Kudoa thyrsites (Myxozoa: Multivalvulida) is a cosmopolitan marine parasite of fishes associated with post-mortem tissue degradation. Financial losses incurred as a result of these infections are of concern to commercial fisheries. There is conflicting evidence whether K. thyrsites represents a cryptic species complex. Myxospore morphology is very similar for K. thyrsites across its range, but preliminary genetic analyses show some differences. Kudoa thyrsites and the morphologically similar Kudoa histolytica were examined from hosts in British Columbia, Canada, Oregon, USA, Chile, England, South Africa, Australia, and Japan. We compared myxospore morphology and DNA sequences of heat shock protein 70 and the small subunit, large subunit, and internal transcribed spacer 1 of the ribosomal DNA. There was some morphological variation between regional representatives, inconsistent with genetic analyses. Phylogenetically, major separations correlated to four broad geographic regions: Japan, Australia, eastern Pacific, and eastern Atlantic. Within these regions there was little additional genetic structure. These data are evidence for regional subdivision of K. thyrsites suggesting global transplantation of fishes has yet to homogenize these distinctions. Within regions, parasite gene flow appears to be high between host species, suggesting little host specificity and minimal cryptic speciation. Our data also indicate that K. histolytica is not a valid species, as it was morphologically and genetically indistinguishable from K. thyrsites.  相似文献

The life cycle of the heterophyid fluke, Haplorchis pumilio is elucidated for the first time from the Indian region. Various stages in the life cycle were established based on observations made on natural infections found in snails and fish in a freshwater stream at Visakhapatnam, India and experimental infections carried out in the laboratory. The thiarid snail, Thiara tuberculata served as the first intermediate host and a wide range of freshwater fish as second intermediate hosts. Natural infections with adult flukes were found in the piscivorous birds Ardeola grayii and Bubulcus ibis. Adults were raised experimentally in day-old chicks. Distinguishing features of the cercaria of H. pumilio are: a large body size (200-224 x 92-96 micro m), body-tail ratio of 1:2.1 and densely distributed pigment granules in the parenchyma imparting a brownish tinge to the body. Natural infections with metacercariae were found in the freshwater fish Channa punctatus, C. orientalis, Puntius sophore, Gambusia affinis and fingerlings of Cyprinus carpio and Liza macrolepis. Additionally, experimental infections were established in Therapon jarbua, Esomus danricus and Oreochromis mossambica. Metacercariae were embedded in the caudal muscles of fish and heavy infections induced mortality. Metacercariae were infective at about 15 days of age.  相似文献

The life history of Echinochasmus bagulai, an echinostomatid trematode in birds, is reported and stages in the life cycle are described. Natural infections with cercariae, which are of the gymnocephalous type, were found in the thiarid snail Thiara tuberculata. Metacercarial cysts were found in the gills of Aplocheilus panchax, Oryzias melastigma, Gambusia affinis and Channa punctata. Adults were obtained in the small intestine of Ardeola grayi. In laboratory experiements development of cercariae into infective metacercariae took 10 days in the gills of A. panchax. Mature flukes were recovered in 10 days in the small intestine of experimentally infected 1-day-old leghorn chicks. The eggs collected from the faeces of these infected chicks were incubated at 39 degrees C. The free-swimming miracidia were found on the fifth day of incubation.  相似文献

A key requirement for several theories involving the evolution of sex and sexual selection is a specificity between host and parasite genotypes, i.e. the resistance of particular host genotypes to particular parasite genotypes and the infectivity of particular parasite genotypes for particular host genotypes. Determining the scope and nature of any such specificity is also of applied relevance, since any specificity for different parasite genotypes to infect particular host genotypes may affect the level of protection afforded by vaccination, the efficacy of selective breeding of livestock for parasite resistance and the long-term evolution of parasite populations in response to these control measures. Whereas we have some evidence for the role of specificity between host and pathogen genotypes in viral and bacterial infections, its role in macroparasitic infections is seldom considered. The first empirical test of this specificity for a vertebrate–nematode system is provided here using clonal lines of parasite and inbred and congenic strains of rat that differ either across the genome or only at the major histocompatibility complex. Although significant differences between the resistance of host genotypes to infection and between the fitness of different parasite genotypes are found, there is no evidence for an interaction between host and parasite genotypes. It is concluded that a specificity between host and parasite genotypes is unlikely in this system.  相似文献

