β-Galactosidase (EC 3.2.1.23) was purified from the cell wall of the fruit of Japanese pear ( Pyrus serotina Rehder var. culta Rehder cv. Hosui) and characterized. Five peaks of β-galactosidase activity, designated as Gal I to V, were separated by hydrophobic chromatography on butyl toyopearl and ion exchange chromatography on Mono S. These isolated β-galactosidases were investigated with regard to their abilities to release monomeric galactose from the fractionated polymers of native cell wall (cyclo-hexane-trans-1,2-diamine tetraacetic acid-, Na2CO3-, guanidine thiocyanate- and KOH-soluble fractions) and arabinogalactan (from larch wood). All the β-galactosidase fractions were active against native cell wall polysaccharides although to varying degrees. Gal I reacted to all fractions of native cell wall polysaccharides although to varying degrees. Gal I reacted to all fractions of native cell wall and arabinogalactan. Gal II released much galactose only from KOH-soluble polymers and arabinogalactan. Gal III released the most galactose. from cyclohexane- trans -1,2-diamine tetraacetic acid-, Na2CO3- and guanidine thiocyanate-soluble cell wall polymers which probably contained galactosyl side chains of pectic polymers, although it did not react much to arabinogalactan. In addition, the activity of Gal Ill dramatically increased as ripening proceeded. Furthermore, Gal III was purified to homogeneity by gel filtration on TSKgel 3000SW and the size of a polypeptide was 80 kDa on SDS-PAGE.  相似文献

The autolytic process in epicotyl cell walls of Cicer arietinum L. cv. Castellana, and also the hydrolysis of heat-inactivated cell walls as mediated by a cell wall β-galactosidase (EC 3.2.1.23) (named βIII and previously characterized as responsible for the autolysis), are maximal on the fourth day of germination and coincide with the maximal growth capacity. They decrease during the following days, in which the growth rate diminishes. In both cases, no differences were observed in the percentages of the different sugars released, galactose being the principal one. The βIII fraction from aged epicotyl cell walls hydrolyzed young walls in proportion to its specific activity, and more efficient than when cell walls from aged material were used as the substrate. The βIII fraction from 4 day-old epicotyls (the time for maximal autolysis) was incapable of hydrolyzing aged epicotyl cell walls to the same extent as young ones. These results, together with the levels and activity of the enzyme throughout growth, allow the assumption that the variations in the autolysis and hydrolysis caused by βIII during growth processes are due to structural modifications in the cells walls, modifications that would limit access of the enzyme to its substrate, thus impeding the release of galactose, even though the enzyme is present.  相似文献

The Hw pectic fraction, extracted with hot water, is the major component of 4 days old epicotyl cell walls of Cicer arietinum L. cv. Castellana and is formed of arabinose and galactose, with smaller amounts of rhamnose, xylose, glucose and mannose. The cell wall 2βIII enzymatic fraction, with β-galactosidase activity (EC 3.2.1.23) and the main enzyme responsible for the autolytic process, essentially acts on the Hw fraction, and is able to hydrolyze 560 μg of this fraction per g of epicotyls, releasing mainly galactose as monosaccharide.
The 2βIII fraction acts very weakly on the other polysaccharide fractions of the cell wall, both pectic and hemicellulosic, releasing 80, 60 and 14 μg per g of epicotyls from the fractions extracted with oxalate (Ox), KOH 10% (KI) and KOH 24% (KII), respectively. It can be concluded that the natural substrate of this enzyme is the Hw pectic fraction, probably an arabinogalactan that is found in the cell wall in isolated form or as side chains of the rhamnogalacturonan I.  相似文献

The products released in cell wall autolysis from 4-day-old epicotyls of Pisum sativum elute in gel filtration chromatography (Bio Gel P.2) as two components, mono and polysaccharides, in a practically constant ratio over the time of incubation. The polysaccharides are mainly composed of arabinose and galactose, with smaller amounts of xylose and glucose, whereas the monosaccharide are almost exclusively composed of galactose. The same results were obtained when inactive cell walls were hydrolyzed by the enzymes extracted from the cell wall with LiCl. The hydrolysis of the different cell wall fractions by these enzymes shows that the autolytic substrates are preferentially located on the pectic fractions.  相似文献

