Chromosomes were analysed in peripheral lymphocytes of 24 workers in a zinc smelting plant who had increased blood levels of lead and cadmium. The number of cells with structural chromosome aberrations was significantly increased as compared with 15 controls. The observed chromosome damage was mainly of the chromatid type (single breaks and exchanges) accompanied by acentric fragments. The observed chromosome aberrations cannot be causally related to cadmium because the workers were additionally exposed to lead and zinc. However, from existing cytogenetic data on heavy metals an effect of cadmium could be well deduced, although possible synergistic effects of several metal compounds, especially under conditions, in vivo, cannot be excluded. 相似文献

In vivo X-ray fluorescence (XRF) techniques were used for biological monitoring of lead, cadmium, and mercury. Lead accumulates
in bone, the level of which may thus be used for monitoring of exposure. However, there was no close association between lead
levels in bone and exposure time, partly because of differences in exposure patterns and partly, probably, because of variations
in the toxicokinetics of lead. There are at least two separate bone lead compartments. The average over-all half-time is probably
5–10 yr. The finger bone level may be an index of the lead status of the total skeleton. In lead workers, the mobilization
of bone lead causes an “internal” lead exposure and affects the blood lead level considerably. In cadmium workers, in vivo
XRF is a sensitive and risk-free method for assessment of accumulation in kidney cortex, the critical tissue as to toxic effects;
workers displayed increased levels. However, there was no clear association with duration and intensity of exposure, cadmium
levels in urine, or microglobulinuria. Determinations of kidney cadmium may add important information on the state of accumulation
and, thus, risk of kidney damage. Workers exposed to elemental mercury vapor, as well as fishermen exposed to methyl mercury,
had mercury levels in bone below the detection limit of the XRF method. 相似文献

ObjectiveThis study aims to observe the effect on urine and sweat excretion levels of cadmium (Cd) and lead (Pb) in healthy men in a maximum incremental test until exhaustion and repeated exposure to heat.Methodstwenty-nine adult men divided into control group (CG; n = 14) and experimental group (EG; n = 15) performing two maximum tests until exhaustion in normothermia (22 °C) and hyperthermia (42 °C). EG experienced 9 sessions of heat exposure at high temperatures (100 °C) (HEHT). After the nine sessions, the initial tests were repeated in both groups. Urine samples were collected before and after each test. After the hyperthermia tests, sweat samples were gathered.ResultsUrinary Cd increased after initial tests in GC and in hyperthermia in EG (p < 0.05). Urinary excretion of Pb rose after HEHT (p < 0.05). Pb in sweat was higher in EG than in CG after HEHT (p < 0.05).ConclusionHeat exercise and constant exposure to heat can be a valid method to increase the excretion of toxic metals. 相似文献

The plasma and erythrocyte lipid peroxide levels were measured in a group of male subjects occupationally exposed to lead
for an average period of 17 yr, and compared to those from an age-matched control group living in the same city in a similar
socioeconomical environment.
The blood lead and plasma zinc levels were measured by atomic absorption spectroscopy. The plasma and erythrocyte lipid peroxide
levels were established by the malondialdehyde determination method. Significant differences were found in the blood lead
levels in lead-exposed workers, 15.00±10.15 μg/dL as compared to controls, 2.37±0.89 μg/dL. The plasma (2.67±0.69 μM) and erythrocyte (27.53±6.28 nmol/g Hb) lipid peroxide levels in workers with occupational exposure to lead were significantly
higher than controls, 1.23±0.61 μM and 14.35±2.08 nmol/g Hb, respectively. There were no significant differences of the zinc levels in both groups.
The blood lead levels had a statistically significant positive correlation with age and with duration of exposure in both
groups, but showed no relationship to the corresponding blood zinc levels. The results presented in this study indicate that
the increase of plasma and erythrocyte lipid peroxide levels in workers exposed to lead may be related to the lead concentration,

