Purification and Characterization of a Novel Brain-Specific 14-kDa Protein   总被引:3,自引:1,他引:3  
A new acidic protein specifically present in the brain was purified to homogeneity from bovine brain. The apparent molecular mass was estimated to be 14 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and 57 kDa by gel filtration, a finding suggesting that it exists as a tetramer under physiological conditions. The protein had a high content of Glu and Pro, and its pI was 4.3. The first six amino acid residues of the protein were Met-Asp-Val-Phe-Met-Lys, and the amino terminal was blocked. The distribution of the protein examined by Ouchterlony gel immunodiffusion indicates that it is present specifically in brain, including rat, human, and bovine, but could not be detected in 10 other rat tissues examined. The protein was absent in Purkinje cell bodies, as examined by electron microscopic immunocytochemistry, but was present in nerve terminals that make synapse-like contacts with Purkinje cells and in neurons with dark granules in the globus pallidus of the rat.  相似文献

 应用抑制性差减杂交技术 ( SSH)克隆两种不同小鼠胸腺基质细胞的差异表达基因 ,获得新基因片段 C55.通过 Gen Bank检索及 RT- PCR扩增出一个全长 1 .4kb的 c DNA.杂交分析认为它是一个完整的 c DNA序列 .c DNA序列分析表明 ,它拥有一个 636bp的开放读码框架 ,编码 2 1 2个氨基酸 .同源序列比较发现 ,它编码一个肌动蛋白相关蛋白的新成员 ,该序列与多种已知的肌动蛋白相关蛋白 SM2 2 α及其同源蛋白在氨基酸水平上有 62 %~ 95%的同源性 .Northern杂交分析显示 ,该基因 m RNA转录本在两种不同胸腺基质细胞中的表达存在显著差异 . 从人胎脑cDNA文库中克隆到一条全长的锌指蛋白新基因的cDNA,命名为ZNF303。序列分析表明,ZNF303的C末端含有7个保守的锌指基序,N末端含有一个KRAB(Krüppel?associated box)结构域。利用肝癌组织表达谱基因芯片杂交证明,该基因在肝癌组织中的表达量有明显降低。认为这种降低可能跟肝癌的形成和转移有密切的关系。利用辐射杂交基因定位技术,得出该基因在人类染色体上的位置是19q13.2。 Abstract: We have cloned a full?length novel zinc finger cDNA of the Krüppel family from human fetal brain cDNA library. Sequence analysis indicates that ZNF303 contains 7 highly conserved zinc finger motifs at the C-terminus and a KRAB(Krüppel-associated box)domain at the N-terminus of the deduced rotein.Hybridization using gene chip of hepatic cancer tissue demonstrates the expressive amount in hepatic cancer tissue is lower than control. We hypothecate that the decreasing of expression amount is related to the formation and metastasis of hepatic cancer.Finally,we show that ZNF303 maps on human chromosome 19q13.2 by radiation hybrid.  相似文献

以G2豌豆幼苗为材料,构建了滴度为6.5×106pfu的cDNA文库,用同源序列筛选法从该文库中得到一个全长465bp的cDNA。杂交分析认为它是一完整的cDNA序列。DNA序列分析表明,它拥有一个282bp的开放读码框,编码94个氨基酸。计算机同源序列比较发现,它可能编码豌豆核糖体小亚基蛋白S21(RS21-PEA),因为该序列与已知的玉米、水稻S21在氨基酸全序列水平上有32%~35%的同源性,并含有与RNA结合的特征结构域。进化树分析显示S21蛋白可在一定程度上反映生物的进化及遗传变异趋势。  相似文献

The present report shows that mediatophore, a nerve terminal membrane protein that translocates acetylcholine on calcium action, forms a complex with a 14-kDa polypeptide. The complex was identified based on the following results. (a) A polyclonal antimediatophore antiserum that immunoprecipitates activity precipitates both the 15- and 14-kDa polypeptides. (b) After HPLC purification of mediatophore, both antigens were found in the same peak. (c) After 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate solubilization of presynaptic membranes or of the purified mediatophore, an immunoaffinity column made with the anti-14-kDa antigen monoclonal antibody retained both the 14-kDa and the 15-kDa polypeptide. Similarly, immunoprecipitation experiments using protein A-coated beads sedimented an immunocomplex in which both antigens were found. (d) The 14-kDa antigen could be localized in the synaptosomal membrane where mediatophore and its 15-kDa component are found.  相似文献

