Extremes of temperature (both heat and chilling) during early inbibitional phase of germination caused disruption of redox-homeostasis by increasing accumulation of reactive oxygen species (superoxide and hydrogen peroxide) and significant reduction of antioxidative defense (assessed in terms of total thiol content and activities of superoxide dismutase, catalase, ascorbate peroxidase and glutathione reductase) in germinating tissues of rice (Oryza sativa L., cultivar Ratna). Imbibitional heat and chilling stress also induced oxidative damage to newly assembled membrane system by aggravating membrane lipid peroxidation and protein oxidation [measured in terms of thiobarbituric acid reactive substances (TBARS), free carbonyl content (C = O groups) and membrane protein thiol level (MPTL)]. Treatment with standardized low titer hydrogen peroxide during early imbibitional phase of germination caused significant reversal in oxidative damages to the newly assembled membrane system imposed by heat and chilling stress [evident from the data of TBARS, C = O, MPTL, ROS accumulation, membrane permeability status, membrane injury index and oxidative stress index] in seedlings of experimental rice cultivar. Imbibitional H2O2 pretreatment also caused up-regulation of antioxidative defense (activities of superoxide dismutase, catalase, ascorbate peroxidase, glutathione reductase and total thiol content) in the heat and chilling stress-raised rice seedlings. When the parameters of early growth performances were assessed (in terms of relative growth index, biomass accumulation, relative germination performance, mean daily germination, T50 value), it clearly exhibited significant improvement of early growth performances of the experimental rice cultivar. The result proposes that an ‘inductive pulse’ of H2O2 is required to switch on some stress acclimatory metabolism through which plant restores redox homeostasis and prevents or repairs oxidative damages to newly assembled membrane system caused by unfavorable environmental cues during early germination to the rice cultivar Ratna. The importance of mitigating oxidative damages to membrane lipid and protein necessary for post-germinative growth under extremes of temperature is also suggested.  相似文献

Characteristics of chlorophyll fluorescence and antioxidative system were investigated in rice (Oryza sativa L.) super-hybrid Liangyoupeijiu (LYPJ), maternal cultivar Peiai64s, and paternal cultivar indica rice 9311 under chilling stress. During 6-d chilling treatment, chlorophyll content of all three genotypes was gradually declined. However, the decrease in photosystem 2 (PS 2) maximum photochemical efficiency (Fv/Fm) and quantum yield of PS 2 (ΦPS2) was less expressive in LYPJ than in parental cultivars The electrolyte leakage and malondialdehyde content in all cultivars increased after chilling treatment, but LYPJ exhibited the least increasing tendency. Activities of superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione reductase (GR) were higher in LYPJ than in parental cultivars. The results demonstrated that tolerance to chilling stress in LYPJ might be adopted mostly from its maternal cultivar.  相似文献

With the imposition of salt stress (0.5 to 3 % NaCl or CaCl2) a decrease in germination rate and accumulation of proline was observed in the root tissue. Both NaCl and CaCl2 solutions induced an increase in the total peroxide content and lipid peroxidation and decrease in catalase, guaiacol peroxidase and superoxide dismutase activities in root tissues suggesting an oxidative stress in the salt sensitive rice cultivar.  相似文献

