A comparison of mycelial extracts of 17 isolates of Penicillium nodositatum collected from the root system of grey alder (Alnus incana (L.) Moench.) and Italian alder (A. cordata Desf.) grown at different sites was made using isoenzyme analysis of esterases, acid phosphatase, malate dehydrogenase and phosphoglucose isomerase. Jaccard similarity coefficient analysis demonstrated that interspecific variability existed both among and within sites. In most cases, the enzyme patterns of isolates were distinct. Comparison of these isolates and three other Penicillium species closely related to P. nodositatum showed no similarity for the enzymes tested.  相似文献

The taxonomy of the Penicillium chrysogenum series is reconsidered. On account of the observations of the available type strains and numerous isolates mainly obtained from food products, Penicillium notatum Westling, P. meleagrinum Biourge and P. cyaneofulvum Biourge are placed in synonymy with P. chrysogenum Thom. Interest in species of the genus Penicillium is related to their ability to produce the mycotoxin patulin and to cause spoilage of fruit products worldwide. The sequence of the isoepoxydon dehydrogenase (idh) gene, a gene in the patulin biosynthetic pathway, was determined for 28 strains representing 12 different Penicillium species known to produce the mycotoxin patulin. Isolates of Penicillium carneum, Penicillium clavigerum, Penicillium concentricum, Penicillium coprobium, Penicillium dipodomyicola, Penicillium expansum, Penicillium gladioli, Penicillium glandicola, Penicillium griseofulvum, Penicillium paneum, Penicillium sclerotigenum and Penicillium vulpinum were compared. Primer pairs for DNA amplification and sequencing were designed from the P. griseofulvum idh gene (GenBank AF006680). The two introns present were removed from the nucleotide sequences, which were translated to produce the IDH sequences of the 12 species for comparison. Phylogenetic relationships among the species were determined from rDNA (ITS1, 5.8 S, ITS2 and partial sequence of 28S rDNA) and from the idh nucleotide sequences minus the two introns. Maximum parsimony analysis showed trees based on rDNA and idh sequences to be congruent. It is anticipated that the genetic information obtained in the present study will aid in the design of probes, specific for patulin biosynthetic pathway genes, to identify the presence of these mycotoxigenic fungi. Inteins are protein-intervening sequences found inside the coding region of different host proteins and are translated in-frame with them. They can self-excise through protein splicing, which ligates the host protein flanks with a peptide bond. In this study, four different species of the genus Penicillium were investigated for the presence of inteins inside the conserved splicing-factor protein PRP8. We identified 157 to 162 amino acid in-frame insertions in the PRP8 protein of Penicillium chrysogenum, Penicillium expansum, and Penicillium vulpinum (formerly Penicillium claviforme). The Penicillium PRP8 inteins are mini-inteins without a conserved endonuclease domain. We demonstrated that the PRP8 mini-inteins of P. chrysogenum, P. expansum, and P. vulpinum undergo autocatalytic protein splicing when heterologously expressed in E. coli, in a model host protein, and in a divided GFP model system. They are, thus, among the smallest known nuclear-encoded, active splicing protein elements. The GFP assay should be valuable as a screening system for protein splicing inhibitors as potential antimycotic agents and as tools for studying the mechanism of protein splicing of fungal mini-inteins.  相似文献

Penicillium species, well-known indoor airborne fungi, are considered to be important causative agents of extrinsic bronchial asthma. For selecting the proper species for clinical investigations, it is important to know which are found indoors. In the present study, 176 plates of modified Mehrlich's Medium were exposed for 20 min in living rooms and bedrooms of 88 homes in Taipei, Taiwan. A total of 5897 colonies of fungi were isolated, varying from one to 329 per house. Of the total, 910 colonies (15.4%) were identified asPenicillium spp. The three most frequently encountered species wereP. citrinum, P. crustosum andP. implicatum. Penicillium citrinum accounted for 40.5% (369 from 28 houses);P. implicatum, for 5.2% (47 from three houses); andP. crustosum, for 2.1% (19 from seven houses).Penicillium citrinum was judged to be the most prevalent airborne fungus in Taiwan. The allergenic significance of this mold is under further investigation.  相似文献

Aims: To assess the ability of fungi isolated from grapes to produce patulin and citrinin. Methods and Results: A total of 446 Aspergillus isolates belonging to 20 species and 101 Penicillium isolates were inoculated in Czapek yeast extract agar and yeast extract sucrose agar and incubated for 7 days at 25°C. Extracts were analysed for patulin and citrinin by thin‐layer chromatography. None of the isolates of Aspergillus spp. produced either patulin or citrinin. Patulin was produced by three isolates of Penicillium expansum and two of Penicillium griseofulvum. Citrinin was produced by five isolates of P. expansum, two of Penicillium citrinum and one of Penicillium verrucosum. Conclusions: Our results show that the Aspergillus and Penicillium species commonly isolated from grapes are not a source of the mycotoxins, patulin and citrinin. Significance and Impact of the Study: The possibility of co‐occurrence of patulin and citrinin with ochratoxin A in grapes and grape products remain low, owing to the low frequency of isolation of potentially producing species.  相似文献

