The oesophagus of Patella vulgata Linnaeus is divisible intotwo histologically and functionally distinct regions, a centralfood channel and a series of lateral pouches forming the oesophagealglands: both are lined by a glandular ciliated columnar epithelium.Subepithelial mucous cells are restricted to the epitheliumlining the food channel and posterior section of oesophagus,whilst amylase-secreting cells occur only in the oesophagealglands. The activity of the ciliated cells lining the food channelmixes ingested food with saliva and mucus to form a food stringthat is carried towards the stomach. Amylase-secreting cellsproduce an extracellular amylase in the form of proteinaceoussecretory granules, which are released by apocrine secretionwithin cytoplasmic blebs. Release is correlated with periodsof immersion when feeding is possible. The free blebs are carriedout of the pouches and into the food channel where the amylaseinitiates carbohydrate digestion. The oesophageal epitheliumabsorbs tritiated D-glucose by a Na+-dependent mechanism inhibitedby 2,4-DNP and phloridzin and the amylase-secreting cells pinocytoseferritin. It is suggested that the oesophagus absorbs smallsolute molecules derived from ruptured algal cells and theirpartially digested polysaccharide storage products. There alsoappears to be pinocytosis of dissolved organic matter. *Current address: The R L. An antiserum against purified rat parotid amylase was used to localize the protein in parotid glands of developing and adult rats. The unlabeled antibody peroxidase-antiperoxidase method and the protein A-gold colloid technique were used at the light and electron microscope levels, respectively. Immunoreactive amylase was detected in a few scattered cells in the glands of 2-day-old rats. During the following days the number of cells stained immunocytochemically for amylase increased rapidly; at 15 days of age all acinar cells revealed amylase, but the intensity of immunostaining varied from cell to cell. Electron microscopically, amylase was localized in the secretory granules, and by using a more concentrated antiserum, in the rough endoplasmic reticulum and Golgi complex. At early stages of development the acinar cells contained fewer and smaller secretory granules than in adult animals; the gold particles indicative of amylase were randomly distributed over the secretory granules. In the glands of adult rats, amylase was distributed inhomogeneously within the secretory granules. In the majority of secretory granules gold colloid particles were located over the electron-dense portions of the granules. However, secretory granules in which an amylase-rich shell surrounded an amylase-poor or amylase-negative "core" were not infrequent.  相似文献

Biotic interactions within benthic megafauna were studied from quantitative dredge samples to investigate their role in the persistence of low scallop, Pecten maximus, stocks in the Bay of Brest. The distributions of 148 species of the megafauna and of four age classes of Pecten maximus were sampled. Suspension feeders dominate all megabenthic assemblages within the bay. This domination can be related to Crepidula fornicata spreading in the bay; however, no direct competition for food between scallops and the slipper limpet (introduced species) has been observed. On the other hand, Crepidula spreading leads to major changes in sediment type (silting) in the bay owing to biodeposit production, and silting would be a major factor inducing the decrease in Pecten maximus distribution. Predation would not be limiting. Within 10 years, one may predict a strong decrease in the area colonized by scallops in the bay.  相似文献

The ventriculus and the midgut caeca of the fed females of Anystis baccarum (L.) were investigated by using light and electron microscopy. In addition to the main type of polyfunctional digestive cells, special secretory cells were detected in the anterior region of the ventriculus. The shape and the ultrastructure of the digestive cells vary depending on their physiological state. Intracellular digestion, absorption or excretion processes prevail at different stages of the cell cycle. The secretory cells are characterized by the presence of extensive rough endoplasmic reticulum, filling whole space of the cell. These cells do not contain the apical network of pinocytotic canals, which are typical for the digestive cells. Three types of secretory granules were found in the cytoplasm of the secretory cells that probably correspond to three sequential stages of granulogenesis. The primary secretory granules are formed by the fusion of Golgi vesicles. The primary granules fuse to form complex vesicles with heterogeneous contents. These secondary granules aggregate to form very large inclusions of high electron density (tertiary secretory granules), which probably represent the storage of the secretory product. All types of secretory granules were observed close to the apical plasmalemma.  相似文献