The Authors examine the variations of the levels of PbB and EPP in 39 workers at known lead exposure and evaluate the capacity of these parameters to follow the measured decreases of the environmental pollution. A study was conducted using a combined testing protocol (CTP), to determine whether short-term biological end-points, singly or in combination, are sufficiently sensitive to identify damage induced by exposure to ambient levels of industrial chemicals. A small-scale inhalation set-up which is both economical and easy to assemble was designed. Mice were exposed to 4 concentrations of a custom-blend mixture of benzene, chloroprene, epichlorohydrin and xylene in a ratio of 2:2:1:2, respectively. The concentrations for benzene, chloroprene and xylene were 0, 0.1, 1.0 and 10 ppm each. Concentrations for epichlorohydrin were half those for the other components. Groups of 22 males and 22 female mice were exposed to each concentration of the mixture for 3 and 6 weeks. Selected biological end-points including urine mutagenesis, bone marrow cell aberrations and micronuclei, spleen lymphocyte aberrations and liver enzyme induction were monitored. The spleen lymphocyte aberrations and liver enzyme induction were the most sensitive end-points. The lymphocytes showed a significant induction of chromosome aberrations from exposure for 3 weeks to all 3 concentrations of the mixtures. After 6 weeks of exposure, significant induction of aberrations was observed after exposure to low and medium concentrations but not to the high concentration. This lack of response at the high concentration after 6 weeks exposure, appeared to correlate with a significant induction of glutathione S-transferase in the liver. Since this enzyme is known to detoxify 3 of the 4 chemicals in our mixture, it may indicate a detoxification mechanism after enzyme induction. The present study indicates that the CTP is sufficiently sensitive to identify toxicological effects after exposure to ambient levels of a gas mixture. 相似文献

The present work is focused on the determination of in vivo doses and studies of genetic effects in workers exposed to epichlorohydrin (ECH). The studied endpoints were hemoglobin (Hb) adducts, frequencies of hprt mutants, micronuclei in cytochalasin B blocked binucleated lymphocytes, sister chromatid exchanges (SCE) and high frequency cells (HFC). Blood samples were collected from office clerks and ECH exposed factory workers at an industrial plant in Germany. The workers were exposed to 0.11–0.23 ppm ECH in the air 45 h per week and to 0.2–2.6 ppm for 3 h per week. Some Swedish non-exposed subjects were also used for Hb adduct measurements. The genetic data, HFC and SCE, showed a significant difference between exposed and unexposed donors. In contrast to earlier studies on SCE, no impact of smoking was observed. Effects on micronuclei were on the borderline of significance, whereas there was no effect for HPRT mutants. The average Hb adduct level was higher in exposed than in non-exposed donors, although the difference was only significant when the exposed group was compared to Swedish controls. Smoking gave significantly increased adduct levels. The absence of significant correlations between individual data for Hb adducts and genetic effects, may be explained by the different periods of time covered by the responses in these endpoints. Whereas Hb adducts reflect the exposure during up to 4 months (i.e. the life span of human erythrocytes), the SCE, and particularly the HFC, seem to accumulate for years in a long-lived fraction of T-lymphocytes without DNA repair. Thus, the adduct data does not reflect the exposure backwards in time unless it can be shown that exposure conditions have remained unchanged. The origin of the background adduct levels in non-smoking control persons is at present not known. 相似文献

This study investigates different effects in roots of Vicia faba (broad bean) after exposure to cadmium. Genotoxic effects were assessed by use of the well-known Vicia root tip micronucleus assay. Cytotoxic effects were evaluated by determining the mitotic index in root tip cells. Finally, molecular induction mechanisms were evaluated by measuring phytochelatins with HPLC. After hydroponical exposure of V. faba roots to a range of cadmium concentrations and during different exposure times, the results of this approach showed large variations, according to the endpoint measured: after 48 h of exposure, genotoxic effects were found between 7.5 x 10(-8) and 5 x 10(-7)M CdCl(2), and cytotoxic effects were observed between 2.5 x 10(-7) and 5 x 10(-7)M CdCl(2). Statistically significant phytochelatin (PC) concentrations were measured at >or=10(-6)M CdCl(2) for PC(2), and at >or=10(-5)M CdCl(2) for PC3 and PC4. 相似文献

