The metabolism of monohalophenols by Penicillium simplicissimum SK9117, isolated from a sewage plant was investigated. In submerged cultures, 3-, 4-chlorophenol, and 4-bromophenol were metabolized in the presence of phenol. 3-Chlorophenol was transformed to chlorohydroquinone, 4-chlorocatechol, 4-chloro-1,2,3-trihydroxybenzene, and 5-chloro-1,2,3-trihydroxybenzene. With 4-chlorophenol only 4-chlorocatechol was observed as transient product. A release of chloride ions was not observed. Whereas monobromo-, and monochlorophenols could not support growth as sole carbon and energy source, growth and release of fluoride ions were observed with monofluorophenols as substrates. In presence of phenol, the degradation of all monofluorophenols was enhanced. Substrate and cosubstrate disappeared simultaneously. 3-Fluorophenol and 4-fluorophenol were completely mineralized as shown by the equimolar release of fluoride ions.Parts of the results have been presented at the annual meeting of the VAAM in Stuttgart, Germany, March 1995.  相似文献

Chlorocatechol 1,2-dioxygenase (CC 1,2-DO), chloromuconate cycloisomerase (CMCI), chloromuconolactone isomerase (CMLI), and dienolactone hydrolase (DELH), the key enzymes of a new modified ortho-pathway in Rhodococcus opacus 1CP cells utilizing 2-chlorophenol via a 3-chlorocatechol branch of a modified ortho-pathway, were isolated and characterized. CC 1,2-DO showed the maximum activity with 3-chlorocatechol; its activity with catechol and 4-chlorocatechol was 93 and 50%, respectively. The enzyme of the studied pathway had physicochemical properties intermediate between the pyrocatechase of ordinary and chlorocatechase of modified pathways described earlier for this strain. In contrast to the enzymes investigated earlier, CMCI of the new pathway exhibited high substrate specificity. The enzyme had K m for 2-chloromuconate of 142.86 M, V max = 71.43 U/mg, pH optimum around 6.0, and temperature optimum at 65°C. CMCI converted 2-chloromuconate into 5-chloromuconolactone. CMLI converted 5-chloromuconolactone into cis-dienolactone used as a substrate by DELH; this enzyme did not convert trans-dienolactone. DELH had Km for cis-dienolactone of 200 M, V max = 167 U/mg, pH optimum of 8.6, and temperature optimum of 40°C. These results confirm the existence of a new modified ortho-pathway for utilization of 2-chlorophenol by R. opacus 1CP.  相似文献

A mixed microbial community, specially designed todegrade a wide range of substituted aromaticcompounds, was examined for its ability to degrademono-chlorophenols as sole carbon source in aerobicbatch cultures. The mixed culture degraded 2-, 3-, and4 -chlorophenol (1.56 mM) via a meta- cleavagepathway. During the degradation of 2- and3-chlorophenol by the mixed culture, 3-chlorocatecholproduction was observed. Further metabolism was toxicto cells as it led to inactivation of the catechol2,3-dioxygenase enzyme upon meta- cleavage of3-chlorocatechol resulting in incomplete degradation.Inactivation of the meta- cleavage enzyme led toan accumulation of brown coloured polymers, whichinterfered with the measurement of cell growth usingoptical denstiy. Degradation of 4-chlorophenol by themixed culture led to an accumulation of5-chloro-2-hydroxymuconic semialdehyde, themeta- cleavage product of 4-chlorocatechol. Theaccumulation of this compound did not interfere withthe measurement of cell growth using optical density.5-chloro-2-hydroxymuconic semialdehyde was furthermetabolized by the mixed culture with a stoichiometricrelease of chloride, indicating complete degradationof 4-chlorophenol by the mixed culture via ameta- cleavage pathway.  相似文献

