Larvae of the elm bark beetle, Scolytus scolytus, were inoculated with conidia of the entomogenous fungi Beauveria bassiana (two strains), Metarhizium anisopliae (two strains), and Paecilomyces farinosus (two strains) and incubated over a range of temperatures (2°, 6°, 10°, 15°, and 20°C). One strain each of B. bassiana and P. farinosus caused infection even at 2°C, whereas the two strains of M. anisopliae caused no infection below 10°C. Infection of adult beetles by B. bassiana (one strain) and M. anisopliae (one strain) was tested at 15°, 20°, and 25°C (B. bassiana) and at 15° and 20°C (M. anisopliae). Fungal infection occurred at all three temperatures, but at 25°C beetles tended to succumb to bacterial infection. The effect of relative humidity on infection of larvae by B. bassiana (one strain), M. anisopliae (one strain), and P. farinosus (one strain) was tested at 51, 74, 86, 90, 95, 97.5, and 100% relative humidity. B. bassiana and M. anisopliae caused some infection at all humidities: with P. farinosus there was no infection at the two lowest humidities. Mortality due to infection by these fungi was most rapid at the highest humidities.  相似文献

Bioassays were conducted to determine the susceptibility of egg masses of Mamestra brassicae and Spodoptera littoralis to different spore doses of Paecilomyces fumoso-roseus and Nomuraea rileyi at 20° and 25°C. P. fumoso-roseus was highly virulent against eggs, whereas N. rileyi provoked only a deferred mortality of larvae hatched from treated eggs. Nevertheless, larval mortality of S. littoralis caused by N. rileyi at 25°C was more effective after first-instar larval contamination than after egg mass treatment. The duration of the egg stage could explain differences of susceptibility between the two noctuids at 25°C. Scanning electron microscopical observations suggested two ways of contamination of newly hatched larvae. First, fungal germinations on the chorion surface suggested that newly hatched larvae might be infected by penetration of the egg integument before hatching. Second, conidia on the egg cuticle could be an entomopathogenic inoculum for newly emerging larvae which fed upon chorions. Results showed that pathogenicity of Hyphomycetes to noctuid eggs might be a promising area of investigation for biological control.  相似文献

Observations on the morphology and infectivity for cattle of Babesia bovis parasites in unfed Boophilus microplus larvae after incubation at various temperatures. International Journal for Parasitology9: 115–120. The temperature of incubation of unfed Boophilus microplus larvae infected with Babesia bovis influenced the morphology and infectivity of the Babesia within the tick. Incubation at 37°C for 1–3 days stimulated the development of parasites morphologically similar to those usually observed in fed larvae harvested from cattle; similar forms appeared more slowly in larvae incubated at 31°C or 25°C. Extracts prepared from larvae after incubation at 37°C for 3–5 days or 30°C for 8 days were consistently infective for cattle. Prior storage of larvae at 14°C for up to 28 days enhanced the development of infectivity at 37°C; infectivity could still be produced after 65 days storage at 14°C but not after 76 days. Larvae released on a host transmitted B. bovis sooner if they had been incubated at 37°C for 4 days. It was concluded that the development of B. bovis to an infective stage in B. microplus is temperature dependent and does not require the stimulus of feeding by the host.  相似文献

The entomogenous fungi Beauveria bassiana (nine isolates), Metarhizium anisopliae (seven isolates), and Paecilomyces farinosus (four isolates) were tested as pathogens of larvae of the elm bark beetle, Scolytus scolytus. Single isolates of B. bassiana and M. anisopliae were also tested against adult beetles. Of the 21 isolates tested as conidial suspensions against larvae, all proved pathogenic. The three most and least virulent isolates were, respectively, isolates of B. bassiana and M. anisopliae. The other isolates fell between these two extremes, with the four P. farinosus isolates all moderately virulent. Spore retention on larvae following inoculation was estimated by washing conidia off the larvae. From the results it was possible to relate larval mortality to the approximate spore dose causing infection at different spore concentrations. Thus, application of spores of the three pathogens at a concentration of 103 spores/ml resulted in limited mortality. At this concentration, an average of only a single spore was recovered from the inoculated larva. Adult bark beetles also proved susceptible to infection by isolates of B. bassiana and M. anisopliae. They were exposed to discs of elm bark dipped in a conidial suspension. It was estimated that a dose of less than 100 spores could cause infection of beetles following feeding on the elm bark discs.  相似文献

