The effect of pH and substrates on the binding of radiolabelled α-cyanocinnamate to mitochondria and submitochondrial particles has been investigated. It has been found that the binding is strongly influenced by the pH of the medium (it decreases on increasing the pH of the medium). The inhibition of pyruvate oxidation by this inhibitor follows the same pH dependence. The pH affects only the affinity of the α-cyanocinnamate binding site without changing their total number. A similar pH dependence has been found in inside-out submitochondrial particles where the binding sites are directly accessible. The quantitative parameters of the binding of α-cyanocinnamate in submitochondrial particles have been determined. The binding can be prevented or displaced by pyruvate and other substrates of the carrier. The turnover number for pyruvate transport in rat-heart mitochondria has been determined.  相似文献

A study is presented of the effect of acidic uncouplers and oligomycin on energy-linked and passive proton translocation, oxidative phosphorylation, and energy-linked nicotinamide-adenine-nucleotide transhydrogenase in EDTA submitochondrial particles from beef-heart. A flow potentiometric technique has been applied to resolve the kinetics of the initial rapid phase of the redox proton pump. Rapid kinetics analysis shows that carbonyl-cyanide-p-trifluoromethoxyphenyl-hydrazone (FCCP) does not exert any direct effect on redox-linked active proton transport. The uncoupling action of FCCP on oxidative phosphorylation and energy-linked transhydrogenase is shown to be quantitatively accounted for by its promoting effect of passive proton-diffusion across the mitochondrial membrane. Oligomycin depresses passive proton diffusion in EDTA sonic particles and this effect accounts for the coupling action exerted by the antibiotic on oxidative phosphorylation and energy-linked transhydrogenase. In fact, rapid kinetic analysis demonstrates that oligomycin does not directly affect the redox-linked proton pump. The present results show that there does not exist any labile intermediate in the redox-linked proton pump which is sensitive to acidic uncouplers.  相似文献

Mitochondrial membranes were incubated with NN'-dicyclohexyl[(14)C]carbodi-imide, which irreversibly inhibited the partial reactions of oxidative phosphorylation by 95-100%. Solutions of the membranes were analysed on polyacrylamide gels. Of the radioactivity recovered from the gels 90% was shown to be associated with a single protein of molecular weight about 10000. The radioactive protein and associated phospholipid was solubilized from the membrane by extraction with chloroform-methanol mixtures and was concentrated 50-fold by solvent fractionation and adsorption chromatography on Sephadex LH-20. Several protein-radioactivity peaks were obtained by Sephadex LH-20 chromatography. However, 90-100% of the radioactivity in each peak was shown to be associated with a single protein similar to the major radioactive protein observed in electrophoretograms of the membrane solutions. It is concluded that dicyclohexylcarbodi-imide inhibits mitochondrial oxidative phosphorylation by reacting covalently with a group on this chloroform-methanol-soluble protein. The possible role of this protein in oxidative phosphorylation is discussed.  相似文献

A novel class of inhibitors of the enzyme γ-glutamyl transpeptidase (GGT) were evaluated. The analog OU749 was shown previously to be an uncompetitive inhibitor of the GGT transpeptidation reaction. The data in this study show that it is an equally potent uncompetitive inhibitor of the hydrolysis reaction, the primary reaction catalyzed by GGT in vivo. A series of structural analogs of OU749 were evaluated. For many of the analogs, the potency of the inhibition differed between the hydrolysis and transpeptidation reactions, providing insight into the malleability of the active site of the enzyme. Analogs with electron withdrawing groups on the benzosulfonamide ring, accelerated the hydrolysis reaction, but inhibited the transpeptidation reaction by competing with a dipeptide acceptor. Several of the OU749 analogs inhibited the transpeptidation reaction by slow onset kinetics, similar to acivicin. Further development of inhibitors of the GGT hydrolysis reaction is necessary to provide new therapeutic compounds.  相似文献

