Continuous cell cultures were established from imaginal wing discs of 2 Lepidoptera, Spodoptera frugiperda and Plodia interpunctella. The S. frugiperda line (IAL-SFD1) grows as multicellular vesicles and responds morphologically and biochemically to the insect hormone, 20-hydroxyecdysone. In contrast, the P. interpunctella cells (IAL-PID2) grow as attached monolayers of small spindle-shaped cells and do not appear to have specific responses to 20-hydroxyecdysone, although growth rates are slowed in these cells upon exposure to the hormone.  相似文献

Fragments of imaginal discs of the fruitfly Drosophila undergo growth and pattern regulation when cultured in vivo in adult female hosts for several days prior to metamorphosis in host larvae. Pattern regulation results in either regeneration of excised pattern elements or duplication of elements whose fate map positions lay within the fragment. Initial wound healing along the cut edge of a fragment is thought to be a crucial first step in the process of pattern regulation. We have examined the capacity for wound healing and pattern regulation of fragments (distal halves) of the wing disc cultured in vitro, using the culture system recently reported to support extensive growth and transdetermination of slightly wounded whole imaginal discs in vitro. Our results suggest that disc fragments and whole discs apparently respond differently in the culture system. With disc fragments, wound healing did not occur in vitro. When fragments were first cultured overnight in adult female hosts to allow initial wound healing prior to explantation in vitro, then some volume increase and regeneration of excised portions occurred during 2–3 weeks of culture in vitro. The extent of apparent growth was much less than that reported for whole discs, and the frequency of regeneration in vitro (19%), while highly significant relative to controls not cultured in vitro (0%), was much less than that observed for fragments cultured in vivo (84%). Furthermore the extent of regeneration which occurred in vitro was considerably smaller than that which occurs during regeneration in vivo.  相似文献

In epidermal cells of embryos of Blabera cultured in a medium without exogenous moulting hormone, 2 peaks in RNA synthesis occur respectively on days 7 and 11, followed by two peaks in cuticular products synthesis: a peak in protein synthesis on day 16 and a peak in chitin synthesis on day 17. Addition of exogenous moulting hormone to the culture medium hastens the two peaks in RNA synthesis and brings them closer together on days 1 and 4. The peaks in protein and chitin synthesis are similarly brought forward, and both appear on day 5.Localisation of the synthesised cuticular products is a function of the stage of differentiation reached in culture by the epidermal cells. Initially, there are little of the synthesised products and they are distributed uniformly throughout the cells. Progressively, the rate of synthesis increases and the products appear concentrated at the tips of the cells and then pass into the developing cuticle. At the beginning, the whole thickness of the deposited cuticle is labelled, then only the basal regions. Finally, the intensity of the label decreases.The results of incorporation rates of uridine, proline and glucosamine, the observation of the localisation of the label, and the secretion of the cuticle show that RNA synthesis occur before protein and chitin syntheses which are tied to the secretion of the procuticle.The hormone acts directly on the epidermal cells by triggering, in a general manner, the events which usually occur under the endogenous moulting hormone control.  相似文献

The effects of alpha-ecdysone (α-E), beta-ecdysone (β-E), and larval fat body on morphogenesis and total RNA synthesis in wing imaginal disks of Galleria mellonella were studied. Both ecdysones induce morphogenesis of disks in vitro. Alpha-ecdysone and β-E (0·3–3·0 μg/ml) stimulate RNA synthesis 30 and 60 per cent above control levels, respectively. While less α-E (0·3 μg/ml) is required to increase RNA synthesis than to induce morphogenesis, the reverse is true for β-E. Morphogenesis (i.e. tracheole migration and evagination) can proceed in the presence of concentrations of β-E (0·03 μg/ml) that are subthreshold for the induction of RNA synthesis (0·3 μg/ml). We conclude, therefore, that the increase in total RNA (presumbly ribosomal) is unrelated to and not a prerequisite for tracheole migration or evagination. If morphogenically active preconditioned medium (i.e. medium in which α-E and fat body have been incubated for 48 hr and the fat body then removed) is added to disk cultures, RNA synthesis is not stimulated. Apparently, increases in total RNA caused by both ecdysones may not be necessary for early in vitro disk development. The independent nature of some ecdysone-induced events and implications of our conclusions are discussed.  相似文献