Antifungal protein is the main inhibitor of fungal infection in the secondary corm of Gastrodia elata B1. was isolated and purified antifungal protein (GAFP) from the plant. Its molecular weight was about 14 kD. Polyclonal antibody against GAFP was produced. In vitro test, this antifungal protein inhibited the growth of some fungi in some crop including Gibberella zeae. cDNA was synthesized from poly (A) mRNA purified from G. elata. The cDNA was ligated into phage vector λgtll DNA and packaged in vitro and the phages were propagated on E. coli Y1090 and a λgtll expression library was constructed. A cDNA clone encoding for antifungal protein was screened out by immunoscreening of the library using the protein as a probe. The λDNA containing insert was digested by Eco RI after isolated and purified recombinants λDNA, the insert was obtained. The cDNA was 300 bp in length. The authors had isolated the cDNA clone encoding antifungal protein from G. elata.  相似文献

The digestibility and gastrointestinal absorption of 14–16-kDa rice allergens (RAs) were investigated. RAs and bovine serum albumin (BSA) were first evaluated for their digestibility. BSA was digested completely by in vitro incubation with some proteases, but RAs remained almost intact. Administered orally (20 mg per mouse), intact RAs were clearly detected in the small intestine even 60 min after the administration, the amount of total RAs in the small intestine being estimated to be 0.59 mg. RAs were then biotinylated and infused into the duodenal lumen of anesthetized mice, and portal blood was collected. The RA concentrations in the portal plasma were respectively estimated to be 0.4–0.9 and 0.3–2.5 μg/ml for 0.4 and 4 mg doses. These results suggest that RAs are highly resistant to digestive enzymes and that about 1/100 of orally administered RAs remain intact in the small intestine, while at least 1/1,000–1/10,000 is absorbed and delivered into circulated blood.  相似文献

 通过检索GenBank的表达序列标签 (EST)数据库并结合cDNA末端快速扩增法 (RACE) ,从小鼠胸腺克隆到一个新的cDNA序列 ,并从人类肝癌组织中克隆出了其同源cDNA .根据读码框架分析 ,这两个cDNA分别编码 541和 555个氨基酸的蛋白质 两个蛋白质之间氨基酸序列一致率为77% ,和已知蛋白无显著同源性 .分子生物学软件和网上分析表明 ,两个蛋白质所含功能序列与STAT家族成员极为相似 ,均含有包括酪氨酸蛋白激酶在内的多种蛋白激酶的磷酸化位点和核定位信号 (NLS) ,可能是一种新型转录因子 .RT PCR分析显示 ,两个基因在正常组织中选择性表达 ,其分布相似 ,而且都具有一定程度的与分化或增殖相关的趋势 .  相似文献

In order to isolate a cDNA clone of ribosome inactivating protein (RIP), a cDNA library was constructed in Uni-ZAP XL vector with poly(A) RNA purified from leaves of Amaranthus viridis. To get the probe for screening the library, PCR of phage DNA was conducted using the vector primer and degenerate primer designed from a conserved putative active site of the RIPs. Twenty-six cDNA clones from about 600,000 plaques were isolated, and one of these clones was fully sequenced. It was 1,047 bp and contained an open reading frame encoding 270 amino acids. The deduced amino acid sequence had a putative signal sequence of 17 amino acids and a putative active site (AIQMVAEAARFFKYIE) conserved in other RIPs. E. coli cells expressing A. viridis RIP cDNA did not grow well as compared to control cells, indicating that recombinant A. viridis RIP presumably inactivated E. coli ribosomes. In addition, recombinant A. viridis RIP cDNA produced by E. coli had translation inhibition activity in vitro.  相似文献

