Summary Bacteria with specific temperature sensitive lethal mutations in the gene for the subunit of RNA polymerase synthesize both the and subunits at a several fold higher rate at 42°C than wildtype cells relative to total protein. Synthesis of the and subunits proceeds at essentially the wild-type rates under these conditions. In contrast, a mutant with a temperature sensitive lethal mutation in the subunit gene synthesizes and at 42°C at slightly lower rates than wild-type, while and synthesis is not significantly altered. In all of the mutants at 42°C, newly synthesized subunits are stable, while the , and subunits are rapidly degraded. The apparent uncoupling of from subunit synthesis seen in the mutants at 42°C might suggest that the synthesis of these subunits is at least in part controlled by different mechanisms.  相似文献

A presumed XY chromosome pair is described fromt estis squashes from the mesopelagic deep-sea fish Bathylagus wesethi, whose 2N chromosome number was determined as 34-XY. Although the metacentric X-chromosome is the largest in the entire compliment, the Y is the smallest and only acrocentric element. The positive heteropycnosis of the sex elements was not easily distinguishable in the nuclei of first meiotic prophase. Tetraploid nuclei were observed in peripheral supporting cells of the testis. Summary The seven possible primary trisomics of Petunia (2 n= 14) located in the progenies of triploid, hypertriploid and hypotriploid plants were distinguished from one another and from diploid on the basis of cytological and morphological criteria. They were provisionally named as Oval, Semi, Slender, Pseudonormal, Arrow, Narrow and Giant. In three of the trisomics, the extra chromosome was identified for the first time at pachytene stage. Postpachytene studies revealed no precise relationship between the length of extra chromosome and the frequency of multiple association.  相似文献

The N-terminal domain (1–318 amino acids) of mouse NFB (p65) has been purified to homogeneity from the soluble fraction of Escherichia coli cells expressing this protein. Its complex with a full-length iB- (MAD3, 1–317 amino acids) molecule was generated by binding the E. coli-derived iB- to the purified NFB and purifying the complex by sequential chromatography. The stoichiometry of NFB to iB in the complex was determined to be 2 to 1 by light scattering and SDS–polyacrylamide gel electrophoresis. The secondary structure of the NFB (p65) determined by Fourier-transform infrared (FTIR) spectroscopy is in good agreement with that of the p50 in the crystal structure of the p50/DNA complex, indicating that no significant structural change in NFB occurs upon binding of DNA. The FTIR spectrum of the NFB/iB complex indicates that its secondary structure is composed of 17% -helix, 39% -strand, 18% irregular structures, and 26% -turns and loops. By comparing these data to the FTIR data for NFB alone, it is concluded that the iB (MAD3) in the complex contains 35% -helix, 27% -strand, 22% irregular structures, and 16% -turns and loops. Circular dichroism (CD) analysis of a shorter form of iB (pp40) indicates that it contains at least 20% -helix and that the iB subunit accounts for nearly all of the -helix present in the NFB/iB complex, consistent with the FTIR results. The stabilities of NFB, iB, and their complex against heat-induced denaturation were investigated by following changes in CD signal. The results indicate that the thermal stability of iB is enhanced upon the formation of the NFB/iB complex.  相似文献

