Two Gram-staining-negative, moderately halophilic bacteria, strains M1-18T and L1-16, were isolated from a saltern located in Huelva (Spain). They were motile, strictly aerobic rods, growing in the presence of 3–25% (w/v) NaCl (optimal growth at 7.5–10% [w/v] NaCl), between pH 4.0 and 9.0 (optimal at pH 6.0–7.0) and at temperatures between 15 and 40 °C (optimal at 37 °C). Phylogenetic analysis based on 16S rRNA gene sequence comparison showed that both strains showed the higher similarity values with Chromohalobacter israelensis ATCC 43985T (95.2–94.8%) and Chromohalobacter salexigens DSM 3043T (95.0–94.9%), and similarity values lower than 94.6% with other species of the genera Chromohalobacter, Kushneria, Cobetia or Halomonas. Multilocus sequence analysis (MLSA) based on the partial sequences of atpA, rpoD and secA housekeeping genes indicated that the new isolates formed an independent and monophyletic branch that was related to the peripheral genera of the family Halomonadaceae, Halotalea, Carnimonas and Zymobacter, supporting their placement as a new genus of the Halomonadaceae. The DNA–DNA hybridization between both strains was 82%, whereas the values between strain M1-18T and the most closely related species of Chromohalobacter and Kushneria were equal or lower to 48%. The major cellular fatty acids were C18:1ω7c/C18:1ω6c, C16:0, and C16:1ω7c/C16:1ω6c, a profile that differentiate this new taxon from species of the related genera. We propose the placement of both strains as a novel genus and species, within the family Halomonadaceae, with the name Larsenia salina gen. nov., sp. nov. In this study the pulp from Solanum lycocarpum fruits was used as raw material for extraction of starch, resulting in a yield of 51%. The starch granules were heterogeneous in size, presenting a conical appearance, very similar to a high-amylose cassava starch. The elemental analysis (CHNS) revealed 64.33% carbon, 7.16% hydrogen and 0.80% nitrogen. FT-IR spectroscopy showed characteristic peaks of polysaccharides and NMR analysis confirmed the presence of the α-anomer of d-glucose. The S. lycocarpum starch was characterized by high value of intrinsic viscosity (3515 mPa s) and estimated molecular weight around 645.69 kDa. Furthermore, this starch was classified as a B-type and high amylose content starch, presenting 34.66% of amylose and 38% crystallinity. Endothermic transition temperatures (To = 61.25 °C, Tp = 64.5 °C, Tc = 67.5 °C), gelatinization temperature (ΔT = 6.3 °C) ranges and enthalpy changes (ΔH = 13.21 J g−1) were accessed by DCS analysis. These results make the S. lycocarpum fruit a very promising source of starch for biotechnological applications.  相似文献

Final temperature preferendum of white shrimp adults were determined with acute and gravitation methods. The final preferendum was similar, independent of method (26.2–25.6 °C). A direct relationship was determined between the critical thermal maxima values and the acclimation temperatures (P<0.05). The end point of Critical Thermal Maxima (CTMax) for adults was defined as the loss of righting response (LRR). The acclimation response ratio (ARR) for adults of white shrimp had an interval of 0.36–0.76, values that agreed with others obtained for crustaceans from tropical and subtropical climates. The oxygen consumption rates increased significantly (P<0.05) from 39.6 up to 90.0 mg O2 kg−1 h−1 wet weight (w.w.) as the acclimation temperature increased from 20 to 32 °C. The range of temperature coefficient (Q10) of the white shrimp between 23 and 26 °C was the lower 1.60. The results obtained in this work are discussed in relation to the species importance in the reproductive scope and maintenance of breeders.  相似文献