Germination of two rice (Oryza sativa L.) cultivars (Ratna and IR 36) in the presence of 10 μM PbCl2 and 10 μM HgCl2 decreased germination percentage, germination index, shoot/root length, tolerance index and dry mass of shoots and roots. Mercury was more toxic than lead. Reduced glutathione (GSH), cysteine (Cys), ascorbic acid, and α-tocopherol alleviated the adverse effects of these metals on plants in the order GSH > Cys > ascorbic acid > α-tocopherol. The effects were more pronounced in tolerant cultivar IR 36 than in the relatively susceptible cultivar Ratna. The present work makes an effort to assess and standardize some redox metabolic and molecular parameters for screening drought tolerant indigenous aromatic rice cultivars of West Bengal, India. PEG-induced dehydration stress during early germination caused disruption of redox-homeostasis and oxidative damage in four IARVs (Jamainadu, Tulaipanji, Sitabhog and Badshabhog) by enhancing the accumulation of pro-oxidants [assessed in terms of oxidation of 2′,7′-dichlorofluorescindiacetate (DCFDA), accumulation of \({\text{O}}_{2}^{ \cdot - }\) and H2O2 and in situ staining of reactive oxygen species (ROS) in germinating tissue], significant reduction of antioxidative defence (total antioxidant and radical scavenging capacity, total thiol content and activities of antioxidative defence enzymes) and aggravating protein oxidation and lipid peroxidation (assessed in terms of free carbonyl content and accumulation of thiobarbituric acid reactive substances). When compared between the indigenous aromatic rice cultivars, a clear trend in differential redox regulatory properties in which ROS-antioxidant interaction acts at metabolic interface for redox homeostasis was observed in the order Badshabhog > Tulaipanji > Sitabhog > Jamainadu. Moreover, when the efficacy of ascorbate–glutathione cycle for scavenging H2O2 generated during dehydration stress was assessed and compared between the landraces exposed to PEG-induced dehydration stress in germinating tissue, it also exhibited almost the same trend with the landrace Tulaipanji and Badsabhog exhibiting maximum and Jamainadu the minimum efficiencies of the redox cycle. The indigenous aromatic rice cultivars Tulaipanji and Badsabhog resist dehydration stress better than the other two landraces due to its early preparedness to combat oxidative stress by up-regulating expression of genes of some enzymes of ascorbate–glutathione cycle along with some other antioxidative enzymes. A model of redox homeostasis in which ROS-antioxidant (ascorbate–glutathione system) acts at metabolic interface for up-regulation of antioxidative gene expression necessary for differential drought stress tolerance among the indigenous aromatic rice varieties is suggested.  相似文献

Water deficit is an important constraint to rice (Oryza sativa L.) productivity. The present study was undertaken to investigate whether the level of oxidative stress, carbonylation of proteins, proteolysis and status of antioxidative defense could serve as a model to distinguish water deficit tolerant and sensitive rice cultivars. When 10-day-grown seedlings of two rice cultivars, Malviya-36 (drought-sensitive) and Brown Gora (drought-tolerant) were subjected to ?1.0 and ?2.1 MPa water deficit treatments for 24–72 h with polyethylene glycol 6000 in the medium, a greater decline in the growth of the seedlings and levels of leaf water potential, relative water content, Chl a, Chl b, carotenoids and greater increase in leaf water loss were observed in the sensitive cultivar than the tolerant. Under similar level of water deficit seedlings of sensitive cultivar showed higher level of superoxide anion generation, H2O2, lipid peroxidation and proteolysis in roots as well as shoots compared to the tolerant. Drought-tolerant cultivar had higher constitutive level of antioxidative enzymes superoxide dismutase and catalase and the activities of these two enzymes alongwith of guaiacol peroxidase showed greater increase in this cultivar under water deficit compared to the sensitive. A significant decline in the level of protein thiol and a higher increase in protein carbonyls content, also confirmed by protein gel blot analysis with an antibody against 2,4-dinitrophenylhydrazine was observed in the seedlings of drought sensitive cv. Malviya-36 compared to the tolerant cv. Brown Gora when subjected to similar level of water deficit. Seedlings of drought sensitive cultivar, under water deficit, showed higher proteolytic activity, higher number of in-gel activity stained proteolytic bands and higher expression of oxidized proteins in roots compared to the tolerant cultivar. Results suggest that poor capacity of antioxidative enzymes could be, at least partly, correlated with water deficit sensitivity of sensitive cultivar and that higher activity of antioxidative enzymes superoxide dismutase, catalase, guaiacol peroxidase, low proteolytic activity, lower level of protein carbonyls and protein thiolation could serve as a model to depict water deficit tolerance in Indica rice seedlings.  相似文献