The sialidase of swine influenza A viruses of N1 and N2 subtypes, isolated from 1930 to 1992, was studied for substrate specificity with ganglio-series, lacto-series type II and GM3 gangliosides containing Neu5Ac2-3Gal, Neu5Gc2-3Gal and Neu5Ac2-6Gal linkages. All viral sialidases tested showed that the activity for hydrolysing substrates with Neu5Ac2-3Gal was higher than the activities with Neu5Gc2-3Gal and Neu5Ac2-6Gal linkages. When GM1b, GM3 and sialylparagloboside were used as substrates, the earliest strain (A/Wisconsin/15/30 H1N1, isolated in 1930) showed the activity ratio of Neu5Ac2-6Gal to Neu5Ac2-3Gal to be 0.13:0.2, and the ratio Neu5Gc2-3Gal/Neu5Ac2-3Gal to be 0.19:0.37, while those strains isolated from 1978 to 1992 exhibited ratios of 0.29:0.58 for Neu5Ac2-6Gal/Neu5Ac2-3Gal and 0.51:0.76 for Neu5Gc2-3Gal/Neu5Ac2-3Gal. Summary A mutant strain of Rhodococcus equi accumulates three metabolites from the androst-4-ene-3,17-dione or from its degradation intermediate, 3a-H-4(3'-propionic acid)-7a-methylhexahydro-1,5-indanedione (MEPHIP). These three metabolites are: 3a-H-4a(3'-propionic acid)-5-hydroxy-7a-methylhexahydro-1-indanone--lactone (HIL); 3a-H-4(3'-trans acrylic acid)-5-hydroxy-7a-methylhexahydro-1-indanone (2'-5-hydroxy-MEPHIP); and 3a-H-4(3'-hydroxy-3'-propionic acid)-5-hydroxy-7a-methylhexahydro-1-indanone (3'-hydroxy-HIL). The behaviour of this mutant allows us to propose a pathway for degradation of the intermediates, methylperhydroindanone propionates. Residual toxicities of registered and selected experimental pesticides used on citrus against Agistemus industani Gonzalez (Acari: Stigmaeidae) were compared. Pesticides considered highly toxic to A. industani were: abamectin 0.15 EC at 731ml/ha+FC 435-66 petroleum oil at 46.8l/ha, pyridaben 75WP at 469g/ha, ethion 4EC at 7.01l/ha+FC 435-66 petroleum oil at 46.8l/ha, propargite 6.55 EC at 3.51l/ha, chlorfenapyr 2SC at 1.46l/ha applied alone or in combination with FC 435-66 petroleum oil at 46.8l/ha, sulphur 80DF at 16.81kg/ha, dicofol 4EC at 7.01l/ha, fenbutatin oxide 50WP at 2.24kg/ha, benomyl 50WP at 2.24kg/ha, benomyl 50WP at 1.68kg/ha+ferbam 76 GF at 5.60kg/ha, ferbam 76GF at 11.21kg/ha, neem oil 90EC at 46.8l/ha, and copper hydroxide DF (40% metallic copper) at 4.48kg metallic copper/ha+FC 435-66 petroleum oil at 46.8l/ha. Pesticides that were moderately to slightly toxic included: copper sulphate 98% at 4.48kg metallic copper/ha+FC 435-66 petroleum oil at 46.8l/ha, fenbuconazole 2F at 280ml/ha+FC 435-66 petroleum oil at 46.8l/ha, FC 435-66 petroleum oil applied alone at 46.8l/ha or 23.4l/ha, and diflubenzuron 25WP at 1.40kg/ha. Pesticides that were non-toxic included: fenbuconazole 2F at 585ml/ha, malathion 57EC at 5.85l/ha, FC 435-66 petroleum oil at 46.8l/ha, carbaryl 80S at 3.36kg/ha, chlorpyrifos 4EC at 4.68l/ha, and formetanate 92SP at 1.12kg/ha. Understanding the toxic effects of field weathered pesticides against key predacious mite species is important for effective IPM. The results of this study provide a comparison of direct and indirect toxic effects of various pesticides to A. industani under field conditions.  相似文献

We have used restriction fragment length polymorphism (RFLP) analysis and DNA sequencing to characterize two distinct DRB1 alleles expressed on DRw52 and DQw7-associated haplotypes but not readily defined by conventional DR serology. These two haplotypes, designated HLA-D HAG and PEV, react variably with DRw13(w6), DRw14(w6), and the more broad DR 3+6 antisera. Analysis of RFLP revealed that HLA-D HAG and PEV are associated with different DRw52 variants, and that HAG is indistinguishable from DRw18(3) haplotypes. Sequencing of the HAG and PEV DRB1 genes showed each to represent novel alleles. Nevertheless, these sequences show similarities with the other alleles of the DR5, w6, and w8 family. HAG (DRB1*1303) appears to have arisen either from two recombinational events involving at least three DRB1 sequences (DRB1*1101, DRB1*0803, DRB1*0401) or from a single recombinational event together with multiple point mutational events. PEV appears to represent a DRB1*1301-1302/DRB1*1101 recombinant allele, with recombination having occured in the region of bases 175 – 198. The results of this study suggest that the DRw52 family haplotypes is derived from a relatively restricted number of ancestral sequences, with diversity among DRB1 alleles within this family arising through gene conversion or recombination events.  相似文献

