Ethephon and the ethylene inhibitors Ag+ and aminoethoxyvinylglycine (AVG) inhibited outgrowth of the axillary bud of thefirst trifoliate leaf in decapitated plants of Phaseolus vulgaris.Endogenous ethylene levels decreased in the stem upon decapitationalthough it is not conclusive that a causal relationship existsbetween this decrease and the release of axillary buds frominhibition. The proposition that auxin-induced ethylene is responsiblefor the suppression of axillary bud growth in the decapitatedplant when the apical shoot is replaced by auxin is not borneout in this study. Application of IAA directly to the axillarybud of intact plants gave rise to a transient increase in budgrowth. This growth increment was annulled when AVG was suppliedwith IAA to the bud despite the fact that the dosage of AVGused did not affect the normal slow growth rate of the bud ofthe intact plant or bud outgrowth resulting from shoot decapitation.  相似文献

Correlative Inhibition in the Shoot of Agropyron repens ( L.) Beauv   总被引:1,自引:0,他引:1  
Correlative inhibition was investigated in plants of Agropyronrepens at two temperatures. Reciprocal inhibition ocrurred betweenthe main shoot apex and the outgrowing axillary shoots, withthe balance of inhibition varying with temperature. Apical dominancewas stronger at 10 °C than at 20 °C , but even at 10°C release of apical dominance by decapitation had onlyminor effects on the timing of outgrowth, growth pattern andrate of dry weight aocumulation of the axillary shoots. Dominanceof the main shoot apex by the axillary shoots was stronger at20 °C than at 10 °C. Removal of axillary buds preventeddecline in size and activity of the main shoot apex ard resultedin increased rates of primordium initiation, leaf emergenceand dry weight accumulation in the main shoot. It is suggestedthat a system of reciprocal dominance provides a mechanism formaintaining the characteristic habit of the grass plant andlimits growth in height of vegetative shoots. The effect of axillary bud age on the development and potentialfor growth of the bud into a shoot was studied in roses. Ageof the buds occupying a similar position on the plant variedfrom 'subtending leaf just unfolded' up to 1 year later. Withincreasing age of the axillary bud its dry mass, dry-matterpercentage and number of leaves, including leaf primordia, increased.The apical meristem of the axillary bud remained vegetativeas long as subjected to apical dominance, even for 1 year. The potential for growth of buds was studied either by pruningthe parent shoot above the bud, by grafting the bud or by culturingthe bud in vitro. When the correlative inhibition (i.e. dominationof the apical region over the axillary buds) was released, additionalleaves and eventually a flower formed. The number of additionalleaves decreased with increasing bud age and became more orless constant for axillary buds of shoots beyond the harvestablestage, while the total number of leaves preceding the flowerincreased. An increase in bud age was reflected in a greaternumber of scales, including transitional leaves, and in a greaternumber of non-elongated internodes of the subsequent shoot.Time until bud break slightly decreased with increasing budage; it was long, relatively, for 1 year old buds, when theysprouted attached to the parent shoot. Shoot length, mass andleaf area were not clearly affected by the age of the bud thatdeveloped into the shoot. With increasing bud age the numberof pith cells in the subsequent shoot increased, indicatinga greater potential diameter of the shoot. However, final diameterwas dependent on the assimilate supply after bud break. Axillarybuds obviously need a certain developmental stage to be ableto break. Kinetin (1•10–4 M and 1•10–3 M) was appliedto the plumules of 6-day-old Chenopodium rubrum plants. Effectson growth, anatomical structure and organogenesis in the apicalmeristem were followed. Floral differentiation as affected bykinetin was also investigated in plants induced to flower byshort-day treatment. Kinetin increased mitotic activity in the apical meristems inboth induced and non-induced plants. The effect was most pronouncedin the peripheral and subcentral zone. An increase in nucleolussize and a higher degree of pyroninophilia in the peripheralzone was also observed, indicating a localized promotion ofRNA synthesis. A higher rate of leaf initiation and a stimulationof leaf and stem growth was subse quentiy recorded. The growthof axillary meristems and of bud primordia was promoted onlyat the lower concentration of kinetin (1•10–4 M),in both photoperiodically-induced and non-induced plants. However,the pattern of lateral bud growth differed from that found innormal floral differentiation. In kinetintreated plants, thebud primordia are isolated from the summit of the shoot apexby a succession of rapidly growing leaves. The enhancement ofleaf growth leads to correlative inhibition of axillary budpriniordia and results, finally, in a suppression of floraldifferentiation. The inhibitory effect of kinetin on floweringwas compared with that of auxin. Inhibition of flowering occurredin both cases but is achieved in two different ways.  相似文献