 Two different types of glandular system in the proboscidial epithelium of Riseriellus occultus have been investigated by transmission electron microscopy. As expected, most of the epithelial cells are glandular in nature. With regard to differences in the ultrastructure of these gland cells and in the formation and morphology of their secretory granules, we have categorized and described four types of gland cell, indicated as G1, G2, G3, and G4. Each gland cell has a completely intraepithelial body characterized by a prominent nucleus, developed rough endoplasmic reticulum, Golgi complexes, and numerous secretory granules at different stages of maturation. These four types of gland cell appear associated in pairs forming numerous glandular systems of two types (A, B). These glandular systems are restricted to the ventral surface of the proboscis and are scattered irregularly throughout its length. Each glandular system consists of two gland cells of different types. The gland cell necks in each glandular system extend together to the epithelial surface; they protrude onto this and form a papilla where they open in a common area. The epithelial supportive cells adjacent to the glandular systems have long, stout microvilli which have a core of tonofilaments. These tonofilaments gather into dense bundles which pass vertically through the supportive cells and attach to the extracellular matrix underlaying the cells by hemidesmosomes. Moreover, a single sensory process stands close to each papilla. The ultrastructural morphology of the type A glandular systems suggests that they have an adhesive function operating in a similar way to that of the duo-gland adhesive systems in other invertebrate groups, although they are not homologous with these. The spatial arrangement of the secreted products of the type B glandular systems suggests that these may contribute to increasing the grip of the proboscis on the prey. The secretory granules (=pseudocnids) of the type G3 gland cells are very likely an autapomorphy of the Anopla, providing a character by which the relationships within the Nemertea can be evaluated. Orbital glands are found in many tetrapod vertebrates, and are usually separate structures, consisting of individual glands lying in the eyelids and both canthi of the orbit. In cetaceans, however, the orbital glandular units are less distinct and have been described by numerous authors as a single, periorbital mass. There are few histochemical and immunhistochemical studies to date of these structures. In this study, we examined the orbital glandular region of both the bowhead whale (Balaena mysticetus: Mysticeti) and the beluga whale (Delphinapterus leucas: Odontoceti) using histological, histochemical, and immunohistochemical techniques. Histologically, in the bowhead, three glandular areas were noted (circumorbital, including Harderian and lacrimal poles), palpebral (midway in the lower eyelid), and rim (near the edge of the eyelid). In the beluga, there was only a large, continuous mass within the eyelid itself. Histochemical investigation suggests neither sexual dimorphism nor age-related differences, but both whales had two cell types freely intermingling with each other in all glandular masses. Large cells (cell type 1) were distended by four histochemically distinct intracellular secretory granules. Smaller cells (cell type 2) were not distended (fewer granules) and had two to three histochemically distinct intracellular secretory granules. The beluga orbital glands had additional lipid granules in cell type 1. Counterintuitively, both lipocalin and transferrin were localized to cell type 2 only. This intermingling of cell types is unusual for vertebrates in whom individual orbital glands usually have one cell type with one to two different secretory granules present. The heterogeneity of the orbital fluid produced by cetacean orbital glands implies a complex function, or series of functions, for these orbital glands and their role in producing the tear fluid.  相似文献

The absence or decreased expression of cystic fibrosis transmembrane conductance regulator (CFTR) induces increased Na(+) absorption and hyperabsorption of the airway surface liquid (ASL) resulting in a dehydrated and hyperviscous ASL. Although the implication of abnormal airway submucosal gland function has been suggested, the ion and water content in the Cystic Fibrosis (CF) glandular secretory granules, before exocytosis, is unknown. We analyzed, in non-CF and CF human airway glandular cell lines (MM-39 and KM4, respectively), the ion content in the secretory granules by electron probe X-ray microanalysis and the water content by quantitative dark field imaging on freeze-dried cryosections. We demonstrated that the ion content (Na(+), Mg(2+), P, S and Cl(-)) is significantly higher and the water content significantly lower in secretory granules from the CF cell line compared to the non-CF cell line. Using videomicroscopy, we observed that the secretory granule expansion was deficient in CF glandular cells. Transfection of CF cells with CFTR cDNA or inhibition of non-CF cells with CFTR(inh)-172, respectively restored or decreased the water content and granule expansion, in parallel with changes in ion content. We hypothesize that the decreased water and increased ion content in glandular secretory granules may contribute to the dehydration and increased viscosity of the ASL in CF.  相似文献

