A facile and efficient method has been developed for the optical resolution of racemic jasmonic acid (JA) on a relatively large scale and was successfully utilized for the preparation of optically pure (+)-JA and (?)-JA. We indicated that (+)-JA has lower growth inhibitory activity than (?)-JA in the rice seedling growth test and confirmed in line with an earlier observation that their respective biologically-active forms, (+)-JA-Ile and (?)-JA-Ile, show comparable inhibitory activities. We compared the metabolism of (+)-JA and (?)-JA into (+)-JA-Ile and (?)-JA-Ile, respectively, in the JA-deficient rice cpm2, and found that the exogenously applied (+)-JA was metabolized to the corresponding Ile conjugate less efficiently as compared with (?)-JA. Such metabolic rate difference may cause a discrepancy between biological potencies of (+)-JA and (?)-JA in rice.

Summary 1. Our aim was to test the hypothesis that selectivity for D3 dopamine (DA) receptors may contribute to limbic anti-DA selectivity ofS-(+)-aporphine DA partial agonists.2. Affinity was tested with3H-emonapride, using human D3 receptors in mouse fibroblasts and D2 receptors in rat striatal tissue.3. D3 receptors showed a picomolar affinity for3H-emonapride, Na+ dependence, and reversible saturability, as well as stereoselectivity. Confirmatory or novel D3/D2 pharmacologic selectivity was found with several benzamides, thioxanthenes, buspirone, GBR-12909, and DA agonists including hydroxyaminotetralins [ADTN, (+)-7-OH-DPAT, (–)-PPHT and its fluorescein derivative], (–)-N-propylnorapomorphine, (–)-3-PPP, (–)-quinpirole, and SDZ-205-502, but neither aminoergoline nor (+)-aporphine partial agonists.4. The biotransformation of [2-14C](±)9, 10-dihydrojasmonic acid (DJA) was studied in excised shoots of 6-day-old barley seedlings after 72 h. From the ethyl acetate extract, some minor metabolites were isolated and purified by DEAE-Sephadex A-25 chromatography, thin-layer chromatography (TLC), C18-cartridges, and high-performance liquid chromatography (HPLC). The structural identification of these metabolites was performed by gas chromatography-mass spectrometry (GC-MS), circular dichroism (CD), and amino acid analysis, and the following amino acid conjugates were found:N-[(–)9,10-dihydrojasmonoyl]valine,N-[(–)9,10-dihydrojasmonoyl]isoleucine,N-[9,10-dihydrojasmonoyl]leucine,N-[11-hydroxy-9,10-dihydrojasmonoyl]valine,N-[11-hydroxy-9,10-dihydrojasmonoyl]isoleucine,N-[12-hydroxy-9,10-dihydrojasmonoyl]isoleucine; and the cucurbic acid-related compoundsN-{[3-hydroxy-2(4-hydroxypentyl)-cyclopent-1-yl]-acetyl}isoleucine andN-{[3-hydroxy-2(5-hydroxypentyl)-cyclopent-1-yl]-acetyl}isoleucine. The results suggest conjugation with isoleucine and valine, as well as preferential hydroxylation at position C-11 or hydrogenation at position C-6, as being important steps in the metabolism of (±)DJA in barley shoots.  相似文献

