The Mycobacterium avium complex (MAC) is a group of opportunistic human pathogens that may thrive in engineered water systems. MAC has been shown to occur in drinking water supplies based on surface water, but less is known about the occurrence and persistence of live cells and DNA in public hot water systems based on groundwater. In this study, we examined the occurrence of MAC in hot water systems of public day care centers and determined the persistence of live and dead M. avium cells and naked DNA in model systems with the modern plumbing material cross-linked polyethylene (PEX). The occurrence of MAC and co-occurrence of Legionella spp. and Legionella pneumophila were determined using cultivation and qPCR. Co-occurrences of MAC and Legionella were detected in water and/or biofilms in all hot water systems at temperatures between 40 and 54 °C. Moderate correlations were observed between abundance of culturable MAC and that of MAC genome copies, and between MAC and total eubacterial genome copies. No quantitative relationship was observed between occurrence of Legionella and that of MAC. Persistence in hot water of live and dead M. avium cells and naked DNA was studied using PEX laboratory model systems at 44 °C. Naked DNA and DNA in dead M. avium cells persisted for weeks. Live M. avium increased tenfold in water and biofilms on PEX. The results suggest that water and biofilms in groundwater-based hot water systems can constitute reservoirs of MAC, and that amplifiable naked DNA is relatively short-lived, whereas PEX plumbing material supports persistence and proliferation of M. avium.  相似文献

Pseudomonads adapt to various ecological niches by forming biofilms, which first requires bacterial adhesion on surfaces. We studied the influence of growth temperature on surface physicochemical properties of Pseudomonas fluorescens MF37 and on its adhesive capacities onto inert surfaces. It presented a global hydrophilic character, measured by microbial adhesion to solvent (MATS), and showed a cell surface more hydrophilic at 8 and 28°C than at 17°C. Moreover, P. fluorescens MF37 was more adhesive at 17°C. This critical temperature thus should be carefully taken into account in food safety. Adhesion onto inert surfaces is thus influenced by the growth temperature, which modifies the bacteria cell wall properties through changes in the outer membrane components. Therefore, we studied the effect of the loss of OprF, the major outer membrane protein, known to act as an adhesin (root, and endothelial cells). The OprF-deficient mutant was able to adhere to surfaces, but showed the same physicochemical and adhesion properties on abiotic surfaces whatever the growth temperature. OprF is thus not essential in this adhesion process. Potential nitrogen limitation to chl a production in surface waters of Saronicos Gulf, Aegean Sea was assayed using the alga Pavlova lutheri as the test organism. The oligotrophic and eutrophic water types of this area were compared by in situ and in vitro chl a production estimations. Additions of ammonium alone as well as in combination with complete nutrient enrichment were made to the oligotrophic waters and the algal growth yield was determined and compared with the corresponding yield in unenriched water cultures. The results from routine nutrient analysis and bioassay experimentation support the view that nitrogen has a priority among the factors limiting phytoplankton growth in the Eastern Mediterranean Sea.  相似文献

