Summary A vesicular-arbuscular mycorrhizal fungus, identified asGlomus mosseae Gerdemann and Trappe, was found to occur in groundnut and some other hosts. In groundnut roots under experimental conditions, the fungus showed three phases of development-a lag phase of 3–4 weeks by the end of which formation of vesicles was noticed, a phase of gradual development upto 12 weeks, by which time an average of 6 vesicles per centimeter of root developed and a constant phase where there was no further increase of the fungus. Pigeon-pea, black gram, green gram, angular gourd, onion, maize, sorghum and pearl millet also formed mycorrhizae with this fungus, but tomato and egg-plant did not. The lag phase was longer and the average number of vesicles developed per unit root length was less in the non-leguminous hosts.  相似文献

The potato tuber moth (PTM),Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), is a major pest of processing tomatoes,Lycopersicon esculentum Mill. (Solanaceae), in Israel. The larvae penetrate the tomato fruit through the stem end and present a serious threat to crop quality. Foliage and fruit samples were taken in nine commercial tomato fields located in Israel's three main tomato growing areas, two of which are potato growing areas as well. PTM was not found where potatoes were absent. Potato harvest in nearby fields was found to be the most significant factor affecting seasonal trends in PTM population density in tomatoes. All four larval instars were found in foliage on all sampling dates. Significantly higher proportions of first instars were found during the population density increase which followed potato harvest. Damaged fruits did not contain first instar larvae, indicating that PTM never undergoes complete development within tomato fruit. Fruit damage levels at harvest were positively correlated to the peak mean population densities on foliage and the date they were observed. In tomato fields not adjacent to potatoes, infestation was first observed at the edge of the field. Both before and after the potato harvest in nearby fields, population density at the edge of the field was significantly higher than at the center. In tomato fields adjacent to potatoes, no significant differences were found between population densities at the edge and center before the potatoes were harvested. Plant growth, mycorrhizal development and vesicular arbuscular spore production were examined in five tropical crop host species inoculated with Glomus clarum and grown in a glasshouse. In one of the two experiments, sequential harvests of maize, sorghum and chickpea were made in order to study spore production in relation to plant growth and mycorrhizal development. Spore numbers in each of these hosts increased at a fairly constant rate until maximum plant dry weight, when spore production ceased. Sorghum and maize produced considerably more spores than chickpea, with spore numbers being closely correlated with mycorrhizal root length. In the second experiment, Glomus clarum was cultured on each of maize, millet, sorghum, groundnut and chickpea for three consecutive generations before cross-inoculation of the spores from each host onto all five hosts. Sporulation with respect to host size was generally greatest when the inoculum used to infect a host had been produced on that host. The growth-promoting effects of the fungus were not influenced by the source of the inoculum. More spores were produced on the cereals than the legumes. Differences in spore numbers amongst hosts and plant generations were apparently influenced mainly by infected root length and by the growth period.  相似文献

Summary A total of 17 species and varieties of Fusarium have been isolated from groundnut kernels, rhizosphere and geocarposhere and from the soil of groundnut fields in Israel.F.solani predominated in the Fusarium flora of soil, rhizosphere and fresh kernels, and in particular on stored kernels. In the geocarposhere,F. solani, F. oxysporum andF. equiseti occurred in fairly equal amounts. Fusaria made up 16.2 to 32.6 per cent of the total mycoflora of kernels, soil, rhizosphere and geocarposphere.In tests with 14 isolates from groundnuts, production of toxins inducing visible reactions on rabbit skin was strongest when cultures were kept at 24 to 30°C. Only one isolate failed to produce a toxic reaction.The mortality of seedlings induced in inoculation tests with 23 groundnut isolates ofF. oxysporum, F. solani andF. equiseti was severe (mostly averaging 20 to 40%) in tomato and eggplant, moderate (mostly 10 to 20%) in bean, cucumber, watermelon and onion, weaker in cotton and pepper, and least in maize and wheat.  相似文献

