Microorganisms are mainly responsible for the transformation and mineralization of degradable organic pollutants within constructed wetlands (CWs). There is still a lack of knowledge concerning microbial community composition within CWs. In order to elucidate the diversity of bacteria inhabiting subsurface vertical flow CWs, the molecular fingerprint technique “terminal restriction fragment length polymorphism” (T-RFLP) derived from total community DNA, was applied.A comparison of the bacterial communities from a full-scale outdoor vertical flow CW with planted and unplanted indoor pilot-scale vertical flow CWs, operated under similar conditions, revealed that both systems are colonized by similar populations showing only little variation in their composition over filter depth. A comparison of bulk soil from an unplanted CW with the rhizosphere soil from the outdoor and indoor CWs showed differences in the bacterial composition, demonstrating the influence of the plants on the rhizosphere community. A comparison of the wastewater before and after the CW passage demonstrated that the bacterial diversity was clearly reduced within the planted outdoor system only.  相似文献

This study examined the performance of pilot-scale vertical subsurface flow constructed wetlands (VSF–CWs) planted with three indigenous plants, i.e. Typha latifolia, Cyperus alternifolius, and Cynodon dactylon, in removing heavy metals from secondary treated refinery wastewater under tropical conditions. The T. latifolia-planted VSF–CW had the best heavy metal removal performance, followed by the Cyperus alternifolius-planted VSF–CW and then the Cynodon dactylon-planted VSF–CW. The data indicated that Cu, Cr, Zn, Pb, Cd, and Fe were accumulated in the plants at all the three VSF–CWs. However, the accumulation of the heavy metals in the plants accounted for only a rather small fraction (0.09–16%) of the overall heavy metal removal by the wetlands. The plant roots accumulated the highest amount of heavy metals, followed by the leaves, and then the stem. Cr and Fe were mainly retained in the roots of T. latifolia, Cyperus alternifolius, and Cynodon dactylon (TF < 1), meaning that Cr and Fe were only partially transported to the leaves of these plants. This study showed that VSF–CWs planted with T. latifolia, Cyperus Alternifolius, and Cynodon dactylon can be used for the large-scale removal of heavy metals from secondary refinery wastewater.  相似文献

In Northern Ireland, phosphorus enrichment of lakes due to agriculture is a significant problem. Heavy metal exports from landfill sites, often located on water-logged land, are also of concern. Locally available laterite, a low grade bauxite which is rich in iron and aluminium, is used in acid solution with subsequent precipitation to remove phosphorus and heavy metals at several sewage treatment works. Constructed wetlands offer an attractive alternative to conventional waste water treatment in certain circumstances but removal of phosphorus is strongly dependent on the bed medium. Calcium-, iron- and aluminium-rich solid media are recommended. A brief introduction to the use and cost-effectiveness of constructed wetlands (CWs) in treating a range of effluents is given. This study, using both laboratory tests and pilot-scale constructed wetlands, reports the effectiveness of granular laterite in removing phosphorus and heavy metals from landfill leachate. Initial laboratory studies have shown that laterite is capable of 99% removal of phosphorus from solution. A pilot-scale experimental CW containing laterite achieved 96% removal of phosphorus. This removal is much greater than that reported in other systems. Initial removals of aluminium and iron by pilot-scale CWs have been up to 85% and 98% respectively. Percolating columns of laterite reduced Cd, Cr and Pb to undetectable concentrations. Possible application of this low cost, low technology, visually unobtrusive yet efficient system to rural areas with dispersed point sources of pollution is discussed.  相似文献

