Xylulose-1,5-bisphosphate in preparations of ribulose-1,5-bisphosphate (ribulose-P2) arises from non-enzymic epimerization and inhibits the enzyme. Another inhibitor, a diketo degradation product from ribulose-P2, is also present. Both compounds simulate the substrate inhibition of ribulose-P2 carboxylase/oxygenase previously reported for ribulose-P2. Freshly prepared ribulose-P2 had little inhibitory activity. The instability of ribulose-P2 may be one reason for a high level of ribulose-P2 carboxylase in chloroplasts where the molarity of active sites exceeds that of ribulose-P2. Because the KD of the enzyme/substrate complex is ≤1 μM, all ribulose-P2 generated in situ may be stored as this complex to prevent decomposition.  相似文献

The temperature dependence of C3 photosynthesis is known to vary with growth environment and with species. In an attempt to quantify this variability, a commonly used biochemically based photosynthesis model was parameterized from 19 gas exchange studies on tree and crop species. The parameter values obtained described the shape and amplitude of the temperature responses of the maximum rate of Rubisco activity (Vcmax) and the potential rate of electron transport (Jmax). Original data sets were used for this review, as it is shown that derived values of Vcmax and its temperature response depend strongly on assumptions made in derivation. Values of Jmax and Vcmax at 25 °C varied considerably among species but were strongly correlated, with an average Jmax : Vcmax ratio of 1·67. Two species grown in cold climates, however, had lower ratios. In all studies, the Jmax : Vcmax ratio declined strongly with measurement temperature. The relative temperature responses of Jmax and Vcmax were relatively constant among tree species. Activation energies averaged 50 kJ mol?1 for Jmax and 65 kJ mol?1 for Vcmax, and for most species temperature optima averaged 33 °C for Jmax and 40 °C for Vcmax. However, the cold climate tree species had low temperature optima for both Jmax(19 °C) and Vcmax (29 °C), suggesting acclimation of both processes to growth temperature. Crop species had somewhat different temperature responses, with higher activation energies for both Jmax and Vcmax, implying narrower peaks in the temperature response for these species. The results thus suggest that both growth environment and plant type can influence the photosynthetic response to temperature. Based on these results, several suggestions are made to improve modelling of temperature responses.  相似文献

Ribulose-1,5-bisphosphate car?ylase (E.C. 4.1.1.39) from 2 blue-green algae, Plectonema boryanum and Anabaena variabilis, was isolated by sucrose density gradient centrifugation. Both enzymes had a sedimentation value of about 18s, similar to that of Chromatium enzyme. The presence of two subunits (A, B) in the algal enzyme was demonstrated by Nadodecyl sulfate polyacrylamide gel electrophoresis. The molecular weight of the two subunits was determined: for Plectonema A, 5.4 × 104 and B, 1.3 × 104 and Anabaena A, 5.2 × 104 and B, 1.3 × 104, respectively. The car?ylase reaction catalysed by the algal enzyme was similar to the higher plant enzyme in exhibiting the Mg2+-effect, the optimal pH shifting from alkaline to neutral by elevating the concentration of Mg2+ in the assay mixture. The rabbit antisera developed against the spinach ribulose-1,5-bisphosphate car?ylase and its catalytic oligomer exhibited significant inhibitory effects on the car?ylation reaction catalysed by the algal enzyme.  相似文献

2-carboxy-D-arabinitol-1-phosphate (CA1P) bound to Rubisco either in leaf extracts or after purification can be displaced by SO42? ions. Thus, treatment of leaf extracts with a buffer containing 200 mol m?3 SO42? displaces any bound CA1P and enables measurement of maximum car-boxylation potential. In tobacco leaves, the activity following treatment with SO4?2 ions (‘maximal activity’) is greater than the total Rubisco activity. The ratio of the two activities altered in a dynamic way with fluctuations in irradiance. Even in species which do not produce significant amounts of CA1P, the maximal activity greatly exceeded the total activity. Anion exchange separation of components in acid extracts confirmed the absence of CA1P in tobacco leaves harvested above an irradiance of 300 μmol quanta m?2 s?1, but the presence of another inhibitor of Rubisco. These results are consistent with the regulation of Rubisco activity by inhibitors other than CA1P which, like CA1P, can be displaced by SO42? ions.  相似文献