1. A comparison was made of the binding of 5alpha-dihydrotestosterone (17beta-hydroxy-5alpha-androstan-3-one) and cyclic AMP in the rat prostate gland. Distinct binding mechanisms exist for these compounds, and cyclic AMP cannot serve as a competitor for the 5alpha-dihydrotestosterone-binding sites and vice versa. In contrast with the results obtained with 5alpha-dihydrotestosterone, very small amounts of cyclic AMP are retained in nuclear chromatin and the overall binding of this cyclic nucleotide is not markedly affected by castration. 2. Androgenic stimulation does not lead to major increases in the adenylate cyclase activities associated with any subcellular fraction of the prostate gland. Accordingly, changes in the concentration of cyclic AMP in the prostate gland after hormonal treatment are likely to be small, but these were not measured directly. 3. When administered to whole animals in vivo, small amounts of non-degraded cyclic AMP are found in the prostate gland but sufficient to promote an activation of certain carbohydrate-metabolizing enzymes in the cell supernatant fraction. The stimulatory effects of cyclic AMP were not evident with cytoplasmic enzymes engaged in polyamine synthesis or nuclear RNA polymerases. These latter enzymes were stimulated solely by the administration of testosterone. 4. By making use of antiandrogens, a distinction can be drawn between the biochemical responses attributable to the binding of 5alpha-dihydrotestosterone but not of cyclic AMP. Evidence is presented to suggest that the stimulation of RNA polymerase, ornithine decarboxylase and S-adenosyl-l-methionine decarboxylase is a consequence of the selective binding of 5alpha-dihydrotestosterone. Only the stimulation of glucose 6-phosphate dehydrogenase can be attributed to cyclic AMP or other metabolites of testosterone. 5. Overall, this study indicates that the formation of cyclic AMP is not a major feature of the androgenic response and affects only a restricted number of biochemical processes. Certainly, cyclic AMP cannot be considered as interchangeable with testosterone and its metabolites in the control of the function of the prostate gland. This difference is additionally emphasized by the failure of cyclic AMP to restore the morphology of the prostate gland in castrated animals; morphological restoration only follows the administration of androgens.  相似文献

  • 1.1. Isolated mitochondria from rat liver were incubated in the presence of [U-14C]palmitate, ATP, CoA, carnitine, EGTA (ethylene glycol bis (β-aminoethyl ether) N,N′-tetraacetic acid) and varying amounts of calcium.
  • 2.2. When a KCl-based incubation medium was used, the oxidation of palmitate was inhibited when the concentration of free calcium was increased from about 0.1–10μM.
  • 3.3. When a sucrose-based incubation medium was used, the basal rate of palmitate oxidation was about half of that observed with the KCl-medium and calcium had a stimulatory effect.
  • 4.4. With the KCl-medium the rate of oxygen consumption was inhibited by calcium with α-ketoglutarate as well as palmitate as the respiratory substrate.
  • 5.5. No inhibitory effect of calcium was observed with succinate or β-hydroxybutyrate.
  • 6.6. With the KCl-medium and with α-ketoglutarate as the respiratory substrate, state 3 respiration but not state 4 respiration was inhibited by calcium.
  • 7.7. When the sucrose-medium was used, state 3 respiration was first inhibited by calcium, but this inhibition was gradually relieved and the respiratory rate finally became higher than it was before calcium addition.

The capacities ofHalobacterium cutirubrum and a moderate halophile NRC 41227 to survive and recover from treatment with N-methyl-N-nitro-N-nitrosoguanidine have been compared.Halobacterium cutirubrum is resistant to this chemical and its mutation frequency is only slightly affected, whereas NRC 41227 is highly sensitive and its mutation frequency is markedly increased. The chemically treated extreme halophile fully regains viability during liquid holding, in notable contrast to its known failure to recover from the effects of ultraviolet irradiation.  相似文献

The soluble fraction of rat testis homogenates is rich in enzymes catalysing the phosphorylation by ATP of many different proteins. The phosphorylation of certain histones, and also of a homogeneous basic protein from guinea-pig seminal-vesicle secretion, is markedly enhanced by 3':5'-cyclic AMP. Various factors affecting these reactions as catalysed by partially purified testicular enzyme preparations are described, and their possible physiological significance is discussed.  相似文献

Summary In vitro studies have been performed with 5-ethyl-2-deoxyuridine (EDU) concerning its incorporation into the cell nuclear material and its effects on the chromosome morphology of cultivated human lyphocytes and skin fibroblasts. This compound is presumably incorporated in the DNA without visible chromosome aberrations as is seen with many other pyrimidine analogues. A slight inhibition of the cell growth was noted at high concentration (120g/ml) of EDU. A similar degree of cell growth inhibition was found with corresponding doses of deoxythymidine.
Zusammenfassung Es wurden in vitro-Untersuchungen hinsichtlich des Äthyldeoxyuridineinbaus (ÄDU) und dessen Einfluß auf menschliche Lymphocyten- und Fibroblastenchromosomen durchgeführt. Diese Substanz wird in die DNS eingebaut und führt nicht zu sichtbaren Chromosomenaberrationen wie bei einigen anderen Pyrimidinanaloga. Eine leichte Hemmung des Zellwachstums wurde mit 120 g/ml ÄDU festgestellt. Eine ähnliche Hemmung des Zellwachstums ließ sich auch mit gleicher Deoxythymidinkonzentration nachweisen.