The results presented in this study indicate that host choice significantly increases one of the fitness components (preadult developmental time) in females of the bean weevil (Acanthoscelides obtectus). The host choice also affects the net replacement rate (Ro), as well as the intrinsic rate of increase (r). Using the method developed by Lande and Arnold (1983), we also measured the intensity of nonrandom association between host choice phenotypes and preadult developmental time.  相似文献

The majority of Kudoa species infect the somatic muscle of fish establishing cysts. As there is no effective method to detect infected fish without destroying them, these parasited fish reach the consumer. We have developed this work to determine whether this parasite contains antigenic compounds capable of provoking an immune response in laboratory animals, in order to consider the possible immunopathological effects in man by the ingestion of Kudoa infected fish. BALB/c mice were injected by the subcutaneous route with the following extracts suspended in aluminium hydroxide: Group 1 (black Kudoa sp. pseudocyst extract), group 2 (white Kudoa sp. pseudocyst extract). Specific IgE levels were measured by ELISA. IgE detected in both groups 1 and 2 showed the possible allergenic nature of some of the components of the parasitic extracts.  相似文献

The life cycle of Brachylaima ruminae n.sp. (Trematoda: Brachylaimidae), a duodenal parasite of rodents on the Mediterranean island of Formentera (Spain) is elucidated. The new species follows a terrestrial triheteroxenous life cycle. Eggs passed in the faeces of the definitive host must be ingested by a specific first intermediate host, the land snail Rumina decollata. Branched cercariogenous sporocysts develop in the digestive gland. Microcercous cercariae come out through the terminal birth pores of the branches. Cercariae shed by the snail are terrestrial, crawling on humid substratum. They contact the second intermediate host, another land snail, principally the species R. decollata and less frequently slugs and Helicids. Cercariae enter via the excretory pore and kidney duct to their specific final location, the kidney. Unencysted metacercariae develop in the kidney (also, less frequently, in the pedal glands) to the mature, infective stage. Infective metacercariae infest the definitive host when ingested together with the snail.  相似文献

In small forest streams of West Nigeria the snail species Bulinus globosus was found to be the first intermediate host of a clinostomatid species which proved in successful infection experiments with the cichlid fish Tilapia zillii as second intermediate host to be Euclinostomum heterostomum (Rudolphi, 1809). Final hosts are herons. The morphology of the redia and the morphology and behaviour of the cercaria are described for the first time. The metacercaria which settles dorsal to the swim-bladder or in the kidney of the host fish requires more than two months (at tropical temperatures) for full development. Histological sections of the metacercaria revealed that its pharynx is provided with only few muscle fibres but surrounded by many myoblast-like cells, and the fact that the interior surface of this organ has a dense brush-like lining consisting of long microtriches. The pathology of infected fish is briefly described, and remarks about the geographical distribution of the parasite are given.  相似文献

The IgG1 was the predominant subclass. IgE detected in groups 1 and 2 showed the possible allergenic nature of some of the components of the parasitic extract. High IgA levels and medium IgG2a and IgG3 levels were obtained in groups 1 and 2. Low IgG2b responses were shown. No cross-reactions between Kudoa sp. pseudocyst extracts and the non-infected hake meat extract were observed.  相似文献

Gilt-head sea bream, Sparus aurata L., the Mediterranean's most important mariculture species, has been cultured for the last 30 yr in Eilat (Israeli Red Sea). Kudoa sp. was the first myxosporean parasite reported from this species. In recent years, an increase in prevalence in both land-based and sea-cage facilities in Eilat has been observed. Infections with the same Kudoa species appeared in cultured European sea bass Dicentrarchus labrax (L.) and grey mullet Mugil cephalus in the same farms, as well as in 10 species of wild Red Sea reef fish, indicating that Kudoa sp. is not fastidious with regard to its host. All affected species displayed 1- to 2-mm (up to 5 mm) whitish, spherical, or oval polysporous plasmodia. The parasite established multiple site infections, most commonly in the muscles and intracranial adipose tissue of the brain and eye periphery. Other sites were subcutaneous adipose tissue, nerve axons, mouth, eye, mesenteries, peritoneum, swim bladder, intestinal musculature, heart, pericardium, kidney, and ovary. On the basis of spore morphology, the parasite was identified as Kudoa iwatai Egusa and Shiomitsu, 1983. Ultrastructural features were comparable to those of previously studied Kudoa species. The 18S rDNA from 7 Red Sea isolates was sequenced and compared with the sequence of the same gene from K. iwatai isolated from cultured red sea bream, Pagrus major, in Japan. The phylogenetic position of K. iwatai within the genus was determined using sequence analysis of all related taxa available in GenBank. The 3 isolates of K. iwatai clustered together on a newly formed, highly supported clade. The Red Sea strain of K. iwatai is apparently native to the region. In the absence of records of this Kudoa sp. from the extensive Mediterranean sea bream and sea bass production industries, introduction with its Mediterranean hosts seems unlikely. Therefore, we conclude that K. iwatai is an Indo-Pacific species that, in the Red Sea, has extended its host range to include the allochthonous gilt-head sea bream, European sea bass, and grey mullet.  相似文献

