Tritiated ribosomal RNA (rRNA) was prepared from hypocotyls of Phaseolus coccineus grown in liquid culture in the dark and in presence of 5-3H-uridine. A mixture of the 18S and 25S 3H-rRNA fractions was used for hybridization with DNA in the polytene chromosome cells of the embryo suspensor of P. coccineus. It was shown that the ribosomal cistrons (rDNA) are located in the nucleolus organizing system (satellite, nucleolar constriction and organizer) of the satellited chromosome pairs I (S1) and V (S2), in the proximal heterochromatic segment of the long arm of chromosomes S1 and in the terminal heterochromatic segment of chromosome pair II. The micronucleoli which are produced by the satellite and nucleolus organizer of the chromosome pair S1 contain rDNA; on the contrary, no rRNA-DNA hybridization is found in the DNA containing granules which are produced by the satellite and nucleolus organizer of chromosome pair S2. The DNA which is amplified during production of DNA puffs at some chromosomal regions apparently does not code for ribosomal RNA (no detectable rRNA-DNA hybridization).Publication no. 62 from the Laboratorio di Mutagenesi e Differenziamento, Consiglio Nazionale delle Ricerche, Pisa. Summary DNA was extracted from three root segments ofAllium cepa: i) an apical portion 500 m long from the tip (meristem); ii) a second portion 4 mm long (I root segment containing metaxylem cells in the initial stages of differentiation); iii) a third portion 6 mm long (II root segment containing metaxylem cells in further stages of differentiation). A mixture of homologous 18 S and 25 S3H-rRNA was used for invitro DNA-rRNA hybridization. The following percent saturation values were detected in the three samples: 0.08 in meristem DNA (samplea), 0.129 in I root segment DNA (sampleb), and 0.105 in II root segment DNA (samplec).Thermal denaturation of DNA and the derivative curves of the melting profiles evidenced five DNA families which were differently represented in the three DNA samples. DNA elution by thermal chromatography on hydroxyapatite followed by hybridization with3H-rRNA, revealed that ribosomal cistrons melt between 90 and 91 °C, corresponding to a G-C content of 50.7%. Moreover, the amount of the DNA family containing ribosomal cistrons was greater in sampleb andc, in sampleb to a greater extent, as compared with samplea. On the other hand, one DNA family melting at a higher temperature (92–93 °C) was drastically increased in samplec.Buoyant density profiles of unsonicated DNA showed no peaks in the three DNA samples. Upon somcation, a heavy shoulder was observed in the profile of sampleb. As the density of ribosomal cistrons and that of shoulder were very similar, it seems possible that the two fractions contain many DNA sequences in common.The present studies demonstrate that the proportion of ribosomal cistrons and other DNA families does not keep constant during the development of the metaxylem cell line.  相似文献

Summary In Schizosaccharomyces pombe the experiments performed in order to assess some possible influence of caffeine on biological processes related to the induced or spontaneous mutations and meiotic recombinations, have shown that caffeine decreases UV- and NG-induced forward mutations at ad-6 and ad-7 loci and UV-induced reverse-mutations of his - mutants whereas spontaneous mutations to adenine or histidine independence were not affected; intergenic meiotic recombinations in the MT chromosomal region were also decreased when caffeine was present in the crossing medium.Publication No. 42 from the Laboratorio di Mutagenesi e Differenziamento, Consiglio Nazionale delle Ricerche (C. Summary Fifty five purple spontaneous mutants of the haploid fission yeast Schizosaccharomyces pombe have been isolated and distributed, by genetic analyses, between two loci (ad-6 and ad-7) which control two independent steps in the purine biosynthesis.The molecular nature of these mutants has been determined by both their physiological behaviour (leakiness, temperature-sensitivity, pH-sensitivity) and their interallelic complementation ability.It is concluded that during the mitotic cycle the large majority of spontaneous mutations are base-pair substitutions.Publication n. 6 from the Laboratorio di Mutagenesi e Differenziamento del Consiglio Nazionale delle Ricerche (C.N.R.), Pisa, Italy.  相似文献

