In Caryedon serratus (Coleoptra; Bruchidae) a large spermatophore is deposited into the female bursa copulatrix during mating. After mating, the size of the spermatophore decreases and the central part becomes vacuolated. Electrophoretic studies show quantitative but not qualitative modifications of the spermatophore secretions. Within 24 h after mating, some male secretions are transferred from the spermatophore to the haemolymph of females as shown by immunoblotting and autoradiographic studies. Some of these secretions or their derivatives are observed in mature oöcytes. This production constitutes a heavy investment on the part of the males. Mating stimulates vitellogenesis and egg laying and it is possible that the male investment represents a contribution to ovarian production. In the absence of food during imaginal life, the number of matings influences the female's reproductive capacity. The supply of male secretions thus constitutes an important trophic contribution that increases females' fecundity and vitellogenesis. In the presence of food (pollen), repetitive matings do not influence the female's reproduction level. The influence of this trophic source and the extent of its importance are analyzed in this study.  相似文献

Newly eclosed Drosophila melanogaster females contain only previtellogenic stage oöcytes and no immunologically detectable female specific haemolymph protein. During the subsequent 48 hr the concentration of female specific protein in the haemolymph rises to a plateau value of 21 μg/μl; at this time yolk protein represents about one third of the total haemolymph protein in adult females. The first mature (stage 14) oöcytes are observed at 48 hr post eclosion. The female specific haemolymph protein and the major protein from mature oöcytes are electophoretically and immunologically the same or very similar. Injection of alpha amanitin into newly eclosed females inhibits the development of mature oöcytes and the degree of inhibition depends on the age of the female at the time of injection. Phenocopies of non-vitellogenic mutants result when alpha amanitin is injected into newly eclosed females; after 36 hr post eclosion no visible inhibition of vitellogenesis (as observed morphologically at 72 hr post eclosion) can be produced by alpha amanitin.  相似文献

In Acanthoscelides obtectus, some male secretions deposited in the spermatophore during mating reach the blood of the females and stimulate oögenesis. Water extracts from spermatophores injected into a female abdomen stimulate oögenesis but do not influence egg-laying or sexual receptivity. After column chromatograph of spermatophores, aqueous extracts on Sephadex G 25 Coarse, G 25 Superfine, and G 15, an active fraction has been isolated. This injected into the abdomen of virgin females stimulates oögenesis at low concentrations, but it is toxic at higher concentrations. This fraction was examined by paper electrophoresis at low voltage and then chromatographed on G 10 Sephadex. Two peaks were obtained: the first corresponds to the paragonial substance A which stimulates oögenesis at 0,2 10?3 μg/μl concentration. The second contains the paragonial substance B. At a 0,3 10?3 ug/μl concentration this substance is toxic. First this toxicity inhibits oögenesis and then causes the death of most females at higher concentrations. The toxic effect appears 2 or 3 days after injection. These two substances are purified on paper chromatography and the biological activities are contained in a zone of Rf 0.25 to 0.45 (paragonial substance A) and in a zone of 0.16–0.30 Rf (paragonial substance B).The paragonial substances disappear from the spermatophore after mating. Aqueous extracts of spermatophores obtained 6 hr after mating do not stimulate oögenesis and do not have any toxic effect. The chemical nature of these both fractions is not yet determined because the quantity of extracts obtained at the end of the purification is very low.The action of both paragonial substances is similar to the action of hormones. The paragonial substances influence unknown receptors at low concentration after a latent period. The origin of the paragonial B substance was not determined, but this substance which inhibits oögenesis at low concentrations could be an antagonist of paragonial A substance.  相似文献

D. melanogaster females homozygous for the ap4 mutant synthesize yolk protein and circulate this protein in the haemolymph at concentrations not different from concentrations found in normal females. However, ap4 females deposit little or no yolk protein into developing oöcytes. Topical application of a juvenile hormone analogue (JHA), ZR-515, stimulated sequestration of yolk protein by developing oöcytes of ap4 females. JHA had no detectable effects on haemolymph concentrations of yolk protein in either normal or ap4 females nor on the protein profiles obtained from electrophoresis of haemolymph samples.  相似文献