Vigorous organogenesis can be induced from hypocotyl and root explants of Arabidopsis thaliana using a two-step culture procedure consisting of preculture on callus-inducing medium (CIM) and subsequent culture on shoot-inducing medium (SIM) or root-inducing medium (RIM). With this culture system, we examined the influence of 5-bromo-2′-deoxyuridine (BrdU), a thymidine (dT) analogue, on plant organogenesis in vitro. Treatment with BrdU during SIM or RIM culture had negative effects on shoot and root redifferentiation over a broad range of concentrations. When explants were exposed to low concentrations of BrdU during preculture and then transferred onto BrdU-free SIM, shoot redifferentiation was accelerated significantly. At higher doses, BrdU treatment during the pre-culture inhibited shoot redifferentiation strongly in hypocotyl explants, but not in root explants. This suggests that a target of the BrdU action lies within the process of acquisition of cell proliferation competence specifically involved in hypocotyl dedifferentiation. These effects of BrdU were counteracted by the simultaneous addition of excess dT. BrdU-pretreated and untreated explants did not differ significantly in the phytohormone dependency of shoot redifferentiation. Our results provide a basis for future studies on plant organogenesis combining pharmacological analysis with BrdU as a probe and molecular genetics with Arabidopsis mutants.  相似文献

Chloroplasts from sunflower leaves senescing in vivo during 7 days in dark do not show a blue shift of the red absorption band; in contrast, the shift becomes apparent within 24 h of in vitro ageing of isolated organelles. Photosynthetic activity by chloroplasts is lost much faster during in vitro than in vivo ageing. The in vitro binding of α-tocopherol to microsomes of lung, liver, heart and brain of the rat was studied with the insoluble tocopherol ligand presented as a complex with bovine serum albumin. Under these conditions, all microsomes showed nonsaturable binding of α-tocopherol and the amount bound to microsomes was linearly proportional to the concentration of albumin-complexed tocopherol. Increasing the amount of α-tocopherol bound to microsomes in this manner reduced the extent of lipid peroxidation induced by added ferrous iron. The apparent affinities of the microsomes for α-tocopherol, as indicated by the amount bound at a given concentration of albumin-complexed tocopherol, decreased in the order brain > liver ≈ heart > lung. The differences in affinity did not correlate with total fatty acid content (r = − 0.39), total unsaturated fatty acid content (r = − 0.26), or with the content of fatty acids containing two or more double bonds (r = − 0.01). A high positive correlation was found with the content of fatty acids containing three or more double bonds (r = + 0.96). Since lung microsomes contain approx. 6-times the tocopherol levels of liver and brain and about twice that of heart microsomes, these results show that the in vivo levels of microsomal tocopherol do not reflect microsomal affinity for this biological antioxidant.  相似文献

Intracerebroventricular (icv) injection of purified recombinant human tumour necrosis factor (TNF , 4–8g) in conscious rats, produced increases in colonic temperature (1.0°C) and resting oxygen consumption (VO2, 14%) which were maximal after 80–90 minutes. Pretreatment with propranolol (10mg/kg s.c) significantly inhibited the rise in VO2, and prevented the increase in body temperature. Icv injection of an antagonist to corticotropin releasing factor (-helical CRF 9-41, 25 g), which prevents the pyrogenic and thermogenic actions of interleukin-1, did not influence the effects of TNF on temperature or VO2. Injection of a fragment of TNF (113–130 amino acid sequence) did not affect body temperature or VO2. TNF injection (icv) significantly increased brown adipose tissue (BAT)in vitro mitochondrial GDP binding, and this effect was slightly inhibited, but not prevented, by surgical denervation of the tissue, and was unaffected by pretreatment with -helical CRF 9-41. These data indicate that TNF can stimulate thermogenesis by a direct central action. The effects are largely, but not totally, dependent on the sympathetic nervous system but, unlike the thermogenic actions of interleukin they do not require release of CRF.  相似文献

Subarachnoid administration of clonidine and xylazine produces antinociception in several species and in humans. The present study compares these two drugs administered by the subarachnoid route to goats. Goats (n=6) were randomly assigned to three treatment groups. All animals of each group received 0.06 mg kg−1 clonidine (CLO), 0.1 mg kg−1 xylazine (XIL) and 0.9% saline solution (SS), with a minimum interval of 1 week between treatments. All injections were made into the subarachnoid space between the last lumbar vertebra and the first sacral vertebra. Analgesia, ataxia, sedation, cardiovascular and respiratory effects, and rectal temperature were evaluated at predefined regular time intervals before drug administration (baseline) and after administration. The onset of analgesia by clonidine and xylazine was observed in 6.8±1.8 and 9.5±2.6 min (mean±S.D.), respectively. Both α2-agonists produced analgesia of dorsocaudal rib areas, flanks, hind limbs, perineum and tail, sedation and ataxia. The duration of antinociception after clonidine administration was 118.8±24 min and after xylazine 88.3±15 min (mean±S.D.). Clonidine and xylazine subarachnoidally administered induced a significant (P<0.05) decrease in heart and respiratory rates and hypothermia in relation to the basal value. Neither drug significantly altered blood pressure. Both α2-agonist drugs induced frequent diuresis and an increase in salivation. We conclude that subarachnoid clonidine produces longer antinociception with less ataxia than xylazine in goats. However, the drug induced bradycardia, a decrease in the respiratory rate and hypothermia, with a small compromise in the blood pressures at the doses studied. Further studies should be done to determine whether this analgesia is sufficient for surgical procedures.  相似文献