The ruthenium complexes, trans-[Ru(phen-NH-phen)(eina)2](PF6)2 and trans-[Ru(phen-NH-phen)(ina)2](PF6)2 where phen-NH-phen = N,N-bis(1,10-phenanthroline-2-yl)amine, ina = isonicotinic acid and eina = ethyl isonicotinate, have been synthesized and characterized by 1H NMR, elemental analysis, and IR spectroscopy. The compounds were non-emissive at room temperature, but displayed intense photoluminescence in 4:1 ethanol/methanol glasses at 77 K with corrected emission maximum at 570-580 nm. A quasi-reversible wave observed in cyclic voltammetry experiments was assigned to the RuIII/II couple, (trans-[Ru(phen-NH-phen)(eina)2)3+/2+ = +1.22 V versus Ag/AgCl. The trans-[Ru(phen-NH-phen)(ina)2](PF6)2 compound was found to bind to nanocrystalline TiO2 thin films from acetonitrile solution. Pulsed 532 nm excitation of trans-[Ru(phen-NH-phen)(ina)2](PF6)2 anchored to mesoporous nanocrystalline TiO2 thin films resulted in an absorption difference spectra consistent with the formation of an interfacial charge separated state trans-[RuIII (phen-NH-phen)(ina)2]+/TiO2 (e). The formation of this state could not be time resolved, consistent with rapid excited state injection into the TiO2, kinj > 108 s−1. Comparative measurements with a thin film actinometer yielded an injection quantum yield (?inj) of 0.8. Charge recombination required milliseconds for completion and followed a bi-second-order equal concentration kinetic model with k1 = 1.0 × 108 s−1, and k2 = 3.0 × 105 s−1. In regenerative solar cells with 0.5 M LiI and 0.005 M I2 in acetonitrile, incident photon-to-current efficiencies were typically less than 10%.  相似文献

Industrial wastewater treatment comprises several processes to fulfill the discharge permits or to enable the reuse of wastewater. For tannery wastewater, constructed wetlands (CWs) may be an interesting treatment option. Two-stage series of horizontal subsurface flow CWs with Phragmites australis (UP series) and Typha latifolia (UT series) provided high removal of organics from tannery wastewater, up to 88% of biochemical oxygen demand (BOD5) (from an inlet of 420 to 1000 mg L−1) and 92% of chemical oxygen demand (COD) (from an inlet of 808 to 2449 mg L−1), and of other contaminants, such as nitrogen, operating at hydraulic retention times of 2, 5 and 7 days. No significant (P < 0.05) differences in performance were found between both the series. Overall mass removals of up to 1294 kg COD ha−1 d−1 and 529 kg BOD5 ha−1 d−1 were achieved for a loading ranging from 242 to 1925 kg COD ha−1 d−1 and from 126 to 900 kg BOD5 ha−1 d−1. Plants were resilient to the conditions imposed, however P. australis exceeded T. latifolia in terms of propagation.  相似文献

Basal metabolic rate (BMR) is thought to be a major hub in the network of physiological mechanisms connecting life history traits. Evaporative water loss (EWL) is a physiological indicator that is widely used to measure water relations in inter- or intraspecific studies of birds in different environments. In this study, we examined the physiological responses of summer-acclimatized Hwamei Garrulax canorus to temperature by measuring their body temperature (Tb), metabolic rate (MR) and EWL at ambient temperatures (Ta) between 5 and 40 °C. Overall, we found that mean body temperature was 42.4 °C and average minimum thermal conductance (C) was 0.15 ml O2 g−1 h−1 °C−1 measured between 5 and 20 °C. The thermal neutral zone (TNZ) was 31.8–35.3 °C and BMR was 181.83 ml O2 h−1. Below the lower critical temperature, MR increased linearly with decreasing Ta according to the relationship: MR (ml O2 h−1)=266.59–2.66 Ta. At Tas above the upper critical temperature, MR increased with Ta according to the relationship: MR (ml O2 h−1)=−271.26+12.85 Ta. EWL increased with Ta according to the relationship: EWL (mg H2O h−1)=−19.16+12.64 Ta and exceeded metabolic water production at Ta>14.0 °C. The high Tb and thermal conductance, low BMR, narrow TNZ, and high evaporative water production/metabolic water production (EWP/MWP) ratio in the Hwamei are consistent with the idea that this species is adapted to warm, mesic climates, where metabolic thermogenesis and water conservation are not strong selective pressures.  相似文献