Responses of cells in the tick salivary gland to parasitism by Theileria parva were studied by electron microscopy. The gland is composed of three distinct types of acini (I, II, III) which together include ten or more different cell types. Of some 30 infected cells observed in the present study, all were E-cells of acinus III. The parasite thus exhibits a high degree of selectivity for acinus and cell type. The glandular cell invaded undergoes massive hypertrophy and accumulates glycogen deposits in its cytoplasm which may serve as an energy source for the growing intracellular parasite. As synthesis of its secretory material declines the product is packaged in progressively smaller secretory granules. The extensive arrays of endoplasmic reticulum are dismantled and eliminated in autophagic vacuoles. Excess secretory granules are also broken down by crinophagy. After 4 days, sporogony is completed and the host cell contains 30,000–50,000 sporozoites in an electron-lucent cytoplasm largely devoid of cytomembranes and secretory granules. Mitochondria are still present and normal in appearance. The loss of basophilia and secretory granules observed heretofore by light microscopy have been attributed to ingestion and destruction of host organelles by the parasite. The pallid appearance of the cytoplasm has been interpreted as a sign of impending degeneration of the host cell. In electron micrographs no ingestion of organelles by the parasite or degenerative changes were found. The host cell clearly remains viable and metabolically active throughout sporogony. The striking changes in its ultrastructure result from active elimination of organelles and inclusions by the host cell itself in response to parasitism.  相似文献

Summary An immunocytochemical study by light- and electron microscopy using the antibody against rat hepatic fatty acid-binding protein (FABP) revealed the brush cells in the gastric epithelium of rats to be intensely immunoreactive. The immunoreactive cells were present in a group in the distal wall of the groove between forestomach and glandular stomach, as well as scattered singly in the surface and foveolar epithelia of the glandular stomach. Almost all immunoreactive brush cells had a thin basal process in contact with the basement membrane. No secretory granules with dense cores, similar to those of endocrine cells, were observed in the brush cells. The specific appearance of FABP-immunoreactivity in the brush cell indicates that this cell type is a distinct entity from other epithelial cells in the stomach and that FABP is a useful histochemical marker of the brush cells. FABP may be involved in the specific function(s) of this cell type related to fatty acid metabolism.  相似文献

The ultrastructure of prostate gland of Lampito mauritii revealed two types of secretory cells. Type 1 cells with a broad basal region and a long apical region contain electron dense oval secretory granules with an increased density at the core region. Numerous electron lucent granules with fine filamentous and electron dense amorphous materials also occur at the basal region of these cells. Type 2 cells contain electron lucent mucous-like secretory granules. This cell type contains exceptionally large Golgi complexes having 20-23 stacked cisternae. Both cell types open into a common lumen and numerous microtubules are visible at the apical end. Junctional complexes, such as desmosomes and septate junctions, are observed in this glandular tissue.  相似文献

By means of electron microscopy cells in the human submandibular glands were studied. It was demonstrated that in acini two types of glandular cells were present: mucosal and seromucosal. In the latter, secretory granules are descrete with electron opaque cores in most of them. Mucocytes are filled with an electron transparent secrete; secretory granules often confluent and their membranes rupture. The acini are surrounded with myoepithelial cells. Intercalated ducts consist of cells with moderately electron opaque granules. In some granules there are dense bodies excentrically situated. In these cells there occur lipid inclusions. Striated ducts are composed of basal (electron transparent) and high cylindric (light and dark) cells. The cylindrical cells have a large amount of mitochondria, deep folds in their basal plasmolemma protruding into cytoplasma. Most of the cells in these parts contain small apically accumulated secretory granules with a dense matrix and separate larger ones scattered in the cell. It is possible to suggest that some secretory granules of ductal or, perhaps, acinar origin contain hormonal products.  相似文献