The relationship between blood lead (Pb) and serum levels of calcium and of neural nutrients such as thiamine and magnesium (Mg) has been determined in a Nigerian population that is occupationally exposed to Pb. Forty-seven male Pb workers were recruited as test subjects and 25 males unexposed to Pb served as controls. The test subjects were classified into three groups, based on severity of exposure to Pb. Blood lead (BPb) and the serum levels of Mg, thiamine, and calcium were determined in both test subjects and controls. The mean blood Pb level was not significantly higher in Pb workers. In contrast, Mg and thiamine levels were significantly decreased (p<0.05; p<0.01, respectively). However, the calcium level was not significantly lower in test subjects than in controls. Also, there was a significant negative correlation between serum thiamine and blood Pb levels (r=-0.50; p<0.01). Furthermore, there was a significant negative correlation between serum calcium and BPb levels (r=-0.41; p<0.01). This study has shown that relatively low BPb levels can enhance Pb absorption and also potentiate Pb neurotoxicity in the presence of decreased serum thiamine and Mg levels. 相似文献

Estrogens are associated with several cancers in humans and are known to induce tumors in rodents. In this review a mechanism of carcinogenesis by estrogens is discussed which features the following key events: (1) Steroid estrogens are metabolized by estrogen 2- and 4-hydroxylases to catecholestrogens. Target organs of estrogen-induced carcinogenesis, hamster kidney or mouse uterus, contain high levels of estrogen 4-hydroxylase activity. Since the methylation of 4-hydroxyestradiol by catechol-O-methyltransferase is inhibited by 2-hydroxyestradiol, it is proposed that a build up of 4-hydroxyestrogens precedes estrogen-induced cancer. (2) The catecholestrogen or diethylstilbestrol (DES) are oxidized to semiquinones and quinones by the peroxidatic activity of cytochrome P-450. The quinones are proposed to be (the) reactive intermediates of estrogen metabolism. (3) The quinones may be reduced to catecholestrogens and DES and redox cycling may ensue. Redox cycling of estrogens has been shown to generate free radicals which may react to form the organic hydroperoxides needed as cofactors for oxidation to quinones. (4) The quinone metabolites of catechol estrogens and of DES bind covalently to DNA in vitro whereas DNA binding in vivo has only been examined for DES. When DES is administered to hamsters, the resulting DES-DNA adduct profile in liver, kidney, or other organs closely matches that of DES quinone-DNA adducts in vitro. In vitro, DES-DNA adducts are chemically unstable and are generated in incubations with organic hydroperoxide as cofactor. It is proposed that the instability of adducts and the lower sensitivity of previous assay methods contributed to the reported failures to detect adducts. Steroid estrogen-DNA adducts in vivo are currently under investigation. (5) Tumors are postulated to arise in cells rapidly proliferating due to the growth stimulus provided by the estrogenic activity of the primary estrogen or of hormonally potent metabolites such as 4-hydroxyestradiol. The covalent modification of DNA in these cells is temporary because of the chemical instability of adducts and will result in altered genetic messages in daughter cells, whereas in non-proliferating cells there may be no lasting genetic damage. The sequence of events described above is a plausible mechanism for tumor initiation by estrogens and is partially substantiated by experimental evidence obtained in vitro and/or in vivo. 相似文献

Chronic exposure (3.5 mo) of mice to cadmium (Cd), lead (Pb), or a cadmium-lead mixture at a concentration of 1 ppm in drinking water induced a highly significant inhibition of antibody response to human serum. The highest immunosuppression (84.4%) was induced by the Cd-Pb mixture, whereas Cd caused the lowest immunosuppression (53.6%). The body burden of Cd and Pb in various organs was investigated in the four groups of mice by atomic absorption spectrometry. The highest level of Cd was found in the kidney of the Cd-treated group, and the highest level of Pb was found in the liver of the Pb- and Cd-Pb-treated groups. It is concluded that when mice are exposed concurrently to Cd and Pb, they develop synergistic immunsuppression. Analysis of Cd levels using atomic absorption spectrometry revealed that it was distributed in the following order: kidney>liver>spleen>heart, whereas Pb was distributed in the following order: liver>kidney>spleen>heart. 相似文献