Infectivity of Echinostoma liei miracidia to NIH albino Biomphalaria glabrata declines significantly from 62% with eggs incubated for 10–24 days to 3% for eggs incubated for 30–42 days. In mass exposures of 25 snails to 125 miracidia in 1 liter of water infectivity was high (54–66 %) and not affected by the presence of lettuce, plastic sheets, chalk, detritus or snail-conditioned water. In distilled water or snail-conditioned water the proportion of infected snails exposed singly to five miracidia per snail in 5 ml was not significantly different from the results of mass exposures of 25 snails in 1 liter to the same snail: miracidia ratio. Some evidence is presented suggesting that infected snails are less likely to suffer mortality than uninfected snails during the first 7–10 days post-exposure.The results suggest that Echinostoma liei miracidial searching efficiency is robust in volumes of at least 1 liter and in a heterogeneous habitat. These aspects enhance the competitive potential of echinostomes as possible biological control agents for Schistosoma mansoni.  相似文献

In the Biomphalaria glabrata-Schistosoma mansoni association, variations in cercarial production, in cercarial infectivity, and in the growth of infected snails are rhythmic. These chronobiological aspects are correlated with the dynamics of the intramolluscan larval stages of the parasite during the course of the infection. Rhythmic variations in the growth kinetics of infected snails are interpreted in terms of host-parasite metabolic exchanges.  相似文献

In field plots in which populations of predaceous species were suppressed using carbaryl, populations of the twospotted spider mite, Tetranychus urticae Koch, on field corn declined in association with aerial dispersal of the mites or with epizootics of the entomogenous fungus, Neozygites floridana Weiser & Muma. The relative importance of these two factors varied from year to year depending on environmental conditions. When moist weather conditions induced epizootics of N. floridana before corn plants became heavily infested with mites, the pathogen appeared to be the major factor causing mite population declines. When dry weather conditions allowed mite populations to expand unchecked until corn plants became entirely infested, mite aerial dispersal was the major factor associated with mite population declines. In every field in which there was an epizootic of N. floridana, routine applications of the fungicide maneb delayed epizootics and reduced or delayed the impact of the pathogen on mite populations. The total number of Neozygites-infected mites produced in maneb-treated plots throughout the test period was significantly greater than that in untreated plots when epizootics were initiated during periods of mite population growth. Fewer total infected mites were produced in maneb-treated plots compared to untreated plots when epizootics were initiated during periods of mite population decline.
Résumé Dans des parcelles de maïs dont les populations d'espèces prédatrices avaient été éliminées par du carbaryl, des populations de T. urticae ont régressé en fonction de la dispersion aérienne des acariens ou d'épizooties provoquées par N. floridana. L'importance relative de ces deux causes varie d'année suivant les conditions écologiques. Le pathogène paraissait la principale cause de déclin des populations quand le temps humide a induit les épizooties par N. floridana avant que le maïs n'ait été largement contaminé par les acariens. Quand le temps sec a permis aux populations d'acariens de se développer sans frein jusqu'à contamination totale du maïs, la dispersion aérienne des acariens était la principale cause de déclin des populations d'acariens. Dans tous les champs où il y avait eu épizootie de N. floridana, des traitements de routine avec le fongicide meneb ont retardé les épizooties et réduit ou retardé l'impact du pathogène sur les populations d'acariens. Pendant toute la période d'étude, le nombre total d'acariens infestés par Neozygites était significativement plus élevé dans les parcelles traitées au maneb que dans les parcelles non traitées, lorsque les épizooties étaient provoquées pendant la période de croissance des populations d'acariens. Quand les épizooties ont été déclenchées pendant la période de déclin des populations d'acariens, un nombre plus faible d'acariens infestés était observé dans les parcelles traitées au maneb que dans les parcelles sans traitement.

