Divalent cations act as bridges among extracellular polymeric substances (EPS) and form cross-linkage for the self-immobilization of microbial biomass. However, their effects on the nitrification performance during the biological nitrogen removal are still unclear. In the present study, the effects of Mg2+ on the nitrifying activity, EPS and floc characteristics were investigated using a lab-scale sequencing batch reactor. The distribution of Mg2+ was quantified at different level of sludge floc. The results indicated that the nitrification activity was significantly improved when influent Mg2+ was below 1.1 mmol/L, but suppressed at 3 mmol/L. The overall performance characterized by COD, NH4+-N and TN, the particle size and sludge flocculation ability rapidly increased with the increase of Mg2+ concentration. Mg2+ was mainly distributed in the pellet and changed slightly in supernatant, LB-EPS and TB-EPS. The four fluorescence peaks detected by three-dimensional excitation-emission matrix spectra were attributed to PN-like substances and humic acid-like substances in the LB-EPS and TB-EPS. The results of XPS analysis demonstrated that LB-EPS and TB-EPS comprised similar elements. Therefore, the types of EPS functional groups was unchanged under varied Mg2+ concentrations, while their proportions changed and LB-EPS/EPS was key factor for the changes of bioflocculation.  相似文献

Laboratory experiments were carried out on activated sludge (AS) to investigate the correlations between the content of extracellular polymeric substances (EPS) and the performance of biosolids–water separation, including sludge flocculation, sedimentation, compression, and dewatering, under non-steady-state conditions. On three stabilized AS reactors changes were made in sludge retention time (SRT), substrate composition, and loading rate, respectively, to bring about unstable operation to the reactors. A two-step heating method was used to extract from the sludge the easily extractable EPS, or loosely bound EPS (LB-EPS), and tightly bound EPS (TB-EPS), respectively. The experimental results demonstrate dynamic changes in sludge characteristic and EPS production under the non-steady-state conditions. During the early phase of transition after a change was imposed, the sludge became generally worse in flocculation, compressibility, and dewaterability. With the acclimatization of the biomass to the new process conditions, biosolids–water separation showed a general trend of improvement. Changes in AS process condition also resulted in considerable variations in EPS production. The change of the LB-EPS content appeared to be more significant than that of the TB-EPS. Throughout the non-steady-state operation, the sludge flocculating behavior, settleability, compressibility, and dewaterability had a positive correlation with the LB-EPS content; however, no correlation could be found between these properties and the TB-EPS content. The results suggest that although EPS is essential to biofloc formation, excessive EPS in the form of LB-EPS would weaken cell attachment and deteriorate the AS floc structure, resulting in poor biosolids–water separation.  相似文献

This study quantified the activities of enzymes in extracellular polymeric substances (EPS) and in pellets. Seven commonly adopted extraction schemes were utilized to extract from aerobic flocs the contained EPS, which were further categorized into loosely bound (LB) and tightly bound (TB) fractions. Ultrasonication effectively extracted the EPS from sludge flocs. Enzyme assay tests showed that the protease activity was localized mainly on the pellets, α-amylase and α-glucosidase activities were largely bound with LB-EPS, and few protease, α-amylase, or α-glucosidase activities were associated with the TB-EPS fraction. There exists no correlation between the biochemical compositions of EPS and the distribution of enzyme activities in the sludge matrix. The 44–65% of α-amylase and 59–100% of α-glucosidase activities noted with the LB-EPS indicate heterogeneous hydrolysis patterns in the sludge flocs with proteins and carbohydrates.  相似文献

The optimization, purification and characterization of an extracellular polysaccharide (EPS) from a bacterium Paenibacillus polymyxa SQR-21 (SQR-21) were investigated. The results showed that SQR-21 produced one kind of EPS having molecular weight of 8.96 × 105 Da. The EPS was comprised of mannose, galactose and glucose in a ratio of 1.23:1.14:1. The ratio of monosaccharides and glucuronic acid was 7.5:1. The preferable culture conditions for EPS production were pH 6.5, temperature 30 °C for 96 h with yeast extract and galactose as best N and C sources, respectively. The maximum EPS production (3.44 g L−1) was achieved with galactose 48.5 g L−1, Fe3+ 242 μM and Ca2+ 441 μM. In addition, the EPS showed good superoxide scavenging, flocculating and metal chelating activities while moderate inhibition of lipid peroxidation and reducing activities were determined. These results showed the great potential of EPS produced by SQR-21 to be used in industry in place of synthetic compounds.  相似文献