Summary In vivo observations ofNicotiana glauca ×Nicotiana langsdorffii hybrid callus cells cultured on hanging-drop slides show a continuous pulsating movement of nucleoli containing vesicles from the nucleus into the cytoplasm and the occasional release of the nucleoli into the cytoplasmic stream. Summary Allelism of nine nitrate reductase deficient (NR) Nicotiana plumbaginifolia cell lines was tested by complementation after protoplast fusion. Complementation was recognized by the appearance of somatic hybrid colonies growing on a selective NH4 +/NO3 medium which cannot support the growth of NR lines. All five apoenzyme defective (NA) lines were non-complementing and therefore allelic. The apoenzyme and the cofactor defective (NX) lines were complementing, as expected, and gave somatic hybrids with restored nitrate reductase activity. The four cofactor defective lines were found to belong to three complementation groups (NX1 and NX9; NX21; NX24). Two of these (NX21 and NX24) are of new types which have not been previously described in flowering plants. In the somatic hybrids restoration of NR activity was accompanied by the restoration of plant regeneration ability. Summary The first step of differentiation in the root segments ofAllium cepa containing metaxylem cells in different stages of differentiation were studied by DNA reassociation curves and compared to meristem cell extracted DNA. Upon sonication of DNA samples to about 400 base pairs, the reassociation profiles of the heat denatured DNA, were spectrophotometrically followed at two different concentrations. The kinetic complexities,i.e., the number of base pairs per haploid genome of a given sequence and its redundancy were calculated. Differences were found at the level of highly and medium repetitive sequences, thus demonstrating that some DNA reassociation classes may undergo amplification during root development.  相似文献

Summary Observations are presented on the patterns of DNA synthesis and mitotic activity in medullary parenchyma cells excised from tubers ofHelianthus tuberosus in four different periods of dormancy. Dormancy break (activation) was induced byin vitro culture on media added with 2,4-dichlorophenoxyacetic acid. The cell cycle responsein vitro to different combinations of growth substances has also been investigated.The results show that remarkable changes in the timing of the first and second cell cycles and their phases occur with the progression of dormancy. With increasing time after tuber harvest, the following behaviours are observed: (i) a lengthening of the first cell cycle, chiefly due to a lengthening of the G2 phase (G2 is absent at the beginning of dormancy) and an increase in the time interval between the start of thein vitro culture and the onset of the first mitotic wave; (ii) an increased duration of the S phase; (iii) a remarkable reduction in the cell synchrony.These behaviours, as indicated also by their comparison with thein vitro response of the cell cycle to different hormonal treatments, seem to depend on the physiological status of the tubers at the time of explant. Summary The number of ribosomal RNA cistrons has been measured in the total DNA extracted from L2 juvenile and adult stages of the free-living nematodePanagrellus silusiae. Saturation hybridization studies with homologous rRNA indicate that both stages have about 275 ribosomal genes per haploid equivalent. Using homologous125I-labelled rRNA for in situ hybridization, the mean number of silver grains per DNA content for oocyte, hypodermis and gut nuclei was similar. The mean DNA contents of maturing oocyte, hypodermis and gut nuclei are about 20C, 2C, and 10C respectively. We conclude that rDNA amplification alone is insufficient to account for the variation in DNA content of oocytes and that postembryonic development in this eutelic organism occurs without a significant differential increase in the number of ribosomal cistrons per worm.Supported by the National Research Council of Canada  相似文献

Summary The differentiation processes of the metaxylem cell line in the root ofAllium cepa are characterized by amplification phenomena of repetitive DNA sequences mainly localized in heterochromatic regions of metaphase chromosomes. Moreover, these sequences are heavily methylated. This paper presents additional results on variation in endogenous DNA methylation in different developing root segments. The results show that methylation is higher in apical meristematic cells than the differentiating segments; contrastingly, total RNA synthesis seems to be correlated with undermethylation. Addition of labelled methyl groups to DNA by eukaryotic methylase, DNA digestions with different restriction enzymes specific for methylated sites and HPLC analysis confirmed the above results. Moreover, variation in methylation levels during differentiation occur not only at the internal cytosine of the-CCG-sites, but also at external cytosine. Furthermore, methylation affects other sites containing the trinucleotides-CXG-. In conclusion, root differentiation inAllium cepa seems to be correlated with gene activation modulated by the methylation/demethylation of particular DNA sequences.  相似文献

Four different food matrices (alfalfa, cilantro, mamey sapote, and mung bean) were contaminated with three different dilutions 106, 104, and 103 cfu/g of Yersinia enterocolitica. DNA was isolated from each food mix and used in chromosomal amplifications. The amplified DNA was used as templates in single PCR reactions of the four genes (virF, ail, yst, and blaA) followed by mixing the four reactions for one PCR primer extension reaction. The presence and the limit of detection of four genes in four food matrices were established by microarray hybridization. Data revealed the diversity of signal intensities. Neither the microarray chip hybridization nor the single PCR amplification could detect virF’s presence located on a plasmid. Ail was detected in 103 cfu/g, whereas blaA and yst were detected from 105 to 106 cfu/g in all food matrices. Therefore, the ail gene could be the gene of choice in identifying Y. enterocolitica in alfalfa, cilantro, mamey, and mung bean. Other genes—blaA, yst, virF—exhibited wide variability in hybridization signals, highlighting the need of a better DNA purification step prior to DNA microarray hybridization.  相似文献