The phenol-assimilating yeast Candida maltosa is able to degrade monochlorophenols but cannot grow on these substrates. 3- and 4-chlorophenol were broken down very rapidly by phenol-grown cells under the formation of 4-chlorocatechol, 5-chloropyrogallol and 4-carboxymethylenebut-2-en-4-olide with concomitant release of chloride.2-Chlorophenol was partially converted into cis,cis-2-chloromuconic acid via 3-chlorocatechol which was also obtained from 3-chlorophenol in low amounts. No further metabolites containing chloride were found.The dehalogenation step in the chlorophenol degradation is the cycloisomerization of the cis,cis-chloromuconic acid to 4-carboxymethylenebut-2-en-4-olide in the ortho fission pathway.Dedicated to prof.Dr. E. A bacterium, CP1, identified as Pseudomonas putida strain, was investigated for its ability to grow on and degrade mono-chlorophenols and phenols as sole carbon sources in aerobic shaking batch culture. The organism degraded up to 1.56 mM 2- and 3-chlorophenol, 2.34 mM 4-chlorophenol and 8.5 mM phenol using an ortho-cleavage pathway. P. putida CP1, acclimated to degrade 2-chlorophenol, was capable of 3-chlorocatechol degradation, while P. putida, acclimated to 4-chlorophenol degradation, degraded 4-chlorocatechol. Growth of P. putida CP1 on higher concentrations of the mono-chlorophenols, ≥1.56 mM 4-chlorophenol and ≥0.78 mM 2- and 3-chlorophenol, resulted in decreases in cell biomass despite metabolism of the substrates, and the formation of large aggregates of cells in the culture medium. Increases in cell biomass with no clumping of the cells resulted from growth of P. putida CP1 on phenol or on lower concentrations of mono-chlorophenol. Bacterial adherence to hydrocarbons (BATH) assays showed cells grown on the higher concentrations of mono-chlorophenol to be more hydrophobic than those grown on phenol and lower concentrations of mono-chlorophenol. The results suggested that increased hydrophobicity and autoaggregation of P. putida CP1 were a response to toxicity of the added substrates. The Penicillium strain Bi 7/2 able to grow on phenol as sole source of carbon and energy was isolated from a contaminated soil in Bitterfeld (East Germany). The strain is adapted to high phenol concentrations. Spores germinated still at a phenol concentration of 1.5 g/l. Phenol is degraded by the ortho-pathway with catechol as first intermediary product. The Penicillium strain metabolizes 4-, 3- and 2-chlorophenol with decreasing rates with phenol or glucose as cosubstrate. In the case of 4-chlorophenol 4-chlorocatechol was detected as intermediary product, further degraded as indicated by release of about 35% of the bound chlorine of the aromatic molecule. The strain also cometabolically metabolizes 4-, 3- and 2-nitrophenol. The final product of 3- and 4-nitrophenol is 4-nitrocatechol.  相似文献

Low average temperatures and temperature fluctuations in temperate soils challenge the efficacy of microbial strains used for clean up of pollutants. In this study, we investigated the cold tolerance of Arthrobacter chlorophenolicus A6, a microorganism previously shown to degrade high concentrations of 4-chlorophenol at 28°C. Luciferase activity from a luc-tagged derivative of the strain (A6L) was used to monitor the metabolic status of the population during 4-chlorophenol degradation. The A6L strain could degrade 200–300 g mL–1 4-chlorophenol in pure cultures incubated at 5°C, although rates of degradation, growth and the metabolic status of the cells were lower at 5°C compared to 28°C. When subjected to temperature fluctuations between 5 and 28°C, A6L continued to degrade 4-chlorophenol and remained active. In soil microcosm experiments, the degradation rates were significantly faster the first week at 28°C, compared to 5°C. However, this difference was no longer seen after 7 days, and equally low 4-chlorophenol concentrations were reached after 17 days at both temperatures. During 4-chlorophenol degradation in soil, CFU and luciferase activity values remained constant at both 5 and 28°C. However, once most of the 4-chlorophenol was degraded, both values decreased by 1–1.5 logarithmic values at 28°C, whereas they remained constant at 5°C, indicating a high survival of the cells at low temperatures. Because of the ability of A. chlorophenolicus A6 to degrade high concentrations of 4-chlorophenol at 5°C, together with its tolerance to temperature fluctuations and stress conditions found in soil, this strain is a promising candidate for bioaugmentation of chlorophenol-contaminated soil in temperate climates.This revised version was published online in November 2004 with corrections to Volume 48.  相似文献