β‐Galactosidases (EC 3.2.1.23) from ripe papaya ( Carica papaya L. cv. Eksotika) fruits having galactanase activities were fractionated by a combination of cation exchange and gel‐filtration chromatography into three isoforms, viz., β‐galactosidase I, II and III. The native proteins of the respective isoforms have apparent molecular masses of 67, 67 and 55 kDa, each showing one predominant polypeptide upon SDS‐PAGE of about 31 and 33 kDa for β‐galactosidases I and III, respectively, and of 67 kDa for β‐galactosidase II. The β‐galactosidase I protein, which was undetectable in immature fruits, appeared to be specifically accumulated during ripening. The β‐galactosidase II protein was present in developing fruits, but its level seemed to decrease with ripening. β‐Galactosidase I seemed to be an important softening enzyme; its activity increased dramatically (4‐ to 8‐fold) to a peak early during ripening and correlated closely with differential softening as related to position in the fruit tissue. The inner mesocarp tissue was softer, and its wall pectins were modified earlier and firmness decreased more rapidly during ripening compared to the outer mesocarp tissue. β‐Galactosidase II also may contribute significantly to softening because of its ability to catalyse increased solubility and depolymerization of pectins as well as through its ability to modify the alkali‐soluble hemicellulose fraction of the cell wall. The physiological significance of both β‐galactosidase isoforms may partly be attributed to their functional capacity as β‐(1,4)‐galactanases.  相似文献

Carbohydrate solubilization and glycosidase activities were investigated in tomato ( Lycopersicon esculentum Mill. cv. Solar Set) locule cell walls to identity processes involved in the liquefaction of this tissue. Cell walls were prepared from the locule tissue of fruit at the immature green, mature green, and breaker stages of development. Enzymically active walls incubated in dilute buffer released high molecular mass pectins, oligomeric carbohydrate, and the neutral sugars rhamnose, glucose, galactose, arabinose, xylose, and mannose. The release was sustained for at least 50 h at 34°C and was inhibited more than 50% by 1 m M Hg2+. Pectins released from the cell walls of locule tissue at progressive stages of liquefaction were similar in molecular mass and showed no evidence of downshifts on a Sepharose CL–2B–300 column during prolonged incubation. A cell-free protein extract prepared from the locule tissue of mature-green fruit promoted a net release of polymeric and monomeric carbohydrates from high-temperature inactivated cell walls. Polygalacturonase activity was not detected in locule protein although glycosidases including β-mannosidase (EC 3.2.1.25), α- and β-galactosidases (EC 3.2.1.22–23), β-arabinosidase (EC 3.2.1.56) and β-glucosidase (EC 3.2.1.21) were present. Pectinmethylesterase (EC 3.1.1.1 1) activity was detected at the immature-green stage but declined to negligible levels in mature-green and breaker locule tissue. Parallels between the in vitro solubilization of carbohydrate from locule tissue cell walls and the changes occurring during locule liquefaction are discussed.  相似文献

Pisum sativum L. (cv. Lincoln) epicotyl cell walls show autohydrolysis and release into the incubation medium up to 120 μg of sugar per mg of cell wall dry weight in 30 h. Cell walls from younger epicotyls with high growth capacity showed higher auto-lytic capacity than older epicotyls. This suggests that both processes, growth and au-tolysis, are related. The proteins responsible for autolysis were extracted from the wall fraction with high saline solution (3 M LiCl) and enzymatic activities associated with the proteins were studied. The highest activity corresponded to α-galactosidase; lower activities were found for β-galactosidase, a-arabinosidase and exoglucanase. Changes in enzymatic activities and changes in the proportion of sugars released in autolysis by cell walls during the growth of epicotyls support the notion that α-galac-tosidase is one of the enzymes involved in the process of autolysis, and that the liberation of arabinose and galactose in this process occurs as arabinogalactan.  相似文献

Coleoptile cell wall proteins from Zea mays L. hybrid B 37 × Mo 17 were extracted and fractionated. Three enzymes identified in that extract were examined to determine their role in cell wall hydrolysis with a goal of evaluating the extent to which they participated in autohydrolytic reactions. Two separate proteins were identified as endo- and exo-glucanases. Incubation of these enzymes with heat inactivated cell walls, liberates products derived from the constitutive (1→3), (1→4)-β- d -glucan. The release of sugars from walls resembles that of cell wall autolysis. A third cell wall protein degraded polysaccharides in a more general manner, releasing carbohydrates containing xylose, arabinose, galactose and glucose. Polyclonal antibodies raised against the exoglucanase protein suppressed autolytic reactions of isolated cell wall.  相似文献

