Degradation of dimethyl sulfide and methanethiol in slurries prepared from sediments of minerotrophic peatland ditches were studied under various conditions. Maximal aerobic dimethyl sulfide-degrading capacities (4.95 nmol per ml of sediment slurry · h−1), measured in bottles shaken under an air atmosphere, were 10-fold higher than the maximal anaerobic degrading capacities determined from bottles shaken under N2 or H2 atmosphere (0.37 and 0.32 nmol per ml of sediment slurry · h−1, respectively). Incubations under experimental conditions which mimic the in situ conditions (i.e., not shaken and with an air headspace), however, revealed that aerobic degradation of dimethyl sulfide and methanethiol in freshwater sediments is low due to oxygen limitation. Inhibition studies with bromoethanesulfonic acid and sodium tungstate demonstrated that the degradation of dimethyl sulfide and methanethiol in these incubations originated mainly from methanogenic activity. Prolonged incubation under a H2 atmosphere resulted in lower dimethyl sulfide degradation rates. Kinetic analysis of the data resulted in apparent Km values (6 to 8 μM) for aerobic dimethyl sulfide degradation which are comparable to those reported for Thiobacillus spp., Hyphomicrobium spp., and other methylotrophs. Apparent Km values determined for anaerobic degradation of dimethyl sulfide (3 to 8 μM) were of the same order of magnitude. The low apparent Km values obtained explain the low dimethyl sulfide and methanethiol concentrations in freshwater sediments that we reported previously. Our observations point to methanogenesis as the major mechanism of dimethyl sulfide and methanethiol consumption in freshwater sediments.  相似文献

Saccharum officinarum bagasse (common name: sugarcane bagasse) and Pennisetum purpureum (also known as Napier grass) are among the most promising feedstocks for bioethanol production in Argentina and Brazil. In this study, both biomasses were assessed before and after acid pretreatment and following hydrolysis with Nasutitermes aquilinus and Cortaritermes fulviceps termite gut digestome. The chemical composition analysis of the biomasses after diluted acid pretreatment showed that the hemicellulose fraction was partially removed. The (hemi) cellulolytic activities were evaluated in bacterial culture supernatants of termite gut homogenates grown in treated and untreated biomasses. In all cases, we detected significantly higher endoglucanase and xylanase activities using pretreated biomasses compared to untreated biomasses, carboxymethylcellulose and xylan. Several protein bands with (hemi) cellulolytic activity were detected in zymograms and two-dimensional gel electrophoresis. Some proteins of these bands or spots were identified as xylanolytic peptides by mass spectrometry. Finally, the diversity of cultured cellulolytic bacterial endosymbionts associated to both Argentinean native termite species was analyzed. This study describes, for the first time, bacterial endosymbionts and endogenous (hemi) cellulases of two Argentinean native termites as well as their potential application in degradation of lignocellulosic biomass for bioethanol production.  相似文献

Tissue morphogenesis comprises the self-organized creation of various patterns and shapes. Although detailed underlying mechanisms are still elusive in many cases, an increasing amount of experimental data suggests that chemical morphogen and mechanical processes are strongly coupled. Here, we develop and test a minimal model of the axis-defining step (i.e., symmetry breaking) in aggregates of the Hydra polyp. Based on previous findings, we combine osmotically driven shape oscillations with tissue mechanics and morphogen dynamics. We show that the model incorporating a simple feedback loop between morphogen patterning and tissue stretch reproduces a wide range of experimental data. Finally, we compare different hypothetical morphogen patterning mechanisms (Turing, tissue-curvature, and self-organized criticality). Our results suggest the experimental investigation of bigger (i.e., multiple head) aggregates as a key step for a deeper understanding of mechanochemical symmetry breaking in Hydra.  相似文献

