Pharmaceuticals: a threat to drinking water? 总被引:9,自引:0,他引:9
Recently, considerable interest has developed regarding the presence of pharmaceuticals in the environment, but there has been comparatively little study on the potential of these substances to enter potable supplies. This is surprising because drinking water would provide a direct route into the body for any drugs that might be present. Although many countries employ advanced treatments, such as granular activated carbon, membrane technologies, ozonation and ultraviolet radiation, for treating water intended for human consumption, some compounds have been shown to be unaffected by such processes. Here, we examine the levels of drug substances reported in drinking water around the world. The possible implications of the presence of these compounds are highlighted and assessed, and recommendations are made for further research. 相似文献

The phenomenon of wildlife mortality at wind turbine installations has been generating increasing concern, both for the continued development of the wind industry and for local ecology. While an increase in aerial insectivore activity in the vicinity resulting from insect attraction to turbines remains a strong possibility, little research exists on the possible causes for such events. In this paper, the relative attraction of a selection of specific turbine colours and other hues is assessed in order to determine if turbine paint colour could be influencing insect numbers at these installations. The common turbine colours ‘pure white’ (RAL 9010) and ‘light grey’ (RAL 7035) were among those found to attract significantly more insects than other colours tested, suggesting colour may well have a role to play in potential mitigation. 相似文献

We have analyzed the stability of the cytochrome c-cytochrome b5 and cytochrome c-cytochrome c oxidase complexes as a function of solvent stress. High concentrations of glycerol were used to displace the two equilibria. Glycerol promotes complex formation between cytochrome c and cytochrome b5 but inhibits that between cytochrome c and cytochrome c oxidase. The results with cytochrome b5 and cytochrome c were expected; the association of this complex is largely entropy driven. Our interpretation is that the cytochrome c-cytochrome b5 complex excludes water. The results with the cytochrome c oxidase and cytochrome c couple were not expected. We interpret them to mean that either glycerol is binding to the oxidase, thereby displacing the cytochrome c, or that water is required at this protein-protein interface. A requirement for substantial quantities of water at the interface of some protein complexes is logical but has been reported only once. 相似文献

The macrocyclic lactone ivermectin (Mectizan(R)) is widely used for the control of human filarial infections, particularly as a donated product for onchocerciasis and lymphatic filariasis. In the case of control of lymphatic filariasis in Africa, it is used in combination with donated albendazole. In areas co-endemic for Onchocerciasis and Loa loa, serious adverse reactions have been observed in patients with apparently high microfilaria counts of Loa loa. Recent findings suggest that the severe central nervous system side effects seen in various vertebrates following ivermectin treatment may be due to an absence of, or functional deficiency in P-glycoprotein. P-glycoprotein is expressed in the apical membrane of brain capillary epithelial cells and is responsible for limiting the brain penetration of a range of compounds. Toxicity of ivermectin in some collie dogs may be explained by a 4-bp deletion mutation of the mdr1 gene resulting in a frame shift, generating stop codons that prematurely terminate synthesis of P-glycoprotein. Additionally, sub-populations of CF-1 identified as expressing reduced levels of P-glycoprotein exhibit increased toxicity to substrates of this transporter. Furthermore, while the traditional view of drug-drug interactions is alteration in drug clearance mediated through a change in hepatic drug metabolism, some of these changes may arise through competition for binding sites on P-glycoprotein in the blood-brain barrier, resulting in reduced extracellular efflux and enhanced CNS toxicity. In conclusion, P-glycoprotein is an integral component of the human blood brain barrier and plays a central role in limiting drug uptake into the brain. Altered expression or function of p-glycoprotein could conceivably allow elevation of brain concentrations of ivermectin and produce severe neurotoxicity. This might arise through a genetic polymorphism in p-glycoprotein or co-administration of ivermectin with a drug or foodstuff that might inhibit this efflux transporter. 相似文献

Tetrahydrobiopterin is one of the most potent naturally occurring reducing agents and an essential cofactor required for enzymatic activity of nitric oxide synthase (NOS). The exact role of tetrahydrobiopterin in the control of NOS catalytic activity is not completely understood. Existing evidence suggests that it can act as allosteric and redox cofactors. Suboptimal concentration of tetrahydrobiopterin reduces formation of nitric oxide and favors "uncoupling" of NOS leading to NOS-mediated reduction of oxygen and formation of superoxide anions and hydrogen peroxide. Recent findings suggest that accelerated catabolism of tetrahydrobiopterin in arteries exposed to oxidative stress may contribute to pathogenesis of endothelial dysfunction present in arteries exposed to hypertension, hypercholesterolemia, diabetes, smoking, and ischemia-reperfusion. Beneficial effects of acute and chronic tetrahydrobiopterin supplementation on endothelial function have been reported in experimental animals and humans. Furthermore, it appears that beneficial effects of some antioxidants (e.g., vitamin C) on vascular function could be mediated via increased intracellular concentration of tetrahydrobiopterin. In this review, the potential role of tetrahydrobiopterin in the pathogenesis of vascular endothelial dysfunction and mechanisms underlying beneficial vascular effects of tetrahydrobiopterin will be discussed. 相似文献