Juvenile hormone (JH) synthesis by the corpora allata of gregarious and solitarious phase females of Schistocerca gregaria was determined in vitro during the penultimate and last stadia as well as during the first gonotrophic period of adults. Generally, the corpora allata of solitarious females showed higher rates of JH synthetic activity. In addition, in adult females there was a temporal difference between the corpora allata activities of gregarious and solitarious locusts, the latter exhibiting relatively higher rates of JH synthesis early in the first gonotrophic period. The corpus allatum volumes of solitarious females were also generally larger than those of their gregarious counterparts; there was no synchrony between fluctuations in JH synthetic activity and changes in corpus allatum volume in either phase.The early onset of relatively high JH synthetic rates in solitarious females was correlated with the early detection, by rocket immunoelectrophoresis, of vitellogenin in the haemolymph and vitellin in the oöcytes. Vitellogenin appeared in the haemolymph on day 4 in solitarious females and on day 6 in gregarious females and vitellin appeared in the oöcytes on days 6 and 8 respectively. Oöcyte length at which vitellogenesis was first detected was 1.8 mm for gregarious and 1.3 mm for solitarious females. However, despite the accelerated onset of both vitellogenin synthesis and uptake, oöcyte maturation time of solitarious females was longer. In both gregarious and solitarious females, vitellogenin titres increased until oöcytes reached a length of about 4 mm and declined thereafter. Vitellin content of ovaries increased proportionately to oöcyte growth until they attained a length of 5.0 mm. The subsequent increase in length of oöcytes to maturity is attributed to postvitellogenic growth, possibly by hydration.  相似文献

Egg chambers from apterous4 (ap4), a female sterile mutant of Drosophila melanogaster, show none of the microvilli or pinocytotic vesicles which are a prominent feature of the membrane of the wild-type vitellogenic oöcyte. The studies reported here show that a juvenile hormone analogue (ZR515) stimulates formation of microvilli and pinocytotic vesicles in oöcytes of ap4 flies. Within 12 hr after topical application of ZR515 to homozygous ap4 females the oöcyte membranes exhibit extensive microvilli and pinocytotic activity. The follicle-cell surface adjoining the oöcyte also shows some changes. In vitro studies in which ap4 ovaries were incubated in Schneider's Drosophila tissue-culture medium in the presence of ZR515 with or without female haemolymph, or in the absence of ZR515, showed that the analogue acts alone directly on the ovary to cause formation of microvilli and pinocytotic vesicles on the oöcyte membrane.  相似文献

Nulliparous females of a normal anautogenous strain of Lucilia cuprina mature all of their primary oöcytes after feeding ad lib on sheep's liver. Females fed measured inadequate amounts of protein-rich materia either fail to mature any oöcytes or mature less than their full complement. These mature oöcytes are smaller than in ad lib fed females. In females maturing no oöcytes, ovarian development ceases with all oöcytes in a pre-vitellogenic or early vitellogenic stage. When females mature only some of their oöcytes, the remainder are resorbed in early vitellogenesis. Few females mature less than 100 oöcytes, if they mature any at all.  相似文献

The corpora allata of castrated females of Nauphoeta grow only very slightly and do not reach a volume greater than that of the glands of normal females during gestation. These small corpora allata are, however, active and are responsible for the synthesis of vitellogenin (female specific protein) in large amounts. Besides vitellogenin the other haemolymph proteins are also synthesized and accumulated in the haemolymph in much higher concentrations than in normal females. Implanted oöcytes grow in castrated as well as in normal females at about the same rate until the tenth day of the oöcyte maturation period. Thereafter they only grow in castrated females. If castrated and normal females are decapitated, their protein content decreases. At the same time the growth stimulating capacity of their haemolymph decreases at a much faster rate. If oöcytes are implanted in castrated and decapitated females after 4 days they cannot grow any more although the vitellogenin titre of the haemolymph is still much higher than it is at any time in normal females. It can be concluded that vitellogenin alone cannot induce oöcyte growth and that juvenile hormone is necessary as well for vitellogenin synthesis as for its incorporation into the oöcytes. However, in insects rich in vitellogenin juvenile hormone leads to a more rapid oöcyte growth than in insects containing only small amounts of this protein.  相似文献

