La3+ and Ce3+, either singly or a mixture, promoted crocin production of Crocus sativus callus but Nd3+ had little effect and all metal ions were toxic above 100 microM. La3+ (60 microM) promoted growth of callus significantly but increased crocin only slightly. Ce3+ (40 microM) significantly promoted crocin production but had little effect on cell growth. La3+ (60 microM) and Ce3+ (20 microM) together gave the highest dry weight biomass (20.4 g l(-1)), crocin content (4.4 mg g(-1)) and crocin production (90 mg l(-1)) which were, respectively, 1.7-fold, 4.2-fold and 7.1-fold of those without additions. Nd3+ (40 microM) only slightly promoted cell growth and crocin production.  相似文献

Excised young intact stigmas plus ovaries of Crocus sativus L. were cultured on Linsmaier-Skoog media supplemented with either a cytokinin or an auxin alone or in combinations. Benzyladenine and kinetin at concentrations of 0.1, 1, and 5 mgl-1 supported growth, and crocin was biosynthesized in the stigmas in vitro. Auxins had little effect. Young excised single stigmas or half ovaries were also cultured so as to form stigma-like structures in order to explore a possible new approach to industrial production of the spice, saffron. On Linsmaier-Skoog and Nitsch media supplemented with kinetin at concentration of 1 or 5 mgl-1 and alpha-naphthalene acetic acid or indole-butyric acid at concentration of 0.1 or 10 mgl-1 in combinations, stigma-like structures appeared directly and indirectly (through meristematic tissue), grew and matured. The maximum number of structures were 75 per half ovary. Stigma-like structures were produced in tissue cultures (TC stigmas) from the ovary explants of C. sativus on MS medium supplemented with NAA and BA. The size of these structures was 2 to 3 cm in length. At higher concentrations of both NAA (54 µM) and BA (44 µM) white tubular abnormal structures were observed from ovary explants in addition to the TC stigmas. Crocin and picrocrocin, responsible for colour and bitter taste respectively, were found to be 6 and 11 times lower in TC stigmas than in the natural stigmas. The saffron obtained from tissue cultures was subjected to sensory analysis and compared with the data obtained from chemical analysis. The sensory data indicated that the saffron pigments produced in tissue cultures were one tenth that of natural stigmas. Sensory profile test showed that the tissue culture saffron was low in floral, spicy and fatty characteristics as compared to saffron obtained from flowers. The aluminum (Al) tolerance of saffron (Crocus sativus L.) in hydroponics and the method of improving Al tolerance were investigated. Compared with the Al-free control, saffron root elongation was decreased by 59.3 and 75% at 0.05 and 0.2 mM Al stress, respectively. At 0.5 mM Al stress, the root elongation was inhibited completely. Addition of 1 mM polyamines improved saffron root growth markedly at 0.2 mM Al stress. Putrescine (Put) showed better amelioration effect than spermidine (Spd) and spermine (Spm). The root elongation in Put treatment was only 15% lower than that of Al-free control. The alleviation of Al rhizotoxicity by polyamines might be attributed to lower Al content in the root tips, and subsequent less lipid peroxidation and oxidative stress. Higher activities of amine oxidases and Hydrogen peroxide (H2O2) content might decrease the effects of Spd and Spm on alleviating oxidative damage compared with that of Put.  相似文献

为提高藏红花(Crocus sativus)胚性愈伤组织的繁殖系数与出芽率, 以建立藏红花离体快繁体系, 解决其资源短缺问题, 采用两步法, 用稀土调控其胚性愈伤组织的生长与分化。结果表明, 在添加了0.25 mg·L–1 NAA、3 mg·L–1 6-BA和400 mg·L–1 CH的B5固体培养基中, 0.04 mmol·L–1 La3+促进胚性愈伤组织生长的效果最佳, 繁殖系数为12, 是不添加稀土处理组的1.48倍; 在添加了0.25 mg·L–1 NAA、3 mg·L–1 6-BA和400 mg·L–1 CH的1/2 B5固体培养基中, 0.06 mmol·L–1 Ce3+促进胚性愈伤组织分化出芽的效果最佳, 出芽率高达84.5%, 是不添加稀土处理组的1.81倍, 且高于国外报道的出芽率(40%)。初步解决了藏红花胚性愈伤组织生长慢和出芽率低等问题, 为建立高效稳定的藏红花离体快繁体系奠定了基础。  相似文献

Saffron corms have been demonstrated to contain a proteoglycan that inhibits growth of human tumor cells. In this study, we show strong evidence indicating that callus cultures of saffron corm also synthesize such glycoconjugate. This compound is cytotoxic against human cervical epithelioid carcinoma cells (IC50 = 7 mg ml-1), and consists of approximately 90% carbohydrate and 10% protein. In order to optimize the glycoconjugate purification to complete functional and therapeutic studies, we have designed an improved method consisting of anion-exchange chromatography and reversed-phase HPLC, which can be easily scaled-up.  相似文献