研究了外源GSH对盐胁迫下耐盐性不同的水稻品种Pokkali(耐盐)和Peta(盐敏感)叶绿体中抗氧化酶活性和抗氧化剂含量的影响.结果表明:盐胁迫下,外源GSH可以提高水稻叶绿体中活性氧清除系统中SOD、APX、GR的活性以及AsA、GSH的含量,降低叶绿体中H2O2和MDA的含量,从而降低了叶绿体膜脂过氧化的水平,缓解盐胁迫对叶绿体膜的伤害.外源GSH对盐胁迫下盐敏感品种Peta叶绿体中上述指标增加或减少的幅度大于耐盐品种Pokkali.  相似文献

Indices of oxidative stress viz., superoxide radical and H2O2 content increased in leaves of all the cultivars with the rise in salinity level, the increase was more pronounced and significant in salt-sensitive varieties and non-significant in resistant cultivars. Except for glutathione reductase (GR), basal activities of all other antioxidative enzymes viz. superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), ascorbate peroxidase (APX) and glutathione reductase (GR) were significantly higher in leaves of all the resistant cultivars as compared to the sensitive ones. A differential response of salinity was observed on various enzymatic and non-enzymatic components of antioxidant system in leaves of salt-tolerant and salt-sensitive cultivars of rice (Oryza sativa L.). Activities of superoxide dismutase and glutathione reductase enhanced in all the tolerant cultivar while declined in the sensitive cultivars with increasing salinity from 0 to 100 mM. Salt-stress induced the activities of catalase and peroxidase in all the cultivars but the magnitude of increase was more pronounced in the sensitive cultivars than in the tolerant cultivars. Contrarily, APX activity increased in the salt-sensitive cultivars but showed no significant change in the salt-tolerant cultivars. The amount of ascorbic acid content, reduced glutathione (GSH), reduced/oxidized glutathione (GSSG) ratio was higher in leaves of the tolerant cultivars than that of the sensitive cultivars under saline conditions. It is inferred that leaves of salt-tolerant cultivars tend to attain greater capacity to perform reactions of antioxidative pathway under saline conditions to combat salinity-induced oxidative stress.  相似文献

Crop yield is severely affected by soil salinity, as salt levels that are harmful to plant growth occur in large terrestrial areas of the world. The present investigation describes the studies of enzymatic activities, in-gel assays, gene expression of some of the major antioxidative enzymes, tocopherol accumulation, lipid peroxidation, ascorbate and dehydroascorbate contents in a salt-sensitive rice genotype PB1, and a relatively salt-tolerant cultivar CSR10 in response to 200 mM NaCl. Salt solution was added to the roots of hydroponically grown 5-day-old etiolated rice seedlings, 12 h prior to transfer to cool white fluorescent?+?incandescent light (100 μmol photons m?2 s?1). Total tocopherol and ascorbate contents declined in salt-stressed rice seedlings. Among antioxidative enzymes, an increase in the activities of superoxide dismutase (EC 1.15.1.1), catalase (EC 1.11.1.6), ascorbate peroxidase (EC 1.11.1.11), glutathione reductase (EC 1.6.4.2), and their gene expression was observed in both cultivars in response to salt stress. The salt-tolerant cultivar CSR10 resisted stress due to its early preparedness to combat oxidative stress via upregulation of gene expression and enzymatic activities of antioxidative enzymes and a higher redox status of the antioxidant ascorbate even in a non-stressed environment.  相似文献