Predicting bioproduction titers from microbial hosts has been challenging due to complex interactions between microbial regulatory networks, stress responses, and suboptimal cultivation conditions. This study integrated knowledge mining, feature extraction, genome-scale modeling (GSM), and machine learning (ML) to develop a model for predicting Yarrowia lipolytica chemical titers (i.e., organic acids, terpenoids, etc.). First, Y. lipolytica production data, including cultivation conditions, genetic engineering strategies, and product information, was manually collected from literature (~100 papers) and stored as either numerical (e.g., substrate concentrations) or categorical (e.g., bioreactor modes) variables. For each case recorded, central pathway fluxes were estimated using GSMs and flux balance analysis (FBA) to provide metabolic features. Second, a ML ensemble learner was trained to predict strain production titers. Accurate predictions on the test data were obtained for instances with production titers >1 g/L (R2 = 0.87). However, the model had reduced predictability for low performance strains (0.01–1 g/L, R2 = 0.29) potentially due to biosynthesis bottlenecks not captured in the features. Feature ranking indicated that the FBA fluxes, the number of enzyme steps, the substrate inputs, and thermodynamic barriers (i.e., Gibbs free energy of reaction) were the most influential factors. Third, the model was evaluated on other oleaginous yeasts and indicated there were conserved features for some hosts that can be potentially exploited by transfer learning. The platform was also designed to assist computational strain design tools (such as OptKnock) to screen genetic targets for improved microbial production in light of experimental conditions.  相似文献

Multivariate curve resolution with alternating least squares (MCR-ALS) was applied to voltammetric data obtained in the analysis of the competitive binding of glutathione (GSH) and phytochelatins [(γGlu-Cys)n-Gly, PCn, n = 2-5] by Cd2+. The displacements between ligands and chain length influence on the competitive binding of PCn toward Cd2+ were investigated. The analysis of the resulting pure voltammograms and concentration profiles of the resolved components suggests that ligands containing more thiol groups are able to displace the shortest chain ligands from their metal complexes, whereas the opposite does not happen. However, when the length of the chain surpasses that of PC3, the binding capacity of the molecule still increases (i.e., it can bind more metal ions), but the position and shape of the voltammetric signals practically rest unchanged. This suggests that at this level, the stability of metal binding could depend more on the nature of the binding sites separately than on the quantity of the sites (i.e., the chain length).  相似文献

Monitoring the biological processes and microbial diversity is essential for sustaining the soil health for long-term productivity. In the present study, the impact of long-term nutrient management systems on changes in Azotobacter diversity of Indian semi-arid alfisol was assessed. Three soils, i.e., unfertilized control, soils amended with organic manures (OM), and with inorganic chemical fertilizers (IC) from century-old experimental fields were evaluated for Azotobacter diversity by Amplified Ribosomal DNA Restriction Analysis (ARDRA). Bray–Curtis’s similarity index of the ARDRA data of the isolates was analyzed by non-metric multi-dimensional scaling and hierarchical cluster analysis. The results revealed that the long-term organically managed soil recorded significantly higher soil organic carbon, microbial biomass carbon, and total culturable bacterial counts, whereas the chemical fertilized and control soils remained unaffected. Though the Azotobacter population was significantly higher in OM soil than IC and control soils, the genetic diversity was unaffected due to long-term addition of either organic manures or inorganic chemical fertilizers. This result implies the importance of continuous addition of organic manures and also the optimal use of inorganic chemical fertilizers without disturbing the biological properties of the soil.  相似文献