Summary Two cell types can be distinguished in the pars intermedia of Klauberina: (1) Glandular cells, which form a single-layered columnar epithelium on the vascular septum which divides the pars nervosa from the pars intermedia. (2) Marginal cells which form a flattened epithelium over the glandular cells and line the hypophysial cleft. Occasional projections from the marginal cells extend between the glandular cells to contact the basement membrane of the vascular septum, and occasional projections of the vascular septum extend across the glandular epithelium to reach the marginal epithelium. Both cell types are AF negative. The granules of the glandular cells are strongly PAS positive, and acidophilic in response to Mallory's trichrome stain. In electron micrographs, the glandular cells contain large quantities of secretory granules. In one class of cells, they range from 2,000 to 2,500 Å in diameter, in the other, from 4,000 to 5,000 Å. Electron-dense granules 1,000 to 1,500 Å in diameter occur in the cytoplasm of the marginal cells in the region of contact with the vascular septum. Hence more than one active principle may be produced by the pars intermedia.No nerve endings of any kind are present in the pars intermedia. Therefore, synaptic contact of neurons with the secretory cells seems not to be necessary for the regulation of their secretory activity as appears to be the case in other vertebrate groups. The distal wall of the groove between the rat forestomach and glandular stomach is lined with a special type of columnar cells (CCGG) and with fibrillovesicular cells (FVC). The cardiac glands contain cardiac mucosa (CMC) and serous cells (CSC). The CCGG contain small mucous granules and special vesicles and tubules. The CMC are filled with large mucous granules and resemble mucous neck cells. The CSC are filled with large proteinaceous granules. The FVC are characterized by long microvilli, apical bundles of microfilaments and a complex "tubulovesicular system". The pattern of 3H-thymidine incorporation and the presence of immature and transitional forms indicate a possible origin of all the cell types concerned from a common undifferentiated precursor. The membranes of the tubulovesicular system of FVC as well as the apical cell membrane were reactive to Thiéry's carbohydrate stain. However, lanthanum tracing of the extracellular space and ultrastructural stereoscopy did not reveal a permanent continuity between both membrane systems. The absence of 3H-thymidine label showed that FVC were not proliferative. The structural characteristics of FVC do not account for a secretory, resorptive or receptive function. The special arrangement of microfilaments and the tubulovesicular system suggests an ability to fast changes in surface area.  相似文献

1. Two molecular forms of arginine kinase, AK1 and AK2 have been purified from the adductor muscle of the scallop, Pecten maximus. AK2 was retained on a DEAE-cellulose column at pH 7.5, but AK1 was not. 2. Both forms were monomeric (mol. wt. approximately 42,000) and showed the same pH optimum (7.5-8.0) in the direction of phosphoarginine synthesis. 3. AK1 had slower electrophoretic mobility at pH 8.3 towards the anode, higher lysine content, lower glutamate content, lower Km for L-arginine and higher Km for Mg(2+)-ATP than AK2. Unlike AK1, AK2 was strongly inhibited at high concentrations of Mg(2+)-ATP. 4. Both molecular forms cross-reacted with antisera raised against native as well as performic acid-oxidized lobster muscle arginine kinase. However, AK1 showed a greater affinity than AK2 to anti-lobster arginine kinase antibodies, particularly to those raised against the native enzyme.  相似文献

It is well-known that amylase is secreted in response to extracellular stimulation from the acinar cells. However, amylase is also secreted without stimulation. We distinguished vesicular amylase as a newly synthesized amylase from the accumulated amylase in secretory granules by short time pulse and chased with 35S-amino acid. The newly synthesized amylase was secreted without stimulation from secretory vesicles in rat parotid acinar cells. The secretion process did not include microtubules, but was related to microfilaments. p-Nitrophenyl β-xyloside, an inhibitor of proteoglycan synthesis, inhibited the newly synthesized amylase secretion. This indicated that the newly synthesized amylase was secreted from secretory vesicles, not via the constitutive-like secretory route, which includes the immature secretory granules, and that proteoglycan synthesis was required for secretory vesicle formation.  相似文献