Apolipoprotein E (apoE), a ligand for the low-density lipoprotein receptor family, has been implicated in modulating glial inflammatory responses and the risk of neurodegeneration associated with Alzheimer’s disease. Glial cells activated by lipopolysaccharide (LPS) have decreased apoE levels and we recently showed that apoE itself can modulate the inflammatory response by reducing c-Jun N-terminal kinase (JNK) activation. Reduced JNK phosphorylation is vital to overcome the LPS-induced decrease in apoE expression, suggesting that JNK inhibition may be an effective way to increase apoE protein and protract its anti-inflammatory properties. This study investigates the impact of JNK inhibition on apoE production using two JNK inhibitors. Our work in primary glia and in vivo in mice injected with JNK inhibitor demonstrates that inhibition of JNK may be an effective way to increase apoE expression.  相似文献

A technique is described which allows defects to be made in situ in the imaginal discs of immature Drosophila larvae. Bisection of the second leg disc across the upper-lower axis results in regeneration of the remainder of the disc from the upper portion, and mirror-image duplication of the anlagen in the lower half (Figs. 2–5, Table 1). The upper half, retaining its connection to the larval epidermis, is able to evert; the lower half metamorphoses as an uneverted implant. Partial bisection of the disc often results in the production of branched legs in which one branch is complete but the other is a double half (Figs. 7–9). These cases can be interpreted as resulting from regeneration from one cut surface and duplication from the other. Pattern triplications have been obtained by partial bisections of the wing disc (Fig. 10), and these can be interpreted in a similar manner. It is suggested that regeneration and duplication are identical phenomena, resulting from the properties of of the anlagen at the cut edge. Cases of regeneration and duplication in other insect and vertebrate systems are discussed, and interpreted on the basis of gradients of developmental capacity (Fig. 11).  相似文献

Membrane inactivation by freezing has been investigated using intact spinach leaves and isolated thylakoid membranes from chloroplasts of leaf cells as test material. During freezing in vitro in solutions containing neutral solute and a slight excess of inorganic salts such as NaCl, electron transport is stimulated while photophosphorylation is lost. Under more drastic freezing conditions damage increases, affecting dichlorophenolindophenol reduction, the rise in variable fluorescence, ferricyanide reduction and electron transport through Photosystem I, in that order. Semipolar compounds such as phenylalanine or phenylpyruvate exhibit a much higher membrane toxicity during freezing than inorganic salts. The profile of damage caused by this class of compounds is different from that caused by salts. Damage to membranes isolated rapidly from frost-killed leaves is similar to that produced by semipolar compounds during freezing in vitro. A few sites of damage could be identified, among them the site responsible for oxidation of water during photosynthesis. The results support the view that the sensitivity of their membranes limits the ability of cells to withstand freezing and suggest that freezing sensitivity is due to the accumulation in the cells of potentially membrane-toxic organic and norganic cell constituents.  相似文献

The incorporation of [14C]mevalonate and [14C]isopentenyl diphosphate into geranylgeranyl diphosphate was investigated in in vitro systems from Cucurbita pepo (pumpkin) endosperm and from Avena sativa etioplasts. Mevalonate incorporation was effectively inhibited in the pumpkin system by geranylgeranyl diphosphate and geranylgeranyl monophosphate but less effectively by phytyl diphosphate or inorganic diphosphate. Membrane lipids, geranyllinalool, or lecithin enhanced mevalonate incorporation in the Cucurbita system. Incorporation of isopentenyl diphosphate was also enhanced by lecithin and inhibited by geranylgeranyl diphosphate in the Cucurbita system. No lipid enhancement was found in the Avena system; inhibition by GGPP required a much higher GGPP concentration than in the Cucurbita system.  相似文献

Twenty-five adult female Plodia interpunctella infected with Nosema plodiae laid 856 eggs in laboratory tests; 12.5% of the eggs were infected transovarially. The highest level of transmission by an individual female was 14 infected eggs of 27 laid (51.8%); the lowest level of transmission observed was 1 of 43 eggs (2.3%). All stages of N. plodiae were not transmitted with equal frequency; moreover, most eggs harbored predominently only the trophozoite stage of the pathogen. Approximately 80% of the infected eggs contained trophozoites almost exclusively; about 13.1% contained about an equal number of spores and trophozoites, and about 6.5% contained mainly spores. Histological observations indicated that infections may be initiated in nurse cells and subsequently transferred to associated oocytes.  相似文献