The degree of biodegradation of low-density polyethylene (LDPE) films modified with Bionolle® polyester in different soils under laboratory conditions was evaluated. Films were incubated in soils from waste coal, a forest and an extinct volcano crater. Prior to degradation studies, soils underwent chemical and microbiological analysis. Film weight loss and mechanical properties, as well as the surface of the polymeric samples determined via scanning electron microscopy, were evaluated after 75, 150 and 225 days of biodegradation. Important chemical changes in the polymeric chains were detected by Fourier Transform Infrared Spectroscopy (FTIR). Fungal and bacterial species that were able to grow on the film surfaces were monitored in order to see whether the films were easily colonised by autochthonous microorganisms (i.e., typical to each soil). Identification of microorganisms was based on their cellular fatty acid methyl ester (FAME) profiles. Biodegradation of modified polyethylene films in soils led to significant changes (i.e., elongation at brake of 98%) in their mechanical properties that were caused by biochemical modifications of both polyester and polyethylene. Compared to waste coal soil, films underwent rapid biodegradation in soils that were rich in organic matter. Bacteria belonging to the genus, Bacillus, and the fungi, Gliocladium viride, Aspergillus awamori and Mortierella subtilissima, were easily able to colonise both polyethylene and polyethylene modified with Bionolle®.  相似文献

The chemical compositions of plant fossils having trimerophyte (Pertica,Psilophyton princeps, P. cf.jorbesii), rhyniophyte (Taeniocrada), and zosterophyll (Sawdonia) morphological characteristics are chemically analyzed and chemotaxonomically related to vascular ( ?Eohostimella, Renalia, Chahuria) and putative non-vascular plant fossils (Botryococcus, Parka, Pachytheca, Prototaxites,Nematothallus, Spongiophyton, Protosalvinia, Orestovia) whose taxonomic affinities are unknown or speculative. Separation of the material examined into clusters representing higher taxa (i.e., algal, nonalgal, trimerophyte, rhyniophyte, zosterophyll plant groupings) is effected by the weighting of chemical data during cluster analyses. The weighting of phenolic and monohydroxycarboxylic acid constituents is shown to cluster vascular plant material, while the criteria of carbon chainlength ranges and maxima separate vascular from non-vascular plant fossils. Multivariate analysis of the data, using chemical and geological factors, results in the clustering of four groups: (1)Botryococcus, Parka, Pachytheca, (2)Spongiophyton, Prototaxites, Nematothallus, Orestovia, (3)Eohostimella,Taeniocrada, Renalia, and (4)Psilophyton spp.,Pertica, Chaleuria. Sawdonia andProtosalvinia appear as data points showing no observable affinity with any of the above fossils.Protosalvinia, Renalia, andChaleuria are interpreted as being chemotaxonomically intermediate. These data are interpreted as indicating taxonomic affinities on a very broad scale ; possible evolutionary trends in specific chemical compounds, as they relate to vascular and non-vascular plant geochemistry, are discussed.  相似文献

Studies of the genetic structure of the internal noncoding transcribed spacer region 2 (ITS2) of the ribosomal DNA of five entomophage species of the genus Trichogramma, i.e., T. pintoi Voeg., T. evanescens Westw., T. dendrolimi Mats., T. cacoeciae Meyer, and T. semblidis Auriv., are performed. A PCR analysis with subsequent determination of the nucleotide sequence of the ITS2 regions made it possible to identify essential inter-species differences. The data may be used for the identification of the species T. pintoi, T. dendrolini, and T. semblidis.  相似文献

Heterozygosity-fitness correlations (HFCs) provide insights into the genetic bases of individual fitness variation in natural populations. However, despite decades of study, the biological significance of HFCs is still under debate. In this study, we investigated HFCs in a large population of the sexually dimorphic lizard Takydromus viridipunctatus (Lacertidae). Because of the high prevalence of parasitism from trombiculid mites in this lizard, we expect individual fitness (i.e., survival) to decrease with increasing parasite load. Furthermore, because morphological asymmetry is likely to influence individuals'' mobility (i.e., limb asymmetry) and male biting ability during copulation (i.e., head asymmetry) in this species, we also hypothesize that individual fitness should decrease with increasing morphological asymmetry. Although we did not formally test the relationship between morphological asymmetry and fitness in this lizard, we demonstrated that survival decreased with increasing parasite load using a capture-mark-recapture data set. We used a separate sample of 140 lizards to test the correlations between individual heterozygosity (i.e., standardized mean d2 and HL based on 10 microsatellite loci) and the two fitness traits (i.e., parasite load and morphological asymmetry). We also evaluated and excluded the possibility that single-locus effects produced spurious HFCs. Our results suggest male-only, negative correlations between individual heterozygosity and parasite load and between individual heterozygosity and asymmetry, suggesting sex-specific, positive HFCs. Male T. viridipunctatus with higher heterozygosity tend to have lower parasite loads (i.e., higher survival) and lower asymmetry, providing a rare example of HFC in reptiles.  相似文献