Development of axillary shoots was induced in embryonic axesof the dipterocarp Shorea roxburghii G. Don. cultured on a modifiedMS medium containing 6-benzyl-aminopurine (BAP) at an optimumconcentration of 5 mg I–1. Excised axillary shoots wereused in multiplication and rooting experiments. Cucumber explants including at least part of the cotyledon,a short section of hypocotyl, and the apical bud, are capableof producing multiple axillary buds from the seedling apex andadventitious shoots from the hypocotyl base in a medium whichcontains 2·0 mg dm–3 of kinetin. Removal of theapical bud triples the number of shoots produced from the apexof explants with two intact cotyledons but does not affect shootproduction from explants with some or all of their cotyledonsremoved. The area of intact cotyledon also influences morphogenesis,as explants with both cotyledons removed, failed to produceadventitious shoots from the hypocotyl base. Culture in continuousdarkness entirely prevents shoot development from the explantbase, but has little influence on shoot production from theapex. The influence of endogenous growth regulators and apicaldominance on the morphogenesis of shoots in cucumber seedlingsare discussed. The growth of garden orache, A triplex hortensis was studiedunder conditions of mild NaCl or Na2SO4 salinity. Growth, drymatter production and leaf size were substantially stimulatedat 10 mM and 50 mM Na+ salts. Increased growth, however, appearedto be due to a K+-sparing effect of Na+ rather than to salinityper se. The distribution of K+ and Na+ in the plant revealeda remarkable preference for K+ in the roots and the hypocotyl.In the shoot the K/Na ratio decreased strongly with leaf age.However, the inverse changes in K+ and Na+ content with leafage were dependent on the presence of bladder hairs, which removedalmost all of the Na+ from the young leaf lamina. Measurementsof net fluxes of K+ and Na+ into roots and shoots of growingAtriplex plants showed a higher K/Na selectivity of the netion flux to the root compared to the shoot. With increasingsalinity the selectivity ratio SK, Na* of net ion fluxes tothe roots and to the shoots was increased. The data suggestthat recirculation of K+ from leaves to roots is an importantlink in establishing the K/Na selectivity in A. hortensis plants.The importance of K+ recirculation and phloem transport forsalt tolerance is discussed. Suitable cytokinin supplements and culture environments havebeen determined for the initiation and establishment of shootcultures of Quercus robur seedling tissue. Initiation of axillaryshoot development from nodal explants required culture mediumsupplemented with BA (6-benzylamminopurine). The greatest numbersof stem segments for culture proliferation were obtained using1.0 mg I-1 BA after 56 d culture. The frequency of shoot developmentand subsequent formation of multiple shoots at initiation wasinfluenced by the position of the nodal explant in the seedlingshoot, incubation temperature and daylength. Explants from basaland apical regions, which contained multiple axillary buds,produced the lowest frequencies of axillary shoot developmentand multiple shoot formation, many remained quiescent. Axillaryshoot development was greatest in single nodal explants excisedfrom the midstem positions, elongated regions of the shoot wherenodes were formerly associated with a leaf. Higher temperaturesstimulated shoot formation with greater numbers of stem segmentsfor culture multiplication being obtained from nodal explantsincubated at 25C. Axillary shoot development was promoted innodal explants maintained under daylengths of 16 h or more.Stem segments cut from axillary shoots which developed fromnodal explants were used to establish shoot multiplication cultureson medium supplemented with 0.4 mg I-1 BA. Shoot formation fromstem segments was greater at higher incubation temperaturesof 25C and 30C. Multiplication coefficients for stem segmentsincreased after one subculture. The leaf growth, tiller production, light interception, anddry weight increase of a simulated sward of S24 perennial ryegrass(Lolium perenne) were followed during the development of thesward from a collection of two-leaved seedlings to a closedcanopy with an LAI of 23, of which 15 consisted of green leaflaminae. The dry weight of live shoots increased exponentiallyat first, but then entered a long linear phase of increase.This was equivalent to a crop growth rate of 200 Kg ha–1day–1 and a conversion