A laboratory-scale submerged anaerobic membrane bioreactor (SAnMBR) treating thermomechanical pulping whitewater was operated for over 7 months to investigate and compare the characteristics of cake sludge and bulk sludge during stable state operation period. Serial analysis showed that cake sludge had a smaller particle size distribution (PSD), much higher specific filtration resistance (1.34 × 1014 m/kg), 1.5 times higher bound EPS and significantly different microbial community as compared with bulk sludge. Further analysis indicated that small flocs, bound EPS and inorganic materials play important role in cake formation process. The formed cake layer was found to have a heterogeneous structure. The results obtained in this study indicated that cake formation process started from attachment of small flocs and/or specific bacterial clusters which colonize the surface of the membrane and provide enhanced conditions that allow for cake formation to progress.  相似文献

In this study, we examined the long-term effects of copper oxide nanoparticles (CuO NPs) on the production and properties of EPS and the resulting variations in surface physicochemical characteristics of biofilms in a sequencing batch biofilm reactor. After exposure to 50 mg/L CuO NPs for 45 days, the protein (PRO) and polysaccharide (PS) contents in loosely bound EPS (LB-EPS) decreased as the production of LB-EPS decreased from 34.4 to 30 mg TOC/g EPS. However, the production of tightly bound EPS (TB-EPS) increased by 16.47 % as the PRO and PS contents increased. The content of humic-like substances (HS) increased significantly, becoming the predominant constituent in EPS with the presence of 50 mg/L CuO NPs. Furthermore, the results of three-dimensional excitation-emission fluorescence spectra confirmed the various changes in terms of the LB-EPS and TB-EPS contents after exposure to CuO NPs. Fourier transform infrared spectroscopy showed that the –OH and –NH2 groups of proteins in EPS were involved in the reaction with CuO NPs. Moreover, the chronic exposure to CuO NPs induced a negative impact on the flocculating efficiency of EPS and on the hydrophobicity and aggregation ability of microbial cells. The PRO/PS ratios of different EPS fractions were consistent with their hydrophobicities (R 2 >0.98) and bioflocculating efficiencies (R 2 >0.95); however, there was no correlation with aggregation ability. Additionally, the presence of bovine serum albumin (BSA) prevented the physical contact between CuO NPs and EPS as a result of NP aggregation and electrostatic repulsion.  相似文献

Extracellular polymeric substances (EPS) play important roles in regulating the dewaterability of sludge. This study sought to elucidate the influence of EPS on the dewaterability of sludge during bioleaching process. Results showed that, in bioleaching system with the co-inoculation of Acidithiobacillus thiooxidans TS6 and Acidithiobacillus ferrooxidans LX5 (A. t+A. f system), the capillary suction time (CST) of sludge reduced from 255.9 s to 25.45 s within 48 h, which was obviously better than the controls. The correlation analysis between sludge CST and sludge EPS revealed that the sludge EPS significantly impacted the dewaterability of sludge. Sludge CST had correlation with protein content in slime and both protein and polysaccharide contents in TB-EPS and Slime+LB+TB layers, and the decrease of protein content in slime and decreases of both protein and polysaccharide contents in TB-EPS and Slime+LB+TB layers improved sludge dewaterability during sludge bioleaching process. Moreover, the low sludge pH (2.92) and the increasing distribution of Fe in the solid phase were another two factors responsible for the improvement of sludge dewaterability during bioleaching. This study suggested that during sludge bioleaching the growth of Acidithiobacillus species resulted in the decrease of sludge pH, the increasing distribution of Fe in the solid phase, and the decrease of EPS content (mainly including protein and/or polysaccharide) in the slime, TB-EPS, and Slime+LB+TB layers, all of which are helpful for sludge dewaterability enhancement.  相似文献

This study investigated soluble (Sol-EPS), loosely bound (LB-EPS), and tightly bound extracellular polymeric substances (TB-EPS) harvested from biofilm and planktonic cultures of the marine bacterium Pseudoalteromonas ulvae TC14. The aim of the characterization (colorimetric methods, FTIR, GC-MS, NMR, HPGPC, and AFM analyses) was to identify new anti-biofilm compounds; activity was assessed using the BioFilm Ring Test®. A step-wise separation of EPS was designed, based on differences in water-solubility and acidity. An acidic fraction was isolated from TB-EPS, which strongly inhibited biofilm formation by marine bacterial strains in a concentration-dependent manner. The main constituents of this fraction were characterized as two glucan-like polysaccharides. An active poly(glutamyl-glutamate) fraction was also recovered from TB-EPS. The distribution of these key EPS components in Sol-EPS, LB-EPS, and TB-EPS was distinct and differed quantitatively in biofilm vs planktonic cultures. The anti-biofilm potential of the fractions emphasizes the putative antifouling role of EPS in the environment.  相似文献