Data were evaluated by powerspectrum and transfer function analyses. During tilt, 27 subjectsdeveloped syncope or presyncope and 25 remained asymptomatic. Subjectswith tilt-induced syncope had significantly greater increases inlow-frequency (0.04-0.15 Hz) systolic BP, diastolic BP, and HRpower during tilt than the asymptomatic subjects(P 0.01). This difference waspresent during spontaneous but not paced breathing. However, averagetidal volume, respiratory rate, minute ventilation, proportion ofbreaths below 0.15 Hz, and low-frequency respiratory power during tilt did not differ between syncopal and nonsyncopal subjects. Transfer magnitudes between low-frequency respiration and BP, and between BP andinterbeat interval, were also similar between groups. Thus vasomotorinstability before syncope is not related to alterations in respirationor the cardiovagal baroreflex but may reflect oscillating centralsympathetic outflow to the vasculature. 相似文献

Background
Kounis syndrome (KS) has been described as the coincidental occurrence of acute coronary syndromes during an allergic reaction with cardiac anaphylaxis. It is caused by inflammatory mediators released after exposure to drugs, food, environmental and other triggers. Oxidative stress occurring in various inflammatory disorders causes molecular damage with the production of advanced oxidation products (AOPPs) and advanced glycation end products (AGEs).Case presentation
Markers of oxidative stress were evaluated in a patient who had experienced KS after antibiotic administration in order to investigate the possible role of these molecules in KS. No data, up to now, are available on biomarkers of oxidative stress in patients with drug-induced KS.Conclusions
AOPPs, but not AGEs, were significantly increased in the KS affected patient compared to controls as already reported in mastocytosis affected patients.8.

Pregnancy is characterized by increased plasma adrenocorticotropic hormone (ACTH) and cortisol. Studies suggest that progesterone acts as an antagonist at mineralocorticoid receptors. Therefore, we tested the hypothesis that chronic progesterone, produced by treatment of nonpregnant ewes or during pregnancy, will result in increased plasma ACTH relative to the plasma cortisol concentrations. We studied three groups of ewes: ovariectomized nonpregnant, nonpregnant treated with progesterone, and pregnant ewes. In two series of studies, ewes were adrenalectomized and replaced with 0.35 mg x kg(-1) x day(-1) or 0.5 mg x kg(-1) x day(-1) cortisol. In both studies, aldosterone was infused at 3 microg x kg(-1) x day(-1). In the first study, additional infusions of cortisol over 24 h were used to increase daily replacement doses to 0.5, 1, or 1.5 mg x kg(-1) x day(-1), and intact pregnant and nonpregnant ewes were studied with infusions of cortisol at 0, 0.5, and 1 mg x kg(-1) x day(-1). In adrenalectomized ewes chronically replaced to 0.35 mg x kg(-1) x day(-1) cortisol, plasma ACTH concentrations were decreased significantly in the nonpregnant progesterone-treated ewes compared with the ovariectomized nonpregnant ewes. With 0.5 mg x kg(-1) x day(-1) cortisol, plasma ACTH levels were greater in pregnant ewes than in nonpregnant ewes with or without progesterone. Overall plasma ACTH levels at 0.35 mg x kg(-1) x day(-1) were significantly related to the plasma protein concentration, suggesting that the ACTH levels in the hypocorticoid ewes are most closely related to plasma volume. Across all steroid doses, ACTH was positively related to plasma proteins and progesterone, and negatively related to cortisol. We conclude that increased progesterone does not alter the feedback relation of cortisol to ACTH, but may modulate ACTH indirectly through plasma volume. 相似文献

A large proportion of drinking water is abstracted for treatment from lowland rivers—about 30% in the UK—and this water is at particular risk from sudden and poisonous industrial or agricultural pollution. To cover the range of potential pollutants it may be possible to use biosensors as broadband monitors for toxins. The underlying assumption is that some biological processes, when challenged with a toxin, will be affected in a way analogous to that of man, and that therefore on-line scrutiny of such processes will provide early warning of substances liable to be detrimental to human health. Suitable processes for study might involve multi-cellular organisms, whole cells or enzymes. To date, most practical work has concentrated on fish, but enzymes and single cells hold out the promise of quicker response and, possibly, easier maintenance. 相似文献