Leaves excised from shoot cultures of Prunus avium cvs. F12/1 and Charger and genotype 1908, and from five genotypes of P. serotina and two hybrids of P. avium×P. sargentii developed shoots on Woody Plant medium (WPM) supplemented with either benzyladenine (BA) or thidiazuron (TDZ). Regeneration in both P. avium 1908 and a genotype of P. serotina was improved using TDZ rather than BA in the medium. Regeneration occurred more frequently in P. serotina if leaves were cultured on medium with WPM rather than modified Driver and Kuniyuki walnut medium. The proportions of leaves that regenerated varied between genotypes of the same species. Regenerated shoots of both P. avium and P. serotina developed into shoot cultures following transfer to the media used to produce the shoot cultures used as explant sources. The opportunistic pathogen Mycobacterium avium is a significant inhabitant of biofilms in drinking water distribution systems. M. avium expresses on its cell surface serovar-specific glycopeptidolipids (ssGPLs). Studies have implicated the core GPL in biofilm formation by M. avium and by other Mycobacterium species. In order to test this hypothesis in a directed fashion, three model systems were used to examine biofilm formation by mutants of M. avium with transposon insertions into pstAB (also known as nrp and mps). pstAB encodes the nonribosomal peptide synthetase that catalyzes the synthesis of the core GPL. The mutants did not adhere to polyvinyl chloride plates; however, they adhered well to plastic and glass chamber slide surfaces, albeit with different morphologies from the parent strain. In a model that quantified surface adherence under recirculating water, wild-type and pstAB mutant cells accumulated on stainless steel surfaces in equal numbers. Unexpectedly, pstAB mutant cells were >10-fold less abundant in the recirculating-water phase than parent strain cells. These observations show that GPLs are directly or indirectly required for colonization of some, but by no means all, surfaces. Under some conditions, GPLs may play an entirely different role by facilitating the survival or dispersal of nonadherent M. avium cells in circulating water. Such a function could contribute to waterborne M. avium infection.  相似文献

Mycobacterium avium subsp. paratuberculosis is a pathogen which causes a debilitating chronic enteritis in ruminants. Unfortunately, the mechanisms that control M. avium subsp. paratuberculosis persistence during infection are poorly understood and the key steps for developing Johne's disease remain elusive. A proteomic analysis approach, based on one dimensional polyacrylamide gel electrophoresis (SDS-PAGE) followed by LC-MS/MS, was used to identify and characterize the cell wall associated proteins of M. avium subsp. paratuberculosis K10 and an cell surface enzymatic shaving method was used to determine the surface-exposed proteins. 309 different proteins were identified, which included 101 proteins previously annotated as hypothetical or conserved hypothetical. 38 proteins were identified as surface-exposed by trypsin treatment. To categorize and analyze these proteomic data on the proteins identified within cell wall of M. avium subsp. paratuberculosis K10, a rational bioinformatic approach was followed. The analyses of the 309 cell wall proteins provided theoretical molecular mass and p I distributions and determined that 18 proteins are shared with the cell surface-exposed proteome. In short, a comprehensive profile of the M. avium subsp. paratuberculosis K10 cell wall subproteome was created. The resulting proteomic profile might become the foundation for the design of new preventive, diagnostic and therapeutic strategies against mycobacterial diseases in general and M. avium subsp. paratuberculosis in particular.  相似文献

The saprophyticLeptospira biflexa serovarpatoc 1 did not undergo a major reduction in size upon starvation, but starvation did result in greater adhesiveness of the leptospires. Adhesion may provide a strategy for survival of leptospires in oligotrophic habitats because these bacteria can scavenge fatty acids adsorbed at surfaces. Provision of an energy substrate (fetal calf serum) to starved cells resulted in an increase in cell size and motility and a decrease in adhesion of the leptospires. Background  

Mycobacterium avium includes the subspecies avium, silvaticum, paratuberculosis and hominissuis, and M. avium subspecies has been isolated from various environments all over the world including from biofilms in water distribution systems. The aim of this study was to examine isolates of M. avium subsp. avium and M. avium subsp. hominissuis of different origin for biofilm formation and to look for correlations between biofilm formation and RFLP-types, and to standardise the method to test for biofilm formation. In order to determine the best screening method, a panel of 14 isolates of M. avium subsp. avium and M. avium subsp. hominissuis, were tested for their ability to form biofilm in microtiter plates under different conditions. Subsequently, 83 additional isolates from humans, swine and birds were tested for biofilm formation. The isolates were tested for the presence of selected genes involved in the synthesis of glycopeptidolipids (GPLs) in the cell wall of M. avium, which is believed to be important for biofilm formation. Colony morphology and hsp65 sequvar were also determined.  相似文献

