1. Myelin proteins from the CNS of recent lungfish (Lepidosiren paradoxa, Protopterus dolloi, Neoceratodus forsteri) were separated and analysed by staining and immunoblotting. 2. All species showed a glycosylated component (g-PLP) that cross-reacted with antibodies against tetrapod proteolipid protein (PLP), indicating phylogenetic relationships with amphibia. 3. Actinopterygian IP or teleostean 36k components were not detectable in lungfish CNS myelin. 4. The identical size of g-PLPs from Lepidosiren and Protopterus (Mr = 29,000) underlines the close relationship of the Lepidosirenidae. The smaller size of g-PLP from the ceratodidan Neoceratodus forsteri (Mr = 27,500) pointed to an earlier diversion.  相似文献

The identification of an extrinsic cardiac innervation in Protopterus has been reported for the first time. It has been based on histological, histochemical and ultrastructural methods. The heart of Protopterus receives a rich cholinergic parasympathic innervation which ramifies in the atria and the ventricle. The induced fluorescence did'nt permit us to detect any mono-aminergic fibres ; meanwhile this method has enabled us to distinguish a particular type of cells containing catecholamines in the heart.  相似文献

The interrenal (adrenal) of Ichthyophis beddomei lies on the ventral side of the kidney, distributed in four zones. It is separated from the renal tissue by a thin layer of connective tissue and contains both adrenocortical and chromaffin cells. Adrenocortical tissue constitutes a major portion of the interrenal islets; the chromaffin tissue consists of a few cells located at the peripheries of the interrenal islets. Histochemical studies demonstrate the presence of Δ53β-hydroxysteroid dehydrogenase, 17 β-hydroxysteroid dehydrogenase, glucose-6-phosphate dehydrogenase, succinate dehydrogenase, and sudanophilic lipids in the adrenocortical tissue, suggesting its steroidogenic potential. Annual histometric and histochemical studies show two peaks of interrenal activity: (1) during the breeding phase of the reproductive cycle (January and February) and (2) during the season of heavy monsoon rains (June and July) in the postbreeding phase.  相似文献

Pronephric, opisthonephric, ovarian, and testicular tissue rich in presumed steroid producing cells taken from the brook lamprey during metamorphosis was investigated by histochemical and spectrophotometric methods. Histochemical results seemed to provide evidence for hydroxysteroid dehydrogenase activity using pregnenolone as substrate. Spectrophotometric evidence for hydroxysteroid dehydrogenase activity was obtained from presumed adrenocortical tissue, ovarian and testicular homogenates using pregnenolone, dehydroepiandrosterone, androsterone, and 3 beta, 17 beta-dihydroxy-5 alpha-androstane as substrates.  相似文献

Summary The ultrastructure of the interrenal (adrenocortical) cells of trout (Salmo fario L.) was studied after dexamethasone treatment. A procedure for identifying and isolating interrenal tissue fragments from the surrounding head kidney tissue prior to their preparation for electron microscopy is described. The peripheral plasma cortisol concentrations were measured in order to evaluate the steroidogenic activity of this tissue.The interrenal cells of control animals contain numerous mitochondria with tubular cristae, and a well developed and highly organized smooth endoplasmic reticulum (SER). The scarcity, or absence, of lipid droplets contrasts markedly with the abundance of SER.Treatment with dexamethasone results in a decrease steroidogenic activity of the interrenal cells, as indicated by the fall in plasma cortisol concentrations. The interrenal cells are small, but still contain numerous mitochondria. The SER is poorly developed, but masses of densely intermeshed smooth cisternae subsist. Lipid droplets do not accumulate in these cells; this peculiarity is discussed in connection with the virtual absence of liposomes in teleost interrenal cells.  相似文献

A simple, rapid procedure for the quantitation of the histochemical stain for delta 5-3beta-hydroxysteroid dehydrogenase (3beta-HSD) in cultured cells is described. Adrenocortical cells were stained for 3beta-HSD and then solubilized with 0.5 N NaOH. The absorbance of the solubilized cell solution was measured at 570 nm and a linear relationship was obtained between the number of cells and the intensity of the absorption. It is shown that the regulation of 3beta-HSD activity in primary cultures of normal rat adrenocortical cells can be monitored by the quantitative histochemical assay. The results show a good correlation with the data obtained by a biochemical assay for the enzyme.  相似文献