The haemolymph protein concentration in Bombyx mori decreases normally by about one-fourth during pharate adult development. In females homozygous for the small egg gene, the concentration of haemolymph protein remained constant throughout the pupal and pharate adult stages. The sm gene does not influence the synthesis of vitellogenic female protein of pupal and pharate adult haemolymph (FP). Normal ovaries transferred to the haemocoele of sm females undergo normal vitellogenesis. In the absence of normal alleles of sm, the ovaries encounter difficulties in the incorporation of FP into their oöcytes from pharate adult haemolymph. These results suggest that an active translocation mechanism is involved in the transfer of haemolymph protein into the ovaries.  相似文献

Abstract Female Caloptilia fraxinella exhibit a prolonged reproductive diapause immediately post adult emergence in mid‐summer until the next spring when mating, egg development and oviposition on fresh Fraxinus spp. leaflets occur. Factors that effect the termination of reproductive diapause are investigated in this species. Caloptilia fraxinella diapausing adults held in overwintering conditions (2 °C, LD 0 : 24 h) for 24 weeks terminate diapause after placement for 2 weeks in simulated summer conditions (24 °C, LD 16 : 8 h) only if they are provided with 10% sugar water. Exogenous application of the Juvenile Hormone (JH) analogue methoprene to moths in both early‐ (summer) and mid‐ (autumn) reproductive diapause demonstrates that JH affects diapause termination but a carbohydrate nutrition source also mediates mating and vitellogenesis. Mating between moth pairs early in diapause occurs only after treatment with methoprene and provision with sugar water. However, there is no impact of mating on the propensity of females to produce vitellogenic oöcytes. Moths collected in the autumn in mid‐diapause respond in a dose‐dependent fashion to methoprene treatment and the response is greater than that of moths early in diapause collected in the summer. Treatment with methoprene and access to sugar water results in vitellogenic oöcytes in 18.75% of females from mid‐diapause moth pairs treated with 0.01 μg methoprene per insect and in all females from pairs treated at the two highest doses of methoprene (0.1 and 1 μg per insect). Mating occurs only between moths in mid‐diapause treated with the two highest doses of methoprene and these doses induce 91% and 100% mating, respectively. Both control and methoprene‐treated males in mid‐diapause held under summer conditions mate successfully and pass a spermatophore to their methoprene‐treated female partner. These data demonstrate that female C. fraxinella undergo a prolonged reproductive diapause in which termination is dependent on JH and further mediated by a carbohydrate nutrition source. The production of vitellogenic oöcytes is independent of mating. These data also provide evidence that response of moths in diapause to exogenous applications of methoprene differs throughout the diapause period and between male and female C. fraxinella.  相似文献

During normal oögenesis in many insects some of the oöcytes fail to mature; instead they degenerate and are resorbed. In this work oöctte degeneration was investigated in Drosophila melanogaster females and found to be limited to early vitellogenic stages (stages 8–10). Even when retained for up to 18 days by females, mature (stage 14) oöcytes showed unaltered protein patterns after separation by SDS polyacrylamide electrophoresis, indicating that protein breakdown, which is characteristic of degeneration, does not occur in chorionated oöcytes.A number of environmental parameters were shown to influence the percentage of degenerating oöcytes in females. Strong responses as reflected by increased stage-8 and 9 oöcyte degeneration were found in females subjected to suboptimal (but not starvation) medium, virgin females, females mechanically unable to oviposit, and females unable to locate suitable oviposition sites. Little or no response was seen in females subjected to crowding, however, since all of these environmental parameters except adult crowding have been shown to decrease fecundity, and therefore the rate of oöcyte production, it is suggested that oöcyte degeneration is a strategy for decreasing the rate of oöcyte production in Drosophila.  相似文献

Haemolymph and fat body soluble protein titres have been examined during the reproductive cycle of Diploptera punctata, with particular emphasis on the occurrence of vitellogenin and its uptake into the developing oöcytes. Vitellogenin was first detected in the haemolymph of mated females 2 days after adult eclosion at about the same time that vitellin deposition in basal oöcytes began. Peak haemolymph titres of vitellogenin occurred on day 6, correlated with the completion of yolk uptake. Thereafter vitellogenin levels declined and were generally undetectable throughout most of gestation, rising again shortly before parturition in association with the second gonotrophic cycle. Total haemolymph protein levels were not correlated with vitellogenesis.Soluble fat body vitellogenin titres of mated females remained low during the first oöcyte growth period but then rose several-fold at its completion and remained high throughout pregnancy and the second gonotrophic cycle. Total fat body soluble proteins decline after adult eclosion in association with oöcyte growth.Vitellin accumulation in basal oöcytes was related linearly to increase in volume until the onset of chorion formation. Thus no post-vitellogenic growth period was detected.  相似文献