The effects of the density of Hymenolepis diminuta and the effects of thermal acclimation on the water balance of Tenebrio molitor were examined. Also, the subsequent infectivity of the cysticercoids for rats were investigated. T. molitor beetles were fed known numbers of H. diminuta eggs and then were kept at 15° or 25°C for 14 days. After 14 days, beetles were desiccated and water loss was determined. Parasite density did not significantly affect transpiratory water loss in T. molitor kept at 15° or 25°C following 24 or 48 hr of desiccation. However, after 72 hr of desiccation, beetles maintained at 15°C evidently could not regulate water efficiently since there was a significant increase in the transpiratory water loss as parasite density increased. Beetles acclimated at 15°C produced fewer cysticercoids than did beetles maintained at 25°C. Also, fewer adult worms were recovered from rats intubated with cysticercoids from heavily infected, 15°C-acclimated beetles. Apparently, heavily infected beetles acclimated to 15°C do not produce viable cysticercoids.  相似文献

Levan是一类果聚糖,由大量的果糖单元以β-(2,6)果糖苷键连接构成聚糖主链并含有少量β-(2,1)果糖苷键连接的支链组成。部分微生物来源的Levan具有抗肿瘤、抗病毒、降血糖、降血脂、免疫增强等重要的生物活性,在医药和功能性食品方面具有巨大的应用潜能。微生物发酵液提取和酶法合成是目前大量获得Levan果聚糖的两种方法,其中微生物发酵液提取的Levan果聚糖产量和蔗糖转化率一般较低,且发酵液中同时存在的其他高聚物不利于Levan的规模化纯化;而利用Levan蔗糖酶以蔗糖为底物转果糖基合成的Levan果聚糖产量已经高达200g/L、蔗糖转化率高达50%,并且Levan蔗糖酶合成Levan过程中酶的活性受到pH值、温度、螯合剂、金属离子等多种因素的影响,可以通过控制反应条件促进多糖合成反应的进行。因此,酶法合成将是工业化获得Levan果聚糖的主要方式。  相似文献

Three cell lines of Heliothis zea and one cell line of Heliothis virescens replicated the singleembedded, nuclear polyhedrosis virus (NPV) of H. Zea, (i.e., Baculovirus heliothis) with concomitant production of polyhedral inclusion bodies (PIB). Between 20 and 60% of the H. zea cells produced PIB, whereas only 3% of H. virescens cells were found to produce PIB. The H. zea cell lines produced 10 to 20 times more PIB than did the H. virescens cell line. The PIB from all cell lines produced typical symptoms of an NPV infection when bioassayed against larvae of H. zea. More than 99% of the total viral activity of the final whole culture was due to the PIB.  相似文献

In field populations of the European corn borer, Ostrinia nubilalis, there is a significant inverse correlation between the prevalence of corn borer infection with the microsporidian Nosema pyrausta and the prevalence of parasitism by the introduced braconid Macrocentrus grandii where infections with N. pyrausta exceed 45%. This relationship occurs geographically and from year to year. Corn borer infection with N. pyrausta, the source of infection for M. grandii, is significantly related to corn borer density in the cornfield. These findings strongly suggest that N. pyrausta has a significant adverse affect on field populations of M. grandii and may help explain the diminishing role of this and other introduced parasites as natural controls of the corn borer in the United States.  相似文献