efficiency of radiant energy (400–700nm) of 7.2 per cent. Towards the end of the growth period therate of increase of live shoots declined rapidly to zero anda ceiling yield was reached equivalent to 10 metric tons ha–1.Leaf growth continued at a high rate, but was equalled by therate of leaf death, so that the weight of live leaf tissue remainedconstant. By this time the swards had achieved a stable tillerpopulation (about 1 cm–1), each tiller bore a constantnumber of live leaves (about three), and the length of eachnewly expanded leaf equalled the length of the old leaf it replaced(about 70 cm). The swards were grown in Perlite so that in theabsence of soil fauna dead leaves accumulated at the base ofthe sward where, after 12 weeks, they accounted for 19 per centof the total weight of dry matter produced.  相似文献

In both reproductive and vegetative plants of Lolium temulentumL., the export of 14C-labelled assimilates from each healthyleaf on the main shoot to terminal meristem, stem, tillers,and roots was measured each time a new leaf was expanded, fora period of 5 to 6 weeks. Some labelled assimilates moved fromeach leaf on the main shoot to every meristem in the same shoot,as well as to the tops and roots of adjacent organically attachedtillers. The terminal meristem of the reproductive shoot, which includedthe developing inflorescence, received 70–80 per centof the carbon assimilated by the emerged portion of the growingleaf, 15–25 per cent of the carbon assimilated by thetwo youngest expanded leaves, and 5–10 per cent of thatfrom each of the older leaves. A similar pattern of carbon supplyto the terminal meristem was found in vegetative shoots, exceptthat older leaves on young vegetative shoots supplied even lessof their carbon to the terminal meristem. The general conclusionis that developing leaves at the tip of the shoot receive aboutthe same proportion of carbon from each leaf as does a developinginflorescence. Young expanded leaves provided most labelled assimilates forstem growth; during both reproductive and vegetative growth,expanded leaves increased their export of labelled carbon tostem, and exported less of their 14C to roots and sometimesto tillers. In these reproductive and vegetative shoots, grown in a constantexternal environment, the major changes in the pattern of distributionof labelled assimilates appeared to be the result of increasedmeristematic activity in stem internodes; the development ofan inflorescence had no obvious direct effect on the carboneconomy of shoots.  相似文献

Barley plants (Hordewn vulgare L. cv. Atem) were grown fromseed for 28 d in flowing solution culture, during which timeroot temperature was lowered decrementally to 5?C. Plants werethen subjected to root temperatures of 3, 5, 7, 9, 11, 13, 17or 25 ?C, with common air temperature of 25/15 ?C (day/night).Changes in growth, plant total N, and NO3 levels, andnet uptake of NH4+ and NO3 from a maintained concentrationof 10 mmol m–3 NH4NO3 were measured over 14 d. Dry matterproduction increased 6-fold with increasing root temperaturebetween 3–25 ?C. The growth response was biphasic followingan increase in root temperature. Phase I, lasting about 5 d,was characterized by high root specific growth rates relativeto those of the shoot, particularly on a fresh weight basis.During Phase I the shoot dry weight specific growth rates wereinversely related to root temperature between 3–13 ?C.Phase 2, from 5–14 d, was characterized by the approachtowards, and/or attainment of, balanced exponential growth betweenshoots and roots. Concentrations of total N in plant dry matterincreased with root temperature between 3–25 ?C, moreso in the shoots than roots and most acutely in the youngestfully expanded leaf (2?l–6?9% N). When N contents wereexpressed on a tissue fresh weight basis the variation withtemperature lessened and the highest concentration in the shootwas at 11 ?C. Uptake of N increased with root temperature, andat all temperatures uptake of NH4+, exceeded that of NO3,irrespective of time. The proportions of total N uptake over14 d absorbed in the form of NH4+ were (%): 86, 91, 75, 77,76, 73, 77, and 80, respectively, at 3, 5, 7, 9, Il, 13, 17,and 25 ?C. At all temperatures the preference for NH4+ overNO3 uptake increased with time. An inverse relationshipbetween root temperature (3–11 ?C) and the uptake of NH4+as a proportion of total N uptake was apparent during PhaseI. The possible mechanisms by which root temperature limitsgrowth and influences N uptake are discussed. The length and basal diameter of all lateral and terminal budsof vegetative annual shoots of 7-year-oldJuglans regia treeswere measured. All buds were dissected and numbers of cataphylls,embryonic leaves and leaf primordia were recorded. Each axillarybud was ranked according to the position of its associated