia binoei from northern Queensland, Isospora cytoheteronotis n. sp., with oöcysts of 20.0-26.0×17.5-25.0 m and sporocysts of 10.0-13.5×7.5-11.5 m, undergoes endogenous development in its host cell cytoplasm, whereas I. nucleoheteronotis n. sp., with oöcysts of 17.5-22.5×17.5-21.5 m and sporocysts of 9.0-12.5×6.5-10.0 m, develops in the host cell nucleus. I. oedurae n. sp. from Oedura rhombifer in northern Queensland has oöcysts of 22.5-25.0×22.5-24.0 m and sporocysts of 12.5-14.0×7.5-11.5 m, and undergoes endogenous development in its host cell nuclei.  相似文献