Fucogalactans from edible Agaricus bisporus (RFP-Ab) and wild Lactarius rufus (RFP-Lr) mushrooms were obtained on aqueous extraction followed by purification. RFP-Ab had Mw 43.8 × 104 g mol−1 and RFP-Lr Mw 1.4 × 104 g mol−1. RFP-Lr had a (1 → 6)-linked α-d-Galp main-chain partially substituted at O-2 by nonreducing end-units of α-l-Fucp (29%). While RFP-Ab had a similar main chain, it was partially substituted at O-2 by nonreducing end-units of α-l-Fucp (2.8%) and β-d-Galp (14.5%), and partially methylated at HO-3. Both RFP-Lr and RFP-Ab were tested in mice against polymicrobial sepsis. Lethality rate, myeloperoxidase (MPO) activity and cytokine levels were determined. It was observed a reduction in late mortality rate by 62.5% and 50%, respectively, prevention of neutrophil accumulation in ileum and decreasing in TNF-α and IL-1β serum levels.  相似文献

This study reports temperature effects on paralarvae from a benthic octopus species, Octopus huttoni, found throughout New Zealand and temperate Australia. We quantified the thermal tolerance, thermal preference and temperature-dependent respiration rates in 1-5 days old paralarvae. Thermal stress (1 °C increase h−1) and thermal selection (∼10-24 °C vertical gradient) experiments were conducted with paralarvae reared for 4 days at 16 °C. In addition, measurement of oxygen consumption at 10, 15, 20 and 25 °C was made for paralarvae aged 1, 4 and 5 days using microrespirometry. Onset of spasms, rigour (CTmax) and mortality (upper lethal limit) occurred for 50% of experimental animals at, respectively, 26.0±0.2 °C, 27.8±0.2 °C and 31.4±0.1 °C. The upper, 23.1±0.2 °C, and lower, 15.0±1.7 °C, temperatures actively avoided by paralarvae correspond with the temperature range over which normal behaviours were observed in the thermal stress experiments. Over the temperature range of 10 °C-25 °C, respiration rates, standardized for an individual larva, increased with age, from 54.0 to 165.2 nmol larvae−1 h−1 in one-day old larvae to 40.1-99.4 nmol h−1 at five days. Older larvae showed a lesser response to increased temperature: the effect of increasing temperature from 20 to 25 °C (Q10) on 5 days old larvae (Q10=1.35) was lower when compared with the 1 day old larvae (Q10=1.68). The lower Q10 in older larvae may reflect age-related changes in metabolic processes or a greater scope of older larvae to respond to thermal stress such as by reducing activity. Collectively, our data indicate that temperatures >25 °C may be a critical temperature. Further studies on the population-level variation in thermal tolerance in this species are warranted to predict how continued increases in ocean temperature will limit O. huttoni at early larval stages across the range of this species.  相似文献

An Enterobacter strain (GY-1) with high activity of decolorization of Reactive Black 5 (RB 5) was isolated from textile wastewater treating sludge. The kinetic characteristics of dye decolorization by the strain GY-1 were determined quantitatively using the diazo dye, RB 5. Effects of different operation parameters (inoculum size, pH, temperature and salinity) and various electron donors on decolorization of the azo dye by GY-1 were systematically investigated to reveal the primary factors that determine the performance of the azo dye decolorization. The decolorization of RB 5 was attributed to extracellular enzymes. A kinetic model was established giving the dependence of decolorization rate on cell mass concentration (first order). Decolorization rate increased with increasing temperature from 20 to 35 °C, which can be predicted by Arrhenius equation with the activation energy (Ea) of 8.50 kcal mol−1 and the frequency factor (A0) of 6.28 × 107 mg l g MLSS−1 h−1. Michaelis-Menten kinetics and Eadie-Hofstee plot were used to determine Vmax, 1.05 mg l−1 h−1 and Km, 24.06 mg l−1.  相似文献