The effect of temperature and oxygen on diazotrophic growth of the thermophilic cyanobacterium HTF (High Temperature Form) Chlorogloeopsis was investigated using cells grown in light-limited continuous culture at a dilution rate of 0.02 h-1. Diazotrophy was more sensitive to elevated temperatures than growth with combined nitrogen. The maximum temperature for growth of cultures gassed with CO2-enriched air was more than 55 °C but less than 60 °C with N2 as the sole nitrogen source, but between 60°C and 65°C when nitrate was present in the medium. The effect of temperature on nitrogenase activity, photosynthesis and respiration in the dark was determined using cells grown at 55°C. Maximal rates of all three processes were observed at 55°C and rates at 60°C during shortterm incubations were not less than 75% of the maximum. However, nitrogenase activity at 60°C was unstable and decayed at a rate of 2.2 h-1 under air and at 0.3 h-1 under argon. Photosynthesis and respiration were more stable at 60°C than anoxic nitrogen fixation. The ability of strains of the genusRhodococcus to transform chlorinated phenolic compounds was studied. Noninduced cells of several strains ofRhodococcus, covering at least eight species, were found to attack mono-, di-, and trichlorophenols by hydroxylation at theortho position to chlorocatechols. 3-chlorophenol and 4-chlorophenol were converted to 4-chlorocatechol, 2,3-dichlorophenol to 3,4-dichlorocatechol, and 3,4-di-chlorophenol to 4,5-dichlorocatechol. The chlorocatechols accumulated to nearly stoichiometric amounts. Other mono- and dichlorophenols were not transformed. The ability of the strains to hydroxylate chlorophenols correlated with the ability to grow on unsubstituted phenol as the sole source of carbon and energy. SeveralRhodococcus strains attacked chlorophenolic compounds by both hydroxylation and O-methylation. 2,3,4-, 2,3,5- and 3,4,5-trichlorophenol were hydroxylated to trichlorocatechol and then sequentially O-methylated to chloroguaiacol and chloroveratrole. Tetrachlo-rohydroquinone was O-methylated sequentially to tetrachloro-4-methoxy-phenol and tetrachloro-1,4-dimethoxybenzene. Several of the active strains had no known history of exposure to any chloroaromatic compound. Rhodococci are widely distributed in soil and sludge and these results suggest that this genus may play an important role in transformation of chlorinated phenolic compounds in the environment.  相似文献

When Lactococcus lactis strains were exposed directly to the lethal temperature of 50 C for 30 ;min, 0.1–31% of the cells survived. However, when pre-exposed to 40 °C, prior to exposure at 50 °C, 4–61% of the cells survived. A plasmid carrying a unique heat shock gene from the thermophile Streptococcus thermophilus was cloned into L. ;lactis. When the transformed cells were cultivated at 30 °C the introduction of the plasmid had no obvious effect on the growth of L. ;lactis. However, when the temperature was abruptly shifted from 30 °C to 42 °C at mid-growth phase the growth decreased by 50%.  相似文献

Summary Pseudomonas putida MST, previously isolated in the presence of -methylstyrene, has been shown to transform several substituted aromatic compounds. It was able to modify halogenated aromatic compounds by co-oxidation. It regiospecifically hydroxylates chlorobenzene and 2-chlorophenol to 3-chlorocatechol, and 4-chlorophenol to 4-chlorocatechol; both metabolites were identified in the cultures.  相似文献

Transplasma membrane electron transport activity by fetal rat liver cells (RLA209-15) infected with a temperature-sensitive strain of SV40 has been measured with cells grown at the restrictive temperature (40°C) and permissive temperature (33°C). The transformed cells grown at 33°C had only one-half the rate of external ferricyanide reduction as the nontransformed cells held at 40°C. Both theK m andV max for ferricyanide reduction were changed in the transformed state. The change inV max can be based on a decrease of NADH in the transformed cells. The change in rate with ferricyanide does not depend on change in surface charge. Reduction of external ferricyanide was accompanied by release of protons from the cells. The ratio of protons released to ferricyanide reduced was higher in the transformed cells than in the non-transformed cells. Since the transplasma membrane electron transport has been shown to stimulate cell growth under limiting serum, the changes in the plasma membrane electron transport and proton release in transformed cells may relate to modification of growth control.  相似文献