leaffrom the apex to the base of its parent shoot. Bud size andcontent were analysed in relation to bud position and were comparedwith the size and number of leaves of shoots in equivalent positionswhich extended during the following growing season. Length andbasal diameter of axillary buds varied according to their positionon the parent shoot. Terminal buds contained more embryonicleaves than any axillary bud. The number of leaves was smallerfor apical and basal axillary buds than for buds in intermediatepositions on the parent shoot only. All new extended shootswere entirely preformed in the buds that gave rise to them.Lateral shoots were formed in the median part of the parentshoot. These lateral shoots derived from buds which were largerthan both apical and basal ones. A quantitative analysis of the 14C-labelled assimilate suppliedby the expanded leaves on the primary shoot to growing leaves,stem, lateral shoots (branches or stolons) and roots in redand white clover was conducted during vegetative growth. Stem growth of the primary shoot was inhibited in both cloversand utilized no energy resources. The growing leaves at theprimary shoot apex of white clover imported 4 per cent of theshoot's assimilate compared with 10 per cent in red clover.At the basal end of the primary shoot, the tap root of whiteclover imported 16 per cent of the shoot's assimilate comparedwith 22 per cent in red clover. Branches in red clover and stolonsin white clover were by far the largest sinks for primary shootassimilate, importing 39 per cent and 63 per cent of the labelledassimilate, respectively. Analyses of the translocation of assimilate from individualprimary shoot leaves demonstrated that in both clovers olderleaves exported more of their assimilate to branches or stolons,whereas younger leaves exported more of their assimilate toroots, and possibly in white clover, to growing leaves at thetip of the shoot. Of the labelled assimilate exported to branchesor stolons, each primary shoot leaf exported preferentiallyto the branch or stolon in its own axil, but in addition exportedsubstantial quantities of assimilate to all other axillary shoots,particularly those arising from basal axils where the subtendingleaf had died. A comparative examination has been made under controlled conditionsof the interacting effects on growth in the vegetative phasewhich takes place when the diurnal temperature is either maintainedconstant at 20 and 25 ?C or reduced at night by 5 and 10 ?Cand the plants subjected daily to either 2.16 or 4.32 ? 104lx for 14 h. For each species (Gossypium hirsutum, Hclianthusannuus, Phaseolus vulgaris and Zea mays) the changes in netassimilation rate, the leaf area ratio and the relative growthrates in plant weight and leaf area were recorded. Contrary to some previous findings, none of these componentsof growth are favoured by cool nights. Rather such reductionsin temperature retard the growth processes to a varying degreeaccording to the species and the component. In general, significantreductions are more evident for the two relative growth rateswhen a drop from 20 to 10 ?C is combined with the lower levelof illumination. The implications of these results are considered in relationto a prior study where for similar light conditions the samespecies plants were subjected to constant temperatures rangingfrom 10 to 25 ?C.  相似文献

Factors controlling growth and tuberization of axillary budsin shoots of plantlets of potato (Solarium tuberosum L.) culturedin vitro were investigated. Correlative inhibition restrainedgrowth and tuberization of the axillary buds. Exposure of intactplantlets for various periods (4 to 48 h) to low (2 or 12C)or high (30 C) temperatures as comparedto 18C, did not alleviatecorrelative inhibition. Removal of the apical part of the shoot,the roots or both was generally ineffective Elevating sucroseconcentration from 30 to 80 g dm–3 promoted tuberizationon axillary buds, and the cytokinin 6-(-dimethylallylamino)purine (2iP), alleviated correlative inhibition and enhancedtuberization in intact plantlets. In the whole plantlet mostof the tubers were formed on the basal nodes, however, oncecorrelative inhibition was eliminated by the dissection of theshoot to single node sections, tubers were formed on every axillarybud. The single most effective factor inducing tuberizationin single node sections was the growth retardant ancymidol,an inhibitor of giberellin biosynthesis. The shoot growth of a deciduous tree, Acanthopanax sciadophylloidesFranch. et Savat. shows inter-annual intermittent repetitionof two distinctive phases, a stagnant growth phase (S-phase)and vigorous extension-growth (E-phase). To help understandthe differentiation mechanism, shoot development was studiedover time in both shoot phases. S-phase and E-phase shoots weredistinguished from each