β-Galactosidase (EC 3.2.1.23) has been established as the main enzyme involved in the autolytic process. The enzyme extracted from cell walls of epicotyls of Cicer arietinum L. cv. Castellana with 3 M LiCl is a 45 kDa protein composed of a single subunit, having an optimum pH of 4; an optimum temperature of 45°C and Km and Vmax of 1.72 m M and 18.5 nkat (mg protein)–1 respectively, as evaluated against p -nitrophenyl-β- d -galactopyranoside. The enzyme is inhibited by Hg2+, d -galactono-1,4-lactone and galactose, substances that also inhibit the autolytic process. Ca2+ and EDTA, which do not affect the activity of the β-gaiactosidase, do however modify the hydrolysis of the cell wall mediated by the enzyme, and they also inhibit the autolytic process. Ca2+ decreased both processes, whereas EDTA increased them; and when both substances were added together, their individual effects were neutralized. The effects of both agents is probably due to modifications in the cell wall that prevent access of the enzyme to its substrate.  相似文献

Galactanase from Phytophthora infestans and an arabinosidase isoenzyme from Sclerotinia fructigena attacked the cortical cell walls of apple fruits liberating galactose and arabinose residues, respectively. Other arabinosidase isoenzymes from S. fructigena attacked cell walls very slowly. A S. fructigena polygalacturonase isoenzyme liberated half of the uronic acid residues with few associated neutral residues, while a second polygalacturonase isoenzyme released more uronic acid with a substantial proportion of arabinose and galactose and lesser amounts of xylose, rhamnose and glucose; reaction products of this enzyme could be further degraded by the first isoenzyme to give high MW fragments, rich in arabinose with most of the xylose, rhamnose and glucose, and low MW fragments rich in galactose and uronic acid. Endoglucanase from Trichoderma viride released a small proportion of the glucose residues from cell walls together with uronic acid, arabinose, xylose and galactose; more extensive degradation occurred if walls were pre-treated with the second polygalacturonase isoenzyme. Endoglucanase reaction products were separated into a high MW fraction, rich in arabinose, and lower MW fractions rich in galactose and glucose residues. The high MW polygalacturonase and endoglucanase products could be degraded with an arabinosidase isoenzyme to release about 75% of their arabinose. Cell walls from ripe fruit showed similar susceptibility to arabinosidase and galactanase to those from unripe apples. Cell walls from fruit, ripened detached from the tree were more susceptible to degradation by polygalacturonase than walls from unripe fruit or fruit ripened on the tree. Endoglucanase released less carbohydrate from ripe fruit cell walls than from unripe fruit cell walls.  相似文献

Oryza sativa L. var. bahia coleoptile cell walls show sufficient autolytic activity for the release into the surrounding medium of amounts up to 60 μg of sugars per mg of dry weight of cell wall. The products released elute in Bio-gel P.2 as mono- and polysaccharides with glucose as the sole component. The polysaccharide component releases tri- and tetrasaccharides on treatment with a glucanase specific for β (1–3) (1–4) linkages in the same proportion as that of the mixed glucan of the cell wall. This supports the hypothesis that the polysaccharide component originates from the cell wall glucan and that autolysis is therefore related to the processes of the loss of rigidity of the cell wall. Nojirimycin (a specific glucanase inhibitor and inhibitor of auxin-induced elongation) decreases autolytic activity of the cell walls, reducing it to 30% of its normal value. Bio-gel P. 2 elution of the products released in autolysis in the presence of nojirimycin shows that only the monosaccharide fraction was affected.  相似文献

Abstract: Chromogranin A and two other proteins (A1 and A2) of the soluble proteins of bovine chromaffin granules were isolated by extraction from polyacrylamide gels after electrophoresis. The carbohydrate content of these proteins was 5%, with galactose, N -acetylgalactosamine, and sialic acid as the main sugars. Membranes of chromaffin granules were solubilized with sodium dodecyl sulphate (SDS) and three glycoproteins were isolated by sequential affinity chromatography on Concanavalin A (Con A) and wheat germ lectin (WGL) Sepharose columns. Two glycoproteins, designated GP II and III, were found to have a high carbohydrate content of about 30%. Mannose, galactose, N -acetylgalactosamine, and sialic acid were the main sugars. In addition membrane-bound dopamine β-hydroxylase was isolated by this procedure. No significant differences between the carbohydrate composition of the membrane-bound and the soluble enzyme were revealed. It was shown that all four subunits of dopamine β-hydroxylase possess carbohydrate chains with an affinity for Con A. The isolation methods established in this study will be useful for immunological studies on these glycoproteins.  相似文献