It has recently been documented that, compared to untransformed controls, the roots of oilseed rape (Brassica napus L. CV CrGC5) seedlings transformed by Agrobacterium rhizogenes A4 show a reduced gravitropic reaction (Legué et al. 1994, Physiol Plant 91: 559–566). After stimulation at 90°C or 135°, the transformed root tips curve, but never reach a vertical orientation. In the present study, we investigated the causes of reduced gravitropic bending observed in stimulated transformed root tips. First, we localized the gravitropic curvature in normal and in transformed roots after 1.5 h of stimulation. The cells involved in root curvature (target cells) corresponded at the cellular level to the apical part of the zone of increasing cell length. In transformed roots grown in the vertical position, these cells showed a reduction in cell length compared to controls. Because auxin is considered to be the gravitropic mediator, the response of normal and transformed roots to exogenous auxin was studied. Indole-3-acetic acid (IAA) was applied along the first 3 mm using resin beads loaded with the hormone. In comparison to normal roots, transformed roots showed reduced bending toward the bead at all points of bead application. Moreover, the cells which responded to IAA corresponded to the target cells involved in the gravitropic reaction. The level of endogenous IAA was lower in transformed roots. Photosynthetic activity, in leaf slices and isolated thylakoids, was examined at 25° C after preincubation of the slices at either 25° C or 4° C at a moderate photon flux density (PFD) of 450 mol·m–2·s–1, or at 4° C in the dark. The plants used wereSpinacia oleracea L.,Cucumis sativus L. andNerium oleander L. which was acclimated to growth at 20° C or 45° C. The plants were grown at a PFD of 550 mol·m–2·s–1. Photosynthesis, measured as CO2-dependent O2 evolution, was not inhibited in leaf slices from any plant after preincubation at 25° C at a moderate PFD or at 4° C in the dark. However, exposure to 4° C at a moderate PFD induced an inhibition of CO2-dependent O2 evolution within 1 h inC. sativus, a chilling-sensitive plant, and in 45° C-grownN. oleander. The inhibition in these plants after 5 h reached 80% and 40%, respectively, and was independent of the CO2 concentration but was reduced at O2 concentrations of less than 3%. Methyl-viologen-dependent O2 exchange in leaf slices from these plants was not inhibited. There was no photoxidation of chlorophyll, in isolated thylakoids, or any inhibition of electron transport at photosystem (PS)II, PSI or through both photosystems which would account for the inhibition of photosynthesis. The conditions which inhibit photosynthesis in chilling-sensitive plants do not cause inhibition inS. oleracea, a chilling-insensitive plant, or in 20° C-grownN. oleander. The CO2-dependent photosynthesis, measured at 5° C, was reduced to about 3% of that recorded at 25° C in chilling-sensitive plants but only to about 30% in the chilling-insensitive plants. Methyl-viologen-dependent O2 exchange, measured at 5° C, was greater than 25% of the activity at 25° C in all the plants. The results indicate that the mechanism of the chilling-induced inhibition of photosynthesis does not involve damage to PSII. Four bacterial strains (CA26, CA28, CA37, and CA45), which all were able to use aniline, 3-chloroaniline (3-CA), and 4-chloroaniline (4-CA) as sole sources of carbon, nitrogen and energy, were isolated after enrichment in aerated soil columns and identified as Pseudomonas acidovorans strains. In addition strains CA26 and CA45 were able to degrade 2-chloroaniline (2-CA) at very low rates. At 25°C strain CA28 was grown on aniline and 3-CA with generation times of 3.0 and 7.7 h, respectively, and exhibited complete mineralization of these substrates in degradation rates of 2.25 mmol aniline and 1.63 mmol 3-CA g-1 of biomass per hour, respectively. Degradation of 4-CA occurred at 1.54 mmol 4-CA g-1 of biomass per hour and a generation time of 18.7 h but, in contrast, was not complete due to formation of minor amounts of chlorohydroxymuconic semialdehyde, a meta-cleavage product of 4-chlorocatechol. The initial attack on the substrate, the formation of corresponding chlorocatechols from 3-CA and 4-CA, was found to be the rate-limiting degradation step. Evidence for two different aniline-oxygenase systems in strain CA28 with distinct activity pattern on chlorinated and nonsubstituted anilines was demonstrated by oxygen uptake rate experiments with aniline and chloroaniline pregrown cells. The degradative properties towards chlorobenzoates by a 2-chlorobenzoate-degrading mixed culture (2MC) were investigated. Although 2MC did not grow on 2,3,5-; 2,3,6- and 2,4,6-trichlorobenzoates, it was able to completely oxidise 2,3,5-trichlorobenzoate and 2,5-dichlorobenzoate. This was degraded through the intermediate formation of 4-chlorocatechol and 3-cis,cis-chloromuconate, suggesting a dioxygenation in 1,2 position. 4-Chlorobenzoate; 3,4- and 2,6-dichlorobenzoate; 2,4,6- and 2,3,6-trichlorobenzoate did not undergo any co-metabolic transformation, the only 2,4-dichlorobenzoate was scarcely transformed into a dichlorinated phenol as end-product. 2MC was constituted of three prominent species: Stenotrophomonas maltophilia, Cupriavidus necator and Flavobacterium sp. Ring-hydroxylating dioxygenase genes (BenA) were retrieved in S. maltophilia and C. necator, indicating the presence of a “modified ortho pathway” in the culture. On the basis of its degradative properties, 2MC could be a good candidate for use in the removal of ortho- and/or meta-chlorobenzoates that are the major end-products of PCB co-metabolism.  相似文献