Tomato (C3-plants) and maize (C4-plants) were grown in a nutrient solution to which triacontanol was added twice a week. After about 4 weeks the triacontanol treatment caused a significant increase in the dry weight of the tomato plants. Leaf area and dry weight measurements of tomato leaves at different stages of development showed that the largest increase in growth was obtained when triacontanol treatment was initiated before bud formation. In maize, no effect of the triacontanol treatment on dry wieght was observed. Photosynthesis was inhibited by 27% in young leaves from triacontanol-treated tomato plants and 39% in the controls, when the oxygen concentration was raised from 2% to 21%. In maize no change in photosynthesis could be observed, neither after altered oxygen concentration nor after triacontanol treatment. The difference in the response of C3- and C4-plants to triacontanol indicates that it regulates processes related to photosynthesis.  相似文献

Nitrate concentration and nitrate reductase activity (NRA) were studied in the leaves of soybean (Glycine max), groundnut (Arachis hypogaea and cowpea (Vigna unguiculata) and sorghum (Sorghum bicolor), pearl millet (Pennisetum americanum) and maize (Zea mays) at three nitrogen fertiliser levels in two field experiments. Higher nitrate concentrations were detected in the leaves of groundnut, cowpea and pearl millet than in sorghum and maize. Nitrate content in the leaves and leaf NRA were not related across crop species, nor was a generalised pattern of leaf NRA and leaf nitrate observed within legumes or within cereals. Nitrogen application resulted in higher nitrate availability in the leaves, with varied leaf NRA.  相似文献

The efficiency of transformation was improved by treating immature embryos with heat and centrifugation before infection with Agrobacterium tumefaciens in rice and maize. Because the effects were detected both in the levels of transgene expression after co-cultivation and in the number of independent transgenic plants obtained per embryo, conditions were first optimized based on the transgene expression, and then transformants were produced. The optimal conditions varied considerably depending on species and genotypes, but reasonably good parameters were identified for Japonica rice, Indica rice or maize. As a general tendency, the effect of centrifugation was greater than that of heat in Japonica rice, whereas that of heat was greater than that of centrifugation in Indica rice and maize A188, and the combination of the treatments was the most effective in all of the genotypes tested. The frequency of transformation was improved several fold in rice and maize. In addition, transformation of certain genotypes of maize, which were not transformable before, and transformation of maize with a less efficient vector, which could not transform maize before, became possible by these pre-treatments. In the highest case, 18 independent transgenic plants were obtained from a single immature embryo of Japonica rice. Although nothing is known about the mechanism, these pre-treatments seemed to render cells of rice and maize more competent for transformation mediated by A. tumefaciens.  相似文献

We compared responses of maize, tomato, and bean plants to water stress. Maize reached a severe water deficit (leaf water potential –1.90 MPa) in a longer period of time as compared with tomato and bean plants. Maize stomatal conductance (g s) decreased at mild water deficit. g s of tomato and bean decreased gradually and did not reach values as low as in maize. The protein content was maintained in maize and decreased at low water potential (w); in tomato it fluctuated and also decreased at low w; in bean it gradually decreased. Ribulose-1,5-bisphosphate carboxylase/oxygenase activity remained high at mild and moderate stress in maize and tomato plants; in bean it remained high only at mild stress.  相似文献

Cell-autonomous genes have been used to monitor the excision of both endogenous transposons in maize andAntirrhinum, and transposons introduced into transgenic plants. In tobacco andArabidopsis, the streptomycin phosphotransferase (SPT) gene reveals somatic excision of the maize transposonActivator (Ac) as green sectors on a white background in cotyledons of seedlings germinated in the presence of streptomycin. Cotyledons of tomato seedlings germinated on streptomycin-containing medium do not bleach, suggesting that a different assay for transposon excision in tomato is desirable. We have tested the use of the spectinomycin resistance (SPEC) gene (aadA) and a Basta resistance (BAR) gene (phosphinothricin acetyltransferase, or PAT) for monitoring somatic excision ofAc in tobacco and tomato. Both genetic and molecular studies demonstrate that genotypically variegated individuals that carry clones of cells from whichAc orDs have excised from either SPEC or BAR genes, can be phenotypically completely resistant to the corresponding antibiotic. This demonstrates that these genes act non-cell-autonomously, in contrast to the SPT gene in tobacco. Possible reasons for this difference are discussed.  相似文献