Responses of plant processes to temperature may vary according to the time scale on which they are measured. In this study, both short‐term and seasonal responses of photosynthesis to temperature were examined. A field study of seasonal changes in the temperature response of photosynthesis was conducted on two provenances, French and Moroccan, of mature maritime pine (Pinus pinaster Ait.). Measurements were made every 2 months over a 1‐year period and used to parameterize a mechanistic model of photosynthesis. Temperature responses of maximum Rubisco activity, Vcmax, and potential electron transport rate, Jmax, were obtained for each measurement period, as was the response of stomatal conductance, gs, to water vapour pressure deficit (VPD). Absolute values of Vcmax and Jmax at 25 °C were related to needle nitrogen content, Narea.Narea, and thus Vcmax and Jmax, were negatively correlated with the mean minimum temperature in the month preceding measurements. The ratio of Jmax : Vcmax at 25 °C varied between 1 and 1·7 but did not show any seasonal trend. Nor was there any seasonal trend in the relative temperature response of Vcmax, which had an activation energy Ha of approximately 57 kJ mol?1 throughout the experiment. The activation energy of Jmax was also close to constant throughout the experiment, averaging 39 kJ mol?1. For the French provenance, the optimal temperature of Jmax was positively correlated with the maximum temperature of the previous day, but no such correlation was found for the Moroccan provenance. The response of gs to VPD also varied seasonally, with much stronger stomatal closure in winter months. Taken together, these results implied a translational shift downwards of the photosynthetic temperature response curve with increasing Tprev, and a shift in the temperature optimum of photosynthesis of 5–10 °C between summer and winter. These results illustrate that the short‐term temperature response of photosynthesis varies significantly on a seasonal basis.  相似文献

Mutagenesis in vitro of the gene encoding the large subunit of ribulose-1,5-bisphosphate carboxylase/ oxygenase (EC 4.1.1.39) from Anacystis nidulans was used to generate novel enzymes. Two conserved residues, threonine 4 and lysine 11 in the N-terminus were changed. The substitution of threonine 4 with serine or valine had little effect on the kinetic parameters. The substitution of lysine 11 with leucine, which is non-polar, increased the K m for ribulose-1,5-bisphosphate from 82 to 190 M but its replacement with glutamine, which has polar properties, had no appreciable effect.Abbreviations Rubisco ribulose-1,5-bisphosphate carboxylase/oxygenase - RuBP ribulose-1,5-bisphosphate - LSU large sub-unit of Rubisco - SSU small subunit of Rubisco We thank Dr. S. Uniculm wheat (Triticum aestivum L.) was grown to maturity at four concentrations of nitrogen corresponding to 3 (N1), 6 (N2), 9 (N3) and 12 (N4) g m–2. Penultimate and flag leaves were examined throughout the ontogeny. Sub-optimal concentrations of N resulted in sharp decline in both area and dry mass of the leaves. Decline in leaf area was due to fewer mesophyll cells. Net photosynthetic rate (PN) increased up to full expansion, remained constant for about a week and then declined. PN, nitrogen, ribulose-1,5-bis-phosphate carboxylase/oxygenase (RuBPCO) amount and activity, chlorophyll and soluble protein contents were similar at all the N concentrations. Both amount and activity of RuBPCO in the flag leaf were about two fold higher as compared to penultimate leaf, but PN was similar. This indicates the presence of an excess amount of RuBPCO in the flag leaf.  相似文献

Differences in acclimation to elevated growth CO2 (700 μmol mol−1, EC) and elevated temperature (ambient +4 °C, ET) in successive leaves of wheat were investigated in field chambers. At a common measurement CO2, EC increased photosynthesis and the quantum yield of electron transport (Φ) early on in the growth of penultimate leaves, and later decreased them. In contrast, EC did not change photosynthesis, and increased Φ at later growth stages in the flag leaf. Contents of chlorophyll (Chl), ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), and total soluble protein were initially higher and subsequently lower in penultimate than flag leaves. EC decreased RuBPCO protein content relative to soluble protein and Chl contents throughout the development of penultimate leaves. On the other hand, EC initially increased the RuBPCO:Chl and Chl a/b ratios, but later decreased them in flag leaves. In the flag leaves but not in the penultimate leaves, ET initially decreased initial and specific RuBPCO activities at ambient CO2 (AC) and increased them at EC. Late in leaf growth, ET decreased Chl contents under AC in both kinds of leaves, and had no effect or a positive one under EC. Thus the differences between the two kinds of leaves were due to resource availability, and to EC-increased allocation of resources to photon harvesting in the penultimate leaves, but to increased allocation to carboxylation early on in growth, and to light harvesting subsequently, in the flag leaves.  相似文献

Statistical analysis of Km (CO2) values of ribulose-1,5-bisphosphate (RuBP) carboxylase from 35 C4 grass species shows that the mean value for PEP-carboxykinase (PCK) type C4 species (41.4±s.e. 2.2 μM CO2) is significantly different from that of NAD-malic enzyme (NAD-ME) type species (55.3±3.1 μM CO2) or NADP-malic enzyme (NADP-ME type species (52.5±s.e. 2.0μM CO2). These C4 type differences remain detectable within both the eu-panicoid and chloridoid grass subfamilies. By contrast, no between-subfamily differences were found within C4 types. Variation in Km (CO2) values of RuBP carboxylase may be related to in vivo differences in CO2 concentration at the enzyme site, mediated perhaps by differences in CO2-leakiness of C4 leaf ‘photosynthetic carbon reduction’ (PCR or ‘Kranz’) tissue.  相似文献