The Australian lungfish Neoceratodus forsteri (Dipnoi-Sarcoterygians) is a likely candidate for the extant sister group of Tetrapoda. Transmission electron and light microscopy analysis revealed that the arrangement of somite cells of the lungfish resembles the structure of the urodelan somite. On the other hand, the pattern of early muscle formation in N. forsteri is similar to that found in the Siberian sturgeon (Acipenser baeri). During the early stages of myogenesis of N. forsteri, somite-derived cells fuse to form multinucleated muscle lamellae. During later stages, mononucleated undifferentiated cells are first observed in the intermyotomal fissures and subsequently in the myotomes, among white muscle lamellae. The cells from the intermyotomal fissure differentiate into fibroblasts. The cells which have migrated into the myotomes, differentiate into mesenchyme-derived myoblasts. After hatching, white muscle lamellae are successively converted into polygonal muscle fibres. Conversion of lamellae into fibres may occur through splitting of muscle lamellae, or cylindrical muscle fibres may arise de novo as a result of fusion of mesenchyme-derived myoblasts. No increase in the number of muscle fibre nuclei is observed either in embryonic or juvenile musculature of N. forsteri. We suggest that until the 53 stage of embryonic development, the increase in muscle mass is accomplished mainly through hyperplasy. Thus, lungfish muscle represents the organizational intermediate between fishes and amphibians. This makes it a useful model to study the evolutionary implications of the mechanisms of muscle development.  相似文献

We describe the structure of the lympho-granulocytic tissue associated with the wall of the spiral valve of the African lungfish Protopterus annectens. The study was performed under freshwater conditions and after 6 months of aestivation. The lympho-granulocytic tissue consists of nodes surrounded by reticular tissue. The nodes are formed by an outer and an inner component separated by a thin collagenous layer. The outer component is a reticular-like tissue that contains two types of granulocytes, developing and mature plasma cells and melanomacrophage centres (MMCs). The inner component, the parenchyma, contains a meshwork of trabeculae and vascular sinusoids and shows dark and pale areas. The dark areas contain diffuse lymphoid tissue, with a large number of mitoses and plasma cell clusters. The pale areas contain a small number of macrophages and lymphocytes. Macrophages and sinus endothelial cells are filled with haemosiderin granules and appear to form part of the reticuloendothelial system of the lungfish. The reticular tissue houses granulocytes, plasma cells and MMCs and might serve for the housing and maturation of cells of the white series. After aestivation, the nodes undergo lymphocyte depletion, the suppression of mitosis, granulocyte invasion and the occurrence of cell death. By contrast, few histological changes occur in the reticular tissue. Whereas the nodes appear to be involved in lymphocyte proliferation and plasma cell maturation, the function of the reticular tissue remains obscure.  相似文献

Summary Purified transthyretin has been isolated from sheep serum. Antiserum raised against this protein has been used with an indirect immunoperoxidase histochemical technique to identify transthyretin in newborn lamb kidney tissue. Transthyretin was found in proximal tubule cells and in glomerular peripolar cells. Preabsorption studies using purified transthyretin protein indicate that the immunoreactivity of the antiserum is specific to transthyretin.  相似文献

Follicles of peripheral lymphoid organs (rat) contain a type of non-lymphoid cell which is capable of arresting antigen-antibody complexes at the cell surface. These so-called dendritic cells can be visualized in immunized rats by staining antigen-antibody complexes with immunohistoperoxidase techniques. The present study concerns a classification of these cells and comparison with known non-lymphoid cell types such as macrophages, marginal metallophils and tingible body macrophages in the rat spleen follicles. Immunoenzyme histochemical and (enzyme) histochemical techniques have been combined in the same tissue sections to correlate the functional capacity of binding immune complexes with morphological characteristics or phagocytic capacity. Dendritic cells show silver affinity but do not demonstrate a characteristic pattern of hydrolytic enzymes or phagocytosis.  相似文献