Ferric-Carbon Micro-Electrolysis (Fe/C-M/E) material had been widely used for the pretreatment of wastewater. Therefore, we hypothesized that Fe/C-M/E material could enhance the treatment of domestic sewage when it was integrated into constructed wetlands (CWs). In this study, CWs integrated with Fe/C-M/E material were developed. Druing the experiment of effect of vegetation on the performance of CWs, percentages of NH4+-N, NO3?-N, total nitrogen (TN), and Chemical Oxygen Demand (COD) removed in polyculture (W1) were up to 91.8%, 97.0%, 92.3%, and 85.4%, respectively, which were much higher than those in Lythrum salicaria monoculture (W2) and Canna indica monoculture (W3). In the experiment of temperature influences on the removal efficiency of CWs, temperature substantially influenced the performance of CWs. For example, NO3?-N removal percentages of W1, W2, and W3 at high temperature (25.5°C and 19.8°C) were relatively stable and greater than 85.4%. At 8.9°C, however, a sharp decline of NO3?-N removal percentage was observed in all CWs. Temperature also influenced the Chemical Oxygen Demand (COD) removal and soil microbial activity and biomass. Overall, the polyculture (Lythrum salicaria +Canna indica) showed the best performance during most of the operating time, at an average temperature ≥ 19.8°C, due to the functional complementarity between vegetation. All the CWs consistently achieved high removal efficiency (above 96%) for TP in all experiments, irrespective of vegetation types, phosphorous loadings, and temperatures. In conclusion, polyculture was an attractive solution for the treatment of domestic sewage during most of the operating time (average temperature ≥ 19.8°C). Furthermore, CWs with Fe/C-M/E material were ideally suitable for domestic sewage treatment, especially for TP removal.  相似文献

The Fe(III)/Fe(II) couple can play a significant role in the abiotic reduction of 2-nitrophenol (2-NP) at the cathode chamber of a microbial fuel cell (MFC). Experimental results demonstrate that Fe(II) addition to the cathode chamber contributes to a significant increase in the reaction rate of 2-NP removal and the power performance of MFC. Observed pseudo first-order rate constants and power densities are heavily dependent on the identity of the Fe(II)-complexing ligands. The Fe(II) complex coordinated with citrate results in the highest rate constant up to 0.12 h−1 as compared to other organically complexed iron species including Fe(II)-EDTA, Fe(II)-acetate and Fe(II)-oxalate, and iron species uncomplexed with any organic ligands. In addition, the presence of Fe(II)-citrate species leads to a maximum volumetric power density of 1.0 W m−3, which is the highest value among those obtained with other iron species for the similar MFC system.  相似文献

The performance and reliability of the CWM1-RETRASO model for simulating processes in horizontal subsurface flow constructed wetlands (HSSF CWs) and the relative contribution of different microbial reactions to organic matter (COD) removal in a HSSF CW treating urban wastewater were evaluated. Various different approaches with diverse influent configurations were simulated. According to the simulations, anaerobic processes were more widespread in the simulated wetland and contributed to a higher COD removal rate [72-79%] than anoxic [0-1%] and aerobic reactions [20-27%] did. In all the cases tested, the reaction that most contributed to COD removal was methanogenesis [58-73%]. All results provided by the model were in consonance with literature and experimental field observations, suggesting a good performance and reliability of CWM1-RETRASO. According to the good simulation predictions, CWM1-RETRASO is the first mechanistic model able to successfully simulate the processes described by the CWM1 model in HSSF CWs.  相似文献

This paper describes the development and operation of an Environmental Decision Support System (EDSS) to improve the operation and maintenance of horizontal subsurface-flow constructed wetlands (EDSS-maintenance). Constructed wetlands (CWs) allow wastewater treatment in a sustainable manner since they involve low energy consumption, low construction and functioning costs and low environmental impact. However, operation and maintenance activities are essential to guarantee reliability in CWs performance. The definition of operation and maintenance protocols depends on several quantitative and qualitative aspects such as wastewater treatment plant configuration, CW design, influent characteristics, sensitivity of the receiving media, etc. Bearing this in mind and considering the limited technical knowledge about CWs, the need for a new tool to support CW performance is clear. In this sense, EDSSs offer a new approach because they can tackle problems of complex and uncertain systems. The EDSS-maintenance provides operation and maintenance manuals specifically defined for every CW. To achieve it, the required knowledge was implemented within a rule-based system, which forms the backbone of the EDSS. Several features presented in this paper demonstrate how the EDSS-maintenance provides a proper platform to support the necessary collaborative work in the ecological engineering problem of horizontal subsurface flow CWs operation and maintenance.  相似文献