Factors involved in promoting caulogenesis from hypocotyl explants of Phyllanthus fraternus were studied. Hypocotyl explants were cultured on B5 medium supplemented with 2,4-D or NAA in the presence and absence of BAP (at concentrations 0, 10–7, 10–6 and 10–5M). Adventitious shoots differentiated from callus developed from the cut ends of 12.5% of the hypocotyl segments cultured on medium supplemented with 10–6M BAP in combination with 10–6M 2,4-D or 10–6M NAA. Profuse rooting occurred from the hypocotyl explants on medium supplemented with 10–6M BAP + 10–6M NAA. Incorporation of casein hydrolysate in B5 medium along with 10–6M BAP + 10–7M 2,4-D enhanced the frequency of cultures with adventitious shoots upto 68.0%. Glutamine, glutamic acid or proline could partially substitute for the effect of casein hydrolysate. Amongst the hypocotyls from 3–14 d old seedlings, the best caulogenesis was obtained with hypocotyls from 7 d old seedlings both in presence or absence of casein hydrolysate. Best rooting of shoots was achieved on half-strength B5 medium supplemented with 10–6M IBA. Relatively little is known about the responses of plants to micronutrients when these nutrients are maintained at the very low levels found in soils of low fertility. We have determined the requirement of barley (Hordeum vulgare L. cv Herta) for ionic Mn2+ in plant culture solutions using the chelating agent HEDTA as a buffer for micronutrient metal ions. The chemical activity of Mn2+ was varied approximately 10,000-fold from log(Mn2+)=–10.8 to –6.8 (pMn 10.8 to pMn 6.8), while holding constant the activities of the other micronutrient cations. Growth, appearance, and composition of Herta barley indicated that log(Mn2+) of approximately –8.3 would permit optimal dry matter production and normal plant development. The specific accumulation rate of Mn by 15 to 23 day old seedlings was a linear function of the Mn2+ activity in solution. At log(Mn2+) of about –9.8 or below, barley seedlings were unable to accumulate significant amounts of Mn, and at some harvests, suffered a net loss of Mn to solution. Seedlings younger than 11 days old were ineffective accumulators of several cations, including Mn, Fe, Zn, Cu, Mg, and Ca. The TES ether of the C6-hydroxy derivative of naturally occurring epi-jasmonic acid (epi-JA) was designed as epimerization-free equivalent of epi-JA. The TES ether was synthesized from (1R,4S)-4-hydroxycyclopent-2-enyl acetate in 13 steps. The acid part of the ether was activated with ClCO2Bu i and subjected to condensation with l-amino acid at room temperature for 48 h. The TES group in the condensation product was removed in HCO2H (0°C, 30 min) and the resulting hydroxyl group was oxidized with Jones reagent (acetone, 0°C, 30 min) to furnish the amino acid conjugate of epi-JA. The amino acids examined are l-isoleucine, l-leucine, l-alanine, l-valine, and d-allo-isoleucine, which afforded the conjugates in 48–68% yields with 89–96% diastereomeric purity over the trans isomers. Similarly, the possible three stereoisomers of epi-JA were condensed with l-isoleucine successfully, producing the corresponding stereoisomers in good yields.  相似文献

Jasmonate:amino acid synthetase (JAR1) is involved in the function of jasmonic acid (JA) as a plant hormone. It catalyzes the synthesis of several JA-amido conjugates, the most important of which appears to be JA-Ile. Structurally, JAR1 is a member of the firefly luciferase superfamily that comprises enzymes that adenylate various organic acids. This study analyzed the substrate specificity of recombinant JAR1 and determined whether it catalyzes the synthesis of mono- and dinucleoside polyphosphates, which are side-reaction products of many enzymes forming acyl approximately adenylates. Among different oxylipins tested as mixed stereoisomers for substrate activity with JAR1, the highest rate of conversion to Ile-conjugates was observed for (+/-)-JA and 9,10-dihydro-JA, while the rate of conjugation with 12-hydroxy-JA and OPC-4 (3-oxo-2-(2Z-pentenyl)cyclopentane-1-butyric acid) was only about 1-2% that for (+/-)-JA. Of the two stereoisomers of JA, (-)-JA and (+)-JA, rate of synthesis of the former was about 100-fold faster than for (+)-JA. Finally, we have demonstrated that (1) in the presence of ATP, Mg(2+), (-)-JA and tripolyphosphate the ligase produces adenosine 5'-tetraphosphate (p(4)A); (2) addition of isoleucine to that mixture halts the p(4)A synthesis; (3) the enzyme produces neither diadenosine triphosphate (Ap(3)A) nor diadenosine tetraphosphate (Ap(4)A) and (4) Ap(4)A cannot substitute ATP as a source of adenylate in the complete reaction that yields JA-Ile.  相似文献

Increases of 23- (5.6 mmol acetylene reduced mg dry wt–1) and 16- (4 mmol acetylene reduced mg dry wt–1) fold in nitrogenase activity and 12- (671 l H2 mg dry wt–1 h–1) and 6- (349 l mg dry wt–1 h–1) fold in H2 photoproduction in Rhodopseudomonas palustris JA1 over 24 h were achieved with pyrazine 2-carboxylate (3 mM) and 3-picoline (3 mM), respectively, and were higher than earlier reports of enhancement (1.5 to 5- fold) in biological H2 production using various alternative methods.  相似文献