Saffron (Crocus sativus L.), and its main constituents, crocin, and crocetin have shown promising effects as an antileukemic agent in animal models and cell culture systems. Saffron retards the growth of cancer cells via inhibiting nucleic acid synthesis and enhancing antioxidative system. It can induce apoptosis and chemosensitivity via inhibiting multidrug resistance proteins. Saffron also induces differentiation pathways via inhibiting promyelocytic leukemia/retinoic acid receptor-α, histone deacetylase1, and tyrosyl DNA phosphodiesterase-1 as well. The present review highlights the most recent findings on the antileukemic effects of saffron and its underlying molecular targets. The emerging evidence suggests that saffron has a selective toxicity effect against leukemic cells while is safe for the normal cells.  相似文献

对藏红花(cfocus sativus L)愈伤组织的诱导条件进行了优化.结果表明:Ms是藏红花芽愈伤组织的最佳诱导培养基,而B5是叶子和花愈伤组织的最佳培养基.藏红花芽、叶和花愈伤组织的最佳诱导温度分别是18℃、25℃和21℃.光照是叶子愈伤组织诱导的有利因素,但不利于芽和花愈伤组织的诱导.1.5~2.0 mg·L-1NAA和0.25 mg·L-6-BA是愈伤组织诱导的最佳激素组合.通过目视法和HPLC方法,从229株细胞系中筛选出细胞系Corml,其藏红花素1的含量是1 677 mg·g-,生长较快,且不易褐化.为采用植物细胞工程法解决藏红花素1资源短缺问题打下了基础.  相似文献

In vitro development of parthenocarpic fruits of Crocus sativus L. was induced by culturing ovaries on MS agar medium supplemented with growth-regulators (2,4-D, GA3 and BAP). Amongh these, 2,4-D was the most effective in promoting fructification. The fructigenic activity was independent of both the stage at which the ovaries were excised (before, during or after anthesis) and pollination of the stigmas. Unlike the above compounds, abscisic acid inhibited fructification.  相似文献

Intergeneric Fragaria vesca x Potentilla fruticosa hybrids were produced using in vitro culture. Hybrid plants were not obtained by direct embryo rescue, but were regenerated from cotyledon-derived callus. Experiments with F. vesca indicated that using cotyledon halves was not more productive than using entire cotyledons. A polarity was observed in cotyledons and in cotyledon halves, with callus and regenerated shoots produced more frequently from proximal ends. Cotyledons from 17% of hybrid embryos produced callus and regenerated mature plants. The technique enabled rapid multiplication of some embryos, with the production of more than one hybrid plant. Totipotent calli of a Paphiopedilum hybrid (Paphiopedilum callosum ‘Oakhi’ × Paph. lawrenceanum ‘Tradition’) were induced from seed-derived protocorms on a 1/2 strength Murashige–Skoog medium plus 1–10 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.1–1 mg l−1 1-phenyl-3-(1.2.3-thiadiazol-5-yl)urea (TDZ). These calli grew well when subcultured on the same medium, but proliferated more on 1/2 MS medium plus 5 mg l−1 2,4-D and 1 mg l−1 TDZ. Calli developed further along a route of production of protocorm-like bodies and eventually formed plantlets that could be transplanted to pots and grew well. Sources of carbohydrate nutrition such as sucrose, glucose, and galactose, with the exception of arabinose, were shown to influence positively callus growth and polysaccharide (pectin silenan and acidic arabinogalactan) biosynthesis. Galactose was found to cause a stimulatory effect on yield and productivity of arabinogalactan. Low concentrations of sucrose failed to support the cell growth and polysaccharide biosynthesis. Increasing sucrose concentrations led to biomass accumulation but failed to enhance efficiency of the substrate utilization. The optimal medium for the campion cell culture growth was found to be one containing 30 g/liter of sucrose or a mixture of sucrose with glucose (in 15 g/liter). Increasing sucrose concentrations in the medium from 30 to 100 g/liter failed to significantly influence the polysaccharide yields while the polysaccharide productivity per liter of the medium grew due to promotion of culture productivity in biomass. Variations of the carbon sources in the nutrient media were shown to influence insignificantly the biochemical characteristics of arabinogalactan and silenan while an increase in the sucrose concentration to 50-100 g/liter led to a diminution of the galacturonic acid content in silenan and to changes in contents of the neutral monosaccharide residues in silenan and arabinogalactan.  相似文献