Recombinant β-galactosidase from Lactobacillus plantarum WCFS1, homologously over-expressed in L. plantarum, was purified to apparent homogeneity using p-aminobenzyl 1-thio-β-d-galactopyranoside affinity chromatography and subsequently characterized. The enzyme is a heterodimer of the LacLM-family type, consisting of a small subunit of 35 kDa and a large subunit of 72 kDa. The optimum pH for hydrolysis of its preferred substrates o-nitrophenyl-β-d-galactopyranoside (oNPG) and lactose is 7.5 and 7.0, and optimum temperature for these reactions is 55 and 60 °C, respectively. The enzyme is most stable in the pH range of 6.5-8.0. The Km, kcat and kcat/Km values for oNPG and lactose are 0.9 mM, 92 s−1, 130 mM−1 s−1 and 29 mM, 98 s−1, 3.3 mM−1 s−1, respectively. The L. plantarum β-galactosidase possesses a high transgalactosylation activity and was used for the synthesis of prebiotic galacto-oligosaccharides (GOS). The resulting GOS mixture was analyzed in detail, and major components were identified by using high performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD) as well as capillary electrophoresis. The maximal GOS yield was 41% (w/w) of total sugars at 85% lactose conversion (600 mM initial lactose concentration). The enzyme showed a strong preference for the formation of β-(1→6) linkages in its transgalactosylation mode, while β-(1→3)-linked products were formed to a lesser extent, comprising ∼80% and 9%, respectively, of the newly formed glycosidic linkages in the oligosaccharide mixture at maximum GOS formation. The main individual products formed were β-d-Galp-(1→6)-d-Lac, accounting for 34% of total GOS, and β-d-Galp-(1→6)-d-Glc, making up 29% of total GOS.  相似文献

We report for the first time on the comparative use of pulsed-plasma gas-discharge (PPGD) and pulsed UV light (PUV) for the novel destruction of the waterborne enteroparasite Cryptosporidium parvum. It also describes the first cyto-, geno- and ecotoxicological assays undertaken to assess the safety of water decontaminated using PPGD and PUV. During PPGD treatments, the application of high voltage pulses (16 kV, 10 pps) to gas-injected water (N2 or O2, flow rate 2.5 L/min) resulted in the formation of a plasma that generated free radicals, ultraviolet light, acoustic shock waves and electric fields that killed ca. 4 log C. parvum oocysts in 32 min exposure. Findings showed that PPGD-treated water produced significant cytotoxic properties (as determined by MTT and neutral red assays), genotoxic properties (as determined by comet and Ames assays), and ecotoxic properties (as determined by Microtox™, Thamnotox™ and Daphnotox™ assays) that are representative of different trophic levels in aquatic environment (p < 0.05). Depending in part on the type of injected gas used, PPGD-treated water became either alkaline (pH ≤ 8.58, using O2) or acidic (pH ≥ 3.21, using N2) and contained varying levels of reactive free radicals such as ozone (0.8 mg/L) and/or dissociated nitric and nitrous acid that contributed to the observed disinfection and toxicity. Chemical analysis of PPGD-treated water revealed increasing levels of electrode metals that were present at ≤ 30 times the tolerated respective values for EU drinking water. PUV-treated water did not exhibit any toxicity and was shown to be far superior to that of PPGD for killing C. parvum oocysts taking only 90 s of pulsing [UV dose of 6.29 μJ/cm2] to produce a 4-log reduction compared to a similar reduction level achieved after 32 min PPGD treatment as determined by combined in vitro CaCo-2 cell culture-qPCR.  相似文献