A shallow-water hydrothermal vent system in the Aegean Sea close to the island of Milos (Greece) was chosen to study the diversity and distribution of the chemolithoautotrophic sulfur-oxidizing bacterium Thiomicrospira. Cell numbers in samples from different regions around a solitary vent decreased toward the center of the vent (horizontal distribution), as well as with depth (vertical distribution), corresponding to an increase in temperature (from ca. 25 to 60°C) and a decrease in pH (from ca. pH 7 to 5). Thiomicrospira was one of the most abundant culturable sulfur oxidizers and was even dominant in one region. Phylogenetic analysis of Thiomicrospira spp. present in the highest most-probable-number (MPN) dilutions revealed that most of the obtained sequences grouped in two new closely related clusters within the Thiomicrospira branch. Two different new isolates, i.e., Milos-T1 and Milos-T2, were obtained from high-dilution (10−5) enrichments. Phylogenetic analysis indicated that isolate Milos-T1 is related to the recently described Thiomicrospira kuenenii and Hydrogenovibrio marinus, whereas isolate Milos-T2 grouped with the MPN sequences of cluster 2. The predominance of strain Milos-T2 was indicated by its identification in several environmental samples by hybridization analysis of denaturing gradient gel electrophoresis (DGGE) patterns and by sequencing of one of the corresponding bands, i.e., ML-1, from the DGGE gel. The results shown in this paper support earlier indications that Thiomicrospira species are important members of hydrothermal vent communities.  相似文献

The Synthesis of Cytochrome Oxidase protein, or SCO protein, is required for the assembly of cytochrome c oxidase in many mitochondrial and bacterial respiratory chains. SCOs have been proposed to deliver copper to the CuA site of cytochrome c oxidase. We have reported that Bacillus subtilis SCO (i.e., BsSCO) binds Cu(II) with high-affinity via a two-step process mediated by three conserved residues (i.e., two cysteines and one histidine, or the CCH motif). A remarkable feature in the reaction of reduced (i.e., di-thiol) BsSCO with copper is that it does not generate any of the disulfide form of BsSCO. This molecular aversion is proposed to be a consequence of a binding mechanism in which the initial copper complex of BsSCO does not involve cysteine, but instead involves nitrogen ligands. We test this proposal here by constructing two isomers of BsSCO in which the conserved copper binding residues (i.e., the CCH-motif) are retained, but their positions are altered. In these variants the two cysteines are exchanged with histidine, and both react transiently with copper (II) with distinct kinetic profiles. The reaction generates Cu(I) and the protein is oxidized to its disulfide form. EPR analysis supports a copper binding model in which cysteine, which is at the “histidine position” in the mutant, is part of an initial encounter complex with copper. When cysteine is the initial ligating residue an oxidation reaction ensues. In contrast initial binding to native BsSCO uses nitrogen-based ligands, and thereby avoids the opportunity for thiol oxidation.  相似文献