The composition and distribution of extracellular polymeric substance (EPS) both from suspended sludge and attached biofilm were investigated in a simultaneous nitrification and denitrification (SND) system with the increase of the salinity from 1.0 to 3.0 %. Fourier-transform infrared (FTIR) spectroscopy and three-dimensional excitation-emission matrix (3D-EEM) fluorescence spectroscopy were used to examine proteins (PN), polysaccharides (PS) and humic substances (HS) present in EPS. High total nitrogen removal (above 83.9 %) via SND was obtained in the salinity range of 1.0–2.5 %. Total EPS in the sludge increased from 150.2 to 200.6 mg/gVSS with the increase of salinity from 1.0 to 3.0 %, whereas the corresponding values in the biofilm achieved the maximum of 288.6 mg/g VSS at 2.0 % salinity. Dominant composition of EPS was detected as HS in both sludge and biofilm, having the percentages of 50.6–68.6 and 41.1–69.9 % in total EPS, respectively. Both PN and PS contents in soluble EPS (S-EPS), loosely bound EPS (LB-EPS) and tightly bound EPS (TB-EPS) of sludge and biofilm increased with the increased salinity. The FTIR spectrum and 3D-EEM fluorescence spectroscopy of S-EPS, LB-EPS and TB-EPS in the sludge and biofilm showed the changes of functional groups and conformations of the compositions in EPS with the increase of salinity. The results demonstrated that the characteristics of EPS varied from sludge to biofilm. The obtained results could provide a better understanding of the salinity effect on the EPS characteristics in a SND system.  相似文献

The Iberian Peninsula encompasses more than 80% of the species richness of European aquatic ranunculi. The floristic diversity of the phytocoenosis characterised by aquatic Ranunculus and the main physical–chemical factors of the water were studied in 43 localities of the central Iberian Peninsula. Four aquatic Ranunculus communities are found in most of the aquatic environments. These are species-poor and have an uneven distribution: three species of Batrachium are heterophyllous and their communities are distributed in different aquatic ecosystems on silicated substrates; one species is homophyllous and its community occurs in various aquatic ecosystems with carbonated waters. In the Mediterranean climate, Ranunculus species are present in different habitats, as shown by the results of all the statistical analyses. Ranunculus trichophyllus communities occur in base-rich waters with a high buffering capacity (2273.44 ± 794.57 mg CaCO3 L−1) and a high concentration of cations (Ca2+, 121 ± 33.12 mg L−1; Mg2+, 71.64 ± 82.77 mg L−1), nitrates (2.89 ± 4.80 mg L−1), ammonium (2.19 ± 1.36 mg L−1) and sulphates (216.25 ± 218.54 mg L−1). Ranunculus penicillatus communities are found in flowing waters with a high concentration of phosphates (0.48 ± 0.6 mg L−1) and intermediate buffering capacity (683.66 ± 446.76 mg CaCO3 L−1). Both Ranunculus pseudofluitans and Ranunculus peltatus communities grow in waters with low buffering capacity (R. pseudofluitans, 385.91 ± 209.2 mg CaCO3 L−1; R. peltatus, 263.3 ± 180.36 mg CaCO3 L−1), and a low concentration of cations (R. pseudofluitans: Ca2+, 12.57 ± 9.42 mg L−1; Mg2+, 3.42 ± 1.67 mg L−1; R. peltatus: Ca2+, 15 ± 18.26 mg L−1; Mg2+, 6.26 ± 8.89 mg L−1) and nutrients (R. pseudofluitans: nitrates, 0.23 ± 0.2 mg L−1; phosphates, 0.09 ± 0.1 mg L−1; R. peltatus: nitrates, 0.19 ± 0.21 mg L−1; phosphates, 0.09 ± 0.12 mg L−1); the first in flowing waters, the latter in still waters.  相似文献