other by morphological traits: S-phaseshoots are characterized by higher allocation to leaves anda shorter period of stem growth, while E-phase shoots show continuousstem extension over the growing season. Specific leaf area didnot differ between the two phases. This shoot differentiationwas similar to the morphological differentiation of shoots betweenlong vs. short shoots found in some temperate trees. Leavesof both phases were well-dispersed through adjustment of petiolelength and leaf-blade size to reduce mutual shading within ashoot. Stem-wood density of current-year shoots was lower inE-phase compared with S-phase shoots. Leaves produced earlyin the season affected the growth phase of the following year.These results suggest that annual shoot differentiation of A.sciadophylloides was determined during the previous season andreflects leaf productivity in a given habitat during that growingseason. The properties and mode of action of a new plant growth retardant,(E)-1-(4-chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazol-1-yl)-1-penten-3-ol(S-3307), were investigated. When a dipping method was used,S-3307 at 2.2 ? 10-7 M (0.064 ppm) retarded the growth of riceplants by 50% of the value found for the control. The retardationof growth was removed by a gibberellin application (8.7?10-5M). S-3307 had nearly no effect on the shoot elongation inducedby gibberellin. The amounts of gibberellin-like substances inrice shoots were decreased by S-3307 treatment in proportionto the degree of growth retardation. This observation was confirmedwith GA1 and GA19, the main gibberellins in the rice plant.Our results indicate that S-3307 inhibits gibberellin biosynthesisin rice plants. Plants of Lycoperstcon esculentum Mill. cv. Ailsa Craig wildtype and flacca (flc) were sprayed daily with H2O or 2?10–2mol m–3 abscisic acid (ABA). ABA treatment effected apartial phenotypic reversion of flc shoots; leaf areas wereincreased and transpiration rates decreased. Leaf expansionof wild type shoots was inhibited by ABA. Indoleacetic acid (IAA), ABA and l-aminocyclopropane-l-carboxylicacid (ACC) concentrations were determined by combined gas chromatography-massspectrometry using deuterium-labelled internal standards ABAtreatment for 30 d resulted in greatly elevated internal ABAlevels, increasing from 1?0 to 4?3 and from 0?45 to 4?9 nmolg–1 fr. wt. in wild type and flc leaves respectively.Endogenous IAA and ACC concentrations were much lower than thoseof ABA. IAA content ranged from 0?05 to 0?1 nmol g–1 andACC content from 0?07 to 0?24 nmol g–1 Ethylene emanationrates were similar for wild type and flc shoots. Wilting of detached leaves induced a substantial increase inethylene and ACC accumulation in all plants, regardless of treatmentor type. Ethylene and ACC levels were no greater in flc leavescompared to the wild type. ABA pretreatment did not preventthe wilting-induced increase in ACC and ethylene synthesis. Indol-3yl acetic acid (10–4M) was applied to the plumulesof Chenopodium rubrum. Effects on the anatomical structure andthe growth pattern in the apical meristem, as well as DNA synthesisand nucleolus size were investigated. When auxin is applied before or during photoperiodic inductionit inhibits DNA synthesis and meristematic activity. The axillarymeristem (i.e. a group of cells in the axils of the leaf primordia)is most affected. A similar inhibition of the axillary meristemwas also observed in non-induced control plants grown in continuouslight. Auxin applied simultaneously with photoperiodic inductioncounteracts the reduction of apical dominance in the apex andthus inhibits the onset of floral differentiation. Auxin appliedfollowing induction inhibits the previously-formed buds andmakes possible a more complete development of the apical flower. The dual effect of IAA on flowering, inhibitory and stimulatory,manifests itself as a growth response at different stages ofthe changing shoot apex.  相似文献

The robustness of a leaf elongation bioassay was evaluated byconducting trials with detached shoots of wheat at several differenttemperatures. Leaf elongation rate (LER) was monitored for shootsfed either an artificial xylem solution or xylem solution plus10–3mol m–3 abscisic acid (ABA). Consistent resultswere obtained when periodic ruler measurements of many shootswere made and compared with simultaneous measurements on a singleshoot made with a linearly variable displacement transducer(LVDT). ABA treatment consistently inhibited leaf growth; however,the magnitude of the inhibition was dependent on the temperatureat which the assay was conducted. Interpretation of resultsfrom such bioas-says in terms of ABA concentration suppliedto the detached shoots is complicated by this observation sincethere is no unique relationship between leaf growth inhibitionand ABA concentration.
