水稻梯田生态系统具有多重服务价值,全球尺度上相关研究的系统梳理和总结可促进该领域的理解和认识。本文依据千年生态系统服务分类系统,采用系统文献分析法对全球水稻梯田生态系统服务研究进行整理并综述其进展。已发表的全球水稻梯田生态系统服务研究文献主要分布在中国、越南、菲律宾。从评价方法看,中国偏重于价值量评价,而国外偏重于物质量评价;国内4个梯田的价值量评价显示,梯田的调节和支持服务高于其他服务,但由于计算单元和方法不同难以相互相比;物质量评价方面,调节服务研究开展了较为深入的水土流失、灾害和虫害调节研究,支持服务对水稻生长有益的硅元素的循环转化进行了研究,供给服务以影响水稻生长的因素及其情景预测为主;景观尺度上,增加其他作物种植斑块可提高各类服务;基于生态系统服务矩阵与制图的分析框架具有良好应用前景。未来还应加强水稻梯田生态系统服务多尺度评估体系,从服务形成机理出发研究权衡协同关系,并加强景观尺度上的格局-过程-服务-人类福祉间的系统研究。  相似文献

Discriminant analysis of 23 digital and 15 palmar quantitative dermatoglyphic variables of 1364 Sardinians, 689 males and 675 females, were performed to identify biological relationships among five Sardinian linguistic groups of both sexes. By ANOVA 17 of the 23 digital variables and 4 of the 15 palmar variables were significantly different among males and females of the Sardinian linguistic groups. By MANOVA the results indicate high intergroup heterogeneity also within each sex. By stepwise discriminant analysis with p≤0.01 as F-to-enter and p≤0.05 as F-to-remove, only 4 of the 38 digital and palmar variables were in the model (URC R5, RRC L5, RRC R5, URC R4). The pattern of interpopulation biological relationships shows a clearly distinct position of the Gallurian group (both males and females) which speaks an Italian dialect. The properly Sardinian linguistic groups (Campidanian and Logudorian), the Sassarian group (which speaks and Italian dialect) and the Alghero group (which speaks Catalan) were close to one another. This pattern agrees more with the ethno-historical background than with the linguistic one.  相似文献

本文采用定量PCR方法观察了慢性乙型肝炎患者HBV-DNA的复制水平。结果表明在HBeAg阳性患者血清HBV-DNA检出率和水平显著高于HBeAg阴性患者(P<0.01);血清HBV-DNA水平的高低与ALT异常情况未发现有相关性。  相似文献

A nested polymerase chain reaction (PCR) protocol using unique primers was developed to detect and quantify Myxococcus species from environmental samples. The protocol amplified most species of Myxococcus when 10 pg of DNA representing 1000 cells was present, although over half were amplified with as little as 0.1 pg (10 cells). The protocol did not amplify other myxobacterial species, members of the δ‐proteobacteria or other unrelated organisms tested at significantly higher concentrations of DNA. The primers were also used in quantitative PCRs, which accurately estimated the population levels in soil.  相似文献

生态过渡性是梯度环境的特征之一,过渡带的显著性是地理学和地植物学中长期争论的重要问题,生态交错带位置和宽度的判定是定量研究交错带生态学过程的基础。样带法是采集非连续梯度数据和研究交错带的结构,功能和格局梯度变化的有效方法。生态交错带变化最为显著的特征是植被的变化,包括植物种类组成和植被结构的变化,植被生态学群落分析中的相异系数,β多样性,梯度分析和分类排序技术等已被证明是成熟可靠的方法。介绍两种交错带定量判定的有效方法。游动分割窗技术是通过滑动求取样带上样点间相异系数,通过相异系数变化曲线判定交错带的位置和宽度。分割窗技术提供了敏感地定位非连续梯度的客观且有效的方法,它不仅可用于环境梯度上的等距离样点取样,而且可用于非等距离梯度带多元变量的分析。植被的特征是对生态环境连续体的综合反应,植被在环境梯度上的排序参数提供了一种较好的交错带定量指标,样带植被数据特征参数的变异轮廓图能够反应植被沿样带的物种组成,结构和空间变化格局,可以反应植被沿环境梯度的β多样性和植物群落的梯度分化,植被变异侧面轮廓图的间断或不连续(突变)区间就是群落边界的位置。  相似文献