Nuclear DNA and AgNOR-area quantity have been measured cytophotometrically, the number of silver grains afterin situ 3H rDNA/DNA hybridization has been counted, and the level of rDNA methylation has been estimated in root meristems, differentiated root zones, hypocotyls, cotyledons, and leaves of 7-day-old seedlings ofLupinus luteus. DNA content increases by endoreplication, reaching the average DNA C-value of 4.2 in the meristem, 6.6C in hypocotyl, 15.9C in cotyledon, and 3.3C in leaf. The quantity of AgNOR-area, the highest in root meristem, does not follow DNA C-value. Number of grains afterin situ 3H rDNA/DNA hybridization corresponds to DNA amount, but their distribution differs, showing in many cases perinucleolar localization in organs displaying low AgNOR-area quantity. Southern hybridization has not demonstrated detectable changes in level rDNA methylation. We suggest that the low level of rDNA expression in non-meristematic tissues could be related to rDNA inactivation caused by its translocation into nucleolus-associated heterochromatin.  相似文献

The effect of Cd on oxalate oxidase (OxO) activity and its localisation were analysed in barley root. In Cd-treated roots OxO activity was strongly induced in the region 2–4 mm behind the root tip and in the area toward the root base. In situ analyses showed that Cd-induced OxO activity was localised to the cell wall (CW) of early metaxylem vascular bundles and surrounding parenchyma cells and was accompanied by lignification of metaxylem vessels. OxO activation was also observed during treatment with other heavy metals (HMs), salt treatment and at elevated non-optimal temperature. In contrast to HM activation of OxO and lignification, high temperature and NaCl indeed activated OxO but did not induce lignification of metaxylem vessels. These results suggest that oxalate oxidase as an H2O2-generating enzyme is activated in response to several stresses, however the ectopic lignification of metaxylem vessels is activated specifically by HMs. This HM-induced premature root xylogenesis due to ectopic lignification of metaxylem vessels probably causes shortening of the root elongation zone and therefore a reduction in root growth.  相似文献

The pattern of proliferation and growth of cortical and central metaxylem cells in a radicle and the transitional zone of a wheat embryo was studied during the final stages of embryogenesis. Cell divisions finished nearer the root tip in the central metaxylem than was the case in the cortex. After divisions ceased the cells of both tissues maintained the ability to synthesize DNA and the cells began DNA endoreduplication. The maximal levels of endoreduplication were 4C and 8C in cortical and central metaxylem cells, respectively. As a result of nonsimultaneous cessation of divisions, the metaxylem cells were two or three times longer than cortical cells. The proportion of cells with the maximal DNA content was smaller in the transitional zone than in the radicle. During the final embryonal stages cell growth rate was decreased. It was established that the transition of cells to DNA synthesis was inhibited in all sites of the radicle during the completion of embryogenesis. The cell growth was topped in proximal sites of the radicle. In the division zone the cells which had already begun DNA synthesis were able to complete it and divided. Cell growth stopped simultaneously with completion of proliferation in this zone.  相似文献

DNA amplification fingerprinting of the Azolla-Anabaena symbiosis   总被引:2,自引:0,他引:2  
The Azolla-Anabaena symbiosis has been used for centuries as a nitrogen biofertilizer in rice paddies. Genetic improvement of the symbiosis has been limited by the difficulty in identifying Azolla-Anabaena accessions and Anabaena azollae strains. The recently developed technique of DNA amplification fingerprinting (DAF) was applied to this problem. DAF uses single, short, oligonucleotide primers of arbitrary sequence to direct amplification of a characteristic set of DNA products by a thermostable DNA polymerase in a thermocycling reaction. The products are separated in polyacrylamide gels and detected by silver staining. DAF could easily distinguish and positively identify accessions of Azolla-Anabaena with DNA extracted from the intact symbioses. The contribution of prokaryotic Anabaena sequences to the fingerprint of the intact symbioses, however, ranged from 0 to 77%, depending on the primer sequence. Therefore, DNA extracted from the intact symbioses would not be suitable for Azolla taxonomy studies. The fingerprints of Anabaena strains isolated by sucrose gradient centrifugation from different species of Azolla could be easily distinguished, and DAF patterns were used to confirm the maternal pattern of transmission of Anabaena in a sexual hybrid. Template DNA extracted from roots was used to produce fingerprints for Azolla without interference from the microsymbiont. Comparison of the patterns from the parents and a hybrid gave strong evidence confirming sexual hybridization.  相似文献