从菜花烙铁头蛇(Trimeresurus jerdonii)的毒腺中提取mRNA,采用RT-PCR技术进行体外扩增,将扩增产物克隆到PMD18-T载体中,最后筛选出一个编码凝集素的基因,命名为TJL。由TJL基因序列推导的氨基酸序列中包含分别由23和135个氨基酸残基组成的信号肽和成熟肽。氨基酸序列比较分析表明,TJL含有半乳糖结合位点和钙离子结合位点,与蝰科蛇毒凝集素TSL、PAL、APL和RSL的同源性较高(87.4%-90.4%),与眼镜蛇科蛇毒凝集素BML的同源性较低(61.5%)。  相似文献

在研究狗抗吗啡活性肽PPC过程中,发现它与牛的DBI(diazepambindinginhibitor)的氨基酸序列有很高的同源性,但尚未见到有关狗的DBI的文献报道,为了更好的探讨PPC和DBI相互间的关系,对狗的DBI的cDNA进行了克隆和序列分析。本研究利用大鼠DBI的基因片段为探针,从狗肝脏cDNA文库中,筛选得到了一阳性克隆,并进行了全自动和手工测序,得到了DBI的全长基因。根据EBMLbank序列检索,发现狗的DBI核酸序列与牛的同源性为81%,将其核酸序列翻译成氨基酸序列,进行同源序列比较,结果显示:狗的DBI的氨基酸序列与猪、牛、人、酵母DBI的同源性分别为88.5%、87.4%、83.9%、46.5%。研究还发现狗的DBI序列与抗吗啡活性肽PPCN端62个氨基酸只有两个不同,C端17个氨基酸序列完全相同。只是PPC比DBI中间多了23个氨基酸。  相似文献

对一水稻cDNA 克隆(R1908) 的分析表明, 其可能编码水稻酰基辅酶A 结合蛋白(acylCoAbinding protein,ACBP)。Southern 杂交显示水稻( Oryza sativa L.) 基因组中仅有一个该基因的拷贝。Northern 分析表明水稻的ACBP基因在水稻的根、茎、叶、叶鞘、黄化苗和幼穗中皆表达,而以黄化苗的绿苗叶鞘中的表达强度高于绿苗叶片。  相似文献

为实现人蛋白C cDNA在哺乳动物细胞中的表达以及研究其生物学特性,针对人蛋白C cDNA序列设计引物,运用逆转录聚合酶链反应(RT-PCR)从人胎肝总RNA中钓取人蛋白C cDNA,将其克隆入pIRES neo载体中,通过酶切和PCR鉴定出重组体并进行测序分析。结果表明,获得大小为1386bp的人蛋白C cDNA基因,成功构建人蛋白C cDNA载体pIRES/hPC,为进一步进行人蛋白C cDNA的表达和活性鉴定奠定了基础。  相似文献

A Caenorhabditis elegans cDNA encoding a new insulin/IGF-like peptide was cloned and examined. The predicted peptide shows significant sequence similarity with the peptide Ceinsulin-1 reported previously and contains a characteristic insertion consisting of three residues in the putative B domain as with the Ceinsulin-1. The gene expression pattern during development is almost identical to that of Ceinsulin-1. The predicted tertiary structure of the peptide is quite similar to that of Ceinsulin-1, and their predicted receptor-recognition surfaces also closely match. These facts suggest that both peptides could recognize the same receptor.  相似文献

以黄瓜子房 (幼果 )RNA为模板 ,应用逆转录 聚合酶链式反应 (RT PCR) ,首次扩增出黄瓜生长素结合蛋白基因 (ABP1)cDNA片段 ,并进行测序和同源性分析。对ABP1基因在黄瓜子房 (幼果 )中的mRNA表达水平作了初步探讨 ,结果表明 ,该基因在开花前 1d的子房中表达信号较弱 ,在授粉后 2、4和 6d的幼果中表达增强 ;开花后 2d未经授粉的子房中 ,绿而膨大、能形成单性结实果者信号较强 ,黄而萎蔫、不能形成果实者信号较弱。Southern杂交结果表明 ,黄瓜生长素结合蛋白为小基因家族编码  相似文献