目的:对目前最常用的检测微小RNA(miRNA)的茎环实时定量PCR法和PAP实时定量PCR法进行比较。方法:分别用茎环实时定量PCR法和PAP实时定量PCR法检测人乳腺癌细胞MCF-7中U6和23种miRNA的表达,利用定量PCR分析软件和琼脂糖凝胶电泳方法,将2种方法在引物设计难度、特异性与灵敏度,以及检测通量方面进行比较。结果:茎环法的特异性和灵敏度比PAP法高,但引物设计难度大,检测通量低;PAP法引物设计难度较低,检测通量较高,但特异性和灵敏度较差。结论:茎环法实时定量PCR适于有针对性地检测小规模miRNA,而PAP法则适于大规模miRNA筛选实验。  相似文献

用胸腺素免疫新西兰兔,获得抗胸腺素抗体,建立了胸腺素ELISA定量测定法。该法检测灵敏度为3.2ng/ml,批内和批间变异系数分别为4.4%和8.9%,方法特异性鉴定显示与另外7种激素无明显交叉反应。该方法灵敏度高、专一性强、重复性好,可用于胸腺素产品的质量检测,具有较大实用价值  相似文献

日新月异的生命科学技术的发展及临床医学科学研究的需求,一般的PCR技术已远远不能满足工作的需要。PE公司在进行了大量的PCR动力学研究的基础上,发现了利用荧光标记探针在PCR循环过程中积累的荧光强度达到仪器捡出阈值时,系统的初始模板数量与循环次数之间有线性关系,据此建立了目前的PE 7700 、PE 5700仪器的定量PCR技术,开创了PCR技术的新局面。但是由于这一技术的误差较大,尚不能满足生命科学及临床医学科学研究的需求,因此需要继续研究新的定量PCR技术。PCR动力学数学模型是根据PCR 技术的原理提出的,能够准确描述PCR反应产物分子数量积累规律的动力学方程,给出了PCR产物数量或者荧光强度与初始模板数量及其他反应条件间的函数关系。利用这一关系,根据PCR反应已积累的产物数量,可以实现准确的定量PCR分析,得到初始模板数量达到定量PCR的目的。使用动力学数学模型做定量PCR分析,其结果的误差仅与使用的荧光强度数值的精确度相关。使用精确到6位数的荧光强度数据,模板数自100~1 000 000区间定量结果的准确性可达99%以上。本文根据模拟实验数据进行了初步的定量PCR分析,结果提示,目前的定量PCR仪器使用PCR动力学模型理论处理分析数据,定量分析的结果会比目前的CT值方法在准确性方面提高几十倍以上,可以满足各方面研究工作误差水平的需要。 Abstract:Today standard PCR can't satisfy the need of biotechnique development and clinical research any more.After numerous dynamic research,PE company found there is a linear relation between initial template number and cycling time when the accumulating fluorescent product is detectable.Therefore,they developed a quantitative PCR technique to be used in PE7700 and PE5700.But the error of this technique is too great to satisfy the need of biotechnique development and clinical research.A better quantitative PCR technique is needed.The mathematical model submitted here is combined with the achievement of relative science,and based on the PCR principle and careful analysis of molecular relationship of main members in PCR reaction system.This model describes the function relation between product quantity or fluorescence intensity and initial template number and other reaction conditions,and can reflect the accumulating rule of PCR product molecule accurately.Accurate quantitative PCR analysis can be made use this function relation.Accumulated PCR product quantity can be obtained from initial template number.Using this model to do quantitative PCR analysis,result error is only related to the accuracy of fluorescence intensity or the instrument used.For an example,when the fluorescence intensity is accurate to 6 digits and the template size is between 100 to 1 000 000,the quantitative result accuracy will be more than 99%.The difference of result error is distinct using same condition,same instrument but different analysis method.Moreover,if the PCR quantitative analysis system is used to process data,it will get result 80 times of accuracy than using CT method.  相似文献

基于数据依赖的扫描模式(data-dependent acquisition, DDA)和数据非依赖的扫描模式(data-independent acquisition,DIA)的非标记定量(label-free quantitative,LFQ)和同位素标记TMT (tandem mass tag)定量是蛋白质组学定量中较常见的技术.本文利用最新的Orbitrap Exploris 480质谱,优化了DDA、FAIMS DDA、FAIMS DIA的非标记定量方法以及TMT定量策略的关键质谱参数,并将其应用在人细胞蛋白质组、单细胞蛋白质组、血浆蛋白质组和酵母蛋白质组分析.结果表明,在DDA实验中,设置碰撞能量为27、二级谱图的分辨率为15 K、最大离子注入时间为22 ms是最佳的参数组合.针对极微量样品200 pg~5 ng,可以根据样品量相应设置最佳的质谱参数.使用200 pg和500 pg的HeLa细胞样品,分别鉴定到1 259和1 725个蛋白质,从而实现了单细胞蛋白质组学的深度覆盖.在FAIMS DDA实验中,60 min或90 min梯度时选择CV-45V的补偿电压,120 ...  相似文献