This study provides a critical historical review and analysis of the variety of human expressions which have been erroneously labeled under the grandiose category mass hysteria. It is argued that Western science reductionist approaches to the classification of mass hysteria treat it as an entity to be discovered transculturally, and in their self-fulfilling search for universals systematically exclude what does not fit within the autonomous parameters of its Western-biased culture model, exemplifying what Kleinman (1977) terms a category fallacy. As a result of objectivist methodologies, the etiology of actions labeled as mass hysteria is typically viewed as deviant, irrational or abnormal behavior resulting from a malfunctioning proper social order. However, what constitutes the correct social order is a function of a researcher's historical sociocultural and/or scientific milieu. This study reviews the problem, advocating Geertz's (1973) culturally relativistic approach to understanding various cross-cultural behavior that is sensitive to and tolerant of the unique context and milieu of participants. Mass or epidemic hysteria is viewed as an invention of Western psychiatry and should be abandoned and replaced with the term collective exaggerated emotions. Instead of attempting to discover a neatly packaged, unitary external disease entity, the focus of a meaning-oriented approach emphasizes the deciphering of foreign realities, semantic networks and symbol systems.  相似文献

The carbohydrate-binding specificity ofPseudomonas aeruginosa lectin I (PA-I) in iodinated or biotinylated form was studied. A large number of glycosphingolipids, as well as some glycoproteins and neoglycoproteins were used as ligands. Also, inhibition by free saccharides of PA-I binding to glycosphingolipids was tested. A case of cirrhotic liver harbouring three atypical macroregenerative nodules and an hepatocellular carcinoma was immunocytochemically investigated for the expression of VLA- 1, VLA-2 and VLA-5 integrins and for different extracellular matrix (ECM) components (collagen I, collagen IV, laminin, fibronectin and tenascin). In addition, the proliferative activity within the nodules was evaluated, using the MIB 1 monoclonal antibody (MAb). The cirrhotic liver disclosed a continuous staining pattern of the ECM proteins investigated, as well as a sinusoidal immunostaining of VLA-1, VLA-2 and VLA-5. The macroregenerative nodules showed a discontinued immunoreactivity for ECM proteins while maintaining a VLA-1 sinusoidal immunostaining, coupled with intercellular immunostaining. VLA-2 and VLA-5 expression was lacking. The growth fraction was low in both the above pathological conditions. The hepatocellular carcinoma was devoid of any ECM immunostaining. VLA-1 immunoreactivity exhibited a honeycomb pattern of staining, whereas VLA subunits were absent. MIB1 expression was high, being present in 30% of neoplastic nuclei. Summary The content of free amino acids was determined in erythrocytes of adult Leghorn (Lg, White Rock (WR) and Cornish (Cr) hens, bred under identical conditions. The concentration of total amino acids was twice as high in the erythrocytes as in plasma, amounting to 396 m/100 ml, 424 m/100 ml and 475m/100 ml in White Rock, Cornish and Leghorn hens, respectively.Significant differences were found in the ratio of basic amino acids to acidic amino acids. These values were 0.76, 1.75 and 3.19 in White Rick, Leghorn and Cornish hens, respectively; in the plasma of all 3 breeds the ratio was 1. Statistically significant interbreed differences were expressed more distinctly in erythrocyte than in plasma amino acid concentrations. For absolute concentrations the differences were significant in the case of 9 amino acids.  相似文献