Four of them have been recovered from the air in Madrid, Spain, one from beech leaves litter in a beech forest near Nancy, France, two from sandy soils in Madrid, and one from an andosol in the Spanish Province of Navarra. A new monoverticillate Penicillium species (subgenus Aspergilloides Dierckx), Penicillium viticola, was isolated from a grape cultivated in Yamanashi Prefecture, Japan. Morphologically, P. viticola is characterized by the production of slightly roughened conidia, slightly roughened stipes, and rapid growth on 25% glycerol nitrate agar (G25N). This species is phylogenetically close to Penicillium angulare S.W. Peterson, E.M. Bayer & Wicklow, but differs with respect to colonial characteristics and conidia and penicilli morphology.  相似文献

β-lactam antibiotics (e.g. penicillins, cephalosporins) are of major clinical importance and contribute to over 40% of the total antibiotic market. These compounds are produced as secondary metabolites by certain actinomycetes and filamentous fungi (e.g. Penicillium, Aspergillus and Acremonium species). The industrial producer of penicillin is the fungus Penicillium chrysogenum. The enzymes of the penicillin biosynthetic pathway are well characterized and most of them are encoded by genes that are organized in a cluster in the genome. Remarkably, the penicillin biosynthetic pathway is compartmentalized: the initial steps of penicillin biosynthesis are catalyzed by cytosolic enzymes, whereas the two final steps involve peroxisomal enzymes. Here, we describe the biochemical properties of the enzymes of β-lactam biosynthesis in P. chrysogenum and the role of peroxisomes in this process. An overview is given  相似文献

Penicillium roqueforti, P. crustosum and P. paneum grow on ensiled grain and recycled feed unless properly treated. The former two species occur also on cut lumber in Canada. These are known to produce a number of secondary metabolites including roquefortine. In cooler dairy production areas, including Scandinavia and North America, cattle toxicosis has been associated with silage contaminated by these fungi. We collected strains associated with cow or cattle toxicoses. The principal metabolites were determined making use of a new extraction method and analysis combining HPLC, LC/MS/MS, and LC/NMR. Penicillium roqueforti and P. crustosum required amino acid nitrogen for metabolite formation and their toxins were formed under conditions of low oxygen (20–30% saturation). Production of roquefortine C occurred on depletion of the available nitrogen and penitrem A on depletion of carbon source. Yield was reduced by excess carbon. Medium osmotic tension (aw) affected metabolite production by the two species differently. Penicillium paneum was associated with ill-thrift of dairy cows and P. roqueforti was associated with more serious symptoms. Our data suggest a physiological basis for the common occurrence of roquefortine C in silage without serious consequences and the alternative, the presence of roquefortine C and toxicoses. The strain isolated from lumber was the best producer of the toxins studied. This is the first report of the toxigenic potential of P. roqueforti and P. paneum from Canada.  相似文献

Penicillium glabrum, P. purpurescens, P. decumbens, P. expansum, P. chrysogenum, P. crustosum andP. aurantiogriseum are responsible for some of the alterations noticed on Moroccan table grapes cold stored. Contamination of these table grapes withPenicillium species occurs in the vineyard and inside the cooling station where other fruits which are often fungus-contaminated are also kept. All of thesePenicillium species were able to grow at 0°C apart fromP. glabrum andP. purpurescens. Consequently, refrigeration of grapes during long-term storage is not sufficient in itself in preserving their initial qualities.  相似文献

The type strains Penicillium clavigerum VKM F-447 and P. commune VKM F-3233 are found to produce fumigaclavines A and B. Of the seven other strains of these species, only two strains, P. commune VKM F-3088 and F-3491, possess the ability to synthesize these alkaloids. It is suggested that the five other strains under study either lost such an ability or require very specific conditions for the synthesis of these alkaloids.  相似文献

A study of the mycobiota in the digestive tract of 5 important species of triatomines, Triatoma brasiliensis, T. infestans, T. sordida, T. pseudomaculata and T. vitticeps, was made. The digestive tracts of 164 adults and 535 nymphs of those triatomines were studied and 393 fungal strains were isolated.The genera with the greatest number of species were Penicillium (19 species), Aspergillus (17 species) and Acremonium (5 species) and the most frequent species, in decreasing order, were Penicillium corylophilum, Aspergillus niger, Penicillium fellutanum, Cladosporium herbarum, Penicillium waksmanii, Aspergillus awamori and Paecilomyces variotii. Among the isolated fungi, we found species that are recognized as entomopathogenic and pathogenic for humans and animals.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Production of fungitoxic extrolites was evaluated in culture filtrates of several isolates belonging to Penicillium canescens and P. janczewskii that showed some extent of inhibitory activity against the plant pathogenic fungus Rhizoctonia solani. In addition to griseofulvin and dechlorogriseofulvin that are already known in these species, curvulinic acid, previously unreported in Penicillium, was produced by all isolates assayed. Another extrolite recently characterized from a P. verrucosum strain by the name of Sch 642305 was detected in 5 isolates of P. canescens only. The purified compound completely inhibited mycelial growth of isolates of Rhizoctonia solani and other plant pathogenic fungi in␣vitro. The role of this extrolite as a possible biochemical determinant of antagonism toward plant pathogenic fungi, and implications concerning chemotaxonomy are discussed.  相似文献