We investigate the interplay between gene flow and adaptation in peripheral populations of a widespread species. Models are developed for the evolution of a quantitative trait under clinally varying selection in a species whose density decreases from the center of the range to its periphery. Two major results emerge. First, gene flow from populations at the range center can be a strong force that inhibits peripheral populations from evolving to their local ecological optima. As a result, peripheral populations experience persistent directional selection. Second, response to local selection pressures can cause rapid and substantial evolution when a peripheral population is isolated from gene flow. The amount of evolutionary change depends on gene flow, selection, the ecological gradient, and the trait's heritability. Rapid divergence can also occur between the two halves of a formerly continuous population that is divided by a vicariant event. A general conclusion is that disruption of gene flow can cause evolutionary divergence, perhaps leading to speciation, in the absence of contributions from random genetic drift.  相似文献

The genotypic basis of morphological variation is largely unknown. In this study we examine patterns of pleiotropic effects on mandibular morphology at individual gene loci to determine whether the pleiotropic effects of individual genes are restricted to functionally and developmentally related traits. Mandibular measurements were obtained from 480 mice from the F2 generation of an intercross between the LG/J and SM/J mouse strains. DNA was also extracted from these animals, and 76 microsatellite loci covering the autosomes were scored. Interval mapping was used to detect chromosomal locations with significant effects on various mandibular measurements. Sets of traits mapping to a common chromosomal region were considered as being affected by a single quantitative trait locus (QTL) for mandibular morphology. Thirty-seven such chromosomal regions were identified spread throughout the autosomes. Gene effects were small to moderate with the allele derived from the LG/J strain typically leading to larger size. When dominance was present, the LG/J allele was typically dominant to the SM/J allele. Most loci affected restricted functional and developmental regions of the mandible. Of the 26 chromosomal regions affecting more than two traits, 50% affect the muscular processes of the ascending ramus, 27% affect the alveolar processes carrying the teeth, and 23% affect the whole mandible. Four additional locations affecting two traits had effects significantly associated with alveolar regions. Pleiotropic effects are typically restricted to morphologically integrated complexes.  相似文献

定量PCR是在PCR定性技术基础上发展起来的核酸定量技术,定量检测时常用编码甘油醛-3-磷酸脱氢酶、β-肌动蛋白、28S和18SrRNA等的持家基因作为内部参照,这些基因被认为在某些类型细胞中的表达是恒定的。近年来,很多研究者发现上述持家基因的表达水平并不稳定,利用它们作为内标来定量并不准确。因此,在进行定量实验时应选择适当的2种或2种以上的内参基因,以减少检测标本间的差异。  相似文献

Phytate, being the major storage form of phosphorus in plants, is considered to be an anti-nutritional substance for human, because of its ability to complex essential micronutrients. In the present study, we describe the genetic analysis of phytate and phosphate concentrations in Brassica rapa using five segregating populations, involving eight parental accessions representing different cultivar groups. A total of 25 quantitative trait loci (QTL) affecting phytate and phosphate concentrations in seeds and leaves were detected, most of them located in linkage groups R01, R03, R06 and R07. Two QTL affecting seed phytate (SPHY), two QTL affecting seed phosphate (SPHO), one QTL affecting leaf phosphate and one major QTL affecting leaf phytate (LPHY) were detected in at least two populations. Co-localization of QTL suggested single or linked loci to be involved in the accumulation of phytate or phosphate in seeds or leaves. Some co-localizing QTL for SPHY and SPHO had parental alleles with effects in the same direction suggesting that they control the total phosphorus concentration. For other QTL, the allelic effect was opposite for phosphate and phytate, suggesting that these QTL are specific for the phytate pathway.  相似文献

In a study of the inheritance of a quantitative trait in the general population, data are collected from the three-generational nuclear families of many index cases. The question arises as to the optimal choice of family members to maximise the power of testing a hypothesis of polygenic and environmental effects against an alternative of environmental effects only. The method chosen is to minimise the asymptotic variance of the ratio of polygenic to environmental variance, for fixed total variance and the environmental correlations between family members. Using Taylor expansions in the region where the ratio is close to zero, an approximate expression for the variance of the ratio is obtained. This expression can then be evaluated for all possible structures of a three-generational nuclear family of a given size.  相似文献