RNA synthesis and morphological changes in the follicular epithelial cells of oöcytes of Douglas-fir beetle, Dendroctonus pseudotsugae were studied during the reproductive phase. Inhibition of synthesis of DNA dependent RNA by actinomycin D injections blocked yolk deposition in the oöcytes as well as oviposition within the normal period. A mixture of radioactively labelled haemolymph and ovarial proteins was deposited as yolk proteins in the oöcytes of normal beetles. Such proteins were not deposited in the oöcytes of females injected with actinomycin D; the blockage of yolk deposition persisted even when such females were treated extraneously with juvenile hormone.  相似文献

Menida scotti (Puton) males have been shown to transfer secretions from their bulbus ejaculatorius and reservoir of ectodermal accessory gland to females by mating during hibernation. In the present study, the major components of the secretions were found to be proteins and lipids. To specify the female organ incorporating the male secretions, a radiotracer experiment in which the male secretions were labeled by [14C]valine was conducted in nine tissues of females collected in the fall and spring of the hibernation period. Relatively high radioactivities were detected in the haemolymph and the residual carcass (head, legs, air-sacs, exoskeleton, etc.) in the fall females, and in CO2 gas evolved and carcasses in the spring females. The radioactivities in the fat body were significantly higher in the fall mating females than in the spring mating females, and vice versa in the ovary. The radioactivities in six fractions (lipids, proteins, glycogen, sugars, free amino acids and the residues) were also assayed in the five organs of females that had a relatively high radioactivity. The highest radioactivity was detected in the protein fraction of the haemolymph in fall and spring females. There were significant differences in the radioactivities incorporated into the lipid fractions of the carcass between fall and spring females.  相似文献

Juvenile hormone (JH) synthesized and secreted in vitro by the corpora allata of mated adult Leucophaea maderae females was determined to be JH III (methyl-10,11-epoxy-3,7,11-trimethyl-2,6-dodecadienoate).The haemolymph titre of JH was determined during maturation of the terminal oöcytes in the first reproductive cycle of L. maderae. In virgin females, JH is not detectable in the haemolymph during the first eight days following adult emergence; however, by 10 days after emergence, trace quantities of JH are apparent. Mating stimuli induce a dramatic increase in the concentration of haemolymph JH, with a peak occurring approximately 12 days after mating; thereafter, the JH concentration declines until it has reached an undetectable level 19 days after mating, at the time of chorion deposition.During ovarian maturation, changes in the rates of synthesis of vitellogenin by the fat body and DNA by the ovary correlate closely with the haemolymph titre of JH. However, no such correlation exists between the JH titre and the extensive ovarian protein synthesis that occurs in L. maderae coincident with chorion formation.The effects of JH I and JH III on both vitellogenin synthesis and ovarain DNA synthesis are statistically similar.  相似文献

The transfer during copulation of radioactively labelled male accessory reproductive gland (ARG) protein and its accumulation by the ovary of Melanoplus sanguinipes have been studied. Most of the transferred material leaves the spermatheca within 24 hr and enters the haemolymph from which it can be accumulated by the ovary. Injection of labelled male ARG protein into vitellogenic females demonstrates that during the first 24 hr after injection, accumulation by the ovary is rapid. Immunoprecipitation and immunoelectrophoretic studies indicate that some of the ARG protein is accumulated unchanged. It is proposed that when the male transfers several spermatophores during copulation, he may make a significant contribution of protein to the developing oöcytes.  相似文献

During the early stages of vitellogenesis in Leucophaea, vitellogenin accounted for most if not all of the secreted protein synthesized by the fat body. Synthesis began about 5 days after mating and continued until 24 hr or so before the formation of the oötheca. Ligation resulted in the degeneration of the oöcytes, the first evidence of which was seen within 24 hr. Ligation also curtailed the synthesis of vitellogenin at about the same time. Isolated abdomens treated with an analog of juvenile hormone commenced vitellogenesis within 12 to 24 hr and measureable oöcyte growth occurred after 5 days. Despite continued synthesis of vitellogenin, the oöcytes in isolated abdomens always degenerated.  相似文献