Changes in contents of pigments, chlorophyll-protein complex, and photosynthetic activities were investigated in field grown apple (Malus pumila Mill.) leaves infected by Apple Proliferation phytoplasma. The contents of chlorophyll a+b (Chl) and carotenoids (Car) markedly decreased in infected leaves. Similar results were also observed for content of total soluble proteins and ribulose-1,5-bisphosphate carboxylase activity. When various photosynthetic activities were followed in isolated thylakoids, phytoplasma infection caused a marked inhibition of whole chain and photosystem 2 (PS2) activity. Smaller inhibition of photosystem 1 (PS1) activity was observed even in severely infected leaves. The artificial exogenous electron donors, MnCl2 diphenyl carbazide, and NH2OH, did not restore the loss of PS2 activity in both mildly and severely infected leaves. Similar results were obtained by Chl fluorescence measurements. The marked loss of PS2 activity in infected leaves was due to the reduction of contents of chlorophyll and light-harvesting chlorophyll-protein 2 complexes. Immunochemical probes are widely used to identih different species and to quantify and understand the role that different antigens play within cells. We optimized a single-cell immunofluorescent assay for the carbon fixation enzyme ribulose-1,5-bisphosphate carboxylase (Rubisco) in order to quantify the enzyme by flow cytometry in phytoplankton cells. The criteria for optimization of the immunofluorescent assay for Rubisco in single cells included maximization of Rubisco immunogenicity, minimization of Rubisco diffusion out of the cells, minimization of cell breakage, and maximization of the cell labeling. Several fixatives (cross-linkers and denaturing) and permeabilizing agents were tested on 26 species of phytoplankton. The only fixative / permeabilizing agent that fulfilled the criteria established for the assay was 96% ethanol. Phytoplankton cells collected from the field needed further treatment with a strong oxidant to permeabilize ethanolfixed cells and thus allow the antibody probe to access the Rubisco antigen. This study should have a general applicability to the study of other soluble photosynthetic antigens in single phytoplankton cells.  相似文献

Bacterial microcompartments (BMCs) are polyhedral bodies, composed entirely of proteins, that function as organelles in bacteria; they promote subcellular processes by encapsulating and co-localizing targeted enzymes with their substrates. The best-characterized BMC is the carboxysome, a central part of the carbon-concentrating mechanism that greatly enhances carbon fixation in cyanobacteria and some chemoautotrophs. Here we report the first structural insights into the carboxysome of Prochlorococcus, the numerically dominant cyanobacterium in the world's oligotrophic oceans. Bioinformatic methods, substantiated by analysis of gene expression data, were used to identify a new carboxysome shell component, CsoS1D, in the genome of Prochlorococcus strain MED4; orthologs were subsequently found in all cyanobacteria. Two independent crystal structures of Prochlorococcus MED4 CsoS1D reveal three features not seen in any BMC-domain protein structure solved to date. First, CsoS1D is composed of a fused pair of BMC domains. Second, this double-domain protein trimerizes to form a novel pseudohexameric building block for incorporation into the carboxysome shell, and the trimers further dimerize, forming a two-tiered shell building block. Third, and most strikingly, the large pore formed at the 3-fold axis of symmetry appears to be gated. Each dimer of trimers contains one trimer with an open pore and one whose pore is obstructed due to side-chain conformations of two residues that are invariant among all CsoS1D orthologs. This is the first evidence of the potential for gated transport across the carboxysome shell and reveals a new type of building block for BMC shells.  相似文献

Attention is drawn to literature evidence that leaf virus infection may interfere with synthesis of ribulose-1,5-bisphosphate carboxylase oxygenase, and with the activity of photosystem II, two main requirements for shade-sun acclimation. Preliminary experiments which show that virus infections resulting in these interactions can lead to photosynthetic physiology diagnostic of nitrogen limited plants and/or shade ecotypes are presented. Other observations on the diversity of virus infections in sun populations of wild species commonly found in the shade are presented. Together, this evidence leads to the hypothesis that virus infection may well confine individuals of some species to shaded habitats because infection prevents acclimation to bright light, and predisposes them to photoinhibition.  相似文献