Summary Several iron-regulated envelope proteins (IREPs), 11–180 kDa, have been detected in preparations of walls and membranes ofMycobacterium smegmatis, in an armadillo-derived mycobacterium (ADM) and inM. avium. The same sized proteins fromM. vaccae appeared under both iron-deficient and iron-sufficient growth conditions. Two larger proteins, of 240 and 250 kDa, appeared in the membranes ofM. smegmatis andM. avium only when grown iron-sufficiently but were constitutively present in both ADM andM. vaccae. The IREPs fromM. smegmatis were not induced under zinc-deficient growth conditions. Three of the four IREPs (14, 21 and 29 kDa) recognized inM. avium grown in vitro were also recovered from membrane fractions of the same strain grown in mice. In addition, these membranes contained both the high-molecular-mass proteins associated with iron-sufficient growth conditions. Membranes ofM. leprae, recovered from infected armadillos, showed the faint presence of a possible IREP at 29 kDa and wall preparations showed the presence of a 21-kDa protein. Membranes also contained the two larger proteins at 240 and 250 kDa. An explanation for the simultaneous occurrence of both low-iron-regulated and high-iron-regulated proteins is offered.  相似文献

Whereas the transfer of Listeria from surfaces to foods and vice versa has been well documented, little is known about the mechanism of bacterial transfer. The objective of this work is to gain a better understanding of the forces involved in listerial biofilms adhesion using atomic force microscopy (AFM). L. monocytogenes Scott A was grown as biofilms on stainless steel surfaces by inoculating stainless steel coupons with Listeria and incubating the coupons for 48 h at 32 °C with a diluted 1:20 tryptic soy broth. After growth, biofilms were equilibrated over saturated salt solutions at a constant relative humidity (%RH) before measurement of adhesion forces using AFM. The effects of contact time, loading force, and biofilm relative humidity (%RH) suggested that neither contact time, loading force nor biofilm %RH had a significant effect on biofilm adhesiveness at a cellular level (P > 0.05). In a second set of experiments, the influence of material type on biofilm adhesiveness was evaluated using two different colloidal probes (SiO2 and polyethylene). Results showed that the maximum pull-off force and retraction work needed to retract the cantilever for glass (−85.42 nN and 1.610−15 J, respectively) were significantly lower than those of polyethylene (−113.38 nN and 2.7 × 10–15 J, respectively; P < 0.001). The results of this study suggest that Listeria biofilms adhere more strongly to hydrophobic surfaces than hydrophilic surfaces when measured at a cellular level. These results provide important insights that could lead to new ways to remediate and avoid listerial biofilm formation in the food industry.  相似文献

Bacillus cereus is a ubiquitous endospore-forming bacterium, which mainly affects humans as a food-borne pathogen. Bacillus cereus can contaminate groundwater used to irrigate food crops. Here, we examined the ability of the emetic strain B. cereus F4810/72 to survive abiotic conditions encountered in groundwater. Our results showed that vegetative B. cereus cells rapidly evolved in a mixed population composed of endospores and asporogenic variants bearing spo0A mutations. One asporogenic variant, VAR-F48, was isolated and characterized. VAR-F48 can survive in sterilized groundwater over a long period in a vegetative form and has a competitive advantage compared to its parental strain. Proteomics analysis allowed us to quantify changes to cellular and exoproteins after 24 and 72 h incubation in groundwater, for VAR-F48 compared to its parental strain. The results revealed a significant re-routing of the metabolism in the absence of Spo0A. We concluded that VAR-F48 maximizes its energy use to deal with oligotrophy, and the emergence of spo0A-mutated variants may contribute to the persistence of emetic B. cereus in natural oligotrophic environments.  相似文献