A sulfated polysaccharide fraction was isolated from the hot water extract of the green alga Caulerpa racemosa and designated HWE. This polymer, which contained galactose, glucose, arabinose and xylose as the major component sugars, had [alpha](D)(30) + 46.2 degrees in water and contained 9% sulfate hemiester groups. Sugar linkage analysis indicates that HWE was branched and mainly contained 1,3- and 1,3,6-linked galactose, 1,3,4-linked arabinose, 1,4-linked glucose and terminal- and 1,4-linked xylose residues. Sulfation was deduced from infrared spectroscopy and methylation analysis to occur on O-6 of galactose and O-3 of arabinose. The native polysaccharide could be fractionated by size exclusion chromatography into two overlapping fractions and the major fraction has a hydrodynamic volume similar to that of 70 kDa dextran. HWE was a selective inhibitor of reference strains and TK(-) acyclovir-resistant strains of herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) in Vero cells, with antiviral effective concentration 50% (EC(50)) values in the range of 2.2-4.2 microg/ml and lacking cytotoxic effects. Furthermore, HWE did not exhibit anticoagulant properties at concentrations near the EC(50).  相似文献

The behaviour of β-1,3-glucanase activity during Aspergillus nidulans autolysis was studied in a basal medium and in the same medium supplemented with 0.5 g l-1 of microcrystalline cellulose, laminarin, pectin, seedling of Lycopersicum esculentum extract, chitin and xylan respectively. In any case β-1,3-glucanase activity was detected in the culture fluid before the onset of the autolysis, but afterwards a progressive increase of β-1,3-glucanase activity took place with incubation time. In the media supplemented with pectin and seedling of Lycopersicum esculentum extract higher activity in the first days of autolysis was found. The activity at the end of the studied process by sample was 2.5, 2.1, 2.5, 1.9, 2.2, 2.3 and 2.3 U, and the specific activity 83, 53, 85, 55, 64, 90 and 53 mU mg-1 of protein for each medium respectively. The β-1,3-glucanase activity in Aspergillus nidulans seems to be related to autolysis and not to the presence of different substances in the culture medium. The behaviour of β-1,3-glucanase activity during the degradative process was followed by FPLC ion-exchange chromatography. Three proteins (I, II, III) with β-1,3-glucanase activity were separated and quantified. These proteins have similar behaviour in all the media. Proteins I and II increase progressively with incubation time but protein III is only present at the first and last days of autolysis.  相似文献

Biochemical changes associated with the ripening of hot pepper fruit   总被引:5,自引:0,他引:5  
Hot pepper ( Capsicum annuum L. cv. Chooraehong) fruit underwent a respiratory climacteric during ripening. However, the rate of ethylene production was low, reaching a maximum of approximately 0.7 μl kg−1 h−1 at the climacteric peak when the surface color was 30 to 40% red. Ripening was accompanied by a loss of galactose and arabinose residues from the cell wall. The content of uronic acid and cellulose in the wall changed only slightly during ripening. The average molecular weight of a cell wall hemicellulosic fraction shifted progressively toward a lower molecular weight during ripening. Total β-galactosidase (EC 3.2.1.23) activity increased 50-fold from the immature green to the red ripe stage. No polygalacturonase (EC 3.2.1.15) activity was detected at any stage of ripeness. Thus, the loss of galactose and arabinose residues from the cell wall, as well as the observed modification of hemicelluloses during ripening, seem to be unrelated to active polygalacturonase. Soluble polyuronide content remained relatively constant at approximately 60 μg (g fresh weight)−1 as fruit ripended.  相似文献