Cultivation in vitro of Schistosomatium douthitti (Trematoda : Schistosomatidae). International Journal for Parasitology12: 541–545. We grew S. douthitti in vitro from the cercaria to pairing ovigerous adults, using methods and media previously reported for cultivation of Schistosoma mansoni. Growth of S. douthitti in vitro was much more rapid than that of S. mansoni, but the cultures achieved a similar stage of maximal development. Cultured S. douthitti worms were smaller than those from animals and eggs were inviable. We determined that females of this species, known to produce eggs in unisexual animal infections, will also do so in culture in the absence of males.  相似文献

NaF mimicked the activation by thyrotropin of iodide binding to proteins and of glucose C-I oxidation but not the accumulation of intracellular colloid droplets or the stimulation of secretion in dog thyroid slices in vitro. On the contrary, NaF inhibited the two latter thyrotropin effects. The inhibitory action of F was partially relieved by the addition of glucose to the medium; it was mimicked by sodium oxamate. These data suggest that NaF depresses the endocytosis of colloid and thyroid secretion by inhibiting aerobic glycolysis in the follicular cell. NaF inhibited the activation of colloid droplet accumulation and secretion by N6,O2′-dibutyryl-adenosine 3′,5′-monophosphate (dibutyryl cyclic AMP) and the accumulation of cyclic AMP in thyrotropin-stimulated slices. This suggests an inhibition at the level of both cyclic AMP accumulation and cyclic AMP action. The inhibition by NaF and sodium oxamate of colloid droplet formation and thyroid secretion but not of glucose C-I oxidation in stimulated slices further confirms our conclusion that the latter effect is not merely a consequence of the activation by thyrotropin of colloid endocytosis.  相似文献

The effect of polyamines on the polyadenylation reaction in vitro was investigated. Varying concentrations of spermine were added to the reaction catalyzed by purified poly(A) polymerase using rat liver nuclear RNA, poly(A), Escherichia coli tRNA or (Ap)3A as exogenous primers. The enzyme activity decreased progressively with increasing concentrations of polyamines; complete inhibition was obtained at 0.4 and 1.2 mm spermine for the nuclear RNA- and poly(A)-primed reactions, respectively. No inhibition was observed for the (Ap)3A-primed reaction. Spermidine and putrescine also inhibited polyadenylation but to a lesser extent than spermine. The degree of inhibition by spermine was related to the polynucleotide primer concentrations. Spermine prevented polyadenylation by binding to the primer but not to the poly(A) polymerase molecule as shown by the migration of [14C]spermine through glycerol gradients after preincubation with enzyme or tRNA. At concentrations inhibitory to polyadenylation in vitro, spermine could stimulate the DNA-dependent RNA synthesis catalyzed by RNA polymerase II. The present study suggests that low levels of polyamines could be used as specific inhibitors of the poly(A) synthesis in vitro.  相似文献

Fatty acid hydroperoxide lyase (HPO lyase) was found in green and non-green tobacco cells cultured in vitro. The HPO lyase activity in non-green cells was 13-12 of that in green cells. When the cells were transferred from the light to dark conditions or vice versa, cells turned non-green or green according to the light conditions. The HPO lyase activity also changed according to the light conditions, but the changes in HPO lyase activities were not proportional to the changes in chlorophyll contents. These results suggest that at least two types of HPO lyases are present in the green cells. One type of HPO lyase is perhaps common both to the green and non-green cells; another one is chloroplastic. The fatty acid compositions of cells and substrate specificities of HPO lyase differed between green and non-green cells.  相似文献

Microbial biofilms have become increasingly problematic in the food processing and medical industries where they cause food and surface contamination. Biofilms have also been implicated as the cause of serious infections in humans as their occurrence makes it difficult to treat common infections and the likelihood of recurrent infections is high. Due to emerging resistance, conventional control methods are fast becoming ineffective. In this study, the use of a selection of commercial plant extracts is investigated. The inhibitory effects of eight herbal extracts on the development of microbial biofilms was investigated against clinical and reference strains of Pseudomonas aeruginosa and Candida albicans. The antimicrobial activity was investigated on the planktonic forms using the minimum inhibitory concentration assay. The extracts that showed the highest antimicrobial activity against the two test organisms were Echinacea angustifolia (cone flower), Mentha piperita (peppermint) and Rosmarinus officinalis (rosemary) with minimum inhibitory concentration values between 0.38 and 2.5 mg/ml. The crystal violet assay was used to assess the effect of pre-treating a surface with plant extracts on cell attachment and the extent of biofilm development following exposure to extracts (biofilm biomass). Most of the extracts reduced microbial colonization by at least 50%. In contrast, preformed biofilms were less responsive to the majority of extracts, thus growth inhibition was more difficult to achieve. Mentha piperita was the only extract that showed some antibiofilm activity against both pathogens.  相似文献