Mate preference research often focuses on traits that indicate a romantic partner's personal worth (e.g., their physical attractiveness, resource potential) rather than their tendency to leverage that worth for mutual vs. zero-sum benefit (i.e., their trustworthiness). No one has assessed the contribution of trustworthiness to perceived mate value relative to other personality dimensions. Here we examined the desirability of a partner's trustworthiness relative to five other personality indicators of mate quality during initial partner selection. Participants (n = 918) ranked multivariate partner profiles constructed from the HEXACO model of personality (i.e., honesty-humility, emotionality, extraversion, agreeableness, conscientiousness, and openness to experience) and provided partner ratings for each trait. Using conjoint analysis, we found that honesty-humility influenced participants' ranking decisions substantially more than each other characteristic (all Cohen's ds > 0.62). This was true for both long- (i.e., committed) and short-term (i.e., purely sexual) partner evaluations, though honesty-humility was relatively more important for long- vs. short-term contexts. There were no sex differences. A different pattern, including sex differences, emerged for partner ratings. Based on these findings, we hypothesize that the challenge of avoiding romantic interpersonal predation may have been a relatively stronger selection pressure during the evolution of human mate preference than has the challenge of identifying other valuable partner traits.  相似文献

The effects of local nutrient loading, harvesting, regional temperature patterns and weather induced light conditions were assessed on the interannual dynamics of the commercially important loose-lying red algal community of Furcellaria lumbricalis and Coccotylus truncatus. Higher algal biomasses were due to higher temperatures and number of calm days (i.e. clear water phases) during the active growing season in spring and summer. In addition, higher biomasses were associated with a higher number of stormy days in November-February, which is linked to a reduced ice cover and longer growing season. Commercial harvesting and nutrient load did not appear to drive the interannual dynamics of these red algae. Thus, the dynamics of the loose-lying red algal community is constrained by regional rather than local environmental factors.  相似文献

Background

The analysis of co-authorship network aims at exploring the impact of network structure on the outcome of scientific collaborations and research publications. However, little is known about what network properties are associated with authors who have increased number of joint publications and are being cited highly.

Methodology/Principal Findings

Measures of social network analysis, for example network centrality and tie strength, have been utilized extensively in current co-authorship literature to explore different behavioural patterns of co-authorship networks. Using three SNA measures (i.e., degree centrality, closeness centrality and betweenness centrality), we explore scientific collaboration networks to understand factors influencing performance (i.e., citation count) and formation (tie strength between authors) of such networks. A citation count is the number of times an article is cited by other articles. We use co-authorship dataset of the research field of ‘steel structure’ for the year 2005 to 2009. To measure the strength of scientific collaboration between two authors, we consider the number of articles co-authored by them. In this study, we examine how citation count of a scientific publication is influenced by different centrality measures of its co-author(s) in a co-authorship network. We further analyze the impact of the network positions of authors on the strength of their scientific collaborations. We use both correlation and regression methods for data analysis leading to statistical validation. We identify that citation count of a research article is positively correlated with the degree centrality and betweenness centrality values of its co-author(s). Also, we reveal that degree centrality and betweenness centrality values of authors in a co-authorship network are positively correlated with the strength of their scientific collaborations.

Conclusions/Significance

Authors’ network positions in co-authorship networks influence the performance (i.e., citation count) and formation (i.e., tie strength) of scientific collaborations.  相似文献

Roseobacter/Rhodobacter and SAR11, affiliated with Alphaproteobacteria, and the phylum Bacteroidetes constitute a large proportion of marine planktonic bacteria, but information about their growth and distribution patterns in the Southern Ocean is scarce. The aim of the present study is to determine patterns in the biomass and productivity of Roseobacter/Rhodobacter, SAR11, and Bacteroidetes groups along the steep temperature, salinity, and organic matter gradients in the Southern Ocean by using catalyzed reporter deposition-fluorescence in situ hybridization and bromodeoxyuridine (BrdU) immunocytochemistry FISH. We found that Roseobacter/Rhodobacter, SAR11, and Bacteroidetes are prominent contributors to total bacterial biomass and production. SAR11 bacteria were the predominant lineage, but their biomass was low in the coldest regions. In contrast, the biomasses of Roseobacter/Rhodobacter and Bacteroidetes lineages were positively correlated with organic matter concentrations. The Roseobacter/Rhodobacter had the highest proportion of BrdU-positive (i.e., actively growing) cells among the three phylotypes at all stations, despite their low abundance. The relative contribution of Bacteroidetes to the total bacterial productivity (number of active cells) was negatively correlated with temperature. These results suggest that the growth and distribution patterns of Roseobacter/Rhodobacter, SAR11, and Bacteroidetes were determined by different environmental gradients (e.g., organic matter concentrations or temperature) in the Southern Ocean.  相似文献