Interactions between Vairimorpha necatrix and three other pathogens of Heliothis zea were evaluated with dose-mortality studies in order to find a synergistic combination which could be tested for field control of H. zea. The effect of the microsporidium combined with the bacterium Bacillus thuringiensis was at least additive, with indications of synergism. The interaction between V. necatrix and Heliothis nuclear-polyhedrosis virus was antagonistic except that the highest microsporidian dose overcame the antagonism with a resultant independent action. The interaction between V. necatrix and the fungus Nomuraea rileyi was additive, though response varied some-what with different proportions of the two pathogens. Even though none of these interactions is likely to be valuable in microbial control, V. necatrix has the potential to synergize or antagonize any biological or chemical agent that acts on the midgut epithelium of host insects.  相似文献

Aspergillus niger and Aspergillus oryzae were co-cultivated with each other and with Magnaporthe grisea or Phanerochaete chrysosporium, respectively. Enzyme assays for plant polysaccharide and lignin-degrading enzymes showed that co-cultivation can improve extracellular enzyme production. Highest ??-glucosidase, ??-cellobiohydrolase, ??-galactosidase, and laccase activities were found for A. oryzae in combination with other fungi, in particular with P. chrysosporium. Highest ??-xylosidase activity was obtained when A. niger was co-cultivated with P. chrysosporium. SDS-PAGE protein profiles demonstrated that A. niger and A. oryzae contributed most to the overall enzyme activities found in the culture medium of the mixed cultivations. These data demonstrate that co-cultivation of two major industrial fungi, A. niger and A. oryzae, results in improved production of biotechnologically relevant enzymes.  相似文献

Batches of Bulinus (Physopsis) globosus and Biomphalaria pfeifferi were exposed to Schistosoma haematobium and S. mansoni respectively each month for a 12-month period. The snails were kept out of doors in Salisbury (highveld) and indoors at Chiredzi (lowveld S. haematobium only) and examined weekly to determine the duration of prepatency, and the number of cercariae produced per infected snail. There was a strong seasonal influence in the highveld experiments which showed sporocyst dormancy in winter and the almost simultaneous maturation of infection in nine batches during early summer. In the lowveld the release of cercariae by infected B. globosus continued throughout the year although numbers fell off in winter. The prepatent period was prolonged in winter, but there was no evidence of sporocyst dormancy.  相似文献

Morphometry, catalytic (redox potential) activity, and body surface potential of two infective-stage juvenile populations of Neoaplectana carpocapsae, obtained from Sitophilus granarius adults and Galleria mellonella larvae, were compared. The relationship among these three morphological and physiological parameters and the degree of pathogenicity (=infectivity) and incidence of the nematode infection (=extensity) in Achroia grisella caterpillars, Trogoderma granarium larvae, and Tribolium confusum adults was discussed.  相似文献

红火蚁Solenopsis invicta Buren是一种国际重大外来入侵生物,严重危害农林生产、生命健康、公共安全和生态环境。本研究在室内条件下测定了小卷蛾斯氏线虫Steinernema carpocapsae All品系、长尾斯氏线虫S. longicaudum X-7品系和芫菁夜蛾斯氏线虫S. feltiae SN品系对红火蚁的致病力,并筛选出高致病力品系及其对红火蚁的田间防控效果。结果表明,不同昆虫病原线虫对红火蚁不同虫态的致病力存在显著差异。3种线虫对红火蚁幼虫具有较高的致病力,处理3 d后,幼虫的累计校正致死率均为100%。3种线虫对工蚁的致病力均较弱,处理10 d后的校正死亡率均小于30%。与芫菁夜蛾斯氏线虫SN品系相比,长尾斯氏线虫X-7品系和小卷蛾斯氏线虫All品系对蛹和生殖蚁具有较强的致病能力,10 d后累计校正死亡率均达到90%以上。田间试验表明,施用小卷蛾斯氏线虫All品系对红火蚁工蚁的防控效果最高可达63.7%,且不会增加红火蚁的迁巢风险。综上所述,小卷蛾斯氏线虫All品系和长尾斯氏线虫X-7品系为红火蚁的高致病品系,田间施用小卷蛾斯氏线虫All品系对红火蚁工蚁具有显著的控制作用,可推荐用于红火蚁的生物防治。  相似文献