The present study sought to identify and partially characterize the glycoconjugates specific to the double-layered ciliary body epithelium of the rat eye by lectin histochemistry and lectin blottings. Hydrated paraffin sections of Carnoy-fixed Sprague-Dawley rat eyes were stained with a panel of 21 different biotinylated lectins, followed by streptavidin-peroxidase and the glucose oxidase-diaminobenzidine-nickel staining procedure. The results of lectin histochemistry revealed that the inner epithelial layer was rich in GlcNAc(1,4)GlcNAc, -Gal, Gal(1,3)GalNAc, GalNAc(1,3)GalNAc/Gal, GalNAc(1,6)Gal, Fuc(1,2)Gal(1,4)GlcNAc and Gal(1,4)GlcNAc(1,2)Man(1,6) sugar residues as shown by its positive reactivities with S-WGA, PWA, DSA, GS-I-B4, PNA, DBA, SBA, WFA, UEA-I, LTA and PHA-E. The reactivities of GS-I-B4, PNA, DBA and SBA were restricted to the inner layer at the tips of the ciliary processes. On the other hand, the outer epithelial layer was stained evenly by DSA and Jacalin, and partly by MAA, showing that this epithelial layer was rich in GlcNAc(1,4)GlcNAc, Gal(1,3)GalNAc and NeuAc(2,3)Gal disaccharides. These lectin binding patterns of the ciliary body epithelium suggest a topographical and functional difference in this double cell-layered epithelium. Their possible roles in the secretion of aqueous humour and production of ciliary zonule are discussed. Some identified lectin markers specific to these two cell layers may be useful for further experimental studies. Glycoproteins extracted from the dissected ciliary body were separated by SDS-PAGE electrophoresis and analyzed by protein blottings with 8 different lectins. The results showed that at least 10 major membrane-bound glycoproteins, with molecular weights ranging from 30 to 150kD, rich in -GlcNAc, -Gal, /-GalNAc and NeuAc(2,6)Gal residues, were present in the microsomal fraction.  相似文献

Summary Five subunits (-, -, -, - and -subunits) of the six -and -subunits) in the F1 portion (F1ATPase) of sweet potato (Ipomoea batatas) mitochondrial adenosine triphosphatase were isolated by an electrophoretic method. The - and -subunits were not distinguishable immunologically but showed completely different tryptic peptide maps, indicating that they were different molecular species. In vitro protein synthesis with isolated sweet potato root mitochondria produced only the -subunit when analyzed with anti-sweet potato F1ATPase antibody reacting with all the subunits except the -subunit. Sweet potato root poly(A)+RNA directed the synthesis of six polypeptides which were immunoprecipitated by the antibody: two of them immunologically related to the -subunit and the others to the - and -subunits. We conclude that the -subunit of the F1ATPase is synthesized only in the mitochondria and the -, - and -subunits are in the cytoplasm.  相似文献

Zusammenfassung Uferschwalben kehren aus den afrikanischen Winterquartieren in Trupps beiderlei Geschlechts zurück. Erste Beringungsergebnisse belegen, daß zunächst mehrjährige, vermutlich untereinander bekannte Vögel eintreffen, die den Brutplatz aus vergangenen Brutperioden her kennen. Die Masse der später ankommenden Vögel dürfte weitgehend aus einjährigen oder ortsfremden Uferschwalben bestehen, die sich größtenteils erst während der Paarbildung persönlich kennenlernen. Der anfängliche Schwarmzusammenhalt der nacheinander eintreffenden Trupps führt zur Bildung von Subkolonien, die für Brutplätze ab einer bestimmten Größenordnung typisch sind. Uferschwalben- gründen nacheinander mehrere Reviere, d. h. sie besetzen Steilwandbereiche, in denen sie ausschließlich mit den Füßen eine Röhre oder Mulde graben, singen und Bogenflüge starten. Bis auf singende oder bekannte werden Artgenossen im Revier geduldet. Uferschwalben- suchen besetzte Reviere auf. Ohne Röhrenbindung verhalten sie sich still und unauffällig, ihre Grabungsaktivitäten sind von untergeordneter Bedeutung. Die Bindung an ein bestimmtes Revier entwickelt sich individuell verschieden und entscheidet über den Abschluß des Röhrenbaues (Herstellung der Nistkammer). Reviere ohne dauerhafte -Bindung werden von den aufgegeben. Aktivitäten, die auf wachsende Revierbindung eines hindeuten, sind: häufige oder/und länger dauernde Aufenthalte des in einem besetzten Revier und sporadisches Mitgraben; aggressives Verhalten gegenüber Artgenossen (i. d. R. fremde ), die im Revier landen wollen; gemeinschaftlicher, leiser Gesang von und im Röhrenbereich. Aktivitäten, die für eine vollzogene Paarbildung sprechen, sind: Fertigstellen der Röhre durch Grabung der Nestkammer; längere gemeinsame Aufenthalte innerhalb und außerhalb der Röhre; Voranfliegen des beim Röhrenanflug; Übernachten von und in der Röhre; Nestbau; ausdauernde Verfolgungsflüge während der Kopulationsphase. Die Paarbildung ist demnach ein individueller Prozeß, bei dem die Aktivitäten der im Revier als Werbung, die der als Revierwahl interpretiert werden.