The effect of -ketotriazole, MCPA and carbaryl on the lipid composition and ATPase activity associated with plasma membrane fractions from rice (Oryza sativa cv Bahia) shoots was investigated. With -ketotriazole and MCPA treatments the relative amount of 5-avenasterol (%) was reduced in the plasma membrane, whereas with -ketotriazole a reduction was also found in the sitosterol content, expressed as a percentage of the total free sterol composition. The fatty acyl chain length of phosphatidylcholine fractions from MCPA-treated plants was also reduced. The plasma membrane Mg2+-ATPase activity was only stimulated in MCPA-treated plants. No changes were observed in lipid composition or ATPase activity with carbaryl treatment.  相似文献

The inhibitory effect of three SH reagents, mersalyl, 5,5-dithio-bis-nitrobenzoate, andN-ethylmaleimide, on Pi transport in rat liver mitochondria was investigated under a variety of conditions. Mersalyl binds at room temperature with both high (K d<10 µM) and low affinity to mitochondria. Inhibition of Pi transport by mersalyl goes in parallel with titration of the high-affinity sites, inhibition being complete when 3.5–4.5 nmol/mg protein is bound to the mitochondria. At concentrations of mersalyl equal to or higher than 10 µM, inhibition of Pi transport occurs in less than 10 sec. At concentrations of mersalyl lower than 10 µM, the rate of reaction with the Pi carrier is considerably decreased. At a concentration of 100 µM, 5,5-dithio-bisnitrobenzoate fully inhibits Pi transport in about 1 min at room temperature. Nearly total inhibition is attained when as little as 40–50 pmol/mg is bound to mitochondria. Upon incubation longer than 1 min, additional SH groups, not belonging to the Pi carrier, begin to react. The uncoupler carbonyl cyanidep-trifluoromethoxyphenylhydrazone decreases the rate of reaction of mersalyl, 5,5-dithio-bis-nitrobenzoate, andN-ethylmaleimide with the Pi carrier. Preincubation with Pi has a similar effect. We propose that both carbonyl cyanidep-trifluoromethoxyphenylhydrazone and Pi act by increasing the acidity of the mitochondrial matrix. Protonation of the Pi carrier at the matrix side would change the accessibility of its SH groups at the outer surface of the inner membrane. This might correspond to a membrane-Bohr effect, possibly related to the opening of a gating pore in the Pi carrier.  相似文献

1. The effect of pH on the co-operative activation of the NAD-specific isocitrate dehydrogenase from pea mitochondria by isocitrate is shown. 2. The interlinked effects of pH on the affinity of the NAD-specific isocitrate dehydrogenase for isocitrate and the dependence of the pH optimum on the substrate concentration are presented. 3. A consideration of the conditions of pH and substrate concentration under which citrate activates the NAD-specific isocitrate dehydrogenase demonstrates similarities between the binding of isocitrate and citrate. 4. A comparison of the effects of citrate and pH on the gross structure of the enzyme is investigated by density-gradient centrifugation. 5. The kinetic interpretations of these results are briefly considered. 6. The metabolic significance of these studies is discussed.  相似文献

1. The mitochondrial NADH dehydrogenase (EC 1.6.99.3) of Candida utilis exhibited altered properties when the organism was grown under iron-limited conditions. No suitable acceptor was found for assay of this enzyme from iron-limited cells. 2. Mitochondrial membrane proteins from C. utilis were analysed by polyacrylamide-gel electrophoresis. Compared with glycerol-limited cells, iron limitation resulted in the loss of at least two polypeptides from the mitochondrial membrane. 3. Neither of the polypeptides affected by iron limitation was part of a cytochrome, although one of them was part of the mitochondrial NADH dehydrogenase. 4. Non-haem iron of mitochondrial membranes was released in the presence of sodium dodecyl sulphate, and electrophoresis in solutions of this detergent cannot be used directly to identify iron-sulphur proteins. Non-ionic detergents do not release non-haem iron but nor do they provide a satisfactory system for electrophoretic separation.  相似文献