The quaternary structure of ribulose-1,5-bisphosphate carboxylase-oxygenase (rubisco) from Rhodospirillum rubrum, an enzyme consisting of two large subunits, L2, was investigated by small-angle X-ray scattering. In the presence of HCO 3 - and Mg2+, rubisco is in the active state and displays a radius of gyration of 2.96 nm, a maximum diameter of 9.5 nm and a volume of 170 nm3. A model is presented where the subunits are arranged back-to-back, rotated relative to each other by 90°, and shifted by 1.3 nm. Upon inactivation by removal of HCO 3 - and Mg2+, the model swells slightly without any distinct changes in configuration. The changes of protein and amino acid contents of young and old leaves from field citrus trees [ Citrus sinensis (L.) Osbeck cv. Washington Navel] were studied throughout the year. the total protein content of old leaves decreased during the spring and summer growths, whereas in young developing leaves it increased rapidly. The end of the spring and summer flushes was followed by a quick recovery of the initial protein content due to a process of reversible senescence. The evolution of SDS-PAGE proteinograms indicated that most of the foliar proteins contributed to the nitrogen mobilization during the spring and summer growth periods. A protein band of molecular weight ca 55,000, probably containing the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase, decreased relatively more than the others. The amino acid content of the leaves increased during winter time due to the accumulation of proline, which was mobilized during the spring flush. The possible nitrogen storage function of proline is discussed.  相似文献

Wild-type and antisense rbcS tobacco (Nicotiana tabacum) plants were grown in a glasshouse in midsummer in Portugal with an irradiance of 1500–2000 μmol m−2s−1 and daytime temperatures of 30–35 °C. The Rubisco content of the transformants was lower by 35, 80 and over 90% than that of the wild-type. Gas exchange was measured over three separate days. There was a near-linear relation between Rubisco content and photosynthetic rate during the period of high irradiance, allowing a flux control coefficient of 0.83–0.89 to be estimated. The relation deviated slightly from linearity, because the internal CO2 concentration (c;) was higher in the transformants than in the wild-type (190 and 275 μmol mol−1 in plants with 35 and 80% less Rubisco, respectively, compared with 175 μmol mol−1 for wild-type), compensating to some extent for the decreased Rubisco content. This increase in ci occurred because the stomatal conductance (g) remained unaltered or was even higher in plants with decreased Rubisco, despite the lower rate of CO2 assimilation. As a consequence, water use efficiency declined. The decreased rate of photosynthesis was not accompanied by a stoichiometric decrease in apparent growth rate. These results are discussed in relation to earlier studies of the plant set in growth cabinets. It is concluded that tobacco can adjust over a wide range of growth conditions to avoid a onesided limitation by Rubisco, but that in extreme environmental conditions this capacity to adapt is exhausted.  相似文献

The distributions of bacterial form IA and form IC ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) were investigated using Lowes Cove intertidal mudflat and Damariscotta Lake littoral sediments by PCR amplification of 492-495 bp fragments of the large subunit RuBisCO gene, cbbL. Genomic extracts for amplification were obtained from lake surface (upper 2 mm), mudflat surface (upper 2 mm), subsurface (5-7 cm), and soft-shell clam (Mya arenaria) burrow-wall sediments, as well as from a sulfide-oxidizing mat. Phylogenetic analyses of cbbL clone libraries revealed that Lowes Cove sediments were dominated by form IA cbbL-containing sequences most closely related to cbbL genes of sulfur-oxidizing bacteria or sulfide-oxidizing mats. In contrast, Damariscotta Lake cbbL clones contained primarily form IC cbbL sequences, which typify aerobic CO- and hydrogen-oxidizing facultative chemolithotrophs. Statistical analyses supported clear differentiation of intertidal and lake chemolithotroph communities, and provided evidence for some differentiation among intertidal communities. amova and libshuff analyses of Lowes Cove libraries suggested that M. arenaria burrow-wall sediments did not harbour distinct communities compared with surface and subsurface sediments, but that surface and subsurface libraries displayed moderate differences. The results collectively support a conceptual model in which the relative distribution of form IA- and IC-containing bacterial chemolithotrophs depends on sulfide availability, which could reflect the role of sulfate reduction in sediment organic matter metabolism, or the presence of geothermal sulfide sources.  相似文献

Ribulose-1,5-biphosphate (RuBP) carboxylases from 38 grass species (26 genera), isolated via affinity chromatography, compare well in amino acid compositions with the enzyme extracted and purified from the same and other species (46 species, 38 genera) by an alternative procedure. Taxonomic differences in the amino acid composition of the enzyme exist between the major grass groups. Those of pooids are distinguishable from those of chloridoids, eu-panicoids and andropogonoids, while those of a bamboo, Oryza, Microlaena and Stipeae bear closer resemblance to those of pooids. The amino acid composition of RuBP carboxylases does not resemble that of total leaf proteins of the grasses. Variations detected in the amino acid compositions of RuBP, carboxylases are independent of observed differences in kinetics between C3 and C4 versions of the enzyme.  相似文献