We are developing a system for isolating tomato genes by transposon mutagenesis. In maize and tobacco, the transposon Activator (Ac) transposes preferentially to genetically linked sites. To identify transposons linked to various target genes, we have determined the RFLP map locations of Ac- and Dissociation (Ds)-carrying T-DNAs in a number of transformants. T-DNA flanking sequences were isolated using the inverse polymerase chain reaction (IPCR) and located on the RFLP map of tomato. The authenticity of IPCR reaction products was tested by several criteria including nested primer amplification, DNA sequence analysis and PCR amplification of the corresponding insertion target sequences. We report the RFLP map locations of 37 transposon-carrying T-DNAs. We also report the map locations of nine transposed Ds elements. T-DNAs were identified on all chromosomes except chromosome 6. Our data revealed no apparent chromosomal preference for T-DNA integration events. Lines carrying transposons at known map locations have been established which should prove a useful resource for isolating tomato genes by transposon mutagenesis.  相似文献

Summary Fewer larvae ofMeloidogyne incognita invaded and fewer galls were formed when seedlings of nonresistant varieties of tomato, eggplant and chilli were growing in soil to which oilcakes of mahua, castor, neem/margosa, mustard and groundnut had been added. Using a soil debris isolation method, populations of Rhizoctonia solani were monitored over a 4 -yr period in four fields which were initially cropped to sugar beet and in which four areas of Rhizoctonia crown rot diseased beets (DA) and four areas of apparently healthy beets (AH) had been selected and precisely located. Soil from these areas was assayed during the subsequent crops, which included sugar beet, tomato, cucumber, maize and soybean. No significant differences in colony counts were found between the soils in DA and AH on any of 30 sampling dates. R. solani population counts were, in general, quite low, except under sugar beet and following tomato harvest. Areas of diseased beet and high R. solani soil populations that developed in subsequent sugar beet crops did not necessarily coincide with the previously selected diseased areas. High R. solani populations developed from parasitic activity on sugar beet or saprophytically on tomato crop residues. Of the other crops, both maize and soybean may have slightly increased the low R. solani residual populations in soil. The monitoring of R. solani populations in the season prior to, and during the early season of sugar beet cropping did not provide a basis for forecasting disease in fields or sites within fields. The initiation of disease patches in these sugar beet fields was therefore governed by factors other than inoculum density.  相似文献

Summary As an initial step towards developing a transposon mutagenesis system in tomato, the maize transposable element Ac was transformed into tomato plants via Agrobacterium tumefaciens. Southern analysis of leaf tissue indicated that in nine out of eleven transgenic plants, Ac excised from the T-DNA and reintegrated into new chromosomal locations. The comparison of Ac banding pattern in different leaves of the same primary transformant provided evidnece for transposition during later stages of transgenic plant development. There was no evidence of Ds mobilization in tomato transformants.  相似文献

A study on the evaluation of some fungal diseases and yield of groundnut in groundnut-based cropping systems was conducted in 2002 and 2003 planting seasons. Analysis of variance indicated that intercropping was highly significant on leaf spot disease severity 0.76; 0.75, rust 2.75; 2.69, as well as percentage defoliation 78.42%; 78.10% in 2002 and 2003, respectively. Plant population was significant on leaf spot severity 4.52, 4.60 rust 2.76; 366 and defoliation 226.5; 441.1 fungal as well as yield, while interactions were not significant on the fungal diseases and yield. Sole groundnut recorded significantly high severity of the fungal diseases investigated but low yield, when groundnut was intercropped with maize and melon and recorded the lowest yield in 2002 and 2003 respectively. 250,000 plants/ha recorded the lowest severity of the diseases investigated, while 444,444 plants/ha recorded the highest. Sole groundnut also recorded the highest percentage defoliation 79.37%, 79.25% when groundnut was intercropped with maize recorded the lowest 77.06%; 77.60%. 250,000 plants/ha had the lowest defoliation when 444,444 plants/ha had the highest 80.75% 82.13% in 2002 and 2003, respectively. Sole groundnut recorded the lowest in the majority of yields when intercropped with maize and with melon recorded a high yield and yield components in 2002 and 2003, respectively. The microorganisms identified were Cercospora spp., Aspergillus linked and Blastomyces.  相似文献