Fluxes of oxygen, inorganic nitrogen (DIN) and denitrification (isotope pairing) were measured from January 1997 to February 1998 via intact cores incubation in a shallow brackish area within the eutrophic Valli di Comacchio (northern Adriatic coast, Italy). Rates were measured in the light and in the dark in sediments colonized by the rooted macrophyte Ruppia cirrhosa and in adjacent sediments with benthic microalgae. Ruppia biomass (25-414 g DW m− 2) exhibited a seasonal evolution whilst that of microphytobenthos (12-66 mg chl a m− 2) was more erratic. Net (NP) and gross (GP) primary productivity was 1.15 and 6.89 mol C m− 2y− 1 for bare and 25.4 and 51.7 mol C m− 2y− 1 for Ruppia vegetated sediments. Nitrogen pools in Ruppia standing stock varied from 43.6 to 631.4 (annual average 201.2) mmol N m− 2; the macrophyte N content was correlated with DIN concentration in the water column. Estimated N pool in microphytobenthos was one order of magnitude lower (from 2.4 to 14.5 mmol N m− 2, annual average 7.2). Theoretical DIN assimilation calculated from NP was 127.8 and 1112.6 mmol N m− 2y− 1 whilst that calculated from GP was 765 and 2282 mmol N m− 2y− 1 for microphytobenthos and Ruppia respectively. Measured annual fluxes of DIN were 974.6 and − 577 mmol N m− 2y− 1 in bare and Ruppia vegetated sediments meaning that the two sites were a source and sink for DIN and that from 25 to 50% of Ruppia annual DIN requirements came from the water column. During the period of this study total denitrification was lower in the macrophyte colonized (92.3 mmol N m− 2y− 1) compared to bare sediments (163.3 mmol N m− 2y− 1) as a probable consequence of higher competition between denitrifiers and phanerogams. At both sites the ratio between denitrification of water column nitrate (DW) and denitrification coupled to nitrification (DN) was >1.6 due to little oxygen penetration in reducing sediments (< 1.2 mm) and scarce nitrification activity. DW (0-35 µmol N m− 2h− 1) was significantly correlated with water column NO3−  (2-16 µM). Theoretical DIN assimilation to denitrification ratio varied from 12.0 to 24.8 for Ruppia vegetated and from 0.8 to 4.7 for unvegetated sediments.At Valle Smarlacca, Ruppia may influence nitrogen cycling by incorporating large DIN pools in biomass which is scattered in surrounding areas and fuels intense bacterial activity. With increasing anthropogenic nutrient input and insignificant organic matter export in the open sea the already severe eutrophic conditions are enhanced and may accelerate the decline of the macrophyte meadow.  相似文献

The ultrafast (< 100 fs) conversion of delocalized exciton into charge-separated state between the primary donor P700 (bleaching at 705 nm) and the primary acceptor A0 (bleaching at 690 nm) in photosystem I (PS I) complexes from Synechocystis sp. PCC 6803 was observed. The data were obtained by application of pump-probe technique with 20-fs low-energy pump pulses centered at 720 nm. The earliest absorbance changes (close to zero delay) with a bleaching at 690 nm are similar to the product of the absorption spectrum of PS I complex and the laser pulse spectrum, which represents the efficiency spectrum of the light absorption by PS I upon femtosecond excitation centered at 720 nm. During the first ∼ 60 fs the energy transfer from the chlorophyll (Chl) species bleaching at 690 nm to the Chl bleaching at 705 nm occurs, resulting in almost equal bleaching of the two forms with the formation of delocalized exciton between 690-nm and 705-nm Chls. Within the next ∼ 40 fs the formation of a new broad band centered at ∼ 660 nm (attributed to the appearance of Chl anion radical) is observed. This band decays with time constant simultaneously with an electron transfer to A1 (phylloquinone). The subtraction of kinetic difference absorption spectra of the closed (state P700+A0A1) PS I reaction center (RC) from that of the open (state P700A0A1) RC reveals the pure spectrum of the P700+A0 ion-radical pair. The experimental data were analyzed using a simple kinetic scheme: An* [(PA0)*A1 P+A0A1] P+A0A1, and a global fitting procedure based on the singular value decomposition analysis. The calculated kinetics of transitions between intermediate states and their spectra were similar to the kinetics recorded at 694 and 705 nm and the experimental spectra obtained by subtraction of the spectra of closed RCs from the spectra of open RCs. As a result, we found that the main events in RCs of PS I under our experimental conditions include very fast (< 100 fs) charge separation with the formation of the P700+A0A1 state in approximately one half of the RCs, the ∼ 5-ps energy transfer from antenna Chl* to P700A0A1 in the remaining RCs, and ∼ 25-ps formation of the secondary radical pair P700+A0A1.  相似文献