The present investigation evaluated the ability of an antioxidative defense system in terms of the tolerance against salinity-induced oxidative stress and also explored a possible relationship between the status of the components of an antioxidative defense system and the salt tolerance in Indica rice (Oryza sativa L.) genotypes. When the seedlings of a salt-sensitive cultivar was grown in sand cultures containing different NaCl concentrations (7 and 14 dS m?1) for 5–20 days, a substantial increase was observed in the rate of superoxide anion (O 2 ·? ) production, elevated levels of H2O2 and thiobarbituric acid reactive substances (TBARS) which indicated an enhancement in lipid peroxidation. A declination in the level of thiol clearly indicated an increase in the protein oxidation as well as a decline in the reduced forms of ascorbate (AsA) and glutathione (GSH) and the ratios of their reduced to oxidized forms occurred in the salt-sensitive seedlings. Similar treatment caused a very little alteration or no change in the levels of these components in the seedlings of salt-tolerant cultivar. The activity of antioxidative enzymes superoxide dismutase (SOD), its isoform Cu/Zn-SOD and ascorbate peroxidase (APX) increased in both the cultivars against salinity. In salt-sensitive seedlings, the activity of the various enzymes, guaiacol peroxidase (GPX), catalase (CAT), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), and glutathione reductase (GR) increased at moderate salinity treatment of 7 dS m?1 NaCl while the activities of these enzymes declined with higher salinity level of 14 dS m?1 NaCl. However, a consistent increase was observed in the activities of these enzymes of salt-tolerant seedlings with an increase in the duration and the level of the salinity treatment. The results suggest that a higher status of antioxidants (AsA and GSH) and a coordinated higher activity of the enzymes (SOD, CAT, GPX, APX, and GR) can serve as the major determinants in the model for depicting salt tolerance in Indica rice seedlings.  相似文献

Salinity stress is a major limiting factor in cereal productivity. Many studies report improvements in salt tolerance using model plants, such as Arabidopsis thaliana or standard varieties of rice, e.g., the japonica rice cultivar Nipponbare. However, there are few reports on the enhancement of salt tolerance in local rice cultivars. In this work, we used the indica rice (Oryza sativa) cultivar BR5, which is a local cultivar in Bangladesh. To improve salt tolerance in BR5, we introduced the Escherichia coli catalase gene, katE. We integrated the katE gene into BR5 plants using an Agrobacterium tumefaciens-mediated method. The introduced katE gene was actively expressed in the transgenic BR5 rice plants, and catalase activity in T1 and T2 transgenic rice was approximately 150% higher than in nontransgenic plants. Under NaCl stress conditions, the transgenic rice plants exhibited high tolerance compared with nontransgenic rice plants. T2 transgenic plants survived in a 200 mM NaCl solution for 2 weeks, whereas nontransgenic plants were scorched after 4 days soaking in the same NaCl solution. Our results indicate that the katE gene can confer salt tolerance to BR5 rice plants. Enhancement of salt tolerance in a local rice cultivar, such as BR5, will provide a powerful and useful tool for overcoming food shortage problems.  相似文献

The responses of antioxidative system and photosystem II photochemistry of rice (Oryza sativa L.) to paraquat induced oxidative stress were investigated in a chilling-tolerant cultivar Xiangnuo no. 1, and a chilling-susceptible cultivar, IR-50. Electrolyte leakage and malondialdehyde (MDA) content of Xiangnuo no. 1 were little affected by paraquat, but they increased in IR-50. After paraquat treatment, superoxide dismutase (SOD) activity remained high in Xiangnuo no. 1, while it declined in IR-50. Activities of catalase (CAT), ascorbate peroxidase (APX) and glutathione reductase (GR) declined with oxidative stress in both cultivars, but Xiangnuo no. 1 had higher GR activity than IR-50. Under paraquat induced oxidative stress, ascorbic acid (AsA) and reduced glutathione (GSH) concentrations remained high in Xiangnuo no. 1, but decreased in IR-50. The results indicated that higher activities of SOD and GR and higher contents of AsA and GSH in Xiangnuo no. 1 under paraquat induced oxidative stress were associated with its tolerance to paraquat, while paraquat induced damage to IR-50 was related to decreased activities of SOD, APX and GR and contents of AsA and GSH. F v/F m, Φ PSII, and qP remained high in Xiangnuo no. 1, while they decreased greatly in IR-50 under paraquat induced oxidative stress.  相似文献