Plant oils (cottonseed, soybean, corn, groundnut and palm) at different dosages were evaluated in the laboratory for their ability to suppress the populations ofCryptolestes pusillus andRhyzopertha dominica in maize and sorghum. Exposure of adults of both beetle species to grains treated with 10 ml/kg of the different oils induced 100% mortality within 24 h. A dose of 5 ml/kg of each oil significantly decreased the progeny produced byR. dominica. Complete protection was achieved on grains treated with 10 ml/kg. These oils also repelled the adults of both species. Percentage weight loss caused byR. dominica in grains treated with 5 ml/kg and 10 ml/kg levels were significantly lower than in untreated grains. Oil treatment did not affect the germination of, or water absorption by, maize and sorghum grains compared with untreated grains. The potential use of plant oils in the management of insect pests in traditional grain storage is discussed.  相似文献

The molecular weight exclusion limit of plasmodesmata in subveinal epidermal cells of Nicotiana clevelandii (Gray) leaves was estimated by microinjection and fluorescence microscopy using fluorescein isothiocyanate-peptide conjugates, carboxyfluorescein and Lucifer Yellow CH. The largest fluorochrome which moved symplastically between cells had a molecular weight of 749, although movement did not appear to depend purely on molecular weight parameters. Systemic infection of plants by tobacco rattle tobravirus, tomato black ring nepovirus or potato Y potyvirus did not alter the limits of plasmodesmatal conductance of the fluorochromes. However, carrot mottle umbravirus and groundnut rosette umbravirus diminished the symplastic mobility of some fluorescent tracers. Field trials were designed to investigate the effect of direct nitrate fertilisation and mucuna fallow on maize yield and borer attacks in the humid forest zone of Cameroon. A traditional maizecassava- groundnut system (farmers’ practice) was compared with a maize-cassava + 120 Kg N ha-1, a rotation system in which maize-cassava followed a mucuna fallow as well as with a maize monocrop grown after mucuna fallow and with a maize monocrop grown with 120 Kg N ha-1. Average egg batch densities of Busseola fusca (Lepidoptera: Noctuidae) were lower by 35–55% in inter- than monocrops but the effect of nitrogen on B. fusca oviposition was not different from that of mucuna fallow. Highest larval infestations were found in sole maize with nitrogen and sole maize after mucuna. Yield losses were 4–10 times higher in the farmers’ practice compared to maize-cassava after mucuna and maizecassava with nitrogen. Mixed cropping systems including farmers’ practice yielded higher total gross and net benefits compared to sole maize crops. But, they were higher in maize-cassava intercrop + N than for farmer’s practice.  相似文献

Necrotic lesions surrounded by yellow areas on leaves followed by stem necrosis were observed on chrysanthemum plants cultivated in Atibaia, Sào Paulo State, Brazil. The host range, in vitro properties and particle morphology of the causal virus were typical of a Tospovirus. Serological studies demonstrated that the virus differed from tomato spotted wilt. tomato chlorotic spot, groundnut ringspot and inpatiens necrotic spot Tospoviruses. The virus isolate is thus possibly a representativeof a new serogroup of a new virus.  相似文献

Antibiotic resistance genes can act as either cell autonomous or non-cell autonomous genetic markers with which to monitor the excision of plant transposons. To convert spectinomycin resistance from a noncell autonomous resistance to cell autonomous resistance, a transit peptide for chloroplast localization from a petunia ribulose bisphosphate carboxylase (rbcS) gene was fused in-frame to the aadA gene, which confers spectinomycin and streptomycin resistance. Constructs were generated in which the expression of this chimeric gene was prevented by the presence, in the 5 untranslated leader, of the maize transposons Activator (Ac) or Dissociation (Ds). When progeny of tobacco or tomato plants transformed with these constructs were germinated on spectinomycin-containing medium, germinally revertant and somatically variegated individuals could be distinguished.  相似文献