The feasibility of composite hydrolysis enzymes in enhanced dewatering of waste-activated sludge (WAS) was verified in this study. A Pearson correlation analysis was conducted to explore the roles of different extracellular polymeric substance (EPS) fractions on WAS dewaterability. The results indicated that tightly bound EPS (TB-EPS) was released into the liquid phase consistently during enzymatic hydrolysis to form soluble EPS (S-EPS) and loosely bound EPS and that the TB-EPS content was positively correlated with the capillary suction time of WAS. A kinetic analysis was carried out to gain further insights into the kinetic variation in TB-EPS removal. It was found that TB-EPS reduction fit a first-order kinetic model and that mild temperature (25–30 °C) and a slightly acidic condition were favorable for the improvement of enzyme activity. Solid phase extraction combined with UV–Vis spectroscopy analysis was used to characterize the processes of migration and transformation of the hydrophobic (HPO), transphilic and hydrophilic (HPI) fractions in EPS during the enzymatic process. The results revealed that HPO and HPI were mainly composed of PN and PS, respectively, and that the enzymatic hydrolysis could enhance the transformation of HPI from TB-EPS to S-EPS, which was the dominant mechanism of improving WAS dewaterability.  相似文献

Bacillus megaterium RB-05 was grown on glucose and on “tossa-daisee” (Corchorus olitorius)-derived jute, and production and composition of extracellular polysaccharide (EPS) were monitored. An EPS yield of 0.065 ± 0.013 and of 0.297 g ± 0.054 g−1 substrate after 72 h was obtained for glucose and jute, respectively. EPS production in the presence of jute paralleled bacterial cellulase activity. High performance liquid chromatography (HPLC), matrix assisted LASER desorption/ionization-time of flight (MALDI-ToF) mass spectroscopy, and fourier transform infrared (FT-IR) spectroscopy demonstrated that the EPS synthesized in jute culture (JC) differed from that synthesized in glucose mineral salts medium (GMSM). While fucose was only a minor constituent (4.9 wt.%) of EPS from GMSM, it a major component (41.9 wt.%) of EPS synthesized in JC. This study establishes jute as an effective fermentation substrate for EPS production by a cellulase-producing bacterium.  相似文献

Optimum culture conditions which ease the synthesis of a novel exopolysaccharide (EPS) from a potent marine strain Streptococcus phocae was proposed in this study. The strain grows well at 35 °C, pH 7.0 and NaCl (2%) with lactose and yeast extract as best carbon and nitrogen sources. The maximum yield of EPS (11.75 and 12.14 g/L) was obtained in the presence of lactose and yeast extract at a concentration of 20 g/L respectively. EPS was refined by gel filtration chromatography using phenyl Sepharose column which revealed the presence of arabinose, fructose and galactose sugar units with molecular mass about 2.8 × 105 Da. Emulsifying and flocculating stability of EPS compared with three commercial hydrocolloids. EPS exhibited better activities which are similar to that of commercial hydrocolloids. Both crude and purified EPS exhibited strong antioxidant potential by quenching hydroxyl and superoxide anion radicals. Antibiofilm activity by inhibition of Gram positive and Gram negative biofilm forming bacteria was evident in our studies. Potential antioxidant activity and biofilm inhibiting property of EPS may lead to the development of novel food grade adjuncts.  相似文献

The main goal of this research was to investigate how different factors influence membrane fouling. The impact of the different concentrations of activated sludge and the amount of extracellular polymer substances (EPS) were monitored. Two pilot plants with submerged membrane modules (hollow fiber and flat sheet) were operated and the raw wastewater was used.Humic substances were identified as the major components of EPS in the activated sludge (more than 34%) in both pilot plants. As the basic constituent in permeate, humic substances were identified as the most dominant components in the effluent (61%) in both pilot plants. Conversely, proteins were mostly analyzed in permeate and supernatant below the detection limit. The total amount of EPS [mg g−1 (VSS)] was similar for concentrations of activated sludge 6, 10 and 14 g L−1. Carbohydrates were identified as the component of EPS which tends most to clog membranes.  相似文献