The physiological responses and adaptive strategies of Populus euphratica Oliv. (arbor species), Tamarix ramosissima Ldb. (bush species), and Apocynum venetum L. (herb species) to variations in water and salinity stress were studied in the hyper-arid environment of the Tarim River in China. The groundwater table, the saline content of the groundwater, as well as the content of free proline, soluble sugars, plant endogenous hormones (abscisic acid (ABA), and cytokinins (CTK)) of the leaves of the three species were monitored and analyzed at the lower reaches of the Tarim River in the study area where five transects were fixed at 100 m intervals along a vertical sampling line before and after water release. Saline stress dramatically increased soluble sugar concentration of the three species. Differences in sugar accumulation were determined among the species at different transects. The free proline concentration of the leaves of T. ramosissima and P. euphratica showed a proportional decrease with various degrees of elevation of the groundwater table after water release. There was a least correlation between the soluble sugars and proline stimulation in T. ramosissima. It was strongly suggested that T. ramosissima developed a different strategy to accumulate organic solutes to adapt to the stress environment. The soluble sugars and proline accumulation responded to the changes of groundwater table independently: the former occurred under salt stress, whereas the latter was more significant under drought stress. The concentration and the increase in concentration of ABA and CTK involved in stress resistance of the three species were also determined. This increase in the hormone concentration in P. euphratica was different from that of the other two species. Expressed as a function of increase of ABA concentration in leaves, A. venetum and T. ramosissima showed a different solute accumulation in response to groundwater table. There was a significant correlation between ABA accumulation and Δ [proline] in A. venetum as well as between ABA accumulation and Δ [sugar] in T. ramosissima. Population densities and total phosphorus concentrations in samples from different lakes of south-eastern Norway were determined. In addition some transplant experiments with dilute phytoplankton populations were carried out. A laboratory batch culture method was used.The diatoms studied may be divided into three ecological groups based on their cell densities and total phosphorus concentrations in the samples. This classification was supported by the experimental results. Cyclotella spp., Asterionella formosa and Tabellaria fenestrata did not grow or had low growth rates above pH 9. Synedra cf. acus and Fragilaria crontonensis had often high growth rates within the pH 9–10 range, but were not able to grow at pH values above 10. High pH-values had no effect on the growth rate of Oscillatoria. Oscillatoria, Synedra and Stephanodiscus were severely growth-limited in filtered water from oligotrophic lakes. Maximum growth rates of all the populations studied were often obtained after addition of phosphate and chelated iron (FeEDTA) in combination to filtered water samples from oligotrophic/mesotrophic lakes.  相似文献

干旱区因降水稀少,地下水成为荒漠植被重要且稳定的水源。选取古尔班通古特沙漠南缘建群种植物梭梭(Haloxylon ammodendron)为研究对象,通过测量不同地下水埋深(3.45、9.08、10.47、13.27 m和15.91 m)下生长季前期和后期同化枝生理生化指标(黎明水势、正午水势、含水量、氯离子、钠离子、脯氨酸和非结构性碳水化合物)和生长与形态特征(生长速率和胡伯尔值),旨在认识荒漠植物对地下水埋深增加和季节性干旱的响应特征和调节适应机制。结果表明:(1)梭梭应对地下水埋深变化的生理调节对策,是采取先降低后升高黎明前同化枝水势、降低新枝形成期同化枝生长速率、增大胡伯尔值和积累非结构性碳水化合物的策略;(2)梭梭应对生长季大气干旱的生理调节对策,是通过降低黎明前同化枝水势、维持较高胡伯尔值、积累钠离子和消耗淀粉抵御季节性干旱;(3)在大气干旱与地下水水文干旱交互作用下,梭梭是采取降低正午同化枝水势、维持较高的同化枝含水量和积累可溶性糖的生态策略。综上所述,梭梭在响应地下水水文干旱和季节性大气干旱的生理特征间存在差异。研究结果丰富了水文和大气干旱对梭梭生理和生长影响的认知,可以为基于地下水资源管理的干旱区荒漠植被保育提供参考。  相似文献

Mycobacterium avium is an environmental mycobacterium that can be divided into the subspecies avium, hominissuis, paratuberculosis and silvaticum. Some M. avium subspecies are opportunistic pathogens for animals and humans. They are ubiquitous in nature and can be isolated from natural sources of water, soil, plants and bedding material. Isolates of M. avium originating from humans (n = 37), pigs (n = 51) and wild birds (n = 10) in Norway were examined by IS1245 and IS1311 RFLP using new and specific probes and for the presence of IS901 and ISMpa1 by PCR. Analysis and generation of a dendrogram were performed with the software BioNumerics.  相似文献