A fractionation of non-cellulosic sugars of Oryza sativa L. coleoptile cell walls was carried out and the composition of each fraction was studied during coleoptile growth.Percentages of fractions extracted with boiling water and with oxalate (pectic substances) were almost constant throughout development. An increase in the K II hemicellulosic fraction (extracted with 24% KOH) content, and a decrease in the K I hemicellulosic fraction (extracted with 10% KOH) were detected, when coleoptile growth finished.The percentage of glucose content in the K I hemicellulosic fraction was highest in young coleoptiles and lowest in old ones. The neutral sugars and amino sugars, released by acid hydrolysis of walls and polysaccharidic fractions, of six species of Talaromyces and the infrared spectra have been used to study their interspecific relationships. In whole cell walls neutral sugars ranged from 23 to 39.6% dry weight and were identified as glucose, galactose and mannose. Glucosamine varied from 8 to 19.8% in the samples. Galactosamine (2% or less) was found in T. emersonii and T. rotundus and no galactosamine in the other species. Sequential fractionation of the cell walls with alkali and acid gave several polysaccharidic fractions. The main differences among species were found in the alkali-soluble fraction at 20° (F1). This fraction represented 8 to 33.2% of the whole cell wall and was characterized as an -glucan in T. bacillisporus, T. emersonii, T. luteus and T. rotundus (Group A) and as a -galactofuranosyl containing glucan in T. ohiensis and T. stipitatus (Group B). The alkali-insoluble residue (F4) represented the bulk of the cell wall in all species tested (33.2% to 57.3%) and was characterized as a -glucan/chitin complex. An exhaustive cell wall fractionation of Fusarium oxysporum f. sp. lycopersici race 2 ( Fol 2) with alkali in a sequential procedure yields only three polysaccharide fractions: F1s (alkali and water soluble), F11 (alkali soluble and water insoluble) and F4 (alkali-insoluble residue). These fractions amounted respectively to 15, 1.3 and 52% of the cell wall and have been characterized by infra-red spectroscopy and gas liquid chromatography-mass spectrometry (GLC-MS). F1s is a β-gluco-galacto-mannan, F11 is mainly composed of a β-1, 3-glucan and F4 is a β-1,3-glucan-chitin complex. The F1s is a very complex polysaccharide and its hydrolysis requires the action of different enzymes. The lysis of the cell wall and its three fractions with lytic enzymes from Fol 2 has been studied and a correlation between the lysis of the cell wall and the lysis of these fractions was found. The amount of glucose, galactose and mannose in F1s and cell wall hydrolysates were quantified by GLC and they indicated the hydrolysis of the gluco-galacto-mannan polysaccharide. In the hydrolysis of F4 and cell walls N -acetylglucosamine was also found and quantified. When chlamydospores of this fungus were treated with Fol 2 lytic enzymes, the sugars liberated were principally mannose and N -acetylglucosamine. These results indicate that Fol 2 produces during its autolysis the necessary enzymes to hydrolyse its own cell walls. This fact suggests that a biological control of Fol 2 with its own lytic enzymes, conveniently immobilized, could be developed.  相似文献

Cell walls prepared from the endosperm tissue of hydrated lettuce (Lactuca sativa L.) seeds undergo autohydrolysis. Release of carbohydrates is most rapid (0.4-0.6 [mu]g per endosperm) within the 1st h of incubation in buffer, but substantial autolysis is sustained for at least 10 h. Autolysis is temperature sensitive, and the optimum rate occurs at pH 5. The rate of autolysis increases markedly in the period just prior to radicle emergence. The cell-wall polysaccharide composition in micropylar and lateral endosperm regions differs significantly; the micropylar walls are rich in arabinose and glucose with substantially lower amounts of mannose. Although walls prepared from both micropylar and lateral regions undergo autolysis, micropylar walls release carbohydrates at a higher rate than lateral walls. Autolysis products elute as large polymers when subjected to size-exclusion chromatography, suggesting that endo-enzyme activity is responsible for release of fragments containing arabinose, galactose, mannose, and uronic acids. Arabinose, galactose, mannose, and glucose are also released as monomers. As a function of time, the ratio of polymers to monomers decreases, indicating that exo-enzyme activity is also present. Thermoinhibition or treatment with abscisic acid suppresses germination and reduces the rates of autolysis of walls isolated from the endosperm by about 25%. Treatments that alleviate thermoinhibition (kinetin and gibberellic acid) increase the rates of autolysis by 20 to 30% when compared to thermoinhibited controls.  相似文献

The polysaccharidic effect of a purified 1,3- β -glucanase, a purified β -glucosidase, and of partially purified endo-1,3- β -glucanase from autolysed Penicillium oxalicum cultures on cell wall isolate fractions from the same fungus were studied.
Fractionation of 5-day-old cell wall gave rise to a series of fractions that were identified using infrared spectrophotometry. The fractions used were: F1, an α -glucan; F3, a β -glucan; F4, a chitin-glucan; and F4b, a β -glucan. The fractions were incubated with each of the enzymes and with a mixture of equal parts of the three enzymes and the products of the enzymatic hydrolysis were analyzed after 96 h incubation.
The enzymes were found to degrade fraction F4b ( β -glucan); the greatest degree of hydrolysis was reached when the three enzymes were used together, suggesting the need for synergic action by these enzymes in the cell wall degradation process.  相似文献