为了解耐盐水稻HD96-1幼苗期耐盐生理调控特性,该研究以籼型水稻HD96-1(耐盐性强)和93-11(耐盐性弱)为材料,采用营养液水培法,设置3种NaCl盐浓度(0、60、120 mmol·L-1),对3叶期幼苗进行了7 d盐处理,测定和分析了两个材料的生长参数和生理生化指标。结果表明:(1)在盐胁迫下,水稻幼苗均表现为株高和假茎宽减小,根冠比增加;与93-11比,HD96-1株高和茎宽减小幅度低,根冠比增加幅度高;地上部和根系干重,HD96-1增加,而93-11减少。(2)盐胁迫后,水稻幼苗的丙二醛(MDA)、超氧阴离子(O2-)和过氧化氢(H2O2)含量均上升,但HD96-1增幅较93-11低。(3)在盐胁迫下,水稻幼苗体内的超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)的活性,以及抗坏血酸(AsA)、谷胱甘肽(GSH)、脯氨酸、可溶性糖和可溶性蛋白的含量均升高,HD96-1增幅高于93-11。综上表明,两个水稻材料幼苗...  相似文献

两个不同抗旱性的水稻品种对PEG6000渗透胁迫(-0.5MPa,-0.8MPa)的反应具有一定差异。渗透胁迫下SOD,POD和CAT活性及Vc,Car含量与膜脂过氧化水平及膜透性呈一定负相关性,表明这些指标可作为水稻抗旱育种的参考依据。  相似文献

To identify biochemical markers for salt tolerance, two contrasting cultivars of rice (Oryza sativa L.) differing in salt tolerance were analyzed for various parameters. Pokkali, a salt-tolerant cultivar, showed considerably lower level of H2O2 as compared to IR64, a sensitive cultivar, and such a physiology may be ascribed to the higher activity of enzymes in Pokkali, which either directly or indirectly are involved in the detoxification of H2O2. Enzyme activities and the isoenzyme pattern of antioxidant enzymes also showed higher activity of different types and forms in Pokkali as compared to IR64, suggesting that Pokkali possesses a more efficient antioxidant defense system to cope up with salt-induced oxidative stress. Further, Pokkali exhibited a higher GSH/GSSG ratio along with a higher ratio of reduced ascorbate/oxidized ascorbate as compared to IR64 under NaCl stress. In addition, the activity of methylglyoxal detoxification system (glyoxalase I and II) was significantly higher in Pokkali as compared to IR64. As reduced glutathione is involved in the ascorbate–glutathione pathway as well as in the methylglyoxal detoxification pathway, it may be a point of interaction between these two. Our results suggest that both ascorbate and glutathione homeostasis, modulated also via glyoxalase enzymes, can be considered as biomarkers for salt tolerance in Pokkali rice. In addition, status of reactive oxygen species and oxidative DNA damage can serve as a quick and sensitive biomarker for screening against salt and other abiotic stresses in crop plants.  相似文献

The changes in the activity of antioxidant enzymes such as superoxide dismutase (SOD: EC 1.15.1.1), catalase (CAT: EC 1.11.1.6), peroxidase (POX: EC 1.11.1.7), ascorbate peroxidase (APOX: EC 1.11.1.11) and glutathione reductase (GR: EC 1.6.4.2), free proline content, and the rate of lipid peroxidation level in terms of malondialdehyde (MDA) in roots of two rice cultivars (cvs.) differing in salt tolerance were investigated. Plants were subjected to three salt treatments, 0, 60, and 120 mol m−3 NaCl for 7 days. The results showed that activated oxygen species may play a role in cellular toxicity of NaCl and indicated differences in activation of antioxidant defense systems between the two cvs. The roots of both cultivars showed a decrease in GR activity with increase in salinity. CAT and APOX activities increased with increasing salt stress in roots of salt-tolerant cultivar Pokkali but decreased and showed no change, respectively, in roots of IR-28 cultivar. POX activity decreased with increasing NaCl concentrations in salt-tolerant Pokkali but increased in IR-28. SOD activity showed no change in roots of both cultivars under increasing salinity. MDA level in the roots increased under salt stress in sensitive IR-28 but showed no change in Pokkali. IR-28 produced higher amount of proline under salt stress than in Pokkali. Increasing NaCl concentration caused a reduction in root fresh weight of Pokkali and root dry weight of IR-28. The results indicate that improved tolerance to salt stress in root tissues of rice plants may be accomplished by increased capacity of antioxidative system.  相似文献