Summary Deficiency of inorganic phosphate caused the hyper production of invertase and the derepression of acid phosphatase in a continuous culture ofSaccharomyces carlsbergensis. The specific invertase activity was 40,000 enzyme units per g dry cell weight at a dilution rate lower than 0.05 h–1 with a synthetic glucose medium of which the molecular ratio of KH2PO4 to glucose was less than 0.006. This activity is eight fold higher than in a batch growth and 1.5 fold as much as the highest enzyme activity observed so far in a glucose-limited continuous culture.For the hyper production of invertase, it is necessary to culture the yeast continuously by keeping the Nyholm's conservative inorganic phosphate concentration at less than 0.2 m mole per g dry weight cell. Bacterial inoculants of the commercially available plant growth promoting rhizobacteria (PGPR) Arthrobacter mysorens 7, Flavobacterium sp. L30, and Klebsiella mobilis CIAM 880 were selected to obtain ecologically safe barley crop production on cadmium (Cd) polluted soils. All the PGPR immobilized 24–68% soluble cadmium from soil suspension. A. mysorens 7 and K. mobilis CIAM 880 were highly resistant to Cd and grew in up to 1 and 3 mmol CdCl2 on DAS medium respectively. All PGPR were able to fix nitrogen (276–1014 nmol mg–1 bacterial DW) and to produce indole acetic acid (IAA) (126–330 nmol mg–1 bacterial DW) or ethylene (4.6–13.5 nmol bacterial DW). All the PGPR actively colonized barley root system and rhizosphere and significantly stimulated root elongation of barley seedlings (up to 25%), growing on soil containing 5 or 15 mg Cd kg–1 of soil. Created in the simulation mathematical model confirms our hypothesis that PGPR beneficial effect on barley growing under Cd-stress is a complex process. One of mechanisms underlying this effect might be increase of bacterial migration from rhizoplane to rhizosphere, where PGPR bind soluble free Cd ions in biologically unavailable complex forms. Among the studied PGPR K. mobilis CIAM 880 was the most effective inoculant. Inoculation with K. mobilis CIAM 880 of barley plants growing on Cd contaminated soil (5 mg Cd kg–1 of soil) under field conditions increased by 120% grain yield and 2-fold decreased Cd content in barley grain. The results suggest that the using K. mobilis CIAM 880 is an effective way to increase the plant yield on poor and polluted areas.  相似文献

Studies of the degradation of the two isomeric forms of butyrate in different anaerobic environments showed isomerization betweenn- andi-butyrate. Degradation rates were similar for the different examined systems and degradation rates forn-butyrate degradation were generally higher than fori-butyrate. Degradation rates forn-butyrate ranged from 0.52 to 1.39 day–1, while the rates fori-butyrate were from 0.46 to 1.15 day–1. Production of isomers was not observed when the volatile fatty acid degradation was inhibited by addition of bromoethane sulfonic acid, indicating that isomerization was coupled to the methanogenic degradation of the acid. The degree of isomerization observed duringn-butyrate degradation was similar to the degree duringi-butyrate degradation. Experiments indicated that the isomerization degree was higher for the thermophilic than for the mesophilic inocula.  相似文献

The death of the hybridoma VO 208 in a continuous culture at pH 7 and 6.8 was investigated by measuring both the appearance of visible dead cells which do not exclude the trypan blue dye and the release of lactate dehydrogenase (LDH) in the culture medium. The intracellular LDH was found to be completely released either when live cells lysed or when they were transformed into visible dead cells. No significant lysis of blue dead cells could be observed at the two different pH. Using a LDH balance over the culture system, cell lysis was found negligible at pH 7, but accounted for 20% of the total cell death at pH 6.8. A methodology is proposed to evaluate the rate constants of hybridoma lysis and total death. A comparison of the binding of (–)- and (+)-[3H]nicotine to rat brain membranes revealed that only the (–)-enantiomer showed high affinity binding; while the (+)-enantiomer was at least 1/10 as effective as the (–)-enantiomer when in competition with (–)-[3H]nicotine as the ligand. Positive cooperativity, which is observed with (–)-[3H]nicotine as the presence of low concentrations of (+)-nicotine, may account for the seeming paradox.  相似文献