The adrenocortical homolog in the chondrostean fish, Acipenser fulvescens, is confined within yellow corpuscles located at the dorsal junction of the two kidneys near the cardinal veins. The concentration and distribution of this tissue in chondrosteans appear to be a condition intermediate between that of cyclostomes and teleosts. The homology of this tissue to adrenocortical tissue of higher vertebrates is demonstrated in its delta5-3beta-hydroxysteroid dehydrogenase activity and the ultrastructure of the cells. The cells contain large lipid droplets and an abundance of smooth endoplasmic reticulum and mitochondria. The abundance of smooth endoplasmic reticulum, a large Golgi apparatus and an extensive elaboration of the plasma membrane suggest the high degree of steroidogenic activity of these cells. Apposed plasma membranes make contacts through the unique cell contacts described in the adrenocortical cells of mammals (Friend and Gilula, '72). The Golgi apparatus is described as a most conspicuous component of the cytoplasm and is presented as further evidence for the important involvement of this organelle in corticosteroidogenesis in vertebrates. These morphological observations are discussed in relation to the known secretory products of this tissue in the sturgeon, and it is suggested that there is a need for further examination of the corticosteroids produced by lower vertebrates.  相似文献

The head kidneys of adult Periophthalamus koelreuteri contain many functional glomerular nephorns which on the basis of this histochemical study, are indistinguishable from those of the typical opistonephros of marine teleosts. The anterior lobes resemble, in location and in the presence of intertubular erythropoeitic tissue, the pronephros in other species. The single nephron found there, however, does not differ significantly from the other nephrons of the head kidney. It is suggested, therefore, that the nephrons of the head kidney in this species are opisthonephric in origin.  相似文献

The adrenal components of C. mrigala are embedded in the pronephric cephalic kidney around the post cardinal vein. The cortical cells responded positively to the lipids, ascorbic acid, delta 5-3 beta-HSD, G-6-PD, MAO, acid and alkaline phosphatase tests. The presence of intense MAO activity may suggest the possible involvement of monoamines in the adrenocortical function. Localization of lipids and delta 5-3 beta-HSD show the sites of corticosteroid synthesis. In the chromaffin cells, MAO, acid and alkaline phosphatase activity was moderate whereas they gave a strong reaction to ascorbic acid test in comparison to the cortical cells. Noradrenaline (NA) and adrenaline (A) storing cells were differentiated adopting glutaraldehyde silver, dichromate and iodate techniques. NA and A storing cells are almost totally depleted of their contents after reserpine treatment. The histochemical response of the adrenal gland of this species is largely comparable to that of higher vertebrates.  相似文献

Cell transplantation provides a way to compare the regulation of cell proliferation in the same cell type in cell culture and in a vascularized tissue structure in a host animal. The cyclin-dependent kinase inhibitors p57(KIP2), p21(WAF1/CIP1/SDI1) and p27(KIP1) have been extensively studied in cell culture but their role in growth control in tissues is less well understood. In the present experiments we compared the behavior of cell cycle inhibitors in human and bovine adrenocortical cells in culture and following cell transplantation in scid mice. p57 was expressed in the majority of cells in the intact human adrenal cortex. However, double immunofluorescence showed that cells that are in the cell cycle are p57(-) adrenocortical cells, p57 and p27 levels were not affected by inhibition of growth at high cell density, whereas p21 was higher in dividing than growth-inhibited cells. However, p21 was also high in senescent adrenocortical cells. After transplantation of human adrenocortical cells in scid mice, p57 and p27 were observed in most cells in the transplant tissue. Over time the number of p21(+) cells decreased greatly in human adrenocortical cells, but not in bovine adrenocortical cells. This difference correlated with lower levels of cell division (assessed by Ki-67 or incorporation of bromodeoxyuridine) in the human cells in transplant tissues in comparison to bovine cells. The differences between human and bovine cells were observed both when cells were transplanted beneath the kidney capsule and when cells were injected subcutaneously in collagen gel. We conclude that the behavior of p57, but not p21, is consistent with a role as a physiological mediator of proliferative quiescence in the adrenal cortex. The high level of p21 in dividing adrenocortical cells in culture, and in bovine adrenocortical cells in transplant tissues, may be a response to conflicting positive and negative growth influences. Cells may enter the cell cycle under the influence of a strong positive mitogenic signal, but coexisting negative growth stimuli trigger a p21-dependent block to further progression through the cell cycle. This model suggests that bovine adrenocortical cells respond to positive growth stimuli in transplant tissues but human cells lack this response.  相似文献