This study reports the development of a tool to characterise and differentiate northern Adriatic waters, particularly oligotrophic, high-salinity waters, based on the cellular fatty acids of culturable heterotrophic bacterioplankton. The growth abilities and population dominance were observed for particle-attached and free-living bacteria cultured in three types of media: Marine Broth, diluted Marine Broth (1:10) and R2 broth. Three groups of water layers were distinguished by hierarchical clustering analysis: eutrophic, oligotrophic and oligotrophic nutrient-selected. Significant differences between the resulting groups were tested by two-way ANOVA (with replication). Eutrophic layers were characterised by readily culturable particle-attached and free-living fractions of the bacterial community in all three media, all dominated by fast-growing γ-Proteobacteria. In contrast, oligotrophic water layers with low productivity had a much weaker culturability and a different population dominance for the free-living community, as compared to their attached or growth-arrested counterparts, for all media. The free-living bacteria from strictly oligotrophic environments demonstrated minimum culturability in Marine Broth, while those from selective oligotrophic environments were culturable and were dominated either by CytophagaFlavobacter complex, α-Proteobacteria or γ-Proteobacteria. The conclusive evidence regarding the selective and refractory nature of organic compounds in these waters demonstrates the dominant culturability of the CytophagaFlavobacter complex and α-Proteobacteria in free-living communities in all growth-media. The response of fatty acid dominance ratios depends significantly on the trophic state and fraction (p < 0.05), although the effect of the trophic state is completely different in attached and free fractions. Both fractions were tested separately, demonstrating a significant influence of the trophic state (p < 0.05), while the effect of the media on the fatty acid response was not significant (p > 0.05). An interaction between media and trophic status was present in the attached fraction (p < 0.05), yet this was not observed in the free fraction (p > 0.05), indicating that any systematic difference between trophic states was the same for each media tested. Accordingly, the free-living fraction of bacterioplankton is a more informative attribute and can be used solely as an indicator of the water layer trophic condition.  相似文献

Summary Examples are given of initial guanotrophy in acid oligotrophic environment and of stabilized guanotrophy of long standing, both in acid oligotrophic and eutrophic environment.Characteristic of a guanotrophic environment is the accumulation of phosphate. In hard waters phosphate is accompanied by large amounts of saline nitrogen compounds. In soft waters the saline nitrogen production seems to be inhibited through low bacteriological activity. In eutrophic guanotrophic environments oxygen production and biochemical oxygen demand are both high. In oligotrophic guanotrophic environments the oxygen content as a rule does not reach saturation and biochemical oxygen demand is fairly low. The amounts of chloride and calcium do not change significantly by the addition of excrements of birds.Diatoms, blue green algae and desmids are very scarce in guanotrophic environments. In hard waters mesosaprobic forms are present in the plankton community. In soft waters no strong saprobic tendencies could be found.During the initial stage of guanotrophy in an acid oligotrophic environment, the desmids disappear together with other sensitive species as Dinobryon pediforme and Bosmina obtusirostris. Unicellular flagellates as Chlamydomonas appear in increasing numbers. Several factors in the environment respond to the changes by increasing fluctuations of the values recorded.There was evidence of transport and introduction of microorganisms by waterfowl.  相似文献

The photosynthetic rates of Trentepohlia odorata (L.) Martius growing on wall surfaces in Singapore changed throughout the day with a maximum in midmorning and decreasing thereafter during the day. Optimum temperature for photosynthesis was 25° C. Different levels of air humidity also affected photosynthetic rates with low relative humidity reducing the rates and efficiency of photosynthesis. Our results suggested that T. odorata was able to maximize its rate of photosynthesis before photoinhibitory light levels were reached and that its growth might be dependent on high levels of atmospheric relative humidity, which may serve as a source of water supply for the alga.  相似文献