White oaks (Quercus section, Quercus subgenus) are widely distributed in Europe. Quercus petraea (sessile oak), an economically important species is predicted to be affected by climate change. Q. pubescens (pubescent oak) and Q. virgiliana (Italian pubescent oak) are economically less important, drought tolerant species. Frequent hybridization of white oaks was observed and currently the introgression of Q. pubescens and Q. virgiliana in non-mediterranean regions of Europe has been reported. Our goal was to use tissue cultures established from individual trees of the above taxa and their putative hybrids, all present in the forest stand of Síkf?kút LTER Research Area (NE Hungary) as simple experimental model systems for studying drought/osmotic stress tolerance. Tissue cultures are more suitable models for such studies, than seedlings, because they are genetically identical to the parent plants. Polyethylene glycol (PEG6000) treatments were used for this purpose. The identification of taxa was based on leaf morphological traits and microsatellite analysis and showed that Q. petraea is genetically distinct to all other taxa examined. We established six callus lines of Quercus. As expected, in Q. petraea cultures PEG6000 induced severe loss of fresh weight and the ability to recover after removal of the osmoticum, which was not characteristic for Q. pubescens and Q. virgiliana. Putative hybrids exhibited an intermediate response to osmotic stress. Activity gels showed the increase of single-strand preferring (SSP) nuclease and no significant change of guaiacol-peroxidase activities in drought-sensitive genotypes/cultures and no significant increase of SSP nuclease activities accompanied with increases of guaiacol-peroxidase activities in drought-tolerant ones. This indicates that drought/osmotic stress tolerance is associated to increased capacity of scavenging reactive oxygen species and hence less susceptibility to DNA damage. Our results confirm that tissue cultures of oak are suitable model systems for studying drought/osmotic stress responses.  相似文献

Hydroxycinnamate: CoA ligase was extracted from stems of in vitro willow cultures and characterized. One peak of activity was obtained after column chromatography on Sephadex G 100 or DEAE Sephacel. p-Coumaric acid gave the highest Vmax among the cinnamates examined. The Kmvalues for p-coumaric, caffeic and ferulic acid were 31.0, 4.7 and 46 μM, respectively. The MW of the CoA ligase was 57 000 and the pH optimum was 7.0. The characteristics of the enzyme correspond to its physiological role in lignin biosynthesis.  相似文献

Summary Wing imaginal discs isolated from last instar larvae of the Indian meal moth,Plodia interpunctella, produced chitin when incubated in vitro with 2×10–7 M 20-hydroxyecdysone. Chitin biosynthesis was initiated 8 h after the conclusion of a 24-h treatment with hormone. Simulataneous incubation of wing discs with 20-hydroxyecdysone and either inhibitors of RNA synthesis (alpha-amanitin, actinomycin-D) or inhibitors of protein systhesis (cycloheximide, puromycin) prevented chitin biosynthesis. We conclude from our results that RNA and protein synthesis must continue undiminished during the hormone-contact period, and that synthesis of protein, but not of new RNA is required during the posthormone culture period. Our findings are consistent with the hypothesis that ecdysteroids stimulate insect metamorphosis by promoting the synthesis of new RNA and protein during a hormone-dependent phase followed by hormone-independent protein synthesis.  相似文献

Explants of the central nervous system of Drosophila have been shown to produce nerve fibres in vitro. The effects of various culture conditions on fibre outgrowth have been examined. Nervous tissue could form nerve fibres in vitro when the explants were obtained from mid-embryonic or early- and mid-pupal stages, but not when they were obtained from larvae or late-pupae. The effect of the temperature-sensitive mutation shibirets has been investigated by placing mutant explants into culture at permissive (17°C) or restrictive (28°C) temperatures. No differences in the extent of fibre outgrowth between wild-type and shibirets were observed, regardless of the temperature of cultivation.  相似文献