The structure of the intestine and its adnexa in the Australian lungfish Neoceratodus forsteri (Krefft), is described from four adult specimens and compared with that of other lungfish. In all lungfish genera the intestine is thick and straight and contains a complicated spiral valve; a pyloric fold separates the foregut from the midgut. In Neoceratodus the coiling of the intestinal lumen begins in the prepyloric or gastric region, unlike in other vertebrates with a spiral valve, where it begins behind the pylorus. The spiral valve of Neoceratodus begins as a deep groove on the right side of the foregut, just behind the glottis. Such a prepyloric groove is present but poorly developed also in the lungfishes Protopterus and Lepidosiren and contains a prepyloric spleen in all genera. A separate postpyloric spleen is situated in the free margin of the spiral valve, i.e. in the axis of the intestine. The spleen has a heavily pigmented cortex containing large amounts of iron. The pancreas, buried in the spiral valve in front of the pylorus in Neoceratodus , contains numerous islets of Langerhans, similar to those of tetrapods. This is unexpected because the islets of Protopterus , described by Gabe in 1969, are more like the large, encapsulated "Brockmann bodies" of teleosts.  相似文献

自由基引发的生物膜不饱和脂肪酸脂质过氧化反应涉及多种疾病过程,多年来检测脂质过氧化反应一直沿用生物化学(如硫代巴比妥酸法测定丙二醛)或生物物理技术(如分光光度法测定共轭双烯)。自从冷Schiff组织化学染色技术用于自由基研究以来,使形态学方法研究脂质过氧化反应成为可能,当前,应用冷Schiff组织化学法进行组织细胞的脂质过氧化反应检测大多限于动物实验研究,本研究对多种人体离体新鲜组织的冰冻切片应用冷Schiff组织化学法进行检测,未发现被组织存在组织化学水平上的脂质过氧化反应;胆对被测组织人为施加氧化攻击(用Fe-NADPH促氧化剂孵育)后,肝、肾及胃的泌酸细胞与其它组织相比呈现较明显的脂质过氧化反应;皮肤、脂肪组织几科不出现脂质过氧化反应;甲状腺C细胞、肌肉、骨骼等与钙代谢、贮存及利用相关的组织也出现较明显的脂质过氧化反应。结论:冷Schiff组织化学方法检测人体组织脂持过氧化反应具有简便易行、同时可以形态学定位的优点,在医学生物学、肿瘤学及老年医学研究中具有应用前景。  相似文献

The immunohistochemical localization of carbonic anhydrase isoenzymes has never been investigated in avian renal tissue previously. Enzyme activity has largely been documented by histochemical and physiological reports. In this investigation, specific antisera were used to study the distribution of the cytosolic carbonic anhydrase II and III isoenzymes in the quail kidney. Comparison between the present findings and the corresponding histochemical patterns, previously obtained in the same species by a cobalt phosphate precipitation method, resulted in the bulk of renal carbonic anhydrase activity being attributed to the carbonic anhydrase II isoenzyme. Conversely, moderate carbonic anhydrase III immunostaining appeared to be confined to the smooth muscle cells of ureteral and arteriolar walls. Indirect evidence of the occurrence, in the quail kidney, of a membrane-associated carbonic anhydrase form, antigenically distinct from the II and III isoforms, was inferred.  相似文献

Anomalous giant granules of beige (bg) mice have been used as a cell marker in the study of cell lineage of mast cells. Similar granules are known to exist in other tissues including kidney proximal tubules and liver parenchymal cells. In the present study, these granules were found to give yellow or orange autofluorescence when the tissue had been fixed with formaldehyde and embedded in paraffin. Thus, the granules can be used as a cell marker that can be visualized in serial paraffin sections without any specific histochemical staining. Chimaeric mice were produced by aggregation of 8-cell-stage embryos of beige (C57BL/6J-bgJ/bgJ) and A/J strains. The chimaeric liver showed beige cell patches with complicated shapes, although the patches frequently conformed to the shape of parenchymal cell cord or plate structures. In chimaeric kidney, beige cells formed coherent patches in the proximal tubules. The tubules were found to contain more than one clone. The patches frequently had long extended shapes suggesting growth of the clone along the tubule axis. Three-dimensional image reconstruction from the serial paraffin sections was carried out with the aid of a computer-assisted image analysis system, resulting in a clearer image of the patch shape.  相似文献

Summary This paper reports the existence of granulated cells in the bovine gallbladder epithelium. The histochemical studies performed on these cells showed that their cytoplasmic granules contain heparin. Optical- and electron microscopic observations demonstrated that these mast cells migrated from the adjacent connective tissue, and crossed the basal membrane in order to enter the epithelium and reach the luminal surface.  相似文献