Mortierella ramanniana var. angulispora accumulates triacylglycerol (TG) in lipid bodies. Studies on lipid transport into lipid bodies are essential for elucidating mechanisms of lipid body formation. We used fluorescent dyes and fluorescent lipid analogs to visualize lipid body formation with a confocal laser scanning microscope. Different sizes of lipid bodies were stained by Nile red, a lipid body marker – one with a diameter of about 1 μm and the other with a diameter of about 2–3 μm. Lipid bodies matured into larger ones with culture. To metabolically monitor lipid bodies, we used 1-palmitoyl, 2-[5-(5,7-dimethyl boron dipyrromethene difluoride)-1-pentanoyl]-phosphatidic acid (C5-DMB-PA), and C5-DMB-phosphatidylcholine (C5-DMB-PC). These were taken up into fungal cells and incorporated into intracellular organelles at 30°C. C5-DMB-PA was quickly incorporated into lipid bodies while C5-DMB-PC was initially incorporated into internal membranes, presumably endoplasmic reticulum membranes, and fluorescence was then gradually transported into lipid bodies. The transport of fluorescent lipids accompanied their metabolism into diacylglycerol (DG) and TG, which, taken together with the fluorescence distribution, suggested that conversion to TG was not necessary for transport into lipid bodies. It is likely that the synthesized DG was mainly located in lipid bodies and the conversion to TG took place in lipid bodies. C5-DMB-PA and C5-DMB-PC were converted to DG and TG in the membrane and lipid body fractions of this fungus, which agreed with in vivo metabolism of these fluorescent lipids and in vitro enzyme activity related to PA and PC metabolism. These results indicate that transport and metabolism of C5-DMB-PA and C5-DMB-PC represent two different routes for lipid body formation in this fungus.  相似文献

Grape anthracnose, which is caused by Elsinoë ampelina, is a disease that negatively affects grape production. This study aimed to investigate the effects of aeration, temperature, light, and preculture period on the formation of E. ampelina conidia and conidial germination and virulence. The colony morphology on potato dextrose agar (PDA) plates was more diverse than that in PDA bottles. The assessment of different culture methods, temperatures, light conditions, and preculture periods revealed that optimal conidial production occurred on 25‐day‐old colonies grown in PDA bottles at 21°C for 24 hr in the dark. The cultures in PDA bottles consistently produced approximately 5.0 × 106 conidia under these conditions. No conidial formation occurred when the cultures were kept at 25°C in the dark. The highest germination rate of E. ampelina was 80% at 25°C after 24 hr, whereas no germination was observed at 17°C after 12 hr. Pathogenicity tests revealed that symptoms of the disease were observed 4 days postinoculation (dpi) on leaves of Vitis vinifera cv. Red Globe. New conidia were observed on the lesions at 8 dpi. This study provides an effective method for the conidial production of E. ampelina that may also be applicable for other Elsinoë fungal species.  相似文献

以紫潮泥和红黄泥两种不同质地的水稻土壤作为研究对象,通过室内培养试验,分析施用硝态氮肥对N2O释放和反硝化基因(narG/nosZ)丰度的影响,并探讨反硝化基因丰度与N2O释放之间的关系。结果表明,施用硝态氮显著增加两种水稻土的N2O释放量。在72h培养过程中,施氮改变了紫潮泥反硝化基因(narG/nosZ)的丰度,但并未明显影响红黄泥反硝化基因(narG/nosZ)丰度。通过双变量相关分析发现,除了紫潮泥narG基因外,其它的反硝化基因丰度和N2O释放之间并没有显著相关性。  相似文献