Studies were carried out to determine the effect of dietary vanadium on chicks fed phosphorus deficient and control diets.
Vanadium at 50 mg/kg of diet decreased growth of both control and deficient chicks. The high mortality among the phosphorus
deficient chicks was significantly alleviated by the presence of vanadium. The increased relative ventricular weights found
among the deficient chicks was also alleviated by the presence of dietary vanadium. Vanadium fed at 10 or 20 mg/kg diet did
not reduce growth rate but significantly reduced mortality among chicks fed the deficient diet and decreased the relative
ventricle weights. Time course studies revealed that chicks are hatched with high relative ventricular weights (.83% of body
wt) and remain at that level among chicks fed the phosphorus deficient diet. The addition of vanadium or phosphate to the
diet resulted in a progressive decrease in relative ventricular weights. The inclusion of vanadium in the diet resulted in
increased serum phosphorus levels among the deficient chicks that may be related to the decrease in mortality and relative

Biological monitoring of exposure to ambient environmental lead and cadmium was performed using feathers of 26 species of birds native to Western Uttar Pradesh in Northern India. The rationale of this study was to address three questions. First, is there any avian species that can be treated as a suitable bioindicator of lead or cadmium present in the environment? Second, do the birds selectively accumulate lead and cadmium in their feathers and exhibit interspecies variation? Third, is there any threat to endangered species of this region from metal pollution? Average concentration of lead in the feathers of selected birds ranged from 3.40 µg/g in parrot to 301.6 µg/g in golden pheasant, whereas cadmium concentration was higher ranging from 40.20 µg/g in red crow to 450 µg/g in blue macaw. A comparison of results made through ANOVA revealed a significant difference in the concentration of lead (df = 25; F = 3965.54) and cadmium (df = 25; F = 8537.27) in their feathers. We hypothesize that feathers of synanthropic birds may be treated as suitable noninvasive tool to monitor the ambient environmental contamination by lead and cadmium. Their accumulation in endangered birds may lead to population decline causing serious ecological disturbances in the region. 相似文献

Aim of the study was to investigate the genotoxic effects of methyl isothiocyanate (MITC), a compound widely distributed in the environment as a constituent of certain vegetables, a soil fumigant and breakdown product of carbamate pesticides. MITC caused only marginal mutation induction in reversion assays with Salmonella strains TA100 and TA98 and, the maximum effect (<2-fold increase over the background rate) was seen at 100microg/ml. In differential DNA-repair assays with E. coli (strains 343/763 uvrB/recA and 343/765 uvr(+)/rec(+)), a pronounced dose-response effect (induction of repairable DNA-damage) was seen at low concentrations (>/=4microg/ml). In both bacterial assays, addition of activation mix (rat liver S-9) led to a reduction of the genotoxic effects. In micronucleus assay and in single cell gel electrophoresis assay with human hepatoma cells (HepG2), clear cut positive results were obtained at exposure concentrations of <5microg/ml. On the contrary, only marginal effects were seen in differential DNA-repair host-mediated assays where E. coli indicator cells were recovered from different inner organs of mice that had been treated orally with a high dose (90mg/kg bw) of the test compound. Further in vitro experiments showed that MITC is inactivated by body fluids (saliva, gastric juice) and that its DNA-damaging properties are attenuated by non-enzymatic protein binding. Since exposure of HepG2 cells to MITC led to formation of thiobarbituric acid reactive substances, it is likely that its DNA-damaging effects involve lipid peroxidation processes. Overall, our findings show that MITC induces only marginal effects at extremely high (almost lethal) doses in inner organs in vivo, but it causes DNA-damage at low concentrations in vitro. 相似文献