In rural areas, insufficient wastewater treatment often causes serious environmental problems, especially for human health. Due to this problem, new wastewater treatment plants (WWTP) should be constructed or upgraded. When considering economic and maintenance factors, constructed wetlands (CWs) are more desirable processes. The available WWTP located in the Northern Israeli town of Sakhnin was redesigned and upgraded. To rehabilitate the WWTP, six new CWs having different operating conditions were constructed as a pilot project. The most appropriate place of construction of the CWs was thought to be at the end of the WWTP. From the beginning of the system, inlet and outlet pollution parameters such as chemical oxygen demand (COD), ammonia (NH4-N+), total suspended solids (TSS), and phosphorus (PO4-P3−) were monitored in the CWs from August 2005 to February 2006. As a result the most appropriate CWs were found having Phragmites as plant and volcanic tufa as media material. The maximum removal efficiencies were 71.8% on COD, 92.9% on TSS, 63.8% on ammonia for CW5 tank. Adversely, phosphorus removal was not so high in CW5. Phosphorus removal was negligible in the study.  相似文献

Metallic engineered nanomaterials (ENMs) undergo various transformations in the environment which affect their fate, toxicity and bioavailability. Although constructed wetlands (CWs) are applied as treatment systems for waste streams potentially containing metallic ENMs, little is known about the fate and effects of ENMs in CWs. Hence, literature data from related fields such as activated sludge wastewater treatment and natural wetlands is used to predict the fate and effects of ENMs in CWs and to analyze the risk of nanomaterials being released from CWs into surface waters. The ENMs are likely to reach the CW (partly) transformed and the transformations will continue in the CW. The main transformation processes depend on the type of ENM and the ambient environmental conditions in the CW. In general, ENMs are expected to undergo sorption onto (suspended) organic matter and plant roots. Although the risk of ENMs being released at high concentrations from CWs is estimated low, caution is warranted because of the estimated rise in the production of these materials. As discharge of (transformed) ENMs from CWs during normal operation is predicted to be low, future research should rather focus on the effects of system malfunctions (e.g. short-circuiting). Efficient retention in the CW and increasing production volumes in the future entail increasing concentrations within the CW substrate and further research needs to address possible adverse effects caused.  相似文献

According to the characteristics of the reverse osmosis concentrate (ROC) generated from iron and steel company, we used three sets of parallel horizontal subsurface flow (HSF) constructed wetlands (CWs) with different plants and substrate layouts to treat the high-salinity wastewater. The plant growth and removal efficiencies under saline condition were evaluated. The evaluation was based entirely on routinely collected water quality data and the physical and chemical characteristics of the plants (Phragmites australis, Typha latifolia, Iris wilsonii, and Scirpus planiculmis). The principal parameters of concern in the effluent were chemical oxygen demand (COD), total nitrogen (TN), and total phosphorus (TP). The results showed that the CWs were able to remove COD, TN, and TP from ROC. S. planiculmis was not suitable for the treatment of high-saline wastewater. The sequence of metals accumulated in CW plants was K>Ca>Na>Mg>Zn>Cu. More than 70% of metals were accumulated in the aboveground of P. australis. The CW filled with gravel and manganese ore and planted with P. australis and T. latifolia had the best performance of pollutant removal, with average removal of 49.96%, 39.45%, and 72.01% for COD, TN, and TP, respectively. The effluent water quality met the regulation in China. These results suggested that HSF CW planted with P. australis and T. latifolia can be applied for ROC pollutants removal.  相似文献

Mercury sulfides (cinnabar and metacinnabar) are the main ores of Hg and are relatively stable under oxic conditions (Ksp = 10?54 and 10?52, respectively). However, until now their stability in the presence of micro‐organisms inhabiting acid mine drainage (AMD) systems was unknown. We tested the effects of the AMD microbial community from the inoperative Hg mine at New Idria, CA, present in sediments of an AMD settling pond adjacent to the main waste pile and in a microbial biofilm on the surface of this pond, on the solubility of crystalline HgS. A 16S rRNA gene clone library revealed that the AMD microbial community was dominated by Fe‐oxidizing (orders Ferritrophicales and Gallionellas) and S‐oxidizing bacteria (Thiomonas sp.), with smaller amounts (≤6%) being comprised of the orders Xanthomondales and Rhodospirillales. Though the order Ferritrophicales dominate the 16S rRNA clones (>60%), qPCR results of the microbial community indicate that the Thiomonas sp. represents ~55% of the total micro‐organisms in the top 1 cm of the AMD microbial community. Although supersaturated with respect to cinnabar and metacinnabar, microcosms inoculated with the AMD microbial community were capable of releasing significantly more Hg into solution compared to inactivated or abiotic controls. Four different Hg‐containing materials were tested for bacterially enhanced HgS dissolution: pure cinnabar, pure metacinnabar, mine tailings, and calcine material (processed ore). In the microcosm with metacinnabar, the presence of the AMD microbial community resulted in an increase of dissolved Hg concentrations up to 500 μg L‐1 during the first 30 days of incubation. In abiotic control microcosms, dissolved Hg concentrations did not increase above 100 ng L?1. When Hg concentrations were below 50 μg L‐1, the Fe‐oxidizing bacteria in the AMD microbial community were still capable of oxidizing Fe(II) to Fe(III) in the AMD solution, whereas concentrations above 50 μg L?1 resulted in inhibition of microbial iron oxidation. Our experiments show that the AMD microbial community contributes to the dissolution of mercury sulfide minerals. These findings have major implications for risk assessment and future management of inoperative Hg mines worldwide.  相似文献