Myriophyllum spicatum and Potamogeton crispus are common species of shallow eutrophic lakes in north-eastern Germany, where a slow recovery of the submersed aquatic vegetation was observed. Thus, the characterisation of the root oxygen release (ROL) as well as its implication for geochemical processes in the sediment are of particular interest. A combination of microelectrode measurements, methylene blue agar and a titanium(III) redox buffer was used to investigate the influence of the oxygen content in the water column on ROL, diel ROL dynamics as well as the impact of sediment milieu. Oxygen gradients around the roots revealed a maximum oxygen diffusion zone of up to 250 μm. During a sequence with a light/dark cycle as well as alternating aeration of the water column, maximum ROL with up to 35% oxygen saturation at the root surface occurred under light/O2-saturated conditions. A decrease to about 30% was observed under dark/O2-saturated conditions, no ROL was detected at dark/O2-depleted conditions and only a weak ROL with 5–10% oxygen saturation at the root surface was measured under light but O2-depleted water column. These results indicate, that during darkness, ROL is supplied by oxygen from the water column and even during illumination and active photosynthesis production, ROL is modified by the oxygen content in the water column. Visualisation of ROL patterns revealed an enhanced ROL for plants which were grown in sulfidic littoral sediment in comparison to plants grown in pure quartz sand. For both plant species grown in sulfidic littoral sediment, a ROL rate of 3–4 μmol O2 h−1 plant−1 was determined with the Ti(III) redox buffer. For plants grown in pure quartz sand, the ROL rate decreased to 1–2 μmol O2 h−1 plant−1. Hence, aside from the oxygen content in the water column, the redox conditions and microbial oxygen demand in the sediment has to be considered as a further major determinant of ROL.  相似文献

Previous work demonstrated that a mixture of NH4Cl and KNO3 as nitrogen source was beneficial to fed-batch Arthrospira (Spirulina) platensis cultivation, in terms of either lower costs or higher cell concentration. On the basis of those results, this study focused on the use of a cheaper nitrogen source mixture, namely (NH4)2SO4 plus NaNO3, varying the ammonium feeding time (T = 7-15 days), either controlling the pH by CO2 addition or not. A. platensis was cultivated in mini-tanks at 30 °C, 156 μmol photons m−2 s−1, and starting cell concentration of 400 mg L−1, on a modified Schlösser medium. T = 13 days under pH control were selected as optimum conditions, ensuring the best results in terms of biomass production (maximum cell concentration of 2911 mg L−1, cell productivity of 179 mg L−1 d−1 and specific growth rate of 0.77 d−1) and satisfactory protein and lipid contents (around 30% each).  相似文献

A Bacillus sp., isolated from sludge and sediments of pulp and paper mill, was found to produce xylanase in a synthetic culture media containing oat spelt xylan (1% w/v) and 10% black liquor as inducers along with 2.5% (w/v) sucrose as additional carbon source. The purified enzyme was highly thermostable with half-life of 10 min at 90 °C and pH 8. The enzyme was stable over a broad range of pH (pH 6-10) and showed good thermal stability when incubated at 70 °C. Chemicals like EDTA, Hg2+, Cu2+ and solvents like glycerol and acetonitrile completely inhibited enzyme activity at high concentration. The molecular weights of the purified enzyme, determined by matrix-assisted laser desorption/ionization coupled with time-of-flight mass spectrometry (MALDI-TOF/MS) analysis was analogous to the results obtained from SDS-PAGE, i.e. 55 kDa. Kinetic parameters were determined by using oat spelt xylan as substrate. The KM and Vmax values of the enzyme were 4.4 mg/ml and 287 U/mg respectively. At high xylan concentrations (>70 mg/ml) a substrate inhibition phenomenon of the enzyme was observed. In addition, crude xylanase showed enormous potential for decolorization of various recalcitrant dyes.  相似文献

A novel nanocomposite material of multiwalled carbon nanotubes (MWCNTs) and room temperature ionic liquid (RTIL) N-butylpyridinium hexafluorophosphate (BPPF6) was explored and used to construct a novel microperoxidase-11 (MP-11) biosensor for the determination of hydrogen peroxide (H2O2). Cyclic voltammetry (CV) and differential pulse voltammetry (DPV) were used to characterize the performance of the biosensor. Under the optimized experimental conditions, H2O2 could be detected in a linear calibration range of 0.5 to 7.0 × 10−7 mol L−1 with a correlation coefficient of 0.9949 (n = 9) and a detection limit of 3.8 × 10−9 mol L−1 at 3σ. The modified electrodes displayed excellent electrochemical response, high sensitivity, long-term stability, and good bioactivity and selectivity.  相似文献