Pyroptosis, a type of programmed cell death mediated by gasdermin, is characterized by the swelling and rupture of cells, release of cellular contents and a strong inflammatory response, which is critical for controlling microbial infection. Pattern recognition receptors recognize the intracellular and extracellular pathogenic microbial components and stimulate the organism's inflammatory response by activating the pyroptosis signaling pathway and releasing interleukin-1β (IL-1β), IL-18, and other inflammatory factors to promote pathogen clearance and prevent infection. In the process of continuous evolution, pathogens have developed multiple strategies to modulate the occurrence of pyroptosis and thus enhance their ability to induce disease; that is, the competition between host cells and pathogens controls the occurrence of pyroptosis. Competition can directly affect tissue inflammation outbreaks and even alter cell survival. Studies have shown that various bacterial infections, including Shigella flexneri, Salmonella, Listeria monocytogenes, and Legionella pneumophila, can lead to pyroptosis. Pyroptosis is associated with the occurrence and development of various diseases caused by microbial infection, and the identification of molecules related to the pyroptosis signaling pathway may provide new drug targets for the treatment of related diseases. Extracellular vesicles (EVs) contain specific proteins, lipids, and nucleic acids that can be passed to other cells as signal molecules to alter their function. However, there are many problems and challenges in the conversion and clinical application of EVs. Storage and protection of EVs is one of the issues that need further research. To adapt to potential clinical applications, this type of problem must be solved. This review summarizes the storage practices of EVs in recent years, and explains the impact of temperature on the quality and stability of EVs during storage based on current research, and explains the potential mechanisms involved in this effect as much as possible. 相似文献

During pregnancy, the heavily glycosylated surfaces of the implanting blastocyst and maternal uterine epithelium interact
in a highly controlled and specific manner. Examination of this interface in species that show interdigitation of embryonic
and maternal surfaces (epitheliochorial placentation) shows that each has its own particular pattern of glycosylation or glycotype,
and that closely related and/or interbreeding species e.g. horse and donkey or llama and guanaco, have very similar glycotypes. Implantation of interspecies hybrids is facilitated,
when the blastocyst has an outer cell layer bearing glycans that are compatible with the maternal host. We refer to this mutual
compatibility as a glycocode. The probability that hybrid embryo glycotypes differ from those normally associated with the
host species may account for the high pregnancy failure rates seen in interspecies breeding. We suggest the maternal host
selects between genotypically distinct embryos, and this selection depends partly on cell surface glycosylation. We infer
that the glycocode plays a critical role in implantation, for if the survival of modified genotypes results in fitter offspring
with altered placental glycosylation, selection pressure downstream may in turn act to drive adaptations in the maternal surface
glycotype to produce a complementary glycocode, thus leading eventually to the creation of new species. We speculate that

Nucleic acid structure and dynamics are known to be closely coupled to local environmental conditions and, in particular, to the ionic character of the solvent. Here we consider what role the discrete properties of water and ions play in the collapse and folding of small nucleic acids. We study the folding of an experimentally well-characterized RNA hairpin-loop motif (sequence 5'-GGGC[GCAA]GCCU-3') via ensemble molecular dynamics simulation and, with nearly 500 micros of aggregate simulation time using an explicit representation of the ionic solvent, report successful ensemble folding simulations with a predicted folding time of 8.8(+/-2.0) micros, in agreement with experimental measurements of approximately 10 micros. Comparing our results to previous folding simulations using the GB/SA continuum solvent model shows that accounting for water-mediated interactions is necessary to accurately characterize the free energy surface and stochastic nature of folding. The formation of the secondary structure appears to be more rapid than the fastest ionic degrees of freedom, and counterions do not participate discretely in observed folding events. We find that hydrophobic collapse follows a predominantly expulsive mechanism in which a diffusion-search of early structural compaction is followed by the final formation of native structure that occurs in tandem with solvent evacuation. 相似文献

Prions are multi-stable proteins that can assume at least two stable conformations: a normal one that is detergent-soluble and is protease-sensitive, and a pathogenic one that is protease-resistant and that forms detergent-insoluble fibrillar aggregates. The fibrillar aggregates have been implicated in various neurodegenerative disorders. The normal biological function of prions is not known, yet the high conservation of the sequences of prions among distantly related animals strongly suggests a common and important function. There is now increasing evidence that prions, which are abundant in nervous tissue, may in fact be involved in memory retention. We propose that electrical activity at the synapse induces the surrounding prion molecules to aggregate. The aggregates serve to hold together the synaptic connection between neurons which are interacting at the instant the sensory stimulus is received. The set of neurons connected in this manner then form the neuronal circuit which is associated with the particular stimulus. We propose that the stronger the electrical activity, the greater will be the aggregations. Long-lasting memory will result from traumatic, or exciting, experiences. Further, the aggregations will be maintained, or reinforced, by repeated stimulation of the same set of neurons. Memory loss will occur when those aggregates dissolve. 相似文献