A new approach to study dynamic interactions between transpiration and xylem pressure in intact plants is presented. Pressure probe measurements were preformed in living (immature) late metaxylem of maize roots rather than in adjacent mature xylem. This eliminated technical limitations related to the measurement of negative pressures. Water relations of single cells showed that turgor and volumetric elastic modulus were significantly larger in living metaxylem than in cortical cells; hydraulic conductivity was similar in both types of root cells. Increasing transpiration induced an immediate decrease of xylem pressure, and vice versa. Turgor in the living metaxylem could be continuously recorded for more than 1 h. The relationship between xylem pressure and transpiration yielded a root hydraulic resistance of 1.3 x 109 MPa s m-3. Control experiments indicated that the response of living xylem in the positive pressure range essentially paralleled that of mature root xylem in the negative range. In mature xylem, pressures as low as -0.55 MPa were recorded for short periods (several minutes). Several tests verified that the pressure probe was in contact with mature xylem during the measurements of tensions. The results demonstrate convincingly that transpiration generates an effective driving force for water uptake in roots, a central feature of the cohesion theory.Key words: Hydraulic conductivity, negative pressure, root development, turgor, water transport, Zea mays.   相似文献

The exceptionally high capacity for transformation to autotrophyfor hormones (habituation) discovered in Nicotiana bigeloviiled us to a comparison of the responses of 2 varieties (quadrivalvisand bigelovii) to different hormone treatments applied to theirseeds. Hormones used were: kinetin (0.8, 1.6, 3.2 ppm), 2,4-D(0.1, 0.4 ppm), IAA (2 ppm) and NAA (1, 2, 4 ppm). Callus formation and habituation were scored after transferon minimal medium (without hormones) 3 months after sowing.Var. quadrivalvis showed a much higher ability to form callusbut the callus formed was autonomous in a lower percentage ofcases when IAA or kinetin treatment was considered. Analysisof auxin-like substances in leaves of the 2 varieties showeda higher content in N. bigelovii var. quadrivalvis. Summary DNA sequences reassociating within a Cot value of 1.8×10–1 and those producing a light satellite in a CsCl density gradient were isolated fromVicia faba DNA and hybridizedin situ on squashes of roots of the same species. Silver grains were seen to be scattered over both the interphase nuclei and the metaphase chromosomes after hybridization with fast renaturing DNA sequences, indicating these are fairly regularly interspersed in theV. faba genome. Clustered labeling occurred after hybridization with satellite DNA sequences, indicating these are clustered in the genome. The localization of satellite DNA in chromosomes appeared to correspond closely to the position of the bright bands detectable after staining with quinacrine mustard. After hybridization with both DNA probes, labeling intensity over the nuclei of meristematic cells was higher than that over the nuclei of differentiating and/or differentiated cells. These results are discussed in relation to the structure of the cell nucleus, the mechanism of quinacrine banding and to previous data suggesting underrepresentation of nuclear repeated DNA sequences in differentiatingV. faba root cells.  相似文献

Using 3-day-old seedling roots of Zea mays L., cv. Kelvedon 33, it was possible to remove the root cap by a simple surgical manipulation without damage to the root proper. By a further small cut, the quiescent center (QC) itself was isolated. This double-convex lens-shaped tissue piece 100 X 250 μm is composed of 1000–1500 cells representing only 0.25 mm3 in volume. The explant was demonstrated unequivocally by 3H-thymidine incorporation before excision and then by autoradiography to be composed of the specific cells usually designated the quiescent center. Using sterile techniques, the QC's were placed on nutrient agar slants and allowed to grow in culture. Of a number of nutrient media tested, only a medium supplemented with organic nitrogen components, indoleacetic acid, kinetin and inorganic nutrients plus sucrose (S2M + K -2,4-D) was effective in eliciting development. Thirty to 40 percent of the 150 isolated QC's grown on this medium formed elongated roots, up to 2 cm in length in 3–4 weeks. Roots developing on agar medium showed in their proximal portion a vascular pattern with 5–6 metaxylem elements or variations of this pattern, but as the root elongated, the vascular pattern was progressively reduced in complexity at the more distal end to a small central group of metaxylem elements. When agar-grown roots were transferred after one week in culture to a liquid nutrient medium of the same composition, the initially reduced vascular pattern evident in the proximal tissues became progressively more complex in the distal portion of the root and after 2 cm of elongation, showed an essentially normal primary vascular tissue pattern characteristic of the seedling root.  相似文献