Anaerobic ammonium oxidation (ANAMMOX) may provide an effective nitrogen removal pathway for constructed wetlands with low C/N influent. In a study of domestic sewage treatment, anaerobic ammonium oxidation process was identified in the pilot-scale constructed wetland of a bio-ecological process which was composed of a bio-contact oxidation reactor and a horizontal subsurface flow constructed wetland (CW). To investigate the ANAMMOX establishment in the bio-ecological process, two new CWs (planted and unplanted) were developed to be a control for the pre-existing CW. Under operational conditions of DO 2-3 mg/l, HRT 3.5 h for the bio-contact oxidation reactor, HRT 3 days for CWs, and domestic sewage as influent, the process achieved more than 90% TN removal rate after the ANAMMOX was established. The ANAMMOX bacteria on the media of the constructed wetlands were analyzed by specific polymerase chain reaction (PCR) with ANAMMOX specific primer set AMX818F-AMX1066R. The result of the genetic sequencing showed that the PCR product was related to Candidatus B. anammoxidans (AF375994.1) with 98% sequence similarity. Copy numbers of 16S rRNA gene of ANAMMOX bacteria in the pre-existing CW, the new planted CW and new unplanted CW were 3.47 × 105, 3.02 × 105 and 1.30 × 105, respectively. These results demonstrated that the ANAMMOX process was successfully established and operated consistently in the constructed wetlands with a bio-contact oxidation reactor as a pretreatment, and that vegetation positively affected the growth and enrichment of ANAMMOX bacteria.  相似文献

An integrated wetland system (IWS) including constructed wetlands (CWs) and modified natural wetlands (NWs) for wastewater treatment to replenish water to wetlands located at the Beijing Wetland School (BWS) in Beijing, China, is presented in this paper. The synergistic effects of CWs and NWs on treated water quality are investigated. The IWS is proved to be an effective wastewater treatment technique and a better alternative to alleviate the water shortage for conservation of wetlands based on the monitoring data obtained from October 2007 to 2008. The results show that CWs and NWs play different roles in removing contaminants from wastewater. The COD removal efficiency in CWs is higher than that in modified NWs, whereas the modified NWs can compensate for the deficiency of CWs where a stable and sufficient rhizosphere is not fully formed in the start-up period. All removal rates of COD, TN, and TP in CWs and modified NWs vary from 50 to 70%, while the total removal rate of COD, TN, and TP in IWS is about 85–90%. The operational results show that the maximum area loading of organic pollutants in modified NWs (65 kg/ha d) is slightly higher than the empirical one (60 kg/ha d) recommended by USEPA (2000) for free water surface wetlands.  相似文献

Azo dyes are commonly generated as effluent pollutants by dye using industries, causing contamination of surface and ground water. Various strategies are employed to treat such wastewater; however, a multi-faceted treatment strategy could be more effective for complete removal of azo dyes from industrial effluent than any single treatment. In the present study, rice husk material was used as a substratum in two constructed wetlands (CWs) and augmented with microorganisms in the presence of wetland plants to effectively treat dye-polluted water. To evaluate the efficiency of each process the study was divided into three levels, i.e., adsorption of dye onto the substratum, phytoremediation within the CW and then bioremediation along with the previous two processes in the augmented CW. The adsorption process was helpful in removing 50% dye in presence of rice husk while 80% in presence of rice husk biocahr. Augmentation of microorganisms in CW systems has improved dye removal efficiency to 90%. Similarly presence of microorganisms enhanced removal of total nitrogen (68% 0 and Total phosphorus (75%). A significant improvement in plant growth was also observed by measuring plant height, number of leaves and leave area. These findings suggest the use of agricultural waste as part of a CW substratum can provide enhanced removal of textile dyes.  相似文献