The synthesis, chemical characterization and functional evaluation are reported for dichloro(6-aminoethylaminopurine)platinum(II) and dichloro(6-hydroxyethylaminopurine)platinum(II) and dichloro(6-hydroxyethylamethylaminopurine)platinum(II) (i.e. Pt(6-AEAP), Pt(6-HEAP) and Pt(6-MHEAP) new complexes of platinum(II). Certain reaction conditions favored the formation of the tripurine platinum complex, but the monopurine complex could be obtained either by hydrolysis of the tripurine or by reacting at reduced temperature and concentration. Although neither compound was as effective as cis-diamminedichloroplatinum(II) (i.e. DDP) at reducing tumor cell viability or proliferation, both were associated with much less renal toxicity than DDP in the mouse kidney (i.e. Pt(6-AEAP):~20 × less; Pt(6-MHEAP): ~100 × less).  相似文献

The aim of the present study was to assess the population structure and admixture levels in the Vrindavani composite population in India by using Bovine50KSNP BeadChip data. Genotypic data were generated for randomly selected animals (n = 72) of Vrindavani population and the data for parental breeds i.e., Hariana (n = 10), Holstein-Friesian (n = 63), Jersey (n = 28) and Brown Swiss (n = 22) were retrieved from a public repository. The indices of population structure were calculated using PLINK software and R-program. The merged dataset was analysed for assessing admixture levels and population stratification using three different approaches i.e., principal component analysis (PCA), multi-dimensional scaling (MDS) approach and the model-based approach in STRUCTURE software. The average minor allele frequency (MAF) value for Vrindavani population was estimated to be 0.235. Vrindavani population was found to possess an average ancestry of 39.5, 22.9, 26.9, and 10.7% inheritance levels from Holstein Friesian, Jersey, Hariana and Brown Swiss cattle breeds, respectively.  相似文献

The chemical composition of the essential oils from Mentha piperita L., Mentha crispa L., Origanum vulgare L., Dracocephalum moldavica L., and Hyssopus officinalis L. cultivated under the conditions of the Western Siberia Region was studied using the chromatography-mass spectrometry approach. The results on the composition of the cultivated O. vulgare, D. moldavica, and H. officinalis are comparable with the data for wild plants, and for M. piperita and M. crispa, with the plants grown in the Southern regions of Russia. The data on the chemical composition of the essence oil, or attar, from M. crispa (Inna breed) were obtained for the first time, and the data for H. officinalis were substantially extended.  相似文献

With the availability of public databases that store compound-target/compound-toxicity information, and Traditional Chinese medicine (TCM) databases, in silico approaches are used in toxicity studies of TCM herbal medicine. Here, three in silico approaches for toxicity studies were reviewed, which include machine learning, network toxicology and molecular docking. For each method, its application and implementation e.g., single classifier vs. multiple classifier, single compound vs. multiple compounds, validation vs. screening, were explored. While these methods provide data-driven toxicity prediction that is validated in vitro and/or in vivo, it is still limited to single compound analysis. In addition, these methods are limited to several types of toxicity, with hepatotoxicity being the most dominant. Future studies involving the testing of combination of compounds on the front end i.e., to generate data for in silico modeling, and back end i.e., validate findings from prediction models will advance the in silico toxicity modeling of TCM compounds.  相似文献