During a systematic ultrastructural study on the endocrine/paracrine cells of the gastro-intestinal tract of ruminants, the aim of present work is to describe these cells in the abomasum of suckling calf. Taking into account all the cytological details, and especially the morphological appearance and the cytochemical reactivity of secretory granules, several endocrine cell types can be distinguished. Some of them (EC, D, D1) are common to all three of the glandular regions, whereas others are typical of cardiac and proper gastric glands: they are ECL, X, A-like and a fourth type, whose classification is uncertain. The last type (G cells) is detectable in pyloric glands only. The cardiac and proper gastric glands of the suckling calf abomasum contain two additional cell types, not present in bulls, A-like cells and the fourth type, and contain D1 cells which form a heterogeneous family. These data show a morphofunctional similarity between the abomasum of the suckling calf and the stomach of non-ruminant mammals.  相似文献

The paired female accessory glands of Phlebotomus perniciosus (Diptera : Psychodidae) were investigated by light microscopy, and by scanning and transmission electron microscopy. These glands undergo morphological and functional changes during oocyte development. After the blood meal, the monostratified glandular epithelium differentiates and starts to secrete. Well-developed rough endoplasmic reticulum, Golgi complexes, and membrane-bounded exocytic vesicles suggest that these secretory cells are involved in protein synthesis. As the secretory cells differentiate, the glandular lumen increases in size and fills with secretory material, consisting of globular granules of different sizes in an amorphous electron-dense matrix. The granules have an electron-translucent core and an electron-dense cortex. The morphological characteristics of the glandular epithelium and the functional role of the glands are discussed in relation to their possible contribution to the reproductive process.  相似文献

The ultrastructure of male reproductive accessory glands was investigated in the scorpionfly Sinopanorpa tincta (Navás, 1931) (Mecoptera: Panorpidae) using light and transmission electron microscopy. The male accessory glands comprise one pair of mesodermal glands (mesadenia) and six pairs of ectodermal glands (ectadenia). The former opens into the vasa deferentia and the latter into the ejaculatory sac. The mesadenia consist of a mono-layered elongated columnar epithelium, the cells of which are highly microvillated and extrude secretory granules by means of merocrine mechanisms. The epithelium of ectadenia consists of two types of cells: the large secretory cells and the thin duct-forming cells. These two types of cells that join with a cuticular duct constitute a functional glandular unit, corresponding to the class III glandular cell type of Noirot and Quennedey. The cuticular duct consists of a receiving canal and a conducting canal. The secretory granules were taken up by the receiving canal and then plunged into the lumen through the conducting canal.  相似文献

Excretory granules from the kidney of the scallop, Pecten maximus (L.), have been isolated and purified. Their morphology has been determined by electron microscopy and their elemental composition by electron-probe X-ray micro-analysis. The granules, varying in size from 5–15 gmm diameter, are inorganic in nature and contain Ca, Mn, Zn, and P, equivalent to 75% of their dry weight, with Mg, Cu, Fe, Cd, K, S, and Cl also present in much smaller amounts. The granules show large variations in elemental composition but the sum of the metals is always close to equimolarity with the phosphorus content. Electron microscopy of the kidney tissue showed that there is a gradual development of the granules from lysosomal membranous vesicles to highly mineralized membrane-limited vacuoles or residual bodies, which are eventually excreted as such into the urinary tract.  相似文献

Summary Glandular cells in the prostate of the intact, adult dog contain numerous, large secretory granules that are released by exocytosis. Following hypophysectomy or castration, the glandular epithelium atrophies and the secretory granules degenerate and eventually disappear. Pharmacologic doses of estradiol-17 17-cyclopentylpropionate cause the regressed glandular cells to synthesize a new population of smaller granules that are also released by exocytosis, even though estrogen is known to inhibit fluid secretion by the canine prostate. Thus, the mechanisms involved in prostatic synthesis and exocytosis of secretory granules are independent of those regulating fluid secretion and are operative in the absence of androgen or pituitary hormones.  相似文献