Deterioration of membranes caused by lipoxygenase (LOX) activity under 10 μM PbCl2 or 10 μM HgCl2 was partially alleviated by the exogenous application of 100 μM salicylic acid (SA). In two cultivars of rice (Oryza sativa L. cvs. Ratna and IR 36), the presence of SA ameliorated the increased leakage of electrolytes, injury index, and the content of malondialdehyde caused by these heavy metals. Lead decreased H2O2 content whereas Hg increased it in both cultivars. Application of SA increased H2O2 in presence of Pb, while decreased it in presence of Hg. Both Pb and Hg decreased superoxide dismutase activity, while increased peroxidase activity. The activity of catalase was decreased by Hg but increased by Pb and SA reversed their effects. Effects of nitrogen (N)-deficiency on midday photoinhibition in flag leaves were compared between two contrastive Japanese rice cultivars, a traditional japonica cultivar with low yield, cv. Shirobeniya (SRB), and a japonica-indica intermediate type with high yield, cv. Akenohoshi (AKN). Both cultivars were grown under high-N and low-N conditions. At midday, low-N supply resulted in more intensive reductions in net photosynthetic rate, stomatal conductance, maximal quantum yield of photosystem II (PSII) and quantum yield of PSII electron transport in SRB than in AKN, indicating that SRB was more strongly photoinhibited than AKN under low-N condition. At midday, the low-N plants of two cultivars showed higher superoxide dismutase (SOD) activities than the high-N plants. However, ascorbate peroxidase (APX) activity was maintained in AKN but significantly decreased in SRB under low-N condition (N-deficiency). In contrast, hydrogen peroxide (H2O2) content in SRB significantly increased under low-N condition, indicating that the susceptibility to midday photoinhibition in the low-N plants of SRB is related to the increased H2O2 accumulation. It is suggested that the midday depression in photosynthesis may be a result of oxidative stress occurring in the low-N plants in which antioxidant capacity is not enough to cope with the generation of H2O2. Therefore, H2O2-scavenging capacity could be an important factor in determining the cultivar difference of midday photoinhibition in flag leaves of rice under low-N condition.  相似文献

Changes in the starch and sucrose contents, and the sucrose phosphate synthase, acid invertase, and starch phosphorylase activities were studied in the seedlings of salt sensitive and salt tolerant rice cultivars growing under two NaCl concentrations (7 and 14 dS m-1) for 20 d. Under salinity, the starch content in roots declined more in salt sensitive cvs. Ratna and Jaya than in salt tolerant cvs. CSR-1 and CSR-3 and was unchanged in shoots. The contents of reducing and non-reducing sugars, and the activity of sucrose phosphate synthase was increased more in the sensitive than in the tolerant cultivars. Acid invertase activity decreased in shoots of the salt tolerant cultivars, whereas increased in salt sensitive cultivars. Starch phosphorylase activity decreased in all cultivars. We studied the effect of water stress imposed at anthesis and pre-anthesis stages on oxidative stress and antioxidant activity in four wheat cultivars, two hexaploid Triticum aestivum cultivars, drought resistant cv. C 306 and drought susceptible cv. Hira, and two tetraploid cultivars, T. durum cv. A 9-30-1 and T. dicoccum cv. HW 24. Water stress decreased relative water content (RWC), membrane stability index (MSI), and increased H2O2 and malondialdehyde (MDA) contents as well as activity of superoxide dismutase (SOD), catalase (Cat) and peroxidase (POX) in all the genotypes at all the stages. Both the tetraploid cultivars showed higher RWC, MSI and SOD activity, and lower H2O2 and MDA contents under water stress than hexaploid ones. Cat and POX activities were highest in C 306.  相似文献