The contributions of loosely bound extracellular polymeric substances (LB-EPS), tightly bound EPS (TB-EPS), residual sludge (the sludge left after EPS extraction) and functional groups such as amine, carboxyl, phosphate and lipid on aerobic granules on biosorption of four different dyes (Reactive Brilliant Blue KN-R (KN-R), Congo Red (CR), Reactive Brilliant Red K-2G (RBR) and Malachite Green (MG)) were investigated. EPS may be responsible for biosorption of cationic dyes. However, residual sludge always made greater contribution than that of EPS. The biosorption mechanisms were dependent on the functional groups on aerobic granules and dyes’ chemical structures. The lipid and phosphate groups might be the main binding sites for KN-R biosorption. Amine, carboxyl, phosphate and lipid were all responsible for the binding of CR. The lipid fractions played an important role for RBR biosorption. For MG, the phosphate groups gave the largest contribution.  相似文献

The major operational problem associated with membrane bioreactors (MBR) is membrane fouling, for which extracellular polymeric substances (EPS) are primarily responsible. In this work both the soluble and bound EPS (i.e. SMP and EPS) produced in an MBR system operating under sludge retention times (SRT) of 10, 15, 20 and 33 days were fractionized by means of membranes having variable molecular weight cutoffs (300 kDa, 100 kDa, 10 kDa & 1 kDa). The results show that increasing the SRT leads to a reduction of SMP and EPS and that these reductions are more pronounced for the SRTs in the range 10–20 days. This reduction is more significant for carbohydrates than for proteins. The decrease of SMP and EPS with increasing SRT from 10 to 20 days led to a significant decrease of the level of fouling. The further increase of SRT to 33 days did not significantly impact on the level of fouling as the SMP and EPS concentrations did not change much.  相似文献

Microbial community developments and biomass characteristics (concentration, particle size, extracellular polymeric substances (EPS), and membrane fouling propensity) were compared when three MBRs were fed with the synthetic wastewater at different organic loadings. Results showed that the bacterial communities dynamically shifted in different ways and the EPS displayed dissimilar profiles under various organic loadings, which were associated with the ratios of food to microorganism and dissolved oxygen levels in the MBRs. The membrane fouling tendency of biomass in the low-loading MBR (0.57 g COD/L day) was insignificantly different from that in the medium-loading MBR (1.14 g COD/L day), which was apparently lower than that in the high-loading MBR (2.28 g COD/L day). The membrane fouling propensity of biomass was strongly correlated with their bound EPS contents, indicating cake layer fouling (i.e., deposition of microbial flocs) was predominant in membrane fouling at a high flux of 30 L/m2 h.  相似文献

Soluble extracellular polymeric substances (EPSs) cause membrane fouling in membrane bioreactors (MBRs), correlated with MBR sludge characteristics. Effects of F/M ratios on the evolution of soluble EPSs, fouling propensity of supernatants, and sludge metabolic activity were measured in this study in a two-period sequencing batch reactor (SBR). The experimental results show that fouling propensity was directly correlated with soluble-EPS concentration and composition. Sludge that had entirely lost active cells by long-term starvation released 64.4 ± 0.9 mg/L of humic acids, which caused a rapid increase in membrane resistance (40.67 ± 2.24 × 1011 m−1) during fouling tests. During short-term starvation, induced by incubation at a normal to low F/M ratio of 0.05 d−1, sludge can use previously secreted utilization-associated products (UAPs) to maintain endogenous respiration. Therefore, the strategies of accumulating sludge and prolonging sludge retention time in MBRs may create long-term starvation and promote membrane fouling.  相似文献

A phthalylated ion-exchange biopolymer was obtained by adding cellulose to molten phthalic anhydride in a quasi solvent-free procedure. Through this route 2.99 ± 0.07 mmol g−1 of pendant groups containing ester and carboxylic acid moieties were incorporated into the polymeric structure that was characterized by elemental analysis, solid-state carbon nuclear magnetic resonance (CP/MAS), infrared spectroscopy, X-ray diffraction, and thermogravimetry. The chemically modified polysaccharide is able to exchange cations from aqueous solution as demonstrated by batchwise methodology. The data were adjusted to a modified Langmuir equation to give 2.43 ± 0.12 and 2.26 ± 0.11 mmol g−1 for divalent cobalt and nickel cations, respectively. The net thermal effects obtained from calorimetric titration measurements were also adjusted to a modified Langmuir equation, and the enthalpy of the interaction was calculated to give endothermic values of 2.11 ± 0.28 and 2.50 ± 0.31 kJ mol−1 for these cations, respectively. The spontaneity of this ion-exchange process is reflected in negative Gibbs energy and with a contribution of positive entropic values. This set of thermodynamic data at the solid-liquid interface suggests a favorable ion-exchange process for this anchored biopolymer for cation exchange from the environment.  相似文献