The aim of this study was to investigate the removal of ibuprofen in laboratory scale constructed wetlands. Four (planted and unplanted) laboratory‐scale horizontal subsurface flow constructed wetlands were supplemented with ibuprofen in order to elucidate (i) the role of plants on ibuprofen removal and (ii) to evaluate the removal performance of a bioaugmented lab scale wetland. The planted systems showed higher ibuprofen removal efficiency than an unplanted one. The system planted with Juncus effusus was found to have a higher removal rate than the system planted with Phalaris arundinacea. The highest removal rate of ibuprofen was found after inoculation of gravel previously loaded with a newly isolated ibuprofen‐degrading bacterium identified as Sphingobium yanoikuyae. This experiment showed that more than 80 days of CW community adaptation for ibuprofen treatment could be superseded by bioaugmentation with this bacterial isolate.  相似文献

Lab‐scale experiments were conducted to investigate the effects of ferrous iron on nutrient removal performance and variations in the microbial community inside aerobic granular sludge for 408 days. Two reactors were simultaneously operated, one without added ferrous iron (SBR1), and one with 10 mg Fe2+ L?1 of added ferrous iron (SBR2). A total of 1 mg Fe2+ L?1 of added ferrous iron was applied to SBR1 starting from the 191st day to observe the resulting variations in the nutrient removal performance and the microbial community. The results show that ammonia‐oxidizing bacteria (AOB) could not oxidize ammonia due to a lack of iron compounds, but they could survive in the aerobic granular sludge. Limited ferrous iron addition encouraged nitrification. Enhanced biological phosphorus removal (EBPR) from both reactors could not be maintained regardless of the amount of ferrous iron that was applied. EBPR was established in both reactors when the concentration of mixed liquor suspended solid (MLSS) and the percentage of Accumulibacteria increased. A total of 10 mg Fe2+ L?1 of added ferrous iron had a relatively adverse effect on the growth of AOB species compared to 1 mg Fe2+ L?1 of added ferrous iron, but it encouraged the growth of Nitrospira sp. and Accumulibacteria, which requires further study. It could be said that the compact and stable structure of aerobic granular sludge preserved AOB and NOB from Fe‐deficient conditions, and wash‐out during the disintegration period. © 2017 American Institute of Chemical Engineers Biotechnol. Role of phosphate in initial iron deposition in apoferritin   总被引:1,自引:0,他引:1
Y G Cheng  N D Chasteen 《Biochemistry》1991,30(11):2947-2953
Ferritins from microorganisms to man are known to contain varying amounts of phosphate which has a pronounced effect on the structural and magnetic properties of their iron mineral cores. The present study was undertaken to gain insight into the role of phosphate in the early stages of iron accumulation by ferritin. The influence of phosphate on the initial deposition of iron in apoferritin (12 Fe/protein) was investigated by EPR, 57Fe M?ssbauer spectroscopy, and equilibrium dialysis. The results indicate that phosphate has a significant influence on iron deposition. The presence of 1 mM phosphate during reconstitution of ferritin from apoferritin, Fe(II), and O2 accelerates the rate of oxidation of the iron 2-fold at pH 7.5. In the presence or absence of phosphate, the rate of oxidation at 0 degrees C follows simple first-order kinetics with respect to Fe(II) with half-lives of 1.5 +/- 0.3 or 2.8 +/- 0.2 min, respectively, consistent with a single pathway for iron oxidation when low levels of iron are added to the apoprotein. This pathway may involve a protein ferroxidase site where phosphate may bind iron(II), shifting its redox potential to a more negative value and thus facilitating its oxidation. Following oxidation, an intermediate mononuclear Fe(III)-protein complex is formed which exhibits a transient EPR signal at g' = 4.3. Phosphate accelerates the rate of decay of the signal by a factor of 3-4, producing EPR-silent oligonuclear or polynuclear Fe(III) clusters. In 0.5 mM Pi, the signal decays according to a single phase first-order process with a half-life near 1 min.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