The application of abscisic acid (ABA), either as a racemic mixture or as optically resolved isomers, increases freezing tolerance in a bromegrass (Bromus inermis Leyss) cell culture and induces the accumulation of several heat-stable proteins. Two stereoisomers of an ABA analog, 23 dihydroacetylenic abscisyl alcohol (DHA), were used to study the role of ABA-induced processes in the acquisition of freezing tolerance in these cells. Freezing tolerance was unchanged in the presence of (–) DHA (LT50 -9°C), and no increase in heat-stable protein accumulation was detected; however, the (+) enantiomer increased the freezing tolerance (LT50 -13°C) and induced the accumulation of these polypeptides. All three forms of ABA increased freezing tolerance in the bromegrass cells, although (–) ABA was less effective than either (+) or (±) ABA when added at equal concentrations. Cells pretreated with 20 or 50 M (–) DHA displayed lower levels of freezing tolerance following the addition of 2.5, 7.5 or 25 M (±) ABA. Full freezing tolerance could be restored by increasing the concentration of (±) ABA to > 25 M. Pretreatment of cells with (–) DHA (20 or 50 M) had no effect on freezing tolerance when 25 M (+) ABA was added. The induction of freezing tolerance by 25 M (–) ABA was completely inhibited by the presence of 20 M (–) DHA. The accumulation of ABA-responsive heat-stable proteins was inhibited by pretreatment with 20 M (–) DHA in cells treated with 2.5 or 7.5M (+) ABA, and in cells treated with 25 M (–) ABA. The accumulation of these polypeptides was restored when (±) or (+) ABA was added at a concentration of 25 M. The analysis of proteins which cross-reacted with a dehydrin antibody revealed a similar inhibitory pattern as seen with the other ABA-responsive proteins. The effects of the various isomers of ABA and DHA on cell osmolarity and sucrose uptake was also investigated. In both cases, (±) and (+) ABA had pronounced effects on the parameters measured, whereas (–) ABA treated cells gave substantially different results. In both sucrose uptake and cell osmolarity, DHA had no significant effect on the results obtained following (±) or (+) ABA treatment. The fate of nitrogen from15N-labelled sheep manure and ammonium sulfate in small lysimeters and plots in the field was studied during two growth seasons. In April 1991,15N-labelled sheep faeces (87 kg N ha–1) plus unlabelled (NH4)2SO4 (90 kg N ha–1), and (15NH4)2SO4 (90 kg N ha–1) were each applied to three soils; soil 1 (100% soil + 0% quartz sand), soil 2 (50% soil + 50% quartz sand) and soil 3 (25% soil + 75% quartz sand). The lysimeters were cropped with spring barley (Hordeum vulgare L.) and undersown ryegrass (Lolium perenne L.). The barley crop recovered 16–17% of the labelled manure N and 56% of the labelled (NH4)2SO4-N. After 18 months 30% of the labelled manure N and 65% of the labelled (NH4)2SO4-N were accumulated in barley, the succeeding ryegrass crop and in leachate collected below 45 cm of soil, irrespective of the soil-sand mixture. Calculating the barley uptake of manure N by difference of N uptake between manured and unmanured soils, indicated that 4%, 10% and 14% of the applied manure N was recovered in barley grown on soil-sand mixtures with 16%, 8% and 4% clay, respectively. The results indicated that the mineralization of labelled manure N was similar in the three soil-sand mixtures, but that the manure caused a higher immobilization of unlabelled ammonium-N in the soil with the highest clay content. Some of the immobilized N apparently was remineralized during the autumn and the subsequent growth season. After 18 months, 11–19% of the labelled manure N was found in the subsoil (10–45 cm) of the lysimeters, most of this labelled N probably transported to depth as organic forms by leaching or through the activities of soil fauna. In unplanted soils 67–74% of the labelled manure N was recovered in organic form in the 0–10 cm soil layer after 4 months, declining to 55–64% after 18 months. The lowest recovery of labelled N in top-soil was found in the soil-sand mixture with the lowest clay content. The mass balance of15N showed that the total recovery of labelled N was close to 100%. Thus, no significant gaseous losses of labelled N occurred during the experiment.  相似文献

Uptake of 3H-labelled (±)-abscisic acid (ABA) into isolated barley (Hordeum vulgare L.) epidermal cell protoplasts (ECP) was followed over a range of pH values and ABA concentrations. The present results show that ABA uptake is not always linearly correlated with the external concentration of undissociated ABA (ABAH). At pH 7.25, ABA uptake exhibited saturation kinetics with an apparent K m value of 75 mmol·m–3 to tal ABA. This saturable transport component was inhibited by pretreating the protoplasts with 1 mol·m–3 p-chloromercuribenzenesulfonic acid at pH 8.0, conditions that minimized the uptake of this acid sulfhydryl reagent. Moreover, the rate of (±)-[3]HABA uptake was reduced by addition of 0.1 mol·m–3 (±)-ABA to 41%, whereas the same concentration of (±)-ABA was approximately half as effective (46% of the inhibitory effect). Thus, it was concluded that only (±)-ABA competes for an ABA carrier that is located in the epidermal cell plasma membrane. The permeability of the epidermal cell plasma membrane was studied by performing a Collander analysis. High-resolution growth measurements were conducted using a linear variable displacement transformer in conjunction with a temperature-programmed meristem-cooling collar. Chilling and rewarming profiles were determined for a range of Gramineae, in the presence and absence of varying concentrations of gibberellic acid (GA3). In wheat (Triticum aestivum L.) seedlings, the growth-constraining temperature (Pe) was progressively lowered by increasing GA3 concentration, with a difference of-4.8°C between controls and material treated with 10–4 M GA3. Dwarf-5 maize (Zea mays L.) seedlings had a higher Pe than tall segregates and the difference was markedly reduced by exposure to a saturating concentration of GA3. A similar effect was observed with Tanginbozu dwarf rice (Oryza sativa L.). The growth ratetemperature responses of Rht3 gibberellin-insensitive dwarf wheat seedlings were unaffected by GA3 and the Pe values for these segregates were around 5° C higher than for normals. Slender (s1) barley (Hordeum vulgare L.) genotypes had Pe values of-7° C, compared with +4° C for wild-type material, and did not show positive hysteresis for growth rate during the rewarming phase.