Summary Vicia faba callus line (VFS 1), isolated from expiants of immature embryo, grew satisfactorily onMurashige andSkoog complete medium with 1.38 M 2,4-D, or with 0.92 M 2,4-D to which 1.0 M kinetin was added. It also grew well on the B 5 modified medium containing 2.3 M 2,4-D and 25.0 M kinetin. On the last of these media the cultures grew more uniformly and without necrosis. They also showed diminishing variation in polyploidy in favour of diploids and corresponding aneuploids (hypodiploids).After being cultured for nearly three years on MS containing 1.38 M 2,4-D, 8–33% of cultures of VFS 1 were able to regenerate roots when transferred to either MS half strength with 5.37 M NAA, or to a medium without 2,4-D, or else to media with the addition of kinetin only (in various concentrations).  相似文献

Callus cultures from stem explants of six Lathyrus sativus L. cultivars were tested for their morphogenic capacity. Shoot-buds were formed in calli of only one cultivar. Maximum response was observed in the medium containing 10-8 M picloram and 10-6 M benzylaminopurin. Supplementation with adenine sulphate was required for shoot-bud formation. The greatest frequency of shoot-bud formation was detected at the second passage and complete lack of regeneration capacity was observed after the 8th passage. 采用水蒸气蒸馏法从藏红花花瓣和雄蕊中提取挥发油,用GC-MS技术结合计算机检索对其二者化学成分进行分离和鉴定,用色谱峰面积归一化法计算各组分的相对含量.花瓣中共鉴定出16种化合物,主要成分为正二十六烷(11.60%)、正十五烷(11.31%)、棕榈酸甲酯(10.82%)、油酸甲酯(10.35%)、2,4-二叔丁基苯酚(9.63%)、亚油酸甲酯(7.18%)、藏红花醛(5.66%);雄蕊中共鉴定出20种化合物,主要成分为油酸甲酯(30.83%)、亚油酸甲酯(24.12%)、环已醇(16.80%)、硬脂酸甲酯(12.88%)、棕榈酸甲酯(8.97%)、花生酸甲酯(1.18%)、苯并噻唑(1.01%).  相似文献

An efficient protocol has been developed for rapid mass propagation of Tylophora indica from leaf derived callus. Optimal callus was developed from leaf explants on Murashige and Skoog (MS) basal medium supplemented with 10 2,4,5-T. Adventitious shoots were regenerated (85%) from the surface of the callus on MS medium supplemented with 5 M Kinetin. Individual elongated shoots were rooted on half-strength MS medium containing 0.5 M IBA. Regenerated plantlets with well developed shoots and roots were successfully transferred to soil. The study demonstrated a dedifferentiated callogenic propagation route via adventitious shoot development in T. indica, which could be useful for large scale multiplication of this endangered medicinal plant.  相似文献

Protoplasts were isolated from callus tissue of Hibiscus syriacus L. using a solution of 3% Onozuka cellulase, 1% Onozuka macerozyme, and 0.5% hemicellulase. Highest yields of viable protoplasts were obtained from friable, white or yellow callus 8–9 days after subculture on Murashige & Skoog medium with 0.5 mg l-1 2,4-dichlorophenoxyacetic acid and 0.1 mg l-1 kinetin. Protoplasts cultured in thin liquid layers of this medium with mannitol continued dividing for longer than those cultured in droplets or in an agar medium. Cultures were maintained until protoplasts had divided to form groups of more than ten cells. Cell groups developed into callus and continued to grow on an agar medium, but failed to differentiate on a regeneration medium with 2 mg l-1 naphthalene acetic acid and 1 mg l-1 benzylaminopurine.  相似文献

A system was developed for protoplast isolation and culture from suspension cultured cells of winged bean,Psophocarpus tetragonolobus. Cells from a three-day-old suspension were incubated in an enzyme mixture containing 6% cullulysin, 1% Macerase, 1% desalted Rhozyme, 0.4M sorbitol, and 0.1M CaCl2 at pH 5.5. Average yields of protoplasts were 6.5 × 106 per gram fresh weight of cells. Protoplasts were cultured in modified B5 medium containing 68.4 g/l glucose, 250 mg/l xylose, 0.1 mg/l 2,4-D, 0.5 mg/l BAP, 250 mg/l N-Z amine type AS, and 20 ml/l coconut water. After 24 h of culture, the protoplasts had synthesized a new wall, and in three days had begun division. The optimum plating density was 1–2 × 103 protoplasts/ml. The division frequency ranged between 40%–60% for most experiments with a high of 72% in one experiment. After three weeks, cell colonies could be transferred to solid MS medium containing N-Z amine and coconut water where callus developed. This protoplast system is technically comparable to soybean for experiments concerned with genetic manipulation involving legumes.  相似文献

基于已经建立的藏红花细胞悬浮培养体系,研究了其悬浮培养动力学.结果表明,悬浮培养时细胞的生长周期约为20 d,在20 d时生物量达到最大,为12.3 g DW/L,但是藏红花素的合成周期大约为28 d,在28 d时藏红花素的含量和产量达到最大,分别为95.8 mg/g和0.92 g/L.藏红花素的积累与细胞的生长之间的关系为半生长偶联型,为其生物反应器放大培养提供了参数等理论依据.  相似文献