Inhibition and recovery of net CO2 uptake and three photosynthetic electron transport reactions as well as plant survival following high-temperature treatments were investigated for Opuntia ficus-indica. For plants maintained at 30°C/20°C day/night air temperatures, treatment at 60°C for 1 h irreversibly inhibited net CO2 uptake and photosynthetic electron transport, resulting in plant death in about 60 days. When a plant maintained at 30°C/20°C was treated at 55°C for 1 h, net CO2 uptake was completely inhibited 1 d after the treatment but fully recovered in 60 d. Differential inactivation of photosystem (PS) I, PSII, and whole chain electron transport activities occurred; PSI was the most tolerant of 55°C and took the least time (45 d) for total recovery. All 30°C/20°C plants survived a 1-h treatment at 55°C, although some pale green areas were observed on the cladode surfaces. In contrast to growing at 30°C/20°C, plants acclimated to 45°C/35°C survived 60°C for 1 h without showing any necrotic or pale green areas on the cladode surfaces. When such a plant was transferred to 30°C/20°C following the high-temperature treatment, recovery in net CO2 uptake began in 1 d and progressed to complete recovery by 30 d. NIN1 is an essential gene for growth of the yeastSaccharomyces cerevisiae and was recently found to encode a component of the regulatory subunit of the 26S proteasome. Thenin1-1 mutant is temperature sensitive and its main defect is in G1/S progression and G2/M progression at non-permissive temperatures. One of the two multicopy suppressors ofnin1-1, SUN2 (SUppressor of Nin1-1), was found to encode a protein of 523 amino acids whose sequence is similar to those ofDrosophila melanogaster diphenol oxidase A2 and the mouse mast-cell Tum transplantation antigen, P91A. The C-terminal half of Sun2p was found to be functional as Sun2p at 25° C, 30° C, and 34° C but not at 37° C. The open reading frame (ORF) of theDrosophila diphenol oxidase A2 gene (Dox-A2) was obtained from a lambda phage cDNA library using the polymerase chain reaction technique. TheDox-A2 ORF driven by theTDH3 promoter complemented the phenotype of a strain deleted forsun2. ThisDox-A2-dependent strain was temperature sensitive and accumulated dumb-bell-shaped cells, with an undivided nucleus at the isthmus, after temperature upshift. This morphology is similar to that ofnin1-1 cells kept at a restrictive temperature. These results suggest thatSUN2 is a functional counterpart ofDox-A2 and that these genes play a pivotal role in the cell cycle in each organism.  相似文献

Rhodospirillum rubrum grown either chemotrophically or phototrophically at 14°C and 30°C, was employed to study the effect of temperature on fatty acid composition as well as on several membrane bound functions involved in energy metabolism. Upon growth at both temperatures the fatty acid composition of membranes showed differences, which could be attributed to an incomplete formation of photosynthetically active membranes rather than specifically to the growth temperature. Activities of NADH dependent respiration and light induced proton extrusion by cells did not show discontinuities in Arrhenius plots down to temperatures of 15°C and 5°C, respectively. In contrast, coupling factor Mg2+- and Ca2+-ATPase as well as succinate cytochrome c oxidoreductase showed significant breaks at 20°C and 18°C, respectively. Similarly, in Rhodopseudomonas sphaeroides. NADH dependent respiration and light induced proton extrusion by cells was continuous over the entire range of temperatures applied. ATPase as well as succinate cytochrome c oxidoreductase, on the other hand, featured discontinuities in Arrhenius plots at 20°C and 19°C.