Summary European Sand Martins arrive at their breeding sites in flocks of usually unmated and . Ringing results of a large population in NW-Germany and own observations indicate that the first flocks about a dozen individuals with an approximately balanced sex ratio appear at traditional breeding places and consist of older, experienced resident birds (presumably acquainted with one another). The birds arriving over the next several weeks are mainly first-year or non-resident individuals. The flocks arrive separately in areas with suitable sandcliffs, synchronize the pair-formation activities and avoid disturbances among paired and unpaired birds. This behaviour causes the formation of subcolonies, which are typical for all densely occupied breeding places. Each settles on a fixed area on the sandcliff (territory) in order to excavate a burrow, to sing the territory-song (fig. 2 b) and to perfor the territory-circle-flight (fig. 2 c, 4 a). Silent birds (normally ) are welcomed or tolerated by the resident . The sexes are monomorphic and therefore courtship displays of the are non-aggressive until establishment of pair-bonds. Only intruding singing or individually known neighbouring are driven away, usually at early stages of territory occupation. Ontogenetic investigations on centric diatoms IVThe planktonic diatomStephanopyxis turris — its treatment and life history
This paper presents a detailed account of the life cycle, development and cellular mechanics of the centric diatomS. turris. Special attention is paid to culture methods, nutritional requirements and the mechanism of vegetative cell enlargement. Instructions are outlined for experimental manipulations of developmental features. Various aspects of development are treated in details, e. g. cellular structures, cell division and morphogenesis, development and germination of resting spores, differentiation of gametangia (spermatogonangia, spermatogonia and oogonia), meiosis in the gametocytes, fertilization and auxospore differentiation (including the formation of the rejuvenated first cell and the accompanying metagamic mitoses).S. turris has one-egged oogonia. Its spermatogonangia develop their spermatogonia according to theBiddulphia granulata-type and their spermiums according to theMelosira-type (Fig. 18). Two new termini, i. e. heterovalvate cytokinesis and depauperizing mitosis are introduced (p. 232, p. 238). Among the more important results are observations on karyokinesis in vivo, meiosis and karyogamy, and on the peculiar process of destruction of supernumerary nuclei following each karyokinesis in the oocyte, and later in the young auxospore. Relations between osmotic cell rhythms, karyokinetic cycle and morphogenesis are discussed at the end of the paper.

A -amylase (EC 3.2.1.2) was identified in the outer pericarp (P) of developing seeds of wheat (Triticum aestivum L.) and compared with the well known -amylase which is synthesized during seed development in the starchy endosperm (E). The enzyme P already exists in the tissues before anthesis and vanishes at the time when E starts to accumulate. The isoelectric-focusing patterns of P and E are very similar. The relative molecular weight (Mr) of P is slightly higher than that of E (66 and 64.5 kDa, respectively). Both P and E exhibit common epitopes in addition to epitopes specific for each of them. The two enzymes were identified in small amounts in the green tissues of the developing seeds (inner pericarp and testa). Incubation of synthetic Man\1-4GlcNAc-OMe, GalNAc1-4GlcNAc-OMe, Glc1-4GlcNAc-OMe, and GlcNAc1-4GlcNac-OMe with CMP-Neu5Ac and rat liver Gal1-4GlcNAc (2-6)-sialyltransferase resulted in the formation of Neu5Ac2-6Man1-4GlcNAc-OMe, Neu5Ac2-6GalNAc1-4GlcNAc-OMe, Neu5Ac2-6Glc1-4GlcNAc-OMe and Neu5Ac2-6GlcNAc1-4GlcNAc-OMe, respectively. Under conditions which led to quantitative conversion of Gal1-4GlcNAc-OEt into Neu5Ac2-6Gal1-4GlcNAc-OEt, the aforementioned products were obtained in yields of 4%, 48%, 16% and 8%, respectively. A numerical measure for evidence is defined in a probabilistic framework. The established mathematical concept of information or entropy (as defined in ergodic theory) can be obtained from this definition in a special case, although in general information is greater than evidence. In another, somewhat complementary, special case a numerical measure for surprise is derived from the definition of evidence. Some applications of the new concept of evidence are discussed, concerning statistics in general and the special kind of statistics performed by neurophysiologists, when they analyze the response of neurons, and perhaps by the neurons themselves.  相似文献