The relationships between the release of factors from the head after blood-feeding, subsequent levels of ecdysteroids and vitellin, and the ultimate maturation of eggs in Aedes aegypti were investigated. Females were decapitated at various times after a blood meal, at 20 or 48 h after feeding the animals were dissected and divided into two groups, those with arrested oöcytes (yolk length < 100 μm) and those with maturing oöcytes (yolk length > 100 μm). These yolk lengths correspond with the levels of oöcyte growth believed to accompany the proposed initiation and promotion phases of egg development. Animals dissected at 20 h were assayed for ecdysteroid by radioimmunoassay; those dissected at 48 h were assayed for vitellin by rocket immunoelectrophoresis.Non-blood-fed unoperated females contained 8% as much ecdysteroid as blood-fed controls and no measurable vitellin. Females with arrested oöcytes (< 100 μm) were obtained only if decapitations were performed before 8 h; these females had about 20% of the ecdysteroids and 8% of the vitellogenin normally found in blood-fed animals. Females decapitated between 2 and 8 h with maturing oöcytes contained 50–60% as much ecdysteroid and vitellin as blood-fed unoperated controls. Normal ecdysteroid and vitellin levels were reached only when decapitations were delayed for 12 and 24 h, respectively. The number of developing oöcytes was also decreased by early decapitation and was closely correlated with vitellin levels.We conclude that the egg development neurosecretory hormone is released twice, once before 8 h and once after 8 h, to control ecdysteroid levels. We also suggest the presence of other factors from the head that control vitellin levels, the number of developing oöcytes, and the early growth of the oöcyte (initiation).  相似文献

In females of the Blaberidae sexual receptivity is inhibited during the preoviposition period and during gestation. Similarly affected are oöcyte development and corpora allata activity during pregnancy. Roth (1964b) believes causation to be due to the mechanical action of the oötheca, but Engelmann (1964) believes it is due to the intervention of a humoral agent.In normal females of Blabera craniifer sexual receptivity is linked to the presence or absence of the oötheca in the brood sac. If the oötheca is not placed in the brood sac receptivity appears sooner than in a pregnant female.If certain afferent pathways are suppressed (by severing the nerve cord), or upset (by anaesthesia or in surgical controls) during pregnancy, only a partial receptivity is shown: straddling of the female on the male abdomen, ‘licking’ of tergal secretions, but no insertion by the male occurs. Such reactions are repeated over several days, giving an alternation of partial receptivity/non-receptivity which differs from that shown in normal animals. At times desynchronization between the receptivity and laying cycles is observed. This leads to the same conclusion as Roth and Barth (1964) that receptivity is not directly controlled by the corpora allata. We believe that the control ‘centre’ of receptivity has an autonomous rhythm, which is normally hidden but which appears when the afferent pathways are lacking or upset. The control mechanism of mating behaviour is probably a neuro-endocrine mechanism.  相似文献

Female grasshoppers of acoustically communicating species assume series of reproductive states that are associated with particular behaviours. Studies on laboratory populations of Chorthippus biguttulus (L.) revealed that females of this species lack the period of ‘passive copulatory readiness’, increase their attractiveness to males by sound production and mate multiple times before their first oviposition. In particular, female Ch. biguttulus display a period of ‘primary rejection’ after their imaginal moult during which they reject male mating attempts followed by a period of ‘active copulatory readiness’ in which they produce acoustic signals and may copulate with courting males. Female stridulation generally stimulated male mating activity and stridulating females attracted more male mating attempts than mute females in the same cage, indicating that males preferentially court females that signal ‘active copulatory readiness’. After receipt of a spermatophore, Ch. biguttulus females displayed periods of ‘secondary rejection’ followed by re-establishment of ‘active copulatory readiness’. Acoustic responses of females to male songs, an indicator of reproductive readiness, were significantly reduced until 2 days after mating and remained slightly reduced in comparison to pre-mating levels. Some females mated multiple times before their first oviposition and cycled between ‘secondary rejection’ and ‘active copulatory readiness’.  相似文献