A new species of Kudoa lutjanus n. sp. (Myxosporea) is described from the brain and internal organs of cultured red snapper Lutjanus erythropterus from Taiwan. The fish, 260 to 390 g in weight, exhibited anorexia and poor appetite and swam in the surface water during outbreaks. Cumulative mortality was about 1% during a period of 3 wk. The red snapper exhibited numerous creamy-white pseudocysts, 0.003 to 0.65 cm (n = 100) in diameter, in the eye, swim bladder, muscle and other internal organs, but especially in the brain. The number of pseudocysts per infected fish was not correlated with fish size or condition. Mature spores were quadrate in apical view and suboval in side view, measuring 8.2 +/- 0.59 microm in width and 7.3 +/- 0.53 microm in length. The 4 valves were equal in size, each with 1 polar capsule. Polar capsules were pyriform in shape, measuring 3.62 +/- 0.49 microm in length and 2.2 +/- 0.49 microm in width. Mild inflammatory responses or liquefaction of host tissue were associated with K. lutjanus n. sp. infection. The junction of shell valves appeared as overlapping, straight lines. The polar filament formed 2 to 3 coils. A general PCR (polymerase chain reaction) primer for Kudoa amplified the small subunit (SSU) rDNA sequences, and the amplified gene was sequenced. It was evident from the phylogenetic tree that the 3 strains tested, AOD93020M, AOD93028M and AOD93028B, were identical and belonged to the Kudoa SS rRNA subgroup. The evolutionary tree showed that these strains form a unique clade, at a distance from other Kudoa species and myxosporeans. The spore's morphological and ultrastructural characteristics, as well as the SS rDNA properties of the isolates, were also essentially identical and served to distinguish them from representative Kudoa. It is, therefore, proposed that the strains isolated from the diseased red snapper be assigned to a new species.  相似文献

The prevalence and intensity of experimental infections of Aedes aegypti with the digenean Plagiorchis noblei increased significantly with the level of trickle exposure to cercariae. Daily exposure to doses of 16 cercariae/day yielded a mean infection intensity of 13.0 metacercariae; doses of 1 cercaria/day resulted in only 2.4 metacercariae per infected mosquito larva. The prevalence of infection rose from 46% at an exposure of 1 cercaria/day to 99% at 16 cercariae/day. Host mortality rose concomitantly from 25% to 88%. Host mortality and parasite acquisition were independent of environmental temperatures (21-29 C), despite the fact that developmental times, and consequently the number of daily exposures, were more than 50% greater at the low end of the temperature scale. This may be attributable to low activity of mosquito larvae and the resulting decrease in the number of encounters with cercariae.  相似文献

We describe Kudoa ovivora n. sp. from ovaries of bluehead wrasse, Thalassoma bifasciatum, and record its presence in 6 species (Labroidei) collected in the San Blas Islands. Panama. Kudoa ovivora spores are quadrate with rounded edges in apical view, oval-shaped with apical valve extensions in side view (mean spore dimensions: length 6.5 microns, width 7.7 microns, thickness 6.9 microns; mean polar capsule dimensions: length 2.1 microns, width 1.5 microns). This is the first Kudoa species from gonads of fishes. Prevalence of infection varied among labrids (Thalassoma bifasciatum, Halichoeres bivittatus, Halichoeres garnoti, Halichoeres poevi), with T. bifasciatum exhibiting the greatest prevalence. Density of infection, measured as percent infected eggs, also varied among species with highest densities occurring in H. garnoti. Kudoa ovivora may not require an intermediate host because fishes fed infected tissue developed more infections than unfed fish. Infected eggs are inviable and larger and heavier than uninfected eggs. Infected eggs contain more organic and inorganic material, indicating that K. ovivora increases resource allocation to eggs. Therefore, infected females may have reduced growth, fecundity, and/or spawning activity. Because males were uninfected and all identified hosts are protogynous sequential hermaphrodites, further studies of K. ovivora may provide new insights on the costs/benefits of sex change.  相似文献
